Abstract
métodos para sintetizar polissacarídeos anticoagulantes. a presente invenção inclui métodos para preparar polissacarídeos anticoagulantes usando várias enzimas sulfotransferases modificadas que não ocorrem naturalmente que são designadas para reagir com compostos de sulfato de arila ao invés do substrato natural, 5'-fosfossulfato de 3'-fosfoadenosina, para facilitar a transferência de grupo sulfo para polissacarídeos aceptores de grupo sulfo. os compostos de sulfato de arila adequados incluem, mas não são limitados a sulfato de p-nitrofenila ou sulfato de 4-nitrocatecol. polissacarídeos anticoagulantes produzidos pelos métodos da presente invenção compreendem resíduos de glucosamina n-, 3-o-, 6-o-sulfatada e resíduos de ácido hexurônico 2-o sulfatado, têm atividade anticoagulante comparável em comparação a polissacarídeos anticoagulantes comercialmente disponíveis, e podem ser utilizados para formar polissacarídeos anticoagulantes truncados tendo um peso molecular reduzido.
