Abstract
a presente invenção refere-se a um ácido nucleico indutor do rna de interferência e o uso do mesmo, e mais particularmente a um ácido nucleico indutor do rna de interferência compreendendo pelo menos uma fita simples de fitas duplas, pelo menos uma fita simples, compreendendo uma modificação substituída por um espaçador, que não é capaz de formar um par de base na região de extremidade 5' ou extremidade 3'. o ácido nucleico indutor do rna de interferência da presente invenção é uma nova forma modificada do nucleotídeo provido para evitar os efeitos ligação-inespecífica, oferecendo um método para reprimir seletivamente a expressão de genes alvo. além disso, de acordo com a presente invenção, o ácido nucleico indutor do rna de interferência provê novas formas modificadas que são seletivas e alvo-específicas como um método para bloquear efeitos ligação-inespecífica enquanto silencia a expressão de genes alvo, enquanto o uso de ácido nucleico de indução de interferência dos rnas convencional causa imprecisão e efeitos adversos por meio das ligações não específicas, assim, o ácido nucleico indutor do rna de interferência foi oferecido para resolver o problema, em que ele será amplamente usado como um método para reprimir a expressão de genes em pesquisa e para terapia de genes sem se preocupar com efeitos ligação-inespecífica.
