resumo método para identificar um polipeptídeo tendo uma funcionalidade ou especificidade de ligação desejadas, biblioteca de moléculas de polinucleotídeo, método para gerar uma biblioteca de polinucleotídeos, vetor, célula hospedeira, anticorpo e método para gerar uma população de células hospedeiras a presente invenção refere-se a um método que oferece eficiência, flexibilidade, utilidade e velocidade incomparáveis para a descoberta e otimização de polipeptídeos com funcionalidade e/ou especificidade de ligação desejadas, incluindo moléculas de ligação a antígeno, tais como anticorpos e fragmentos dos mesmos, para fenótipos funcionais e/ou de ligação desejados. o novo método baseia-se em construções de transposição e bibliotecas de adn diversificadas clonadas em vetores de transposição e em sua transfeção em células hospedeiras por expressão transitória concomitante de uma enzima transponase funcional. isto assegura uma introdução eficiente e estável de vetores de expressão baseados na transposição de células hospedeiras de vertebrados em uma etapa, que podem depois ser classificados em relação a um fenótipo funcional ou de ligação desejado das proteínas expressas, após o que as sequências codificantes relevantes para as proteínas expressas, incluindo anticorpos e fragmentos dos mesmos, podem ser identificadas por técnicas de clonagem e de sequenciação de adn convencionais. ?? ?? ?? ?? 1/1
