Abstract
"MOLéCULA DE áCIDO NUCLEICO ISOLADA, CéLULA HOSPEDEIRA, POLIPEPTìDEO ISOLADO, ANTICORPO, MéTODOS PARA PRODUZIR UM POLIPEPTìDEO E PARA DETECTAR A PRESENçA DE UM POLIPEPTìDEO E DE UMA MOLéCULA DE áCIDO NUCLEICO, KIT MéTODOS PARA PRODUZIR ACETALDEìDO E ETANOL, EXTRATO DE ENZIMA, MéTODO PARA SELECIONAR UMA ENZIMA DE PIRUVATO DESCARBOXILASE, E, PLASMìDEO". A invenção fornece moléculas de ácido nucleico isoladas que codificam as enzimas da piruvato descarboxilase tendo atividade de descarboxilase, afinidade de substrato, termoestabilidade e atividade em pH diferente melhoradas. Os ácidos nucleicos da invenção também têm um uso de códon que permite a alta expressão em uma variedade de células hospedeiras. Conseq³entemente, a invenção fornece vetores de expressão recombinantes contendo tais moléculas de ácido nucleico, células hospedeiras recombinantes que compreendem os vetores de expressão, células hospedeiras que compreendem ainda outras enzimas etanologênicas e métodos para produzir substâncias úteis, por exemplo, acetaldeído e etanol, usando tais células hospedeiras.
