La présente invention concerne des dérivés d'amino-alcools organiques répondant à la formule :

dans- laquelle R est un groupe acylamido dérivant d'un acide aliphatique inférieur éventuellement substitué par un ou des atomes d'halogène ou -un groupe benzoylamido ou analogue.
De tels dérivés contiennent deux atomes de carbone asymétriques et la formule ci-dessus représente par conséquent aussi bien les formes isomères de structure (érythro ou thréo) que les isomères optiques (D et L) et les racémiques des deux séries érythro et thréoo
Un objet de la présente invention est de fournir un procédé pour l'épimérisation des formes érythro de ces composés. Un autre objet., correspondant à un mode de réalisation préféré de la présente invention, est de fournir un procédé technique avantageux pour la transformation des formes DL et L érythro du dichloracétamido-2 p-nitrophényl-1 propanediol- 1,3 dans les formes correspondantes DL et D-thréo qui sont des composés thérapeutiques de valeur, L'isomère D-thréo étant l'antibiotique connu sous le nom de Chloramphénicol.

c'est-à-dire des érythro acylamido-2 nitrophényl-1 propanediols-1,3 le- quel procédé comporte l'estérification d'un isomère optique ou d'un racémi- que de la forme érythro d'un tel diol pour former le diester nitrique 1,3 de ce dernier., le traitement du diester par un alcali dans des conditions favorisant l'hydrolyse sélective du groupement ester nitrique en position 1 (par opposition à l'élimination des deux groupes esters) avec cyclisa'- tion simultanée en une oxazoline, le traitement de l'oxazoline produite dans la réaction par un acide, ce qui;
, par scission de l'oxazoline, donne naissance à un propanol, le traitement dudit propanol par un alcali, ce qui cause l'hydrolyse du groupe ester nitrique en position 3 avec cyclisation simultanée en une oxazoline le traitement de l'oxazoline résultante par un acide pour en effectuer la scission et enfin la neutralisation du mélange réactionnel.
L'invention concerne spécialement l'épimérisation de l'érythro dichloracétamido-2 p-nitrophényl-1 propanediol-1,3 et dans ce cas la suite des réactions peut être représentée comme suit :

L'inversion de la structure conduisant à la forme thréo se pro- duit au stade de formation de l'oxazoline-1,2 désignée par (c) par opposi- tion à la formation de l'oxazoline-2,3 désignée Ce)
La phase initiale de la formation du diester nitrique est effec- tuée en traitant l'érythrodiol par l'acide nitrique de densité 1,50 à une température voisine de 0 et de préférence nexcédant pas 5 . La deuxième pha-

se c'est-à-dire la transformation du diester en monoester d'oxazoline par traitement avec un alcali est de préférence effectuée à froid en solution ou en suspension dans un milieu approprié tel qu'un alcool aqueux, le métha- nol ou l'éthanol convenant bien. L'hydroxyde de sodium est normalement l'al- cali de choix.
La scission de l'oxazoline résultante, par exemple celle qui est appelée (c) c'est-à-dire la troisième phase du procédé, est comme on l'a déjà indiqué effectuée par traitement au moyen d'un acide, de préféren- ce un acide minéral tel que l'acide chlorhydrique, et de préférence à froid.
Si on désire isoler le produit résultant, par exemple celui qui est appelé ci-dessus (d), le mélange réactionnel est neutralisé au moyen d'une base convenable et le produit solide ainsi obtenu alors traité par un alcali dans les mêmes conditions que celles qui sont décrites ci-dessus pour la seconde phase c'est-à-dire la conversion du diester en monoester d'oxazo- lire. Alternativement la solution acide résultant de la troisième phase du processus peut être traitée directement au moyen d'un excès d'alcali avec conversion in situ du produit réactionnel du type (d) en hydroxy-oxazoline du type (e).
La dernière phase de la réaction est effectuée en traitant l'hy- droxyoxazoline du type (e) au moyen d'un acide, de préférence l'acide chlor- hydrique à froid, et neutralisant ensuite le mélange réactionnel au moyen d'une base convenable telle que l'ammoniaque ou le bicarbonate de sodium. Un excès de base forte doit être évité car le diol résultant est sensible à une telle base. La phase de neutralisation est de préférence effectuée au voisi- nage de la température ordinaire.
Le procédé de la présente invention permet de passer facilement de la série érythro à la série thréo. Un mode de réalisation préféré de la présente invention consiste à traiter le DL-érythro ou L-érythro dichlora- cétamido-2 p-nitrophényl-1 propanediol-1,3 selon le mode opératoire décrit ci-dessus, et à isoler du mélange réactionnel le diol DL thréo ou D thréo correspondant. Ce mode de réalisation préféré permet de façon relativement simple et efficace la préparation des composés thréo à partir des formes DL- et L-érythro de l'amino-2 p-nitrophényl-1 propanediol-1,3 qui peuvent être transformés facilement en les formes DL- et L-érythro du dichloracé- tamido-2 p-nitrophényl-1 propanediol-1,3.
Le procédé de la présente invention offre un certain nombre d'avantages techniques. C'est ainsi, par exemple, que toutes les phases de la réaction peuvent être effectuées dans des conditions de température modérées et conduisent à des produits contenant relativement peu d'impure- tés accessoires; il en résulte qu'il est avantageux et même préférable, tel qu'il est décrit dans les exemples suivants, d'isoler les intermédiaires sans les purifiero Un avantage supplémentaire est qu'on obtient des rende- ments élevéso
Le procédé de la présente invention est illustré par les exem- ples suivants :
EXEMPLE 1 -
6,8 g de diester nitrique du DL-érythro dichloracétamido-2 o- nitrophényl-1 propanediol-1,3 finement pulvérisé (P.F. 152 avec décompo- sition) sont mis en suspension dans 50 cm3 de méthanol et on ajoute 10 cm3 d'une solution de soude 2N. On abandonne une heure à la température ordinai- re et précipite plus complètement le produit par addition d'eau. Le solide jaune pâle est filtré et lavé d'abord à l'acide acétique dilué, puis à l'eau.
Le rendement (5,5 g) représente 95 % de la théorie. Le point de fusion est de 95=97 , le produit pur fondant à 103 . Le produit possède la structure suivante :

Le produit ci-dessus est broyé avec 90 cm3 d'acide chlor- hydrique concentré et après repos d'une heure à la température ordinai- re on filtre le chlorhydrate solide et on le lave avec une petite quanti- té d'acide chlorhydrique concentré. Le chlorhydrate humide est mis en suspension dans 50 cm3 d'eau et on ajoute du bicarbonate de sodium jus- qu'à ce que le milieu devienne alcalin au tournesol. Après avoir laissé reposer quelques temps, on filtre le produit (5,6 g) et on le lave à l'eau. Le produit brut fond à 127-134 (P.F du produit pur 139-140 ).
On l'utilise tel quel dans la phase suivante :
Le produit obtenu précédemment est dissous dans 45 cm3 de méthanol; on ajoute 1,45 cm3 d'une solution de soude à 500 g par litre, et maintient la solution pendant deux heures à 30-40 . Pendant ce temps le mélange réactionnel devient jaune et des cristaux se séparent. La suspension est diluée à l'eau et le solide filtré, puis lavé, d'abord à l'acide acétique dilué et ensuite à l'eau (le point de fusion est 166 , le produit pur fondant à 169 ). Le rendement (3,9 g) représente 83 % de la théorie. Le produit possède la structure suivante

Dans la phase finale on hydrolyse l'oxazoline au moyen d'a- cide chlorhydrique en triturant le produit ci-dessus avec 9 cm3 d'acide chlorhydrique concentré et laissant la suspension reposer 10 minutes à la température ordinaire.
L'excès d'acide chlorhydrique est alors neutralisé au moyen de bicarbonate de sodium solide ajouté au mélange réactionnel dilué. L'huile qui précipite se solidifie par repos de 4 heures à la tem- pérature ordinaire. On filtre le solide obtenu., on le lave à l'eau et on le sèche. Il fond à 116-117 , le point de fusion du produit pur étant 121 . Le rendement en DL-thréo dichloracétamido-2 o-nitrophényl-1 propa- nediol-1,3 (3,7 g) représente 89% de la théorie. Le rendement global de l'inversion est 70 % de la théorie.
Le diester nitrique utilisé comme matière première est obte- nu de la façon suivante :
25 g de DL-érythro dichloracétamido-5 phényl-4 méthyl-2 dio- xane -1,3 sont ajoutés en une demi-heure sous agitation à 75 cm3 d'acide nitrique de densité 1,50 refroidi à -40 . La suspension agitée s'échauffe spontanément jusqu'à 0 et est maintenue à cette température pendant une demi-heure. La solution résultante est versée sur un mélange agité de glace pilée et d'eau.
Le solide ainsi obtenu est filtré et lavé à fond à l'eau- Deux cristallisations dans le méthanol fournissent à l'état pur le dinitrate du DL-érythro dichloracétamido-2 o-nitrophényl-1 propanediol-

1,3 fondant à 152 avec décomposition.
EXEMPLE 2.
6,46 g de DL-érytho dichloracétamido-2 p-nitrophényl-1 propa- nediol-1,3 en poudre sont ajoutés en 10 minutes sous agitation, à 12 cm3 d'acide nitrique (D = 150) à une température d'environ -30 . L'acide ni- trique a été préalablement traité à l'acide sulfamique pour éliminer l'a- cide nitreux. On agite le mélange réactionnel et laisse la température mon- ter à + 10 . La solution claire incolore résultante est versée sur un mé- lange glace-eau agité ce qui provoque la précipitation du produit sous for- me d'un solide granuleux blanc. Le dinitrate de DL-érythro dichloracétamido- 2 p-nitrophényl-1 propanediol-1,3 brut est filtré, lavé à l'eau et séché.
Le'rendement en produit brut (8,05 g) représente 97,5 % de la théorie. Le produit fond à 133-136 . Le produit recristallisé fond à 145-146 . Toute- fois,, le produit brut est suffisamment pur pour la phase suivante.
4,03 g de dinitrate du DL-érythro dichloracétamido-2 p-nitro- phényl-1 propanediol-1,3 brut sont mis en suspension dans25 cm3 de métha- nol à la . A la suspension agitée on ajoute 6 cm3 d'une solution de soude 2N ce qui fluidifie considérablement la suspension mais après quelques minutes, des cristaux cubiques jaune pâle précipitent. La suspension jaune pâle est abandonnée au repos pendant une heure à 0 , les cristaux sont séparés par filtration et lavés avec un peu d'eau froide, puis avec du méthanol. Le pro- duit de réaction brut (3,12 g) est obtenu avec un rendement de 91 % de la théorie. Il fond à 101-102 , le produit recristallisé fondant à 104-105 .
Le produit obtenu est vraisemblablement l'ester nitrique de la DL-thréo dichlorométhyl-2 hydroxyméthyl-4 p-nitrophényl-5 # 2-oxazoline ayant la structure suivante ;

Le produit brut est suffisamment pur pour être utilisé dans la pha.se suivante sans autre purification. La potasse, le carbonate de potas- siumle triéthylamine, etc.., peuvent être utilisés comme agents d'élimina- tion de 1'acide nitrique mais le rendement n'est pas aussi élevé que lors- qu'on emploie la soude dans ce but.
Les 3el2 g d'ester nitrique de la DL-thréo dichlorométhyl-2 hy- droxyméthyl-4 p-nitrophényl-5 # 2 oxazoline obtenus ci-dessus sont tritu- rés avec 20 cm3 d'acide chlorhydrique concentré et la suspension crémeuse du chlorhydrate abandonnée au repos à la température ordinaire pendant une de- mi-heure. Le solide est filtré et lavé avec quelques gouttes d'acide chlorhy- drique concentré froid. Le chlorhydrate est remis en suspension dans 40 cm3 d'eau froide et on ajoute un excès de bicarbonate de sodium solide suffisant pour rendre le milieu juste alcalin. Le produit de réaction brut est filtré, lavé avec un peu / d'eau et séché à l'air. Le rendement (3,22 g) est 98 % de la théorie.
Le moncester en 3 du DL-thréo dichloracétamido-2 p-nitrophényl-1 propanediol-1,3 brut fond à 115-120 , le produit recristalliséfondant à 119 .
Le produit brut est suffisamment pur pour la phase suivante.
2,46 g de l'ester nitrique en 3 du DL-thréo dichloracétamido-2 p-nitrophényl-1 propanediol-1,3 brut obtenus ci-dessus sont dissous dans 6 cm3 de méthanol et additionnés de 3,5 cm3 d'une solution de soude 2N. Cette solu- tion, après repos d'une demi-heure à la température ordinaire, laisse déposer des cristaux de DL-thréo dichlorométhyl-2 p-nitrophénylhydroxyméthyl-4 # 2 oxazolineo Après repos de deux heures et demie la suspension épaisse est diluée par un peu,de méthanol et les cristaux jaune pâle sont filtrés et lavés au méthanol.
La DL-tbréo dichlorométhyl-2 p-nitro-phénylhydroxyméthyl-4 # 2-oxa- zoline brute est obtenue avec un rendement de 82 % de la théorie (1,7 g) et fond à 159-161 , l'oxazoline pure fondant à 162-163 . Le produit brut peut

être employé sans autre purification dans la phase suivante.

de 1,5 g de DL-thréo dichlorométhyl-2 p-nitrophényl-hYdroxymé-thYl-4,a 2- oxazoline brute dans 5 cm3 d'acide chlorhydrique concentré froid. Après deux minutes de repos, il se sépare un chlorhydrate cristallisé. Après repos d'une demi-heure on obtient une bouillie épaisse. Pour faciliter la filtration, la masse est diluée au moyen d'acide chlorhydrique concen- tré et le chlorhydrate est filtré et lavé avec une petite quantité supplé- mentaire d'acide chlorhydrique concentré. Le chlorhydrate humide est dis- sous dans la quantité minimum d'eau distillée, la solution est filtrée, puis neutralisée à l'ammoniaque ou au bicarbonate de sodium.
La suspen- sion est abandonnée au repos jusqu'à fin de cristallisation. On sépare le chloramphénicol racémique, on le filtre et on le sèche.
Le rendement en chloramphénicol racémique (1,5 g) est 94,5 % de la théorie. Le point de fusion est 149-150 . Le produit possède la struc- ture suivante :

qui est la même que celle du produit de départ sauf qu'il appartient à la série thréo et non à la série érythro.
Le rendement global pour l'inversion complète est 68 % de la théorie.
EXEMPLE 3.
4 g de L-érythro dichloracétamido-2 p-nitrophényl-1 propane- diol-1,3 [[alpha]]D 26 = -20,6 ; ( c 4% dans l'acétone) sont ajoutés en l'espace de deux minutes, vers -40 , à 7,45 cm3 d'acide nitrique (D 1,50), préalablement traité à l'acide sulfamique pour éliminer toute tra- ce d'acide nitreux. L'agitation est poursuivie et la suspension s'échauffe jusqu'à 0 en 40 minutes environ. On obtient à 0 une solution complète et incolore qu'on verse sur un'mélange agité de glace et d'eau. Après repos d'une nuit, le produit légèrement collant durcit complètement. On le fil- tre et sèche sous vide.
On obtient 4,80 g de diester nitrique du L-érythro dichloracétamido-2 p-nitrophényl-1 propanediol-1,3 brut ce qui représente 93,5. de la théorie. Le point de fusion est 126-128 et le pouvoir rota- toire + 8,92 ( c = 4 % dans l'acétone).
3,57 g de diester nitrique brut obtenus comme ci-dessus sont mis en suspension à la dans 21 cm3 de méthanol et on ajoute 4,02 cm3 d'une solution de soude 2N. L'ester se dissout rapidement et après 30 secondes il se dépose une huile. Après repos d'une heure à 0 , le liquide surnageant est éliminé et remplacé par 17,8 cm3 d'acide'

chlorhydrique concentré. Par trituration on provoque la précipitation d'un chlorhydrate blanc. Après repos d'une demi-heure le solide est filtré et lavé avec un peu d'acide chlorhydrique.

fond à 136-139 et a un pouvoir rotatoire de -169,9 (c = 1 % dans l'acétate d'éthyle). Par cristallisation dans le méthanol on ob- tient un produit ayant un point de fusion de 141-142 et un pouvoir rota- toire [[alpha]] D 20 = 172 ( c = 1 % dans l'acétate d'éthyleo)
0,5 g d'oxazoline brute sont mélangés avec 2,5 cm3 d'acide chlorhydrique concentré et la suspension blanche qui se forme rapidement est abandonnée au repos sur la glace pendant une heure. Le solide est fil- tré et lavé avec 1 cm3 d'acide chlorhydrique concentré. Le résidu est dis- sous dans 5 cm3 d'acide chlorhydrique N/2 à 30 et la solution est filtrée.
Elle est rendue juste alcaline au tournesol par addition de bicarbonate de sodium solide. Après concentration sous pression réduite à volume moitié, le chloramphénicol formé est filtré, layé à l'eau glacée et séché au vide.
Le produit fond à 150-152 et le pouvoir rotatoire [[alpha]] 4 = -19,81 (c 4 % dans l'acétate d'éthyle). / D EXEMPLE 4.
Le DL-érythro dichloracétamido-2 p-nitrophényl-1 propanediol- 1,3 est transformé en dinitrate comme décrit dans l'exemple 2.*2,06 g du di- nitrate de DL-érythro dichloracétamido-2 p-nitrophényl-1 propanediol-1,3 brut sont mis en suspension dans 20 cm3 de méthanol à 10 et on ajoute 2,75 cm3 d'une solution de soude 2N. Le solide blanc se redissout et, au bout d'une minute, il se sépare des cristaux jaune pâle de l'ester nitrique de la DL-thréo dichlorométhyl-2 hydroxyméthyl-4 p-nitrophényl-5 # 2-oxazo- lineo Après avoir maintenu la suspension pendant une demi-heure à 10 , on la laisse se réchauffer jusqu'à la température du laboratoire pendant une demi-heure.
La suspension est juste alcaline au Jaune Brillante
La suspension est rendue neutre au tournesol par addition de 0,3 cm3 d'acide chlorhydrique 2N et on ajoute encore 2,75 cm3 d'acide chlor- hydrique 2N. La suspension commence par se fluidifier considérablement mais elle s'épaissit par la suite avec séparation d'aiguilles de l'ester nitr.- que en 3 du DL-thréo dichloracétamido-2 p-nitrophényl-1 propanediol-1,3.
L'hydrolyse de l'oxazoline se produit à la température ordinaire en une heu- re.
La. suspension est neutralisée au moyen d'une solution de soude 2N puis on ajoute une quantité égale de la même solution de soude. La solu- tion est abandonnée au repos pendant une nuit à la température ordinaire et il se forme des cristaux jaune pâle de DL-thréo dichlorométhyl-2 p-ni- trophénylhydroxyméthyl-4 % 2-oxazoline. La suspension n'est plus alcaline au Jaune Clayton. On ajoute alors 0,5 cm3 d'une solution de soude 2N et aban- donne la suspension au repos pendant une heure.
A ce moment il peut être parfois avantageux d'isoler l'oxazoline ce qui en permet une purification efficace;, car l'oxazoline est peu soluble dans ce milieu. Toutefois dans l'essai correspondant au présent exemple la suspension orange est neutralisée au moyen d'acide chlorhydrique 2N, puis on ajoute encore 2,5 cm3 d'acide chlorhydrique 2N. Après agitation pendant 10 minutes le solide 'se redissout en donnant une solution louche orange.
Le chloramphénicol racémique ainsi obtenu est extrait à l'acétate d'éthy-
