Abstract

La presente divulgación proporciona composiciones y métodos para editar un sitio diana de un genoma de planta mediante el suministro de componentes de edición funcionales utilizando virus mosaico del tabaco modificado (mTMV). Los métodos divulgados en la presente pueden utilizarse para suministrar un sistema de edición de genes, tal como una ADN endonucleasa, a una célula de planta de tabaco para la modificación de un sitio diana del genoma de la planta. Además, los métodos y composiciones divulgados en la presente proporcionan la producción de una molécula que codifica una meganucleasa in vitro antes del suministro del ARN a una célula de planta. Después de la introducción de la molécula de ácido nucleico que codifica un componente de edición funcional y subsiguiente expresión de los componentes de edición funcionales, la planta puede cultivarse y permitírsele producir semillas que tienen una edición en el sitio diana genómico. Las semillas pueden seguidamente experimentar un rescate de embriones y se los puede cultivar para producir una planta modificada sin material genético heterólogo.
