Abstract
Como resultado de los descubrimientos, es posible medir con precisión la actividad del FVIII en el plasma de un paciente que padece hemofilia A por medio de un análisis de la coagulación con una sola etapa basado en el APTT, y también es posible medir con precisión la titulación de los inhibidores del FVIII en el plasma de un paciente que padece hemofilia A y que ha sido sometido a un tratamiento con inhibidores del FVIII por medio de un análisis de Bethesda basado en el APTT. Reivindicación 1: Un método para medir la reactividad del factor de coagulación VIII, caracterizado porque comprende el paso de poner en contacto (1) una muestra derivada de la sangre que contiene una sustancia que tiene una actividad relacionada con la sustitución de la función del factor de coagulación VIII, con (2) una o más sustancias útiles para neutralizar la sustancia que tiene una actividad relacionada con la sustitución de la función del factor de coagulación VIII. Reivindicación 3: El método ole acuerdo con la reivindicación 1 ó 2, caracterizado porque el anticuerpo biespecífico toma la forma de cualquiera de los anticuerpos que se describen más adelante, que comprenden un primer polipéptido asociado a un tercer polipéptido y un segundo polipéptido asociado a un cuarto polipéptido: un anticuerpo biespecífico en el cual el primer polipéptido es una cadena H que consiste en una secuencia de aminoácidos como la que se detalla en SEQ ID Nº 9, el segundo polipéptido es una cadena H que consiste en una secuencia de aminoácidos como la que se detalla en SEQ ID Nº 11 y el tercer polipéptido y el cuarto polipéptido son cadenas L comunes como la que se detalla en SEQ ID Nº 10 (Q499-z121 / J327-Z119 / L404-k); o un anticuerpo biespecífico en el cual el primer polipéptido es una cadena H que consiste en una secuencia de aminoácidos como la que se detalla en SEQ ID Nº 36, el segundo polipéptido es una cadena H que consiste en una secuencia de aminoácidos como la que se detalla en SEQ ID Nº 37 y el tercer polipéptido y el cuarto polipéptido son cadenas L comunes como la que se detalla en SEQ ID Nº 38 (Q153-G4K / J142-G4H / L180-K). Reivindicación 8: Un conjunto de elementos caracterizado porque resulta útil en un método de acuerdo con cualquiera de las reivindicaciones 1 a 7, que comprende uno o más anticuerpos que se seleccionan del grupo que consiste de un anticuerpo que puede unirse a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación IX, un anticuerpo que puede unirse a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación IX activado, un anticuerpo que puede unirse a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación X, un anticuerpo que puede unirse a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación X activado y un anticuerpo biespecífico que puede unirse a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación X y/o el factor de coagulación IX activado y a un fragmento Fab que comprende un sitio de unión al antígeno al que puede unirse el factor de coagulación X y/o el factor de coagulación X activado. Reivindicación 10: Un método para diagnosticar la gravedad de la enfermedad que padece un paciente que ha sido sometido a un tratamiento con una sustancia que tiene una actividad relacionada con la sustitución de la función del factor de coagulación VIII, caracterizado porque está basado en el uso de un método de acuerdo con cualquiera de las reivindicaciones 1 a 7.
