Abstract
Se describen vectores de expresión y métodos de uso de los mismos para mejorar la producción de proteínas recombinantes en plantas o células de las plantas. La producción puede mejorarse en forma adicional ante la co-expresión de la proteína P19 que suprime el silenciamiento génico del virus del enanismo ramificado del tomate. Preferentemente, las proteínas recombinantes son enzimas y/o anticuerpos terapéuticos, y los métodos se llevan a cabo en Nicotiana benthamiana -opcionalmente, una cepa glicomodificada sobre la base de ARNi- o en Nicotiana tabacum, cultivar Little Crittenden. Reivindicación 1: Un vector de expresión caracterizado porque comprende: (a) un promotor seleccionado entre (i) el promotor 35S del virus del mosaico de la coliflor (CaMV) o (ii) el promotor del gen de subunidad pequeña ribulosa bisfosfato carboxilasa (rbc) de Chrysanthemum morifolium; (b) una región no traducida (UTR) 5 seleccionada entre (i) la UTR 5 de 35S de CaMV o (ii) la UTR 5 del gen de subunidad pequeña rbc de C. morifolium; y (c) una UTR 3 y secuencia terminadora seleccionada entre (i) la UTR 3 y secuencia terminadora del gen nopalina sintasa (nos) de Agrobacterium, (ii) la UTR 3 y secuencia terminadora del gen osmotina (osm) de Oryza sativa, (iii) la UTR 3 y secuencia terminadora del gen de subunidad pequeña rbc de C. morifolium o (iv) una versión truncada, por 162 pb de acuerdo con lo definido por un sitio de reconocimiento de BspEl, de la UTR 3 y secuencia terminadora del gen de subunidad pequeña rbc de C. morifolium. Reivindicación 7: El vector de expresión de acuerdo con la reivindicación 6, caracterizado porque el anticuerpo es trastuzumab o bevacizumab.
