Abstract
Se proporciona a-amilasa fúngica de Aspergillus clavatus (AcAmy1). La AcAmy1 tiene un pH óptimo de 4,5 y puede operar a 30 - 75ºC, lo que permite usar la enzima en combinación con una glucoamilasa en una reacción de sacarificación. Esto obvia la necesidad de realizar una reacción de sacarificación en forma de un proceso discontinuo, donde se deben reajustar el pH y la temperatura para el uso óptimo de la alfa-amilasa o glucoamilasa. La AcAmy1 también cataliza la sacarificación de los sustratos de almidón a una composición de oligosacárico enriquecida significativamente en DP2 y (DP1 + DP2) en comparación con los productos de la sacarificación catalizados por una a-amilasa de Aspergillus kawachii. Esto facilita, por ejemplo, la utilización de la composición de oligosacáridos por parte de un organismo fermentador en un proceso de sacarificación y fermentación simultáneos.
