Abstract
Se proporciona a-amilasa fúngica de Aspergillus clavatus (AcAmy1). La AcAmy1 tiene un pH óptimo de 4,5 y puede operar a 30 - 75ºC, lo que permite usar la enzima en combinación con una glucoamilasa y una isoamilasa en una reacción de sacarificación. Esto obvia la necesidad de realizar una reacción de sacarificación en forma de un proceso discontinuo, donde se deben reajustar el pH y la temperatura para el uso óptimo de la a-amilasa o glucoamilasa. La AcAmy1 también cataliza la sacarificación de los sustratos de almidón a una composición de oligosacárico enriquecida significativamente en DP2 y (DP1 + DP2) en comparación con los productos de la sacarificación catalizados por una a-amilasa de Aspergillus kawachii. Esto facilita, por ejemplo, la utilización de la composición de oligosacáridos por parte de un organismo fermentador en un proceso de sacarificación y fermentación simultáneas.
