Abstract
En la actualidad zotepina solo se encuentra disponible en tabletas. Sin embargo, la observancia del tratamiento por parte del paciente es un problemamuy conocido con las drogas anti-psicopatías. Es conveniente proveer la zotepina en una formulacion líquida que pudiese ser agregada en forma de gotas a losalimentos o administrada a través de una bebida. No obstante, la zotepina posee una pobre solubilidad en los disolventes que comunmente se utilizanen la industria farmacéutica a los efectos de preparar formulaciones líquidas orales. La solubilidad de la zotepina puede ser mejorada conviertiéndola enuna sal de adicion ácida, pero las soluciones de sales de adicion ácidas son inestables en almacenamiento debido a la hidrolisis que produce 8-clorodibenzo[b,f]tiepi-10(11H)nona. Además, la zotepina en solucion puede soportar una oxidacion para dar el N-oxido con el tiempo. Sorprendentemente, se ha descubiertouna formulacion que provee zotepina en forma líquida, la cual es estable en almacenamiento. En consecuencia, la presente invencion provee una formulacionfarmacéutica líquida que comprende: a) entre 2 y 7% en peso/volumen de zotpeina; b) entre 0,5 y 35% en peso/volumen de un ácido orgánico seleccionadodel grupo integrado por ácido ascorbico, ácido cítrico, ácido fumárico, ácido glutárico, ácido láctico, ácido málico, ácido sorbico y ácido tartárico; c)entre 15 y 60% volumen/volumen de etanol, y d) un diluyente líquido hasta el 100%. Preferiblemente, el ácido orgánico es seleccionado del grupo integradopor ácido cítrico, ácido málico y ácido láctico, y más aun el ácido orgánico es ácido láctico. Preferiblemente, el diluyente líquido es agua, polietilenglicolo aceite de sésamo o mezclas de los mismos. Más aun, el diluyente esagua o polietilenglicol. Es sumamente conveniente que el diluyente seapolietilenglicol.
