Abstract

IZRAŽAVANJE HEPATITIS B PRES2 PROTEINA U METILLTROFNIM KVASCIMA, naznačen time, što obuhvata: a) transformisanje metilotrofnog kvasca sa najmanje jednim vektorom koji ima najmanje jednu kasetu za izražavanje koja sadrži strukturni gen za preS2, operativno povezanu za regulatorni region 3' terminacionu sekvencu; i posle toga b) kultivisanje dobivenog transformisanog soja kvasca pod podesnim uslovima tako da se ostvaruje proizvodnja pomenutog HBsAg preS2 proteina. 2. Postupak iz zahteva 1, naznačen time, što je pomenuti vektor izabran iz grupe koja sadrži plazmide linearnih, integrativni vektora specifičnih za mesto. 3. Postupak iz Zahteva 2, naznačen time, što je vektor linearan integrativan vektor specifičan za mesto. 4. Postupak iz Zahteva 3, naznačen time, što pomenuti linearan integrativan vektor specifičan za mesto sadrži sledeću serijsku konstrukciju: a) prvi DNA fragment koji se može umetnuti, b) gen markera i najmanje jednu kasetu za izražavanje koja sadrži strukturni gen za preS2, operativno povezanu za regulatorni region i 3' terminacionu sekvencu, i c) drugi DNA fragment koji se može umetnuti; pri čemu se redosled gena markira i komponente kasete (b) može zameniti.
