Abstract

본 발명은 유전자 발현 조절 DNA 및 그것을 사용한 단백질의 제조법을 제공한다.
즉, 코리네형 세균의 이소시트르산 리아제(ICL) 유전자에 유래하는 DNA로서, 단백질을 코딩하는 구조 유전자와 함께 벡터 DNA에 삽입되어서 숙주 코리네형 세균에 도입되었을 때에 이 구조 유전자의 발현을 조절하는 작용을 갖는 DNA 및 DNA를 사용하여 유용 단백질을 효율 좋게 제조하는 방법을 제공한다.

유전자 발현 조절 디엔에이(DNA)
본 내용은 요부공개 건이므로 전문내용을 수록하지 않았음
제1도는 ICL 유전자의 Hind Ⅲ 6.0kb 클론화 DNA 단편의 제한효소지도를 나타낸다.
제2도는 pKT10의 제작공정을 나타낸다.
제3도는 ICL 유전자의 Hind Ⅲ 6.0kb 클론화 DNA 단편의 서브클로닝에 있어서의 제한효소지도를 나타낸다.

Claims (11)

  1. 코리네형 세균의 이소시트르산 리아제(ICL) 유전자에 유래하는 DNA로서, 단백질을 코딩하는 구조 유전자와 함께 벡터 DNA에 삽입되어서, 숙주 코리네형 세균에 도입되었을 때에, 이 구조 유전자의 발현을 조절하는 작용을 갖는 DNA.
  2. 제1항에 있어서, 구조 유전자의 발현이 배지중의 탄소원을 당질로 하였을 때에 억제되고, 배지중의 탄소원을 비당질로 하였을 때 또는 당질 비존재 배지에서 유도되는 것인 DNA.
  3. 제1항에 있어서, 배열번호 3으로 표시되는 염기배열의 제1번에서 제702번까지의 염기배열에 포함되는 DNA인 DNA.
  4. 제1항에 있어서, 구조 유전자가 ICL, β-갈락토시다제, 클로람페니콜 아세틸트란스페라제, 인슐린, 성장호르몬, 인터페론 및 과립구 콜로니자극 호르몬으로부터 선택되는 효소 또는 단백질을 코딩하는 유전자인 DNA.
  5. 제1항에 있어서, ICL 유전자를 채쥐하는 코리네형 세균이 코리네박테륨 속, 브레비박테륨 속, 및 미크로박테륨 속으로부터 선택되는 것인 DNA.
  6. 제5항에 있어서, 코리네형 세균이 코리네박테륨 글루타이쿰 ATCC 13032, 코리네박테륨 아세토아시도필름 ATCC 13870, 코리네박테륨 아세토글루타미쿰 ATCC 15806, 코리네박테륨 칼루나에 ATCC 15991, 코리네박테륨 헤르쿨리스 ATCC 13868, 코리네박테륨 멜랏세콜라 ATCC 17965, 코리네박테륨 릴륨 ATCC 15990, 브레비박테륨 이마리오필룸 ATCC 14068, 브레비박테륨 사카롤리티쿰 ATCC 14066, 브레비박테륨 티오게니탈리스 ATCC 19240, 브레비박테륨 디바리카툼 ATCC 14020, 브레비박테륨 플라붐 ATCC 14067, 브레비박테륨 락토페르멘툼 ATCC 13869, 브레비박테륨 로세움 ATCC 13825 및, 미크로박테륨 암모니아필룸 ATCC 15354로부터 선택되는 DNA.
  7. 제1항에 있어서, 숙주 코리네형 세균이 코리네박테륨 속, 브레비박테륨 속 및 미크로박테륨 속으로부터 선택되는 것인 DNA.
  8. 제7항에 있어서, 코리네형 세균이 코리네박테륨 글루타미쿰 ATCC 13032, 코리네박테륨 아세토아시도필름 ATCC 13870, 코리네박테륨 아세토글루타미쿰 ATCC 15806, 코리네박테륨 칼루나에 ATCC 15991, 코리네박테륨 헤르쿨리스 ATCC 13868, 코리네박테륨 멜랏세콜라 ATCC 17965, 코리네박테륨 릴륨 ATCC 15990, 브레비박테륨 이마리오필룸 ATCC 14068, 브레비박테륨 사카롤리티쿰 ATCC 14066, 브레비박테륨 티오게니탈리스 ATCC 19240, 브레비박테륨 디바리카툼 ATCC 14020, 브레비박테륨 플라붐 ATCC 14067, 브레비박테륨 락토페르멘툼 ATCC 13869, 브레비박테륨 로세움 ATCC 13825, 브레비박테륨 암모니아게네스 ATCC 6872 및 미크로박테륨 암모니아필룸 ATCC 15354로부터 선택되는 DNA.
  9. (1) 코리네형 세균의 이소시트르산 리아제(ICL) 유전자에 유래하는 DNA로서, 단백질을 코딩하는 구조유전자와 함께 벡터 DNA에 삽입되어서 숙주 코리네형 세균에 도입되었을때에 이 구조유전자의 발현을 조절하는 작용을 갖는 DNA 및 (2) 단백질을 코딩하는 구조유전자가 벡터 DNA에 삽입된 재조합 DNA.
  10. (1) 코리네형 세균의 이소시트르산 리아제(ICL) 유전자에 유래하는 DNA로서, 단백질을 코딩하는 구조유전자와 함께 벡터 DNA에 삽입되어서 숙주 코리네형 세균에 도입되었을때에 이 구조유전자의 발현을 조절하는 작용을 갖는 DNA 및 (2) 단백질을 코딩하는 구조유전자가 숙주에서 자동복제 가능한 벡터 DNA에 삽입된 재조합 DNA를 함유하는 코리네형 세균 형질전환제.
  11. (1) 코리네형 세균의 이소시트르산 리아제(ICL) 유전자에 유래하는 DNA로서, 단백질을 코딩하는 구조유전자와 함께 벡터 DNA에 삽입되어서 숙주 코리네형 세균에 도입되었을때에 이 구조유전자의 발현을 조절하는 작용을 갖는 DNA 및 (2) 단백질을 코딩하는 구조유전자가 숙주에서 자동 복제 가능한 벡터 DNA에 삽입된 재조합 DNA를 함유하는 코리네형 세균 형질 전환체를 배지에 배양하여, 배양물중에 해당 단백질을 생성 축적시켜, 이 배양물로부터 해당 단백질을 채취하는 것을 특징으로 하는 단백질의 제조법.
