Abstract

본 발명은 갯벌에서 새롭게 분리한 균주에서 추출한 새로운 액티노플라보사이드 B, C, D와 그 항균 용도 및 암과 면역 관련 질환의 예방 및 치료 용도에 대한 것이다.
해양 미생물들은 독특한 화학구조와 2차 대사산물의 다양한 생리활성으로 인해 화학적 발견을 위한 천연 자원으로서 매우 가치가 있으며, 항생제, 항진균제, 항종양제 및 치료제로 개발되어 왔다. 천연 제품 중에서 플라보노이드는 생체 활성 폴리페놀의 크고 다양한 그룹이고 대부분의 플라보노이드는 항암, 항산화, 항균 및 항염증과 같은 생물학적 활성을 나타내므로 약학적 및 화학적 중요성을 지닌 천연 화합물로서 큰 관심을 받고 있다. 그러나 플라보노이드는 양치식물과 고등식물에 존재하는 것으로 오래전부터 알려져 왔지만 미생물에서는 상대적으로 덜 존재하는 것으로 알려졌다.
1997년, 액티노플라보사이드 A로 명명된, glycoside 7-{2,3,6-trideoxy-3-[3-(R)-hydroxy-2-(R)-methylbutanoic acid] amino-α-D-ribo-hexopyranosyl}-8-hydroxy-5-hydroxymethyl-2-phenyl-chroman-4-one가 해양 미생물인 Streptomyces sp.에서 분리되었다. 상기 액티노플라보사이드 A는 희귀한 아미노당 리스토사민 (aminosugar ristosamine)과 결합된 전례 없는 5-hydroxymethyl-7,8-dihydroxyflavonone으로 구성된다. 상기 리스토사민 성분은 erythro-2-methyl-3-hydroxy butyric acid으로 추가로 아실화되어 해당 아미드를 형성한다.
그러나 상기 액티노플라보사이드 A가 발견된지 25년이 넘게 지나도록 해양 방선균류로부터 유래된 새로운 플라보노이드형 배당체는 발견되지 않은 실정이다.
본 발명자들은 갯벌은 조석차에 의한 염분, 온도, 수압 등의 극한 환경변화에 크게 영향을 받아 생물다양성이 높아 다양한 2차 대사산물을 생산하는 방선균 발견을 기대할 수 있다는 점에 착안하여, 대한민국 무안의 갯벌의 해양 미생물로부터 새로운 생리 활성 2차 대사산물을 발견하기 위해 방선균 균주를 분리하였고, 상기 균주로부터 새로운 화합물, 즉 액티노플라보사이드 (Actinoflavoside) B, C, D를 고성능 액체 크로마토그래피로 정제한 후 NMR을 통하여 그 구조를 확인하였을 뿐만 아니라, 상기 액티노플라보사이드들의 항균 작용과 면역 조절 기능을 확인하여 발명을 완성하였다.
따라서, 본 발명의 목적은 신규한 액티노사이드 B, C, D 및 그의 염을 제공하는 데 있다.
본 발명의 또 다른 목적은 상기 신규한 액티노사이드 B, C, D 및 그의 염을 유효성분으로 포함하는 항균제를 제공하는 것이다.

본 발명에서 “그람 양성균”이라 함은, 그람 염색법에 의하여 자색으로 염색되는 세균을 통칭하는 것으로, 세포벽의 외면에 펩티도글리칸 층을 포함하고 있으나 이에 제한되는 것은 아니다. 본 발명에서 상기 그람 양성균은 대표적으로 유산균, 고초균, 탄저균, 나병균, 포도상구균, 디프테리아균, 연쇄상 구균, 파상풍균, 방선균 등이 알려져 있으나 이에 제한되는 것은 아니다.
본 발명에서 상기 “그람 염색”이라 함은 모든 세균을 크게 두 가지로 구분할 수 있도록 하는 가장 중요한 세균 동정법을 의미하며 통상 그람 염색법에 의하여 자색으로 염색되는 세균을 그람 양성균, 자색으로 염색되지 않으며, 대조색인 사프라닌에 의하여 적색으로 염색되는 세균을 그람 음성균이라 한다. 그람 염색법은 세균을 크리스탈 바이올렛으로 염색한 다음, 매염제인 아이오딘으로 처리하고, 에탄올 탈색 및 사프라닌 대조염색의 과정을 거쳐 진행된다. 그람 염색은 세포벽의 펩티도글리칸의 함량차이에 의하여 달라지는 것으로, 일반적인 그람 양성균은 1개의 세포막위에 두터운 펩티도글리칸 층을 가지고 있으며, 그람 음성균은 2개의 세포막을 가지고 있는 것으로 알려져 있다. 이러한 그람염색을 수행하는 가장 큰 이유는 그람염색의 양성 및 음성 여부에 따라 사용되는 배양법 및 항생제가 달라지기 때문이다.
본 발명에서 “그람 음성균”은 살모넬라균, 이질균, 티푸스균, 대장균, 콜레라균, 페스트균, 임균, 수막염균, 백일해균 등이 알려져 있으나 이에 제한되는 것은 아니다.

한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 약학적 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥 내, 근육 내, 동맥 내, 골수 내, 경막 내, 심장 내, 경피, 피하, 복강 내, 비강 내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다.
본 발명에서, "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 안구내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학적 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명의 약학적 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학적 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약물 형태, 투여 경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 1,000 mg/kg 또는 0.001 내지 500 mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 의약 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형될 수 있다.
본 발명의 약학적 조성물은 단독으로, 또는 수술, 방사선 치료, 호르몬 치료, 화학 치료 및 생물학적 반응 조절제를 사용하는 방법들과 병용하여 사용할 수 있다.
본 발명에서 상기 "투여"는 임의의 적절한 방법으로 대상체에 소정의 본 발명의 화합물을 제공하는 것을 의미한다.
본 발명에서 상기 투여가 필요한 "대상체"는 포유동물 및 비-포유동물을 모두 포함할 수 있다. 여기서, 상기 포유동물의 예로는 인간, 비-인간 영장류, 예컨대 침팬지, 및 다른 유인원 및 원숭이 종; 축산 동물, 예컨대 소, 말, 양, 염소, 돼지; 사육 동물, 예컨대 토끼, 개 및 고양이; 실험 동물, 예를 들어 설치류, 예컨대 래트, 마우스 및 기니아 피그 등을 포함할 수 있으나, 이에 제한되는 것은 아니다. 또한, 본 발명에서 상기 비-포유동물의 예로는 조류 및 어류 등을 포함할 수 있으나, 이에 제한되는 것은 아니다.
본 발명에서 상기와 같이 투여되는 화합물의 제제는 특별히 제한하지 않으며, 고체 형태의 제제, 액체 형태의 제제 또는 흡인용 에어로졸 제제로 투여될 수 있으며, 사용하기 바로 전에 경구 또는 비경구 투여용 액체 형태 제제로 전환되도록 의도되는 고체 형태 제제로 투여될 수 있고, 예를 들면, 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 투여될 수 있으나, 이에 제한되는 것은 아니다.
또한, 본 발명에서 상기 투여 시 본 발명의 화합물과 함께 약학적으로 허용 가능한 담체를 추가로 투여할 수 있다. 여기서, 상기 약학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 화합물의 제형은 상술한 바와 같은 약학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있다.

본 발명에서, "약학적으로 유효한 양"은 바람직한 생물학적 결과를 제공하기 위한 작용제의 충분한 양을 지칭한다. 상기 결과는 질환의 징후, 증상 또는 원인의 감소 및/또는 완화, 또는 생물계의 임의의 다른 바람직한 변화일 수 있다. 예를 들어, 치료 용도를 위한 "유효량"은 질환에서 임상적으로 유의한 감소를 제공하는데 요구되는, 본 발명에 개시된 화합물의 양이다. 임의의 개별적인 경우에서 적절한 "효과적인" 양은 일상적인 실험을 사용하여 당업자에 의해 결정될 수 있다. 따라서, 표현 "유효량"은 일반적으로 활성 물질이 치료 효과를 갖는 양을 지칭한다.

본 발명의 특징 및 이점을 요약하면 다음과 같다.
(a) 새롭게 3가지 액티노플라노사이드를 분리하여 구조를 밝혔으며, 그 중 액티노플라노사이드 D가 가지는 플라보노이드(flavonoid)에 연결된 isobutyl alkylated chormone 구조는 플라보노이드가 주로 생산되는 식물부터 미생물에서도 아직까지 보고된 적이 없는 매우 독특하고 희귀한 구조임을 확인하였다.
(b) 본 발명에서 새롭게 발견하여 구조를 밝힌 액티노플라보사이드들은 그람 음성균인 P. aeruginosa 및 그람 양성균인 B. subtilis의 성장을 억제할 수 있다.
(c) 액티노플라보사이드를 처리한 경우, T 세포의 성장과 분화에 중요한 역할을 담당하는 T 세포 성장인자이면서 Th-1 세포에서 생산되는 사이토카인인 IL-2의 생산이 비장에서 증가하고, 상기 IL-2의 생산은 액티노플라보사이드의 농도 의존적으로 증가함을 확인할 수 있었다. 종합하면, 이러한 결과는 액티노플라보사이드가 Th2 세포 활성화 없이 일차 T 세포 활성화 및 IL-2 사이토카인 생산을 증가시킴으로써 T 세포 면역 반응에서 중요한 역할을 담당할 수 있다.
(d) 액티노플라보사이드 처리시 알레르기와 천식과 관계되어 있다고 알려져 있고 Th-2 세포에서 생산되는 IL-4의 생산은 비장에서 감소함을 확인할 수 있었다.
(e) 비장 세포의 증식은 ConA로 비장 세포가 자극되거나(도 6b) 자극되지 않거나 이와 무관하게 액티노플라보사이드 처리시 증가함을 확인할 수 있었다.

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하고자 한다. 이들 실시예는 오로지 본 발명을 보다 구체적으로 설명하기 위한 것으로, 본 발명의 요지에 따라 본 발명의 범위가 이들 실시예에 의해 제한되지 않는다는 것은 당업계에서 통상의 지식을 가진 자에 있어서 자명할 것이다.
실시예
[실시예 1] 박테리아 분리 및 배양
[실시예 1-1] 일반적 화합물 분석 방법
일반 회전(General rotations)은 1.0cm 셀이 있는 PerkinElmer Model 343 plus 선광계(polarimeter)(Waltham, MA, USA)로 측정했다. 또한 UV 스펙트럼은 Agilent Technologies 1260 시리즈 Infinity II LC 시스템(Agilent Technologies, Santa Clara, CA, USA)을 사용하여 기록했고, 적외선(IR) 스펙트럼은 PerkinElmer 스펙트럼 400 FT-IR 및 FT-NIR 분광계(Waltham, MA, USA)로 측정되었다. 1H, 13C, 2D NMR 스펙트럼은 오창에 있는 광주 한국기초과학연구원(KBSI)의 Varian Unity INOVA 600MHz 분광계와 2014년 12월 1일에 Bruker Avance II 800MHz NMR 분광계(Bruker, Billerica, MA, USA)에서 획득했다. 전기분무 이온화 (Electrospray ionization, ESI) 저해상도 LC/MS 데이터는 Phenomenex Luna 역상(reversed-phase) C18 컬럼(100x4.6mm, 5㎛)을 사용하여 Agilent Technologies 1260 시리즈 Infinity II LC 시스템과 결합된 Agilent G6125B MSD 시스템에서 획득했다. 고해상도 TOF Mass Spectrum Field Desorption 이온 소스 데이터는 전남대학교 공동 연구 시설 센터(CCRF)에서 JMS-T200GC(Jeol, Akishima, Tokyo)를 사용하여 수집되었다. 고해상도 전자분무 이온화(HR-ESI) 질량 스펙트럼은 Agilent 6530 iFunnel Q-TOF LC/MS 시스템(Agilent Technologies, Santa Clara, CA, USA)에 결합된 Agilent Technologies 1290 시리즈 HPLC를 사용하여 얻었다.
[실시예 1-2] 박테리아 분리
균주 JML48 및 JMS33은 2020년 8월 대한민국 무안의 갯벌 샘플에서 분리되었다. 먼저 건조된 침전물(2g)을 멸균된 해수 4mL에 희석하고 혼합물을 방선균(actinomycete) 분리 배지에 뿌렸다. JML48 균주는 A1 한천배지(한천 18g, 시클로헥시미드(cycloheximide) 25mg, 날리딕식산(nalidixic acid) 10mg, 해수 1L)에서 배양하여 분리하였고, JMS33 균주는 TWYE 한천배지(효모 추출물 0.25g, K2HPO4 0.5g, 18g 한천, 시클로헥시미드 25mg, 날리딕식산 10mg, 해수 1L)에서 배양하여 분리하였다. JML48 및 JMS33 균주는 16S rDNA 시퀀싱 분석 데이터를 기반으로 Streptomyces althioticus(동일성 99.9%, 수탁 번호 #LN864578) 및 Streptomyces sanglieri(동일성 99.0%, 수탁 번호 #AB735535)와 가장 밀접한 관련이 있었다는 것을 확인할 수 있었다.
[실시예 1-3] 박테리아 배양 및 추출
JML48 균주를 100mL의 삼각 플라스크 내에서 50mL의 YEME 배지 (효모 추출물 3g, 맥아 추출물 3g, 펩톤(peptone) 5g, 소이톤(soytone) 2g, 포도당 10g, 해수 1L)에서 배양하였다. 이어서 해당 균주를 25℃에서 3일간 190rpm으로 회전 진동기에서 배양한 후, 배양액 3.5mL를 YEME 배지 150mL가 담긴 500mL 삼각 플라스크에 접종하고 2일 동안 섞었다. 그 다음 상기 배양액 20mL를 2.5L Ultra Yield 플라스크에 담긴 YEME 배지 1L에 접종했다. JML48 (8L) 균주는 25℃에서 190rpm 속도로 8일간 배양되었다. 상기 배양액 전체를 에틸아세테이트(EtOAc)로 추출하고, 분리깔대기 (용량, 3L)를 이용하여 기질을 분리한 후, 무수 Na2SO4를 첨가하여 에틸아세테이트 층에 남아있는 물을 제거하였다. 이어서 해당 추출물을 진공에서 농축하여 3.5g의 건조 추출물을 얻었으며 이 절차를 다섯 번 반복하였다. JMS33 균주 역시 4일 동안 동일한 절차로 배양 및 추출하여 건조 추출물 2.8g을 얻었다.
[실시예 2] 액티노플라보사이드(Actinoavosides) B, C, D 구조 결정
[실시예 2-1] 액티노플라보사이드(Actinoavosides) 분리
앞서 수득한 JML48과 JMS33 건조 추출물을 C18 역상 개방형 컬럼 (C18 reversed-phase open column, YMC ODS-A-C18, 50 ㎛ 실리카 겔)에서 20%, 40%, 60%, 80% 및 100%의 메탄올 수용액과 1:1 MeOH/CH2Cl2를 이용하여 분획화하였다. 액티노플라보사이드 B-D는 60% 및 80% MeOH/CH2Cl2 분획에서 검출되었다. 상기 60% 및 80% 분획은 등용매 용매 시스템(an isocratic solvent system, 33% CH3CN/H2O, 254nm에서 UV 검출, 8 mL/min 유속), tR= 30 (42 mg), 35 (33mg) min을 사용하여 예비 역상 HPLC(preparative reversed-phase HPLC, YMC-Pack ODS-A-C18 column, 5㎛, 250 x 20 mm)에서 분리되었다. 추가적으로 액티노플라보사이드 B-D는 등용매 조건(30% CH3CN/H2O, 254nm에서의 UV 검출, 유속: 2 mL/min)을 사용하여 반 준비 역상 HPLC(a semi-preparative reversed-phase HPLC, YMC-Pack ODS-A-C18 column, 5 ㎛, 250 x 10 mm)에서 추가로 정제되었다. 그 결과 액티보블라보사이드 B, C 및 D는 40분(15mg), 36분(5.2mg), 48분(13mg)의 각 체류 시간에서 순수한 화합물로 수집되었다.

액티보플라보사이드 C (2): 노란색 기름(yellow oil), [α]D-37.7 (c 0.02, MeOH); UV (MeOH) λmax (log ε) 215 (3.08)nm, 235 (3.36)nm, 285 (3.36)nm; IR (neat) vmax 3373, 2929, 1601, 1063 cm-1, HRMS (TOF) m/z: [M]+ 515.2172(calcd for C24H27NO8, 457.1737)이고 1H와 13C NMR 결과는 표 1에 나타내었다.

[실시예 2-2] 액티노플라보사이드(Actinoavosides) B와 D의 MTPA 에스테르화
액티노플라브사이드 B(1)의 샘플을 2개의 40mL 바이알(각 바이알에 3mg의 화합물)로 옮기고 고진공 하에서 밤새 완전히 건조시켰다. 그 다음, 무수 피리딘 (anhydrous pyridine) 1mL를 질소 가스 하에서 각 바이알에 첨가하였다. 상기 혼합물을 실온에서 대략 5분 동안 교반하였다. 이어서, R- 과 S-α-methoxy-α-(trifluoromethyl) phenylacetyl chloride (MTPA-Cl) (30 uL)를 각각 2개의 바이알 중 하나에 첨가하고 상기 혼합물을 35℃에서 4시간 동안 교반하였다. 그 다음 상기 생성물은 단계별 구배 용매 시스템(45분 동안 50-100% CH3CN/H2O, 45-70분 동안 100% CH3CN, 유속 2mL/min, UV 280에서 검출)을 통한 역상 HLPC (a reversed phase HPLC, YMC-Pack-C8 column, 5 ㎛, 250 x 10.0 mm)를 이용하여 정제하였고 액티노플라보사이드 B의 S-MTPA 에스테르(1a)와 R-MTPA 에스테르(1b)는 모두 51분에 용출되었다. 1H NMR 및 1H-1H S-R COSY NMR 스펙트럼을 분석하여 입체 중심 주변의 △δS-R 값을 지정했다. 액티노플라보사이드 D(3)의 경우 동일한 절차를 따라 3a 및 3b를 생성하였다.

[실시예 2-3] 액티노플라보사이드(Actinoavosides) B, C, D의 구조 결정 및 분석 결과
이를 통해 분석된 액티노플라노사이드 B, C, D의 구조를 도 1에 도시하였다. 특히 해당 분석을 통해 밝혀진 액티노플라노사이드 D가 가지는 플라보노이드(flavonoid)에 연결된 isobutyl alkylated chormone 구조는 플라보노이드가 주로 생산되는 식물부터 미생물에서도 아직까지 보고된 적이 없는 매우 독특하고 희귀한 구조임을 확인할 수 있었다.
[실시예 3] 항균 활성 분석
그람 양성균(B. subtilis ATCC 6051)과 그람 음성균(P. aeruginosa KCTC 22073, E. coli ATCC 11775, Er. rhapontici ATCC 29283)을 Luria-Bertani 한천(LB)에서 배양하였다. 30℃에서 밤새 배양한 후, 상기 세포를 LB에서 30℃에서 24시간 동안 배양하고 수확된 미생물 세포를 초기 광학 밀도(OD600) 값이 0.0008인 Mueller Hilton 배양액에 접종했다. 화합물 용액은 DMSO에서 제조하였다. 상기 각 화합물 용액을 Mueller-Hilton로 희석하여 200~0.8g/mL 범위에서 연속 2배 희석 용액을 제조했다. 각 플레이트를 30℃ 20시간 동안 배양하고 OD 값을 600 nm에서 관찰하였다. 겐타마이신 (Gentamicin)을 기준 화합물 (reference compound)로 사용하여 항균 활성을 분석하였다. 그 결과를 하기 표 2에 나타내었다.

상기 표 2에서 화합물 1 내지 3은 각기 액티노플라보사이드 B 내지 D를 의미하며, 최소 억제 농도 (Minimum inhibitory concentration, MIC) 값을 겐타마이신 (Gentamicin)과 비교하여 나타내었다.
그 결과 액티노플라보사이드 B와 D는 그람 음성균인 P. aeruginosa에 대하여 MIC값을 각각 0.29와 0.30mM을 나타내는 바, 그람 음성균인 P. aeruginosa의 성장을 억제할 수 있음을 확인할 수 있었다. 또한 액티노플라보사이드 B는 그람 양성균인 B. subtilis에 대하여 MIC값을 0.14mM 나타내는 바, 그람 양성균인 B. subtilis의 성장을 억제할 수 있음을 확인할 수 있었다.
[실시예 4] 면역 조절 활성 분석(Immunomodulatory Activity Assay)
7주령의 BALB/c 암컷 마우스(Damool Science, 대전, Korea)를 온도가 조절되고 12시간의 명암 주기가 있는 병원균이 없는 동물실에서 준비하였다. 모든 동물 절차는 전남대학교 동물 관리 및 사용 위원회(CNU IACUC-YB-2022-01)의 지침을 따랐다. 경추 탈구로 마우스를 희생시켰고 단세포 현탁액은 50㎛ 스트레이너 메쉬 (BD Falcon, San Diego, CA, USA)를 사용하여 분리된 비장으로부터 준비되었다. 상기 세포 펠렛을 실온에서 5분 동안 적혈구 용해 완충액(Biolegend, San Diego, CA, USA)에 완전히 재현탁시켰다. 비장 세포를 RPMI 배지에 현탁시키고 T 세포를 비특이적으로 활성화시키는 미토겐(mitogen) 즉, concanavalin A, ConA (1ug/mL)로 유도하였다. 비장 세포를 48-웰 플레이트(SPL life sciences Co., 포천, 경기도, 한국)에 2 x 105 세포/웰로 접종하였다. 상기 세포들을 액티노플라보사이드 B, C 또는 D(10, 30 또는 100ug/mL) 및 ConA(1ug/mL)로 72시간 동안 처리하였다. ELISA 키트(R&D 시스템, 미국 미네소타주 미네아폴리스 소재)를 사용하여 세포 상층액의 IL-2 및 IL-4 사이토카인을 측정하였다. ELISA 마이크로플레이트 판독기(ELx808)(Bio Tek Instruments, Inc., inooski, VT, USA)를 사용하여 450 nm에서 흡광도를 판독하였다. 마우스 비장 세포는 96-웰 플레이트(SPL life sciences Co., 포천, 경기도, 한국)에 1 x 105 세포/웰로 접종하였다. 상기 세포들을 ConA의 부재 또는 존재 하에 72시간 동안 액티노플라보사이드 D(3, 10, 30 또는 100g/mL)로 처리하였다. 제조업체의 지침에 따라 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxy-methoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) 시약을 사용하여 세포 증식을 분석했다. 이어서 ELISA 마이크로플레이트 판독기(ELx808)를 사용하여 490 nm에서 흡광도를 판독하였다. 모든 실험은 다른 날에 세 번 이상 반복되었다. 달리 명시되지 않는 한 결과는 평균의 평균 표준 오차(SEM)로 표현되었다. 통계적 차이는 다중 그룹 비교를 위한 일원 분산 분석(ANOVA)을 사용하여 평가한 다음 Tukey의 사후 테스트(p < 0.05가 통계적으로 유의한 것으로 간주됨)를 수행했다.
그 결과를 도 4 내지 도 6b에 나타내었다. 도 4에 나타낸 바와 같이 T 세포의 성장과 분화에 중요한 역할을 담당하는 T 세포 성장인자이면서 Th-1 세포에서 생산되는 사이토카인인 IL-2의 생산이 액티노플라보사이드 D를 ConA (concanavalin A, T 세포 활성제: 1ug/mL)와 함께 처리한 경우 비장에서 증가함을 확인할 수 있었다. 또한, 도 5a에 나타낸 바와 같이, IL-2의 생산은 ConA로 자극된 비장에서 액티노플라보사이드 D의 농도 의존적으로 증가함을 확인할 수 있었다. 종합하면, 이러한 결과는 액티노플라보사이드 D가 Th2 세포 활성화 없이 일차 T 세포 활성화 및 IL-2 사이토카인 생산을 증가시킴으로써 T 세포 면역 반응에서 중요한 역할을 한다는 것을 시사한다.
또한, 도 5b에 나타낸 바와 같이, 알레르기와 천식과 관계되어 있다고 알려져 있고 Th-2 세포에서 생산되는 IL-4의 생산은 ConA로 자극된 비장에서 액티노플라보사이드 D 처리시 감소함을 확인할 수 있었다.
마지막으로 도 6a와 도 6b에서 나타낸 바와 같이, 비장 세포의 증식은 ConA로 비장 세포가 자극되거나(도 6b) 자극되지 않거나(도 6a) 이와 무관하게 액티노플라보사이드 D 처리시 증가함을 확인할 수 있었다.
