Abstract
본 발명은, 신규 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염을 제공한다. 본 발명에 따른 신규 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염은 인간 암 세포주에 대한 세포 독성을 나타내어 암 세포의 성장을 억제함으로써, 위 언급된 종류의 암에 대하여 우수한 예방 또는 치료 효과를 가지며, LPS에 의해 유도된 염증실험에서 미세아교세포의 신경염증 저해 활성을 나타냄으로써 위 언급된 종류의 신경염증 질환에 대하여 우수한 예방 또는 치료 효과를 가진다.
Description
본 발명은 신규 다이페나진 화합물 및 이의 용도에 관한 것이다.
해양 곰팡이는 지속적으로 연구되어 오고 있으며, 많은 수의 신규화합물을 생산하는 것으로 알려져 있다. 심해 미생물 유래의 신규 생리활성물질을 찾기 위한 연구를 진행하던 중 본 발명자들은, 인도양 중앙 해령대에서 분리된 흔하지 않은 효모성 곰팡이 시스토바시디움 라린지스(Cystobasidium laryngis) IV17-028을 발견하였다. 선행문헌(Mar. Drugs,
2019, 17, 482)에서, IV17-028 균주에서 새로운 페나진(phenazine) 계열의 유도체 3개를 분리하였고, 이 유도체들이 LPS 자극된 RAW 264.7 세포에서 항염활성을 나타내는 것을 확인하였다. 페나진 계열의 화합물들은 C. laryngis IV17-028 균주에서 분리되기 전까지, 세균(스트렙토마이세스와 슈도모나스)과 일부 고세균에서 생산되는 것으로 알려져 있었다. 산화 환원력(redox activity)을 가진 페나진 골격은 전자 셔틀(electron shuttle), 활성 산소(reactive oxygen species)의 감소, 유전자 발현 조절, 생물막(biofilm) 형성, 쿼럼센싱(quorum sensing) 신호 및 세포내 산화환원 항상성의 유지 등에 관여한다. 따라서, 페나진 계열 화합물들은 앞서 언급한 다양한 역할과 폭 넓은 생리활성 때문에 관심을 받고 있다. 특히, 에스메라딘 A-B, 페나조스타틴 A-D, 이주미페나진 A-B, 페나지오린 A-E 및 다이아스타페나진과 같은 페나진 이량체(dimeric phenazine)는 상대적으로 보고된 바가 적고, 페나조스타틴 D(슈도노카르디아 종에서 보고됨)를 제외한 나머지는 스트렙토마이세스 종에서만 분리된 것으로 보고되어 있다. 보고된 페나진 이량체 중에서, 페나조스타틴 A-C는 뇌 허혈성 손상을 막기 위한 글루타메이트 독성으로부터 뉴런의 하이브리도마 N-18-RE-105 세포를 보호하는 것으로 보고되어 있다.
본 발명자들은, 신규 다이페나진 화합물인 페나조스타틴 E-J의 분리 및 구조를 동정하고, 그중 일부 화합물을 반합성 하였으며, 이들 화합물의 항암효과 및 신경염증에 대한 생리활성을 확인하여 본 발명을 완성하였다.

본 발명이 해결하고자 하는 과제는 신규 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염을 제공하는 것이다.
또한, 본 발명은 상기 신규 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염의 용도를 제공하는 것이다.
또한, 본 발명은 상기 신규한 다이페나진 화합물, 이의 광학 이성질체 및 이의 약학적으로 허용 가능한 염을 필요로 하는 개체에 투여하는 단계를 포함하는 암 관련 질환의 예방 또는 치료방법을 제공하는 것을 그 목적으로 한다.

본 발명의 화학식 1로 표시되는 화합물은 1개 이상의 비대칭 탄소를 함유할 수 있으며, 이에 따라 라세미체, 라세믹 혼합물, 단일의 에난티오머, 부분입체이성체 혼합물 및 각각의 부분입체이성체로서 존재할 수 있다.
이러한 이성질체는 종래 기술, 예를 들어 화학식 1로 표시된 화합물은 관크로마토그래피 또는 HPLC 등의 분할에 의해 분리가 가능하다. 또는, 화학식 1로 표시되는 화합물 각각의 입체 이성질체는 공지된 배열의 광학적으로 순수한 출발 물질 및/또는 시약을 사용하여 입체 특이적으로 합성할 수 있다.
또한, 본 발명의 구체예에 따르면, 상기 화학식 1의 화합물은 하기 표 1의 화합물들로 이루어진 군으로부터 선택된 어느 하나일 수 있다.
본 발명에서, 약학적으로 허용가능한 염은 의약업계에서 통상적으로 사용되는 염을 의미하며, 예를 들어 칼슘, 포타슘, 소듐 및 마그네슘 등으로 제조된 무기이온염, 염산, 질산, 인산, 브롬산, 요오드산, 과염소산 및 황산 등으로 제조된 무기산염; 아세트산, 트라이플루오로아세트산, 시트르산, 말레인산, 숙신산, 옥살산, 벤조산, 타르타르산, 푸마르산, 만데르산, 프로피온산, 젖산, 글리콜산, 글루콘산, 갈락투론산, 글루탐산, 글루타르산, 글루쿠론산, 아스파르트산, 아스코르브산, 카본산, 바닐릭산, 하이드로 아이오딕산 등으로 제조된 유기산염; 메탄설폰산, 에탄설폰산, 벤젠설폰산, p-톨루엔설폰산 및 나프탈렌설폰산 등으로 제조된 설폰산염; 글리신, 아르기닌, 라이신 등으로 제조된 아미노산염; 및 트리메틸아민, 트라이에틸아민, 암모니아, 피리딘, 피콜린 등으로 제조된 아민염 등이 있으나, 열거된 이들 염에 의해 본 발명에서 의미하는 염의 종류가 한정되는 것은 아니다.
화학식 1의 화합물의 제조방법
본 발명은 시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) KCTC 13720BP 균주 또는 이의 배양물로부터 하기 화학식 1로 표시되는 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염을 분리하는 단계;를 포함하는 화학식 1의 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염의 제조방법을 제공한다.
본 발명에서 시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) KCTC 13720BP 균주의 배양물은 균주를 액체 배지 또는 고체 배지에서 배양하여 얻을 수 있다. 상기 배지는 탄소원으로서 비제한적인 예로 글루코오스, 물엿, 덱스트린, 전분, 당밀, 동물유 또는 식물유를 포함할 수 있다. 또한, 상기 배지는 질소원으로서 비제한적인 예로 밀기울, 대두박, 소맥, 맥아, 면실박, 어박, 콘스팁리커, 육즙, 효모 추출물, 황산암모늄, 질산소다 또는 요소를 포함할 수 있다. 또한, 상기 배지는 필요에 따라 식염, 칼륨, 마그네슘, 코발트, 염소, 인산, 황산 또는 기타 이온 생성을 촉진하는 무기염류를 포함할 수 있다. 배양은 진탕 또는 정치하면서 배양하는 것일 수 있고, 배양 온도는 약 20℃ 내지 약 37℃일 수 있고, 바람직하게는 약 25℃ 내지 약 30℃일 수 있다.
화학식 1의 화합물은 상기 균주 또는 이의 배양물을 용매 추출, 농축, 컬럼 크로마토그래피를 수행하여 얻을 수 있다. 상기 농축은 균주 또는 이의 배양물에 용매를 가하여 추출액을 감압 증발시켜 농축하는 것일 수 있다. 상기 용매는 에틸아세테이트, 클로로포름, 메틸렌 클로라이드, 탄소 수 1 내지 4개의 저급 알코올을 사용할 수 있으며, 바람직하게는 에틸아세테이트, 클로로포름, 메틸렌 클로라이드 및 메탄올을 사용할 수 있다. 상기 크로마토그래피는 정지상의 형태에 따라 컬럼 크로마토그래피, 평판 크로마토그래피, 종이 크로마토그래피 또는 얇은 막 크로마토그래피일 수 있다. 또는 이동상의 물리적 특성에 따라 HPLC (고성능 액체 크로마토그래피), 가스 크로마토그래피일 수 있다.
본 발명의 구체예에 따르면, 균주 또는 이의 배양물의 에틸아세테이트 추출물을 이용하여 실리카 플래시 컬럼 크로마토그래피로 분획한 후 메탄올 및 물의 혼합 용매를 이용하여 HPLC로 정제하여 화학식 1의 화합물을 얻을 수 있다.
따라서 본 발명은 (a) 시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) KCTC 13720BP 균주를 배양하는 단계;
(b) 상기 (a) 단계의 배양액을 에틸아세테이트로 추출하는 단계;
(c) 상기 (b) 단계의 추출물을 클로로포름으로 추출하는 단계;
(d) 상기 (c) 단계의 클로로포름 추출물을 CH2Cl2/MeOH로 단계적으로 용출시키는 단계로서
상기 단계적 용출은 CH2Cl2와 MeOH의 v/v비가 10:0, 20:1, 10:1, 5:1, 1:1인 용매로 용출하는 것인 단계;
(e) 상기 (d) 단계에서 용출된 결과물을 MeOH/H2O로 단계적으로 용출시키는 단계로서
상기 단계적 용출은 MeOH와 H2O의 v/v비가 1:4, 2:3, 3:2, 4:1, 5:0인 용매로 용출하는 것인 단계; 및
(f) 용출된 분획을 80% 메탄올로 용출시키고 분리된 화학식 1의 화합물을 정제하는 단계를 포함하는, 화학식 1의 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염의 제조 방법을 제공한다.

본 발명의 약학 조성물은 투여를 위해서 상기 화학식 1의 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염 외에 추가로 약학적으로 허용가능한 담체를 1 종 이상 더 포함할 수 있다. 약학적으로 허용 가능한 담체는 식염수, 멸균수, 링거액, 완충 식염수, 덱스트로즈 용액, 말토 덱스트린 용액, 글리세롤, 에탄올 및 이들 성분 중 1 성분 이상을 혼합하여 사용할 수 있으며, 필요에 따라 항산화제, 완충액, 정균제 등 다른 통상의 첨가제를 첨가할 수 있다. 또한 희석제, 분산제, 계면활성제, 결합제 및 윤활제를 부가적으로 첨가하여 수용액, 현탁액, 유탁액 등과 같은 주사용 제형, 환약, 캡슐, 과립 또는 정제로 제제화할 수 있다. 따라서, 본 발명의 약학 조성물은 패치제, 액제, 환약, 캡슐, 과립, 정제, 좌제 등일 수 있다. 이들 제제는 당 분야에서 제제화에 사용되는 통상의 방법 또는 Remington's Pharmaceutical Science(최근판), Mack Publishing Company, Easton PA에 개시되어 있는 방법으로 제조될 수 있으며 각 질환에 따라 또는 성분에 따라 다양한 제제로 제제화 될 수 있다.

본 발명의 치료방법은 상기 화학식 1의 화합물을 투여함으로써, 징후의 발현 전에 질병 그 자체를 다룰 뿐만 아니라, 이의 징후를 저해하거나 피하는 것을 또한 포함한다. 질환의 관리에 있어서, 특정 활성 성분의 예방적 또는 치료학적 용량은 질병 또는 상태의 본성(nature)과 심각도, 그리고 활성 성분이 투여되는 경로에 따라 다양할 것이다. 용량 및 용량의 빈도는 개별 환자의 연령, 체중 및 반응에 따라 다양할 것이다. 적합한 용량 용법은 이러한 인자를 당연히 고려하는 이 분야의 통상의 지식을 가진 자에 의해 쉽게 선택될 수 있다. 또한, 본 발명의 치료방법은 상기 화학식 1의 화합물과 함께 질환 치료에 도움이 되는 추가적인 활성 제제의 치료학적으로 유효한 양의 투여를 더 포함할 수 있으며, 추가적인 활성제제는 상기 화학식 1의 화합물과 함께 시너지 효과 또는 보조적 효과를 나타낼 수 있다.
본 발명은 상기 화학식 1의 다이페나진 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 암의 예방 또는 개선용 식품 조성물을 제공한다.
본 발명은 상기 화학식 1의 다이페나진 화합물, 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경염증 질환의 예방 또는 개선용 식품 조성물을 제공한다.
본 발명의 식품 조성물은 건강기능식품으로서 사용될 수 있다. 상기 "건강기능식품"이라 함은 건강기능식품에 관한 법률 제6727호에 따른 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 제조 및 가공한 식품을 의미하며, "기능성"이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻을 목적으로 섭취하는 것을 의미한다.
본 발명의 식품 조성물은 통상의 식품 첨가물을 포함할 수 있으며, 상기 "식품 첨가물"로서의 적합 여부는 다른 규정이 없는 한, 식품의약품안정청에 승인된 식품 첨가물 공전의 총칙 및 일반시험법 등에 따라 해당 품목에 관한 규격 및 기준에 의하여 판정한다.
본 발명의 식품 조성물은 암 및/또는 신경염증 질환의 예방 및/또는 개선을 목적으로, 조성물 총 중량에 대하여 상기 화학식 1의 화합물을 0.01 내지 95 %, 바람직하게는 1 내지 80 % 중량백분율로 포함할 수 있다. 또한, 암 관련 질환 및/또는 신경염증 질환의 예방 및/또는 개선을 목적으로, 정제, 캅셀, 분말, 과립, 액상, 환, 음료 등의 형태로 제조 및 가공할 수 있다.
또한, 본 발명은 암 및/또는 신경염증 질환의 치료용 약제의 제조를 위한 상기 화학식 1의 다이페나진 화합물, 또는 이의 약학적으로 허용가능한 염의 용도를 제공하고자 한다. 약제의 제조를 위한 상기 화학식 1의 화합물은 허용되는 보조제, 희석제, 담체 등을 혼합할 수 있으며, 기타 활성제제와 함께 복합 제제로 제조되어 활성 성분들의 상승 작용을 가질 수 있다.
본 발명의 조성물, 용도, 치료방법에서 언급된 사항은 서로 모순되지 않는 한 동일하게 적용된다.

본 발명에 따른 신규 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염은 선택적으로 우수하게 암세포주에 세포독성을 나타냄으로써 위 언급된 종류의 암에 대하여 우수한 예방 또는 치료 효과를 가지며, LPS-유도된 미세아교세포에서의 신경염증 질환 저해 활성을 나타냄으로써 위 언급된 종류의 신경염증 질환에 대하여 우수한 예방 또는 치료 효과를 가진다.

이하, 본 발명이 속하는 기술 분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있도록 본 발명의 실시예에 대하여 상세히 설명한다. 그러나 본 발명은 여러 가지 상위한 형태로 구현될 수 있으며 여기에서 설명하는 실시예에 한정되지 않는다.
실시예 1.다이페나진 화합물의 생산
실험 장치
비선광도는 Rudolph analytical사의 Autopol Ⅲ S2 편광계를 사용하여 측정하였다. CD 스펙트럼 측정은 대한민국 창원에 있는 창원대학교 공동실험관에 있는 JASCO사의 J-1500 편광계를 이용하였다. UV 스펙트럼은 Shimadzu 사의 UV-1650PC 분광광도계를 통해 얻었고, IR 스펙트럼은 JASCO 사의 FT/IR-4100 분광광도계를 이용하여 얻었다. NMR 스펙트럼은 Bruker사의 AVANCE Ⅲ 600 분광기로 1H은 600 MHz, 13C은 150 Hz에서 측정하였고, 3 mm 프로브를 사용하여 얻었다. 화학적 이동 값은 CDCl3의 용매피크(δH 7.24, δC 77.23)를 기준으로 하였다. EIMS 데이터는 Agilent사의 6100 단일 사중극자 질량분석기를 사용하여 얻었고, HRESIMS 데이터는 대한민국 청주에 있는 한국기초과학지원연구원 (KBSI)의 Waters사의 SYNPT G2 Q-TOF 질량분석기를 통하여 얻었다. PrimeLine사의 바이너리 펌프와 Shodex사의 RI-101 굴절률 검출기와 S3210 가변형 UV 검출기로 구성된 고성능 액체 크로마토그래피(HPLC)를 사용하였다. HPLC용 칼럼은 YMC-pack Silica(250 mm × 10 mm, 5 μm), YMC-Triart C18(250 mm × 10 mm, 5 μm 와 250 mm × 4.6 mm, 5 μm)를 사용하였다. 개방형 칼럼 크로마토그래피 용 충진제는 Silica gel 60(Merck, 230-400 mesh)과 역상형 실리카젤(YMC-Gel ODS-A, 12 nm, S-75 μm)을 사용하였다. 대량배양은 퍼멘텍 사의 100 L 발효기를 이용하여 진행하였다. 모든 용매는 HPLC 등급 또는 증류된 용매를 사용하였다.
IV17-028 균주의 분리 및 대량 배양
시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) (GenBank 수탁번호: MK131277)는 인도양 수심 4,317 m에서 채집된 심해 퇴적토에서 분리하였다.
시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) 균주를 Bennett (BN) 한천 배지에서 28℃에서 7일 동안 배양하였고, 이후 균주를 600 mL의 BN 액체 배지 (증류수 1L 중에 글루코오스 10g, 효모 추출물 1g, 트립톤 2g, 쇠고기 추출물 1g, 글리세롤 5g 및 해수염 32g 포함)가 들어 있는 2 L 플라스크에 접종하여 140rpm으로 교반하면서 28℃에서 5일 동안 종균 배양하였다. 종균 배양액을 60 L BN 액체 배지가 든 100 L 발효기에 접종하였고, 28℃, 50 rpm, 20 LPM의 조건으로 10일간 대량배양을 진행하였다.
상기 시스토바시디움 라린지스 IV17-028 (Cystobasidium laryngis IV17-028) 균주를 2018년 11월 20일에 기탁하여 기탁번호 KCTC 13720BP를 부여받았다.
화합물의 분리
60 L 대량배양액을 동량의 에틸아세테이트(EtOAc)로 2번, 총 120 L를 추출하였다. EtOAc 추출물은 농축하여 조추출물 약 46 g을 수득하였다. 이 중, 조추출물 33.7 g을 물 1 L와 n-헥산(n-hexane), 클로로포름(chloroform), 에틸아세테이트 (EtOAc), n-부탄올(n-BuOH) 등과 순차적으로 분획하여, 각각 5.8, 14.5, 1.1, 0.9 g을 얻었다. 클로로포름 농축물 5.8 g을 실리카 플래시 컬럼 크로마토그래피를 메틸렌 클로라이드(CH2Cl2):메탄올(MeOH) (v/v, 10:0, 20:1, 10:1, 5:1, 1:1)의 비율로 혼합한 유기용매를 단계적으로 용출하였다. CH2Cl2/MeOH (20:1) 분획물을 총 9개로 나누어 받았다. 이 중, 6-7번 소분획물 4.8 을 역상 플래시 컬럼 크로마토그래피를 메탄올:물 (v/v, 1:4, 2:3, 3:2, 4:1, 5:0)의 비율로 혼합한 유기용매를 단계적으로 용출하였고, 각각의 분획물을 3개씩 나누었다. 3번째 80% 메탄올 분획물 468.5 mg을 순상 HPLC (YMC-Pack SIL 250 mm × 10 mm, 5 μm; EtOAc/Hex/MeOH (v/v, 45:55:0.02); 유속: 2.0 mL/min; 검출기: RI)하여 6개의 화합물 (1: 15.6 mg, tR 34 min, 2: 1.9 mg, tR 39 min, 3: 3.0 mg, tR 40 min, 4: 4.0 mg, tR 37 min, 5: 1.9 mg, tR 29 min, 6: 1.8 mg, tR 23 min)을 분리정제 하였다. 화합물 1,2,4-6의 MTPA 에스터화
화합물 1,2,4-6의 절대구조를 확인하기 위해 변형된 모셔의 방법을 이요하기 위해 화합물 1,2,4-6을 MTPA 에스터화 시켰다.
화합물 1(0.2 mg 씩, 0.4 μmol)을 두개의 350 μL 튜브에 넣은 뒤, N2 가스로 건조시켰다. 소량의 4-다이메틸아미노피리딘 (DMAP)과 교반 자석을 넣은 뒤, 16시간 동안 진공건조 시켰다. 무수 피리딘(pyridine) 80 μL 넣은 뒤, 상온에서 5분간 교반 하였다. 그리고나서, S- 또는 R-MTPA-Cl 를 3 μL씩 각각의 튜브에 첨가한 뒤, 상온에서 16시간 동안 교반 하였다. 반응물은 40 ℃, N2 가스로 농축하였다. 반응물을 MC에 녹인 뒤, 1N 염화수소 (HCl), 포화 탄산수소나트륨 (NaHCO3) 용액과, 소금물 (Brine)로 씻어내었다. 유기층은 무수 황산마그네슘 (MgSO4)으로 수분을 제거한 뒤, 필터하여 감압 농축하였다. 나머지 화합물 2, 4-6도 동일한 방법으로 S- 또는 R-MTPA 에스터화 하였다. MTPA 에스터화된 화합물들의 입체중심 주변의 화학적 이동값은 1H와 COSY 스펙트럼 분석을 통하여 확인하였다. (도 3)

화합물 1의 산화를 통한 화합물 3의 확인
화합물 3의 절대구조는 화합물 1을 산화시켜 화합물 3을 제조함으로써 확인하였다.
화합물 1(1.5 mg, 2.89 μmol)을 2 mL 갈색 바이알에 넣고, N2 가스로 건조하였다. Dess-Martin periodinane(7.4 mg, 17.36 μmol)을 1이 든 바이알에 넣은 뒤, 16시간 진공건조 시켰다. 무수 MC(800 μL)를 넣은 뒤, 상온에서 18시간 동안 교반 하였다. 반응물을 MC로 희석한 뒤, 1:1 10% 싸이티오황산 나트륨(Na2S2O3)/포화 탄산수소나트륨(NaHCO3)과 소금물(Brine)로 씻었다. MC 층은 무수 황산 마그네슘(MgSO4)로 수분을 제거한 뒤, 필터하여 감압 농축하여 3이 포함된 혼합물 0.8 mg을 얻었다. 혼합물을 순상 HPLC(YMC-Pack SIL, 250 mm × 4.6 mm, 5 μm; EtOAc/Hex/MeOH(v/v, 50:50:0.02); 유속: 0.7 mL/min; 검출기: UV; 파장: 254 nm)를 이용하여 3(0.3 mg, t

화합물 6의 가수분해
화합물 6의 C-12의 절대구조를 확인하고자 (R)-사페닉산과 비선광도 값을 비교하기 위해 화합물 6을 6N 염화수소(HCl)로 가수분해하였다.
화합물 6(0.2 mg, 0.4 μmol)이 든 4 mL 갈색 바이알에, 6N 염화수소(HCl) 200 μL을 넣고 80 ℃에서 30분 동안 교반하였다. 반응이 종료됨은 LC-MS를 통해 확인하였고, 6의 가수분해물은 40 ℃, N2 가스로 농축하였다.

화합물 6의 반합성 과정 및 화합물 7의 합성
화합물 8로서 (R)-사페닉산 (17.8 mg, 0.066 mmol)을 무수 디메틸포름아미드(dimethylformamide, DMF) 1 mL에 녹인 뒤, 하이드록시벤조트리아졸(HOBt, 10.2 mg, 0.066 mmol), 1-(3-다이에틸아미노프로필)-3-에틸카보디이미드 염산염(EDCI·HCl, 12.6 mg, 0.066 mmol)과 4-다이메틸아미노피리딘(DMAP, 4.8 mg, 0.039 mmol)을 순차적으로 넣고, 0 ℃에서 10분간 교반 하였다. 그리고 나서 화합물 8(21.2 mg, 0.079 mmol)을 넣고, 상온에서 21시간 동안 교반 하였다. 반응물을 감압 농축한 뒤, 에틸 아세테이트(EtOAc)에 다시 녹인 뒤, 소금물(Brine)로 세번 씻었다. 에틸 아세테이트 층은 무수 황산 마그네슘(MgSO4)으로 수분을 제거한 뒤, 감압 농축하였다. 농축된 혼합물을 순상 HPLC(YMC-pack SIL; 250 × 10 mm; 5 μm; EtOAc:Hex:MeOH (v/v, 50:50:0.1); 유속: 2.5 mL/min; 검출기: RI)로 합성된 화합물 6 (6.9 mg, t

구조결정을 위한 전산 분석
초기 기하학 최적화(initial geometry)와 구조 검색(conformational search)은 Conflex 8(Rev. B, Conflex Corp., 도쿄, 일본) 프로그램을 사용하여 얻었다. ECD를 위한 최적화와 계산은 Gaussian 16(Rev. B.01, Gaussian Corp., 코네티컷, 미국) 프로그램을 사용하여 수행하였다. 구조 검색(conformational search)은 MMFF94s force field를 사용하여 계산하였고, 검색범위는 10 Kcal/mol로 제한하였다. 화합물 1-2와 화합물 4-5는 ground state method에서 Hatree-Fock/6-31+G(d,p), 가스 상으로 최적화 하였다. ECD 스펙트럼의 이론적 계산은 TD-SCF method에서 Hartree-FoCK/6-31+G(d,p), gas phase로 계산하였다. ECD 스펙트럼은 SpecDis(v. 1.71) 프로그램으로 σ = 0.30 - 0.50 eV의 값으로 시뮬레이션 되었다. 모든 계산된 곡선들은 실험 스펙트럼과 맞추기 위하여, +20 - 60 nm 정도 조절하였다.
화합물 1 내지 6의 입체구조 결정
화합물 1은 연녹색의 무정형 고체로 분리되었다. 화합물 1의 분자식은 HR-ESIMS에서 C30H22N4O5의 [M + Na]+ 피크에 해당하는 m/z 541.1487 (calcd for 541.1488)를 확인하였다. 화합물 1은 22의 불포화도를 가졌다. 화합물 1의 1H (표 2)과 HSQC 스펙트럼은 11개의 방향족의 수소 신호[δ

max 253과 371 nm), 1H과 13C NMR 데이터 [δH 8.95 - 7.7 ppm 대의 벗겨진 방향족 수소 신호, 8개의 질소를 포함하는 4차탄소 신호(δC 142.9, 142.3, 141.3, 140.5, 140.4, 140.2×2, 140.0), 2개의 카복실산(δC 166.1과 166.0), 2개의 메틴(δC 67.8과 34.1), 2개의 메틸기(δC 22.9와 21.4)]는 페나진 링을 포함하는 1,6-이중치환된 사페닉산과 매우 유사하였고, 화합물 1이 사페닉산의 이량체임을 확인하였다. COSY 스펙트럼 분석을 통해 6개의 스핀 시스템[H-2(δH 8.93)/H-4(δH 8.33), H-7(δH 7.67)/H-9(δH 8.13), H-12(δH 7.07)/H-13(δH 2.00), H-2'(δH 8.95)/H-3'(δH 8.02), H-7'(δH 7.77)/H-9'(δH 8.15) 및 H-12'(δH 5.24)/H-13'(δH 1.12)]을 확인하였다. 두 카복실산의 위치는 HMBC의 상호관계[H-2(δH 8.93)와 C-11 (δC 166.0), H-2'(δH 8.95)과 C-11'(δC 166.1)]를 통해 결정되었다. 산소가 치환된 메틴 수소 H-12'(δH 5.24)와 메틸 수소 H-13'(δH 1.12)는 C-6'(δC 143.7)와 C-7'(δC 127.9)에 각각 연결됨을 HMBC를 통해 확인하였다. 두 사페닉산의 연결됨은 H-13(δH 2.00)에서 C-고리의 C-6 (δC 145.8), A'-고리의 C-4'(δC 150.9)으로, C-고리의 H-7(δH 7.67)과 A'-고리의 H-3'(δH 8.02)에서 C-12(δC 34.1)로의 HMBC 상호연결을 통해 확인하였다. 화합물 1의 1H, 13C NMR 스펙트럼은 기존에 보고된 페나조스타틴 C(phenazostatin C)와 매우 유사하였다. 이 둘의 차이는 화합물 1이 카복실릭 메틸 에스터 대신 카복실산과 C-6'의 위치에 이차 알콜을 가지고 있는 것이다.
짝지음 상수(coupling constatnts)나 NOESY 데이터에서의 불충분한 신호로 화합물 1의 상대구조를 분명하게 정할 수 없었다. 이론적으로 예상 가능한 화합물 1의 구조는 두 개의 카이랄(C-12 와 C-12')의 존재로 4가지의 가능성이 있다. 먼저, C-12'의 절대구조는 변형된 모셔의 방법(Mosher's method)을 이용하여 결정하였다. 1을 S- 또는 R-MTPA 에스터 1a 와 1b로 각각 반응시켰다. 1a와 1b의 1H NMR 신호들은 COSY 스펙트럼 분석을 통해 정하였고, ΔδH값은 δS-esterδR-ester으로 계산하였다(도 3). 이러한 결과를 바탕으로 C-12'의 절대구조는 R 인 것으로 확인되었다. 남은 두 개의 부분 입체 이성질체(12R, 12'R 과 12S, 12'R)는 가우시안 16을 이용하여 이론적인 ECD 스펙트럼을 계산하여 측정한 ECD 스펙트럼과 비교하였다. 화합물 1의 실제 ECD 곡선은 12R, 12'R-형태의 이성질체와 일치하였다(도 5). 따라서 C-12의 절대구조는 R인 것으로 결정하였다.
화합물 2는 연녹색의 무정형 고체로 얻어졌다. 화합물 2의 분자식은 HR-ESIMS 데이터를 기반으로 화합물 1과 같은 C30H22N4O5이었다. 화합물 2의 1H과 13C NMR 스펙트럼(표 2) 또한 1과 매우 유사하였으나, H-12'신호(ΔδH 4.95, m)만 달라 화합물 2의 평면구조는 화합물 1과 동일하였다. 그러나, 화합물 2의 비선광도(optical rotation)의 값이 화합물 1과는 반대로 나왔으며, ECD 스펙트럼 또한 곡선이 반대로 나타났다[화합물 1: [α]25
D-167 (c 0.1, CHCl3); ECD (CHCl3) λmax (Δε) 242 (-38.20), 268 (+148.53), 화합물 2: [α]25
D+88 (c 0.1, CHCl3); ECD (CHCl3) λmax (Δε) 247 (+15.90), 272 (-24.16)]. 화합물 1과 화합물 2의 유사점들과 차이점들은 이들이 서로 입체 이성질체임을 보여주었다. 화합물 2의 C-12'에 대한 절대구조는 화합물 1과 동일한 방식으로 변형된 모셔의 방법으로 R-구조임을 확인하여, 화합물 2의 절대구조는 12S, 12'R-구조임을 확인하였다. 흥미롭게도, 에스터화된 2a 와 2b의 H-13'의 화학적 이동값은 각각 δH 0.26 와 δH 0.24로 1a(δH 1.49) 과 1b(δH 1.46)의 값보다 1.2 ppm 더 업 필드(upfield)로 이동하였다. 이는 12S-배향으로 인한 독특한 입체장애 때문인 것으로 보인다(도 5).
화합물 3은 연녹색의 무정형의 고체로, HRESIMS 데이터를 바탕으로 분자식 C30H20N4O5(불포화도 23)임을 확인하였다. 화합물 3의 1D NMR 스펙트럼(표 2)은 화합물 1 및 화합물 2와 유사하였다. 그러나, 화합물 1, 2에 존재하였던 이차 알콜 대신, 아세틸 그룹 (C-12', δC 200.3)이 존재하였다. 따라서, 화합물 3의 구조는 1 또는 2의 산화된 형태임을 알 수 있었다. C-12의 절대구조를 결정하기 위해, 산화된 1과 화합물 3의 비선광도 값을 비교하였다. 화합물 1을 산화시킨 뒤, HPLC를 이용하여 순수하게 분리하였고, LC-MS와 1H NMR 스펙트럼을 통해 화합물 3과 동일 함을 확인하였다. 산화된 화합물 1([α]25
D-10)의 비선광도 값은 3([α]25
D-30)과 같은 음의 값을 보였다. 따라서 C-12의 절대구조는 R인 것으로 결정하였다.
화합물 4-6은 HRESIMS 데이터에서 화합물 1, 2와 동일한 분자식 C30H22N4O5를 나타냈으나, 1H NMR 스펙트럼의 갈라짐 양식(splitting pattern)이 화합물 1, 2 와는 달랐다. 이는 화합물 4-6이 화합물 1, 2의 부분 입체 이성질체임을 의미한다. 화합물 4의 1H NMR 스펙트럼에서, 2개의 단일항(singlet) 수소 신호 (δH 9.16, s, H-2'과 δH 8.41, s, H-4')를 나타내었다. A'고리에 존재하는 카복실산의 위치는 H-2'(δH 9.16)과 C-11'(δC 166.2) 사이의 HMBC 상관관계를 통하여 확인하였다. 두 사페닉산의 연결을 H-2'(δH 9.16)에서 C-12 (δC 39.7)로, H-12(δH 5.97)에서 C-3'(δC 149.6), C-4'(δC 131.7), C-5a(δC 142.2), C-6(δC 144.2), 그리고 C-7(δC 129.1)로의 HMBC 신호를 통하여 확인하였다 (도 2). 화합물 4와 달리, 화합물 5는 두개의 단일항 수소 신호 δH 7.90(s, H-7'과 H-9')가 겹쳐 나타났다. 두 카복실산의 위치는 H-2(δH 8.93)에서 C-11(δC 165.7)로, H-2'(δH 8.90)에서 C-11'(δC 165.9)으로의 HMBC 상관관계를 통해 확인하였다. 따라서 중첩된 두개의 단일항 수소는 C'고리에 속해 있었다. 두 사페닉 산의 연결은 H-12(δH 6.78)에서 C-7(δC 128.5)과 C-8'(δC 143.6)으로의 HMBC 상관관계를 통해 확인하였다. 이러한 결과는, 화합물 4는 C고리와 A'고리가 C-12를 통해 연결되어 있는 반면, 화합물 5는 C고리와 C'고리가 C-12를 통해 연결되어 있음을 나타내었다. 화합물 4와 5의 절대구조는 화합물 1, 2와 동일한 방식으로 결정하였다(도 6, 7). 따라서, 화합물 4와 5의 절대구조는 12S, 12'R과 12R, 12'R인 것으로 각각 결정하였다.
마지막으로, 화합물 6 또한, 1D 및 2D NMR 데이터 분석을 통하여 결정하였다. 화합물 6의 1H NMR 스펙트럼은 산소가 치환된 메틴 수소(δH 7.65, H-12)가 다른 화합물 1-5의 값보다 다운필드(downfield)로 이동해 있었다. 두 사페닉산 간의 연결은 H-7(δH 8.54)에서 C-12(δC 70.0)로, H-12(δH 7.65)과 H-2'(δH 8.33)에서 A'고리에 연결된 카복실산의 C-11'(δC 165.8)로의 HMBC 연결 신호를 통하여 에스터 결합을 이루고 있음을 확인하였다. C-12'의 절대구조는 변형된 모셔의 방법을 이용하여 R임을 확인하였다. C-12의 절대구조를 확인하기 위해, 화합물 6을 6N 염화수소(HCl)로 가수분해한 뒤, 그 가수분해물을 화합물 8인 (R)-사페닉산과 함께 비선광도 값을 비교하였다. 화합물 6의 가수분해물의 비선광도 값([α]25
D-10)은 (R)-사페닉산(화합물 8)([α]25
D-13)과 동일한 음의 값을 보였다. 결과적으로, 화합물 6의 절대구조는 12R, 12'R 로 결정되었다.
실험예 1. 다이페나진 화합물의 항뇌신경염증 활성 및 세포 생존력 평가
항뇌신경염증 활성을 확인하기 위한 NO생성은 Griess 시약을 사용하여 배지 내의 아질산염(nitrite) 농도를 측정함으로써 평가하였고, 세포 생존력은 MTT assay를 이용하여 평가하였다. 간략하게, BV-2 미세아교세포를 5% heat-inactivated FBS와 페니실린-스트렙토마이신 50 μg/mL를 첨가한 DMEM(Dulbecco's modified Eagle's medium)배지에서 37℃, 5% CO2 인큐베이터에서 배양하였다. 미리 배양한 세포(2.5 Х 104 cells/mL)를 분주한 96-well plate에 다양한 농도(10, 20, 50 및 100 μM)의 화합물들을 1시간 동안 처리하고, LPS(200 ng/mL)를 처리하고 24시간 배양하였다. 상층액을 Griess 시약과 섞고, 10분간 상온에서 배양하였다. Nitrite 농도는 세포배양 배지에 준비된 sodium nitrite 표준용액을 사용하여 결정하였다. 마이크로플레이트 리더기로 540 nm 파장에서 흡광도를 측정하였다. 세포 생존력을 평가하기 위해서, 세포(2.5 × 104 cells/mL)를 분주한 96-well plate에 다양한 농도(10, 20, 50 및 100 μM)의 화합물들을 1시간 동안 처리하고, LPS(200 ng/mL)를 처리하고 24시간 배양하였다. 각각의 well에 MTT 용액(0.5 mg/mL)을 첨가한 뒤, 37 ℃, 4 시간 동안 배양하였다. 상층액을 제거하고, 형성된 formazan crystal을 DMSO에 녹였다. 이를 마이크로플레이트 리더기를 사용하여 550 nm에서 흡광도를 측정하였다. 화합물 6의 IC50값을 구하기 위해서, NO 생성 및 MTT 분석을 앞서 기술한 것과 동일한 방식으로 농도(0.25, 0.5 및 1 μM)만 다르게 진행하였다. (표 4) 실험은 3반복 진행하였다.
BV-2 미세아교세포(microglia)에 대한 세포독성을 MTT 분석을 이용해 측정하였다. 모든 화합물들은 사페닉산 부분으로 구성되어 있으나, 두 사페닉산의 연결된 패턴에 따라 활성 세기에 큰 차이를 보였다(도 8 및 표 4). 특히, 화합물 6이 0.3 μM 농도의 가장 뛰어난 IC50 값을 보였다(도 9). 이는 6의 활성이 다른 화합물들 보다 최소 54배(8, 16.3 μM)에서 최고 552배(1, 165.6 μM)까지 더 뛰어난 활성을 나타냄을 의미한다. 화합물 1과 화합물 2는 서로 부분 입체 이성질체 관계이고, 화합물 3은 화합물 1의 산화된 형태였으나, 화합물 3(34.7 μM)은 화합물 1(165.6 μM)보다 5배 더 강하고, 화합물 2(90.2 μM)는 화합물 1(165.6 μM) 보다 약 2배 더 강한 활성을 보였다(표 4). 앞서 언급한 바와 같이, C-12의 입체구조로부터 기인되는 독특한 입체적 장애의 차이(도 5)가 활성에 영향을 준다는 것을 발견하였다. 세포 생존력(cell viability) 시험에서, 화합물 6이 1 μM의 농도까지 강력한 세포독성을 보였다.
화합물 6은 다른 화합물들과 다르게 항뇌신경염증 활성을 보였기 때문에, 추가적인 생리활성 실험을 위해 화합물 6을 합성하고자 하였다. 반합성된 6은 사페닉산(화합물 8)을 이용하여 EDCI 커플링 반응을 통해 합성되었고, 천연물 6과 동일한 1H, 13C 및 MS 스펙트럼을 보였다. 화합물 6의 정제과정 중, 화합물 6의 산화된 형태의 부산물인 화합물 7도 얻게 되었다(도 10). 화합물 7의 1H NMR 스펙트럼이 화합물 6과 비슷하였으나, 산소가 치환된 메틴(δH 5.72, 1H)과 연결된 메틸의 신호(δH 1.97, d, 3H)가 아세틸기의 신호(δH 3.08, s, 3H)로 변한 것을 확인하였다. 화합물 7의 분자량 또한 화합물 6보다 적은 분자량 516 인 것을 LRESIMS 스펙트럼에서 확인하여 화합물 7이 화합물 6의 산화된 형태임을 나타내었다. 따라서 화합물 7의 구조는 (R)-6-(1-((6-아세틸페나진-1-카르보닐)옥시)에틸)페나진-1-카르복실산 ((R)-6-(1-((6-acetylphenazine-1-carbonyl) oxy)ethyl)phenazine-1-carboxylic acid)로 결정하였다.
실험예 2. 다이페나진 화합물의 항암활성 분석
세포독성 테스트는 6개의 인간 암세포주에 대해 술포로다민 B (sulforhodamine B, SRB) 분석으로 평가하였다. 인간의 암세포주, ACHN(신장), NCI-H23(폐), PC-3(전립선), NUGC-3(위), MDA-MB-231(유방) 및 HCT-15(결장)는 ATCC(American Type Culture Collection)에서 구입하였다. 세포는 10% FBS(fetal bovine serum)이 첨가된 RPMI 1640배지로 37℃, 5% CO2 인큐베이터에서 배양하였다. 세포(8 × 103 cells/well)를 96-well plate에 분주하고 하루동안 배양한 뒤, 0.1% DMSO에 녹인 화합물 1-8 및 화합물 9로써 종래 공지된 (R)-6-(1-((2-아미노벤조일)옥시)에틸)페나진-1-카르복실산 (R)-(6-(1-((2-aminobenzoyl)oxy)ethyl)phenazine-1-carboxylic acid)과 양성대조군 아드리아마이신을 세포에 처리하였다. 48 시간동안 배양한 뒤, 50% 트리클로로초산(trichloroacetic acid, 50 μg/mL)으로 고정하고 1% 아세트산(acetic acid)에 녹인 0.4% surfohodamine B로 염색하였다. 염색되지 않은 것은 1% 아세트산으로 씻어 제거하고, 단백질과 결합된 것은 10 mM Tris base(pH 10.5)로 추출하여 광학밀도(optical density)를 측정하였다. 마이크로플레이트 리더기를 사용하여 540 nm에서 흡광도를 측정하고, GI50값은 GraphPad Prism 4.0을 사용하여 계산하였다.
테스트된 화합물은 세포독성을 보이지 않았으나, 화합물 6과 화합물 6이 산화된 형태인 화합물 7에서만 양성대조군 아드리아마이신(Adriamycin)보다 2-19배 더 강력한 세포독성을 나타내었다. 또한 화합물 6은 화합물 7보다 4-6배 더 강력한 활성을 보였다. 특히, 위암세포인 NUGC-3에서는 GI50 값이 7.7 nM으로 가장 강력한 활성을 보였으며, 이는 아드리아마이신보다 19배 더 강력하였다. 이러한 결과는 C'고리의 하이드록시 그룹과 에스터 결합이 활성에 큰 역할을 함을 나타내었다. 하기의 표 5에 화합물 1-9의 6종의 암세포들에 대한 세포독성 결과를 나타내었다 (GI50 = μM).
이상으로 본 발명 내용의 특정한 부분을 상세히 기술 하였는바, 당업계의 통상의 지식을 가진 자에게 있어서, 이러한 구체적 기술은 단지 바람직한 실시 양태일 뿐이며, 이에 의해 본 발명의 범위가 제한되는 것이 아닌 점은 명백할 것이다. 따라서 본 발명의 실질적인 범위는 첨부된 청구항들과 그것들의 등가물에 의하여 정의된다고 할 것이다.

- 제1항에 있어서,상기 화학식 1의 화합물이 하기 화합물로 이루어진 군으로부터 선택되는 어느 하나인 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염:4-((R)-1-(6-카르복시페나진-1-일)에틸)-6-((R)-1-히드록시에틸)페나진-1-카르복실산;4-((S)-1-(6-카르복시페나진-1-일)에틸)-6-((R)-1-히드록시에틸)페나진-1-카르복실산;(R)-6-아세틸-4-(1-(6-카르복시페나진-1-일)에틸)페나진-1-카르복실산.3-((S)-1-(6-카르복시페나진-1-일)에틸)-6-((R)-1-히드록시에틸)페나진-1-카르복실산;8-((R)-1-(6-카르복시페나진-1-일)에틸)-6-((R)-1-히드록시에틸)페나진-1-카르복실산;6-((R)-1-((6-((R)-1-히드록시에틸)페나진-1-카르보닐)옥시)에틸)페나진-1-카르복실산; 및(R)-6-(1-((6-아세틸페나진-1-카르보닐)옥시)에틸)페나진-1-카르복실산.

- 제11항에 있어서, 상기 신경염증 질환은 뇌염증 질환, 다발성 경화증, 우울증, 뇌졸중, 알츠하이머 병, 파킨슨 병, 루게릭 병, 헌팅턴 병, 크로이츠펠트야콥병, 외상 후 스트레스 장애, 정신분열증 및 근위축성측색경화증으로 이루어진 군에서 어느 하나 이상 선택되는 것인, 암 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제5항 중 어느 한 항에 따른 다이페나진 화합물, 이의 광학이성질체 또는 이의 약학적으로 허용가능한 염을 유효성분으로 포함하는 신경염증 질환 예방 또는 개선용 식품 조성물.
- 제13항에 있어서, 상기 신경염증 질환은 뇌염증 질환, 다발성 경화증, 우울증, 뇌졸중, 알츠하이머 병, 파킨슨 병, 루게릭 병, 헌팅턴 병, 크로이츠펠트야콥병, 외상 후 스트레스 장애, 정신분열증 및 근위축성측색경화증으로 이루어진 군에서 어느 하나 이상 선택되는 것인, 신경염증 질환 예방 또는 개선용 식품 조성물.
- 제1항 내지 제5항 중 어느 한 항에 따른 다이페나진 화합물, 이의 광학이성질체 및 이의 약학적으로 허용 가능한 염을 이를 필요로 하는 개체에 투여하는 단계를 포함하는, 암 관련 질환의 예방 또는 치료 방법.
