Abstract

본 발명은 네오아가로올리고당을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 조성물에 관한 것이다.
한천(Agar)은 오래 전부터 식품첨가물, 의약품, 화장품, 가축사료 및 공업원료 등으로 널리 이용되고 있는 대표적인 해조류 유래 다당류로, 국내의 경우 해마다 그 생산량이 약 3,600톤에 이르고 있는 비교적 풍부한 수산자원의 하나이다. 그러나 실제 이용 면에서는 전체생산량의 일부만이 단순가공 처리되어 값싼 원료로 사용될 뿐이며, 그 나머지는 대부분 방치되고 있어 부존자원량에 비해 부가가치가 매우 낮은 실정이다. 따라서 풍부한 국내 한천의 새로운 용도개발과 부가가치 향상에 관한 연구가 크게 요구되고 있는 실정이다.
한천은 아가로스(agarose)와 아가로펙틴(agaropectin)으로 구성되어있는데, 아가로스는 갈락토스(Dgalactose)와 3,6-anhydro-L-galactose가 β-1, 4 형태로 결합한 단위체인 아가로비오스(agarobiose)가 반복되어 α-1, 3 결합으로 연결된 직쇄구조로 되어있고 겔(gel)화력이 강한 반면, 아가로펙틴은 아가로스와 마찬가지로 아가로비오스 단위로 되어 있으나 황산기 등의 산성기를 함유하며 겔화력이 약하다.
이중 아가로스는 β-1, 4 결합에 작용하는 베타-아가레즈(β-agarase)에 의해서 네오아가로테트라오스(neoagarotetraose)를 거쳐 네오아가로비오스(neoagarobiose)로 분해되고 계속해서 α-1, 3 결합에 작용하는 알파-아가레즈(α-agarase)에 의해서 갈락토스(D-galactose)와 3,6-anhydro-L-galactose로 최종 분해된다.

본 발명자들은 상기와 같은 요구를 충족시키기 위하여 연구를 거듭한 결과 DagA 효소반응으로 제조한 한천 유래의 네오아가로올리고당이 박테리아의 증식 억제뿐 아니라, 죽은 박테리아로부터 분리된 내독소를 제거하는 우수한 효과를 가지고 있어, 항생제와 병용할 경우 항생제에 의한 부작용을 최소화할 수 있으며, 단독 투여에 비해 현저하게 우수한 패혈증 치료 효과를 보임을 확인함으로써 본 발명을 완성하였다.
본 발명의 목적은 네오아가로올리고당을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 네오아가로올리고당을 유효성분으로 포함하는 면역 증강용 조성물을 제공하는 것이다.
상기와 같은 과제를 해결하기 위해, 본 발명은 네오아가로올리고당을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물을 제공한다.
또한, 본 발명은 네오아가로올리고당을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 개선용 식품 조성물을 제공한다.
또한, 본 발명은 네오아가로올리고당을 유효성분으로 포함하는 면역 증강용 약학적 조성물을 제공한다.
또한, 본 발명은 네오아가로올리고당을 유효성분으로 포함하는 면역 증강용 기능성 식품을 제공한다.
본 발명에 따른 네오아가로올리고당은 우수한 패혈증 예방 효과를 나타낼 뿐만 아니라 염증발생을 효과적으로 억제함으로써 면역증진 효과가 탁월하므로, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료 및 면역 증진을 위한 의약품 및 기능성 식품에 유용하게 사용될 수 있다.

이하, 본 발명에 대해 상세히 설명한다.
본 발명은 한천(agar) 또는 아가로스(agarose)로부터 DagA 효소반응으로 제조한 네오아가로올리고당(neoagarooligosaccharide)을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 조성물을 제공한다.
상기 조성물은 약학적 조성물 및 식품 조성물을 포함한다.
본 발명의 DagA는 베타-아가레즈(β-agarase)로 서열번호 2의 아미노산 서열을 갖는 단백질을 의미하며, 방선균인 스트렙토마이세스 시리칼라(Streptomyces coelicolor)로부터 생산되거나 이종균주로부터 생산된 단백질을 포함한다. 또한, 기능이 전혀 다른 것으로 변경되거나 베타-아가레즈 활성을 잃지 않는 범위 내에서 통상적인 유전자 재조합 방법 즉, 정제를 유리하게 하기 위해 표지 아미노산을 포함시키거나 이종 발현을 위해 아미노산 서열을 변경하는 등의 방법에 따라 재조합된 단백질을 포함한다.
보다 구체적으로, 본 발명의 DagA는 스트렙토마이세스 시리칼라가 생산하는 베타-아가레즈로 유전자로부터 번역될 때 서열번호 2의 309개 아미노산을 갖고 분자량이 약 35kDa인 상태로 생산되며, N-말단 30개의 아미노산 시그널 펩티드가 잘려져서 서열번호 2의 31 내지 309번의 아미노산 서열을 갖는 완성된 세포외형 단백질(약 32kDa)의 상태로 분비된다.
스트렙토마이세스 시리칼라의 dagA 유전자가 상기 DagA를 암호화하며, dagA 유전자는 서열번호 1의 염기서열로 표시될 수 있다. 서열번호 1은 스트렙토마이세스 시리칼라 A3(2)의 게놈에 존재하는 유전자의 염기서열로 NCBI(미국 국립생물정보센터) 데이터베이스 상에 "SCO3471"로 명명되어 있다. In vitro 실험을 통해 확인된 바에 따르면, dagA 유전자의 전사는 RNA 중합효소의 적어도 다른 3가지의 완전효소(holoenzyme)에 의해 인지되는 4 또는 5가지의 다른 프로모터(promoter)에 의해 조절된다. dagA 유전자의 전사단계 분석을 통해, 전사가 암호화 서열의 상부 32, 77, 125 및 220번째 염기에서 개시되는 것으로 나타났다.
본래 DagA의 생산 균주인 스트렙토마이세스 시리칼라의 배양을 통해 본 발명의 DagA를 생산할 수 있으나, 생산효율을 높이기 위해 이종균주인 스트렙토마이세스 리비던스(Streptomyces lividans)의 발현 시스템을 이용하는 것이 바람직하며, 상기 dagA 유전자를 방선균용 벡터에 삽입하여 재조합 벡터를 제조한 다음 이 재조합 벡터로 스트렙토마이세스 리비던스를 형질전환하고, 이 형질전환체를 배양하는 방법을 사용할 수 있다. 이 경우 재조합 벡터는 dagA 유전자가 방선균 유래 프로모터에 의해 전사가 조절될 수 있도록 구성하는 것이 바람직하다.
방선균 유래 프로모터에는 sgtR 프로모터(sgtRp), ermE 프로모터(ermEp), tipA 프로모터(tipAp) 등 다양한 프로모터가 존재하므로 재조합 벡터를 제조할 때 이들을 선택하여 사용할 수 있다. 이들 프로모터에 의해 전사가 조절될 수 있도록 구성된 여러 종류의 벡터들이 개발되어 있어, 이 벡터에 SCO3471를 클로닝하면 방선균 유래 프로모터에 의해 전사가 조절되는 구조의 재조합 벡터를 제조할 수 있다.
형질전환체는 숙주균주를 재조합 벡터로 형질전환시켜 제조할 수 있는데, 숙주균주에 따라 형질전환을 위한 방법이 다양하게 존재하므로, 적절한 방법을 선택하여 사용할 수 있다. 예를 들어, 스트렙토마이세스 리비던스를 숙주균주로 사용하는 경우 PEG(polyethylene glycol)를 매개로한 형질전환 방법을 사용할 수 있다.
형질전환체 등과 같은 DagA 생산균주를 액체배지에서 배양하면 DagA를 생산할 수 있으며, 배양액을 수득하여 한외여과법과 같은 통상의 단백질 정제 방법을 이용하면 고순도의 DagA를 생산할 수 있다. 이때 액체배지에 한천(agar) 또는 아가로스(agarose)를 포함시키면 보다 효율적으로 DagA를 생산할 수 있다.
한천 및 아가로스와 같은 다당류는 상기 DagA의 효소반응을 통해 상대적으로 크기가 작은 올리고당이 생성될 수 있으며, 상기 효소반응은 35 내지 45℃의 온도 및 pH 6 내지 8에서 이루어지는 것이 바람직하나 이에 한정하지 않는다.
상기 효소반응의 산물로서 네오아가로올리고당이 생성되며, 상기 네오아가로올리고당은 네오아가로비오스(neoagarobiose), 네오아가로테트라오스(neoagarotetraose) 및 네오아가로헥사오스(neoagarohexaose) 각각이 될 수 있으며, 이들이 혼합된 상태가 될 수 있으나, 이에 한정하지 않는다.
본 발명에 따른 네오아가로올리고당은 전염증성 사이토카인을 감소시키고, 항염증성 사이토카인을 증가시키고, 패혈증 또는 패혈증 쇼크로 인한 사망률을 현저히 낮춤으로써 패혈증의 예방 또는 치료 효과를 나타낸다.
본 발명에 있어서, "패혈증"이란 미생물에 감염되어 전신에 심각한 염증 반응이 나타나는 상태를 말한다. 체온이 38도 이상으로 올라가는 발열 증상 혹은 36도 이하로 내려가는 저체온증, 호흡수가 분당 24회 이상으로 증가(빈호흡), 분당 90회 이상의 심박수(빈맥), 혈액 검사상 백혈구 수의 증가 혹은 현저한 감소 중 두 가지 이상의 증상을 보이는 경우, 이를 전신성 염증 반응 증후군(systemic inflammatory response syndrome; SIRS)이라고 부른다. 이러한 전신성 염증 반응 증후군이 미생물의 감염에 의한 것일 때 패혈증이라고 한다. 신체의 감염병소에서 병원균이 지속적 또는 단속적으로 혈류에 들어와 여러 장기조직에 정착하여 병소를 만들고 심한 전신 증상을 보이는 것이다. 원인균으로는 포도상구균, 연쇄상구균, 대장균, 녹농균, 결핵균, 폐렴간균, 진균, 혐기성균 등이 있다.
또한, 본 발명은 한천(agar) 또는 아가로스(agarose)로부터 DagA 효소반응으로 제조한 네오아가로올리고당(neoagarooligosaccharide)을 유효성분으로 포함하는 면역 증강용 조성물을 제공한다.
상기 조성물은 약학적 조성물 및 건강보조식품을 포함한다.
본 발명에 따른 네오아가로올리고당은 전염증성 사이토카인의 분비를 억제하고 항염증성 사이토카인의 분비를 증가시킴으로써 면역 증강 효과를 나타낸다.
상기 조성물은 면역 질환의 예방 또는 질환에 이용될 수 있으며, 상기 면역질환은 생체의 면역체계 이상으로 유발되는 다양한 염증성 질환, 알러지성 질환 및 암 등을 포함하며, 예로서 감기, 천식, 폐렴, 알러지성 비염, 알러지성 결막염, 아토피성 피부염, 알러지, 류마티스 관절염, 알츠하이머병, 자가면역성 갑상선 질환, 염증성 장질환, 재생불량성 빈혈, 홍반성 루푸스 및 건선 등을 포함하나 이에 한정하지 않는다.
본 발명에 따른 네오아가로올리고당은 우수한 패혈증 예방 효과를 나타낼 뿐만 아니라 면역증진 효과가 탁월하므로, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료, 및 면역 증진을 위한 의약품 및 기능성 식품에 유용하게 사용될 수 있다.
본 발명의 조성물은 약학적 조성물의 제조에 통상적으로 사용하는 적절한 담체, 부형제 및 희석제를 더 포함할 수 있다. 본 발명에 따른 약학적 조성물은 각각 통상의 방법에 따라 산제, 과립제, 정제, 캡슐제, 현탁액, 에멀젼, 시럽, 에어로졸 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 당해 기술 분야에 알려진 적합한 제제는 문헌(Remington's Pharmaceutical Science, Mack Publishing Company, Easton PA)에 개시되어 있는 것을 사용하는 것이 바람직하다.
본 발명의 약학적 조성물에 포함될 수 있는 담체, 부형제 및 희석제로는 락토오스, 덱스트로오스, 수크로오스, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말티톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로오스, 메틸 셀룰로오스, 미정질 셀룰로오스, 폴리비닐 피롤리돈, 물, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 탈크, 마그네슘 스테아레이트 및 광물유를 들 수 있다.
본 발명의 약학적 조성물을 제제화할 경우에는 보통 사용하는 충진제, 증량제, 결합제, 습윤제, 붕해제, 계면활성제 등의 희석제 또는 부형제를 사용하여 조제된다. 경구 투여를 위한 고형제제에는 정제, 환제, 산제, 과립제, 캡슐제 등이 포함되며, 이러한 고형제제는 상기 유효성분에 적어도 하나 이상의 부형제 예를 들면, 전분, 칼슘카보네이트, 수크로오스, 락토오스, 젤라틴 등을 섞어 조제된다. 또한 단순한 부형제 이외에 마그네슘 스테아레이트, 탈크 같은 윤활제들도 사용된다. 경구 투여를 위한 액상 제제로는 현탁제, 내용액제, 유제, 시럽제 등이 해당되는데 흔히 사용되는 단순희석제인 물, 리퀴드 파라핀 이외에 여러 가지 부형제, 예를 들면 습윤제, 감미제, 방향제, 보존제 등이 포함될 수 있다. 비경구 투여를 위한 제제에는 멸균된 수용액, 비수성용제, 현탁제, 유제, 동결건조 제제, 좌제가 포함된다. 비수성용제, 현탁제로는 프로필렌글라이콜(propylene glycol), 폴리에틸렌 글라이콜, 올리브 오일과 같은 식물성 기름, 에틸올레이트와 같은 주사 가능한 에스테르 등이 사용될 수 있다. 좌제의 기제로는 위텝솔(witepsol), 마크로골, 트윈(tween) 61, 카카오지, 라우린지, 글리세로제라틴 등이 사용될 수 있다.
본 발명에서 사용되는 용어 "투여"는 임의의 적절한 방법으로 개체에게 소정의 본 발명의 조성물을 제공하는 것을 의미한다.
본 발명의 약학적 조성물의 바람직한 투여량은 개체의 상태 및 체중, 질병의 정도, 약물형태, 투여경로 및 기간에 따라 다르지만, 당업자에 의해 적절하게 선택될 수 있다. 바람직한 효과를 위해서, 본 발명의 조성물은 1일 0.001 내지 1000 mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다.
본 발명의 약학적 조성물은 개체에게 다양한 경로로 투여될 수 있다. 투여의 모든 방식은 예상될 수 있는데, 예를 들면, 경구, 직장 또는 정맥, 근육, 피하, 자궁 내 경막 또는 뇌혈관 내 주사에 의해 투여될 수 있다. 본 발명의 약학적 조성물은 바람직하게는 주사제 형태로 투여될 수 있다.
본 발명에 따른 약학적 조성물은 상기 유효성분 외에 패혈증 또는 패혈증성 쇼크의 예방 또는 치료 또는 면역 증강 효과를 갖는 공지된 물질을 하나 이상 추가로 포함할 수 있다.
본 발명에 따른 약학적 조성물은 상기 유효성분 외에 기관지 확장제, 항히스타민제 또는 소염진통제를 추가로 포함할 수 있다.
예를 들어, 상기 기관지 확장제로는 β-효능제, 항콜린제, 메틸잔틴 등을 사용할 수 있고, 상기 항히스타민제로는 아크리바스틴(acrivastine), 세티리진(cetirizine), 데슬로라타딘(desloratadine), 펙소페나딘(fexofenadine), 레보세티리진(levocertirizine), 로라타딘(loratadine), 미졸라스틴(mizolastine), 아일메마진(ailmemazine), 클로세티리진(chlocertirizine), 클레마스틴(clemastine), 시프로펩타딘(cypropheptadine), 하이드록시진(hydroxyzine), 케토티펜(ketotifen), 프로멘타진(promenthazine) 등을 사용할 수 있고, 상기 소염진통제로는 아스피린(aspirin), 디클로페낙(diclofenac), 페노프로펜(fenoprofen), 플루비프로펜(flurbiprofen), 이부프로펜(ibuprofen), 인도메타신(indomethacin), 케토프로펜(ketoprofen), 나프록센(naproxen), 피록시캄(piroxicam), 술린닥(sulindac), 셀레콕시브(celecoxib), 발데콕시브(valdecoxib), 로페콕시브(rofecoxib) 등을 사용할 수 있다.
본 발명에 있어서 "기능성 식품"이란 식품에 물리적, 생화학적, 생물공학적 수법 등을 이용하여 해당 식품의 기능을 특정 목적에 작용, 발현하도록 부가가치를 부여한 식품군이나 식품 조성이 갖는 생체방어리듬조절, 질병방지와 회복 등에 관한 체내조절기능을 생체에 대하여 충분히 발현하도록 설계하여 가공한 식품을 의미한다.
상기 기능성 식품에는 식품학적으로 허용 가능한 식품 보조 첨가제를 포함할 수 있으며, 기능성 식품의 제조에 통상적으로 사용되는 적절한 담체, 부형제 및 희석제를 더욱 포함할 수 있다.
본 발명의 조성물을 식품 첨가물로 사용할 경우, 상기 조성물을 그대로 첨가하거나 다른 식품 또는 식품 성분과 함께 사용될 수 있고, 통상적인 방법에 따라 적절하게 사용될 수 있다. 유효성분의 혼합양은 사용 목적 (예방, 건강 또는 치료적 처치)에 따라 적합하게 결정될 수 있다. 일반적으로, 식품 또는 음료의 제조 시에 본 발명의 조성물은 원료에 대하여 15중량 % 이하, 바람직하게는 10 중량 % 이하의 양으로 첨가된다. 그러나 건강 및 위생을 목적으로 하거나 또는 건강 조절을 목적으로 하는 장기간의 섭취의 경우에는 상기 범위 이하일 수 있으며, 안전성 면에서 아무런 문제가 없기 때문에 유효성분은 상기 범위 이상의 양으로도 사용될 수 있다.
상기 외에 본 발명의 조성물은 여러 가지 영양제, 비타민, 전해질, 풍미제, 착색제, 펙트산 및 그의 염, 알긴산 및 그의 염, 유기산, 보호성 콜로이드 증점제, pH 조절제, 안정화제, 방부제, 글리세린, 알코올, 탄산 료에 사용되는 탄산화제 등을 포함할 수 있다. 그 밖에 본 발명의 조성물은 천연 과일주스, 과일주스 음료 및 야채 음료의 제조를 위한 과육을 포함할 수 있다. 이러한 성분은 독립적으로 또는 조합하여 사용할 수 있다. 이러한 첨가제의 비율은 크게 중요하진 않지만 본 발명의 조성물 100 중량부당 0.01 내지 0.1 중량부의 범위에서 선택되는 것이 일반적이다.
이하, 본 발명을 실시예에 의하여 더욱 상세하게 설명한다. 단, 하기 실시예는 본 발명을 더욱 쉽게 이해할 수 있도록 예시하는 것으로 본 발명의 내용이 실시예에 의해 한정되는 것은 아니다.
실시예 1. DagA 생산
스트렙토마이세스 시리칼라 A3(2)의 염색체 DNA를 주형으로 하고 하기의 Asm-F 및 Asm-R 프라이머를 사용한 PCR을 통해 dagA 유전자 단편(DagA의 시그널 펩타이드 및 완성형 펩타이드가 암호화된 947bp 단편, 서열번호 1에 프라이머의 일부 서열이 포함된 상태)을 증폭하였고, 단편의 제한효소 부위(NdeI/BamHI)를 이용하여 도 1에 나타낸 pUWL201pw 벡터에 dagA 유전자를 클로닝하여 dagA 유전자의 전사가 ermE 프로모터에 의해 조절되도록 재조합 벡터를 제조하였으며, 이를 도 2에 나타내었다.

상기 재조합 벡터로 스트렙토마이세스 리비던스(Streptomyces lividans) TK24를 형질전환하여 수득한 형질전환균주를 DagA의 생산을 위해 사용하였다.
이 균주를 0.3%(w/v)의 아가(agar)가 포함된 R2YE 액체배지에 접종하여 28℃에서 48 내지 72시간 120rpm으로 진탕배양하였다. 전배양 과정을 거친 다음 본배양을 실시하였으며, 각 배양시간은 2.5일로 하였다. 본배양을 통해 수득한 배양액을 원심분리하여 균체를 제거한 다음, 상층액을 한외여과막(5kDa cut-off membrane)으로 여과하여 여과막을 통과하지 못한 단백질을 분리 및 정제하였다. 수득한 농축액(효소액)은 동결건조하여 보관하면서 사용하였다.
실시예 2. DagA의 아가레즈 활성 측정
환원당량 검정방법(DNS method)으로 상기 실시예 1의 DagA에 대한 아가레즈 활성을 측정하였다. 상기 실시예 1에서 수득한 효소액 10㎕에 0.5%(w/v)로 아가로스를 녹인 20mM Tris-HCl 용액(pH 7) 0.5㎖를 섞어 40℃에서 15분간 반응시킨 후 DNS 시약(dinitrosalicylic acid 6.5g, 2M NaOH 325㎖, glycerol 45㎖ / 1ℓ 증류수) 0.5㎖를 넣고 10분간 끓인 다음 식혀 흡광도(Å540㎚)을 측정하였다. 효소 1U은 15분 반응 후에 흡광도(Å540㎚) 0.001을 생성하는 활성으로 정의하였다.
실시예 3. DagA 효소반응을 통한 한천 또는 아가로스의 분해
0.5 내지 5%(w/v) 한천 또는 아가로스를 녹인 20mM Tris-HCl 용액 1ℓ를 제조하여 100℃에서 약 10분간 가열하여 충분히 녹인 다음 40℃로 온도를 떨어뜨리고 DagA 효소 2,000 내지 50,000U을 처리하여 24시간 효소반응을 실시하였다.
효소반응산물에서 미분해된 아가로스를 제거하기 위해 원심분리하여 상등액을 회수한 다음 TLC(thin layer chromatography)를 이용하여 효소반응 생성물을 확인하였다. 회수한 상등액을 5KDa cut-off membrane으로 한외여과방법을 통해 부분 정제하였다.
실시예 4. HPLC-ELSD 분석을 통한 DagA 효소반응산물의 조성 확인
상기 실시예 3에서 수득한 효소반응산물의 조성을 확인하기 위하여 HPLC-ELSD 분석을 수행하였다. NH2 P-50 4E multimode column(250 x 4.6mm)을 사용하였으며 이동상으로 아세토니트릴(acetonitrile)과 물의 혼합용액(acetonitrile : water = 65 : 35)을 사용하였으며, 그 결과를 도 3에 나타내었다.
도 3에 나타낸 바와 같이 반응산물 중 네오아가로비오스(neoagarobiose, 이하 'DP2'라 한다), 네오아가로테트라오스(neoagarotetraose, 이하 'DP4'라 한다) 및 네오아가로헥사오스(neoagarohexaose, 이하 'DP6'라 한다)로 구성된 네오아가로올리고당(neoagarooligosaccharides)의 총량이 65±20%(중량)를 차지하는 것으로 나타났고, 이 네오아가로올리고당의 총량을 기준으로 DP2, DP4, DP6가 각각 0 ~ 10%, 50 ~ 70%, 30 ~ 50%(중량)를 차지하는 것으로 나타났다.
실험예 1. 네오아가로올리고당 투여에 의한 패혈증 예방 효과 분석
네오아가로올리고당 투여에 의한 패혈증의 예방 효과를 분석하기 위하여, 하기와 같은 실험을 수행하였다.
1-1. 실험 디자인 및 생존율 분석
실험 동물로는 6주령의 암컷 BALB/c 마우스를 이용하였다. 마우스의 복강으로 네오아가로올리고당을 복강주사하여 전처리한 후, 1시간 뒤 107 CFU/mice의 E. coli K1 (dissolved in LB broth)을 주사하여 패혈증을 유도하였다. 패혈증이 유도된 실험군 및 대조군 각 마우스의 시간에 따른 생존율을 도 4에 나타내었다.
도 4에 나타낸 바와 같이, 대조군은 패혈증 유도 18시간 후 모두 사망하였다. 반면, 네오아가로올리고당을 전처리한 군은 생존율이 대조군에 비해 현저하게 증가하는 것을 확인하였다.
1-2. 혈청 분석
패혈증 유도 12시간 후, 각 마우스로부터 혈청을 분리하여 혈청 내 염증성 사이토카인(TNF-α, IL-6, 및 IL-1β, IL-12p70, IL-10), AST, ALT, BUN 수치를 분석하였다. 이때, 사이토카인의 농도는 ELISA 키트(eBiosciences, San Diego, CA)를 이용하여 측정하였으며, 그 결과를 도 5 및 6에 나타내었다.
도 5에 나타낸 바와 같이, 패혈증에 의해 증가되는 마우스 혈청 내 전염증성 사이토카인인 TNF-α, IL-6, IL-1β 및 IL-12p70의 양이 네오아가로올리고당을 전처리한 경우 현저하게 억제함을 확인하였다. 반면 항염증성 사이토카인인 IL-10의 분비에는 네오아가로올리고당 전처리에 의해 증가함을 확인하였다.
또한, 도 6에 나타낸 바와 같이, 간 및 신장 독성과 관련된 혈청 내 AST, ALT 및 BUN 농도가 패혈증에 의해 증가하나 마우스 네오아가로올리고당을 전처리한 경우 현저하게 억제됨을 확인하였다.
실험예 2. 네오아가로올리고당 투여에 의한 면역 증강 효과 분석
네오아가로올리고당 투여에 의한 면역 증강 효과를 분석하기 위하여, 하기와 같은 실험을 수행하였다.
2-1. 실험 디자인 및 생존율 분석
실험동물로는 6주령의 암컷 BALB/c 마우스를 이용하였다. 마우스의 복강으로 네오아가로올리고당 100 mg/kg을 복강 주사하여 전처리한 후, 1시간 뒤 지질다당체ipopolysaccharide;LPS) 20 mg/kg를 주사하였다. 그 후 실험군 및 대조군 각 마우스의 시간에 따른 생존율을 도 7에 나타내었다.
도 7에 나타낸 바와 같이, 대조군은 LPS 투여 24시간 후 모두 사망하였다. 반면, 네오아가로올리고당을 전처리한 군은 생존율이 대조군에 비해 현저하게 증가하는 것을 확인하였으며, 네오아가로올리고당의 농도 의존적으로 증가함을 확인하였다.
2-2. 혈청 분석
LPS 투여 12시간 후, 각 마우스로부터 혈청을 분리하여 혈청 내 염증성 사이토카인(TNF-α, IL-6, 및 IL-1β, IL-12p70, IL-10), AST, ALT, BUN 수치를 분석하였다. 이때, 사이토카인의 농도는 ELISA 키트(eBiosciences, San Diego, CA)를 이용하여 측정하였으며, 그 결과를 도 8 및 9에 나타내었다.
도 8에 나타낸 바와 같이, LPS 투여에 증가되는 마우스 혈청 내 전염증성 사이토카인인 TNF-α, IL-6, IL-1β 및 IL-12p70의 양이 네오아가로올리고당을 전처리한 경우 현저하게 억제됨을 확인하였다. 반면 항염증성 사이토카인인 IL-10의 분비에는 네오아가로올리고당 투여에 의해 증가함을 확인하였다.
또한, 도 9에 나타낸 바와 같이, 간 및 신장 독성과 관련된 혈청 내 AST, ALT 및 BUN 농도가 패혈증에 의해 증가하나 마우스 네오아가로올리고당을 전처리한 경우 현저하게 억제됨을 확인하였다.
2-3. 조직 분석
LPS 투여 12시간 후, 각 마우스로부터 폐 및 비장 조직을 적출하고, 종래 공지된 방법에 따라 파라핀 섹션 후 H & E 염색을 수행하였으며, 현미경을 통해 관찰하였으며, 그 결과를 도 10 및 도 11에 나타내었다.
도 10에 나타낸 바와 같이, 네오아가로올리고당을 전처리한 경우, 대조군에 비해 폐 조직에서 염증 유발 세포의 침윤이 현저하게 억제됨을 확인하였다.
또한, 도 11에 나타낸 바와 같이, 네오아가로올리고당을 전처리한 경우, 대조군에 비해 비장 조직에서의 세포 사멸이 현저하게 억제됨을 확인하였다.
상기 실험 결과를 통하여, 네오아가로올리고당의 우수한 패혈증 및 패혈증성 쇼크의 예방 효과를 확인하였으며, 또한 염증발생을 효과적으로 억제함으로써 면역증진 효과가 탁월함을 확인하였다.
이하 본 발명의 약학적 조성물 및 식품 조성물의 제제예를 설명하나, 이는 본 발명을 한정하고자 함이 아닌 단지 구체적으로 설명하고자 함이다.
제제예 1. 약학적 조성물의 제조
1-1. 산제의 제조
네오아가로올리고당 20 mg
유당 100 mg
탈크 10 mg
상기의 성분을 혼합하고 기밀포에 충진하여 산제를 제조하였다.
1-2. 정제의 제조
네오아가로올리고당 10 mg
옥수수전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
상기의 성분을 혼합한 후, 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조하였다.
1-3. 캡슐제의 제조
네오아가로올리고당 10 mg
결정성 셀룰로오스 3 mg
락토오스 14.8 mg
마그네슘 스테아레이트 0.2 mg
상기의 성분을 혼합한 후, 통상의 캡슐제의 제조방법에 따라서 젤라틴 캡슐에 충전하여 캡슐제를 제조하였다.
1-4. 주사제의 제조
네오아가로올리고당 10 mg
만니톨 180 mg
주사용 멸균 증류수 2974 mg
Na2HPO4·2H2O 26 mg
통상의 주사제의 제조방법에 따라 1 앰플당 (2 ml) 상기의 성분 함량으로 제조한다.
1-5. 액제의 제조
네오아가로올리고당 10 mg
이성화당 10 g
만니톨 5 g
정제수 적량
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고 레몬향을 적량 가한 다음 상기의 성분을 혼합한 다음 정제수를 가하여 전체를 정제수를 가하여 전체 100 ml로 조절한 후 갈색병에 충진하여 멸균시켜 액제를 제조한다.
제제예 2. 식품 조성물의 제조
2-1. 건강식품의 제조
네오아가로올리고당 100 mg
비타민 혼합물 적량
비타민 A 아세테이트 70 g
비타민 E 1.0 mg
비타민 B1 0.13 mg
비타민 B2 0.15 mg
비타민 B6 0.5 mg
비타민 B12 0.2 g
비타민 C 10 mg
비오틴 10 g
니코틴산아미드 1.7 mg
엽산 50 g
판토텐산 칼슘 0.5 mg
무기질 혼합물 적량
황산제1철 1.75 mg
산화아연 0.82 mg
탄산마그네슘 25.3 mg
제1인산칼륨 15 mg
제2인산칼슘 55 mg
구연산칼륨 90 mg
탄산칼슘 100 mg
염화마그네슘 24.8 mg
상기의 비타민 및 미네랄 혼합물의 조성비는 비교적 건강식품에 적합한 성분을 바람직한 실시예로 혼합 조성하였지만, 그 배합비를 임의로 변형 실시하여도 무방하며, 통상의 건강식품 제조방법에 따라 상기의 성분을 혼합한 다음, 과립을 제조하고, 통상의 방법에 따라 건강식품 조성물 제조에 사용할 수 있다.
Claims (9)
- 한천(agar) 또는 아가로스(agarose)로부터 DagA 효소반응으로 제조한 네오아가로올리고당(neoagarooligosaccharide)을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 DagA 효소는 스트렙토마이세스 시리칼라(Streptomyces coelicolor) 유래인 것을 특징으로 하는, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 제2항에 있어서, 상기 DagA 효소는 서열번호 2의 31 내지 309번의 아미노산 서열로 표시되는 것을 특징으로 하는, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 제2항에 있어서, 상기 DagA 효소는 서열번호 1의 염기서열로 암호화되는 것을 특징으로 하는, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 제1항에 있어서, 상기 효소반응은 35 내지 45℃의 온도 및 pH 6 내지 8에서 이루어지는 것을 특징으로 하는, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 상기 네오아가로올리고당은 네오아가로비오스(neoagarobiose), 네오아가로테트라오스(neoagarotetraose) 및 네오아가로헥사오스(neoagarohexaose)로 구성된 군으로부터 선택된 1종 이상인 것을 특징으로 하는, 패혈증 또는 패혈증성 쇼크의 예방 또는 치료용 약학적 조성물.
- 한천 또는 아가로스로부터 DagA 효소반응으로 제조한 네오아가로올리고당을 유효성분으로 포함하는 패혈증 또는 패혈증성 쇼크의 예방 또는 개선용 식품 조성물.
- 한천 또는 아가로스로부터 DagA 효소반응으로 제조한 네오아가로올리고당을 유효성분으로 포함하는 면역 증강용 약학적 조성물.
- 한천 또는 아가로스로부터 DagA 효소반응으로 제조한 네오아가로올리고당을 유효성분으로 포함하는 면역 증강용 기능성 식품.
