본 출원은 근육 손실 저해 및 근육량 증진 활성을 갖는 펩타이드 및 이의 용도에 관한 것으로서, 서열번호 1의 아미노산 서열로 이루어지는 펩타이드, 및 상기 펩타이드를 유효 성분으로 포함하는 근육 질환의 예방 또는 치료용 약제학적 조성물, 및 상기 펩타이드를 유효 성분으로 포함하는 근육 질환의 예방 또는 개선용 식품 조성물을 제공한다.
Description
본 출원은 근육 손실 저해 및 근육량 증진 활성을 갖는 펩타이드 및 이의 용도에 관한 것이다. 본 출원은 2022년 6월 10일 출원된 대한민국 특허출원 제10-2022-0071033호를 우선권으로 주장하고, 상기 특허출원의 개시 사항은 본 명세서에 참조로서 삽입된다.
최근 고령 인구가 전세계적으로 급증하는 추세에 있는 가운데, 국제연합 (United Nations)의 보고에 따르면 2050년에는 60세 이상 인구가 20억 명이 넘을 것으로 예측되고 있다. 골격근은 전체 몸무게의 50%에 달하는 가장 큰 기관이며, 근육 감소와 근력 약화는 노화로 인해 일어나는 주요 신체 변화 중 하나이다. 이에, 골격근은 30세 이후가 되면 우리 몸에서 매년 약 1%씩 감소하는 경향을 보이다가 65세 이후부터는 급격하게 감소한다. 이러한 근육의 급격한 감소는 신체 능력을 저하시키고 낙상과 골절의 위험을 높이는 등 삶의 질에 큰 영향을 미칠 수 있으며, 이러한 우려를 반영하여, 2017년 세계보건기구 (WHO)는 정식 질병으로 근 감소증(sarcopenia)을 인정한 바 있다.
근육 질환과 연관된 척수신경, 운동신경 또는 골격근 섬유의 퇴행에 의해 유발되는 근감소증은 아직까지 발병 원인이 규명되지 않은 대표적인 난치성 질환 중 하나이다. 지금까지 연구된 바에 의하면, 골격근의 수축을 유도하는 운동 신경이 퇴행되어 골격근의 수축이 진행되지 않거나 또는 골격근 내에서 근육의 수축에 관여하는 단백질의 발현이 감소되거나 상기 단백질이 변형되어 정상적인 골격근의 수축이 진행되지 않으며, 장기적으로는 상기 운동신경 또는 골격근이 섬유성 조직으로 변형되는 것으로 알려져 있다. 이와 같은 근감소증의 근본적인 발병 원 인이 아직 규명되지 않았고, 운동 신경이나 골격근의 퇴행을 방지하거나 또는 회복시킬 수 있는 방법이 개발되지 않고 있기 때문에, 현재로서는 상기 근감소증의 진행을 둔화시키는 방법을 개발하기 위한 연구가 활발히 진행되고 있다. 현재 근감소증에는 운동, 단백질 및 칼로리 보충이 도움이 된다고 알려져 있으나, 근감소증 환자의 대부분을 차지하는 노인들에서는 크게 도움이 되지 않아 근감소증 치료제가 절실히 필요하다.
한편, 근육의 크기 또는 근육량은 근육 내에서 일어나는 동화작용이나 이화작용을 유도하는 세포 내 신호전달 과정에 의해 조절된다. 구체적으로, 근육 단백질의 분해에 비해 근육 단백질의 합성을 유도하는 신호전달 반응이 우세한 경우, 근 단백질 합성이 증가되는데, 이는 근육 크기 증가(hypertrophy)나 근섬유 수 증가(hyperplasia)로 나타난다. 또한, 근육 세포의 분화와 근육 형성은 다양한 근육 조절 인자(muscle regulatory factors)에 의해 조절된다. MyoD는 근육 특이 유전자의 발현을 개시하게 하고, 근육위성세포(muscle satellite cells)가 근원세포(myoblast)로 분화되는 것을 유도하며, MyoD의 활성에 의한 myogenin 발현의 유도는 근원세포의 융합(fusion)에 가장 중요한 요소로서, 근관세포(myotube)의 형성에 관여한다. 이와 같은 과정을 통해 형성된 근섬유는 다발을 이루어 최종적으로 근육을 형성하게 된다.
이러한 기술적 배경 하에서, 외부적 또는 내인적 요인으로부터 야기된 근육의 손실을 저해하고, 근육 단백질의 합성 촉진시키기 위한 다각적인 연구가 진행되고 있으나(한국 등록특허 제 10-2064387 호), 아직은 미비한 실정이다.
일 양상은 서열번호 1의 아미노산 서열로 이루어지는 펩타이드를 제공하는 것이다.
다른 양상은 서열번호 1의 아미노산 서열로 이루어지는 펩타이드를 유효 성분으로 포함하는 근육 손실 저해 및 근육량 증진용 조성물을 제공하는 것이다.

한편, 종래의 기능성 펩타이드는 유효한 생물학적 활성에도 불구하고 펩타이드 자체의 크기로 인해, 표적 조직 또는 세포에 효과적으로 유입되지 못하거나 반감기가 짧아 단기간에 체내에서 소멸되는 단점을 보여 주었다. 반면, 일 실시예에 따른 약제학적 조성물은 약 10개 이하의 아미노산으로 이루어지는 펩타이드를 유효 성분으로 포함하며, 이에 따라, 유효 성분의 세포 침투율 등이 매우 우수하고, 예를 들어, 국소적으로 투여하는 경우, 유효한 근육 질환의 치료 효과를 얻을 수 있다.
일 실시예에 따르면, 본원의 펩타이드는 근원세포의 분화 초기 마커(예를 들어, MyoG, 및 Myf5)와, 분화 후기 마커 또는 근 섬유 구성 단백질(예를 들어, Myosin heavy chain, α-actinin)의 발현을 유의적으로 증가시켜, 근육 세포 또는 근육 섬유로의 분화를 촉진시킬 수 있다. 또한, 본원의 펩타이드는 외인성 요인에 의한 근육 손상 시, 근육량 회복/재생을 향상시킬 뿐만 아니라, 손상된 근육 조직 내 면역세포의 침윤과 콜라겐 조직의 축적을 감소시켜, 근육 질환의 증상을 완화하고 질병의 진행을 저해시킬 수 있는 바, 상기 펩타이드는 근육 질환 치료를 위한 약제학적 조성물의 유효 성분으로 활용될 수 있다.
상기 약제학적 조성물은 상기 펩타이드의 약제학적 유효량(pharmaceutically effective amount); 및/또는 약제학적으로 허용되는 담체를 포함하는 것일 수 있으나, 이에 제한되는 것은 아니다.
본 명세서에서 사용되는 용어 "약제학적 유효량"은 상기 약제학적 조성물의 골 질환의 치료 효능을 달성하는 데 충분한 양을 의미할 수 있다.

상기 약제학적으로 허용되는 담체는 제제 제조 시에 통상적으로 이용되는 것으로서, 락토스, 덱스트로스, 수크로스, 솔비톨, 만니톨, 전분, 아카시아 고무, 인산 칼슘, 알기네이트, 젤라틴, 규산 칼슘, 미세결정성 셀룰로스, 폴리비닐피롤리돈, 셀룰로스, 물, 시럽, 메틸 셀룰로스, 메틸히드록시벤조에이트, 프로필히드록시벤조에이트, 활석, 스테아르산 마그네슘 및 미네랄 오일 등을 포함하나, 이에 한정되는 것은 아니다. 적합한 약제학적으로 허용되는 담체 및 제제는 Remington's Pharmaceutical Sciences (19th ed., 1995)에 상세히 기재되어 있다.
상기 약제학적 조성물은 상기 성분들 이외에 윤활제, 습윤제, 감미제, 향미제, 유화제, 현탁제, 보존제 등을 추가로 포함할 수 있으나, 이에 제한되는 것은 아니다.
상기 약제학적 조성물은 경구 또는 비경구로 투여할 수 있고, 비경구 투여인 경우에는 근육 주입, 정맥 내 주입, 피하 주입, 복강 주입, 국소 투여, 경피 투여 등으로 투여할 수 있으나, 이에 한정되는 것은 아니다.
상기 약제학적 조성물의 투여량은 1 일당 0.0001 내지 1000 ug(마이크로그램, 0.001 내지 1000 ug, 0.01 내지 1000 ug, 0.1 내지 1000 ug, 또는 1.0 내지 1000 ug일 수 있으나, 이에 제한되는 것은 아니며, 제제화 방법, 투여 방식, 환자의 연령, 체중, 성, 병적 상태, 음식, 투여 시간, 투여 경로, 배설 속도 및 반응 감응성과 같은 요인들에 의해 다양하게 투여될 수 있다.
상기 약제학적 조성물은 당해 발명이 속하는 기술분야에서 통상의 지식을 가진 자가 용이하게 실시할 수 있는 방법에 따라, 약제학적으로 허용되는 담체 및/또는 부형제를 이용하여 제제화함으로써 단위 용량 형태로 제조되거나 또는 다용량 용기 내에 내입시켜 제조될 수 있다.
상기 제형은 오일 또는 수성 매질 중의 용액, 현탁액 또는 유화액 형태이거나, 연고, 크림, 겔, 경피흡수제, 카타플라스마제, 첩부제, 페이스트제, 엑스제, 분말제, 과립제, 정제 또는 캅셀제 형태일 수도 있으며, 분산제 및/또는 안정화제를 추가적으로 포함할 수 있다.

상기 식품 조성물에는 건강기능식품이 포함될 수 있다. 본 명세서에서 용어, "건강기능식품"은 인체에 유용한 기능성을 가진 원료나 성분을 사용하여 정제, 캅셀, 분말, 과립, 액상 및 환 등의 형태로 제조 및 가공한 식품을 말한다. 여기서 '기능성'이라 함은 인체의 구조 및 기능에 대하여 영양소를 조절하거나 생리학적 작용 등과 같은 보건 용도에 유용한 효과를 얻는 것을 의미한다. 상기 건강기능식품은 통상의 기술분야에서 통상적으로 사용되는 방법에 의하여 제조 가능하며, 상기 제조시에는 통상의 기술분야에서 통상적으로 첨가하는 원료 및 성분을 첨가하여 제조할 수 있다. 또한 상기 건강기능식품의 제형 또한 건강기능식품으로 인정되는 제형이면 제한없이 제조될 수 있다. 상기 식품 조성물은 다양한 형태의 제형으로 제조될 수 있으며, 일반 약품과는 달리 식품을 원료로 하여 약품의 장기 복용시 발생할 수 있는 부작용 등이 없는 장점이 있고, 휴대성이 뛰어나, 일 실시예에 따른 건강기능식품은 근육 질환의 치료 효과를 증진시키기 위한 보조제로 섭취가 가능하다.
또한, 일 실시예에 따른 조성물이 사용/적용될 수 있는 건강 식품의 종류에는 제한이 없다. 상기 펩타이드를 유효 성분으로 포함하는 식품 조성물은 당업자의 선택에 따라 건강기능식품에 함유될 수 있는 적절한 기타 보조 성분과 공지의 첨가제를 혼합하여 제조할 수 있다. 첨가할 수 있는 식품의 예로는 육류, 소세지, 빵, 쵸코렛, 캔디류, 스낵류, 과자류, 피자, 라면, 기타 면류, 껌류, 아이스크림류를 포함한 낙농 제품, 각종 스프, 음료수, 차, 드링크제, 알콜 음료 및 비타민 복합제 등이 있으며, 이 밖에도 즙, 차, 젤리 및 주스 등에 첨가하여 제조할 수 있다.

이하 본 발명을 실시예를 통하여 보다 상세하게 설명한다. 그러나, 이들 실시예는 본 발명을 예시적으로 설명하기 위한 것으로 본 발명의 범위가 이들 실시예에 한정되는 것은 아니다.
실시예 1. 펩타이드의 합성
자동 펩타이드 합성기(Milligen 9050, Millipore, 미국)를 이용하여 하기의 [표 1]에 기재된 서열번호 1의 아미노산 서열을 갖는 펩타이드를 합성하고, C18 역상 고성능액체크로마토그래피(HPLC)(Waters Associates, 미국)를 이용하여 이들 합성된 펩타이드를 순수 분리하였다. 컬럼은 ACQUITY UPLC BEH300 C18 (2.1㎜ Х 100㎜, 1.7㎛, Waters Co, 미국)을 이용하였다.

실시예 2. 초기 분화 단계에서 근원세포의 분화 촉진 효과 확인
본 실시예에서는 마우스 근원세포인 C2C12 세포의 초기 분화 마커 단백질의 발현 변화를 평가함으로써, 초기 분화 단계에서 근원세포로부터 근육 세포/근육 섬유로의 분화에 일 실시예에 따른 펩타이드가 미치는 영향을 확인하고자 하였다.
2-1. 단백질 발현 분석을 통한 평가
C2C12 세포를 1% P/S, 및 2% BCS를 포함한 DMEM 배지에서 배양한 후, cell confluency가 70-80% 가량 되었을 때, 이를 6-웰 배양 플레이트에 seeding 하였다. 이후, 6-웰 배양 플레이트 내 세포의 cell confluency가 100%가 되었을 때, 배양 배지를 분화 배지 (1% P/S, 2% Horse serum)로 교체하면서, 여기에 5, 50, 또는 100ug/ml의 서열번호 1의 펩타이드를 첨가하였다. 이후, 2일에 한번씩 분화 배지와 동량의 서열번호 1의 펩타이드를 교체 및 첨가하였다. 배지 교체를 처음으로 수행한 날로부터 3일 경과 후, 배양물에 용해 버퍼를 첨가하여, 배양물을 용해시킨 후 4℃, 12,000 rpm에서 30분간 원심분리하여 단백질을 수득하고, 이를 BCA 키트를 사용하여 정량하였다. 상기 단백질을 포함하는 샘플을 대상으로 SDS-PAGE를 실시하여 각각의 단백질을 분리한 후, 이를 membrane에 electrotransfer하였다. 상기 단백질이 부착된 membrane을 5% skim milk로 처리하여 blocking한 후, 1차 항체를 4℃에서 overnight 반응시켰다. PBS-T 로 세척한 다음 2차 항체를 상온에서 1시간 반응시키고 다시 PBS-T로 세척한 후, 이를 Gel Doc (Bio-Rad, Hercules, CA, USA), 및 Western 검출 시약 (Elpis Biotech, Daejeon, Korea)을 통해 검출하였다. 한편, 1차 항체로는 Anti-MyoG (santa cruz, USA), 및 Anti-Myf5 (Abcam, UK), 및 Anti-GAPDH (santa cruz, USA)를 사용하였으며, 대조군으로 비처리군 (Non), 비교군으로 분화 배지만을 처리한 군 (CM)을 사용하였다.
그 결과, 도 1 및 도 2에 나타내 바와 같이, 서열번호 1의 펩타이드 처리에 의해, 근원세포의 분화 초기 마커인 Myogenin(MyoG) 및 Myf5의 발현이 증가됨을 확인하였다.
2-2. 면역형광법을 통한 평가
C2C12 세포를 1% P/S, 및 2% BCS를 포함한 DMEM 배지에서 배양한 후, cell confluency가 70-80% 가량 되었을 때, 이를 6-웰 배양 플레이트에 seeding하였다. 이후, 6-웰 배양 플레이트 내 세포의 cell confluency가 100%가 되었을 때, 배양 배지를 분화 배지 (1% P/S, 2% Horse serum)로 교체하면서, 여기에 100ug/ml의 서열번호 1의 펩타이드를 첨가하였다. 이후, 2일에 한번씩 분화 배지와 100ug/ml의 서열번호 1의 펩타이드를 교체 및 첨가하였다. 배지 교체를 처음으로 수행한 날로부터 3일 경과 후, 상기 배양된 세포를 실온에서 10분 동안 4% 파라포름알데히드 (PFA)로 고정하고 0.1% Triton X-100에서 5분 동안 투과 과정 (permeabilization)을 실시하였다. 이후, 상기 고정된 세포에 PBS, 10% FBS 및 0.1% Triton X-100을 첨가하여 실온에서 1시간 동안 blocking한 후, 1차 항체인(MyoG, santa cruz, USA)을 처리하고, 이를 실온에서 1시간 동안 인큐베이션하였다. 이후, 상기 세포를 세척하고, 다시 2차 항체(goat anti-mouse IgG-TR, Santa cruz, USA)와 함께 실온에서 30분 동안 인큐베이션하였다. 이후, 세포의 핵을 DAPI(Santa cruz, USA)로 염색하고, 이를 LEICA 형광 현미경(TCS SP8, LEICA, Germany)으로 관찰하였다.
그 결과, 도 3 및 도 4에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, 근원세포의 분화 초기 마커인 Myogenin(MyoG)의 발현이 증가되었을 뿐만 아니라, 근원세포의 핵 영역에서의 발현이 증가됨을 확인하였다.
상기 실험 결과를 종합하면, 일 실시예에 따른 펩타이드는 근원세포로부터 근육 세포로의 초기 분화를 촉진시키는데 기여함을 알 수 있었다.
실시예 3. 후기 분화 단계에서 근원세포의 분화 촉진 효과 확인
본 실시예에서는 마우스 근원세포인 C2C12 세포의 초기 분화 마커 단백질의 발현 변화를 평가함으로써, 후기 분화 단계에서 근원세포로부터 근육 세포/근육 섬유로의 분화에 일 실시예에 따른 펩타이드가 미치는 영향을 확인하고자 하였다.
3-1. 단백질 발현 분석을 통한 평가
용해 버퍼 첨가 시점 (배지 교체를 처음으로 수행한 날로부터 7일 경과 시점)을 제외하고, 상기 실시예 2-1과 동일한 방식으로 수행하여, 근원세포의 후기 분화 마커인 Myosin heavy chain(MyHC) 단백질의 발현 수준을 확인하였다. 한편, 1차 항체로 Anti-MyHC (santa cruz, USA), 및 Anti-GAPDH (santa cruz, USA)를 사용하였으며, 대조군으로 비처리군 (Non), 비교군으로 분화 배지만을 처리한 군 (CM)을 사용하였다.
그 결과, 도 5 및 도 6에 나타내 바와 같이, 서열번호 1의 펩타이드 처리에 의해, 근원세포의 분화 후기 마커인 Myosin heavy chain (MyHC)의 발현이 증가됨을 확인하였다.
3-2. 면역형광법을 통한 평가
고정화 시점 (배지 교체를 처음으로 수행한 날로부터 7일 경과 시점)을 제외하고, 상기 실시예 3-1과 동일한 방식으로 수행하여, 근원세포의 후기 분화 마커인 Myosin heavy chain(MyHC) 단백질의 발현 수준을 확인하였다. 한편, 1차 항체로는 Anti-α-actinin (santa cruz, USA), 2차 항체로는 goat anti-mouse IgG-TR(Santa cruz, USA)를 사용하였으며, 비교군으로 분화 배지만을 처리한 군 (CM)을 사용하였다.
그 결과, 도 7 및 도 8에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, 근원세포의 분화 후기 마커인 α-actinin 의 발현이 증가됨을 확인하였다.
상기 실험 결과를 종합하면, 일 실시예에 따른 펩타이드는 근원세포로부터 근 세포로의 후기 분화를 촉진시키는데 기여함을 알 수 있었다.
실시예 4. 동물 모델을 이용한 근육 손실 저해 효과 확인
본 실시예에서는 BaCl2에 의해 근육 손상이 유도된 동물 모델을 대상으로, 근육량 변화, 근육 조직 내 면역세포의 침윤 영역과 축적된 콜라겐의 수준, 및 근 섬유 구성 단백질 및 염증성 단백질의 발현 수준을 평가함으로써, 근육 손실 저해에 일 실시예에 따른 펩타이드가 미치는 영향을 확인하고자 하였다.
4-1. 근육량 평가
8주령 수컷 C57BL/6 마우스의 TA (Tibialis anterior) 근육에 1.2%(w/v) 50μl의 BaCl2 수용액을 주사하여, 1일간 마우스의 근육 손상을 유도하였다. 이후, 상기 근육 손상이 유도된 마우스에 20mg의 서열번호 1의 펩타이드를 0.1 mg/μl의 농도로 1일 간격으로 총 6회 경구 투여하였다. 양성 대조군에 대해서는 0.5ug의 IGF-I를 0.01ug/μl의 농도로 근육 주사하였다. 근육 손상을 유도한 날로부터 7일째, 마우스를 희생시키고, TA 근육의 변화를 육안으로 확인하고, 이의 중량을 측정하였다. 대조군으로는 비처리군 (Non), 음성 대조군으로는 BaCl2에 의한 근육 손상만 유도된 군 (N.C), 양성 대조군으로는 BaCl2에 의한 근육 손상이 유도된 군에 IGF-I를 처리한 군 (P.C)를 사용하였다.
그 결과, 도 9 및 도 10에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, BaCl2에 의한 근육 손상이 유도된 마우스의 TA 근육의 근육량이 증가됨을 확인하였으며, 이로부터 손상된 근육의 재생을 촉진시킬 수 있음을 알 수 있었다.
4-2. 근육 조직 염색을 통한 평가
상기 실시예 4-1의 근육 손상이 유도된 마우스로부터 수득한 근육 조직을 실온에서 PBS로 세척한 후, 4% 파라포름알데히드(PFA)로 고정하였다. 이후, 고정화된 근육 조직을 PBS로 3회 세척하였으며, 이를 gradient ethanol series(from 70% to 100%)으로 탈수(dehydration)시켰다. 상기 근육 조직 샘플을 4μm의 크기로 절편화하였다. 상기 절편화된 표본을 포함하는 조직 슬라이드는 gradient ethanol series를 통해 dewaxing 및 hydration 되었으며, 순차적으로 hematoxlin 용액에서 1분 Eosin 용액에서 10초 동안 염색되었다. 염색을 마친 후, 조직 슬라이드를 순차적으로 90%, 100% ethyl alcohol에 침지시키고, 마지막으로 xylene에 5분간, 2회 침지시킨 후, 조직 슬라이드에 대한 mounting을 진행하였다(H&E 염색).
또한, 상기 절편화된 표본을 포함하는 조직 슬라이드를 전술한 바와 같이 dewaxing 및 hydration시킨 후, Picro Sirius Red Solution 에서 60분 염색한 후 1% acetic acid solution에 2분간 2회 침지시키고, 이를 D.W로 세척하였다. 이후, 조직 슬라이드를 ethyl alcohol에 침지시키고, 마지막으로 xylene에 5분간, 2회 침지시킨 후, 조직 슬라이드에 대한 mounting을 진행하였다 (Sirius red 염색).
그 결과, 도 11 및 도 12에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, BaCl2에 의한 근육 손상이 유도된 조직 내 면역세포의 침윤 영역이 감소됨을 확인하였다. 또한, 도 13 및 도 14에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, BaCl2에 의한 근육 손상이 유도된 조직 내 콜라겐의 축적이 감소되었음을 확인하였다.
4-3. 근육 조직 내 단백질 발현 분석을 통한 평가
상기 실시예 4-1의 근육 손상이 유도된 마우스로부터 수득한 근육 조직을 homogenizer를 이용하여 파쇄하고, 여기에 용해 버퍼를 첨가하여 파쇄물을 용해시킨 후 4℃, 12,000 rpm에서 30분간 원심분리하여 단백질을 수득하고, 이를 BCA 키트를 사용하여 정량하였다. 상기 단백질을 포함하는 샘플을 대상으로 SDS-PAGE를 실시하여 각각의 단백질을 분리한 후, 이를 membrane에 electrotransfer하였다. 상기 단백질이 부착된 membrane을 5% skim milk로 처리하여 blocking한 후, 1차 항체를 4℃에서 overnight 반응시켰다. PBS-T 로 세척한 다음 2차 항체를 상온에서 1시간 반응시키고 다시 PBS-T로 세척한 후, 이를 Gel Doc (Bio-Rad, Hercules, CA, USA), 및 Western 검출 시약(Elpis Biotech, Daejeon, Korea)을 통해 검출하였다. 한편, 1차 항체로는 Anti-MyHC(santa cruz, USA), Anti-α-actinin(santa cruz, USA), Anti-α-SMA(Abcam, UK), Anti-IL-6(santa cruz, USA), Anti-α-tubulin(santa cruz, USA)를 사용하였다.
그 결과, 도 15 및 도 16에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, BaCl2에 의한 근육 손상이 유도된 조직 내 근 섬유 구성 단백질인 MyHC, 및 α-actinin의 발현이 증가됨을 확인하였다. 또한, 도 17 및 도 18에 나타낸 바와 같이, 서열번호 1의 펩타이드 처리에 의해, BaCl2에 의한 근육 손상이 유도된 조직 내 염증성 단백질인 α-SMA, 및 IL-6의 발현이 감소됨을 확인하였다.
상기 실험 결과를 종합하면, 일 실시예에 따른 펩타이드는 손상된 근육의 재생 또는 회복시킬 뿐만 아니라, 근육 손상을 야기하는 염증 반응 등과 같이, 근육 손상의 진행을 억제하는데 기여할 수 있음을 알 수 있었다.
제형예 1. 펩타이드 나노좀의 제조
상기 실시예 1의 펩타이드 50 mg을 증류수 500 ml로 충분히 교반하여 용해하였다. 배합체 용액을 레시틴 5 g, 소듐 올리에이트(sodium oleate) 0.3 ml, 에탄올 50 ml 그리고 약간의 유상과 함께 혼합한 후, 총량이 1 L가 되도록 증류수로 양을 맞춰 준 다음, 마이크로플루다이저로 고압을 이용하여 유화시켜 사이즈 100 nm 정도의 펩타이드 나노좀을 제조하였다.
제형예 2. 약학적 제제
2-1. 산제의 제조
하기의 성분들을 혼합하고 기밀포에 충진하여 산제를 제조한다.
본 발명의 펩타이드 20 mg
유당 100 mg
탈크 10 mg
2-2. 정제의 제조
하기의 성분들을 혼합한 후 통상의 정제의 제조방법에 따라서 타정하여 정제를 제조한다.
본 발명의 펩타이드 10 mg
옥수수전분 100 mg
유당 100 mg
스테아린산 마그네슘 2 mg
2-3. 캡슐제의 제조
통상의 캡슐제 제조방법에 따라 하기의 성분을 혼합하고 젤라틴 캡슐에 충전하여 캡슐제를 제조한다.
본 발명의 펩타이드 10 mg
결정성 셀룰로오스 3 mg
락토오스 14.8 mg
마그네슘 스테아레이트 0.2 mg
2-4. 주사제의 제조
통상의 주사제의 제조방법에 따라 1 앰플당 (2 ml) 하기의 성분 함량으로 제조한다.
본 발명의 펩타이드 10 mg
만니톨 180 mg
주사용 멸균 증류수 2974 mg
Na2HPO4·2H2O 26 mg
2-5. 액제의 제조
통상의 액제의 제조방법에 따라 정제수에 각각의 성분을 가하여 용해시키고, 하기의 성분을 혼합한 다음 정제수를 가하여 전체를 100 ml로 조절한 후, 갈색병에 충진하여 멸균시켜 액제를 제조한다.

만니톨 5 g
정제수 적량
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
Claims (10)
- 서열번호 1의 아미노산 서열로 이루어지는, 펩타이드.
- 청구항 1에 있어서, 상기 펩타이드의 N-말단은 아세틸기(acetyl group), 플루오레닐메톡시카르보닐기(fluoreonylmethoxycarbonyl group), 포르밀기(formyl group), 팔미토일기(palmitoyl group), 미리스틸기(myristyl group), 스테아릴기(stearyl group), 부톡시카르보닐기(butoxycarbonyl group), 아릴옥시카르보닐기(allyloxycarbonyl group) 및 폴리에틸렌글리콜(polyethylene glycol; PEG)로 이루어지는 군으로부터 선택되는 어느 하나의 보호기와 결합된 것인, 펩타이드.
- 청구항 1에 있어서, 상기 펩타이드의 C-말단은 아미노기(amino group, -NH2), 삼차 알킬기(tertiary alkyl group) 및 아자이드(azide, -NHNH2)로 이루어지는 군으로부터 선택되는 어느 하나의 보호기와 결합된 것인, 펩타이드.
- 청구항 1에 있어서, 상기 펩타이드는 하기와 같은 특성으로부터 선택되는 어느 하나 이상을 나타내는 것인, 펩타이드:(a) Myogenin 및 Myogenic factor 5의 발현 증진;(b) Myosin heavy chain, 및 α-actinin의 발현 증진; 및(c) α-Smooth muscle actin, 및 interleukin-6의 발현 감소.
- 청구항 1 내지 4 중 어느 한 항의 펩타이드를 유효 성분으로 포함하는 근육 질환의 예방 또는 치료용 약제학적 조성물.
- 청구항 5에 있어서, 상기 근육 질환은 근 기능 저하, 근육 소모 또는 근육 퇴화로 인한 근육 질환인 것인, 약제학적 조성물.
- 청구항 6에 있어서, 상기 근육 질환은 근감소증(Sarcopenia), 긴장감퇴증(Atony), 근이영양증(Muscular dystrophy), 근위축증(Muscular atrophy), 근육 퇴화, 근경직증, 근위축성 축삭경화증, 근무력증 및 악액질(Cachexia) 중 어느 하나인 것인, 약제학적 조성물.
- 청구항 1 내지 4 중 어느 한 항의 펩타이드를 유효 성분으로 포함하는 근육 질환의 예방 또는 개선용 식품 조성물.
- 청구항 8에 있어서, 상기 근육 질환은 근 기능 저하, 근육 소모 또는 근육 퇴화로 인한 근육 질환인 것인 식품 조성물.
- 청구항 8에 있어서, 상기 식품 조성물은 비타민 C, 비타민 B1, 비타민 B2, 비타민 B6, 비타민 H, 비타민 PP, 프로-비타민 B5, 비타민 A, 비타민 D, 비타민 E, 비타민 K1 또는 카로틴 또는 이들의 혼합물로부터 선택된 어느 하나의 성분을 추가로 포함하는, 식품 조성물.
