보툴리눔 유래 펩타이드를 포함하는 통증개선용 조성물에 관한 것으로, 본 발명에 따른 보툴리눔 독소 재조합 단백질을 포함하는 조성물은 근육 마비 효과를 가지는 보툴리눔 독소 경쇄를 통증 부위에 빠르게 전달함으로써 효과적으로 근육 긴장을 완화시켜 통증을 감소시킬 수 있으므로 통증 예방, 개선, 또는 치료 물질로 유용하게 이용될 수 있다.
Description
본 발명은 보툴리눔 유래 펩타이드를 포함하는 통증개선용 조성물에 관한 것이다.
본 발명은 2021년 10월 25일에 출원된 대한민국 특허출원 제10-2021-0143024호 및 2022년 5월 25일에 출원된 대한민국 특허출원 제10-2022-0064311호에 기초한 우선권을 주장하며, 상기 출원들의 명세서 및 도면에 개시된 모든 내용은 본 출원에 원용된다.
일반적으로 통증은 위험에서 신체를 보호하고 손상받은 조직의 회복에 중요한 역할을 하는 것으로 알려져 있다. 그러나 바이러스나 박테리아의 감염, 말초신경 주변의 심각한 염증 반응, 또는 말초신경의 직접적인 손상 등에 의해서 발생하는 병증성 통증은 인체에 유해하며, 환자의 생활의 질에 지대한 영향을 미친다.
인류의 역사를 통틀어 오래 전부터 통증을 완화하거나 무감각하게 하기 위한 진통제가 개발되어 이용되어 왔는데, 이러한 진통제는 구강 투여가 일반적이다. 그러나, 구강 투여는 간손상, 신장손상 등의 문제점을 유발하는 경향이 있으며, 혈류에 진입하기 전에 소화계를 통하여 흡수되어야 하기 때문에, 효능을 발휘하는데 시간 지체가 발생한다.
이러한 구강 투여의 문제점을 해결하기 위해, 비경구 투여방법, 예를 들어, 혈관 내 주입 또는 근육 내 주입 방식들이 제안되었으나, 이들 방법은 편리하지 않고 비용이 많이 소요되는 문제점이 있으며, 병원 외에서는 실시하기가 매우 불편한 문제점이 있다.
따라서, 국소(topical) 투여, 특히, 경피(transdermal) 투여 방식으로 투여하려는 시도들이 이루어지고 있다.
한편, 보툴리눔 독소는 클로스트리디움 보툴리늄(Clostridium botulinum)에 의해 생성되는 신경독소 단백질이며, 신경세포의 신경전달물질인 아세트콜린 및 카테콜아민의 분비를 억제하는 것으로 보고 되고 있다. 보툴리늄 독소는 A, B, C, D, E, F, G 및 H 타입의 8종류가 있으며, A 및 B 타입이 상업적으로 이용된다.
보툴리눔 독소는 신경독 단백질로 인간의 반수 치사량은 정맥주사나 근육 주사 시 1.3 ∼ng/kg, 흡입할 경우 10 ∼ng/kg 로 독성이 매우 높은 물질이나, 적정한 양으로 조절 시 신경장애, 근육 질환, 다한증, 사각턱 등을 치료하는 약물로 사용될 수 있고, 주름, 종아리 근육 축소 등 미용 용도로 가장 많이 사용되고 있다.
이러한 보툴리눔 독소는 경쇄와 중쇄를 합쳐 150 kDa으로 매우 큰 분자이기 때문에 피부를 투과하기 어려워 주사제를 통해서만 활용되고 있으며, 독소 지속 시간은 3~6개월 이내로 정기적인 처치가 필요하므로, 이용 대상자에 대한 편의성을 제공할 수 있는 다른 효과적인 전달 수단을 찾기 위해 많은 연구가 이루어지고 있으나, 아직 미비한 실정이다.
본 발명자들은 본 발명에 따른 보툴리눔 독소 재조합 단백질(피부 투과형 보툴리눔 유래 성분 펩타이드)의 통증 개선 효과를 확인한 결과, 두통, 만성 통증 등의 통증 환자에 대하여 우수한 통증 감소 효과를 나타냄을 확인하여 본 발명을 완성하였다.
따라서, 본 발명의 목적은 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 약학적 조성물로서, 상기 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄의 일측 말단 또는 양측 말단에 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드가 융합된 것을 특징으로 하는, 통증개선용 약학적 조성물을 제공하는 것이다.
본 발명의 다른 목적은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 의약외품 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 피부외용제 조성물을 제공하는 것이다.
본 발명의 또 다른 목적은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 화장료 조성물을 제공하는 것이다.
그러나, 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 본 발명이 속하는 기술 분야의 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.
상기의 목적을 달성하기 위하여, 본 발명은 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 약학적 조성물로서, 상기 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄의 일측 말단 또는 양측 말단에 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드가 융합된 것을 특징으로 하는, 통증개선용 약학적 조성물을 제공한다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 의약외품 조성물을 제공한다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 피부외용제 조성물을 제공한다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 화장료 조성물을 제공한다.
본 발명의 일 구현예로, 상기 보툴리눔 독소 재조합 단백질은 서열번호 31 내지 서열번호 58로 이루어진 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어질 수 있으나, 이에 제한되지 않는다.
본 발명의 다른 구현예로, 상기 보툴리눔 독소 경쇄는 서열번호 3 내지 서열번호 9로 이루어진 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어질 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 보툴리눔 독소 경쇄는 일측 말단에 헥사히스티딘(hexahistidine) 태그(tag)를 더 포함할 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 상기 보툴리눔 독소 경쇄는 보툴리눔 독소 혈청형(serotype) A, B, C, D, E, F 및 G로 이루어진 군으로부터 선택될 수 있으나, 이에 제한되지 않는다.

본 발명의 또 다른 구현예로, 상기 통증은 기계적 통증, 열 통증, 만성 통증, 급성 통증, 통각과민 통증, 체인성 통증, 신경병성 통증, 심인성 통증, 열 유도된 통증, 물리적 통증, 통각수용, 통각과민, 류마티스성 통증, 두통, 요통, 골반통, 근막통, 혈관성 통증, 편두통, 상처 연계된 통증, 염증성 통증, 관절염 통증, 당뇨병성 통증, 암 유래의 통증, 신체 내장 통증, 섬유근통, 수술 후 통증, 환상통, 삼차신경통, 포진 후 신경통, 통증이 있는 당뇨병성 신경병증, 통증이 있는 당뇨병성 말초 신경병증, 당뇨병성 다발성 신경증, 좌골 통증, 신경근병증, 척수 신경근통, 근육통, 및 요추 통증으로 이루어진 군으로부터 선택될 수 있으나, 이에 제한되지 않는다.
본 발명의 또 다른 구현예로, 본 발명에 따른 조성물은 경피 투여용일 수 있으나, 이에 제한되지 않는다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 조성물을 이를 필요로 하는 개체에 투여하는 단계를 포함하는 통증개선 방법을 제공한다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 조성물의 통증개선 용도를 제공한다.
또한, 본 발명은 상기 보툴리눔 독소 재조합 단백질의 통증개선용 약제 제조를 위한 용도를 제공한다.
본 발명에 따른 보툴리눔 독소 재조합 단백질은 세포투과성 펩타이드와의 융합을 통해 근육 마비 효과를 가지는 보툴리눔 독소 경쇄를 경피로 전달할 수 있는바, 간편하게 보툴리눔 독소 경쇄를 통증 부위에 빠르게 전달함으로써 효과적으로 근육 긴장을 완화시켜 통증을 감소시킬 수 있으므로 통증 예방, 개선, 또는 치료 물질로 유용하게 이용될 수 있다.

본 발명자들은 보툴리눔 독소 경쇄에 세포투과성 펩타이드를 융합시킨 보툴리눔 독소 재조합 단백질(피부 투과형 보툴리눔 유래 성분 펩타이드)의 통증 개선 효과를 확인한 결과, 두통, 만성 통증 등의 통증 환자에 대하여 우수한 통증 감소 효과를 나타냄을 확인하여 본 발명을 완성하였다.
이하, 본 발명을 상세히 설명한다.
보툴리눔 독소는 하나의 폴리펩타이드로 발현이 되지만 발현 후 재구성 과정에 의해 약 100 kDa의 중쇄(Heavy chain; H 사슬)와 약 50 kDa의 경쇄(Light chain; L 사슬)로 나뉘어 지게 되며 H 사슬과 L 사슬은 디설파이드 결합으로 연결되어 있다. H 사슬은 신경세포의 수용체와 결합하여 내포작용(endocytosis)을 통해 보툴리눔 독소가 내부로 들어갈 수 있도록 한다. 보툴리눔 독소의 L 사슬은 세포 안으로 들어간 후 엔도좀을 빠져나와 세포질로 들어가고, 세포질 내의 스네어 단백질(SNARE protein)을 절단하여 아세틸콜린 분비를 억제시켜 근육 마비 효과를 나타낸다. 따라서, 신경세포의 아세틸콜린 분비 억제는 L 사슬 단독으로도 가능하며 H 사슬과 L 사슬은 각각 독립적으로 기능할 수 있다.
그러나, 분리된 분자량 50 kDa의 보툴리눔 독소 경쇄는 세포막을 투과할 수 없어 그 자체로는 기능을 할 수 없다. 일반적으로 보툴리눔 독소 경쇄가 신경세포 내 세포질로 전달되어 보툴리눔 독소 특유의 활성을 나타내기 위해서는 약 100 kDa의 보툴리눔 독소 중쇄의 도움이 반드시 필요하다.
이에, 본 발명의 일 실시예에서는, 보툴리눔 독소 경쇄를 피부 내부 및 신경세포에 효율적으로 전달할 수 있도록 보툴리눔 독소 경쇄와 세포투과성 펩타이드(MTD; Macromolecule Transduction Domain)가 융합된 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 조성물을 두통, 만성통증, 근육통, 섬유근통, 또는 근막통이 있는 환자의 통증이 있는 부위에 도포한 결과, 우수한 통증 감소 효과를 나타내는 것을 확인하였다.
따라서, 본 발명은 세포 내로의 도입이 용이하지 않은 보툴리눔 독소 경쇄에 세포투과성 펩타이드를 융합시켜 세포투과성을 부여함으로써 보툴리눔 독소 경쇄가 세포 내로 고효율로 전달될 수 있는 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 약학적 조성물로서,
상기 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄의 일측 말단 또는 양측 말단에 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드가 융합된 것을 특징으로 하는, 통증개선용 약학적 조성물을 제공한다.
본 발명에서 “보툴리눔 독소 재조합 단백질”이란 세포투과성 펩타이드와 보툴리눔 독소 경쇄(light chain)를 포함하며, 이들의 펩타이드 결합이나 공유결합과 같은 화학적 결합으로 형성된 결합체를 의미한다. 구체적으로, 본 발명에 따른 보툴리눔 독소 재조합 단백질은 세포 내로의 도입이 용이하지 않은 거대분자인 보툴리눔 독소 경쇄에 특정한 세포투과성 펩타이드를 융합시켜 세포투과성을 부여함으로써 보툴리눔 독소 경쇄를 세포 내로 고효율로 전달할 수 있으며, 이때 상기 융합은 상기 보툴리눔 독소 경쇄의 카복시 말단, 아미노 말단 또는 이들 모두에 상기 세포투과성 펩타이드가 융합될 수 있다.
본 발명에 따른 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄와 세포투과성 펩타이드의 융합을 통해 세포 내로 고효율로 전달될 수 있으며, 보툴리눔 독소 경쇄의 활성 및 안정성이 향상되어 생체 내에서 보툴리눔 독소의 고유의 효능이 극대화되는 것을 특징으로한다.
본 발명에서의 보툴리눔 독소 재조합 단백질(피부투과형 보툴리눔 유래 성분 펩타이드)의 구성은 대한민국 등록특허 제10-1882461호와 동일하며, 상기 문헌에 개시된 모든 내용은 본 출원에 참조로 원용된다.
본 발명에 있어서, 상기 보툴리눔 독소 재조합 단백질은 근육 마비 효과가 있는 보툴리눔 독소 경쇄가 세포 내로 고효율로 전달됨으로써 근육 긴장을 완화하고 근육 회복을 촉진시킴으로써 통증을 감소시킬 수 있다.
본 발명에 있어서, 상기 보툴리눔 독소 재조합 단백질은 피부를 투과하여 SNARE complex의 구성요소인 SNAP-25에 접근하고, 보툴리눔 독소 경쇄는 SNAP-25를 절단함으로써 아세틸콜린(acetylcholine)의 분비를 차단함으로써 통증을 개선할 수 있다.
본 발명에서, "보툴리눔 독소"는 세균에 의해 또는 재조합 기술에 의해 생성되거나 조작된 변이체들 또는 융합 단백질을 포함하여 뒤이어 발견될 수 있는 것인지 여부에 관계없이 임의의 공지된 종류의 보툴리눔 독소를 의미한다.
본 발명에서, 보툴리눔 독소 경쇄는 보툴리눔 독소 혈청형(serotype) A, B, C, D, E, F 및 G로 이루어진 군으로부터 선택될 수 있으며, 이때, 상기 보툴리눔 독소 경쇄는 서열번호 3 내지 서열번호 9으로 이루어진 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어질 수 있다. 또한, 상기 보툴리눔 독소 경쇄는 서열번호 3 내지 서열번호 9으로 이루어진 군으로부터 선택되는 하나의 아미노산 서열로 이루어질 수 있다. 이 때, 상기 보툴리눔 독소 경쇄는 서열번호 10 내지 16으로 이루어진 군으로부터 선택되는 염기서열로 이루어진 폴리뉴클레오티드에 의해 인코딩될 수 있으나, 이에 제한되지 않는다.
또한, 상기 보툴리눔 독소 경쇄는 일측 말단에 헥사히스티딘(hexahistidine) 태그(tag)를 더 포함할 수 있다. 본 발명에서, 상기 보툴리눔 독소 경쇄의 일측 말단에 헥사히스티딘 태그를 더 포함한 형태는 서열번호 17 내지 서열번호 23으로 이루어진 군으로부터 선택되는 아미노산 서열로 이루어질 수 있으며, 서열번호 24 내지 서열번호 30으로 이루어진 군으로부터 선택되는 염기서열에 의해 인코딩될 수 있으나, 이에 제한되지 않는다.
본 발명에서, 보툴리눔 독소 경쇄는 대안적으로 보툴리눔 독소 유도체, 즉, 보툴리눔 독소 활성을 가지나 임의로 부분 또는 서열 상에 하나 이상의 변형을 갖는 화합물일 수 있다. 예를 들어, 7종의 혈청형의 보툴리눔 독소 경쇄 단백질에 대비해, 아미노산 서열 상의 결실(deletion), 수정(modification), 치환(replacement) 또는 키메라 융합(chimeric fusion) 등의 방법을 적용하여 경쇄의 endopeptidase의 활성이 유지됨과 동시에 특성을 강화하거나 부작용을 감소시키는 방법으로 변형된 형태일 수 있다. 또는 재조합 또는 화학적인 합성으로 제조된 보툴리눔 독소 경쇄 또는 보툴리눔 독소 경쇄의 일부분이 이용될 수 있다.
본 발명에서, 상기 보툴리눔 독소 재조합 단백질은 서열번호 31 내지 서열번호 58로 구성된 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어질 수 있으며, 이들을 인코딩하는 폴리뉴클레오티드는 서열번호 59 내지 서열번호 86으로 구성된 군으로부터 선택되는 염기서열로 이루어질 수 있으나, 이에 한정되는 것은 아니다.
또한, 본 발명에서 상기 보툴리눔 독소 재조합 단백질은 서열번호 31 내지 서열번호 58로 구성된 군으로부터 선택되는 하나의 아미노산 서열로 이루어질 수 있다.
본 발명의 일 실시예에 따르면, 상기 보툴리눔 독소 재조합 단백질은 바람직하게는 서열번호 45로 표시되는 아미노산 서열로 이루어질 수 있으며, 이들을 인코딩하는 폴리뉴클레오티드는 서열번호 73로 표시되는 염기서열일 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 상기 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드(MTD; Macromolecule Transduction Domain)는 생물학적 활성 분자의 세포 내 운반을 매개할 수 있는 펩타이드로서, 인간 피부 각질 형성 세포와 신경세포에 대한 투과성을 동시에 보유하고 있을 수 있으나, 이에 제한되지 않는다.
상기 세포투과성 펩타이드는 바람직하게는 정의된 효소 또는 치료적 생물학적 활성을 갖지 않으나, 세포막을 통해 세포 내 전송을 가능하게 하는 전송체(carrier)로써 역할을 한다. 이는 세포 내로 전송하고자 하는 cargo의 N-말단 또는 C-말단 그리고, 양말단에 부착될 수 있으며, 각각의 말단에서 정방향 또는 역방향으로 부착될 수 있다. 또한, 본 발명에 따른 펩타이드는 바람직하게는 단위체(monomer)로 적용될 것이나, 이에 제한되지 않으며, 이합체(dimer) 또는 중합체(polymer) 형태의 구성으로도 이용될 수 있다. 더욱이, 본 발명에 따른 펩타이드는 서열번호 1의 아미노산 서열을 최소의 단위로 포함하는 펩타이드일 수 있다.
본 발명에 있어서, 상기 세포투과성 펩타이드는 서열번호 2의 염기서열로 이루어진 폴리뉴클레오티드에 의해 인코딩될 수 있으나, 이에 제한되지 않는다.
본 명세서에서 “유효성분”은 단독으로 목적으로 하는 활성을 나타내거나 또는 그 자체는 활성이 없는 담체 등과 함께 목적으로 하는 활성을 나타낼 수 있는 성분을 의미한다.
본 발명의 일 실시예에 따르면, 본 발명에 따른 조성물이 통증개선 효과를 내기 위한 가장 적절한 투여 경로는 피부를 통해 흡수되는 것이므로, 바람직하게는 본 발명에 따른 조성물은 경피 투여용일 수 있으나, 이에 제한되지 않는다.
본 발명에 있어서, 본 발명에 따른 조성물은 경피흡수촉진제를 더 포함할 수 있으나, 이에 제한되지 않는다.
상기 "경피흡수촉진제"는 유화제 중에서 피부투과에 영향을 주는 성분들로 통상적으로 경피 패치제에 사용되는 성분이다. 본 발명에서는 보툴리눔 독소 재조합 단백질의의 피부 투과 및 세포 침투를 증대시키는 역할을 하는 것으로, 레시틴(Lecithin), 라우릴 피롤리돈(Lauryl Pyrrolidone), 글리세롤 모노올리에이트(Glycerol Monooleate), 글리세롤 모노라우레이트(Glycerol Monolaurate), 프로필렌 글리콜 모노라우레이트(Propylene Glycol Monolaurate), 폴리옥시에틸렌 소르비탄 모노올리에이트(Polyoxyethylene Sorbitan Monooleate), 폴리옥시에틸렌 소르비탄 모노스테아레이트(Polyoxyethylene Sorbitan Monostearate), 폴리옥시에틸렌 소르비탄 모노라우레이트(Polyoxyethylene Sorbitan Monolaurate), 소르비탄 모노올리에이트(Sorbitan Monooleate), 소르비탄 모노스테아레이트(Sorbitan Monostearate), 소르비탄 모노라우레이트(Sorbitan Monolaurate)일 수 있으나, 이에 제한되지 않는다.
본 발명에서, "통증"은 강한 자극이나 신체 손상으로 인하여 불쾌한 감정을 느끼는 것을 총칭하는 개념이다. 본 발명의 목적상 상기 통증은 신체적인 통증을 의미할 수 있다. 구체적으로, 상기 통증은 기계적 통증, 열 통증, 만성 통증, 급성 통증, 통각과민 통증, 체인성 통증, 신경병성 통증, 심인성 통증, 열 유도된 통증, 물리적 통증, 통각수용, 통각과민, 류마티스성 통증, 두통, 요통, 골반통, 근막통, 혈관성 통증, 편두통, 상처 연계된 통증, 염증성 통증, 관절염 통증, 당뇨병성 통증, 암 유래의 통증, 신체 내장 통증, 섬유근통, 수술 후 통증, 환상통, 삼차신경통, 포진 후 신경통, 통증이 있는 당뇨병성 신경병증, 통증이 있는 당뇨병성 말초 신경병증, 당뇨병성 다발성 신경증, 좌골 통증, 신경근병증, 척수 신경근통, 근육통, 및 요추 통증으로 이루어진 군으로부터 선택될 수 있으나, 이에 제한되지 않는다, 본 발명의 일 실시예에 따르면, 상기 통증은 두통, 만성 통증, 근육통, 섬유근통, 또는 근막통일 수 있다.
본 발명에서 사용되는 용어, “만성 통증”은 통증의 기간과 강도가 환자의 기능이나 생활에 유해한 효과를 일으키거나 또는 정상적인 조직 치유기간, 보통 3개월을 넘어서 지속되는 경우를 의미한다.
본 발명에서 사용되는 용어, “섬유근통(fibromyalgia)”은 만성적으로 전신의 근골격계 통증, 뻣뻣함, 감각 이상, 수면 장애, 피로감을 일으키고, 신체 곳곳에 압통점(누르면 아픈 부분)이 나타나는 힘줄 및 인대 근막과 근육, 지방조직 등 연부조직의 통증 증후군을 의미한다.
본 발명에서 사용되는 용어 “근막통(myofascial pain syndrome)”은 근육이 딱딱하게 뭉쳐지면서 통증을 유발하는 질환으로 잘못된 자세와 스트레스에 의해 어깨나 뒷목 주변 근육이 쉬지 못하고 장시간 긴장함에 따라 근육에 영양분과 산소가 부족해져 발생하는 것을 의미한다.
본 명세서에 기재된 특정 서열번호로 표시되는 아미노산 서열로 이루어진 단백질(펩타이드)은 해당 아미노사 서열에만 제한되지 않으며, 상기 특정 서열번호로 표시되는 아미노산 서열로 이루어진 단백질들의 기능적 동등물도 포함한다. 본 발명의 피부외용제 조성물에서 약학적으로 허용되는 담체로는 그의 제형에 따라 다르나, 바셀린, 유동 파라핀, 겔화 탄화수소(플라스티베이스) 등의 탄화수소류; 중쇄지방산트리글리세라이드, 돈지, 하드 팻, 카카오지 등의 동식물성 오일; 세탄올, 스테아릴알코올, 스테아린산, 팔미틴산이소프로필 등의 고급지방산 알코올 및 지방산 및 그의 에스테르류; 폴리에틸렌글리콜, 1,3-부틸렌글리콜, 글리세롤, 젤라틴, 백당, 당알코올 등의 수용성 기제; 글리세린 지방산 에스테르, 스테아린산폴리옥실, 폴리옥시에틸렌경화 피마자유 등의 유화제; 아크릴산에스테르, 알긴산나트륨 등의 점착제; 액화석유가스, 이산화탄소 등의 분사제; 파라옥시벤조산에스테르류 등의 방부제 등을 들 수 있다. 또한, 이들 외에도 안정제, 향료, 착색제, pH 조정제, 희석제, 계면활성제, 보존제, 항산화제 등을 필요에 따라 배합할 수 있다. 본 발명의 피부외용제 조성물의 사용은 통상의 방법에 의해 환부 또는 통증이 발현하는 부위의 피부에 도포하는 것이 바람직하다.
또한, 본 발명에 따른 피부외용제 조성물은 통상적인 반창고의 창상 박리 커버 등과 같은 고체 지지체 상에 점착되어 사용될 수 있다. 점착은 본 발명의 조성물을 고체 지지체에 포화시킨 후 탈수시켜 달성할 수 있다. 바람직하게는 고체 지지체를 먼저 점착제로 피복하여 고체 지지체에 본 발명의 조성물의 부착을 향상시킬 수 있다. 점착제의 예로는 폴리아크릴레이트 및 시아노아크릴레이트 등이 있다.
또한, 본 발명은 본 발명의 다른 양태로 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 화장료 조성물로서, 상기 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄의 일측 말단 또는 양측 말단에 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드가 융합된 것을 특징으로 하는, 통증개선용 화장료 조성물을 제공한다.
본 발명에 따른 화장료 조성물은 당업계에서 통상적으로 제조되는 어떠한 제형으로도 제조될 수 있으며, 본 발명에 따른 조성물 이외에 피부과학적으로 허용 가능한 매질, 기제, 보조제, 부형제 등과 함께 피부과학 분야에서 통상적으로 제조되는 국소 적용 또는 전신 적용할 수 있는 보조제 형태로 제조될 수 있다. 예를 들어, 기초 화장품 조성물(화장수, 크림, 에센스, 클렌징폼 및 클렌징 워터와 같은 세안제, 팩, 보디오일), 색조 화장품 조성물(화운데이션, 립스틱, 마스카라, 메이크업 베이스), 두발 제품 조성물(샴푸, 린스, 헤어컨디셔너, 헤어젤) 및 비누 등의 형태로 제조될 수 있다.
또한, 본 발명의 화장료 조성물은 추가로 지방 물질, 유기 용매, 용해제, 농축제 및 겔화제, 연화제, 항산화제, 현탁화제, 안정화제, 발포제(foaming agent), 방향제, 계면활성제, 물, 이온형 또는 비이온형 유화제, 충전제, 금속이온 봉쇄제 및 킬레이트화제, 보존제, 비타민, 차단제, 습윤화제, 필수 오일, 염료, 안료, 친수성 또는 친유성 활성제, 지질 소낭 또는 화장품에 통상적으로 사용되는 임의의 다른 성분과 같은 화장품학 또는 피부과학 분야에서 통상적으로 사용되는 보조제를 함유할 수 있다. 그리고 상기의 성분들은 피부과학 분야에서 일반적으로 사용되는 양으로 도입될 수 있다.
상기 부형제로는 이에 한정되지는 않으나 예를 들어, 피부연화제, 피부 침투 증강제, 착색제, 방향제, 유화제, 연화제, 농화제, 농축제, 겔화제, 현택화제, 안정화제, 발포제(foaming agent), 계면활성제, 충전제 또는 용매를 포함할 수 있다. 또한 향료, 색소, 살균제, 산화방지제, 방부제, 보조제 및 보습제 등을 추가로 포함할 수 있으며, 물성 개선을 목적으로 점증제, 무기염류, 합성 고분자 물질 등을 포함할 수 있다. 예를 들면, 본 발명의 화장료 조성물로 세안제 및 비누를 제조하는 경우에는 통상의 세안제 및 비누 베이스에 본 발명의 추출물을 첨가하여 용이하게 제조할 수 있다. 크림을 제조하는 경우에는 일반적인 수중유적형(O/W)의 크림베이스에 본 발명의 보툴리눔 독소 재조합 단백질을 첨가하여 제조할 수 있다. 여기에 향료, 킬레이트제, 색소, 산화방지제, 방부제 등과 물성 개선을 목적으로 한 단백질, 미네랄, 비타민 등 합성 또는 천연 소재를 추가로 첨가할 수 있다.
적합한 화장료 조성물의 제형으로는 예를 들면 용액, 겔, 고체 또는 반죽 무수 생성물, 수상에 유상을 분산시켜 얻은 에멀젼, 현탁액, 마이크로에멀젼, 마이크로캡슐, 미세과립구 또는 이온형(리포좀), 비이온형의 소낭 분산제의 형태, 크림, 스킨, 로션, 파우더, 연고, 스프레이 또는 콘실 스틱(conceal stick)의 형태로 제공될 수 있다. 또한, 포말(foam)의 형태 또는 압축된 추진제를 더 함유한 에어로졸 조성물의 형태로도 제조될 수 있다.
본 발명의 화장료 조성물을 첨가할 수 있는 제품으로는 이에 한정되는 것은 아니나 스킨로션, 스킨 소프너, 스킨토너, 수렴화장수, 유연화장수, 영양화장수, 아스트린젠트, 로션, 밀크로션, 모이스처 로션, 영양로션, 바디크림, 마사지크림, 영양크림, 모이스처 크림, 핸드크림, 에센스, 영양에센스, 팩, 비누, 샴푸, 클렌징폼, 클렌징로션, 클렌징크림, 바디로션, 바디클렌저, 트리트먼트, 미용액, 유액, 프레스파우더, 루스파우더, 아이섀도 등의 제형을 포함한다.
본 발명에 따른 조성물은 멘톨(Menthol), 시어 버터(Shea Butter), 스카치 파인 잎(Scotch Pine Leaf), 알로에 베라(Aloe Vera), 박하(Peppermint), 로즈마리(Rosemary) 등의 추출물 더 포함할 수 있으나, 이에 제한되지 않는다.
이하, 본 발명의 이해를 돕기 위하여 바람직한 실시예를 제시한다. 그러나 하기의 실시예는 본 발명을 보다 쉽게 이해하기 위하여 제공되는 것일 뿐, 하기 실시예에 의해 본 발명의 내용이 한정되는 것은 아니다.
[실시예]
실시예 1. 두통개선 효과 확인
1-1. 실험 재료 및 실험 방법
두통으로 일상 생활에 불편함을 느끼며 진통제를 복용하고 있는 23 ~ 53세 성인 남녀 총 9명(여성 7명, 남성 2명, 평균 연령 36세)을 대상으로 대한민국 등록특허 제10-1882461호에 따라 제조한 보툴리눔 독소 재조합 단백질(피부 투과형 보툴리눔 유래 성분 펩타이드)이 함유된 용액 2.5 cc를 통증이 있는 두피 부위와 이마에 매일 아침마다 직접 도포하였다.
평가는 시험 전 피험자들에게 설문 조사 후 두통학회의 '두통일기 앱'을 통해 두통이 있는 날마다 피험자가 두통횟수 및 진통제 복용횟수를 직접 입력하도록 하였다. 시험기간은 총 12주로 피험자가 직접 입력한 정보로 두통횟수는 사용 전, 사용 후 1 ~ 4주, 사용 후 5 ~ 8주, 사용 후 9 ~ 12주로 4가지를 비교 평가하였다. 진통제 복용횟수는 사용 후 1 ~ 4주, 사용 후 5 ~ 8주, 사용 후 9 ~ 12주로 3가지를 비교 평가하였다.
1-2. 두통횟수
두통횟수를 비교 평가한 결과, 도 1에 나타낸 바와 같이, 피험자 9명 모두 두통이 개선된 것을 확인할 수 있었다. 구체적으로, 사용 후 기간별 평균 두통발생횟수는 사용 전 4주간(14.3회), 사용 후 1 ~ 4주(8.5회), 사용 후 5 ~ 8주(6.9회), 사용 후 9 ~ 12주(2.5회)로 피부 투과형 보툴리눔 유래 성분 펩타이드가 함유된 용액을 사용하기 전 4주간에 비해 사용 후 9 ~ 12주 기간 동안 피험자들의 평균 두통발생횟수는 84% 감소한 것을 확인할 수 있었다.
또한, 도 2에 나타낸 바와 같이, 사용 후 기간별 두통횟수 감소율은 사용 초기인 1 ~ 4주와 사용 마지막인 9 ~ 12주를 비교한 결과, 20% 이상 개선된 피험자가 2명(7명 →9명), 50% 이상 개선된 피험자가 4명(4명 →8명), 70% 이상 개선된 피험자가 4명(1명 →5명), 100% 개선된 피험자가 3명(0명 →3명)으로 증가한 것을 확인할 수 있었다.
1-3. 진통제 복용횟수
사용 후 1 ~ 4주와 9 ~ 12주의 진통제 복용횟수를 비교 평가한 결과, 피험자 9명 중 8명에서 진통제 복용횟수가 감소하였으며, 1명은 1회 증가한 것으로 나타났다.
사용 후 기간별 진통제 복용횟수를 비교한 결과, 도 3에 나타낸 바와 같이, 사용 후 1 ~ 4주 동안은 2회 3명, 1회 3명, 0회 3명이였으며, 사용 후 5 ~ 8주 동안은 3회 1명, 2회 1명, 1회 4명, 0 회 3명으로 복용횟수가 1회 증가한 사람이 1명, 복용횟수가 1회 감소한 사람이 1명이었다. 사용 후 9 ~ 12주 동안은 3회 1명, 2회 0명, 1회 2명, 0회 6명으로 사용 초기 1 ~ 4주에 비해 진통제 복용이 필요 없는 피험자가 3명 증가한 것을 확인할 수 있었다.
상기 결과로부터, 보툴리눔 독소 주사와 유사한 주름 개선 효과를 보이는 피부 투과형 보툴리눔 유래 성분 펩타이드를 두경부에 도포할 경우, 전반적으로 두통횟수와 진통제 복용횟수가 감소하였는바, 두통 개선효과가 있을 것으로 판단되었다.
실시예 2. 통증개선 효과 확인
2-1. 실험 재료 및 실험 방법
30 ~ 75세의 만성 통증, 섬유근통, 근막통, 또는 근육통 환자인 성인 24명을 대상으로 피부에 도포한 피부투과형 보툴리눔 유래 성분 펩타이드가 근육 긴장을 완화하여 통증을 감소시킬 수 있는지 여부 및 근육 경련성 통증을 감소시킬 수 있는지 여부, 및 피부투과형 보툴리눔 유래 성분 펩타이드의 반복적인 움직임, 기계적 충격으로 유발되는 만성 통증의 개선 정도를 확인하였다.
실험 대상은 하기 표 1의 3가지 조건 중 하나를 충족하도록 구성하였으며, 피부투과형 보툴리눔 유래 성분 펩타이드가 함유된 보툴리눔 로션 적당량을 매일 2회 통증 부위에 직접 도포하도록 하였다.
실험 대상 24명을 다음과 같이 3개의 그룹으로 나누어 분석하였다:
Group A: 만성 통증 환자
Group B: 섬유근통, 및 근육통 환자
Group C: 마약성 진통제 사용 그룹(Group C의 경우, Group A, B와 표본 중복 있음).
2-2. Group A
만성 통증 환자 14명(표 1의 각 조건을 만족하는 환자를 적절하게 섞음)에서 보툴리눔 로션 사용 후 6일 간의 통증 변화를 확인한 결과, 도 4에 나타낸 바와 같이, 보툴리눔 로션 사용 후 78.57%의 환자에서 통증이 감소한 것을 확인할 수 있었다 (14명 중 11명이 통증이 감소함). 보툴리눔 로션을 사용하는 동안 통증은 감소된 상태를 유지하였다. 보툴리눔 로션 사용 직후 즉각적인 통증 감소 효과가 나타났으며, 전반적으로 로션을 바르는 동안 통증 감소 효과가 유지되는 것을 확인할 수 있었다.
2-3. Group B
섬유근통, 및 근육통 환자 7명(표 1의 각 조건을 만족하는 환자를 적절하게 섞음)에서 보툴리눔 로션 사용 후, 통증 감소 지속 여부 확인한 결과, 보툴리눔 로션 사용 직후 71.43%의 환자에서 통증이 감소하였으며(7명 중 5명이 통증이 감소한 것으로 나타남, 도면에 나타내지는 않음), 도 5에 나타낸 바와 같이, 57.14%의 환자에서 로션을 바르는 동안 통증 감소 상태가 유지되는 것으로 나타났다 (7명 중 4명이 통증 감소 상태가 유지됨). 전반적으로 보툴리눔 로션 사용 직후 즉각적인 통증 감소 효과를 경험했으며, 보툴리눔 로션을 바르는 동안 효과가 유지되는 것으로 나타났다.
2-4. Group C
마약성 진통제를 사용하는 환자 15명에서 보툴리눔 로션 사용 후, 통증 감소 지속 여부를 확인한 결과, 도 6에 나타낸 바와 같이, 80%의 환자가 즉각적으로 통증이 완화되었다고 응답하였으며(도면에 나타내지는 않음), 도 6에 나타낸 바와 같이, 66.67%의 환자가(15명 중 10명) 보툴리눔 로션을 사용하는 동안 지속적인 통증 개선 효과가 있었다고 응답하였다. 보툴리눔 로션 사용 직후, 전반적으로 즉각적인 통증 감소 효과를 경험했으며, 로션을 바르는 동안 효과가 유지되는 것으로 나타났다.
전술한 본 발명의 설명은 예시를 위한 것이며, 본 발명이 속하는 기술분야의 통상의 지식을 가진 자는 본 발명의 기술적 사상이나 필수적인 특징을 변경하지 않고서 다른 구체적인 형태로 쉽게 변형이 가능하다는 것을 이해할 수 있을 것이다. 그러므로 이상에서 기술한 실시예들은 모든 면에서 예시적인 것이며 한정적이 아닌 것으로 이해해야만 한다.
본 발명에 따른 보툴리눔 독소 재조합 단백질은 세포투과성 펩타이드와의 융합을 통해 근육 마비 효과를 가지는 보툴리눔 독소 경쇄를 경피로 전달할 수 있는바, 간편하게 보툴리눔 독소 경쇄를 통증 부위에 빠르게 전달함으로써 효과적으로 근육 긴장을 완화시켜 통증을 감소시킬 수 있으므로 통증 예방, 개선, 또는 치료 물질로 유용하게 이용될 수 있어 산업상 이용가능성이 있다.
Claims (16)
- 보툴리눔 독소 재조합 단백질을 유효성분으로 포함하는 통증개선용 약학적 조성물로서,상기 보툴리눔 독소 재조합 단백질은 보툴리눔 독소 경쇄의 일측 말단 또는 양측 말단에 서열번호 1의 아미노산 서열로 이루어진 세포투과성 펩타이드가 융합된 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 보툴리눔 독소 재조합 단백질은 서열번호 31 내지 서열번호 58로 이루어진 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어진 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 보툴리눔 독소 경쇄는 서열번호 3 내지 서열번호 9으로 이루어진 군으로부터 선택되는 하나 이상의 아미노산 서열로 이루어진 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 보툴리눔 독소 경쇄는 일측 말단에 헥사히스티딘(hexahistidine) 태그(tag)를 더 포함하는 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 보툴리눔 독소 경쇄는 보툴리눔 독소 혈청형(serotype) A, B, C, D, E, F 및 G로 이루어진 군으로부터 선택되는 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 융합은 상기 보툴리눔 독소 경쇄의 카복시 말단, 아미노 말단 또는 이들 모두에 상기 세포투과성 펩타이드가 융합되는 것을 특징으로 하는, 통증개선용 약학적 조성물.
- 제1항에 있어서,상기 융합은 펩타이드 결합 또는 공유결합에 의해 이루어지는 것을 특징으로 하는, 통증개선용 약학적 조성물.

- 제1항에 있어서,상기 통증은 기계적 통증, 열 통증, 만성 통증, 급성 통증, 통각과민 통증, 체인성 통증, 신경병성 통증, 심인성 통증, 열 유도된 통증, 물리적 통증, 통각수용, 통각과민, 류마티스성 통증, 두통, 요통, 골반통, 근막통, 혈관성 통증, 편두통, 상처 연계된 통증, 염증성 통증, 관절염 통증, 당뇨병성 통증, 암 유래의 통증, 신체 내장 통증, 섬유근통, 수술 후 통증, 환상통, 삼차신경통, 포진 후 신경통, 통증이 있는 당뇨병성 신경병증, 통증이 있는 당뇨병성 말초 신경병증, 당뇨병성 다발성 신경증, 좌골 통증, 신경근병증, 척수 신경근통, 근육통, 및 요추 통증으로 이루어진 군으로부터 선택되는 것을 특징으로 하는, 통증개선용 약학적 조성물.
