Abstract
Description
[1] 培養上清中の超低密度リポタンパク質(VLDL)の増減を指標にして、脂肪肝をはじめとする脂質異常に効果を有する物質のスクリーニング方法に使用するための、ヒト脂肪肝モデル細胞であって、
該細胞の培養上清がVLDL及び低密度リポタンパク質(LDL)を含有し、該VLDLが該LDLよりも多く含むことを特徴とする、細胞。
[2] 単層細胞培養物である、[1]の細胞。
[3] 前記VLDLが、Large-VLDL、Medium-VLDL、及びSmall-VLDLを含み、該Large-VLDLをVLDL全体の70質量%以上の割合で含む、[1]又は[2]の細胞。
[4] 脂肪肝由来のヒト肝細胞を、ジメチルスルホキシドを含む培地中にて培養して得られた細胞である、[1]~[3]のいずれかの細胞。
[4-1] ジメチルスルホキシドを含まないこと以外は同じ条件で培養された脂肪肝由来のヒト肝細胞の2倍以上、3倍以上、4倍以上、5倍以上、6倍以上、7倍以上、8倍以上、又は9倍以上の脂質を内包する、[1]~[4]のいずれかの細胞。
[4-2] 培養上清中に含まれるリポタンパク質(より好ましくはトリグリセリド)量が、ジメチルスルホキシドを含まないこと以外は同じ条件で培養された脂肪肝由来のヒト肝細胞の培養上清中に含まれるリポタンパク質(より好ましくはトリグリセリド)量の5倍以上、6倍以上、7倍以上、8倍以上、又は9倍以上である、[1]~[4]のいずれかの細胞。
[5] 脂肪肝由来のヒト肝細胞が、ヒト肝細胞を有するキメラ非ヒト動物より回収されたものである、[4]、[4-1]又は[4-2]の細胞。
[6] 脂質異常に効果を有する物質のスクリーニング方法であって、
[1]~[5]のいずれかの細胞に被検物質を投与する工程、ならびに、
被験物質を投与した細胞と投与されていない細胞との間で培養上清中の超低密度リポタンパク質(VLDL)の量を比較する工程、を含む方法。
本明細書は本願の優先権の基礎である2021年2月26日に出願された日本国特許出願2021- 30911号の明細書等に記載される内容を包含する。
本明細書で引用した全ての刊行物、特許および特許出願をそのまま参考として本明細書にとりいれるものとする。
本発明におけるヒト脂肪肝モデル細胞は、脂肪肝由来のヒト肝細胞を、ジメチルスルホキシドを含む培地中にて培養する工程を含むヒト脂肪肝モデル細胞の製造方法により製造することができる。以下、本発明のヒト脂肪肝モデル細胞の製造方法について記載する。
本発明において「脂肪肝由来のヒト肝細胞」とは、脂肪肝組織より回収されたヒト肝細胞、あるいは、それを一旦冷凍保存した後解凍したものを用いることができる。脂肪肝組織は、脂肪肝を有するヒト患者に由来する脂肪肝組織、肝障害免疫不全非ヒト動物にヒト肝細胞を移植して得られた非ヒト動物モデル(以下、「キメラ非ヒト動物」と記載する)に由来する脂肪肝組織等を用いることができる。
本発明において利用可能なキメラ非ヒト動物に由来する脂肪肝組織より回収されたヒト肝細胞は以下の手法により調製することができる。
本発明において「キメラ非ヒト動物」とは、肝臓における肝細胞の一部又は全部が、ヒト肝細胞に置換されている非ヒト動物を意味する。
「肝障害免疫不全非ヒト動物」とは、異種動物由来の細胞に対して拒絶反応を示さない免疫不全であるとともに、非ヒト動物由来の肝臓の細胞が障害を受けている動物を意味する。非ヒト動物由来の肝臓の細胞が障害を受けていることにより、移植されたヒト肝細胞は増殖がしやすく、また、移植されたヒト肝細胞により肝機能が維持される。
本発明において「肝障害免疫不全非ヒト動物」は、免疫不全の形質を規定する遺伝子及び肝障害の形質を規定する遺伝子を、それぞれをホモ接合型で有していてもよいし、ヘテロ接合型で有していてもよい。例えば、本発明の肝障害免疫不全非ヒト動物としては、uPA(+/-)/SCID(+/+)、uPA(+/+)/SCID(+/+)などの遺伝子型で表される肝障害免疫不全マウスを好適に利用することができる。
本発明において肝障害免疫不全非ヒト動物に移植される「ヒト肝細胞」は、ヒト由来の肝細胞であればよく、例えば、ヒト肝組織から、コラゲナーゼ灌流法等の常法にしたがって単離されたヒト肝細胞を利用することができる。ヒト肝組織は健常人由来の肝組織であってもよいし、脂肪肝や肝臓がん等の疾患に罹患した患者由来の肝組織であってもよいが、好ましくは健常人由来の肝組織である。肝細胞を単離するヒトの年齢は特に制限されないが、14歳以下の小児の肝組織より単離するのが好ましい。14歳以下の小児の肝細胞を使用することにより、移植後、ヒト肝細胞による高置換率を達成することができる。また、単離した肝細胞を一旦冷凍保存した後解凍して用いることもできる。
肝障害免疫不全非ヒト動物へのヒト肝細胞の移植は、当該非ヒト動物の脾臓を経由して肝臓へ移植することにより行うことができる。あるいは、直接門脈から移植することもできる。移植するヒト肝細胞の数は、1~200万個程度、好ましくは20万~100万個程度とすることができる。肝障害免疫不全非ヒト動物の性別は特に限定されない。また、移植に用いる肝障害免疫不全非ヒト動物の日齢は、特に限定されないが、低週齢のときにヒト肝細胞を移植すると、動物の成長とともにヒト肝細胞がより活発に増殖し得ることから、生後0~40日程度、好ましくは生後8~40日程度のものを使用することができる。
キメラ非ヒト動物からのヒト肝細胞の回収は、コラゲナーゼ灌流法等の常法にしたがって行うことができる。ヒト肝細胞の回収は、回収される肝細胞中にヒト肝細胞が高率で含まれるキメラ非ヒト動物を用いて行うことが好ましくは、例えば、以下の一又は複数の特徴を有するキメラ非ヒト動物を用いて行うことができる。
(i)肝臓における肝細胞の60%以上、好ましくは70%以上、より好ましくは80%以上、さらに好ましくは90%以上、よりさらに好ましくは95%以上、特に好ましくは99%以上がヒト肝細胞に置換されている;
(ii)血中ヒトアルブミン量が0.1mg/mL以上、好ましくは0.5mg/mL以上、より好ましくは1mg/mL以上、さらに好ましくは5mg/mL以上、よりさらに好ましくは10mg/mL以上である;
(iii)ヒト肝細胞の移植後、12週~21週、好ましくは13週~20週、より好ましくは14週~19週、経過している。
本発明において、脂肪肝由来のヒト肝細胞の培養は、動物細胞の培養に一般的に用いられる培地を使用して行うことができる。このような培地としては、例えば、ダルベッコ改変イーグル培地(DMEM)、ウイリアムス培地E等が挙げられるが、これらに限定はされない。好ましくは、DMEMを利用することができる。培地には必要に応じてさらに、ウシ胎児血清、インスリン、上皮成長因子、デキサメタゾン、緩衝剤、抗生物質、pH調整剤、プロリン、アスコルビン酸、ニコチンアミド等を適宜添加することができる。
本発明のヒト脂肪肝モデル細胞は、ヒト脂肪肝における肝細胞と同様に、多くの脂肪滴を内包し、多くの脂質を分泌及び/又は蓄積し、脂肪肝関連遺伝子の発現等が観察され、脂肪肝の症状を維持するヒト由来の培養肝細胞であり、より詳細には単層の肝細胞培養物である。
本発明のヒト脂肪肝モデル細胞は、培養上清中のリポタンパク質のサブクラスの含有量によっても特徴付けることができる。
本発明のヒト脂肪肝モデル細胞はまた、細胞における脂肪肝関連遺伝子の発現量によっても特徴付けることができる。
本発明のヒト脂肪肝モデル細胞は、DMSOを含む培地中にて培養することが好ましい。
本発明のヒト脂肪肝モデル細胞は、ヒト脂肪肝のモデルとして用いることができ、培養上清中の超低密度リポタンパク質(VLDL)含量の増減を指標にして、脂肪肝をはじめとする脂質異常に効果を有する物質をスクリーニングする方法において利用することができる。
1.脂肪肝由来のヒト肝細胞の調製
(ヒト肝細胞を有するキメラマウス(PXBマウス)の作製)
PXBマウスは従来公知の手法(特開2002-45087号公報)に準じて作製した。すなわち、肝臓で合成されるアルブミンのエンハンサー、及びプロモーターに連結したウロキナーゼプラウミノーゲンアクチベーター(uPA)遺伝子(cDNA-uPA)が全細胞に導入された遺伝的肝障害マウスと、免疫不全マウス(SCIDマウス)とを交配させて免疫不全肝障害マウス(cDNA-uPA(+/-)/SCIDマウス)を作製した。
麻酔下のPXBマウスを解剖台に載せ、医療用テープで固定後に開腹した。留置針を門脈に挿し、灌流液Aを送流し、脱血した。灌流液Bを送流し肝組織中のコラーゲンを溶解し、腸管や胃を傷つけないよう肝臓を切り出し、灌流液C中で肝臓を揺らして肝細胞をほぐし、遊離した。未消化組織片を除去するために、セルストレイナーに通過させ、肝細胞をチューブに回収した。
回収した肝細胞(PXB-cells)を遠心分離し、上清を取り除いた後、得られた沈殿に、培地Aを40mL加え静かに混合した。この操作を2回繰り返し、上清中に浮遊した不純物や脂質等を除去した。細胞塊を単離するために、セルストレイナーに通過させ、チューブに回収した。細胞数の計測は、トリパンブルー色素排除法で血球計算盤を使用して行い、計数した値より、細胞密度、総細胞数、及び生存率を求めた。
細胞懸濁液の細胞密度から、目的とする播種密度の希釈倍率を算出し、培地Aにて希釈した。希釈した細胞懸濁液500μLを培養プレートの各Wellに静かに注ぎ、細胞が底面に軽く接着するまで約20分間静置した後、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))または培地C(DMSO(-))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、5日間培養した。培養終了後、顕微鏡カメラを用いて細胞像を撮影後に培養上清を回収し、下記培養上清中のリポタンパク質の解析を行った。
・DMSO含有培地中で5日間培養
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から1日目及び2日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた5日間の培養終了後、下記培養上清中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、及び8日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた9日間の培養終了後、下記培養上清中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、及び8日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた12日間の培養終了後、下記培養上清中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、8日目、及び12日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた14日間の培養終了後、下記培養上清中のリポタンパク質の解析を行った。
・DMSO含有培地中で5日間培養

培地B(DMSO(+))を用いた5日間の培養終了後、細胞内のコレステロール及びトリグリセリド量について、下記細胞中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、及び8日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた9日間の培養終了後、細胞内のコレステロール及びトリグリセリド量について、下記細胞中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、及び8日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた12日間の培養終了後、細胞内のコレステロール及びトリグリセリド量について、下記細胞中のリポタンパク質の解析を行った。
播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。培地B(DMSO(+))を用いた培養開始から5日目、7日目、8日目、及び12日目に、培地を新しい培地B(DMSO(+))に交換した。
培地B(DMSO(+))を用いた14日間の培養終了後、細胞内のコレステロール及びトリグリセリド量について、下記細胞中のリポタンパク質の解析を行った。
既知肝細胞(HepG2細胞、HuH7細胞)を500μLの培地Aに播種し、インキュベーター(37℃,5%CO2)へ静かに移し、7日間培養した。培養終了後、下記培養上清中のリポタンパク質の解析、ならびに、下記細胞中のリポタンパク質の解析を行った。
(培養上清中のリポタンパク質の解析)
PXB-cells、HepG2、及びHuH7の培養上清中に含まれるリポタンパク質の解析には、株式会社スカイライト・バイオテック社のLipoSEARCH(登録商標)法を用いた。
・細胞中トリグリセリド測定
細胞中のトリグリセリドの測定は、コレステスト(登録商標)TG(積水メディカル株式会社)を製造元の指示書に従って用いて行った。すなわち、細胞をPBSで洗浄後、完全に水を切り(測定までは-80℃保存)、細胞Well毎に200μLのTG酵素液(1)を添加し、37℃、10分間保温し、反応させた(フリーのグリセロールを除去)。次いで、ピペットで細胞を剥離させ、遠心チューブに移し10,000rpm×10分間遠心分離した後、上清7.5μLを96穴マイクロプレートに移し、68μLのTG酵素液(1)を添加し、37℃、10分間保温し、完全にフリーのグリセロールを除去した。次いで、25μLのTG酵素液(2)を添加し、37℃、10分間保温した。得られた反応物について、550nmにて吸光度を測定し、HDL-C180Aを標準としてトリグリセリド含量を算出(HDL-C180Aのトリグリセリド濃度は、52.26mg/dL)した。
細胞中のコレステロールの測定は、コレステスト(登録商標)CHO(積水メディカル株式会社)を製造元の指示書に従って用いて行った。すなわち、細胞をPBSで洗浄後、完全に水を切り(測定までは-80℃保存)、細胞Well毎に200μLのCHO酵素液(1)を添加し、37℃、10分間保温した。次いで、ピペットで細胞を剥離させ、遠心チューブに移し10,000rpm×10分間遠心分離した後、上清15μLを96穴マイクロプレートに移し、68μLのCHO酵素液(1)を添加し、37℃、10分間保温した。次いで、25μLのCHO酵素液(2)を添加し、37℃、10分間保温した。得られた反応物について、550nmにて吸光度を測定し、HDL-C180Aを標準としてコレステロール含量を算出(HDL-C180Aのトリグリセリド濃度は、152.67mg/dL)した。
培地Bで3日間又は6日間培養したPXB-cellsの総RNAを、TRIzol(登録商標)+Direct zol(Thermo Fisher Scientific社)を、製造元の指示書に従って用いて抽出した。
灌流液A、灌流液B、灌流液C、培地A、培地B、培地C、及び培地Dは以下の組成のものを用いた。
1.DMSO含有培地中で培養した培養上清中のリポタンパク質の解析結果
図1は、培地B(DMSO(+))又は培地C(DMSO(-))で5日間培養したPXB-cellsを示す。培地B(DMSO(+))で培養したPXB-cells(図1、左)においては、培地C(DMSO(-))で培養した場合(図1、右)と比べて多くの(およそ2倍)脂肪滴(白色で示される)が観察された。いずれの場合においても、PXB-cellsは単層細胞培養物を形成した。
図3に、培地B(DMSO(+))で5、9、12、及び14日間培養したPXB-cells、ならびに既知肝細胞(HepG2細胞、HuH7細胞)の各培養上清中のリポタンパク質の解析結果を示す。

図6に、培地B(DMSO(+))で3日間、及び6日間培養したPXB-cellsにおける脂肪肝関連遺伝子(FASN、SREBF1、G6PC)の発現量を解析した結果を示す。結果は、培養3日目の各遺伝子の発現量を「1」とする相対値にて示す。いずれの遺伝子についても、培養3日目より培養6日間において発現量の上昇が認められた。また、図6には示さないが、CYP7A1、CETP、GCK、及びPCK1についても同様に、培養3日目より培養6日間において発現量の上昇が認められた。
(1-1)オレイン酸の添加
・細胞の調製
PXB-cellsを、培地Aにて希釈した。希釈した細胞懸濁液500μLを培養プレートの各Wellに静かに注ぎ、細胞が底面に軽く接着するまで約20分間静置した後、インキュベーター(37℃,5%CO2)へ静かに移し、培養した。播種の翌日培地Aを除去して、500μLの培地B(DMSO(+))を添加し、インキュベーター(37℃,5%CO2)へ静かに移し、13日間培養した。培養終了後、下記試験に用いた。
被験物質として、肝細胞における脂質蓄積を促すことが知られているオレイン酸を用いた。
上記調製したPXB-cellsの培地を、オレイン酸を0.75mMの量で含む培地Dに交換し、インキュベーター(37℃,5%CO2)へ静かに移し、2日間培養した。
培養終了後、上記「培養上清中のリポタンパク質の解析」及び「細胞中のリポタンパク質の解析」に記載の手法にて、培養上清中及び細胞中の総コレステロール及び総中性脂肪(トリグリセリド)をそれぞれ測定した。
対照には、オレイン酸の代わりに、培地D(5μL)のみを添加した。
図7に、オレイン酸を添加して、2日間培養したPXB-cellsの培養上清におけるリポタンパク質の各サブグループにおける中性脂肪(トリグリセリド)含量について解析した結果(A)、ならびに、培養上清のVLDLについて、Large、Medium、Smallの各サイズの中性脂肪(トリグリセリド)含量について解析した結果(B)を示す。
・細胞の調製
上記(1-1)にて記載した「細胞の調製」の方法において、培地B(DMSO(+))での培養を6日間とした以外は、同様にしてPXB-cellsを調製し、下記試験に用いた。
被験物質として、高脂血症治療薬として知られるロミタピド(東京化成)を用いた。
上記調製したPXB-cellsの培地を、ロミタピドを1nM、5nMの量でそれぞれ含む培地Dに交換し、インキュベーター(37℃,5%CO2)へ静かに移し、2日間培養した。培養終了後、上記「培養上清中のリポタンパク質の解析」及び「細胞中のリポタンパク質の解析」に記載の手法にて、培養上清中及び細胞中のコレステロール及び中性脂肪(トリグリセリド)をそれぞれ測定した。
対照には、ロミタピドの代わりに、エタノール(5μL)のみを添加した。
図8に、ロミタピド添加して、2日間培養したPXB-cellsの培養上清におけるリポタンパク質の各サブグループにおける中性脂肪(トリグリセリド)含量について解析した結果(A)、ならびに、培養上清のVLDLについて、Large、Medium、Smallの各サイズの中性脂肪(トリグリセリド)含量について解析した結果(B)を示す。
・細胞の調製
上記(1-1)にて記載した「細胞の調製」の方法と同様にしてPXB-cellsを調製し、下記試験に用いた。
被験物質として、高脂血症治療薬と知られるフェノフィブラート(sigma-aldrich)を用いた。
対照には、フェノフィブラートの代わりに、エタノール(5μL)のみを添加した。
図9に、フェノフィブラート添加して、2日間培養したPXB-cellsの培養上清におけるリポタンパク質の各サブグループにおける中性脂肪(トリグリセリド)含量について解析した結果(A)、ならびに、培養上清のVLDLについて、Large、Medium、Smallの各サイズの中性脂肪(トリグリセリド)含量について解析した結果(B)を示す。
Claims (6)
- 培養上清中の超低密度リポタンパク質(VLDL)の増減を指標にして、脂質異常に効果を有する物質のスクリーニング方法に使用するための、ヒト脂肪肝モデル細胞であって、
該細胞の培養上清がVLDL及び低密度リポタンパク質(LDL)を含有し、該VLDLが該LDLよりも多く含むことを特徴とする、細胞。 - 単層細胞培養物である、請求項1に記載の細胞。
- 前記VLDLが、Large-VLDL、Medium-VLDL、及びSmall-VLDLを含み、該Large-VLDLをVLDL全体の70質量%以上の割合で含む、請求項1又は2に記載の細胞。
- 脂肪肝由来のヒト肝細胞を、ジメチルスルホキシドを含む培地中にて培養して得られた細胞である、請求項1~3のいずれか一項に記載の細胞。
- 脂肪肝由来のヒト肝細胞が、ヒト肝細胞を有するキメラ非ヒト動物より回収されたものである、請求項4に記載の細胞。
- 脂質異常に効果を有する物質のスクリーニング方法であって、
請求項1~5のいずれか1項に記載の細胞に被検物質を投与する工程、ならびに、
被験物質を投与した細胞と投与されていない細胞との間で培養上清中の超低密度リポタンパク質(VLDL)の量を比較する工程、を含む方法。
