低pH且つ高スクロース環境下で良好に生育する乳酸菌を提供する。受託番号:NITE BP-03116(識別の表示:WR16-4)で寄託されているラクトバチルス属に属する種(Lactobacillus sp.)である乳酸菌。
Description
本発明は、新規な乳酸菌及びその取得方法並びに前記乳酸菌を含有する乳酸菌含有飲食品に関するものである。
一般的な乳酸菌の最適生育pH領域はpH6~7であり、酸性領域での生育限界値はpH4、アルカリ性領域での生育限界値はpH8と言われている。しかしながら、乳酸菌の中には優れた耐酸性を有し、一般的な乳酸菌では生育が難しい苛酷な環境下(pH3前後)でも生育可能なものもある(例えば特許文献1,2)。
また、味噌や醤油などの塩分濃度が高い環境下でも生育が可能な優れた耐塩性を有する乳酸菌もある(例えば特許文献3,4)。
このように乳酸菌の中には過酷な環境下でも生育可能なものが多く存在するが、低pH(高酸性)且つ高スクロース濃度という苛酷な環境下で良好に生育することができる乳酸菌の存在については本出願人の知る限りこれまで報告されていない。
そこで、本出願人は、このような乳酸菌の存在により、例えば、植物と糖を混合し圧搾して得られる液やこの液を基にした飲料、或いはジャム、フルーツソースなどの酸性値及びスクロース濃度が高く、一般的な乳酸菌では発酵が難しい素材を容易に発酵させることができ、新たな発酵食品や発酵飲料の提供が可能となると考え、上述のような低pH環境且つ高スクロース環境下で良好に生育することができる乳酸菌について研究を進め、ついに低pH且つ高スクロース濃度という苛酷な環境下で良好に生育することができる乳酸菌の取得に成功した。
本発明は、低pH且つ高スクロース環境下で良好に生育することができる乳酸菌を提供することを目的とする。

また、請求項1~10いずれか1項に記載の乳酸菌において、pH3.5~5.5の低pH環境下で増殖し、且つスクロース濃度30~60wt%の高濃度スクロース環境下でも増殖し、さらに、pH3.5~5.5の低pH環境下で増殖し、且つフルクトース濃度及びグルコース濃度が夫々10~30wt%のフルクトース・グルコース高濃度環境下でも増殖することを特徴とする乳酸菌に係るものである。
また、滅菌した液体培地に植物発酵エキスを加えて培養液を得、続いて、この培養液を集積培養して集積培養液を得、続いて、この集積培養液を滅菌した分離用培地と混釈して平板培地を作成し、続いて、この平板培地に出現したコロニーを釣菌して低pH環境且つ高濃度スクロース環境下で増殖可能な乳酸菌を取得することを特徴とする乳酸菌の取得方法に係るものである。
また、請求項14記載の乳酸菌の取得方法において、前記植物発酵エキスは、大根と上白糖と菌液とを混合し発酵させた大根発酵エキスであることを特徴とする乳酸菌の取得方法に係るものである。
また、請求項15記載の乳酸菌の取得方法において、前記菌液は酵母菌からなるものであることを特徴とする乳酸菌の取得方法に係るものである。
また、請求項14~16いずれか1項に記載の乳酸菌の取得方法において、前記液体培地は、野菜発酵エキス及び果物発酵エキスのいずれか一方若しくは双方を用いて得られた混合発酵エキスと、野草の煮出液と上白糖とを混合して得られた野菜・上白糖混合液とを混合したものに抗生物質を添加したものであることを特徴とする乳酸菌の取得方法に係るものである。
また、請求項17記載の乳酸菌の取得方法において、前記抗生物質はシクロヘキシミドであることを特徴とする乳酸菌の取得方法に係るものである。

本発明の乳酸菌は、低pH且つ高スクロース環境下で優位に生育(増殖)するから、例えば、植物と糖を混合し圧搾して得られる液やこの液を基にした飲料、或いはジャム、フルーツソースなどの酸性値及びスクロース濃度が高く、一般的な乳酸菌では発酵が難しい素材を容易に発酵させることができ、新たな発酵食品や発酵飲料の提供が可能となる。
好適と考える本発明の実施形態を、図面に基づいて本発明の作用を示して簡単に説明する。
本発明の乳酸菌は、ラクトバチルス属に属する種(Lactobacillus sp.)であり、受託番号:NITE BP-03116(識別の表示:WR16-4)で寄託されている。
本発明の乳酸菌から得られた配列データ(16SrDNA配列)をもとにNCBI(National Center for Biotechnology Information)の遺伝子データベースにて相同性検索を行った結果、前記配列と相同性が最も高かったもので93%であり(図12参照)、この結果から本発明の乳酸菌は従来にない新種の乳酸菌(ラクトバチルス属に属する種)と言える。
本発明の乳酸菌は、一般的な乳酸菌の最適生育環境とされるpH6~7では生育し難く、これよりも低いpH(例えばpH3.5~5.5)の環境下で良好に生育する特徴を有する。
さらに、本発明の乳酸菌は、一般的な乳酸菌では生育し難い低pH(例えばpH5以下)で且つ高スクロース(例えば30wt%以上)環境下でも良好に生育する特徴を有する。
このように、本発明の乳酸菌は、低pHで且つ高スクロース環境下で優位に増殖する特徴を有するから、例えば、植物と糖を混合し圧搾して得られる液やこの液を基にした飲料、或いはジャム、フルーツソースなどの酸性値及びスクロース濃度が高く、一般的な乳酸菌では発酵が難しい素材を容易に発酵させることができるものとなる。
本発明の具体的な実施例について図面に基づいて説明する。
本実施例の乳酸菌は、配列番号1に示す16SrDNA領域の塩基配列を有し、受託番号:NITE BP-03116(識別の表示:WR16-4)で寄託されているラクトバチルス属に属する種(Lactobacillus sp.)の乳酸菌である。
本実施例の乳酸菌は、細胞形態が短桿状であり、また、コロニーの状態では白色をしていて、菌体外多糖(粘性物質)を生産する。
以下、本実施例の乳酸菌(以下、WR16-4株という。)について詳述する。
<WR16-4株の菌種について>
WR16-4株の16SrDNAデータをもとに微生物同定検査を行い、WR16-4株の菌種について調査した。なお、微生物同定検査は株式会社ファスマックに依頼した。

微生物同定検査の結果、最も高い相同値を示した菌種は、Lactobacillus kefiri(ATCC=35441)であった(相同値93.14%)。
<WR16-4株の新規性について>
WR16-4株の16SrDNAデータをもとにNCBIの遺伝子データベースにて相同性検索を行い、WR16-4株の新規性について調査した。

図12はこの相同性検索においてサンプル由来配列と高い相同性を示したデータの一覧表である。この図12に示すように、このWR16-4株と相同性が最も高かったものでも93%程度であった。一般的に、相同値が98.7%以上の種がない場合は、新種と判断されることから、このWR16-4株は新規(新種)の乳酸菌であると言える。
<WR16-4株の分離源>
WR16-4株は植物発酵エキス、具体的には、大根発酵エキスを分離源とするものである。

分離源となる大根発酵エキスは、大根、上白糖及び菌液を混合し発酵させたものであり、具体的には、裁断した大根と、この大根と同量の上白糖とを混合し、これに酵母菌を発酵させた後、圧搾して得られた液分をさらに発酵、熟成させたものである。なお、分離源となる植物は大根以外の野菜でも良い。
<WR16-4株の分離方法>
WR16-4株は、上記大根発酵エキスから分離する際、一般的な乳酸菌の分離方法では分離できず、本出願人が見出した以下の分離方法により分離することができる。

(1) 湿式滅菌(121℃,15分)した液体培地10mlに上記の大根発酵エキス80μlを加え、25℃で6日間保温し培養液を得る。
(2) 得られた培養液80μlを上記の液体培地10mlに加え、再び25℃で3~4日間保温し培養液を得る。
(3) 上記2の処理を数回(本実施例では8回)繰り返す集積培養を行い、集積培養液を得る。
(4) 得られた集積培養液1mlを湿式滅菌(121℃,15分)した分離用培地15mlと混釈した後、冷却固化して平板を作成する。
(5) この平板を25℃で10日間保温する。
(6) 上記平板に出現したコロニーを釣菌することで、WR16-4株が得られる。
なお、上記液体培地は、野菜発酵エキス及び果物発酵エキスのいずれか一方若しくは双方を用いて得られた混合発酵エキスと、野草の煮出液と上白糖とを混合して得られた野草・上白糖混合液とを混合したものに抗生物質を添加したもの、具体的には、上記の混合発酵エキスと、複数の野草類の煮出液に上白糖を1:1で混合して得た野草・上白糖混合液と、1g/Lシクロヘキシミド溶液を、10:90:1で混合した混合液である。
また、上記分離用培地は、下表に示す培地A及び培地Bを湿式滅菌し混合したものである。
上記分離方法により取得したWR16-4株の特性を確認するため、一般的な乳酸菌を用い、これらとの比較によるpH耐性試験及びスクロース耐性試験を行った。
具体的には、WR16-4株の比較対象の乳酸菌として、乳酸菌の中では広く知られている属に属する種であり、一般的な乳酸菌と同様、最適生育pH領域をpH6~7とする、ラクトバチルス(Lactobacillus)属のLactobacillus acidophilus JCM 1132T、ラクトコッカス(Lactococcus)属のLactococcus lactis subsp. lactis NBRC12007、ロイコノストック(Leuconostoc)属のLeuconostoc mesenteroides subsp. dextranicum NBRC100495Tの3種の乳酸菌を用いた。
<各乳酸菌の前培養>
上記2種の耐性試験を行うにあたり、各乳酸菌に対して以下のような前培養を行った。

(1) 図9に示すMRS培地を2倍濃縮で調整した。この際、4N水酸化ナトリウム溶液又は4N塩酸を添加することにより、WR16-4株用のものはpH4.5に、比較対象用のものはpH6.5に調整した。
(2) 4wt%スクロース溶液又は4wt%グルコース溶液を調整した。
(3) 上記(1),(2)について、それぞれオートクレーブを用いた湿熱滅菌(121℃,20分)を行った。
(4) 湿熱滅菌した(1)及び(2)の溶液を冷却した後、クリーンベンチ内で1:1(v/v)となるように混合し、2wt%スクロース含有MRS培地(pH4.5)又は2wt%グルコース含有MRS培地(pH6.5)を調整した。
(5) WR16-4株は2wt%スクロース含有MRS培地(pH4.5)、比較対象用は2wt%グルコース含有MRS培地(pH6.5)を用いて、30℃で24~48時間の嫌気条件で行った。
<pH耐性試験>
前培養した各乳酸菌のpH耐性試験を以下のようにして行った。

(1) 図9に示すMRS培地を2倍濃縮で調整した。この際、4N水酸化ナトリウム溶液又は4N塩酸を添加することにより、pH3,3.5,4,4.5,5,5.5,6,6.5に調整した。
(2) 4wt%スクロース溶液を調整した。
(3) 上記(1),(2)について、それぞれオートクレーブを用いた湿熱滅菌(121℃,20分)を行った。
(4) 湿熱滅菌した(1)及び(2)の溶液を冷却した後、クリーンベンチ内で1:1(v/v)となるように混合し、2wt%スクロース含有MRS培地(pH3~6.5)を調整した。
(5) 前培養した各乳酸菌株を遠心分離(15,000×g,10分)して上清を取り除いた後、(3)で調整した培地に初期濃度がA660nm≒0.05~0.10となるように再懸濁した。
(6) 96穴マイクロプレートに200μl/wellとなるように、各3wellずつ分注した後(n=3)、30℃、嫌気条件で培養した。
(7) マイクロプレートリーダーを用いて経時的に濁度測定を行った。
図1~3は比較対象用(Lactobacillus acidophilus JCM 1132T(図1)、Lactococcus lactis subsp. lactis NBRC12007(図2)、Leuconostoc mesenteroides subsp. dextranicum NBRC100495T(図3))の乳酸菌の濁度測定結果を示すグラフであり、図4はWR16-4株の濁度測定結果を示すグラフである。
上記2wt%スクロースを炭素源としたpH耐性試験においては、各乳酸菌の濁度測定結果に示されるように、比較対象用の乳酸菌はpH5~6.5の中性に近い酸性領域で生育が良好であったのに対し、WR16-4株はこれらの乳酸菌が良好に生育する領域よりもより酸性側の領域、具体的には、pH3.5~5の低pH領域で生育し、これよりpHが高い中性に近い領域ではほとんど生育しないことが確認された。
<スクロース耐性試験>
前培養した各乳酸菌のスクロース耐性試験を以下のようにして行った。

(1) スクロースを添加した図9に示すMRS培地を、スクロース濃度が5wt%,10wt%,20wt%,30wt%,40wt%,50wt%,60wt%,70wt%となるように調整した後、それぞれに対して4N塩酸を添加しpH4.5となるように調整した。
(2) 上記(1)について、それぞれオートクレーブを用いた湿熱滅菌(121℃,20分)を行った。
(3) 前培養した各乳酸菌株を遠心分離(15,000×g,10分)して上清を取り除いた後、(2)で調整した培地に初期濃度がA660nm≒0.05~0.10となるように再懸濁した。
(4) 96穴マイクロプレートに200μl/wellとなるように、各3wellずつ分注した後(n=3)、30℃、嫌気条件で培養した。
(5) マイクロプレートリーダーを用いて経時的に濁度測定を行った。
図5~8は比較対象用(Lactobacillus acidophilus JCM 1132T(図5)、Lactococcus lactis subsp. lactis NBRC12007(図6)、Leuconostoc mesenteroides subsp. dextranicum NBRC100495T(図7))の乳酸菌の濁度測定結果を示すグラフであり、図8はWR16-4株の濁度測定結果を示すグラフである。
上記初期pH4.5の酸性条件下でのスクロース耐性試験においては、各乳酸菌の濁度測定結果に示されるように、比較対象用の乳酸菌はスクロース濃度が20wt%以下での生育が確認されたもののその生育状態(増殖率)は低く、30wt%以上の高スクロース環境ではほぼ生育できない結果であったのに対し、WR16-4株はスクロース濃度40wt%程度までは良好な生育状態を示し、60wt%の高スクロース環境でも生育することが確認された。
<フルクトース・グルコース耐性試験>
スクロースは、単糖のフルクトースとグルコースとが結合した二糖類であり、WR16-4株は上記のように高スクロース環境で生育可能なことが確認されたことから、フルクトース及びグルコースが夫々単糖状態で存在するフルクトース・グルコース高濃度環境下でも、スクロースの場合と同様に生育可能であるのではないかと考え、上記スクロース耐性試験と同様に、フルクトース・グルコース耐性試験を以下のようにして行った。
スクロースは、単糖のフルクトースとグルコースとが結合した二糖類であり、WR16-4株は上記のように高スクロース環境で生育可能なことが確認されたことから、フルクトース及びグルコースが夫々単糖状態で存在するフルクトース・グルコース高濃度環境下でも、スクロースの場合と同様に生育可能であるのではないかと考え、上記スクロース耐性試験と同様に、フルクトース・グルコース耐性試験を以下のようにして行った。
(1) スクロース、フルクトース、グルコースの糖を夫々単独で添加した図10に示すMRS培地を、夫々糖濃度が20wt%となるように調整した後、それぞれに対して4N塩酸を添加しpH4.5となるように調整した。
(2) 別途、フルクトース及びグルコースを添加した図10に示すMRS培地を、フルクトース、グルコース夫々の糖濃度が10wt%(合計で20wt%)となるように調整した後、それぞれに対して4N塩酸を添加しpH4.5となるように調整した。
(3) 上記(1),(2)について、それぞれオートクレーブを用いた湿熱滅菌(121℃,20分)を行った。
(4) 前培養したWR16-4株を遠心分離(15,000×g,10分)して上清を取り除いた後、調整した培地に初期濃度がA660nm≒0.05~0.10となるように再懸濁した。
(5) 96穴マイクロプレートに200μl/wellとなるように、各3wellずつ分注した後(n=3)、30℃、嫌気条件で培養した。
(6) マイクロプレートリーダーを用いて経時的に濁度測定を行った。
この初期pH4.5の酸性条件下でのフルクトース・グルコース耐性試験における上記濁度測定の結果を図11に示す。
この図11に示されるように、WR16-4株は、フルクトース、グルコースのいずれかが単独で含まれている場合は増殖率が低いが(低生育状態であるが)、フルクトース、グルコースの両方が含まれる場合は、スクロースを含む場合と同様、増殖率が高くなり(高生育状態になり)、この結果から、WR16-4株は、スクロース耐性だけでなく、フルクトース・グルコース耐性も有することが確認された。
また、前述したようにスクロースはフルクトースとグルコースが結合した二糖類であることから、WR16-4株は、スクロースの場合と同様、例えば、フルクトースとグルコースを夫々30wt%ずつ含む(糖濃度の合計が60wt%)高糖濃度環境でも十分生育可能と考える。
<WR16-4株の用途>
WR16-4株は乳酸菌含有飲食品、具体的には、発酵飲食品に用いることができる。

WR16-4株は、上述したように、低pHで且つ高スクロース環境下で優位に増殖する特徴を有する乳酸菌であるから、例えば、植物と糖を混合し圧搾して得られる液やこの液を基にした飲料、或いはジャム、フルーツソースなどの酸性値及びスクロース濃度が高く、一般的な乳酸菌では発酵が難しい素材(酸性且つ高糖濃度抽出液)を容易に発酵させることができ、これにより前記抽出液に有機酸を含有させることができ、酸味の付与(甘さの軽減やさわやかさの付与)や防腐(添加物としての保存料が不要になる)などの効果が得られる。
また、WR16-4株は、低pHで且つ高フルクトース・グルコース環境下でも優位に増殖する特徴を有する乳酸菌であるから、上記の他に、フルクトースとグルコースの両方を含む糖(異性化糖、ブドウ糖及び果糖の夫々の精製糖を混合したものなど)を用いた飲食品にも用いることができる。
また、上記酸性且つ高糖濃度抽出液の取得方法の一例を以下に示す。なお、抽出液の取得方法はこれに限定されるものではない。
(1) 裁断した野菜や果物と上白糖とを同比率で混合し、菌液を加えて発酵させる。
(2) 発酵後、圧搾して液分を回収し、この回収液をさらに発酵・熟成させて発酵液Aを得る。
(3) 野草の煮出し液に上白糖を加えたものに発酵液Aを加え、発酵・熟成させて発酵液Bを得る。
この発酵液Aや発酵液Bが酸性且つ高糖濃度抽出液となる。なお、この発酵液A及び発酵液Bは飲料として製造されるものであり、WR16-4株によって発酵させることで、酸味を付与すると共に、保存料を添加することなく防腐効果が得られ、これまでにない飲料の提供が可能となる。
また、WR16-4株は、上記試験結果に示すように、一般的な乳酸菌の最適pH環境であるpH6~7の環境下でほとんど生育しない特性を有するから、例えば、スクロース濃度が高くてpHが中性付近の素材を、WR16-4株と他の微生物(例えば一般的な乳酸菌や酵母など)で発酵させようとする場合、WR16-4株は至適pHに到達するまでの間は生育しないので、他の微生物の生育を阻害しない。よって、複数種の微生物による複合的な発酵を利用する場合に有用となる。
また、上記特性から、WR16-4株は、素材のpHを中性付近に調整することで発酵を非加熱的に停止させることができ、加熱できない発酵製品への利用も有用となる。
なお、本発明は、本実施例に限られるものではなく、各構成要件の具体的構成は適宜設計し得るものである。

- 受託番号:NITE BP-03116(識別の表示:WR16-4)で寄託されているラクトバチルス属に属する種(Lactobacillus sp.)であることを特徴とする乳酸菌。
- 請求項1記載の乳酸菌において、前記乳酸菌は配列番号1に示される16SrDNA領域の塩基配列を有することを特徴とする乳酸菌。
- 請求項1記載の乳酸菌において、植物発酵エキスから取得されたものであることを特徴とする乳酸菌。
- 請求項2記載の乳酸菌において、植物発酵エキスから取得されたものであることを特徴とする乳酸菌。
- 請求項3記載の乳酸菌において、前記植物発酵エキスは大根発酵エキスであることを特徴とする乳酸菌。
- 請求項4記載の乳酸菌において、前記植物発酵エキスは大根発酵エキスであることを特徴とする乳酸菌。
- 請求項5記載の乳酸菌において、前記大根発酵エキスは大根、上白糖及び菌液を混合し発酵させたものであることを特徴とする乳酸菌。
- 請求項6記載の乳酸菌において、前記大根発酵エキスは大根、上白糖及び菌液を混合し発酵させたものであることを特徴とする乳酸菌。
- 請求項7記載の乳酸菌において、前記菌液は酵母菌からなるものであることを特徴とする乳酸菌。
- 請求項8記載の乳酸菌において、前記菌液は酵母菌からなるものであることを特徴とする乳酸菌。
- 請求項1~10いずれか1項に記載の乳酸菌において、pH3.5~5.5の低pH環境下で増殖し、且つスクロース濃度30~60wt%の高濃度スクロース環境下でも増殖することを特徴とする乳酸菌。
- 請求項1~10いずれか1項に記載の乳酸菌において、pH3.5~5.5の低pH環境下で増殖し、且つフルクトース濃度及びグルコース濃度が夫々10~30wt%のフルクトース・グルコース高濃度環境下でも増殖することを特徴とする乳酸菌。
- 請求項1~10いずれか1項に記載の乳酸菌において、pH3.5~5.5の低pH環境下で増殖し、且つスクロース濃度30~60wt%の高濃度スクロース環境下でも増殖し、さらに、pH3.5~5.5の低pH環境下で増殖し、且つフルクトース濃度及びグルコース濃度が夫々10~30wt%のフルクトース・グルコース高濃度環境下でも増殖することを特徴とする乳酸菌。
- 滅菌した液体培地に植物発酵エキスを加えて培養液を得、続いて、この培養液を集積培養して集積培養液を得、続いて、この集積培養液を滅菌した分離用培地と混釈して平板培地を作成し、続いて、この平板培地に出現したコロニーを釣菌して低pH環境且つ高濃度スクロース環境下で増殖可能な乳酸菌を取得することを特徴とする乳酸菌の取得方法。
- 請求項14記載の乳酸菌の取得方法において、前記植物発酵エキスは、大根と上白糖と菌液とを混合し発酵させた大根発酵エキスであることを特徴とする乳酸菌の取得方法。
- 請求項15記載の乳酸菌の取得方法において、前記菌液は酵母菌からなるものであることを特徴とする乳酸菌の取得方法。
- 請求項14~16いずれか1項に記載の乳酸菌の取得方法において、前記液体培地は、野菜発酵エキス及び果物発酵エキスのいずれか一方若しくは双方を用いて得られた混合発酵エキスと、野草の煮出液と上白糖とを混合して得られた野菜・上白糖混合液とを混合したものに抗生物質を添加したものであることを特徴とする乳酸菌の取得方法。
- 請求項17記載の乳酸菌の取得方法において、前記抗生物質はシクロヘキシミドであることを特徴とする乳酸菌の取得方法。
- 請求項1~10いずれか1項に記載の乳酸菌を含有することを特徴とする乳酸菌含有飲食品。
- 請求項11記載の乳酸菌を含有することを特徴とする乳酸菌含有飲食品。
- 請求項12記載の乳酸菌を含有することを特徴とする乳酸菌含有飲食品。
- 請求項13記載の乳酸菌を含有することを特徴とする乳酸菌含有飲食品。
- 請求項19記載の乳酸菌含有飲食品において、前記乳酸菌含有飲食品は発酵飲食品であることを特徴とする乳酸菌含有飲食品。
- 請求項20記載の乳酸菌含有飲食品において、前記乳酸菌含有飲食品は発酵飲食品であることを特徴とする乳酸菌含有飲食品。
- 請求項21記載の乳酸菌含有飲食品において、前記乳酸菌含有飲食品は発酵飲食品であることを特徴とする乳酸菌含有飲食品。
