本発明は、徐放性基剤等に関する。
徐放性製剤は、投与後に長時間にわたり薬物が少しずつ体内で放出される製剤であり、速放性製剤に比べて投与回数を減らすことができるという特徴を有している。これにより患者の負担軽減およびコンプライアンスの向上が期待できるため、国内外で広く用いられている。
徐放性製剤に用いられる基剤としては、様々なポリマーが用いられており、ポリビニルアルコール(以下、PVAとも略記する)が使用された例もある。
例えば、特許文献1及び2には、PVAやPVA誘導体と微結晶セルロース(以下、MCCとも略記する)の混合物を直接圧縮することで優れた成形性と徐放性を示す製剤が得られることが開示されている。
本発明の目的は、新規な徐放性基剤を提供することにある。
本発明の他の目的は、新規な製剤を提供することにある。
本発明のさらに他の目的は、新規な製剤の製造方法を提供することにある。
特許文献1及び2に記載の製剤は、成形性や徐放性が向上しているとはいえ、その効果は十分ではない場合があった。
本発明者らは、上記課題を解決するために鋭意研究を重ねた結果、PVAと低置換度ヒドロキシプロピルセルロースを組み合わせることにより、徐放性製剤の基剤として機能できること等を見出し、さらに研究を重ねて本発明を完成した。
すなわち、本発明は、次の発明などに関する。

[徐放性基剤]
本発明の徐放性基剤は、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを組み合わせている。
本発明の徐放性基剤は、これら両成分が組み合わせられていればよく、単一の基剤中に両成分を含有していてもよいし、複数基剤の組み合わせ全体として両成分を含有するように組み合わせてなる基剤であってもよい。換言すれば、複数基剤を組み合わせてなる系は、低置換度ヒドロキシプロピルセルロースと組み合わせるための、ポリビニルアルコール系重合体を含む徐放性基剤であってもよい。
本発明の徐放性基剤は、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを組み合わせている。
本発明の徐放性基剤は、これら両成分が組み合わせられていればよく、単一の基剤中に両成分を含有していてもよいし、複数基剤の組み合わせ全体として両成分を含有するように組み合わせてなる基剤であってもよい。換言すれば、複数基剤を組み合わせてなる系は、低置換度ヒドロキシプロピルセルロースと組み合わせるための、ポリビニルアルコール系重合体を含む徐放性基剤であってもよい。
(ポリビニルアルコール系重合体)
ポリビニルアルコール系重合体(PVA系重合体、PVAなどということがある)は、通常、ビニルエステル系重合体(少なくともビニルエステルを重合成分とする重合体)のけん化物であってもよい。
ポリビニルアルコール系重合体(PVA系重合体、PVAなどということがある)は、通常、ビニルエステル系重合体(少なくともビニルエステルを重合成分とする重合体)のけん化物であってもよい。
PVA系重合体の平均けん化度は、製剤における低置換度ヒドロキシプロピルセルロースの崩壊性の発現を抑制しやすくなるためか、製剤が徐放性効果をより発揮しやすい等の観点から、例えば、90.0モル%以下(例えば、89.5モル%以下)、好ましくは89.0モル%以下(例えば、88.5モル%以下、88.0モル%以下)等であってもよい。
また、PVA系重合体の平均けん化度の下限値は、例えば、60.0モル%以上(例えば、62.0モル%以上)、好ましくは65.0モル%以上(例えば、68.0モル%以上、70.0モル%以上、75.0モル%以上、80.0モル%以上)等であってもよい。
また、PVA系重合体の平均けん化度の下限値は、例えば、60.0モル%以上(例えば、62.0モル%以上)、好ましくは65.0モル%以上(例えば、68.0モル%以上、70.0モル%以上、75.0モル%以上、80.0モル%以上)等であってもよい。
なお、PVA系重合体の平均けん化度は、これらの上限値と下限値とを適宜組み合わせて適当な範囲(例えば、60.0モル%以上90.0モル%以下等)を設定してもよく(他も同じ)、上記上限値と下限値の全ての組み合わせが含まれる。
なお、PVA系重合体の平均けん化度は、特に限定されないが、例えば、JIS K6726のけん化度測定方法などによって、測定してもよい。
PVA系重合体の平均重合度は、製剤における低置換度ヒドロキシプロピルセルロースの崩壊性の発現を抑制しやすくなるためか、製剤が徐放性効果をより発揮しやすい等の観点から、例えば、800以上(例えば、900以上、1000以上、1100以上、1200以上、1300以上、1400以上)、好ましくは1500以上(例えば、1600以上)、より好ましくは2000以上(例えば、2100以上、2400以上)等であってもよい。
また、PVA系重合体の平均重合度の上限値は、例えば、5000以下(例えば、4900以下)、好ましくは4500以下(例えば、4400以下)、より好ましくは3500以下(例えば、3400以下)等であってもよい。

また、PVA系重合体の4質量%水溶液粘度(JIS K6726に従って測定)の上限値は、例えば、300mPa・s以下(例えば、280mPa・s以下)、好ましくは240mPa・s以下(例えば、220mPa・s以下)、より好ましくは100mPa・s以下(例えば、80mPa・s以下)等であってもよい。
また、PVA系重合体の4質量%水溶液粘度(JIS K6726に従って測定)の上限値は、例えば、300mPa・s以下(例えば、280mPa・s以下)、好ましくは240mPa・s以下(例えば、220mPa・s以下)、より好ましくは100mPa・s以下(例えば、80mPa・s以下)等であってもよい。

PVA系重合体は、1種で又は2種以上組み合わせて使用してもよい。
PVA系重合体は、市販品を利用してよく、合成したものを使用してもよい。PVA系重合体の製造方法としては、ビニルエステル系モノマーを含む重合成分の重合体(ビニルエステル系重合体)をけん化する等の公知の方法が用いられる。
ビニルエステル系モノマー(ビニルエステル系単量体)としては、特に限定されないが、例えば、脂肪酸ビニルエステル[例えば、ギ酸ビニル、酢酸ビニル、プロピオン酸ビニル、酪酸ビニル、カプリル酸ビニル、バーサチック酸ビニル、モノクロロ酢酸ビニル等のC1-20脂肪酸ビニルエステル(例えば、C1-16アルカン酸-ビニルエステル)等]、芳香族カルボン酸ビニルエステル[例えば、安息香酸ビニルなどのアレーンカルボン酸ビニル(例えば、C7-12アレーンカルボン酸-ビニルエステル)等]等が挙げられる。
ビニルエステル系モノマーは、1種で又は2種以上組み合わせて使用してもよい。
ビニルエステル系モノマーは、少なくとも脂肪酸ビニルエステル(例えば、ギ酸ビニル、酢酸ビニル、プロピオン酸ビニル、酪酸ビニル等のC1-10アルカン酸-ビニルエステルなど)を含んでいるのが好ましく、工業的観点等から、特に、酢酸ビニルを含んでいてもよい。
ビニルエステル系重合体は、ビニルエステル単位を有していればよく、必要に応じて、他の単量体(ビニルエステル系モノマーと共重合可能な単量体)由来の単位を有していてもよい(他の単量体により変性されていてもよい)。

重合成分におけるビニルエステル系モノマーの割合は、例えば、50質量%以上、好ましくは70質量%以上、さらに好ましくは90質量%以上であってもよく、100質量%であってもよい。
なお、PVA系重合体は、ビニルアルコール単位の一部が、アセタール化、エーテル化、アセトアセチル化、カチオン化などの反応によって、変性されたものであってもよい。
重合成分(例えば、酢酸ビニル等のビニルエステル系モノマーを含む重合成分)の重合方法としては、特に限定されず、例えば、従来公知の塊状重合、溶液重合、懸濁重合、乳化重合等が挙げられるが、溶剤としてメタノールを用いた溶液重合が工業的に好ましい。該溶液重合には、過酸化物系、アゾ系等の公知の開始剤を用いることができ、重合成分とメタノールの配合比、重合収率を変えることにより、ビニルエステル系重合体の重合度を調整することができる。また、PVA系重合体を得るための原料として、市販のビニルエステル系重合体(ポリ酢酸ビニル樹脂など)を使用することもできる。
ビニルエステル系重合体(例えば、ポリ酢酸ビニル)のけん化方法としては、従来から公知のアルカリ触媒又は酸触媒を用いたけん化方法を適用することができ、中でもビニルエステル系重合体(例えば、ポリ酢酸ビニル)のメタノール溶液又はビニルエステル系重合体(例えば、ポリ酢酸ビニル)のメタノール、水、酢酸メチル等の混合溶液に水酸化ナトリウム等のアルカリを加えて、撹拌して混合しながら、ビニルエステル系重合体(例えば、ポリ酢酸ビニル)のアシル基(例えば、アセチル基)を加アルコール分解する方法が、工業的に好ましい。
その後、得られた塊状物、ゲル状物あるいは粒状物を粉砕し、必要に応じて添加したアルカリを中和した後、固形物と液分を分離し、固形物を乾燥することによりPVA系重合体を得ることができる。
低置換度ヒドロキシプロピルセルロースは、置換度(エーテル化度)が、例えば、0.05~1.0(例えば、0.07~0.8)、好ましくは0.1~0.6(例えば、0.15~0.5)程度であってよい。
なお、置換度は、低置換度ヒドロキシプロピルセルロースにおいて、グルコース単位あたりの置換された水酸基の平均数であってよい。

本発明の徐放性基剤において、ポリビニルアルコール系重合体と低置換度ヒドロキシプロピルセルロースの配合割合は、製剤の徐放性や成形性等の観点から、ポリビニルアルコール系重合体:低置換度ヒドロキシプロピルセルロース(質量比)が、例えば、30:70~90:10(例えば、33:67~87:13)、好ましくは35:65~85:15(例えば、40:60~80:20)、より好ましくは43:57~78:22(例えば、45:55~75:25)程度であってもよい。
本発明の徐放性基剤の対象物としては、特に限定されないが、例えば、医薬品、医薬部外品、食品、農薬等であってもよい。また、対象物は、有効成分、栄養素(又は栄養成分)等であってもよく、薬物(原薬等)であってもよい。

これら添加剤は、1種又は2種以上を使用することができる。他の成分(添加剤)の含有量は、薬剤の種類等に応じて適宜決定することができる。
対象物は、単独で又は2種以上組み合わせて使用してもよい。

なお、粉末の平均粒子径は、レーザー式粒度分布測定装置などにより測定してもよい。

本発明は、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを含む固形製剤をも含有する。なお、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースは、上記例示のものであってよい。また、製剤の対象物は、上記例示の対象物(徐放性基剤の対象物)であってよく、通常、粉体であってよい。
本発明は、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを含む固形製剤をも含有する。なお、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースは、上記例示のものであってよい。また、製剤の対象物は、上記例示の対象物(徐放性基剤の対象物)であってよく、通常、粉体であってよい。
本発明の製剤は、例えば、錠剤、顆粒剤、カプセル剤等であってよいが、錠剤が好ましい。
本発明の製剤は、経口剤として好適に使用してもよい。
本発明の製剤は、ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロース(さらに、製剤の対象物)以外に、他の成分を含んでいてもよい。
他の成分としては、通常、この分野で常用され得る添加剤(例えば、崩壊剤、滑択剤、凝集防止剤、溶解補助剤など)を含んでいてもよい。
崩壊剤としては、例えば、カルメロース又はその塩、クロスカルメロースナトリウム、カルボキシメチルスターチナトリウム、クロスポビドン、結晶セルロース、結晶セルロース・カルメロースナトリウム等が挙げられる。
滑択剤又は凝集防止剤としては、例えば、タルク、ステアリン酸マグネシウム、ステアリン酸カルシウム、コロイダルシリカ、ステアリン酸、ワックス類、硬化油、ポリエチレングリコール類、安息香酸ナトリウムなどが挙げられる。
溶解補助剤としては、例えば、フマル酸、コハク酸、リンゴ酸、アジピン酸などの有機酸などが挙げられる。
これらの添加剤は、1種又は2種以上を使用することができる。
また、これら添加剤の含有量は、製剤の成分の種類などに応じて適宜決定することができる。
製剤において、本発明の徐放性基剤の割合は、例えば、40~99.5質量%(例えば、42~99質量%)、好ましくは45~98.5質量%(例えば、50~98質量%)、より好ましくは52~97.5質量%(例えば、55~97質量%)程度であってもよい。
製剤において、ポリビニルアルコール系重合体の割合は、例えば、20~60質量%(例えば、22~58質量%)、好ましくは25~55質量%(例えば、27~52質量%)、より好ましくは30~50質量%(例えば、33~48質量%)程度であってもよい。
製剤において、低置換度ヒドロキシプロピルセルロースの割合は、例えば、5~60質量%(例えば、7~58質量%)、好ましくは10~55質量%(例えば、12~52質量%)、より好ましくは15~50質量%(例えば、33~48質量%)程度であってもよい。

なお、本発明の製剤の形状は、特に限定されないが、製剤が錠剤である場合、錠剤の形状は、例えば、円盤形、レンズ形、竿形などのいずれであってもよい。
また、本発明の製剤の大きさは、特に限定されないが、特に、製剤が錠剤である場合、錠剤の大きさは、例えば、直径(最大径)で、3mm以上(例えば、4~15mm、5~12mm、8~11mmなど)であってもよい。
錠剤は、ポリビニルアルコール系重合体、低置換度ヒドロキシプロピルセルロース及び製剤の対象物(又は、本発明の徐放性基剤及び製剤の対象物)を少なくとも含む混合物(打錠末)を打錠することによって、製造することができる。
打錠末は、上記他の成分を含んでいてもよい。
打錠末は、上記他の成分を含んでいてもよい。
打錠方法としては、特に限定されず、慣用の打錠方法を採用できるが、特に、直接圧縮法(直接打錠法)が好ましい。直接圧縮(直接打錠)は、打錠末をそのまま打錠機で圧縮成形することにより、実施することができる。
錠剤は、錠剤表面に被覆層を有していてもよい。
被覆錠剤において、被覆層の構成成分としては、特に限定されないが、例えば、セルロース系樹脂(例えば、ヒドロキシプロピルメチルセルロース、ヒドロキシプロピルメチルセルロースアセテートサクシネートなど)、ポリビニルアルコール系重合体などの樹脂材料が挙げられる。
本発明の製剤は、徐放性製剤であってもよい。
本発明の徐放性製剤は、活性成分(例えば、有効成分、栄養素、薬物等)の種類等もよるが、活性成分の溶出率が80%以上(例えば、80%)に到達する時間(溶出時間)が、少なくとも2時間以上であってもよく、好ましくは3時間以上(例えば、4時間以上、5時間以上、6時間以上、7時間以上等であってよい。
本発明の徐放性製剤において、活性成分の溶出率が80%以上(例えば、80%)に到達する時間の上限値は、特に限定されないが、例えば、20時間以下、19時間以下、18時間以下、17時間以下、16時間以下、15時間以下等であってもよい。

また、本発明の徐放性製剤は、活性成分の種類等によるが、活性成分の溶出率が50%に到達する時間が、例えば、20分以上、好ましくは30分以上(例えば、40分以上)、より好ましくは60分以上(例えば、80分以上)等であってもよい。
本発明の徐放性製剤において、活性成分の溶出率が50%に到達する時間の上限値は、特に限定されないが、例えば、20時間以下、19時間以下、18時間以下、17時間以下、16時間以下、15時間以下等であってもよい。

また、溶出時間の測定方法は、特に限定されないが、例えば、日本薬局方溶出試験第2法(パドル法)を使用してもよい。日本薬局方溶出試験第2法(パドル法)は、試験液が蒸留水900mL、パドル回転数が50rpmであってもよい。
以下、本発明について実施例をあげて具体的に説明するが本発明はこれらに限定されるものではない。
なお、以下の実施例及び比較例中、特にことわりのないかぎり、「%」及び「部」は質量基準を表す。

<錠剤硬度の評価>
得られた錠剤の硬度を、錠剤硬度計(TBH125、ERWEKA製)を用いて測定した。6回測定を行い、その平均値を測定値とした。

<溶出率の評価>
以下の試験条件下において溶出試験を実施した。また、溶出率が80%に到達した時間を測定、算出した。

測定サンプル:実施例および比較例で示される錠剤
検出波長:UV444nm(リボフラビン)

以下の実施例及び参考例で使用した各PVA系重合体(PVA1~3)を、表1に示す。
なお、表1に製品名が記載されたPVAは、日本酢ビ・ポバール製である。
また、いずれのPVA系重合体も、粉砕を行い、平均粒子径が約100μmになるように調整した。

また、PVA3の合成方法を、以下に示す。
(合成例1)
攪拌機、コンデンサー、窒素ガス導入口および開始剤投入口を備えた反応槽に、予めメタノール140部および酢酸ビニルモノマー860部を仕込み、系内の窒素置換を行った後60℃に加熱し、還流が発生した時点で、開始剤として2,2’-アゾビス2,4-ジメチルバレロニトリルの1.5%メタノール溶液を2.0部添加し、重合を開始した。重合中は系を70℃に保持し、系内に窒素ガスを流しつつ常圧とし、さらに2,2’-アゾビス2,4-ジメチルバレロニトリルの2%メタノール溶液2.0部を重合開始から15分後に加えた。重合開始から2.3時間後、酢酸ビニルの反応収率が52%になった時点で系を冷却し、重合を終了した。得られた反応物にメタノール蒸気を加えながら、残存する酢酸ビニルモノマーを留出し、ポリ酢酸ビニルの40%メタノール溶液を得た。
次に、上記で得られたポリ酢酸ビニルの40%メタノール溶液500部に、メタノール100部、水6部、水酸化ナトリウムの5.2%メタノール溶液6.3部を加えてよく混合し、40℃で40分間けん化反応を行い、得られたゲル状物を粉砕し、中和後にメタノールで洗浄した後に乾燥して、けん化度65.0モル%、平均重合度2400のPVA系重合体3(PVA3)を得た。
攪拌機、コンデンサー、窒素ガス導入口および開始剤投入口を備えた反応槽に、予めメタノール140部および酢酸ビニルモノマー860部を仕込み、系内の窒素置換を行った後60℃に加熱し、還流が発生した時点で、開始剤として2,2’-アゾビス2,4-ジメチルバレロニトリルの1.5%メタノール溶液を2.0部添加し、重合を開始した。重合中は系を70℃に保持し、系内に窒素ガスを流しつつ常圧とし、さらに2,2’-アゾビス2,4-ジメチルバレロニトリルの2%メタノール溶液2.0部を重合開始から15分後に加えた。重合開始から2.3時間後、酢酸ビニルの反応収率が52%になった時点で系を冷却し、重合を終了した。得られた反応物にメタノール蒸気を加えながら、残存する酢酸ビニルモノマーを留出し、ポリ酢酸ビニルの40%メタノール溶液を得た。
次に、上記で得られたポリ酢酸ビニルの40%メタノール溶液500部に、メタノール100部、水6部、水酸化ナトリウムの5.2%メタノール溶液6.3部を加えてよく混合し、40℃で40分間けん化反応を行い、得られたゲル状物を粉砕し、中和後にメタノールで洗浄した後に乾燥して、けん化度65.0モル%、平均重合度2400のPVA系重合体3(PVA3)を得た。
(実施例1)
表2に記載の配合割合で、ステアリン酸マグネシウム(St-Mg)以外の成分をポリエチレン袋に入れ、空気を入れて袋を膨らませた後、100回混合した(袋を振った)。その後、袋の中にSt-Mgを添加し、空気を入れて袋を膨らませた後、30回混合した(袋を振った)。得られた混合物を打錠し、錠剤を作製した。得られた錠剤について、錠剤硬度および溶出時間を測定した。結果を表2に示す。なお、表2において、L-HPCとは、低置換度ヒドロキシプロピルセルロースを示す。L-HPCとしては、NBD-021(信越化学工業株式会社)を使用した。
表2に記載の配合割合で、ステアリン酸マグネシウム(St-Mg)以外の成分をポリエチレン袋に入れ、空気を入れて袋を膨らませた後、100回混合した(袋を振った)。その後、袋の中にSt-Mgを添加し、空気を入れて袋を膨らませた後、30回混合した(袋を振った)。得られた混合物を打錠し、錠剤を作製した。得られた錠剤について、錠剤硬度および溶出時間を測定した。結果を表2に示す。なお、表2において、L-HPCとは、低置換度ヒドロキシプロピルセルロースを示す。L-HPCとしては、NBD-021(信越化学工業株式会社)を使用した。
(実施例2~8)
表2に記載のPVA系重合体を用い、各成分の種類及び配合割合を表2に記載のものとした以外は、実施例1と同様にして打錠を実施し、錠剤硬度及び溶出時間を測定した。結果を表2に示す。

(参考例1~2)
表2に記載のPVA系重合体を用い、各成分の種類及び配合割合を表2に記載のものとした以外は、実施例1と同様にして打錠を実施し、錠剤硬度及び溶出時間を測定した。結果を表2に示す。なお、表2において、Cl-PVPとは、架橋ポリビニルピロリドン(コリドンCL、BASFジャパン社)を示す。また、参考例1~2の溶出率の経時変化を図2に示す。
表2に記載のPVA系重合体を用い、各成分の種類及び配合割合を表2に記載のものとした以外は、実施例1と同様にして打錠を実施し、錠剤硬度及び溶出時間を測定した。結果を表2に示す。なお、表2において、Cl-PVPとは、架橋ポリビニルピロリドン(コリドンCL、BASFジャパン社)を示す。また、参考例1~2の溶出率の経時変化を図2に示す。
表2に示すように、実施例では、PVA系重合体と低置換度ヒドロキシプロピルセルロースを併用することにより、高い成形性と優れた徐放性を両立した製剤を作製することができた。
本発明によれば、新規な徐放性基剤を提供できる。このよう基剤は、徐放性製剤等を得るのに有用である。
Claims (11)
- ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを組み合わせた徐放性基剤。
- ポリビニルアルコール系重合体及び低置換度ヒドロキシプロピルセルロースを含む固形製剤。
- 徐放性製剤である請求項2記載の製剤。
- 経口剤である請求項2又は3記載の製剤。
- 製剤の対象物が薬物を含む請求項2~4のいずれか記載の製剤。
- ポリビニルアルコール系重合体が、JIS K6726に従って測定した4質量%水溶液粘度が15.0mPa・s以上を満たす請求項1~5のいずれか記載の剤又は製剤。
- ポリビニルアルコール系重合体の平均けん化度が65.0~90.0モル%である請求項1~6のいずれか記載の剤又は製剤。
- ポリビニルアルコール系重合体の平均重合度が1000以上である請求項1~7のいずれか記載の剤又は製剤。
- ポリビニルアルコール系重合体:低置換度ヒドロキシプロピルセルロースの質量比が、40:60~80:20である請求項1~8のいずれか記載の剤又は製剤。
- ポリビニルアルコール系重合体の平均重合度が1000以上であり、ポリビニルアルコール系重合体:低置換度ヒドロキシプロピルセルロースの質量比が40:60~80:20である請求項1~9のいずれか記載の剤又は製剤。
- 請求項1及び6~10のいずれか記載の剤及び製剤の対象物を含む混合物を直接圧縮する工程を含む、固形製剤の製造方法。
