Abstract
本発明は、下記一般式(1)又は(1')[式中、R1及びR2は、それぞれ独立して、水素原子又はヒドロキシ基であり;R3は、炭素数1~6のアルキル基であり;R4は、ハロゲン原子又はアルキル基であり;mはR4の数で、0~4の整数であり;mが2以上のとき複数のR4は同じでも異なってもよく;nは1~6の整数であり;n´は1~4の整数である。]で表される、化合物及び当該化合物を有効成分とする、好中球浸潤抑制剤を提供する。
Description
本発明は、レゾルビンE2と同様の抗炎症作用を有し、かつレゾルビンE2よりも安定な化合物、当該化合物を有効成分とする抗炎症剤に関する。
ω-3多価不飽和脂肪酸の代謝物であるレゾルビン(Rvs)は、強い抗炎症作用を有しており(非特許文献1)、近年、新しい抗炎症薬のリードとして注目を集めている。Rvsは、好中球浸潤を抑制し、著しく低用量でマクロファージの食作用を促進することによって、炎症の消散を積極的に促進する。中でもレゾルビンE2(RvE2:(5S,6E,8Z,11Z,14Z,16E,18R)-5,18-dihydroxyicosa-6,8,11,14,16-pentaenoic acid)は、極めて強力な炎症収束作用を有する脂質メディエーターである。しかし、Rvsは、その多価不飽和構造のために、化学的にも代謝的にも不安定である。したがって、強力な抗炎症活性を維持しつつ、より安定した物質を開発することが求められている。
RvE2の安定等価化合物としては、例えば、RvE2のシクロプロパン同族体(congener)(CP-RvE2)が報告されている(非特許文献2)。RvE2のスキップジエン部分の2つのビスアリルC10及びC13の位置は、酸化されやすい。CP-RvE2は、C11-C12のcis-オレフィンを、cis-シクロプロパンに置換された化合物である。

CP-RvE2は、RvE2と比較して酸素に対する安定性が改善された、強力な抗炎症剤であるが、代謝安定性は不十分である。また、CP-RvE2の合成にはかなり長い反応ステップが必要であり、結果として総収率が低い、という問題がある。
すなわち、本発明は、RvE2の医薬利用の障害となっている2つの問題点、すなわち、化学的及び代謝的な不安定性と化学合成の煩雑さの両方を克服したRvE2の安定等価体を提供することを目的とする。
本発明者らは、RvE2のスキップジエン部分をベンゼン環に置換することによって得られたRvE2のベンゼン同族体(BZ-RvE2)は、RvE2と同様に抗炎症活性を示す上に、化学的及び代謝的な安定性が高く、かつ合成も比較的容易であることを見出し、本発明を完成させた。
すなわち、本発明は、以下の、化合物等を提供するものである。

本発明に係る化合物は、RvE2と同様に抗炎症活性を示す上に、化学的及び代謝的な安定性が高く、かつ合成も比較的容易である。このため、当該化合物は、RvE2と同様に、炎症性疾患等の治療に用いられる医薬用組成物の有効成分として非常に有用である。
本発明及び本願明細書において、「Cy-z(y及びzは、y<zを充たす正の整数である。)」は、炭素数がy以上z以下であることを意味する。
本発明及び本願明細書において、「式(X)で表される化合物」は、「化合物(X)」と表されることがある。
本発明に係る化合物は、下記一般式(1)又は(1′)で表される化合物(以下、「化合物(1)」、又は「化合物(1′)」ということがある。)である。化合物(1)及び(1′)は、RvE2のスキップジエン部分がベンゼン環に置換されたBZ-RvE2とその同族体である。化合物(1)及び(1′)は、安定性の低いジエン部分を安定性の高いベンゼン環に置換しているため、化学的にも代謝的にも安定である。

一般式(1)中、mは0~2の整数であることが好ましく、0又は1であることがより好ましい。一般式(1)中、mが1以上である場合、R4は、フッ素原子、塩素原子、臭素原子、ヨウ素原子、またはメチル基であることが好ましく、mが1である場合、R4は臭素原子であることがより好ましい。R4は、後述する式(1-o)、(1-m)においては、カルボン酸基を有する置換基のメタ位又はパラ位に置換されることが好ましい。
化合物(1)には、化合物(1-o)、化合物(1-m)、及び化合物(1-p)が含まれる。
一般式(1)、(1-o)、(1-m)、及び(1-p)中、R1及びR2は、それぞれ独立して、水素原子又はヒドロキシ基である。化合物(1)、化合物(1-o)、化合物(1-m)、及び化合物(1-p)としては、R1がヒドロキシ基であり、R2が水素原子である化合物、又は、R1及びR2がいずれもヒドロキシ基である化合物が好ましく、R1がヒドロキシ基であり、R2が水素原子である化合物が特に好ましい。
一般式(1)、(1-o)、(1-m)、及び(1-p)中、R3は、炭素数1~6のアルキル基(C1-6アルキル基)である。C1-6アルキル基は、直鎖状であってもよく、分岐鎖状であってもよい。当該C1-6アルキル基としては、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基、イソブチル基、sec-ブチル基、tert-ブチル基、ペンチル基、イソペンチル基、ネオペンチル基、tert-ペンチル基、ヘキシル基等が挙げられる。化合物(1)、化合物(1-o)、化合物(1-m)、及び化合物(1-p)としては、R3が、C1-4アルキル基である化合物が好ましく、直鎖状のC1-4アルキル基である化合物がより好ましく、メチル基、エチル基、又はプロピル基である化合物がさらに好ましく、エチル基である化合物が特に好ましい。
一般式(1)、(1-o)、(1-m)、及び(1-p)中、nは1~6の整数である。化合物(1)、化合物(1-o)、化合物(1-m)、及び化合物(1-p)としては、nが2~5の整数である化合物が好ましく、nが2~4の整数である化合物がより好ましく、nが3である化合物が特に好ましい。

化合物(1)としては、生体内における代謝安定性がより優れていることから、化合物(1-o)又は化合物(1-m)が好ましく、化合物(1-o)がより好ましい。化合物(1-o)としては、化合物(1-o-1)~化合物(1-o-4)が挙げられ、化合物(1-m)としては、化合物(1-m-1)~化合物(1-m-4)が挙げられる。化合物(1)としては、好中球浸潤抑制作用を含む抗炎症作用がより優れていることから、下記一般式(1-o-1)、(1-o-2)、(1-m-1)、又は(1-m-2)で表される化合物が好ましく、下記一般式(1-o-2)又は(1-m-2)で表される化合物がより好ましく、下記一般式(1-o-2)で表される化合物がさらに好ましい。
一般式(1-o-1)~(1-o-4)、(1-m-1)~(1-m-4)中、R3、R4、m及びnは、前記一般式(1)と同じである。これらの一般式で表される化合物としては、R3が直鎖状のC1-4アルキル基であり、nが2~4の整数である化合物が好ましく、R3がメチル基、エチル基、又はプロピル基であり、nが2~4の整数である化合物がより好ましい。
一般式(1′)中、R2、R3、R4、及びmは、前記一般式(1)と同じである。n´は、1~4の整数であり、3であることが好ましい。一般式(1′)中、R2及びR3を有する置換基は、ラクトン環を有する置換基のオルト位又はメタ位に置換されることが好ましい。この場合、R4は、ラクトン環を有する置換基のメタ位又はパラ位に置換されることが好ましい。
化合物(1)は、塩を形成してもよい。当該塩を形成する塩基としては、ナトリウム、カリウム等のアルカリ金属;カルシウム、マグネシウム等のアルカリ土類金属等が挙げられる。また、化合物(1)及び(1´)は、水和物等の形態であってもよい。例えば、化合物(1)中のカルボキシ基は、ナトリウム、カリウム等のアルカリ金属塩を形成していてもよい。
化合物(1)及び(1´)は、例えば、後記実施例に示すように、ベンゼン環に、RvE2のω-末端鎖部分とカルボン酸鎖部分を、Stilleカップリング反応等によって連結することで合成できる。安定なベンゼン化合物を原料とし、比較的穏やかな反応条件で合成反応を行えることから、化合物(1)及び(1´)は、RvE2やCP-RvE2と比較して、化学合成が容易である。
化合物(1)及び(1′)はRvE2の安定等価体であり、RvE2と同様の薬理作用を有する。例えば、化合物(1)及び(1′)は、RvE2と同様に、炎症収束作用を有する。このため、化合物(1)、(1′)及びその塩は、炎症収束作用により治療又は予防効果が期待される疾患の治療に用いられる医薬用組成物の有効成分として非常に有用であり、特に、炎症や炎症性疾患の治療又は予防のための医薬用組成物の有効成分として好適である。炎症性疾患としては、例えば、潰瘍性大腸炎、クローン病等の炎症性腸疾患、アレルギー性皮膚炎、喘息、アレルギー性鼻炎等のアレルギー性疾患、関節リウマチ、膠原病、全身性エリテマトーデス、多発性筋炎・皮膚筋炎、シェーグレン症候群等の自己免疫疾患等が挙げられる。その他、化合物(1)及び(1′)は、RvE2と同様に(非特許文献3)、鬱病の治療又は予防に用いられる医薬用組成物の有効成分として有用である。
化合物(1)、(1′)又はその塩は、低分子化合物であることから、免疫原性等の問題がない。また、経口投与も可能であり、投与経路もあまり制限されない。このため、化合物(1)、(1′)又はその塩は、特に、ヒトを含む哺乳類に対する医薬の有効成分として有用である。
化合物(1)、(1′)又はその塩を医薬組成物に含有せしめる場合、必要に応じて薬学的に許容される担体と配合し、予防又は治療目的に応じて各種の投与形態を採用可能である。該形態としては、例えば、経口剤、注射剤、坐剤、軟膏剤、貼付剤等が挙げられるが、経口剤が好ましい。これらの投与形態は、各々当業者に公知慣用の製剤方法により製造できる。
薬学的に許容される担体としては、固形製剤における賦形剤、結合剤、崩壊剤、滑沢剤、着色剤;液状製剤における溶剤、溶解補助剤、懸濁化剤、等張化剤、緩衝剤、無痛化剤等が用いられる。また、必要に応じて防腐剤、抗酸化剤、着色剤、甘味剤、安定化剤等の製剤添加物を用いることもできる。
経口用固形製剤を調製する場合は、化合物(1)又は(1′)に賦形剤、必要に応じて、結合剤、崩壊剤、滑沢剤、着色剤、矯味・矯臭剤等を加えた後、常法により錠剤、被覆錠剤、顆粒剤、散剤、カプセル剤等を製造することができる。
経口用液体製剤を調製する場合は、化合物(1)又は(1′)に、矯味剤、緩衝剤、安定化剤、矯臭剤等を加えて、常法により内服液剤、シロップ剤、エリキシル剤等を製造することができる。
注射剤を調製する場合は、化合物(1)又は(1′)に、pH調節剤、緩衝剤、安定化剤、等張化剤、局所麻酔剤等を添加し、常法により皮下、筋肉内及び静脈内用注射剤を製造することができる。
坐剤を調製する場合は、化合物(1)又は(1′)に当業界において公知の製剤用担体、例えば、ポリエチレングリコール、ラノリン、カカオ脂、脂肪酸トリグリセリド等を加えた後、常法により製造することができる。
軟膏剤を調製する場合は、化合物(1)又は(1′)に通常使用される基剤、安定剤、湿潤剤、保存剤等が必要に応じて配合され、常法により混合、製剤化される。
貼付剤を調製する場合は、通常の支持体に前記軟膏、クリーム、ゲル、ペースト等を常法により塗布すればよい。

前記の各製剤中の化合物(1)又は(1′)の含有量は、患者の症状、その剤形等により一定ではないが、一般に経口剤では約0.05~1000mg、注射剤では約0.01~500mg、坐剤では約1~1000mg程度である。
また、これらの製剤の1日あたりの投与量は、患者の症状、体重、年齢、性別等によって異なり一概には決定できないが、通常成人(体重60kg)1日あたり、化合物(1)又は(1′)が約0.05~5000mg程度であり、0.1~1000mgが好ましく、これを1日1回又は2~3回程度に分けて投与するのが好ましい。
化合物(1)又は(1′)を有効成分とする好中球浸潤抑制剤、抗炎症剤、及び医薬用組成物が投与される動物は、特に限定されるものではなく、ヒトであってもよく、ヒト以外の動物であってもよい。非ヒト動物としては、ウシ、ブタ、ウマ、ヒツジ、ヤギ、サル、イヌ、ネコ、ウサギ、マウス、ラット、ハムスター、モルモット等の哺乳動物や、ニワトリ、ウズラ、カモ等の鳥類等が挙げられる。
次に実施例を示して本発明をさらに詳細に説明するが、本発明は以下の実施例に限定されるものではない。
<化合物の分析方法>
NMRスペクトルは、NMR装置JEOL ECX400P(400 MHz)、JEOL EXP400(400 MHz)、及びJEOL ECA500(500 MHz)(いずれも、日本電子社製)で測定した。化学シフトは、テトラメチルシラン(TMS)に対してδスケールで、内部標準として、1H(CDCl3)に対して0.00 ppm、残留CHCl3(1Hに対して7.26 ppm、13Cに対して77.16 ppm)、CH3OH(1Hに対して3.31 ppm、13Cに対して49.0 ppm)として報告された。

旋光度は、旋光計DIP-1030(日本分光社製)を用いて測定した。
シリカゲルカラムクロマトグラフィー及びフラッシュカラムクロマトグラフィーは、それぞれ、Wakogel 60N(和光純薬工業社製、中性、63-212μm)及びシリカゲル60N(関東化学社製、球状、中性、40-50m)を用いて測定した。
旋光度は、旋光計DIP-1030(日本分光社製)を用いて測定した。

<動物実験>

<抗炎症性の評価アッセイ>
アクネ菌を腹腔内へ移植して誘発したマウス急性炎症モデルを用いて、各化合物の抗炎症作用の強さを評価した。
マウス急性炎症モデルは、雄のBALB/cマウス(6~7週間、Japan SLC社から入手)を用いて製造した。まず、熱殺菌したPropionibacterium acnes(P.アクネス;マウスあたり500μg)をマウスの腹腔内に注射して、急性炎症モデルマウスを作製した。アクネ菌注射から8又は12時間後に、急性炎症モデルマウスに被験化合物を腹腔内投与した。その後、アクネ菌注射から24時間後に、腹膜滲出液を収集して好中球数をカウントした。
アクネ菌を腹腔内へ移植して誘発したマウス急性炎症モデルを用いて、各化合物の抗炎症作用の強さを評価した。

評価対象の合成化合物のエタノール溶液(1mg/1mL)から60μLを分取し、溶媒を除去した。0.1M PBSバッファー(435μL、pH7.4)、CH3CN(10μL)、NADPH再生システムA溶液(25μL)、及びNADPH再生システムB溶液(5μL)を各化合物に添加し、ボルテックスミキサーで攪拌した。得られた反応液を、37℃で5分間インキュベーションした後、ヒト肝ミクロゾーム(Corning(登録商標)UltraPool(登録商標)HLM 150)(25μL)を添加し、80μLの溶液を1時間ごと(添加から0、1、2、3、6時間後)に分取した。分取された溶液は、CH3CN(80μL)を加えることによりクエンチされた後、室温で3分間遠心分離(10000 rpm)された。上清を回収し、HPLC(COSMOSIL 5C18-MS-II、4.6mm×250mm、CH3CN/H2O/TFA=35/65/0.01、1.0mL/分、検出254nm)を使用して分析した。

[実施例1]
2-ヨードベンジルアルコールをビニルスタナンとStilleカップリングにより順次連結し、脱保護、加水分解を経てo-BZ-RvE2[一般式(1-o-1)のうち、R3がエチル基、mが0、nが3である化合物]を合成した。同じ経路から対応するヨードベンジルアルコールを用いることにより、m-BZ-RvE2[一般式(1-m-1)のうち、R3がエチル基、mが0、nが3である化合物]及びp-BZ-RvE2[一般式(1-p)のうち、R1及びR2がヒドロキシ基、R3がエチル基、mが0、nが3である化合物]も合成した。

<Methyl (S,E)-5-hydroxy-7-(tributylstannyl)hept-6-enoate (S2)>
Cy3PH・BF4(88.0mg、0.24mmol)及びi-Pr2NEt(84μL、0.48mmol)のCH2Cl2(60mL)溶液に、室温で、Pd2(dba)3(55mg、60μmol)を加えた。得られた溶液を、同じ温度で10分間撹拌した。反応混合物に、化合物S1(1.87g、12.0mmol、>99%ee)及びBu3SnH(3.9mL、14.4mmol)を含むCH2Cl2溶液に0℃で加えた後、当該混合物を同じ温度で一晩撹拌して濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(K2CO3-SiO2=1:9、AcOEt-ヘキサン=1:8)で精製して、化合物S2(3.5g、7.83mmol、65%)を無色の油として得た。化合物S2の分析結果は以下の通りであり、これは既報と一致した。

化合物S2(3.5g、7.83mmol)のDMF(52mL)溶液を攪拌した後、室温でイミダゾール(2.13g、31.3mmol)及びTBSCl(2.36g、15.7mmol)を加えた。得られた混合物を、同じ温度で12時間撹拌し、水の添加によりクエンチした後、AcOEt/ヘキサン(1:4)で抽出した。得られた有機抽出物を全て混合し、ブラインで洗浄し、硫酸ナトリウム(Na2SO4)で乾燥させて濃縮した。得られた残渣をフラッシュシリカゲルカラムクロマトグラフィー(K2CO3-SiO2=1:9、AcOEt-ヘキサン=1:8)で精製して、化合物(8)(4.0g、7.12mmol、91%)を無色の油として得た。化合物(8)の分析結果は以下の通りであり、これは既報と一致した。

<抗炎症性の評価アッセイ>
アクネ菌注射から12時間後の急性炎症モデルマウスに、300pgのRvE2、o-BZ-RvE2、m-BZ-RvE2、p-BZ-RvE2、又は溶媒のみを腹腔内投与した(n=4)。その後、アクネ菌注射から24時間後に、腹膜滲出液を収集して好中球数をカウントした。結果を図1(a)に示す。

独立して、アクネ菌注射から12時間後の急性炎症モデルマウスに、300fg、300pg、若しくは300ngのRvE2、o-BZ-RvE2、又は溶媒のみを、腹腔内投与した(n=3又は4)。その後、アクネ菌注射から24時間後に、腹膜滲出液を収集して好中球数をカウントした。結果を図1(b)に示す。
図1(a)に示ように、o-BZ-RvE2、m-BZ-RvE2、及びp-BZ-RvE2はいずれも、溶媒のみを投与したモデルマウスよりも好中球の数が少なく、炎症部位である腹腔内への好中球の浸潤が優位に抑制されていた。この結果から、o-BZ-RvE2、m-BZ-RvE2、及びp-BZ-RvE2はいずれも、RvE2と同様に、炎症収束作用があり、抗炎症剤として有用であることがわかった。特に、o-BZ-RvE2は、RvE2よりも優れた抗炎症作用を有していた(図1(a)及び(b))。
<代謝安定性の評価>
RvE2、o-BZ-RvE2、m-BZ-RvE2、及びp-BZ-RvE2のヒト肝ミクロゾームに対する代謝安定性を調べた。具体的には、RvE2、o-BZ-RvE2、m-BZ-RvE2、及びp-BZ-RvE2を、それぞれ、ヒト肝ミクロゾームと混合し、経時的に残存するRvE2等の割合を調べた。結果を図2に示す。図2に示すように、p-BZ-RvE2はRvE2と同様にヒト肝ミクロゾームによって分解されやすかった。これに対して、o-BZ-RvE2及びm-BZ-RvE2は、ヒト肝ミクロゾームと混合して6時間経過後でも残存率が90%以上であった。特にo-BZ-RvE2は非常に代謝安定性に優れていた。

[実施例2]
RvE類は共通してカルボン酸とω-3位のヒドロキシ基を有しており、またRvE1とRvE2では共通して5位のヒドロキシ基を有する。実施例1において抗炎症活性の最も強かったo-BZ-RvE2についてデオキシ誘導体を合成し、その生物活性への影響を検討した。5-deoxy-o-BZ-RvE2は、一般式(1-o-3)のうち、R3がエチル基、mが0、nが3である化合物であり、17-deoxy-o-BZ-RvE2は、一般式(1-o-2)のうち、R3がエチル基、mが0、nが3である化合物であり、5,17-deoxy-o-BZ-RvE2は、一般式(1-o-4)のうち、R3がエチル基、mが0、nが3である化合物である。

アルゴン雰囲気下、化合物(10)(1.08g、3.0mmol)、2-ヨードベンジルアルコール(4a、702mg、3.0mmol、1.0eq.)、テトラキストリフェニルホスフィンパラジウム(173mg、0.15mmol、5mol%)をトルエン溶液(20mL)として凍結脱気し、加熱還流下、一晩撹拌した。得られた反応液に、飽和塩化アンモニウム水溶液を加えて反応を停止し、酢酸エチルで抽出した。得られた有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを加えて乾燥した。その後、減圧下溶媒を留去し、フラッシュシリカゲルクロマトグラフィー (10%w/w K2CO3-SiOH、ヘキサン:酢酸エチル=10:1)で精製し、化合物(13d)(323mg、0.65mmol、63%)を得た。化合物(13d)の分析結果は以下の通りであり、これは既報と一致した。

アルゴン雰囲気下、化合物(16f)(20mg、0.046mmol)のTHF溶液(92μL)に、テトラブチルアンモニウムフルオリド(1M THF溶液、276μL、0.276mol、6eq.)を加え、室温で24時間撹拌した。次いで、当該反応液に水酸化リチウム一水和物(8mg、0.184mmol、4eq.)の水溶液(92μL)を加え、室温で1時間攪拌した。その後、リン酸緩衝液(pH6)を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄して、無水硫酸ナトリウムで乾燥した。減圧下溶媒を留去して残渣をフラッシュシリカゲルクロマトグラフィー(クロロホルム:メタノール=30:1)で精製し、5-deoxy-o-BZ-RvE2(11.2mg、0.037μmol、81%)を無色油状物質として得た。5-deoxy-o-BZ-RvE2の分析結果は以下の通りであった。

<抗炎症性の評価アッセイ>
アクネ菌注射から8時間後の急性炎症モデルマウスに、300pgのRvE2、o-BZ-RvE2、17-deoxy-o-BZ-RvE2、5-deoxy-o-BZ-RvE2、5,17-dideoxy-o-BZ-RvE2、又は溶媒のみを腹腔内投与した(n=3又は4)。その後、アクネ菌注射から24時間後に、腹膜滲出液を収集して好中球数をカウントした。結果を図3に示す。
アクネ菌注射から8時間後の急性炎症モデルマウスに、300pgのRvE2、o-BZ-RvE2、17-deoxy-o-BZ-RvE2、5-deoxy-o-BZ-RvE2、5,17-dideoxy-o-BZ-RvE2、又は溶媒のみを腹腔内投与した(n=3又は4)。その後、アクネ菌注射から24時間後に、腹膜滲出液を収集して好中球数をカウントした。結果を図3に示す。
図3に示ように、コントロールとしてアクネ菌を接種させたマウスでは、無処置群(図中、左端カラム)と比べて好中球数が顕著に増加したのに対して、RvE2、o-BZ-RvE2、17-deoxy-o-BZ-RvE2、5-deoxy-o-BZ-RvE2、及び5,17-dideoxy-o-BZ-RvE2を投与したマウスでは有意に好中球数が減少した。特に、17-deoxy-BZ-RvE2を投与した群は、RvE2を投与した群及びo-BZ-RvE2を投与した群と同程度に好中球数を減少させており、高い抗炎症活性が維持されていた。一方で、5-deoxy-BZ-RvE2を投与した群及び5,17- dideoxy-BZ-RvE2を投与した群では、RvE2又はo-BZ-RvE2を投与した群よりも、抗炎症活性が減弱した。これらの結果から、o-BZ-RvE2が持つ抗炎症活性の発現には、カルボン酸のδ位に存在する水酸基が重要であることが示唆された。
<代謝安定性の評価>
17-deoxy-o-BZ-RvE2及び5-deoxy-o-BZ-RvE2のヒト肝ミクロゾームに対する代謝安定性を調べた。具体的には、RvE2、o-BZ-RvE2、17-deoxy-o-BZ-RvE2、及び5-deoxy-o-BZ-RvE2について、37℃におけるヒト肝ミクロソーム存在下、0.1M PBS(pH7.4)中での残存率(%)の経時変化を、HPLCで測定した(n=3)。結果を図4に示す。
17-deoxy-o-BZ-RvE2及び5-deoxy-o-BZ-RvE2のヒト肝ミクロゾームに対する代謝安定性を調べた。具体的には、RvE2、o-BZ-RvE2、17-deoxy-o-BZ-RvE2、及び5-deoxy-o-BZ-RvE2について、37℃におけるヒト肝ミクロソーム存在下、0.1M PBS(pH7.4)中での残存率(%)の経時変化を、HPLCで測定した(n=3)。結果を図4に示す。
図4に示すように、RvE2は時間経過と共に残存率が低下したのに対して、5-deoxy-BZ-RvE2及び17- deoxy-BZ-RvE2の残存率は、RvE2と比較して緩やかに減少しており、代謝的な安定性が高いことが示された。また、両デオキシ体の代謝安定性には大きな差異がなかったことから、5-deoxy-o-BZ-RvE2における抗炎症活性の減弱は、代謝抵抗性に起因するものではないと考えられた。

<Methyl 5-bromo-2-iodobenzotate (17-a)>
アルゴン雰囲気下、5-ブロモアントラニル酸メチル(200 mg, 0.869 mmol)を塩酸(12 M, 2.1 mL)に溶かし0℃で撹拌した。15分後、氷(1.0 g)を加え0℃で撹拌した。10分後、亜硝酸ナトリウム(120 mg, 3.82 mmol, 2.0 eq.)を水(1.5 mL)に溶解させたものを加え0 ℃で撹拌した。10分後、ヨウ化カリウム(634 mg, 3.82 mmol, 4.4 eq.)を水(2.9 mL)に溶解させたものを加え、室温で撹拌した。1時間後、反応液に飽和チオ硫酸ナトリウム水溶液を加え反応を停止し、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、芒硝乾燥し、溶媒を減圧下留去した。シリカゲルクロマトグラフィー(ヘキサン:酢酸エチル=9:1)で精製し、化合物(17-a) (283 mg, 0.831 mmol, 96%)を黄色結晶として得た。

アルゴン雰囲気下、化合物(17-a)(7.93 g, 23.0 mmol)をTHF(46 mL)に溶かした溶液にエタノール(5.4 mL, 92 mmol, 4.0 eq.)、LiBH4(1.00 g, 46 mmol, 2.0 eq.)を氷冷下加え1時間撹拌した。さらに室温に昇温し12時間撹拌した。反応液に飽和塩化アンモニウム水溶液を加えて反応を停止し、酢酸エチルで抽出した。得られた有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを加え乾燥した。減圧下溶媒を留去し、シリカゲルクロマトグラフィー(ヘキサン:酢酸エチル=4:1)で精製し化合物(18-a)(6.67 g, 21.3 mmol, 93%)を白色粉末状物質として得た。

アルゴン雰囲気下、化合物(18-a)(250 mg, 0.80 mmol)、化合物(10)(287 mg, 0.80 mmol, 1.0 eq.)をトルエン(5.5 mL)に溶解させ、テトラキストリフェニルホスフィンパラジウム(46 mg, 0.04 mmol, 0.05 eq.)を加え加熱還流下24時間撹拌した。反応液に飽和塩化アンモニウム水溶液を加えて反応を停止し、酢酸エチルで抽出した。得られた有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを加えて乾燥した。減圧下溶媒を留去し、シリカゲルクロマトグラフィー(10%w/w K2CO3-SiOH、ヘキサン:酢酸エチル=19:1 to 9;1)で精製し化合物(19-a)(130 mg, 0.51 mmol, 64%)を黄色油状物質として得た。

アルゴン雰囲気下、化合物(19-a)(50 mg, 0.20 mmol)をジクロロメタン(2.0 mL)に溶かした溶液に、トリエチルアミン(31 μL, 0.22 mmol, 1.1 eq.)、メタンスルホニルクロリド(17 μL, 0.22 mmol, 1.1 eq.)を0 ℃で加え、撹拌した。90分後、飽和食塩水を加えてクエンチし、酢酸エチルで抽出した。得られた有機層に無水硫酸ナトリウムを加え乾燥した。減圧下溶媒を留去して化合物20-aを粗生成物として得た。粗生成物をアセトン(2.0 mL)に溶解させ、臭化ナトリウム(112 mg, 1.09 mmol, 6.0 eq.)を加え12時間還流した。反応液に飽和食塩水を加え、酢酸エチルで抽出した。減圧下溶媒を留去して残渣をシリカゲルクロマトグラフィー(ヘキサン:酢酸エチル=9:1)で精製して化合物(21-a)(48.5 mg, 0.152 mmol, 76% for 2 steps)を黄色油状物質として得た。

アルゴン雰囲気下、化合物(21-a)(24.3 mg, 43.3 μmol, 1.5 eq.)と化合物(8)(9.2 mg, 28.8 μmol, 1.0 eq.)をジメチルホルムアミド(1.0 mL)に溶解させ、テトラキストリフェニルホスフィンパラジウム(2.1 mg, 1.81 μmol, 0.05 eq.)を加えて加熱還流下18時間撹拌した。反応液に飽和食塩水を加えて反応を停止し、酢酸エチルで抽出した。得られた有機層を飽和食塩水で洗浄し、無水硫酸ナトリウムを加え乾燥した。減圧下溶媒を留去し、フラッシュシリカゲルクロマトグラフィー(ヘキサン:酢酸エチル=99:1)で精製し、得られた化合物(22-a)をそのまま次の反応に用いた。化合物(22-a)をテトラヒドロフラン(100 μL)に溶解させ、テトラブチルアンモニウムフルオリド(1 M THF溶液, 107 μL, 107 μmol, 4 eq.)を加え、室温で2日間撹拌した。反応液にリン酸緩衝液(pH 6)を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄して無水硫酸ナトリウムで乾燥した。減圧下溶媒を留去し、シリカゲルクロマトグラフィー(クロロホルム:メタノール=30:1)で精製し、化合物(23-a)(3.6 mg, 9.43 μmol, 33% for 2 steps)を無色油状物質として得た。
