Abstract

Description
本発明は、線維芽細胞増殖因子21(以下「FGF21」という)誘導剤に関する。より具体的には、本発明は、D-ソルビトール、D-プシコース、及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び前記少なくとも1種の糖質の糖残基を含むオリゴ糖のいずれか一方又は両方を含有する、FGF21誘導剤に関する。本発明は、前記FGF21誘導剤を含む、哺乳動物におけるアルコール嗜好性又は単純糖質嗜好性を抑制するための組成物にも関する。
FGF21は、エネルギー恒常性を調節するホルモンであり、飢餓状態及び高炭水化物食を含む様々な代謝性ストレスに応答して肝臓で発現誘導される。FGF21は、細胞膜FGF受容体と1回膜貫通型タンパク質のβ-Klothoとの複合体を介して作用する。FGF21は、アルコール嗜好性及び甘味嗜好性を調節することが知られている(非特許文献1)。本発明者らは、単純糖質に対する嗜好性の抑制が、分泌されたFGF21とβ-Klothoとの複合体形成による脳内のオキシトシン(Oxt)神経の活性化によって引き起こされることを見出した(非特許文献2)。
アルコール飲料は、生活・文化の一部として親しまれている。一方で、アルコールは依存性を有し、慢性的なアルコールへの暴露により臓器障害(例えば、肝疾患又は脳卒中)を引き起こすことがある。わが国では、アルコール依存症患者は約110万人おり、その予備軍が約300万人、高リスク飲酒者は訳1000万人になると推定されている。酒量を抑制するための医薬品として、ジスルフィラム、シアナミド液、アカンプロサート、及びナルメフェンが市販されている。
グルコースは、脳・中枢神経系においてほぼ唯一のエネルギー源として利用され、生命活動に重要な役割を担っている。しかし、過剰に摂取された糖質は、脂肪組織に脂肪として蓄積され、肥満の原因となる。わが国では、肥満症患者及びその予備軍が約2000万人いると報告されている。肥満症を処置するための医薬品として、マジンドール、及び消化管リパーゼ阻害剤が市販されている。
砂糖に対する欲求及び砂糖を多く含むものに対する欲求を止められないという、砂糖に対する渇望感の存在が指摘されている。この砂糖に対する渇望感は、中毒症状の一種であり、砂糖依存症(Sugar Addiction)とも称されている。この中毒症状を惹き起こすのは砂糖であるとされている。このように、グルコース及び砂糖(スクロース)のような単純糖質は、生体においてエネルギー源として利用される一方で、中毒症状をひきおこしたり、又は肥満症の病因になることもある。

本開示は、アルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方を抑制し得る新規組成物を提供することを1つの目的とする。
本発明者らは、アルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方を抑制し得る標的としてFGF21に着目した。本発明者らは、経口摂取可能な様々な糖質について検討し、特定の希少糖又は糖アルコールがFGF21の分泌量を増大させることを見出した。
本開示の例示的な態様は、FGF21誘導剤、アルコール嗜好性を抑制するための組成物、及び単純糖質嗜好性を抑制するための組成物を提供する。本開示の例示的な態様は、より具体的には、以下の実施形態を提供する。

本明細書において「糖質」は、糖又は糖アルコールを含む炭水化物を意味する。本開示に係る糖質は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の化合物を意味する。糖質は、例えば、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の化合物であってよい。1つの実施形態において、糖質は、D-ソルビトール、D-プシコース、又はD-タガトースの1種単独であってよい。1つの実施形態において、糖質は、D-ソルビトール及びD-プシコースの組合せ、D-プシコース及びD-タガトースの組合せ、D-ソルビトール及びD-タガトースの組合せ、又はD-ソルビトール、D-プシコース及びD-タガトースの組合せであってよい。
本明細書において「糖」又は「単純糖質」は、単糖又は二糖を意味する。単純糖質は、限定するものではないが、グルコース、フルクトース、スクロース、ラクトース及びマルトースが挙げられる。本明細書において「多糖」は、複数の糖が結合した糖化合物であって、20糖以上のものを意味する。1種類の単糖のみからなる多糖をホモグリカンといい、複数種類の単糖のみからなる多糖をヘテログリカンともいう。多糖は、限定するものではないが、アミロース、アミロペクチン、デキストリン、スターチ、及びグリコーゲンが挙げられる。
本明細書において「合成甘味料」は、自然界では存在せず、化学的な合成により製造される化合物であって、哺乳動物が経口摂取した場合に、前記哺乳動物に甘みを感じさせる化合物を意味する。合成甘味料は、限定するものではないが、アスパルテーム、スクラロース、ネオテーム、アドバンテーム、サッカリン、及びアセスルファムカリウムが挙げられる。
本明細書において「オリゴ糖」は、複数の糖残基が結合した糖化合物であって、3糖以上20糖未満のものを意味する。本開示に係るオリゴ糖は、好ましくは、3糖以上10糖以下である。オリゴ糖は、例えば、分子量300~3,000である。オリゴ糖は、1種の糖残基からなるもの、又は2種の糖残基からなるものであってよい。オリゴ糖は、限定するものではないが、有機化学合成法又は酵素化学合成法に従って、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質から合成することができる。オリゴ糖は、例えば、糖転移酵素(グルコシルトランスフェラーゼ)を利用した方法、及び/又は糖分解酵素(グリコシダーゼ)の逆反応を利用した方法によって合成することができる。
本明細書において「D-ソルビトール」又は「D-Sorbitol」は、式1で表される糖アルコールを意味する:

本明細書において「鏡像異性体」は、重ね合わすことができない像と鏡像の関係にある立体異性体を意味する。本明細書において、糖の立体異性体は、d-グリセルアルデヒドの立体配置を基準として、この立体配置に対応する立体異性体をD体とし、その鏡像異性体をL体とする。D-ソルビトール、D-プシコース、及びD-タガトースはいずれも、d-グリセルアルデヒドの立体配置に対応するD体である。それぞれ異なる鏡像異性体の混合物から、公知の方法に従って、所望の鏡像異性体を分離して、その純度を高めることができる。そのような公知の方法として、限定するものではないが、キラルHPLCが挙げられる。
本明細書において「FGF21」は、哺乳動物においてFgf21遺伝子にコードされ、肝臓から分泌されるホルモンタンパク質である。FGF21は、線維芽細胞増殖因子(fibroblast growth factor:FGF)の内分泌型FGFクラスに属し、1回膜貫通型タンパク質のβ-Klothoと複合体を形成する細胞膜FGF受容体を介して作用する。FGF21とβ-Klothoとで形成された複合体は、脳内のオキシトシン(Oxt)神経を活性化させ、アルコール嗜好性及び単純糖質嗜好性を抑制し得る。血液中FGF21量の増加は、アルコール嗜好性及び単純糖質嗜好性を抑制し得る。
血液中FGF21量は、哺乳動物の血液(全血、血漿又は血清)中のFGF21濃度(例えばng/ml、nM)、所定量の血液に含有されるFGF21の重量(例えばng、mg)であってよい。血液中FGF21量は、公知の方法に従って測定できる。公知の方法としては、限定するものではないが、抗FGF21抗体を利用するELISAが挙げられる。
本明細書において「誘導剤」は、哺乳動物の体内において所期の生成物の発現を誘導する物質を配合した物を意味する。配合物は、単独成分で構成されていてもよく、2種以上の成分を含む組成物であってもよい。配合物は、限定するものではないが、固体形態または液体形態であってよい。誘導剤は、例えば、哺乳動物の細胞内において、血液中に分泌される所期の生成物を生成させる物質を含む。この例において、前記誘導剤は、哺乳動物の血液中で所期の生成物量の増加をもたらし得る。
本明細書において「FGF21誘導剤」は、哺乳動物の体内においてFGF21の発現を誘導する物質を配合した物を意味する。FGF21誘導剤は、限定するものではないが、哺乳動物の肝細胞内において、血液中に分泌されるFGF21の発現を誘導する物質を含む。本発明に係るFGF21誘導剤は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び前記少なくとも1種の糖質の糖残基を含むオリゴ糖のいずれか一方又は両方を、哺乳動物の体内においてFGF21の発現を誘導する物質として含有する。
1つの実施形態において、FGF21誘導剤は、D-ソルビトール、D-プシコース又はD-タガトースの1種の糖質、及び/又はD-ソルビトール、D-プシコース又はD-タガトースの1種の糖残基を含むオリゴ糖を含む。FGF21を誘導する成分としてD-プシコースのみ、D-プシコースの糖残基を含むオリゴ糖のみ、又はそれらの両方を含むFGF21誘導剤は、比較的速やかに血中のFGF21濃度を増加させ得る。D-ソルビトール及びD-タガトースのいずれか一方の糖質のみを、糖質及び/又は糖残基として含むFGF21誘導剤は、比較的長い時間、血中のFGF21濃度を高く維持させ得る。
1つの実施形態において、FGF21誘導剤は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖質を、糖質及び/又は糖残基として含む。FGF21誘導剤は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖質、及び/又はD-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖残基を含むオリゴ糖を含有する。前記少なくとも2種の糖質又は糖残基は、D-ソルビトール及びD-プシコースの組合せ、D-ソルビトール及びD-タガトースの組合せ、D-プシコース及びD-タガトースの組合せ、並びにD-ソルビトール、D-プシコース及びD-タガトースの組合せであってよい。
前記少なくとも2種の糖質及び/又は前記オリゴ糖は、FGF21の誘導について相乗効果又は相加効果を奏し得る。前記少なくとも2種の糖質及び/又は前記オリゴ糖を含有するFGF21誘導剤は、その摂取量を低減させつつ、高いFGF21誘導作用を奏し得る。D-タガトース及びD-ソルビトール又はD-プシコースを糖質及び/又は糖残基として含むFGF21誘導剤は、その摂取量を低減させつつ、高いFGF21誘導作用を奏させ得る。D-タガトース及びD-プシコースを糖質及び/又は糖残基として含むFGF21誘導剤は、その摂取量を低減させつつ、比較的速やかに血中のFGF21濃度を増加させ、かつ比較的長い時間高いFGF21濃度を維持させ得る。
D-プシコース及びD-タガトース又はD-ソルビトールを糖質及び/又は糖残基として含むFGF21誘導剤は、比較的速やかに血中のFGF21を増加させ、かつ比較的長い時間高いFGF21濃度を維持させ得る。D-ソルビトール及びD-タガトースを糖質及び/又は糖残基として含むFGF21誘導剤は、FGF21を比較的長い時間高い濃度で維持させ得る。D-ソルビトール、D-プシコース及びD-タガトースを糖質及び/又は糖残基として含むFGF21誘導剤は、比較的速やかに血中のFGF21を増加させ、かつ比較的長い時間高いFGF21濃度を維持させ得る。
FGF21誘導剤の形態は、限定するものではないが、経口摂取可能な形態、例えば錠剤、カプセル剤、顆粒剤、散剤、粉剤、シロップ剤、又は液剤であってよい。前記経口摂取上許容される添加剤は、限定するものではないが、賦形剤、崩壊剤、結合剤、湿潤剤、安定剤、緩衝剤、滑沢剤、保存剤、調味料、矯味剤、香料、酸味料、又は着色剤、もしくはそれらの組み合わせであってよい。FGF21誘導剤は、限定するものではないが、その形態に応じた公知の方法に従って製造することができる。FGF21誘導剤が粉末剤の場合、例えば、粉末形態の前記少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖と、必要に応じて、経口摂取上許容される粉末形態の添加剤とを配合することにより製造することができる。FGF21誘導剤が液剤の場合、例えば、前記少なくとも1種の糖質及び/又は前記少なくとも1つの糖質の糖残基を含むオリゴ糖を、水性液剤に配合することにより製造することができる。
FGF21誘導剤における、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖の配合量は、剤の形態に応じて適宜設定される。FGF21誘導剤に配合される前記少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖の1日あたりの摂取量は、限定するものではないが、FGF21誘導剤を摂取する前の哺乳動物の血液中のFGF21量と比較して、FGF21誘導剤を1週間摂取した当該哺乳動物の血液中のFGF21量が有意に増加する量である。有意差検定は、限定するものではないが、Tukey’s testによって行うことができる。前記哺乳動物は、例えばマウスである。マウスにおける血液中のFGF21量を有意に増加せる量は、例えば、1~10g/体重kg、3~7g/体重kg又は5g/体重kgである。マウス以外の哺乳動物における血液中のFGF21量を有意に増加させる量は、本願明細書の開示内容に照らして、当業者により適宜設定され得る。マウス以外の哺乳動物に対する本開示に係る糖質の摂取量は、例えば、体表面積に基づくヒト等価用量(HED)に従って設定することができる。ヒトにおける血液中のFGF21量を有意に増加させる量は、例えば、100mg~1g/体重kg、200~700mg/体重kg、300~500mg/体重kg又は300~400mg/体重kgである。
FGF21誘導剤の1日あたりの摂取回数は、例えば、1日分を1回で摂取してもよく、または複数回に分けて摂取してもよい。1日1回で摂取されるFGF21誘導剤は、例えば、FGF21を誘導する成分としてD-ソルビトール及びD-タガトースのいずれか一方の糖質のみ、前記糖質の糖残基を含むオリゴ糖のみ、又はそれらの両方を、糖質又は糖残基として含む、或いはD-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖質、前記少なくとも2種の糖質の糖残基を含むオリゴ糖、又はそれらの両方を含む。FGF21誘導剤は、限定するものではないが、サプリメントとしてそのまま摂取されてもよく、又は飲食品に添加して摂取されてもよい。
本明細書において「組成物」は、限定するものではないが、固体形態または液体形態であってよい。組成物は、例えば、哺乳動物においてアルコール嗜好性の抑制をもたらし得る物質を配合した物である。この例において、前記組成物は、例えば、哺乳動物の血液中の、アルコール嗜好性の抑制を奏し得る生成物の増加をもたらし得る。
本明細書において「サプリメント」は、哺乳動物において所定の症状の発症を予防し、前記哺乳動物の体調の維持又は健康の増進を目的として経口摂取される、医薬品ではない組成物を意味する。1つの実施形態において、サプリメントは機能性表示食品である。
本明細書において「処置」は、所定の症状又は病態の発症の予防、或いは、所定の症状又は病態の治療を意味する。本明細書において「治療」は、所定の病状又は病態の軽快、緩和又は完治を意味する。
本明細書において「哺乳動物におけるアルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方を抑制するための組成物」は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を、哺乳動物におけるアルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方を抑制し得る物質として含有する。前記組成物は、限定するものではないが、本明細書に記載のFGF21誘導剤を含有する。1つの実施形態において、前記組成物は、哺乳動物におけるアルコール嗜好性を抑制するための組成物である。他の実施形態において、前記組成物は、哺乳動物における単純糖質嗜好性を抑制するための組成物である。他の実施形態において、前記組成物は、哺乳動物におけるアルコール嗜好性及び単純糖質嗜好性を抑制するための組成物である。組成物の形態は、FGF21誘導剤について記載された形態が適宜適用される。
1つの実施形態において、単純糖質嗜好性を抑制するための組成物は、グルコース、フルクトース、スクロース、ラクトース、及びマルトースに対する嗜好性を抑制するための組成物である。他の実施形態において、単純糖質嗜好性を抑制するための組成物は、グルコース、フルクトース、及びスクロースに対する嗜好性を抑制するための組成物である。他の実施形態において、単純糖質嗜好性を抑制するための組成物は、単純糖質に対する嗜好性は抑制するが、合成甘味料及び多糖のいずれか一方又は両方に対する嗜好性を実質的に変化させない、組成物である。1つの実施形態において、単純糖質嗜好性を抑制するための組成物は、D-ソルビトール及びD-タガトースからなる群より選択される少なくとも1種の糖質(例えば1種又は2種)、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を含む。
合成甘味料は、限定するものではないが、アスパルテーム、スクラロース、ネオテーム、アドバンテーム、サッカリン、及びアセスルファムカリウムからなる群より選択される少なくとも1つの化合物である。合成甘味料は、例えばサッカリンである。多糖は、限定するものではないが、アミロース、アミロペクチン、デキストリン、スターチ、及びグリコーゲンからなる群より選択される少なくとも1つの化合物である。多糖は、例えばデキストリンである。1つの実施形態において、単純糖質嗜好性を抑制するための組成物は、単純糖質に対する嗜好性は抑制するが、サッカリン及びデキストリンに対する嗜好性は実質的に変化させない組成物である。
1つの実施形態において、アルコール嗜好性を抑制するための組成物は、炭化水素の水素原子をヒドロキシル基で置き換えた物質(例えばエタノール)を含む水性組成物(例えば酒類)に対する嗜好性を抑制するための組成物である。他の実施形態において、アルコール嗜好性を抑制するための組成物は、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質(例えば1種又は2種)、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を含む。
本明細書に記載のアルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方を抑制するための組成物に配合される前記少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖の1日あたりの摂取量は、限定するものではないが、前記組成物を摂取する前の哺乳動物のアルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方と比較して、前記組成物を1週間摂取した当該哺乳動物のアルコール嗜好性及び単純糖質嗜好性のいずれか一方又は両方が有意に低下する量である。有意差検定は、限定するものではないが、Tukey’s testによって行うことができる。前記哺乳動物は、例えばマウスである。マウスにおける血液中のFGF21量を有意に増加せる量は、例えば、1~10g/体重kg、3~7g/体重kg又は5g/体重kgである。マウス以外の哺乳動物における血液中のFGF21量を有意に増加させる量は、本願明細書の開示内容に照らして、当業者により適宜設定され得る。ヒトにおける血液中のFGF21量を有意に増加させる量は、例えば、100mg~1g/体重kg、200~700mg/体重kg、300~500mg/体重kg又は300~400mg/体重kgである。
本明細書において「医薬」又は「医薬組成物」は、哺乳動物において所定の症状又は兆候を処置するための物質を配合した物を意味する。配合物は、限定するものではないが、固体形態または液体形態であってよい。医薬又は医薬組成物は、例えば、哺乳動物におけるアルコール依存症を処置するための物質を配合した物である。この例において、医薬又は医薬組成物は、アルコール依存症の予防効果、治療効果、又は緩和作用をもたらし得る。
本明細書において「肥満症を処置するための医薬」又は「アルコール依存症を処置するための医薬」は、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を、肥満症又はアルコール依存症を処置するための物質として配合する。前記医薬は、限定するものではないが、本明細書に記載のFGF21誘導剤を配合する。1つの実施形態において、前記医薬は、肥満症処置用の医薬組成物である。肥満処置用の医薬組成物は、例えば、単純糖質の摂取量の低減を目的として投与される。他の実施形態において、前記医薬は、アルコール依存症処置用の医薬組成物である。アルコール依存症処置用の医薬組成物は、例えば、酒類(アルコール飲料)の摂取量の低減を目的として投与される。1つの実施形態において、アルコール依存症処置用の医薬組成物は、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を含む。
前記医薬の形態は、限定するものではないが、経口摂取可能な形態、例えば錠剤、カプセル剤、顆粒剤、散剤、粉剤、シロップ剤、又は液剤であってよい。医薬上許容される経口摂取可能な添加剤は、例えば、医薬分野において公知の添加剤、及びFGF21誘導剤に関して記載した添加剤を適宜用いることができる。医薬は、限定するものではないが、その形態に応じた公知の方法に従って製造することができる。
本明細書に開示した組成物又は医薬の1日あたりの摂取回数は、例えば、1日分を1回で摂取してもよく、または複数回に分けて摂取してもよい。1日1回で摂取される本明細書に開示した組成物又は医薬は、例えば、FGF21を誘導する成分としてD-ソルビトール及びD-タガトースのいずれか一方の糖質のみ、前記糖質の糖残基を含むオリゴ糖のみ、又はそれらの両方、或いはD-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖質、前記少なくとも2種の糖質の糖残基を含むオリゴ糖、又はそれらの両方を含む。
1つの実施形態において、本明細書に記載のFGF21誘導剤又は医薬は、その全量を100質量%として、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を1~99質量%、5~80質量%、10~50質量%含有する。1つの実施形態において、本明細書に記載のFGF21誘導剤又は医薬は、その全量を100質量%として、D-ソルビトール、D-プシコース、又はD-タガトースを1~99質量%、5~80質量%、10~50質量%含有する。
本明細書において「アルコール依存症」は、飲酒がその対象にとって大きな価値をもち、他の行動よりも優先する一群の生理的、行動的、及び/又は認知的現象を意味する。アルコール依存症における生理的状態には、飲酒の中止又は飲酒量の低下によって、痙攣発作及び幻覚を含むアルコール離脱症状群がある。アルコール依存症における認知的現象には、飲酒したいという強烈な欲求、脅迫感(渇望)、飲酒又は飲酒量の抑制喪失、飲酒以外の娯楽への関心の減退若しくは喪失が挙げられる。アルコール依存症は、アルコール耐性の獲得を伴うことがある。アルコール耐性は、同じ程度の酔いを獲得するためには以前より大量の飲酒をしなければならない状態を意味する。
本明細書において「肥満症」は、肥満に起因する又は関連する健康障害(以下「肥満関連疾患」ともいう)を伴う、減量を要する病態を意味する。肥満関連疾患には、耐糖能障害(例えば2型糖尿病、耐糖能異常)、脂質異常症、高血圧、高尿酸血症又は痛風、冠動脈疾患(例えば心筋梗塞、又は狭心症)、脳梗塞(例えば脳血栓症、又は一過性脳虚血発作)、脂肪肝、月経異常又は妊娠合併症(例えば妊娠高血圧症候群、妊娠糖尿病、又は難聴)、睡眠時無呼吸症候群又は肥満低換気症候群、整形外科的疾患(例えば変形性関節症、変形性脊椎症)、及び肥満関連腎臓病が挙げられる。
FGF21誘導剤は、例えば、哺乳動物に経口摂取される。前記医薬は、哺乳動物に投与される。哺乳動物は、限定するものではないが、ヒト、霊長類(サル、チンパンジー)、家畜動物(ウシ、ウマ、ブタ、ヒツジ)、愛玩動物(イヌ、ネコ)であってよい。哺乳動物は、好ましくはヒトである。前記医薬は、好ましくは、ヒトに経口投与される。
1つの実施形態において、アルコール嗜好性を抑制するための組成物は、アルコール依存症患者、依存症予備軍の者又は高リスク飲酒者に経口摂取される。1つの実施形態において、単純糖質嗜好性を抑制するための組成物は、肥満症の患者、又は肥満症の予備軍の者に経口摂取される。
1つの実施形態において、アルコール依存症を処置するための医薬は、アルコール依存症患者に経口投与される。1つの実施形態において、肥満症を処置するための医薬は、肥満症患者に経口投与される。
1つの実施形態において、哺乳動物におけるアルコール依存症又は肥満症を処置するための医薬を製造するための、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖の使用が提供される。本明細書において、FGF21誘導剤、組成物又は医薬について記載した特徴は、本実施形態にも適用される。
1つの実施形態において、その必要のある哺乳動物に、アルコール依存症又は肥満症を処置するための医薬を経口投与することを含む、アルコール依存症又は肥満症を処置する方法であって、前記医薬が、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を含有する、前記方法が提供される。本明細書において、FGF21誘導剤、組成物又は医薬について記載した特徴は、本実施形態にも適用される。

1つの実施形態において、哺乳動物においてFGF21を誘導する方法であって、D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又は前記少なくとも1種の糖質の糖残基を含むオリゴ糖を哺乳動物に投与することを含む、方法が提供される。前記哺乳動物は、例えば、血中でのFGF21量が前記糖質及び/又は前記オリゴ糖の投与前の血中のFGF21量と比較して増大する。本明細書において、FGF21誘導剤、組成物又は医薬について記載した特徴は、本実施形態にも含まれる。
以下、具体的な実施例を記載するが、それらは本開示の例示的な実施形態を示すものであり、添付する特許請求の範囲に記載の発明の範囲をいかようにも限定するものではない。
[実施例1]
(マウス)
9週齢の雄β-Klotho Floxマウスをコントロール群として用いた。Oxt神経特異的にFGF21受容体を欠損した9週齢の雄オキシトシン(Oxt)神経特異的β-Klothoノックアウトマウス(Cell Metab. 2012 Sep 5;16(3):387-93)を実験群として用いた。
(飼育条件)
マウスは、ポリカーボネートのケージ(日本クレア(株))にて個別に飼育した。各マウスに、固形餌(CE-2;日本クレア(株))を自由摂取させた。飼育室および試験溶液の温度は25±1℃にて維持した。飼育室の明暗サイクルは、明期12時間、暗期12時間(06:00と18:00に切り替えた)に維持した。
(マウス)

4%エタノール水溶液、8%エタノール水溶液、及び16%エタノール水溶液を試験溶液として用いた。
(2瓶選択試験)
・第1期間(実験条件への馴化): 滅菌水を入れた微量飲水量測定用給水瓶2本を24時間、4日間提示して、2本の微量飲水量測定用給水瓶からの飲水を学習させた。
・第2期間(ベースラインの試験溶液に対する嗜好性の測定): 滅菌水を入れた微量飲水量測定用給水瓶と試験溶液を入れた微量飲水量測定用給水瓶の2本を24時間、4日間提示した。場所に対する嗜好性の影響を排除するために、2本の微量飲水量測定用給水瓶は、24時間ごとに位置を交換した。
4%エタノール水溶液、8%エタノール水溶液、及び16%エタノール水溶液を試験溶液として用いた。
(2瓶選択試験)
・第1期間(実験条件への馴化): 滅菌水を入れた微量飲水量測定用給水瓶2本を24時間、4日間提示して、2本の微量飲水量測定用給水瓶からの飲水を学習させた。
・第2期間(ベースラインの試験溶液に対する嗜好性の測定): 滅菌水を入れた微量飲水量測定用給水瓶と試験溶液を入れた微量飲水量測定用給水瓶の2本を24時間、4日間提示した。場所に対する嗜好性の影響を排除するために、2本の微量飲水量測定用給水瓶は、24時間ごとに位置を交換した。
(摂取溶液量の測定)
マウスが摂取した溶液量は、マウス・ラット用微量飲水量測定用給水瓶(Drink-O-Measurer、シンファクトリー社製)を用いて測定した。
(嗜好性の計算)
試験溶液に対する嗜好性は、以下の式を用いて計算した。
嗜好性 = (4日間の試験溶液の総飲水量)/(4日間の滅菌水の総飲水量 + 4日間の試験溶液の総飲水量)×100
マウスが摂取した溶液量は、マウス・ラット用微量飲水量測定用給水瓶(Drink-O-Measurer、シンファクトリー社製)を用いて測定した。
(嗜好性の計算)
試験溶液に対する嗜好性は、以下の式を用いて計算した。
嗜好性 = (4日間の試験溶液の総飲水量)/(4日間の滅菌水の総飲水量 + 4日間の試験溶液の総飲水量)×100
(試験結果)
各試験溶液に対する嗜好性について、コントロール群と実験群との間に有意差があるかをStudent’s t-testを用いて検定した。各試験溶液について得られた結果を、図1にまとめた。図1(a)~(c)は、β-Klotho Floxマウス(白抜き)と比較して、オキシトシン(Oxt)神経特異的β-Klothoノックアウトマウス(灰色)においてアルコールに対する嗜好性が増加したことを示す。これらの結果は、FGF21の共受容体であるβ-Klothoを介したFGF21による作用が、マウスにおけるアルコール嗜好性の抑制に必要であることを示唆する。
各試験溶液に対する嗜好性について、コントロール群と実験群との間に有意差があるかをStudent’s t-testを用いて検定した。各試験溶液について得られた結果を、図1にまとめた。図1(a)~(c)は、β-Klotho Floxマウス(白抜き)と比較して、オキシトシン(Oxt)神経特異的β-Klothoノックアウトマウス(灰色)においてアルコールに対する嗜好性が増加したことを示す。これらの結果は、FGF21の共受容体であるβ-Klothoを介したFGF21による作用が、マウスにおけるアルコール嗜好性の抑制に必要であることを示唆する。

(試験溶液)
実施例1と同じ濃度のエタノール水溶液を用いた。
(摂取溶液量の測定及び嗜好性の計算)
実施例1と同じ方法を用いた。
(マウス)

・第4期間(本実験): 粉末餌(CE-2)を摂取するマウスをコントロール群とした。高ショ糖食(HSD)の粉末餌(高ショ糖食;(株)オリエンタルバイオサービス)を摂取するマウスの2群をそれぞれ実験群とした。餌は、マウス用マルチフィーダーにて与え、24時間ごとに新しい粉末餌に交換した。滅菌水を入れた微量飲水量測定用給水瓶と、試験溶液を入れた微量飲水量測定用給水瓶との2本を24時間、4日間提示した。場所に対する嗜好性の影響を排除するために、2本の微量飲水量測定用給水瓶は、24時間ごとに位置を交換した。
・第5期間(採血): 第4期間の後、滅菌水を入れた給水瓶1本を24時間提示した後、麻酔下で心臓から採血を行った。血液はヘパリン(最終濃度が10μg/ml;富士フイルム和光純薬(株))で処理後、4℃、2000rpm、20分間の遠心分離を行い、血漿を採取した。血漿中のFGF21濃度は、Fibroblast Growth Factor 21 Mouse/Rat ELISA(BioVendor社)を用い、添付の指示書に従って測定した。

(試験結果)

アルコール嗜好性と血漿中FGF21濃度との間に相関関係があるかを調べた。図2(g)~(i)は、アルコール嗜好性と血漿中FGF21濃度との間に負の相関関係があることを示す。これらの結果は、血中FGF21濃度が増加するに伴って、アルコール嗜好性が抑制されることを示唆する。
[実施例3]

9週齢の雄β-Klotho Floxマウスをコントロール群として用いた(n=9)。9週齢の雄オキシトシン(Oxt)神経特異的β-Klothoノックアウトマウスを実験群として用いた(n=9)。

[実施例4]
(初代培養肝細胞の作製)
9週齢の雄C57BL/6JJclマウス(日本クレア(株))を用い、以下の手順に従って、初代培養肝細胞を作製した。麻酔下でマウスの門脈にシリコンチューブを挿入し、腹部下大動脈を切断した。大動脈の切断と同時に、門脈に挿入したシリコンチューブを介してEGTA液(37℃)を流し入れ、その後、コラゲナーゼ液(37℃)を流し入れた。次に肝臓を切離し、シャーレに移した。切離した肝臓からピッペッティングによって肝細胞を遊離させた。遊離した肝細胞にHanks液(4℃)を加え、回収した肝細胞を含む懸濁液を100μmセルストレーナー(コーニングファルコン)で濾過し、得られた濾液を低速遠心(50g、3分間)した。その沈渣をHanks液(4℃)に再び分散させて、低速遠心した。この再分散及び遠心処理を3回繰り返した。得られた沈渣を培養液(37℃)に分散し、1×105cells/wellでコラーゲンTypeIコートマイクロプレート24wellに播種した。播種後3時間で新たな培養液に交換し、24時間培養した後、得られた肝細胞を初代培養肝細胞とした。
培養液は、Williams’ medium E(シグマアルドリッチ)にウシ胎児血清(10%;ThermoFischer)、ITS-Gサプリメント(x100)(1μM;富士フイルム和光純薬(株))、デキサメサゾン(1μM;富士フイルム和光純薬(株))を添加して作製したものを用いた。

(初代培養肝細胞への糖又は糖アルコール添加実験及び遺伝子発現解析)
作製した初代培養肝細胞を、25mMとなるように以下の表に示す各糖質1~6の試験溶液を添加した培養液又は同容量のVehicle(蒸留水)を添加した培養液中で、24時間培養した。

24時間培養した後、培養上清を除去し、D-PBS(-)を用いて培養肝細胞を含むwellを洗浄した。培養肝細胞から、RNAiso Plus(タカラバイオ(株))を用いてTotal RNAを抽出した。Total RNA 1μgから、GoScript(登録商標)Reverse Transcriptase(promega社)を用いてcDNAを作製した。作製したcDNAを用いてリアルタイムPCR法を行い、遺伝子発現量を測定した(Light Cycler 480 SYBR Green I;ROCHE社)。FGF21のmRNA量は、β-actinのmRNA量で補正した。リアルタイムPCRに用いたプライマーの配列を以下の表に示す。
(試験結果)
培養肝細胞におけるFGF21のmRNA量について、Vehicleを添加した培養液中で培養した肝細胞群と、各糖質1~6の試験溶液を添加した培養液中で培養した肝細胞群との間に有意差があるかをTukey’s testで検定した(n=3~4)(#:p<0.01)。各肝細胞群について得られた結果を図4にまとめた。図4は、Fgf21遺伝子の発現量が、D-Glucose及びD-Tagatoseにおいて有意に増加したことを示す。また、Fgf21遺伝子の発現増加の程度は、D-Glucose添加による遺伝子発現増加よりも、D-Tagatose添加による遺伝子発現増加の方が大きいことを示す。
培養肝細胞におけるFGF21のmRNA量について、Vehicleを添加した培養液中で培養した肝細胞群と、各糖質1~6の試験溶液を添加した培養液中で培養した肝細胞群との間に有意差があるかをTukey’s testで検定した(n=3~4)(#:p<0.01)。各肝細胞群について得られた結果を図4にまとめた。図4は、Fgf21遺伝子の発現量が、D-Glucose及びD-Tagatoseにおいて有意に増加したことを示す。また、Fgf21遺伝子の発現増加の程度は、D-Glucose添加による遺伝子発現増加よりも、D-Tagatose添加による遺伝子発現増加の方が大きいことを示す。
[実施例5]
(D-Tagatose添加によるFGF21分泌量の測定)
添加するD-Tagatoseの濃度を5mM、10mM、15mM及び25mMの一連の濃度としたことを除き、実施例4と同様にして、初代培養肝細胞を24時間培養し、培養上清を採取した(n=4)。採取した培養上清中のFGF21量は、実施例2と同様、Fibroblast Growth Factor 21 Mouse/Rat ELISAを用いて測定した。
(D-Tagatose添加によるFGF21分泌量の測定)
添加するD-Tagatoseの濃度を5mM、10mM、15mM及び25mMの一連の濃度としたことを除き、実施例4と同様にして、初代培養肝細胞を24時間培養し、培養上清を採取した(n=4)。採取した培養上清中のFGF21量は、実施例2と同様、Fibroblast Growth Factor 21 Mouse/Rat ELISAを用いて測定した。

培養上清中のFGF21の測定結果を図5にまとめた。Tukey’s testを用いて、統計的解析を行った。図5は、Vehicleと25mM D-Mannitolとが群aに分類され、群bに分類された25mM D-Glucose又は5mM D-TagatoseによるFGF21分泌量と有意に異なることを示す。また、図5は、D-Tagatose添加によるFGF21分泌量が、その用量に依存して有意に増加することを示す。実施例4と同様にして、Fgf21遺伝子の発現の程度を調べた結果、Fgf21遺伝子の発現量も、D-Tagatoseの用量依存的に増加していた。これらの結果は、D-TagatoseによるFgf21遺伝子の発現量の増加とFGF21の分泌量の増加との間には、正の相関関係があることを示唆する。

実施例4と同様にして作製した初代肝細胞を、25mMとなるように以下の表に示す各糖又は糖アルコール1~48の試験溶液を添加した培養液又は同容量のVehicle(蒸留水)を添加した培養液中で、24時間培養した。
(初代培養肝細胞への糖又は糖アルコール添加実験及びFGF21分泌量の測定)
実施例4と同様にして作製した初代肝細胞を、25mMとなるように以下の表に示す各糖又は糖アルコール1~48の試験溶液を添加した培養液又は同容量のVehicle(蒸留水)を添加した培養液中で、24時間培養した。
24時間培養した後、培養上清を採取し、培養上清中のFGF21量を測定するまで-30℃にて保存した。培養上清中のFGF21量は、実施例2と同様、Fibroblast Growth Factor 21 Mouse/Rat ELISAを用いて測定した(n=3)。
(試験結果)
各試験溶液を添加した培養上清中のFGF21量の結果を図6にまとめた。糖質を添加してないVehicle(蒸留水)を添加した培養液を用いた試験を陰性対照とし、単糖類D-グルコースを添加した培養液を用いた試験を陽性対照(positive control)とした。図6において、陽性対照の平均値をダッシュラインで示す。図6は、D-Fructose、D-Sorbitol、D-Xylitol、D-Arabitol、D-Psicose、及びD-Tagatoseを培養液に添加した場合に、陽性対照によるFGF21発現量を超えたことを示す。ここで、D-Xylitolは、Fgf21遺伝子発現を増加させることが知られている(Uebanso T et al.PLoS One.2011;6(8):e22976.)。本試験結果のうち、D-XylitolによるFGF21発現量を超える発現量を示した糖又は糖アルコールは、D-Sorbitol、D-Psicose、及びD-Tagatoseの3種類であった。
各試験溶液を添加した培養上清中のFGF21量の結果を図6にまとめた。糖質を添加してないVehicle(蒸留水)を添加した培養液を用いた試験を陰性対照とし、単糖類D-グルコースを添加した培養液を用いた試験を陽性対照(positive control)とした。図6において、陽性対照の平均値をダッシュラインで示す。図6は、D-Fructose、D-Sorbitol、D-Xylitol、D-Arabitol、D-Psicose、及びD-Tagatoseを培養液に添加した場合に、陽性対照によるFGF21発現量を超えたことを示す。ここで、D-Xylitolは、Fgf21遺伝子発現を増加させることが知られている(Uebanso T et al.PLoS One.2011;6(8):e22976.)。本試験結果のうち、D-XylitolによるFGF21発現量を超える発現量を示した糖又は糖アルコールは、D-Sorbitol、D-Psicose、及びD-Tagatoseの3種類であった。
添加するD-Sorbitol、D-Psicose、及びD-Tagatoseの3種類の糖又は糖アルコールについて、培養液中での濃度を5mM、10mM、15mM、25mMの一連の濃度とし、上記と同様にして培養上清中のFGF21量を測定した(n=4)。その結果、FGF21の発現量が、D-Sorbitolの用量依存的に増加することが示された(図7)。D-Tagatoseについては、実施例5と同様、FGF21発現量は、15mMにて最も大きくなったことを除いて、D-Tagatoseの用量依存的にFGF21の発現量も増加した。同じ25mMの濃度では、D-SorbitolとD-Tagatoseは、D-Glucoseに比べてFGF21分泌刺激能力が高かった。
[実施例7]
実施例4で用いた糖質(表1)に代えて、以下の表に示す糖又は糖アルコール1~4の試験溶液を25mMとなるように添加したことを除き、実施例4と同様にして、FGF21のmRNA量を測定した。FGF21のmRNA量の測定は、糖又は糖アルコールの添加後、4時間、8時間、12時間及び24時間に実施した。

培養上清中のFGF21の測定結果を図10にまとめた。Tukey’s testを用いて、統計的解析を行った。図10は、実施例9で用いた糖質の組合せのすべて、D-Fructose、D-Tagatose、D-Sorbitol及びD-Xylitolの糖質単独を用いた場合に、Fgf21遺伝子の発現量が有意(P<0.01)に増加したことを示す。Fgf21遺伝子の発現量は、D-TagatoseとD-Sorbitolとの組合せを用いた場合が一番大きかった。各糖質を用いた場合のFgf21遺伝子の発現量の合計値からの増加量は、D-TagatoseとD-Psicoseとの組合せを用いた場合が一番大きかった。
培養上清中のFGF21の測定結果を図10にまとめた。Tukey’s testを用いて、統計的解析を行った。図10は、実施例9で用いた糖質の組合せのすべて、D-Fructose、D-Tagatose、D-Sorbitol及びD-Xylitolの糖質単独を用いた場合に、Fgf21遺伝子の発現量が有意(P<0.01)に増加したことを示す。Fgf21遺伝子の発現量は、D-TagatoseとD-Sorbitolとの組合せを用いた場合が一番大きかった。各糖質を用いた場合のFgf21遺伝子の発現量の合計値からの増加量は、D-TagatoseとD-Psicoseとの組合せを用いた場合が一番大きかった。
D-TagatoseとD-Sorbitolとの組合せ、及びD-TagatoseとD-Psicoseとの組合せは、各組合せを構成する2つの糖質それぞれのFgf21遺伝子発現量の合計値よりも、当該組合せでのFgf21遺伝子発現量の方が大きかった(図10)。これらの結果は、当該組合せが、Fgf21遺伝子発現において相乗効果を奏することを示す。D-SorbitolとD-Psicoseとの組合せを用いた場合のFgf21遺伝子の発現量は、当該組合せを構成する2つの糖質それぞれのFgf21遺伝子発現量の合計値とほぼ等しかった(図10)。この結果は、当該組合せが、Fgf21遺伝子発現において相加効果を奏することを示す。
一方、D-XylitolとD-Tagatoseとの組合せ、D-XylitolとD-Sorbitolとの組合せ、又はD-XylitolとD-Psicoseとの組合せを用いた場合のFgf21遺伝子発現量は、各組合せを構成する2つの糖質それぞれのFgf21遺伝子発現量の合計値よりも小さい又はほぼ同等であった(図10)。これ等の結果は、当該組合せが、Fgf21遺伝子発現において相乗効果を奏しないことを示す。
[実施例10]
(マウス)
9週齢の雄C57BL/6JJclマウス(日本クレア)を用いた。
(飼育条件)
実施例1と同じ飼育条件を用いた。

(糖質の投与、及び採血)
マウスに、5g/体重kgの蒸留水、又は各種糖質D-Glucose、D-Tagatose、D-Psicose若しくはD-Sorbitolを経口投与した。経口投与の4時間、6時間、8時間、12時間及び24時間後に、80μLの血液をマウスの尾静脈からヘマトクリット毛細管(ヘパリン処理;アズワン)を用いて採取した。採取した血液を2,000g×20分、4℃の遠心分離を行い、血漿を採取して分析まで-30℃で保存した。
(血漿中のFGF21量の測定)
採取した血漿中のFGF21量は、実施例2と同様、Fibroblast Growth Factor 21 Mouse/Rat ELISAを用いて測定した。
マウスに、5g/体重kgの蒸留水、又は各種糖質D-Glucose、D-Tagatose、D-Psicose若しくはD-Sorbitolを経口投与した。経口投与の4時間、6時間、8時間、12時間及び24時間後に、80μLの血液をマウスの尾静脈からヘマトクリット毛細管(ヘパリン処理;アズワン)を用いて採取した。採取した血液を2,000g×20分、4℃の遠心分離を行い、血漿を採取して分析まで-30℃で保存した。
(血漿中のFGF21量の測定)
採取した血漿中のFGF21量は、実施例2と同様、Fibroblast Growth Factor 21 Mouse/Rat ELISAを用いて測定した。
(試験結果)
血漿中のFGF21量について、蒸留水を投与したマウス群と各種糖質を投与したマウス群との間に有意差があるかを、Tukey’s multiple comparison testで検定した(n=4)(*:p<0.05、**:p<0.01)。D-Psicoseを投与した群では、血漿中のFGF21量は投与後4時間でピークに達した(図11a、△)。D-Tagatoseを投与した群では、血漿中のFGF21量は投与後6時間でピークに達した(図11a、□)。D-Sorbitolを投与した群では、血漿中のFGF21量は投与後8時間でピークに達した(図11a、◇)。これらの結果は、投与する糖又は糖アルコールの種類又は組合せによって、血液中のFGF21量を増加させるタイミングを調整可能であることを示唆する。例えば、D-PsicoseとD-Sorbitolとの組合せは、比較的速やかに血中のFGF21濃度を上昇させ、長期間にわたってその濃度を維持し得ることが示唆される。
血漿中のFGF21量について、蒸留水を投与したマウス群と各種糖質を投与したマウス群との間に有意差があるかを、Tukey’s multiple comparison testで検定した(n=4)(*:p<0.05、**:p<0.01)。D-Psicoseを投与した群では、血漿中のFGF21量は投与後4時間でピークに達した(図11a、△)。D-Tagatoseを投与した群では、血漿中のFGF21量は投与後6時間でピークに達した(図11a、□)。D-Sorbitolを投与した群では、血漿中のFGF21量は投与後8時間でピークに達した(図11a、◇)。これらの結果は、投与する糖又は糖アルコールの種類又は組合せによって、血液中のFGF21量を増加させるタイミングを調整可能であることを示唆する。例えば、D-PsicoseとD-Sorbitolとの組合せは、比較的速やかに血中のFGF21濃度を上昇させ、長期間にわたってその濃度を維持し得ることが示唆される。
生体内(マウス)でFGF21量がピークに達したタイミングは、D-Sorbitolを投与した場合よりも(図11a、◇:投与後8時間)、D-Psicoseを投与した場合の方が早かった(図11a、△:投与後4時間)。この結果は、生体外(初代肝臓培養細胞)でのFgf21遺伝子の発現量がD-Sorbitolを投与した場合よりも(図8、投与後12時間)、D-Psicoseを投与した場合の方が早かった(図9、投与後4時間)結果と一致する。これらの結果は、生体外での試験結果から、生体内でのFGF21の発現プロファイル(例えば、量及び経時的な変化)又はFGF21による作用(例えば、単純糖/アルコール嗜好性の抑制作用)を予測可能であることを示唆する。
各マウス群の血漿中FGF21量についてのAUCを算出した(図11b)。図11bは、D-Psicose、D-Tagatose及びD-Sorbitolがいずれも有意に血漿中のFGF21量が増加したこと、ならびに各糖質についてのAUC値がほぼ同等であることを示す。
[実施例11]
(マウス)
9週齢の雄C57BL/6JJclマウス(日本クレア)を用いた。

実施例1と同じ飼育条件を用いた。

50mMスクロース水溶液を試験溶液として用いた。

・第2期間(ベースラインの測定): 5g/体重kgの蒸留水を経口投与したマウスに、滅菌水を入れた微量飲水量測定用給水瓶と50mMスクロース水溶液を入れた微量飲水量測定用給水瓶の2本を24時間、2日間提示した。場所に対する嗜好性の影響を排除するために、2本の微量飲水量測定用給水瓶は、24時間ごとに位置を交換した。
・第3期間(投与実験): 5g/体重kgのvehicle(蒸留水)、又は各種糖質D-Tagatose、D-Psicose若しくはD-Sorbitolを経口投与したマウスに、第2期間と同様の試験を行った。
(摂取溶液量の測定)
マウスが摂取した溶液量は、マウス・ラット用微量飲水量測定用給水瓶(Drink-O-Measurer、シンファクトリー社製)を用いて測定した。
・第1期間(実験条件への馴化): 滅菌水を入れた微量飲水量測定用給水瓶2本を24時間、4日間提示して、2本の微量飲水量測定用給水瓶からの飲水を学習させた。
・第2期間(ベースラインの測定): 5g/体重kgの蒸留水を経口投与したマウスに、滅菌水を入れた微量飲水量測定用給水瓶と50mMスクロース水溶液を入れた微量飲水量測定用給水瓶の2本を24時間、2日間提示した。場所に対する嗜好性の影響を排除するために、2本の微量飲水量測定用給水瓶は、24時間ごとに位置を交換した。
・第3期間(投与実験): 5g/体重kgのvehicle(蒸留水)、又は各種糖質D-Tagatose、D-Psicose若しくはD-Sorbitolを経口投与したマウスに、第2期間と同様の試験を行った。
(摂取溶液量の測定)
マウスが摂取した溶液量は、マウス・ラット用微量飲水量測定用給水瓶(Drink-O-Measurer、シンファクトリー社製)を用いて測定した。
(試験結果)
50mMスクロース水溶液を飲水した量について、第2期間(Pretrial)と第3期間(Posttrial)との間に有意差があるかを、Student’s t-testを用いて検定した(n=6)。得られた結果を図12にまとめた。図12は、D-Sorbitol又はD-Tagatoseを投与したマウス群で、50mMスクロース水溶液を飲水した量が有意に減少したことを示す(*:p<0.05)。図12は、D-Psicoseを投与したマウス群で、50mMスクロース水溶液を飲水した量が減少する傾向があることを示す(p=0.068)。これらの結果は、D-Sorbitol、D-Tagatose又はD-Psicoseが、単純糖質嗜好性を抑制する作用を有することを示唆する。

・第3期間(糖質投与による嗜好性の変化を測定): 第2期間でアルコールに対する嗜好性が66%以上であったマウスに対して、滅菌水を入れた微量飲水量測定用給水瓶と試験溶液を入れた微量飲水量測定用給水瓶の2本を24時間、2日間提示した。第3期間試験中は、5g/体重kgのvehicle(蒸留水)、又は各種糖質D-Tagatose、D-Sorbitol若しくはD-Psicoseを経口投与し(1回/日;14時)、嗜好性を測定した。
試験溶液に対する嗜好性は、以下の式を用いて計算した。
嗜好性 = (2日間の試験溶液の総飲水量)/(2日間の滅菌水の総飲水量 + 2日間の試験溶液の総飲水量)×100

(試験結果)
試験液に対する嗜好性について、蒸留水を投与したマウス群と各種糖質を投与したマウス群との間に有意差があるかをPaired T testを用いて検定した(n=5~7)。得られた結果を図13にまとめた。図13は、D-Tagatose又はD-Psicoseを投与したマウス群で、8%エタノール水溶液に対する嗜好性が有意に減少したことを示す(*:p<0.05)。D-Sorbitolを投与したマウス群では、個々のマウスについてのデータのばらつきが大きく、推定される平均値に有意な減少は観察されなかった。当該群では、8%エタノール水溶液に対する嗜好性が抑制されたマウスが観察された。これは、D-Sorbitolが、8%エタノール嗜好性を示すマウスのその嗜好性を抑制したことを示す。これらの結果は、D-Sorbitol、D-Tagatose又はD-Psicoseが、アルコール嗜好性を抑制する作用を有することを示唆する。
試験液に対する嗜好性について、蒸留水を投与したマウス群と各種糖質を投与したマウス群との間に有意差があるかをPaired T testを用いて検定した(n=5~7)。得られた結果を図13にまとめた。図13は、D-Tagatose又はD-Psicoseを投与したマウス群で、8%エタノール水溶液に対する嗜好性が有意に減少したことを示す(*:p<0.05)。D-Sorbitolを投与したマウス群では、個々のマウスについてのデータのばらつきが大きく、推定される平均値に有意な減少は観察されなかった。当該群では、8%エタノール水溶液に対する嗜好性が抑制されたマウスが観察された。これは、D-Sorbitolが、8%エタノール嗜好性を示すマウスのその嗜好性を抑制したことを示す。これらの結果は、D-Sorbitol、D-Tagatose又はD-Psicoseが、アルコール嗜好性を抑制する作用を有することを示唆する。
Claims (10)
- D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖質、及び/又はD-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも1種の糖残基を含むオリゴ糖を含有する、線維芽細胞増殖因子21(以下「FGF21」という)誘導剤。
- 前記少なくとも1種の糖質が、D-ソルビトール又はD-タガトースを含む、請求項1に記載のFGF21誘導剤。
- 前記少なくとも1種の糖質が、D-ソルビトールである、請求項1に記載のFGF21誘導剤。
- 前記少なくとも1種の糖質が、D-タガトースである、請求項1に記載のFGF21誘導剤。
- D-ソルビトール、D-プシコース及びD-タガトースからなる群より選択される少なくとも2種の糖質が、糖質及び/又は糖残基として含まれる、請求項1に記載のFGF21誘導剤。
- 請求項1~5のいずれか一項に記載のFGF21誘導剤を含む、哺乳動物におけるアルコール嗜好性を抑制するための組成物。
- 請求項1~5のいずれか一項に記載のFGF21誘導剤を含む、哺乳動物における単純糖質嗜好性を抑制するための組成物。
- サプリメントである、請求項6又は7に記載の組成物。
- アルコール依存症を処置するための、請求項6に記載の医薬組成物。
- 肥満症を処置するための、請求項7に記載の医薬組成物。
