Description
本発明は、コラーゲン産生亢進剤、医薬品、化粧品、及びコラーゲン産生亢進剤の製造方法に関する。
近年、ヒトの皮膚の構造やその新陳代謝のメカニズムなどに関する研究が進むにつれて、加齢にともないヒトの皮膚にしわ、小じわ、しみ、及びたるみなどの変化が現れる原因や機構が徐々に明らかになりつつある。皮膚は、外側の薄い表皮(上皮組織)とその下層の厚い真皮(結合組織)とから構成されている。表皮は、体の最外層として、外界から生体を保護するとともに、内部の水分や栄養分が外界に漏出するのを防ぐ。真皮は、主に線維芽細胞、膠原線維(コラーゲン)、弾性繊維(エラスチン)、及びプロテオグリカンなどが複合的に三次元状に広がった構造を持つ結合組織であり、皮膚に強度、伸展性、及び弾力性をもたらす役割を担っている。加齢とともに皮膚における皮脂や水分の量が減少すると、皮膚表面の角質層の保湿力が失われ、乾燥などによる小ジワや肌のかさつきが生じやすくなる。これに対し、発酵品を用いて、皮膚機能の改善を行うことが提案されている(例えば、特許文献1から7参照。)。
本発明は、新規な天然由来成分を有効成分とするコラーゲン産生亢進剤、医薬品、化粧品、及びコラーゲン産生亢進剤の製造方法を提供することを目的とする。
本発明の一側面に係るコラーゲン産生亢進剤は、シソ科植物の発酵物を備える。本発明の一側面に係る医薬品は、シソ科植物の発酵物を備える。本発明の一側面に係る化粧品は、シソ科植物の発酵物を備える。本発明の一側面に係るコラーゲン産生亢進剤の製造方法は、シソ科植物を発酵させることを含む。
本発明によれば、新規な天然由来成分を有効成分とするコラーゲン産生亢進剤、医薬品、化粧品、及びコラーゲン産生亢進剤の製造方法を提供することが可能となる。
以下、本発明の実施の形態について詳細に説明する。なお以下の示す実施の形態は、この発明の技術的思想を具体化するための装置や方法を例示するものであって、この発明の技術的思想は構成部材の組み合わせ等を下記のものに特定するものではない。この発明の技術的思想は、特許請求の範囲において種々の変更を加えることができる。
実施形態に係るコラーゲン産生亢進剤は、シソ科植物の発酵物を備える。シソ科(Lamiaceae又はLabiatae)植物の例としては、ヤマハッカ属(Isodon又はRabdosia)植物が挙げられる。ヤマハッカ属植物の例としては、ヒキオコシ(Isodon japonicus HARA又はRabdosia japonica HARA)が挙げられる。ヒキオコシは、延命草(Isodonis Herba)とも呼ばれる。
実施形態に係るコラーゲン産生亢進剤は、シソ科植物由来の乳酸菌を備えていてもよい。乳酸菌は、例えば、ラクトバシラス(Lactobacillus)属である。ラクトバシラス属は、乳酸桿菌の一種であり、グラム陽性の通性嫌気性菌である。ラクトバシラス属は、糖を発酵して乳酸を産生する。ラクトバシラス属は、ヒトを含む動物の体内にも生息しているが、実施形態に係るラクトバシラス属は、シソ科植物由来のラクトバシラス属である。例えば、実施形態に係るラクトバシラス属は、シソ科植物を発酵させて、抽出される。

実施形態に係るコラーゲン産生亢進剤は、細胞のコラーゲン産生を亢進する。コラーゲンは、例えばI型コラーゲンである。I型コラーゲンは、皮膚にハリ、弾力や強度を与える。I型コラーゲンの減少は、しわ及び老化の原因となる。また、I型コラーゲンは、創傷治癒に関与する。I型コラーゲンは2本のα1鎖及び1本のα2鎖により構成され、α1鎖はCOL1A1遺伝子より発現され、α2鎖はCOL1A2遺伝子より発現される。
実施形態に係るコラーゲン産生亢進剤は、皮膚にハリ、弾力、及び強度の少なくともいずれかを与えるための医薬品、化粧品、及び/又は食品として使用可能である。また、実施形態に係るコラーゲン産生亢進剤は、抗しわ機能向上のための医薬品、化粧品、及び/又は食品として使用可能である。実施形態に係るコラーゲン産生亢進剤は、抗しわ用医薬品、抗老化用医薬品、抗しわ用化粧品、及び抗老化用化粧品として使用可能である。さらに、実施形態に係るコラーゲン産生亢進剤は、創傷治癒のための医薬品、化粧品、及び/又は食品として使用可能である。なお、しわは小じわを含む。
また、実施形態に係るコラーゲン産生亢進剤は、微生物を減少させる機能も有する。微生物の例としては、細菌及びウイルスが挙げられる。細菌の例としては、グラム陰性菌及びグラム陽性菌が挙げられる。
実施形態に係るコラーゲン産生亢進剤は、例えば、グラム陰性菌及びグラム陽性菌を、24時間以内に、80%以上、85%以上、90%以上、あるいは95%以上減少させる。グラム陰性菌としては、大腸菌、サルモネラ菌、腸炎ビブリオ菌、肺炎桿菌、及び緑膿菌等が挙げられるが、これらに限定されない。グラム陽性菌としては、黄色ブドウ球菌、メチシリン耐性黄色ブドウ球菌(MRSA)、芽胞を形成するセレウス菌、及び枯草菌等が挙げられるが、これらに限定されない。
実施形態に係るコラーゲン産生亢進剤は、ウイルスを減少させる機能も有する。実施形態に係るコラーゲン産生亢進剤は、例えば、ウイルスを、24時間以内に、80%以上、85%以上、90%以上、あるいは95%以上減少させる。ウイルスは、エンベロープを有するウイルスであるエンベロープウイルス、及びエンベロープを有さないウイルスであるノンエンベロープウイルスを含む。また、ウイルスは、DNAウイルス及びRNAウイルスを含む。
エンベロープを有するDNAウイルスとしては、ヒトヘルペスウイルス、ワクシニアウイルス、及びB型肝炎ウイルス等が挙げられるが、これらに限定されない。
エンベロープを有するRNAウイルスとしては、インフルエンザウイルス、SARSコロナウイルス、RSウイルス、ムンプスウイルス、ラッサウイルス、デングウイルス、風疹ウイルス、ヒト免疫不全ウイルス、麻疹ウイルス、C型肝炎ウイルス、エボラウイルス、黄熱ウイルス、及び日本脳炎ウイルス等が挙げられるが、これらに限定されない。
エンベロープを有さないDNAウイルスとしては、アデノウイルス、B19ウイルス、パポバウイルス、及びヒトパピローマウイルス等が挙げられるが、これらに限定されない。
エンベロープを有さないRNAウイルスとしては、ノロウイルス、カリシウイルス、ポリオウイルス、エコーウイルス、A型肝炎ウイルス、E型肝炎ウイルス、ライノウイルス、アストロウイルス、ロタウイルス、コクサッキーウイルス、エンテロウイルス、及びサポウイルス等が挙げられるが、これらに限定されない。
実施形態に係るコラーゲン産生亢進剤は、有効量のシソ科植物の発酵物を含む。実施形態に係るコラーゲン産生亢進剤は、シソ科植物の発酵物を、水等の溶媒中に含んでいてもよいし、植物の発酵液中に含んでいてもよい。発酵液を得るための植物としては、ラクトバシラス属と同様に、シソ科植物が挙げられる。有効量とは、細胞のコラーゲン産生を亢進するために必要な量であり、対象とするヒト又は動物の年齢、種類、及び適用部位等によって適宜決定される。
実施形態に係るコラーゲン産生亢進剤に含まれる乳酸菌は、生菌であってもよいし、例えば加熱処理された死菌であってもよい。したがって、実施形態に係るコラーゲン産生亢進剤は、乳酸菌の死菌を含んでいてもよい。乳酸菌は、菌体乾燥物であってもよい。乳酸菌の死菌、あるいは菌体乾燥物も、コラーゲン産生を亢進する効果を奏する。また、乳酸菌の死菌、あるいは菌体乾燥物は、運搬や、長期にわたる保管が容易である。
実施形態に係るコラーゲン産生亢進剤は、例えば、液体、クリーム、軟膏、硬膏、ジェル、ワックス、及びスプレーであってもよい。
実施形態に係るコラーゲン産生亢進剤は、例えば、整肌用化粧品であってもよい。整肌用化粧品の例としては、化粧水、美容液、及びパックが挙げられる。実施形態に係るコラーゲン産生亢進剤は、例えば、保護用化粧品であってもよい。保護用化粧品の例としては、保護用乳液及び保護用クリームが挙げられる。実施形態に係るコラーゲン産生亢進剤は、例えば、ベースメークアップ化粧品であってもよい。ベースメークアップ化粧品の例としては、ファンデーション、白粉、及び化粧下地が挙げられる。実施形態に係るコラーゲン産生亢進剤は、例えば、ポイントメークアップ化粧品であってもよい。ポイントメークアップ化粧品の例としては、口紅、アイメークアップ、頬紅、及びネイルエナメルが挙げられる。
また、実施形態に係るコラーゲン産生亢進剤は、例えば、消毒薬、塗布治療薬等の皮膚外用薬、点眼剤、及び内服薬として供される。実施形態に係るコラーゲン産生亢進剤は、例えば、手足指を含む人体の皮膚、毛、口腔、及び眼球等に投与される。
実施形態に係るコラーゲン産生亢進剤は、シソ科植物の発酵物の他に、液体油脂、固体油脂、ロウ、炭化水素、高級脂肪酸、高級アルコール、エステル、シリコーン、アニオン界面活性剤、カチオン界面活性剤、両性界面活性剤、非イオン性界面活性剤、保湿剤、水溶性高分子、増粘剤、被膜剤、金属イオン封鎖剤、低級アルコール、多価アルコール、糖類、アミノ酸類、有機アミン類、pH調整剤、皮膚栄養剤、ビタミン類、酸化防止剤、香料、粉体、色材、及び水等の化粧品及び医薬品の配合成分を目的に応じて適宜含んでいてもよい。
実施形態に係るコラーゲン産生亢進剤が油性成分を含む場合、実施形態に係るコラーゲン産生亢進剤における油性成分の濃度は、特に限定されないが、例えば、0.1質量%以上90質量%以下、あるいは、0.5質量%以上90質量%以下である。実施形態に係るコラーゲン産生亢進剤が水性成分を含む場合、実施形態に係るコラーゲン産生亢進剤における水性成分の濃度は、特に限定されないが、例えば、0.1質量%以上90質量%以下、あるいは、0.5質量%以上90質量%以下である。実施形態に係るコラーゲン産生亢進剤における油性成分と水性成分の比は、実施形態に係るコラーゲン産生亢進剤がオイルインウォーター(O/W)型か、ウォーターインオイル(W/O)型かによって、適宜設定される。実施形態に係るコラーゲン産生亢進剤が界面活性剤を含む場合、実施形態に係るコラーゲン産生亢進剤における界面活性剤の濃度は、特に限定されないが、例えば2質量%以上10質量%以下である。
実施形態に係るコラーゲン産生亢進剤は、シソ科植物を発酵させ、発酵物を得ることにより製造される。シソ科植物を発酵させる際には、塩、及び糖蜜等の糖がシソ科植物に添加される。発酵温度は、例えば30℃である。得られる発酵液の水素イオン指数(pH)は、4.0前後である。発酵液から、ラクトバシラス属の分泌物を抽出してもよい。
得られた発酵液を加熱し、発酵液に含まれるラクトバシラス属を死菌にしてもよい。したがって、発酵液は、熱処理発酵液であってもよい。また、発酵液を噴霧乾燥し、ラクトバシラス属の菌体乾燥物を得てもよい。菌体乾燥物は、凍結乾燥法及び熱風乾燥法等によっても作製可能である。
さらに、得られた発酵液、ラクトバシラス属の菌体、あるいはラクトバシラス属の菌体乾燥物を豆乳に添加し、豆乳を発酵させて豆乳発酵液を得てもよい。豆乳発酵液も、コラーゲン産生亢進剤効果を奏する。
以上のとおり、本発明の各実施形態に係るコラーゲン産生亢進剤等は、上述したいずれか1つ又は複数の組み合わせによる以下の例による構成及び作用効果を有する。

以下に本発明の実施例を説明する。ただし、本発明は以下の実施例に限定されないことはもちろんである。
(実施例1:シソ科植発酵物)
シソ科植発においては、一日のうち、日の出時間を挟む前後1時間、合計2時間の間に、乳酸菌の数が最大化するとされている。また、この時間帯以外においては、乳酸菌が減少し、光合成菌が増加するとされている。したがって、この2時間の間に、延命草の先端から約20cmの部分を採取した。採取した6.3kgの延命草を、すぐさま、中にビニールの袋を敷いた第1の漬物樽に入れ、延命草に、3.2kgの糖蜜と0.6kgの粗塩を振りかけた後、ビニール袋の口を閉じて密封した。ビニール袋の上から、重石を載せ、延命草を漬け込んだ。
シソ科植発においては、一日のうち、日の出時間を挟む前後1時間、合計2時間の間に、乳酸菌の数が最大化するとされている。また、この時間帯以外においては、乳酸菌が減少し、光合成菌が増加するとされている。したがって、この2時間の間に、延命草の先端から約20cmの部分を採取した。採取した6.3kgの延命草を、すぐさま、中にビニールの袋を敷いた第1の漬物樽に入れ、延命草に、3.2kgの糖蜜と0.6kgの粗塩を振りかけた後、ビニール袋の口を閉じて密封した。ビニール袋の上から、重石を載せ、延命草を漬け込んだ。
漬け汁が延命草の上まで上がった数日後、重石を外した。次に、第2の漬物樽に、すすぎ洗い出し用の塩素を含まない10Lの水を入れ、水の中に、延命草の漬物と、10kgの漬け汁を入れた。さらに、第3の漬物樽を用意し、第3の漬物樽の開口上に金網フィルタを載せた。第2の漬物樽から、手でもみ洗いしながら少しずつ延命草を取り出し、第3の漬物樽の開口上の金網フィルタに延命草を軽く掌で押さえて、漬け汁を絞った。
延命草を全て絞った後、第2の漬物汁に残っていた漬け汁を金網フィルタに通してろ過した。次に、第3の漬物樽の中の漬け汁に、糖蜜(波照間黒糖)を終濃度が10重量%となるよう、また粗塩を終濃度が3重量%となるよう溶かし入れた。その後、第3の漬物樽の周囲温度を約30℃にすることにより、発酵を開始させた。最初に大きな泡の発泡が確認され、徐々に細かい泡の発泡に変わっていき、最後に発泡が収まった。約1週間後、発泡が収まった時のpHは、3.8付近であった。このときの漬け汁を、延命草発酵液とした。得られた延命草発酵液の一部を70℃で30分間加熱し、乳酸菌等の菌を死滅させた熱処理延命草発酵液を得た。
(実施例2:シソ科植発酵物を用いた豆乳発酵液)
豆乳を70℃に加熱し、約30分間過熱滅菌処理を行った。加熱滅菌処理した豆乳に、実施例1で用意した熱処理していない延命草発酵液を、終濃度が約10wt%となるよう加え、十分攪拌した。その後、熱処理していない延命草発酵液を添加した豆乳を、37℃で24時間発酵させた。発酵させた後、濾過によって固形分を除去し、延命草発酵液を含む豆乳発酵液を得た。
豆乳を70℃に加熱し、約30分間過熱滅菌処理を行った。加熱滅菌処理した豆乳に、実施例1で用意した熱処理していない延命草発酵液を、終濃度が約10wt%となるよう加え、十分攪拌した。その後、熱処理していない延命草発酵液を添加した豆乳を、37℃で24時間発酵させた。発酵させた後、濾過によって固形分を除去し、延命草発酵液を含む豆乳発酵液を得た。
(実施例3:シソ科植発酵物によるコラーゲン産生亢進)
正常ヒト真皮線維芽細胞をDMEM培地(+5%FBS)を用いて培養器中でコンフルエントな状態になるまで維持した。コンフルエントに達した後、培養器から培地を除去し、600μmol/Lの過酸化水素(H2O2)を含有するDMEM(0%FBS)を培養器に添加し、37℃にて細胞を1時間培養した。1時間培養後、H2O2含有DMEM(0%FBS)を培養器から除去し、DMEM培地(+10%FBS)を培養器に添加した。この操作を、4日間繰返した後、DMEM(10%FBS)でさらに3日間細胞を培養し、得られた細胞を老化誘導処理細胞とした。
正常ヒト真皮線維芽細胞をDMEM培地(+5%FBS)を用いて培養器中でコンフルエントな状態になるまで維持した。コンフルエントに達した後、培養器から培地を除去し、600μmol/Lの過酸化水素(H2O2)を含有するDMEM(0%FBS)を培養器に添加し、37℃にて細胞を1時間培養した。1時間培養後、H2O2含有DMEM(0%FBS)を培養器から除去し、DMEM培地(+10%FBS)を培養器に添加した。この操作を、4日間繰返した後、DMEM(10%FBS)でさらに3日間細胞を培養し、得られた細胞を老化誘導処理細胞とした。
細胞が老化誘導されたことは、老化マーカーである、senescence-associated beta-galactosidase(SA-β-Gal)染色にて確認した。
老化誘導処理細胞を、DMEM培地(+5%FBS)を用いて、5.0×104cells/wellの細胞密度にて48穴プレートに播種した。播種24時間後、培地を、1.0%及び10.0%の濃度で実施例1で用意した熱処理をしていない延命草発酵液を含むDMEM培地(+0.5%FBS)、25μmol/LでビタミンCリン酸マグネシウムを含むDMEM培地(+0.5%FBS)、25μmol/LでビタミンCを含むDMEM培地(+0.5%FBS)、及びDMEM培地(+0.5%FBS)のそれぞれと交換した。その後、48時間細胞を培養した後、培地を回収し、培地中のI型コラーゲン量をELISA法(Anti-Human Collagen Type I抗体(Rabbit)を用いた直接法)で定量した。
その結果、図1に示すように、ビタミンCリン酸マグネシウム又はビタミンCを含む培地(陽性コントロール)で細胞を培養すると、I型コラーゲンの産生が亢進されたことが示された。また、延命草発酵液を添加された培地で細胞を培養した場合も、I型コラーゲンの産生が亢進されたことが示された。
(実施例4:シソ科植発酵物の抗菌効果)
グラム陰性球菌として大腸菌、サルモネラ菌、腸炎ビブリオ菌、及び肺炎桿菌を用意し、グラム陰性桿菌として緑膿菌を用意した。また、グラム陽性球菌として黄色ブドウ球菌及びMRSAを用意し、グラム陽性桿菌として枯草菌及びセレウス菌を用意した。
グラム陰性球菌として大腸菌、サルモネラ菌、腸炎ビブリオ菌、及び肺炎桿菌を用意し、グラム陰性桿菌として緑膿菌を用意した。また、グラム陽性球菌として黄色ブドウ球菌及びMRSAを用意し、グラム陽性桿菌として枯草菌及びセレウス菌を用意した。
10mLの実施例1で用意した熱処理をしていない延命草発酵液に、上記の菌のいずれかを107個/mLの濃度で含む0.1mLの菌液を接種し、25℃で作用させ経時的に接種した菌の生菌数を24時間測定した。コントロールは、延命草発酵液をリン酸緩衝液(1/15mol/L、pH7.2)に代えた以外は同じである。その結果、図2に示すように、延命草発酵液は、用意した全ての種類の細菌を24時間以内に減少させた。なお、実施例2で用意した延命草発酵液を含む豆乳発酵液を用いた場合も、同様の結果が得られた。
(実施例5:シソ科植発酵物の抗ウイルス効果)
エンベロープウイルスとして、インフルエンザウイルスA型(H1N1)の培養液を用意した。また、ノンエンベロープウイルスとして、ノロウイルスの代替ウイルスであるネコカリシウイルスの培養液を用意した。ウイルスの培養液は、精製水で、10倍ずつ段階希釈した。その後、50%組織培養感染量(TCID50:50% Tissue Culture Infectious Dose)に従い、実施例1で用意した熱処理をしていない延命草発酵液による抗ウイルス試験を室温で実施した。抗ウイルス試験は、日本食品分析センターで実施された。
エンベロープウイルスとして、インフルエンザウイルスA型(H1N1)の培養液を用意した。また、ノンエンベロープウイルスとして、ノロウイルスの代替ウイルスであるネコカリシウイルスの培養液を用意した。ウイルスの培養液は、精製水で、10倍ずつ段階希釈した。その後、50%組織培養感染量(TCID50:50% Tissue Culture Infectious Dose)に従い、実施例1で用意した熱処理をしていない延命草発酵液による抗ウイルス試験を室温で実施した。抗ウイルス試験は、日本食品分析センターで実施された。
その結果、図3に示すように、延命草発酵液は、1時間以内でインフルエンザウイルスとネコカリシの感染価を減少させた。なお、実施例2で用意した延命草発酵液を含む豆乳発酵液を用いた場合も、同様の結果が得られた。
以上説明した各実施形態及び実施例は、本発明の理解を容易にするためのものであり、本発明を限定して解釈するためのものではない。本発明は、その趣旨を逸脱することなく、変更/改良され得るとともに、本発明にはその等価物も含まれる。すなわち、各実施形態及び実施例に当業者が適宜設計変更を加えたものも、本発明の特徴を備えている限り、本発明の範囲に包含される。例えば、各実施形態及び実施例が備える各要素などは、例示したものに限定されるわけではなく適宜変更することができる。また、各実施形態及び実施例は例示であり、異なる実施形態で示した構成の部分的な置換又は組み合わせが可能であることはいうまでもなく、これらも本発明の特徴を含む限り本発明の範囲に包含される。

- 前記コラーゲンがI型コラーゲンである、請求項1から4のいずれか1項に記載のコラーゲン産生亢進剤。
- 前記シソ科植物由来の乳酸菌を備える、請求項1から5のいずれか1項に記載のコラーゲン産生亢進剤。
