2-(4-(2-((tert-ブチルジメチルシリル)オキシ)エチル)ピペラジン-1-イル)エタン-1-オール(中間体1)
窒素雰囲気下、室温にて1,4-ビス(2-ヒドロキシエチル)ピペラジン(CAS番号122-96-3、5.00g,28.7mmol)のクロロホルム(500mL)溶液に塩化tert-ブチルジメチルシリル(4.33g,28.7mmol)とトリエチルアミン(4.0mL,28.7mmol)を加え、一晩攪拌した。反応液を濃縮し、残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール=100/1~5/1)で2回精製し、標記中間体1(1.71g,21%)を無色油状物質で得た。
2-(4-(2-((tert-ブチルジメチルシリル)オキシ)エチル)ピペラジン-1-イル)エチル (R)-5-(1,2-ジチオラン-3-イル)ペンタン酸(中間体2)
窒素雰囲気下、室温にて(R)-α‐リポ酸(1.22g,5.91mmol)、2-(4-(2-((tert-ブチルジメチルシリル)オキシ)エチル)ピペラジン-1-イル)エタン-1-オール(中間体1、1.71g、5.93mmol)、および4-ジメチルアミノピリジン(217mg、1.78mmol)のMTBE(24mL)溶液に、DIC(821mg、6.51mmol)のMTBE(7.5mL)溶液を滴下し、得られた反応液を室温にて一晩攪拌した。反応液をろ過し、ろ液を減圧下濃縮して得られた残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル/メタノール=100/0~9/1)で精製し、標記中間体2(2.35g,83%)を黄色油状物質で得た。
1,3-ジモルホリノプロパン-2-オール(中間体3)
窒素雰囲気下、室温にて1,3-ジクロロプロパン-2-オール(500μl,5.31mmol)とモルホリン(3.0ml)を混合し、マイクロウェーブ装置を用い、150℃で30分間攪拌した。反応液に10%水酸化ナトリウム水溶液を加え、塩化メチレン/メタノール(10/1)で抽出した。有機層を無水硫酸ナトリウムで乾燥した後、ろ過後減圧下濃縮した。得られた残渣をシリカゲルカラムクロマトグラフィー(塩化メチレン/メタノール=100/0~9/1)で精製し、標記参中間体3(872mg,71%)を黄色油状物質で得た。
3-メチル-2-ブテン-1-イル (R)-5-(1,2-ジチオラン-3-イル)ペンタン酸(化合物1)
窒素雰囲気下、室温にて(R)-α‐リポ酸(3.0g,14.6mmol)、3-メチル-2-ブテン-1-オール(1.38g、16.0mmol)、4-ジメチルアミノピリジン(534mg、4.37mmol)のMTBE(24mL)溶液に、DIC(2.02g、16.0mmol)のMTBE(12mL)溶液を滴下し、得られた反応液を室温にて一晩攪拌した。反応溶液をろ過し、ろ液を減圧下濃縮して得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=100/0~9/1)で精製し、標記化合物1(1.71g,43%)を黄色油状物質で得た。
(R)-2-((5-(1,2-ジチオラン-3-イル)ペンタノイル)オキシ)-N-ベンジル-N,N-ジメチルエタン-1-アミニウムクロリド(化合物2)
市販のベンジル(2-ヒドロキシエチル)ジメチルアンモニウムクロリドを用い、溶媒としてMTBEの代わりにクロロホルムを用いて、化合物1と同様の方法で標記化合物2を得た(収率35%)。
2-(4-(2-ヒドロキシエチル)ピペラジン-1-イル)エチル (R)-5-(1,2-ジチオラン-3-イル)ペンタン酸(化合物3)
窒素雰囲気下、室温にて中間体2(2.35g,4.93mmol)の塩化メチレン(24mL)溶液に、フッ化テトラブチルアンモニウム(1Mテトラヒドロフラン溶液、14.8mL、14.8mmol)を加え、得られる反応液を室温にて4時間攪拌した。反応溶液を減圧下濃縮して得られた残渣をシリカゲルカラムクロマトグラフィーで(酢酸エチル~クロロホルム/メタノール=5/1)で精製したのち、再度NHシリカゲルカラムクロマトグラフィーで(酢酸エチル/クロロホルム=9/1~1/1~クロロホルム/メタノール=9/1)で精製し、標記化合物3(1.24g,69%)を黄色油状物質で得た。
2-モルホリノエチル (R)-5-(1,2-ジチオラン-3-イル)ペンタン酸(化合物4)
市販の2-モルホリノエタノールを用い、化合物1と同様の方法で標記化合物4を得た(収率21%)。

窒素雰囲気下、1,3-ジモルホリノプロパン-2-オール(中間体3、491mg、2.13mmol)、α-リポ酸(400mg、1.94mmol)、4-ジメチルアミノピリジン(71.1mg、0.582mmol)のMTBE(3.2ml)溶液に、室温にてDIC(330μl、2.13mmol)のMTBE(1.6ml)溶液を滴下し、得られる反応液を室温にて一晩攪拌した。反応溶液をろ過し、ろ液を減圧下濃縮して得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=9/1~0/100~酢酸エチル/メタノール=9/1)で精製し、標記化合物5(335mg,41%)を黄色油状物質で得た。
(2,2,5-トリメチル-4H-[1,3]ジオキソ[5,4-c]ピリジン-8-イル)メチル (R)-5-(1,2-ジチオラン-3-イル)ペンタン酸(化合物6)
アルゴン雰囲下、室温で(2,2,8-トリメチル-4H-[1,3] ジオキソ[4,5-c] ピリジン-5-yl)メタノール(1.00g、4.77mmol)、α-リポ酸(1.18g、5.72mmol)、4-ジメチルアミノピリジン(0.21g、1.72mmol)をMTBE (9.4ml)でスラリー溶液にし、氷冷下でDIC(0.98ml、4.27mmol)を加えた。5分後、室温に戻して3時間40分撹拌した。反応液をろ過し、ろ液を減圧下濃縮して得られる残渣をシリカゲルカラムクロマトグラフィー(クロロホルム/メタノール=40/1)にて精製し、標記化合物6(1.94g、85%)を黄色油状物質で得た。
化合物1~4による水晶体の弾性に対する作用を検討した。InvestOphthalmol Vis Sci、57、2851-2863、2016に記載の方法を参考に、評価試験を行った。
1)基剤の調製
0.1%(w/v) ピルビン酸エチル、0.269%(w/v) リン酸二水素ナトリウム・一水和物(NaH2PO4・H2O)、0.433%(w/v) リン酸水素二ナトリウム(Na2HPO4)、0.2%(w/v) ヒドロキシプロピルメチルセルロース、0.5%(w/v) NaCl、1.0%(w/v)ポリオキシル35ひまし油(以下、CO35ともいう)、および精製水(適量)を含有する基剤(水溶液)を調製した。
2)化合物1~4試料の調製
化合物1~4にそれぞれ基剤を加えて撹拌し、0.03%(w/v)の溶液を調製した。
3)EV06試料の調製
EV06は、下記式:
EV06に基剤を加えて撹拌し、0.03%(w/v)の溶液を調製した。
1)8ヵ月齢のC57BL/6Jマウスに各被験試料を1日3回(9:00、13:00、17:00を目安)、12~15日間、2.5 μL/eyeずつピペットマンを用いて右眼に点眼した。
2)最終点眼後に、マウスに二酸化炭素を吸引させ安楽殺し、眼球を摘出して、Hank‘s balanced salt solution(HBSS)でリンスした。
3)視神経近くの強膜をカミソリで切り、水晶体を切開部から摘出し、摘出した水晶体はHBSSに浸した。
4)水晶体をスライドガラスの上にのせ、オールインワン蛍光顕微鏡BZ-9000(キーエンス)を用いて水晶体の画像を取り込んだ(画像a)。
5)次に、水晶体の上に1枚のカバーガラス(Corning(登録商標) 22x22mm Square)を載せ、重さにより水晶体の厚さが変動した画像を同様に取り込んだ(画像b)。
6)水晶体直径の変化量を下記計算式1(すなわち、画像bの水晶体直径から画像aの水晶体直径を引く)より算出した。ついで、基剤群と比較した各試料群の水晶体弾性向上量を下記計算式2より算出した。なお、平均値は、基剤群は5例、各化合物1~4試料群は10例、EV06試料群は9例の平均である。

1)基剤の調製

0.1%(w/v) ピルビン酸エチル、0.269%(w/v) リン酸二水素ナトリウム・一水和物(NaH2PO4・H2O)、0.433%(w/v) リン酸水素二ナトリウム(Na2HPO4)、0.2%(w/v) ヒドロキシプロピルメチルセルロース、0.5%(w/v) NaCl、0.2%(w/v) CO35、および精製水(適量)を含有する基剤A(水溶液)を調製した。

化合物3に基剤Cを加えて撹拌し、1.5%(w/v)溶液を調製し、希釈液で希釈して0.3%(w/v)溶液を調製した。さらに、調製した0.3%(w/v)溶液を基剤Aで希釈して0.03%(w/v)溶液を調製した。さらに、調製した0.03%(w/v)溶液を基剤Aで希釈して0.003%(w/v)溶液を調製した。
化合物4に基剤Dを加えて撹拌し、1.5%(w/v)エマルションを調製し、希釈液で希釈して0.3%(w/v)エマルションを調製した。さらに、調製した0.3%(w/v)エマルションを希釈液で希釈して0.03%(w/v)エマルションを調製した。さらに、調製した0.03%(w/v)エマルションを希釈液で希釈して0.003%(w/v)エマルションを調製した。
EV06に基剤Aを加えて撹拌し、1.5%(w/v)溶液を調製し、化合物1および3の試験の比較に用いた。
EV06に基剤Bを加えてソニケートし、1.5%(w/v)エマルションを調製し、化合物4の試験の比較に用いた。
1)7~8ヵ月齢のC57BL/6Jマウスに各被験試料を1日3回(9:00、13:00、17:00を目安)、12~15日間、2.5 μL/eyeずつピペットマンを用いて右眼に点眼した。

EV06に基剤Cを加えて撹拌し、0.03%(w/v)溶液を調製し、化合物5および化合物6の試験の比較に用いた。
1)9ヵ月齢のC57BL/6Jマウスに各被験試料を1日3回(9:00、13:00、17:00を目安)、14日間、2.5 μL/eyeずつピペットマンを用いて両眼に点眼した。

EV06および化合物1~4の1.5%(w/v)点眼液をウサギに単回点眼し、リポ酸の房水移行性を評価した。
薬理試験2の被験試料の調製と同様の方法により、EV06および化合物1、3、4の1.5%(w/v)点眼液を調製した。なお、EV06の1.5%(w/v)点眼液は、基剤Cを用いて調製した。また、化合物2に基剤Cを加えて撹拌し、化合物2の1.5%(w/v)点眼液を調製した。
ウサギ(雄性日本白色種)の片眼に1.5%(w/v)EV06点眼液を、対側眼に1.5%(w/v)化合物1、2、3または4の点眼液をそれぞれ25μLで1回点眼した。点眼0.5、1、2、および4時間後にウサギ眼球へ局所麻酔を施した後、房水を採取した(各時点4眼)。高速液体クロマトグラフ・タンデムマススペクトロメーター(LC-MS/MS)を用いて、房水中リポ酸濃度を測定した。
0.03%(w/v)化合物1点眼液、0.03%(w/v)化合物3点眼液および1.5%(w/v)EV06点眼液をそれぞれウサギに単回点眼し、リポ酸の房水および水晶体移行性を評価した。
薬理試験2の被験試料の調製と同様の方法により、0.03%(w/v)化合物1点眼液、0.03%(w/v)化合物3点眼液および1.5%(w/v)EV06点眼液を調製した。
ウサギ(雄性日本白色種)の片眼に1.5%(w/v)EV06点眼液を、対側眼に0.03%(w/v)化合物1点眼液または0.03%化合物3点眼液をそれぞれ25μLで1回点眼した。点眼0.5および1時間後にウサギを屠殺して房水及び水晶体を採取し、房水および水晶体中のリポ酸濃度を高速液体クロマトグラフ・タンデムマススペクトロメーター(LC-MS/MS)を用いて測定した。
(試料調製)
薬理試験2の被験試料の調製と同様の方法により、0.03%(w/v)および0.3%(w/v)化合物1点眼液、ならびに、0.03%(w/v)および0.3%(w/v)化合物3点眼液を調製した。なお、基剤群には、基剤Aを用いた。
化合物1点眼液および化合物3点眼液の点眼群
0.03%(w/v)および0.3%(w/v)化合物1点眼液、0.03%(w/v)および0.3%(w/v)化合物3点眼液、並びに、基剤をピペットで日本白色種ウサギの左眼に30分間隔で10回点眼(50μL/眼/回)し、最終点眼後1時間の前眼部刺激症状観察(改良Draize法)およびスリットランプ検査を実施した。なお、対側眼は無処置とした。
前眼部刺激症状観察は、以下の基準によりスコア化した。
+0.5:ごく軽度、+1:軽度、+2:中等度、+3:重度
試験結果を表11に示す。化合物1点眼液点眼眼では前眼部刺激症状は認められず、化合物3点眼液点眼眼についても、そのスコアは低かった。また、化合物1点眼液および化合物3点眼液点眼眼のスリットランプ検査では異常は認められなかった。
化合物1点眼液および化合物3点眼液の安全性は、高いことが示された。
(試料調製)
化合物1に基剤Cを加えて撹拌し、0.3%(w/v)溶液を調製し、基剤Cで希釈して0.1%(w/v)溶液を調製した。さらに、調製した0.1%(w/v)溶液を基剤Cで希釈して0.03%(w/v)溶液を調製した。
0.03%(w/v)、0.1%(w/v)および0.3%(w/v)化合物1点眼液をピペットで日本白色種ウサギの左眼に6時間間隔で1日2回、2週間反復点眼(50μL/眼/回)し、最終点眼後1時間にスリットランプを用いた前眼部刺激症状観察(McDonald-Shadduck法)および水晶体観察を実施した。投与期間終了後、眼局所の病理組織学的検査を実施した。なお、対側眼は無処置とした。
前眼部刺激症状観察は、以下の基準によりスコア化した。
+1:軽度、+2:中等度、+3:重度
化合物1点眼液の安全性は、高いことが示された。
