眼の調節力はジオプター(D)として測定され、次の式(1)により求めることができる。

視力は、近見視力(裸眼、遠見矯正下、矯正)として測定され、少数視力、分数視力、logMARより求めることができる。
例えば、点眼剤の場合、1回量1~5滴、好ましくは1~3滴、より好ましくは1~2滴、特に好ましくは1滴を、1日1~4回、好ましくは1日1~3回、より好ましくは1日1~2回、特に好ましくは1日1回を、毎日~1週間毎に点眼投与することができる。ここで、1滴は、通常、約0.01~約0.1mLであり、好ましくは、約0.015~約0.07mLであり、より好ましくは、約0.02~約0.05mLであり、特に好ましくは約0.03mLである。
本発明の化合物は、有機合成の当業者によく知られている多くの方法により製造することができる。本発明の化合物は、有機合成化学の分野にて知られている合成方法とともに、下記の実施例の方法、またはそれについて当業者により十分認識される変形を用いて合成することができる。好ましい方法としては、下記の実施例に記載のものが挙げられるが、これらに限定されない。
アルゴン雰囲気下、室温にてα-リポ酸(2.00g、9.69mmol)のN,N-ジメチルホルムアミド溶液(20ml)にヨウ化メチル(0.728ml、11.7mmol)と炭酸カリウム(2.68g、19.4mmol)を加え、室温にて一晩攪拌した。反応溶液をろ過し炭酸カリウムをろ去する。炭酸カリウムを酢酸エチルで洗浄し、有機層を集めて飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した。ろ過し、ろ液を減圧下濃縮して得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=10/1)で精製し、標記中間体1(1.69g,79%)を黄色油状物質で得た。
アルゴン雰囲下、室温にて(R)-5-(1,2-ジチオラン-3-イル)ペンタン酸メチル 中間体1(796mg、3.63mmol)の無水テトラヒドロフラン溶液(10ml)に水素化ホウ素リチウム(94.8mg、4.35mmol)を加えた。反応溶液を1.5時間還流した後、さらに反応液に水素化ホウ素リチウム(23.7mg、1.09mmol)を加え、1時間還流した。室温まで冷却し、1M塩酸と水を加え、pH5~6とし、酢酸エチルで抽出した。酢酸エチル層を飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥し、ろ過後、ろ液を減圧下濃縮して得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=2/1)にて精製し、標記化合物1(625mg、89%)を無色油状物質で得た。
DL-α-リポ酸(1.38g、6.69mmol)のN,N-ジメチルホルムアミド溶液(10ml)に、室温にてヨウ化メチル(0.54ml、8.67mmol)と炭酸カリウム(1.39g、10.1mmol)を加え、室温にて3時間攪拌した。反応溶液に水(40ml)を加え、酢酸エチル(40ml)で2度抽出した。酢酸エチル層を水(80ml)および飽和食塩水(80ml)で洗浄し、無水硫酸マグネシウムで乾燥し、ろ過した。ろ液を減圧下濃縮し、得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=19/1)で精製し、標記中間体2(3.07g,57%)を黄色油状物質で得た。
室温にて5-(1,2-ジチオラン-3-イル)ペンタン酸メチル 中間体2(0.52g、2.36mmol)のテトラヒドロフラン溶液(5ml)に水素化ホウ素リチウム(151mg、6.93mmol)を加え、5時間還流した。水と1M塩酸を加え、pH5~6とし、酢酸エチルで抽出した。酢酸エチル層を飽和食塩水で洗浄後、無水硫酸マグネシウムで乾燥し、ろ過した。ろ液を減圧下濃縮し、得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=19/1→10/1)にて精製し、標記化合物2(150mg、33%)を無色油状物質として、また標記化合物3(27.1mg、6%)を黄色油状物質として得た。
化合物2(300mg、1.55mmol)をテトラヒドロフラン(200ml)に溶解し、1規定水酸化ナトリウム水溶液(約1ml)を加えてpHを7~8とした後室温で4日間攪拌した。反応液を減圧下濃縮し、水(20ml)およびクロロホルム(20ml)で分配した。クロロホルム層を水(20ml)、0.1規定塩酸(20ml)、水(20ml)および飽和食塩水で洗浄した。クロロホルム層を、無水硫酸マグネシウムで乾燥し、ろ過した。ろ液を減圧下濃縮し、得られる残渣をシリカゲルカラムクロマトグラフィー(ヘキサン/酢酸エチル=10/1)にて精製し、標記化合物3(183mg、62%)を無色油状物質として得た。
1,3-プロパンジチオール(20g, 212.3 mmol), アセトン (13.57 g, 233.6 mmol) および触媒量のp-トルエンスルホン酸をトルエン(400mL)に加え、ディーン・スタークを用いて12時間還流した。反応液を室温まで放冷し、得られた混合物を5%水酸化ナトリウム水溶液(100ml、2回)、水(100ml)および飽和食塩水(100ml)で順次洗浄した。有機層を無水硫酸マグネシウムで乾燥後、減圧下溶媒を留去することにより、標記中間体3 (20.7g, 66%) を無色油状物質で得た。
窒素雰囲気下、2,2-ジメチルメチル-1,3-ジチアン 中間体3(20.7g、139.9mmol) のメタノール溶液 (1500ml)を-5度に冷却し、過ヨウ素酸ナトリウム(29.9g、139.9mmol)水溶液(250ml)を-5度に保ち滴下した。反応溶液を20度以下で30分攪拌した後、塩化メチレンでろ過洗浄(200ml、3回)した。減圧下、ろ液を濃縮し、得られた残留物を逆相クロマトグラフィーにて精製した(Column, C18 silica gel; アセトニトリル/水、10%から50%)。目的物を含むフラクションを回収、減圧下濃縮後、塩化メチレン(1000ml、3回)で抽出した。合わせた有機層を飽和食塩水(4000ml)で洗浄後、無水硫酸ナトリウムで乾燥し、減圧下溶媒を留去することにより、標記中間体4(20.8g、78%)を淡黄色油状物質で得た。
窒素雰囲気下、6‐ブロモ‐1‐ヘキサノール(4.2g、23.2mmol)の塩化メチレン溶液(50ml)に、1H‐イミダゾール(2.37g, 34.8mmol)、塩化tert‐ブチルジフェニルシラン(9.56g、34.8mmol)を順次加えた。 反応液を室温で終夜攪拌した。反応液に水(150ml)を加え、塩化メチレン(150ml、3回)で抽出した。有機層を飽和食塩水(450ml、3回)で洗浄し、無水硫酸ナトリウムで乾燥後、減圧下溶媒を留去した。得られた残留物をシリカゲルカラムクロマトグラフィー(酢酸エチル/石油エーテル、0~8%)にて精製し、標記中間体5-1(8.9g、91%)を油状物質で得た。
上記中間体5-1の合成方法と同様の方法にしたがって、4‐ブロモ‐1‐ブタノール(2.75g、18.0mmol)より標記中間体5-2(3.6g、51%)を無色油状物質として得た。
上記中間体5-1の合成方法と同様の方法にしたがって、2‐(2-ブロモエトキシ)エタノール(4.5g、26.6mmol)より標記中間体5-3(10g、92%)を黄色油状物質として得た。
2,2-ジメチル-1,3-ジチアン-1-イウム-1-オレート 中間体4(1.74g, 10.6mmol)のテトラヒドロフラン溶液(12ml)に、テトラメチルエチレンジアミン(1.85g, 15.9mmol)および2M リチウムジイソプロピルアミド-テトラヒドロフラン溶液(7.94ml, 15.9mmol) を窒素雰囲気下‐78度で順次加えた。反応液を‐78度で20分攪拌した。この反応溶液に、[(6‐ブロモヘキシル)オキシ](tert‐ブチル)ジフェニルシラン 中間体5-1(4.89g, 11.7mmol)を‐78度で加え、更に20分攪拌した。反応液を室温に戻し、90分攪拌した。室温で、反応液に塩化アンモニウム水溶液(150ml)を加えた後、酢酸エチル(150ml、3回)で抽出した。有機層を飽和食塩水(450ml、3回)で洗浄後、無水硫酸ナトリウムで乾燥し、減圧下、溶媒を留去した。得られた残留物をシリカゲルカラムクロマトグラフィー(酢酸エチル/石油エーテル、0~48%)にて精製し、標記中間体6-1(3.2g、60%)を無色油状物質で得た。
上記中間体6-1の合成方法と同様の方法にしたがって、2,2-ジメチル-1,3-ジチアン-1-イウム-1-オレート 中間体4(1.58g、9.61mmol)および[(4‐ブロモブチル)オキシ](tert‐ブチル)ジフェニルシラン 中間体5-2より標記中間体6-2(3.43g、75%)を無色油状物質として得た。
上記中間体6-1の合成方法と同様の方法にしたがって、2,2-ジメチル-1,3-ジチアン-1-イウム-1-オレート 中間体4(2.2g、13.4mmol)および[2-(2‐ブロモエトキシ)エトキシ](tert‐ブチル)ジフェニルシラン 中間体5-3(6.0g、14.7mmol)より標記中間体6-3(5.5g、83%)を無色油状物質として得た。
窒素雰囲気下、6‐(2‐{2‐[(tert‐ブチルジフェニルシリル)オキシ]エトキシ}エチル)‐2,2‐ジメチル‐1,3‐ジチアン‐1‐イウム‐1‐オレート 中間体6-1(3.3g, 66.6mmol)のアセトニトリル溶液(33ml)に2M塩酸(1.64ml, 3.28mmol)を滴下し、40度で終夜攪拌した。反応液に炭酸水素ナトリウム水溶液(10ml)を加えた後、塩化メチレン(100ml、3回)で抽出した。有機層を飽和食塩水(400ml)で洗浄し、無水硫酸ナトリウムで乾燥後、減圧下溶媒を留去した。得られた残留物を逆相クロマトグラフィーにて精製した(Column, C18 silica gel; アセトニトリル/水、5%~95%、45分)。目的物を含むフラクションを回収、減圧下濃縮後、塩化メチレン(100ml、3回)で抽出した。有機層を飽和食塩水(400ml)で洗浄し、無水硫酸ナトリウムで乾燥後、減圧下溶媒を留去し標記化合物5(320mg、24%)を淡黄色油状物質として得た。
上記化合物5の合成方法と同様の方法にしたがって、6‐{4‐[(tert‐ブチルジフェニルシリル)オキシ]ブチル}‐2,2‐ジメチル‐1,3‐ジチアン‐1‐イウム‐1‐オレート 中間体6-2(2.93g、6.2mmol)より標記化合物6(413mg、36%)を黄色油状物質として得た。
上記化合物5の合成方法と同様の方法にしたがって、6‐(2‐{2‐[(tert‐ブチルジフェニルシリル)オキシ]エトキシ}エチル)‐2,2‐ジメチル‐1,3‐ジチアン‐1‐イウム‐1‐オレート 中間体6-3(5.0g、10.2mmol)より標記化合物7(718mg、36%)を黄色油状物質として得た。
リポ酸コリンエステル(EV06)による水晶体の弾性に対する作用を確認した。InvestOphthalmol Vis Sci、57、2851-2863、2016に記載の方法を参考に、評価試験を行った。EV06は、下記式:
1)基剤の調製
0.1%(w/v) ピルビン酸エチル、0.269%(w/v) リン酸二水素ナトリウム一水和物(NaH2PO4・H2O)、0.433%(w/v) リン酸水素二ナトリウム(Na2HPO4)、0.2%(w/v) ヒドロキシプロピルメチルセルロース、0.5%(w/v) NaCl、および精製水(適量)を含有する基剤を調製した。
EV06に基剤を加えてソニケートし、5%(w/v)の懸濁液を調製した。調製した5%(w/v)懸濁液を基剤で希釈して1.5%(w/v)溶液を調製した。さらに、調製した1.5%(w/v)溶液を基剤で希釈して0.5%(w/v)溶液を調製した。それぞれ1日分を用時調製した。
1)8ヵ月齢のC57BL/6Jマウスに各被験試料を1日3回(9:00、13:00、17:00を目安)、15~17日間、2.5 μL/eyeずつピペットマンを用いて右眼に点眼した。
2)最終点眼後に、マウスに二酸化炭素を吸引させ安楽殺し、眼球を摘出して、Hank‘s balanced salt solution(HBSS)でリンスした。
3)視神経近くの強膜をカミソリで切り、水晶体を切開部から摘出し、摘出した水晶体はHBSSに浸した。
4)水晶体をスライドガラスの上にのせ、オールインワン蛍光顕微鏡BZ-9000(キーエンス)を用いて水晶体の画像を取り込んだ(画像a)。
5)次に、水晶体の上に1枚のカバーガラス(Corning(登録商標) 22x22mm Square)を載せ、重さにより水晶体の厚さが変動した画像を同様に取り込んだ(画像b)。
6)画像bの水晶体直径から画像aの水晶体直径を引き、水晶体直径の変化量を下記計算式1より算出した。ついで、基剤群と比較した各試料群の水晶体弾性向上量を下記計算式2より算出した。なお、平均値は、基剤群は6眼、各EV06試料群は12眼の平均である。

化合物1に基剤を加えて撹拌し、0.03%(w/v)溶液を調製した。調製後、冷蔵保管した。
EV06に基剤を加えて撹拌し、0.03%(w/v)の溶液を調製し、化合物1の試験の比較に用いた。調製後、冷蔵保管した。
1)9ヵ月齢のC57BL/6Jマウスに各被験試料を1日3回(9:00、13:00、17:00を目安)、14日間、2.5 μL/eyeずつピペットマンを用いて両眼に点眼した。
2)最終点眼後に、マウスに二酸化炭素を吸引させ安楽殺し、眼球を摘出して、Hank‘s balanced salt solution(HBSS)でリンスした。
3)視神経近くの強膜をカミソリで切り、水晶体を切開部から摘出し、摘出した水晶体はHBSSに浸した。
4)水晶体をスライドガラスの上にのせ、オールインワン蛍光顕微鏡BZ-9000(キーエンス)を用いて水晶体の画像を取り込んだ(画像a)。
5)次に、水晶体の上に1枚のカバーガラス(Corning(登録商標) 22x22mm Square)を載せ、重さにより水晶体の厚さが変動した画像を同様に取り込んだ(画像b)。
6)水晶体直径の変化量を下記計算式1(すなわち、画像bの水晶体直径から画像aの水晶体直径を引く)より算出した。ついで、基剤群と比較した各試料群の水晶体弾性向上量を下記計算式2より算出した。なお、平均値は、基剤群は8眼、化合物1試料群およびEV06試料群は10眼の平均である。

一方、0.03%EV06試料群は基剤群と比べて水晶体直径の増加は殆どなく、水晶体弾性改善作用を示さなかった。
化合物1の1日1回、2週間点眼による水晶体の弾性に対する作用を検討した。

水晶体直径の変化量=各被験試料の画像bの水晶体直径-各被験試料の画像aの水晶体直径
各試料群の水晶体弾性向上量=各被験試料群の水晶体直径の変化量の平均値-基剤群の水晶体直径の変化量の平均値
化合物5、6、7の1日1回、2週間点眼による水晶体の弾性に対する作用を検討した。
1)基剤の調製
1.0%(w/v) CO35、0.1%(w/v) ピルビン酸エチル、0.269%(w/v) リン酸二水素ナトリウム一水和物(NaH2PO4・H2O)、0.433%(w/v) リン酸水素二ナトリウム(Na2HPO4)、0.2%(w/v) ヒドロキシプロピルメチルセルロース、0.5%(w/v) NaCl、および精製水(適量)を含有する基剤を調製した。
2)化合物5、6、7試料の調製
化合物5に基剤を加えて撹拌し、0.1%(w/v)懸濁液を調製した。調製した0.1%(w/v)懸濁液を基剤で希釈して0.03%(w/v)懸濁液を調製した。
化合物6に基剤を加えて撹拌し、0.1%(w/v)懸濁液を調製した。調製した0.1%(w/v)懸濁液を基剤で希釈して0.03%(w/v)懸濁液を調製した。
化合物7に基剤を加えて撹拌し、0.1%(w/v)懸濁液を調製した。
いずれも調製後、冷蔵保存した。
3)EV06試料の調製
EV06に基剤を加えて撹拌し、0.03%(w/v)の溶液を調製し、化合物5、6、7の試験の比較に用いた。調製後、冷蔵保存した。
1)8ヵ月齢のC57BL/6Jマウスに各被験試料を1日1回(13:30を目安)、14日間、2.5 μL/eyeずつピペットマンを用いて両眼に点眼した。
2)最終点眼後に、マウスに二酸化炭素を吸引させ安楽殺し、眼球を摘出して、Hank‘s balanced salt solution(HBSS)でリンスした。
3)視神経近くの強膜をカミソリで切り、水晶体を切開部から摘出し、摘出した水晶体はHBSSに浸した。
4)水晶体をスライドガラスの上にのせ、オールインワン蛍光顕微鏡BZ-9000(キーエンス)を用いて水晶体の画像を取り込んだ(画像a)。
5)次に、水晶体の上に1枚のカバーガラス(Corning(登録商標) 22x22mm Square)を載せ、重さにより水晶体の厚さが変動した画像を同様に取り込んだ(画像b)。
6)画像bの水晶体直径から画像aの水晶体直径を引き、水晶体直径の変化量を下記計算式1より算出した。ついで、基剤群と比較した各試料群の水晶体弾性向上量を下記計算式2より算出した。なお、各群の平均値は、8眼の平均である。
水晶体直径の変化量=各被験試料の画像bの水晶体直径-各被験試料の画像aの水晶体直径
各試料群の水晶体弾性向上量=各被験試料群の水晶体直径の変化量の平均値-基剤群の水晶体直径の変化量の平均値
0.03%EV06試料群は水晶体弾性改善作用を示さなかった。
0.1%化合物7試料群は強力な水晶体弾性改善作用を示した。
(試料調製)
薬理試験1の被験試料の調製と同様の方法により基剤および0.03%(w/v)化合物1点眼液を調製した。
基剤、ならびに、0.03%(w/v)化合物1点眼液をピペットで日本白色種ウサギの片眼に30分間隔で10回点眼(50μL/眼/回)し、最終点眼後1時間にスリットランプを用いた前眼部刺激症状観察(McDonald-Shadduck法)を実施した。なお、角膜上皮障害性観察は最終点眼後3時間に実施した。
前眼部刺激症状観察は、以下の基準によりスコア化した。
+1:軽度、+2:中等度、+3:重度
化合物1点眼液の安全性は、高いことが示された。
(試料調製)
薬理試験4の被験試料の調製と同様の方法により基剤、ならびに、0.01%(w/v)および0.1%(w/v)化合物4点眼液を調製した。
基剤、ならびに、0.01%(w/v)および0.1%(w/v)化合物4点眼液をピペットで日本白色種ウサギの片眼に30分間隔で10回点眼(50μL/眼/回)し、最終点眼後1時間にスリットランプを用いた前眼部刺激症状観察(McDonald-Shadduck法)を実施した。なお、角膜上皮障害性観察は最終点眼後3時間に実施した。
前眼部刺激症状観察は、以下の基準によりスコア化した。
+1:軽度、+2:中等度、+3:重度
化合物4点眼液の安全性は、高いことが示された。
Claims (26)
- 式[I]:

であり、ここでA部は同一または異なった1~3個のR1で置換されていてもよく;
R1は、各々独立して、-OH、ハロ、C1-3アルキル、およびC1-3アルコキシからなる群から選択され;
Wは、C1-10アルキレンであり;ここで該アルキレン中の1または2個のメチレンは、各々独立して、O、NR2(ここでR2はHまたはC1-3アルキルである)、およびS(O)0-2からなる群から選択される二価基と置き換わっていてもよく;また該アルキレンは―OH、ハロ、C1-3アルキル、およびC1-3アルコキシからなる群から独立して選択される少なくとも1つの置換基で置換されていてもよい]
で表される化合物または医薬的に許容されるその塩を有効成分として含有する、老視の治療または予防のための剤/組成物。 - 該眼疾患が眼の調節力の低下を伴う眼疾患である、請求項2に記載の剤/組成物。
- 該眼疾患が老視である、請求項2~4のいずれか一項に記載の剤/組成物。
- Wが、-(CH2)1-10-であるアルキレンであり、該アルキレンは―OH、ハロ、C1-3アルキル、およびC1-3アルコキシからなる群から独立して選択される少なくとも1つの置換基で置換されていてもよい、請求項1~10のいずれか一項に記載の剤/組成物。
- Wが、-(CH2)1-10-であるアルキレンである、請求項1~11のいずれか一項に記載の剤/組成物。
- Wが、
-(CH2)1-5-O-(CH2)2-4-、
-(CH2)1-2-O-(CH2)2-3-O-(CH2)2-3-、
-(CH2)1-5-S-(CH2)2-4-、
-(CH2)1-2-S-(CH2)2-3-S-(CH2)2-3-、
-(CH2)1-5-NH-(CH2)2-4-、または
-(CH2)1-2-NH-(CH2)2-3-NH-(CH2)2-3-であり、
ここで、最左の結合はA部に結合しかつ最右の結合は該ヒドロキシ基の酸素原子に結合するか、あるいは最左の結合は該ヒドロキシ基の酸素原子に結合しかつ最右の結合はA部に結合してもよく、
該Wは―OH、ハロ、C1-3アルキル、およびC1-3アルコキシからなる群から独立して選択される少なくとも1つの置換基で置換されていてもよい、請求項1~10のいずれか一項に記載の剤/組成物。 - Wが、

- 該有効成分の含有量が0.00001~10%(w/v)である、請求項1~20のいずれか一項に記載の剤/組成物。
