i)(4E,8E,12E,16E,20E)-N-{2-[{4-[(2-アミノ-4-{[(3S)-1-ヒドロキシヘキサン-3-イル]アミノ}-6-メチルピリミジン-5-イル)メチル]ベンジル}(メチル)アミノ]エチル}-4,8,12,17,21,25-ヘキサメチルヘキサコサ-4,8,12,16,20,24-ヘキサエンアミド(以下、「化合物A」という)または製薬学的に許容されるその塩;
ii)スクワラン;
iii)アスコルビン酸エステル類(例えば、L-アスコルビン酸ステアリン酸エステル、パルミチン酸アスコルビン酸等)、アスコルビン酸の無機塩(例えば、アスコルビン酸カリウム、アスコルビン酸ナトリウム、アスコルビン酸カルシウム等)、およびアスコルビン酸からなる群から選択される抗酸化剤A;
iv)非還元性の糖類および糖アルコール類(ただしマンニトールは除く)からなる群から選択される賦形剤A;
v)親水性界面活性剤;および
vi)親油性界面活性剤
を含有するエマルションの凍結乾燥製剤が挙げられる。
i)(4E,8E,12E,16E,20E)-N-{2-[{4-[(2-アミノ-4-{[(3S)-1-ヒドロキシヘキサン-3-イル]アミノ}-6-メチルピリミジン-5-イル)メチル]ベンジル}(メチル)アミノ]エチル}-4,8,12,17,21,25-ヘキサメチルヘキサコサ-4,8,12,16,20,24-ヘキサエンアミド(以下、「化合物A」という)または製薬学的に許容されるその塩;
ii)スクワラン;
iii)アスコルビン酸エステル類(例えば、L-アスコルビン酸ステアリン酸エステル、パルミチン酸アスコルビン酸等)、アスコルビン酸の無機塩(例えば、アスコルビン酸カリウム、アスコルビン酸ナトリウム、アスコルビン酸カルシウム等)、およびアスコルビン酸からなる群から選択される抗酸化剤A;
iv)非還元性の糖類および糖アルコール類(ただしマンニトールは除く)からなる群から選択される賦形剤A;
v)ポリオキシエチレンソルビタン脂肪酸エステル類(例えば、ポリソルベート20、ポリソルベート40、ポリソルベート60、ポリソルベート65、ポリソルベート80等);ポリオキシエチレン硬化ヒマシ油類(例えば、ポリオキシエチレン硬化ヒマシ油10、ポリオキシエチレン硬化ヒマシ油20、ポリオキシエチレン硬化ヒマシ油40、ポリオキシエチレン硬化ヒマシ油50、ポリオキシエチレン硬化ヒマシ油60等);ポリオキシエチレンポリオキシプロピレングリコール類(例えば、ポリオキシエチレン(42)ポリオキシプロピレン(67)グリコール、ポリオキシエチレン(54)ポリオキシプロピレン(39)グリコール、ポリオキシエチレン(105)ポリオキシプロピレン(5)グリコール、ポリオキシエチレン(124)ポリオキシプロピレン(39)グリコール、ポリオキシエチレン(160)ポリオキシプロピレン(30)グリコール、ポリオキシエチレン(196)ポリオキシプロピレン(67)グリコール、ポリオキシエチレン(200)ポリオキシプロピレン(70)グリコール等)等の親水性界面活性剤;
vi)ソルビタン脂肪酸エステル類(例えば、ソルビタン脂肪酸エステル、モノラウリン酸ソルビタン、モノパルミチン酸ソルビタン、モノステアリン酸ソルビタン、トリステアリン酸ソルビタン、モノオレイン酸ソルビタン、セスキオレイン酸ソルビタン、トリオレイン酸ソルビタン、中鎖脂肪酸トリグリセリド等);グリセリン脂肪酸エステル類(例えば、グリセリン脂肪酸エステル、モノステアリン酸グリセリン、モノミリスチン酸グリセリン、モノオレイン酸グリセリン、トリイソオクタン酸グリセリン等);ショ糖脂肪酸エステル類(例えば、ショ糖脂肪酸エステル、ショ糖ステアリン酸エステル、ショ糖パルミチン酸エステル等);プロピレングリコール脂肪酸エステル類(例えば、プロピレングリコール脂肪酸エステル、モノステアリン酸プロピレングリコール等)等の親油性界面活性剤;および
iii’)トコフェロール類(例えば、α-トコフェロール、β-トコフェロール、γ-トコフェロール、δ-トコフェロール等)、トコフェロール酢酸エステル、およびブチルヒドロキシアニソールからなる群から選択される抗酸化剤B
を含有するエマルションの凍結乾燥製剤が挙げられる。
当該製剤におけるトコフェロールの含量としては、化合物Aの重量の5倍~250倍であり、好ましくは12.5倍~125倍であり、更に好ましくは20倍~50倍であり、更により好ましくは25倍~50倍である。
当該処方において、更なる抗酸化剤として、チオ硫酸ナトリウムまたはブチルヒドロキシアニソール等を含んでいてもよい。
移動相A:0.1% トリフルオロ酢酸水溶液
移動相B: 0.06% トリフルオロ酢酸を含むアセトニトリル/メタノール混液(8:2)
送液条件:
流速: 毎分0.5 mL
カラム温度: 40℃付近の一定温度
〔実施例1~16、比較例1~3〕
化合物Aを表1~4の組成となるように油性成分(実施例1~9及び15においてはスクワラン、トリオレイン酸ソルビタン及びα-トコフェロール、実施例10~13においてはスクワラン及びトリオレイン酸ソルビタン、実施例14においては、スクワラン、トリオレイン酸ソルビタン、α-トコフェロール及びパルミチン酸アスコルビン酸、実施例16においてはスクワラン、トリオレイン酸ソルビタン、α-トコフェロール及びブチルヒドロキシアニソール、比較例1~3においてはスクワレン、トリオレイン酸ソルビタン及びα-トコフェロール)に溶解させた。表1~4の組成となるように注射用水に水性成分(実施例1~3、14及び16においてはスクロース及びポリソルベート80、実施例4~13においてはスクロース、ポリソルベート80及びアスコルビン酸ナトリウム、実施例15においてはスクロース、ポリソルベート80及びチオ硫酸ナトリウム)を溶解後、上記油性組成物を加えて予備混合を行い、超高圧乳化分散機を用いて乳化分散させた。0.2μmの滅菌フィルターを通してろ過した後、ガラス製バイアルに1 mLずつ充填し、凍結乾燥した。窒素ガスで復圧後、ゴム栓で密封し、凍結乾燥組成物である実施例1~16及び比較例1~3を得た。
凍結乾燥組成物の製造工程において、乳化後で凍結乾燥前のエマルションに含有される油滴粒子の粒度分布を下記の方法により測定した。
注射用水を用いてエマルションを10倍に希釈し、動的光散乱式粒度分布測定装置(Zetasizer Nano ZS)を用いて、散乱強度基準の90%粒子径(D90)を測定した。実施例1~16および比較例1~3のD90(nm)を表5に示す。
調製した凍結乾燥組成物について、開始時及び5℃及び25℃の恒温庫内で6箇月間保存後の粒度分布を下記の方法により測定した。
実施例1~16および比較例1~3で調整した凍結乾燥組成物1バイアルに1 mLの注射用水を加えて復水後、復水後の薬液100μLをマイクロピペットを用いて採取し、900μLの注射用水と混合後、動的光散乱式粒度分布測定装置(Zetasizer Nano ZS)を用いて、散乱強度基準の90%粒子径(D90)を測定した。保存前後の実施例1~16および比較例1~3のD90(nm)を表6に示す。
試験結果より、アスコルビン酸類の抗酸化剤を含む実施例4~14の処方においては、5℃及び25℃の恒温庫内で6箇月間保存後の粒度分布が、保存開始時の値から変化が少なく、粒度分布の安定性が高い処方であると言える。
調製した凍結乾燥組成物について、開始時及び5℃の恒温庫内で6箇月間保存後の不純物量(Uk-1.02の面積百分率値)を下記の方法により測定した。
Phenyl-Hexylカラム(Waters製Xselect CSH Phenyl-Hexyl XP Column、4.6 mm×75 mm、2.5μm、型番:186006134)を用いて、純水、アセトニトリル、メタノール、トリフルオロ酢酸を用いた逆相系高速液体クロマトグラフィー法により、化合物Aとして0.4~2μg相当量を注入し、220 nmの波長で分光学的に検出した。測定条件の詳細を以下に示す。
移動相A:0.1% トリフルオロ酢酸水溶液
移動相B: 0.06% トリフルオロ酢酸を含むアセトニトリル/メタノール混液(8:2)
送液条件:
流速: 毎分0.5 mL
カラム温度: 40℃付近の一定温度
本法で測定したピーク面積及び溶出時間を用い、化合物Aの1.02倍の溶出時間に検出される不純物ピーク(Uk-1.02)の面積百分率値を以下の式により算出した。
Uk-1.02の面積百分率値(%)=Uk-1.02のピーク面積/類縁物質及び化合物Aの総ピーク面積×100
保存前後の実施例1~16および比較例1~3の不純物ピーク(Uk-1.02)の面積百分率値(%)を表7に示す。
試験結果より、アスコルビン酸類の抗酸化剤を含む実施例4~14の処方においては、5℃の恒温庫内で6箇月間保存後の不純物の値(Uk-1.02の面積百分率値)が低く、保存安定性が高い処方であると言える。また、油性成分としてスクワランを含有する実施例1~3の処方と、油性成分としてスクワレンを含有する比較例1~3の処方を比較すると、油性成分としてスクワランを含有する処方の方が不純物の値(Uk-1.02の面積百分率値)が低い事から、スクワランが化合物1の抗酸化安定性に寄与しており、スクワランを含有する処方は保存安定性が高い処方であると言える。
Claims (45)
- i)(4E,8E,12E,16E,20E)-N-{2-[{4-[(2-アミノ-4-{[(3S)-1-ヒドロキシヘキサン-3-イル]アミノ}-6-メチルピリミジン-5-イル)メチル]ベンジル}(メチル)アミノ]エチル}-4,8,12,17,21,25-ヘキサメチルヘキサコサ-4,8,12,16,20,24-ヘキサエンアミド(以下、「化合物A」という)または製薬学的に許容されるその塩;
ii)スクワラン;
iii)アスコルビン酸エステル類(例えば、L-アスコルビン酸ステアリン酸エステル、パルミチン酸アスコルビン酸等)、アスコルビン酸の無機塩(例えば、アスコルビン酸カリウム、アスコルビン酸ナトリウム、アスコルビン酸カルシウム等)、およびアスコルビン酸からなる群から選択される抗酸化剤A;および
iv)非還元性の糖類および糖アルコール類(ただしマンニトールは除く)からなる群から選択される賦形剤A、
を含有するエマルションの凍結乾燥製剤。 - 更に、v)親水性界面活性剤、およびvi)親油性界面活性剤を含む、請求項1に記載の製剤。
- エマルションが、水中油型エマルションである、請求項1または2に記載の製剤。
- 親水性界面活性剤が、ポリオキシエチレンソルビタン脂肪酸エステル類(例えば、ポリソルベート20、ポリソルベート40、ポリソルベート60、ポリソルベート65、ポリソルベート80等);ポリオキシエチレン硬化ヒマシ油類(例えば、ポリオキシエチレン硬化ヒマシ油10、ポリオキシエチレン硬化ヒマシ油20、ポリオキシエチレン硬化ヒマシ油40、ポリオキシエチレン硬化ヒマシ油50、ポリオキシエチレン硬化ヒマシ油60等);またはポリオキシエチレンポリオキシプロピレングリコール類(例えば、ポリオキシエチレン(42)ポリオキシプロピレン(67)グリコール、ポリオキシエチレン(54)ポリオキシプロピレン(39)グリコール、ポリオキシエチレン(105)ポリオキシプロピレン(5)グリコール、ポリオキシエチレン(124)ポリオキシプロピレン(39)グリコール、ポリオキシエチレン(160)ポリオキシプロピレン(30)グリコール、ポリオキシエチレン(196)ポリオキシプロピレン(67)グリコール、ポリオキシエチレン(200)ポリオキシプロピレン(70)グリコール等)である、請求項2または3に記載の製剤。
- 親水性界面活性剤が、ポリソルベート20、ポリソルベート40、ポリソルベート80、ポリオキシエチレン硬化ヒマシ油60、またはポリオキシエチレン(160)ポリオキシプロピレン(30)グリコールである、請求項2または3に記載の製剤。
- 親水性界面活性剤が、ポリソルベート20、ポリソルベート40、またはポリソルベート80である、請求項2または3に記載の製剤。
- 親油性界面活性剤が、ソルビタン脂肪酸エステル類(例えば、ソルビタン脂肪酸エステル、モノラウリン酸ソルビタン、モノパルミチン酸ソルビタン、モノステアリン酸ソルビタン、トリステアリン酸ソルビタン、モノオレイン酸ソルビタン、セスキオレイン酸ソルビタン、トリオレイン酸ソルビタン、中鎖脂肪酸トリグリセリド等);グリセリン脂肪酸エステル類(例えば、グリセリン脂肪酸エステル、モノステアリン酸グリセリン、モノミリスチン酸グリセリン、モノオレイン酸グリセリン、トリイソオクタン酸グリセリン等);ショ糖脂肪酸エステル類(例えば、ショ糖脂肪酸エステル、ショ糖ステアリン酸エステル、ショ糖パルミチン酸エステル等);またはプロピレングリコール脂肪酸エステル類(例えば、プロピレングリコール脂肪酸エステル、モノステアリン酸プロピレングリコール等)である、請求項2~6のいずれかに記載の製剤。
- 親油性界面活性剤が、ソルビタン脂肪酸エステル、モノオレイン酸ソルビタン、セスキオレイン酸ソルビタン、またはトリオレイン酸ソルビタンである、請求項2~6のいずれかに記載の製剤。
- 親油性界面活性剤が、トリオレイン酸ソルビタンである、請求項2~6のいずれかに記載の製剤。
- 更に、トコフェロール類(例えば、α-トコフェロール、β-トコフェロール、γ-トコフェロール、δ-トコフェロール等);トコフェロール酢酸エステル;およびブチルヒドロキシアニソールからなる群から選択される抗酸化剤Bを含む、請求項1~9のいずれかに記載の製剤。
- 抗酸化剤Bが、α-トコフェロール、β-トコフェロール、γ-トコフェロール、およびδ-トコフェロールからなる群から選択されるトコフェロール類である、請求項10に記載の製剤。
- 抗酸化剤Bが、α-トコフェロールである、請求項10に記載の製剤。
- 抗酸化剤Aが、パルミチン酸アスコルビン酸、アスコルビン酸カリウム、アスコルビン酸ナトリウム、またはアスコルビン酸である、請求項1~12のいずれかに記載の製剤。
- 抗酸化剤Aが、アスコルビン酸ナトリウムまたは、アスコルビン酸カリウムである、請求項1~12のいずれかに記載の製剤。
- 賦形剤Aが、非還元性の糖類(例えば、スクロース、またはトレハロース);または糖アルコール類(例えば、ソルビトール、エリスリトール、キシリトール、マルチトール、またはラクチトール)である、請求項1~14のいずれかに記載の製剤。
- 賦形剤Aが、スクロース、トレハロース、ソルビトール、またはキシリトールである、請求項1~14のいずれかに記載の製剤。
- 賦形剤Aが、スクロース、またはトレハロースである、請求項1~14のいずれかに記載の製剤。
- スクワランの配合量が、化合物Aの重量の50倍~500倍である、請求項1~17のいずれかに記載の製剤。
- スクワランの配合量が、化合物Aの重量の100倍~400倍である、請求項1~17のいずれかに記載の製剤。
- スクワランの配合量が、化合物Aの重量の200倍~300倍である、請求項1~17のいずれかに記載の製剤。
- 親水性界面活性剤の配合量が、化合物Aの重量の0.5倍~250倍である、請求項2~20のいずれかに記載の製剤。
- 親水性界面活性剤の配合量が、化合物Aの重量の5倍~100倍である、請求項2~20のいずれかに記載の製剤。
- 親水性界面活性剤の配合量が、化合物Aの重量の10倍~50倍である、請求項2~20のいずれかに記載の製剤。
- 親油性界面活性剤の配合量が、化合物Aの重量の0.5倍~250倍である、請求項2~21のいずれかに記載の製剤。
- 親油性界面活性剤の配合量が、化合物Aの重量の5倍~100倍である、請求項2~22のいずれかに記載の製剤。
- 親油性界面活性剤の配合量が、化合物Aの重量の10倍~50倍である、請求項2~23のいずれかに記載の製剤。
- 抗酸化剤Aの配合量が、アスコルビン酸ナトリウムの重量に換算した場合に、化合物Aの重量の0.5倍~500倍である、請求項1~26のいずれかに記載の製剤。
- 抗酸化剤Aの配合量が、アスコルビン酸ナトリウムの重量に換算した場合に、化合物Aの重量の2.5倍~250倍である、請求項1~26のいずれかに記載の製剤。
- 抗酸化剤Aの配合量が、アスコルビン酸ナトリウムの重量に換算した場合に、化合物Aの重量の5倍~100倍である、請求項1~26のいずれかに記載の製剤。
- 賦形剤Aの配合量が、化合物Aの重量の50倍~1000倍である、請求項1~29のいずれかに記載の製剤。

- 化合物Aの重量が、化合物Aを含まない凍結乾燥物の重量の0.0005倍~0.065倍である、請求項1~35のいずれかに記載の製剤。
- エマルション調製直後の粒子径D90値、および凍結乾燥製剤として25℃で6カ月保存後に復水したエマルションの粒子径D90値が、いずれも1000 nm以下である、請求項1~38のいずれかに記載の製剤。
- 凍結乾燥製剤を5℃で6か月保存した時の、不純物UK-1.02の面積百分率値における増加量が、5.0%以下である、請求項1~39のいずれかに記載の製剤。
- 凍結乾燥製剤を5℃で6か月保存した時の、不純物UK-1.02の面積百分率値における増加量が、1.0%以下である、請求項1~39のいずれかに記載の製剤。
- 請求項1~41のいずれかに記載の製剤を含むワクチンアジュバント。
