Definitions
- the present invention relates to an oral composition containing reduced coenzyme Q10 and suppressing discoloration, and a method for producing the same.
- the present invention also relates to a discoloration suppressing method and a discoloration suppressing agent for a composition containing reduced coenzyme Q10.
- Coenzyme Q10 (hereinafter, also referred to as “CoQ10”) is a benzoquinone derivative widely distributed in nature.
- CoQ10 is an essential substance as a coenzyme when mitochondria of animal cells produce energy, and is also known as a vitamin-like acting substance having an action of improving oxygen utilization efficiency.
- the main pharmacological actions of CoQ10 include an antioxidant action, an energy production promoting action, a cardiopulmonary function enhancing action, an immunostimulating action and an aging preventing action, and they are also used as components of health foods and the like.
- CoQ10 has an oxidized type and a reduced type.
- Oxidized CoQ10 (molecular formula: C 59 H 90 O 4 , molecular weight 863.365) is also referred to as ubiquinone 10.
- reduced CoQ10 (molecular formula: C 59 H 92 O 4 , molecular weight 865.381) is also called as ubiquinol 10 and is a two-electron reduced form of oxidized CoQ10.
- Oxidized CoQ10 is a yellow to orange crystalline powder, whereas reduced CoQ10 is a white to pale yellow crystalline powder.
- CoQ10 is a nutrient that is contained in meat and eggs at about several ⁇ g / g and is also synthesized in vivo. However, the amount of synthesis in vivo decreases with age and tends to be deficient. However, since there are very few natural food materials having a high content of CoQ10, it is difficult to ingest a sufficient amount of CoQ10 to supplement the deficiency with a normal diet alone. Further, it is desirable to supplement CoQ10 from a dietary supplement, a nutrition-enriched food or the like in order to prevent or reduce symptoms such as ischemic heart disease which is common in the elderly.
- Oxidized CoQ10 has been conventionally used for such nutritional supplements and fortified foods, but oxidized CoQ10 has a problem of low absorbability upon ingestion. - the color difference was measured by the following method.
- ⁇ Color difference measurement method> The L value, a value, and b value of the sample were measured using a color difference meter (product name Spectrophotometer SE7700, manufactured by Nippon Denshoku Industries Co., Ltd.). ⁇ Unless otherwise specified, the number of n is 3 per level. The average value of 3 samples was made into L value of each level, a value, and b value. The measurement was performed 5 times for each sample, and the average value was output as a result. At that time, the sample was stirred for each measurement so that the entire sample could be measured. -For the measurement, about 1 g of the sample was weighed from the petri dish and used. At that time, the sample in the petri dish was mixed and homogenized, and then taken out.
- ⁇ E ⁇ (L value 0time levels A - L value of 1week level A) 2 + (a value of 0time levels A - a value of 1week level A) 2 + (b value of 0time levels A - B value of 1 week of level A) 2 ⁇ 1/2
- a smaller ⁇ E value means that discoloration (change in color tone) is suppressed.
- the reduced CoQ10 (level S-1) increased in ⁇ E value over time. When the appearance was visually observed, all levels were white at the start of storage. Level S-1 became dark yellow over time and became orange after 7 days. Theanine, sesamin and dextrin were visually white to pale yellow even after 7 days. Therefore, it was confirmed that the theanine, the sesamin-class compounds and the dextrin have almost no change in color tone due to storage.
- Example 1 The measured raw materials were put in a bag and mixed to prepare three levels of compositions shown in Table 2. Approximately 2 g of each composition was placed in a Petri dish, and allowed to stand at a temperature of approximately 20 ° C. and a humidity of approximately 50% RH for storage. L value and a value of the sample immediately after preparation (at the start of storage: 0 hours of storage), and after 8 hours, 1 day (24 hours), 4 days (96 hours) and 7 days (168 hours) of storage And b value were measured to obtain ⁇ E value. Table 3 shows the time-dependent changes in the ⁇ E value at each level.
- compositions of Levels 1-1 to 1-3 containing reduced CoQ10 increased the ⁇ E value with the passage of time, but when sesamin compounds, or sesamin compounds and theanine were added, the ⁇ E value was suppressed. That is, discoloration (change of color tone) could be suppressed by adding sesamin, or sesamin and theanine to reduced CoQ10.
- Example 2 In an experiment under the same conditions as in Example 1, the mixing ratios of reduced CoQ10 and sesamin were changed as shown in Table 4 and the mixture was placed in a petri dish, and the samples immediately after preparation (0 hours storage) and 7 days (168 hours) storage were tested. The L value, the a value, and the b value were measured to obtain the ⁇ E value. Table 4 shows ⁇ E values after 7 days of storage. Regarding the ⁇ E value after 7 days of storage, the ratio (%) of the ⁇ E value of levels 2-2 to 2-4 to the ⁇ E value of level 2-1 (control) was determined as the change rate with respect to the control. This change rate (%) is also shown in Table 4.
- Levels 2-2 to 2-4 the change in color tone of the composition was suppressed.
- the blending weight ratio of sesamin-class to reduced CoQ10 was 0.01 in Level 2-2, 0.1 in Level 2-3, and 0.5 in Level 2-4.
- level 2-4 corresponds to the composition indicated by N (67,33,0) in the three-phase diagram shown in FIG.
- Example 3 The measured raw materials were put in a bag and mixed to prepare a composition of each level.
- the composition (% by weight) of each level of composition is shown in Table 5. Approximately 2 g of each composition was placed in a petri dish and allowed to stand under the same conditions as in Example 1 and stored. Immediately after preparation (0 hour storage), 1 week (7 days) storage, 2 weeks (14 days) storage, and 4 weeks (28 days) storage, the L value, a value, and b value of the sample were measured, and the ⁇ E value was calculated. I asked. In the evaluations 1 to 5 shown in Table 5, the ⁇ E value was obtained in the same evaluation system with the same control as a reference. Levels AM and OP shown in Table 5 are shown in the three-phase diagram of FIG.
- Table 6 shows the time-dependent changes in the ⁇ E value of each sample.
- the ratio of the ⁇ E value of each level when the ⁇ E value of the control was 100% was determined as the change rate with respect to the control. This change rate is also shown in Table 6.
- the change in color tone was suppressed in the levels A to M and O to P.
- the present invention is useful in fields such as foods and drinks and pharmaceuticals.

ΔE={(ΔL)2+(Δa)2+(Δb)2}1/2
(ΔL=保管前のL値-保管後のL値、Δa=保管前のa値-保管後のa値、Δb=保管前のb値-保管後のb値)

ΔE={(ΔL)2+(Δa)2+(Δb)2}1/2
ΔE={(水準Aの0timeのL値-水準Aの1weekのL値)2+(水準Aの0timeのa値-水準Aの1weekのa値)2+(水準Aの0timeのb値-水準Aの1weekのb値)2}1/2
還元型CoQ10(水準S-1)は、経時的にΔE値が増加した。
