(2)炎症性サイトカインとしてインターロイキン-1(IL-1)、インターロイキン-6(IL-6)、又は腫瘍壊死因子(TNF-α)である上記(1)に記載の炎症性サイトカイン産生抑制剤;
(3)トマト抽出物が、生トマトの搾汁液の分子量分画による分子量1万以上の画分である上記(1)又は(2)に記載の炎症性サイトカイン産生抑制剤;
(4)上記(1)ないし(3)に記載の炎症性サイトカイン産生抑制剤を含有する、医薬品又は飲食品;
(5)経口投与或いは経口的摂取の形態にある上記(4)に記載の医薬品又は飲食品;
である。
本発明が提供する炎症性サイトカインの産生抑制剤は、例えば、インターロイキン-1(IL-1)、インターロイキン-6(IL-6)、並びに腫瘍壊死因子(TNF-α)などの炎症性サイトカインの過剰の産生を抑制することから、これらサイトカインの過剰産生に起因するリウマチ性関節炎、潰瘍性大腸炎、クローン病、更には、2型糖尿病、うつ病、肥満症など様々な疾病に対する有効な治療薬となり得るものであり、また、日常的に経口摂取することにより、日々の健康管理を有効に行える利点を有するものである。
本発明が提供する炎症性サイトカイン産生抑制剤における有効成分であるトマト抽出物は、成熟した生トマトをホモジネートし、固形分を除去したいわゆる搾汁液から得られ抽出物である。
原料トマトの品種は特に限定されるものではなく、一般に市場に流通している生食用、加工用の品種のいずれであっても使用することが可能である。
生トマトの搾汁には糖質が多く含まれていることから、分子量を基準として、1万以上の抽出物とするのがよい。
用いる遠心式限外濾過フィルターとしては、目的とする分子量分画に適応し得る種々のフィルターを挙げることができるが、例えば、Merck Millipore社のアミコンウルトラ(登録商標)シリーズを使用することができる。
乾燥にあたっては、減圧乾燥、凍結乾燥、噴霧乾燥等の通常の乾燥手段を採用することができるが、なかでも凍結乾燥により乾燥物とするのが好ましい。
本発明の炎症性サイトカインの産生抑制剤の投与は、投与目的、疾患の種類、症状によって異なり、特に限定されるものではないが、剤型は、錠剤、カプセル剤、顆粒剤、散剤、粉剤、液剤等にして直接投与したり、食品や飲水に混ぜて投与したりすることができ、経口的に投与することが望ましい。
また、これらの剤型は、従来から知られている通常の方法で製造することができ、本発明の効果を損なわない範囲で、必要に応じてデキストリン、乳糖、コーンスターチ、乳化剤、防腐剤、賦形剤、増量剤、甘味剤、香味剤、着色剤等の添加剤を配合することも可能である。
かかる飲食品には、機能的に特定保健用食品(いわゆる、「トクホ」)、栄養機能食品、及び機能性表示食品等が含まれる。
なお、本発明の生トマト抽出物は、日常的に摂食されている成熟トマトの搾汁から得られるものであり、安全性が高いものであり、本発明において使用される量では毒性は問題にならない。
成熟生トマト(鹿児島県において生産)103.96gを裁断し、乳鉢ですり潰した。そのものを、4,000×g/15分間の遠心分離を行い、液体部分として27.5mLを回収した。
液体部分20mLをAmicon Ultra-15(MWCO:10,000)の遠心式限外濾過フィルターを用いた遠心式限外濾過に供し、4,000×gで遠心した。
遠心後、分子量1万以上の高分子画分および分子量1万以下の低分子画分を得、それぞれ凍結乾燥を行い、生トマト抽出物を得た。
本発明の生トマト抽出物は上記の方法により得られた分子量1万以上の分子量画分であるが、比較のため、得られた分子量1万以下の分子量画分も、以下の試験に供した。
なお、以下の試験では、得られた生トマト抽出物をDMEN培地(ダルベッコ改変イーグル培地+10%ウシ血清+0.1%抗生物質・抗菌薬)で100μg/mLの濃度に調整し、細胞に供した。
以下に、具体的手法を記載する。
(1)マウスマイクログリア細胞株(MG6)を6-well plateに、1×105細胞/mLを撒く。
(2)3日から4日後に、コントロール(DMEN培地、1mL/well)、LPS(5ng/mL)、LPS(5ng)+生トマト高分子抽出物(100μg)[液量は1mL]及び生トマト高分子抽出物(100μg/mL)の4群の溶液を細胞に適用する。
(3)3時間後および6時間後に培地を抜き取り、0.3mLのRLT溶液(RNeasy Mini kit, QIAGEN)(1%β-メルカプトエタノールを含む)をwellに加える。
(4)セルスクレーパーで細胞をこすり取る。
(5)細胞懸濁液に70%エタノールを0.3mL加え、混和する。
(6)スピンカラム(RNeasy Mini kit, QIAGEN)に上記溶液を入れて、8,000×gで1分間遠心する。
(7)スピンカラムに700μLのRW1溶液(RNeasy Mini kit, QIAGEN)を入れて、8,000×gで1分間遠心する。
(8)スピンカラムに500μLのRPE溶液(RNeasy Mini kit, QIAGEN)を入れて、8,000×gで1分間遠心する。このステップを2回繰り返す。
(10)この水溶液の11μLを用いて、Versco cDNA Synthesis Kit(thermoscientific)により、cDNAを作製する。
(11)上記で得られたcDNA溶液をRNase-free水で10倍希釈する。
(12)この希釈したcDNA溶液を用いて、GAPDH mRNAとTNF-α mRNAの発現を、スタンダード法によるリアルタイムRT-PCR法で測定した。
(14)Wellには、水:8.5μL、cDNAサンプル溶液:2.5μL、GAPDH mRNA検出プライマーあるいはTNF-α mRNA検出プライマー(forward+reverse):1.5μL、Sybergreen mater mix:12.5μLを加える。
(15)PCRのサーマルサイクルの条件は、Hold、94℃、10分+3 step PCR(40サイクル;94℃、30分+55℃、30秒+72℃、30秒)+Hold、72℃、1分とした。
(16)各群のサンプルごとに、TNF-α mRNA/GAPDH mRNAの値を求め、コントロールの平均値を100%として、それぞれの値を%に換算した。
(17)統計学的有意差検定は、分散分析後、Turkey-Kramer法を用いた。
図1は3時間後の結果であり、図2は6時間後の結果である。
図1に示した結果からも判明するように、本発明の生トマト抽出物は、3時間後の結果において、TNF-α mRNAの発現を効果的に抑制していることが理解される。
以下に、具体的手法を記載するが、その手法は、上記試験例1に準ずるものである。

(10)この水溶液の11μLを用いて、Versco cDNA Synthesis Kit(thermoscientific)により、cDNAを作製する。
(11)上記で得られたcDNA溶液をRNase-free水で10倍希釈する。
(12)この希釈したcDNA溶液を用いて、GAPDH mRNAとIL-1β mRNAの発現を、スタンダード法によるリアルタイムRT-PCR法で測定した。

図3は3時間後の結果であり、図4は6時間後の結果である。
図に示した結果からも判明するように、本発明の生トマト抽出物は、3時間後の結果において、IL-1β mRNAの発現を効果的に抑制していることが理解される。

図5は3時間後の結果であり、図6は6時間後の結果である。
図に示した結果からも判明するように、本発明の生トマト抽出物は、3時間後及び6時間後の結果において、IL-6 mRNAの発現を効果的に抑制していることが理解される。
比較のため、生トマト抽出物として低分子量(分子量1万以下)画分の炎症性サイトカインの産生抑制作用を検討した。
低分子量の生トマト抽出物として100μg/mL用いて試験例1と同様の手法より検討した。
その結果を図7に示した。
図7に示した結果からも判明するように、低分子の生トマト抽出物には炎症性サイトカインの産生抑制効果は認められず、分子量として1万以上の高分子量画分の生トマト抽出物に炎症性サイトカインの産生抑制効果が認められることが判明した。
試験例1におけるマウスマイクログリア細胞株(MG6)に代えて、マウスマクロファージ細胞株(Raw264)を用いて、同様の手法により検討した。
すなわち、以下の具体的手法により検討した。
(1)マウスマクロファージ細胞株(Raw264)を6-well plateに、1×105細胞/mLを撒く。
(2)3日から4日後に、コントロール(DMEN培地、1mL/well)、LPS(5ng/mL)、LPS(5ng)+生トマト高分子抽出物(100μg)[液量は1mL]及び生トマト高分子抽出物(100μg/mL)の4群の溶液を細胞に適用する。
(3)3時間後および6時間後に培地を抜き取り、0.3mLのRLT溶液(RNeasy Mini kit, QIAGEN)(1%β-メルカプトエタノールを含む)をwellに加える。
(4)セルスクレーパーで細胞をこすり取る。
(5)細胞懸濁液に70%エタノールを0.3mL加え、混和する。
(6)スピンカラム(RNeasy Mini kit, QIAGEN)に上記溶液を入れて、8,000×gで1分間遠心する。

図9に示した結果からも判明するように、本発明の生トマト抽出物は、6時間後の結果において、TNF-α mRNAの発現を効果的に抑制していることが理解される。
試験例5の手法に順じ、検討した。
すなわち、
(1)マウスマクロファージ細胞株(Raw264)を6-well plateに、1×105細胞/mLを撒く。
(2)3日から4日後に、コントロール(DMEN培地、1mL/well)、LPS(5ng/mL)、LPS(5ng)+生トマト高分子抽出物(100μg)[液量は1mL]及び生トマト高分子抽出物(100μg/mL)の4群の溶液を細胞に適用する。
(3)3時間後および6時間後に培地を抜き取り、0.3mLのRLT溶液(RNeasy Mini kit, QIAGEN)(1%β-メルカプトエタノールを含む)をwellに加える。

本発明が提供する炎症性サイトカインの産生抑制剤は、例えば、インターロイキン-1(IL-1)、インターロイキン-6(IL-6)、並びに腫瘍壊死因子(TNF-α)などの炎症性サイトカインの過剰の産生を抑制することから、これらサイトカインの過剰産生に起因するリウマチ性関節炎、潰瘍性大腸炎、クローン病、2型糖尿病、肥満症(特に、インスリン抵抗性)、更にはうつ病など様々な疾病に対する有効な治療薬となり得るものであり、また、日常的に経口摂取することにより、日々の健康管理を有効に行える利点を有するものであり、その産業上の利用性は多大なものである。
Claims (5)
- トマト抽出物を有効成分とする炎症性サイトカイン産生抑制剤。
- 炎症性サイトカインとしてインターロイキン-1(IL-1)、インターロイキン-6(IL-6)又は腫瘍壊死因子(TNF-α)である請求項1に記載の炎症性サイトカイン産生抑制剤。
- トマト抽出物が、生トマトの搾汁液の分子量分画による分子量1万以上の画分である請求項1又は2に記載の炎症性サイトカイン産生抑制剤。
- 請求項1ないし3に記載の炎症性サイトカイン産生抑制剤を含有することを特徴とする医薬品又は飲食品。
- 経口投与或いは経口的摂取の形態にある請求項4に記載の医薬品又は飲食品。
