Abstract
中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含むことを特徴とする組成物。
Description
本発明は、消化管症状を抑制するための組成物及び方法に関する。より具体的には、本発明は、中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物と方法に関する。
ケトン体は、アセト酢酸、β-ヒドロキシ酪酸、アセトンの総称であり、生体内では肝臓において脂肪酸のβ酸化によって合成される。生成されたケトン体は、肝臓以外の、脳や心臓、骨格筋といった多くの組織で、ブドウ糖等と同様にエネルギーとして利用される。特に脳ではブドウ糖とケトン体しか利用されず、ケトン体は重要なエネルギー源となる(非特許文献1)。
体内でのケトン体生成の主たる原料となる脂肪酸は、炭素鎖の長さによって、長鎖脂肪酸、中鎖脂肪酸、短鎖脂肪酸に分類される。中鎖脂肪酸は、吸収・代謝経路の違いにより、長鎖脂肪酸に比べて効率的にケトン体を生成できると考えられている。また、ヒトや動物において、長鎖脂肪酸を含む油脂よりも中鎖脂肪酸を含む油脂を摂取する方が、血中のケトン体濃度が上昇することが知られている(非特許文献2)。
しかしながら、中鎖脂肪酸を含む油脂を摂取する場合、胃をはじめとする消化管において不快な症状を体感し得ることが知られている。

本発明の目的は、中鎖脂肪酸を含む油脂を摂取する場合に、胃をはじめとする消化管における不快な症状を抑制するための組成物及び方法を提供することにある。
本発明者らは、上記課題に鑑み、鋭意研究したところ、中鎖脂肪酸を含む油脂に長鎖脂肪酸を含む油脂を配合させた組成物をヒトが摂取した場合に、中鎖脂肪酸を含む油脂を摂取したときの消化管症状を抑制できることを見出した。
したがって、本発明によれば、以下の消化管症状を抑制するための組成物等を提供できる。
1.中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含むことを特徴とする組成物。
2.前記消化管症状が上部消化管症状である、1に記載の組成物。
3.前記消化管症状が、食道、胃、及び十二指腸のうち少なくとも一つにおける症状である、1又は2に記載の組成物。
4.前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が6~12であり、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が13~30である、1~3のいずれかに記載の組成物。
5.前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてオクタン酸及びデカン酸を含み、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてパルミチン酸、オレイン酸、及びリノール酸を含む、1~4のいずれかに記載の組成物。
6.前記中鎖脂肪酸を含む油脂の配合量が、組成物100gあたり3g以上15g以下であり、前記長鎖脂肪酸を含む油脂の配合割合が、前記中鎖脂肪酸を含む油脂の配合量に対して、30質量%以上200質量%以下である、1~5のいずれかに記載の組成物。

したがって、本発明によれば、以下の消化管症状を抑制するための組成物等を提供できる。
1.中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含むことを特徴とする組成物。
2.前記消化管症状が上部消化管症状である、1に記載の組成物。
3.前記消化管症状が、食道、胃、及び十二指腸のうち少なくとも一つにおける症状である、1又は2に記載の組成物。
4.前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が6~12であり、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が13~30である、1~3のいずれかに記載の組成物。
5.前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてオクタン酸及びデカン酸を含み、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてパルミチン酸、オレイン酸、及びリノール酸を含む、1~4のいずれかに記載の組成物。

本発明によれば、中鎖脂肪酸を含む油脂を摂取する場合に、胃をはじめとする消化管における不快な症状を抑制するための組成物及び方法を提供することができる。
[消化管症状を抑制するための組成物]
本発明の組成物は、中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含むことを特徴とするものである。

本発明の組成物によれば、中鎖脂肪酸を含む油脂を摂取することに起因する消化管(食道・胃・腸)の各種症状を抑制できる。消化管症状とは、これらに限定されないが、例えば、刺激、痛み、熱感、むかつき、膨満感、げっぷ、吐き気、違和感、不快感等が挙げられる。本発明の組成物によれば、消化管におけるこれらの症状を抑制できる。
本発明の一態様において、消化管とは、特に上部消化管(食道・胃・十二指腸)である。したがって、本発明の一態様によれば、中鎖脂肪酸を含む油脂を摂取することに起因する、食道、胃、及び十二指腸のうち、少なくとも一つにおける各種症状を抑制できる。
体内でケトン体が多く生成されるように考案された食事として、古くから高脂質低炭水化物食(ケトン食)が知られている。ケトン食の摂取により、血中のケトン体濃度は上昇する。ケトン食はGLUT1欠損症の治療や難治性てんかんにおける発作抑制に有用であることが知られている(藤井達哉、「ケトン食の基礎から実践まで」、診断と治療社、2011年3月)。さらに最近では、アルツハイマー病等の神経変性疾患の予防や治療にも有用であることが報告されている(Maciej Gasior、外2名、「Neuroprotective and disease-modifying effects of the ketogenic diet」、Behav Pharmacol.、2006年9月、17(5-6)、p.431-439)。したがって、上記疾患の予防や症状の軽減のために中鎖脂肪酸を含む油脂を多量に摂取する際、本発明の組成物、及び後述する本発明の方法により消化管症状を抑制でき、QOLを向上することができる。
本発明において、中鎖脂肪酸を含む油脂とは、油脂中の構成脂肪酸の長さが中鎖であるものをいい、MCT(Medium Chain Triglyceride)とも呼ぶ。MCTは、代表的には炭素数が6~12、好ましくは炭素数が8~12、より好ましくは炭素数が8~10の脂肪酸で構成されるものを意味する。中鎖脂肪酸としては、例えば、ヘキサン酸、ヘプタン酸、オクタン酸、ノナン酸、デカン酸、ドデカン酸等が挙げられる。油脂を構成する1つのグリセロールに結合する3つの中鎖脂肪酸は、同じでも、異なっていてもよい。複数の中鎖脂肪酸が結合する場合、これらの割合は特に限定されない。
本発明の一態様において、中鎖脂肪酸を含む油脂は、例えば、オクタン酸及びデカン酸の少なくとも一方を含む油脂であり、好ましくは、オクタン酸とデカン酸を含む油脂である。
また、本発明の一態様において、中鎖脂肪酸を含む油脂は、好ましくは、オクタン酸とデカン酸を50~100:50~0の範囲の質量比で含む油脂であり、より好ましくは、オクタン酸とデカン酸を70~90:30~10の範囲の質量比で含む油脂であり、特に好ましくは、オクタン酸とデカン酸を75:25の質量比で含む油脂である。
また、本発明の一態様において、中鎖脂肪酸を含む油脂は、好ましくは、オクタン酸とデカン酸を50~100:50~0の範囲の質量比で含む油脂であり、より好ましくは、オクタン酸とデカン酸を70~90:30~10の範囲の質量比で含む油脂であり、特に好ましくは、オクタン酸とデカン酸を75:25の質量比で含む油脂である。
本発明において、長鎖脂肪酸を含む油脂とは、油脂中の構成脂肪酸の長さが長鎖であるものをいい、LCT(Long Chain Triglyceride)とも呼ぶ。LCTは、代表的には炭素数が13~30、好ましくは炭素数が13~24、より好ましくは炭素数が13~18の脂肪酸で構成されるものを意味する。長鎖脂肪酸としては、例えば、テトラデカン酸、ペンタデカン酸、ヘキサデカン酸、9-ヘキサデセン酸、オクタデカン酸、cis-9-オクタデセン酸、11-オクタデセン酸、テトラコサン酸、cis-15-テトラコサン酸、トリアコンタン酸等が挙げられ、不飽和脂肪酸であってもよい。油脂を構成する1つのグリセロールに結合する3つの長鎖脂肪酸は、同じでも、異なっていてもよい。複数の長鎖脂肪酸が結合する場合、これらの割合は特に限定されない。
本発明の一態様において、長鎖脂肪酸を含む油脂は、好ましくは、パルミチン酸、オレイン酸、リノール酸を含む油脂であり、より好ましくは、パルミチン酸、オレイン酸、リノール酸を20~30:50~60:10~20の範囲の質量比で含む油脂である。
中鎖脂肪酸を含む油脂は、ココナッツ、パームフルーツ等の植物体の種子や、牛乳、乳製品等に存在するため、これらから抽出(粗抽出を含む)あるいは精製(粗精製を含む)したものを本発明に使用することができる。長鎖脂肪酸を含む油脂は、大豆、菜種、オリーブ等の植物体の種子や、牛脂やラード中に存在するため、これらから抽出(粗抽出を含む)あるいは精製(粗精製を含む)したものを本発明に使用することができる。
本発明において、組成物とは、経口又は経管(胃瘻、腸瘻、経鼻経管)摂取するものである。組成物の形態は特に限定されず、例えば、固体、半固体物、液体、液状物、ペースト等であることができる。したがって、本発明の組成物は、例えば、食事、飲料、菓子、栄養剤、サプリメント、医薬品等として、経口又は経管(胃瘻、腸瘻、経鼻経管)摂取できる。具体的には、本発明の組成物は、流動食、半流動食、ゼリー、ゲル、粉末、調製粉乳、発酵乳、バー、ムース、チョコレート、ビスケット、アイスクリーム等の形態にすることができる。
本発明において、組成物中における中鎖脂肪酸を含む油脂の配合割合は、特に限定されず、組成物の用途や機能に依存して適宜決定することができるが、例えば、組成物100gあたりに油脂が3g以上15g以下で含まれることが好ましく、5g以上12g以下で含まれることがより好ましく、7g以上10g以下で含まれることがさらに好ましい。
本発明において、組成物中における長鎖脂肪酸を含む油脂の配合割合は、MCTの配合量に対して、30質量%以上200質量%以下で配合されることが好ましく、50質量%以上120質量%以下で配合されることがより好ましく、70質量%以上100質量%以下で配合されることがさらに好ましい。
本発明において、組成物は、中鎖脂肪酸を含む油脂及び長鎖脂肪酸を含む油脂のほか、任意の成分を含むことができる。これら任意の成分としては、例えば、水、タンパク質、脂質、糖質、食物繊維、ビタミン類、ミネラル類、アミノ酸、甘味料、フレーバー類、酸化防止剤、乳化剤、増粘剤、安定化剤、ゲル化剤、pH調整剤等が挙げられる。
本発明の組成物を摂取した場合、中鎖脂肪酸のみを含む油脂を摂取した場合と比較して、血中ケトン体濃度(特に、β-ヒドロキシ酪酸濃度)に有意差はみられず、同様に、血中ケトン体濃度(特に、β-ヒドロキシ酪酸濃度)を上昇させることができる。
[消化管症状を抑制するための方法]
本発明の方法は、中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための方法であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含む組成物を摂取することを特徴とするものである。
本発明の方法は、中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための方法であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含む組成物を摂取することを特徴とするものである。
本発明の方法によれば、中鎖脂肪酸を含む油脂を摂取することに起因する消化管(食道・胃・腸)の各種症状(例えば、刺激、痛み、熱感、むかつき、膨満感、げっぷ、吐き気、違和感、不快感等)を抑制できる。
本発明の一態様において、消化管とは、特に上部消化管(食道・胃・十二指腸)である。したがって、本発明の一態様によれば、中鎖脂肪酸を含む油脂を摂取することに起因する、食道、胃、及び十二指腸のうち、少なくとも一つにおける各種症状を抑制できる。

本発明の方法において、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含む組成物とは、既に説明した本発明の組成物をいう。本発明の組成物に関する具体的な特徴は、既に説明したとおりである。
以下、実施例を示して本発明をさらに具体的に説明するが、本発明の範囲はこれら実施例の記載に何ら限定されるものではない。
試験例1 上部消化管症状の評価
中鎖脂肪酸及び長鎖脂肪酸を配合した試験食品2品と、中鎖脂肪酸を配合した対照となる試験食品2品を調製し、被験者15人(男性13名、女性2名、年齢26~57歳)にクロスオーバー試験法により摂取させ、摂取後の上部消化管症状の体感の発現を比較した。
クロスオーバー試験法において、各被験者は、試験食品4品すべてを摂取しているが、試験食品4品を摂取する順番はランダム化されており、被験者によって異なる。
調製した試験食品4品の成分組成を表1に示す。
中鎖脂肪酸及び長鎖脂肪酸を配合した試験食品2品と、中鎖脂肪酸を配合した対照となる試験食品2品を調製し、被験者15人(男性13名、女性2名、年齢26~57歳)にクロスオーバー試験法により摂取させ、摂取後の上部消化管症状の体感の発現を比較した。
クロスオーバー試験法において、各被験者は、試験食品4品すべてを摂取しているが、試験食品4品を摂取する順番はランダム化されており、被験者によって異なる。
調製した試験食品4品の成分組成を表1に示す。
脂質のうち、中鎖脂肪酸を含む油脂(MCT)は、オクタン酸(炭素数8)とデカン酸(炭素数10)を75:25の質量比で含む油脂であり、長鎖脂肪酸を含む油脂(LCT)は、パルミチン酸(炭素数16)、オレイン酸(炭素数18)、リノール酸(炭素数18)、その他の脂肪酸を21:52:16:11の質量比で含む油脂である。
各試験日において、被験者は、試験開始12時間前から絶食とした。被験者は、試験食品の摂取前と、摂取してから3時間後まで15分おきに、合計13ポイント(摂取前、摂取15分後、30分後、45分後、60分後、75分後、90分後、105分後、120分後、135分後、150分後、165分後、180分後)において、上部消化管症状の体感を評価した。
上部消化管症状に関する評価項目は、「上腹部への刺激がある(胃の痛み)」、「上腹部に熱感がある」、「胃がムカムカする(胃のむかつき)」、「腹部の膨満を感じる」、「げっぷが出る」、「吐き気がする」の6つで、評価項目それぞれについて3段階評価(「あてはまらない」:0点、「わずかにあてはまる」:1点、「あてはまる」:2点)で体感スコアを付けた。
体感スコアの結果を表2に示す。表2において、各評価項目の数値は、被験者ごとに13ポイントの体感スコアの合計点を算出し、全被験者間で「平均値±標準偏差」を算出したものである。したがって、各評価項目の体感スコアの上限値は26点(2点×13ポイント)、上部消化管症状に関する評価項目6つの合計スコアの上限値は156点(26点×6項目)となる。
流動食形状の試験食品を摂取した場合の上部消化管症状の合計スコアは、長鎖脂肪酸を配合していない対照である試験食品1と比較して、長鎖脂肪酸を配合した試験食品2では統計的有意差があり、上部消化管症状が有意に抑制された。
また、ゼリー形状の試験食品を摂取した場合の上部消化管症状の合計スコアは、長鎖脂肪酸を配合していない対照である試験食品3と比較して、長鎖脂肪酸を配合した試験食品4では差があり、上部消化管症状が抑制された。
また、ゼリー形状の試験食品を摂取した場合の上部消化管症状の合計スコアは、長鎖脂肪酸を配合していない対照である試験食品3と比較して、長鎖脂肪酸を配合した試験食品4では差があり、上部消化管症状が抑制された。
試験例2 血中ケトン体濃度の評価
中鎖脂肪酸及び長鎖脂肪酸を配合した試験食品2品と、中鎖脂肪酸を配合した対照となる試験食品2品を調製し、被験者2人(男性2名、年齢37歳、48歳)にこれらを摂取させ、摂取後の血中β-ヒドロキシ酪酸濃度の推移を観察した。血中β-ヒドロキシ酪酸濃度は、簡易測定器(プレシジョン、アボット社)にて測定した。
調製した試験食品4品の成分組成は表1と同じである。
中鎖脂肪酸及び長鎖脂肪酸を配合した試験食品2品と、中鎖脂肪酸を配合した対照となる試験食品2品を調製し、被験者2人(男性2名、年齢37歳、48歳)にこれらを摂取させ、摂取後の血中β-ヒドロキシ酪酸濃度の推移を観察した。血中β-ヒドロキシ酪酸濃度は、簡易測定器(プレシジョン、アボット社)にて測定した。
調製した試験食品4品の成分組成は表1と同じである。
被験者は、試験開始12時間前から絶食とした。被験者は、試験食品の摂取前と、摂取してから3時間後まで30分おきに、合計7ポイント(摂取前、摂取30分後、60分後、90分後、120分後、150分後、180分後)において、血中β-ヒドロキシ酪酸濃度を測定した。
β-ヒドロキシ酪酸の最高血中濃度(Cmax:単位「mmol/L」)と、血中濃度-時間曲線下面積(AUC:単位「mol・hour/0.5・L」)の結果をそれぞれ図1、図2に示す。図1、図2ともに、2人の被験者における測定値の平均値を示す。いずれの試験食品を摂取した場合でもCmax、AUCに有意差はみられなかった。
このことから、本発明の組成物を摂取した場合、消化管症状を抑制することができるとともに、MCTのみを摂取した場合と比較して血中ケトン体であるβ-ヒドロキシ酪酸濃度に有意差はみられず、血中ケトン体であるβ-ヒドロキシ酪酸濃度を上昇させることが可能であることが分かった。
このことから、本発明の組成物を摂取した場合、消化管症状を抑制することができるとともに、MCTのみを摂取した場合と比較して血中ケトン体であるβ-ヒドロキシ酪酸濃度に有意差はみられず、血中ケトン体であるβ-ヒドロキシ酪酸濃度を上昇させることが可能であることが分かった。
本発明によれば、中鎖脂肪酸を含む油脂を摂取することに起因する消化管(食道・胃・腸)の各種症状を抑制できる。また、本発明によれば、特定の疾患の予防や症状の軽減のために中鎖脂肪酸を含む油脂を多量に摂取する際、消化管症状を抑制でき、QOLを向上することができる。
上記に本発明の実施形態及び/又は実施例を幾つか詳細に説明したが、当業者は、本発明の新規な教示及び効果から実質的に離れることなく、これら例示である実施形態及び/又は実施例に多くの変更を加えることが容易である。従って、これらの多くの変更は本発明の範囲に含まれる。

Claims (9)
- 中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制するための組成物であって、中鎖脂肪酸を含む油脂と、長鎖脂肪酸を含む油脂とを含むことを特徴とする組成物。
- 前記消化管症状が上部消化管症状である、請求項1に記載の組成物。
- 前記消化管症状が、食道、胃、及び十二指腸のうち少なくとも一つにおける症状である、請求項1又は2に記載の組成物。
- 前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が6~12であり、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸の炭素数が13~30である、請求項1~3のいずれかに記載の組成物。
- 前記中鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてオクタン酸及びデカン酸を含み、前記長鎖脂肪酸を含む油脂が、油脂中の構成脂肪酸としてパルミチン酸、オレイン酸、及びリノール酸を含む、請求項1~4のいずれかに記載の組成物。
- 前記中鎖脂肪酸を含む油脂の配合量が、組成物100gあたり3g以上15g以下であり、前記長鎖脂肪酸を含む油脂の配合割合が、前記中鎖脂肪酸を含む油脂の配合量に対して、30質量%以上200質量%以下である、請求項1~5のいずれかに記載の組成物。
- 流動食、半流動食、ゼリー、ゲル、粉末、調製粉乳、発酵乳、バー、ムース、チョコレート、ビスケット、又はアイスクリームの形態である、請求項1~6のいずれかに記載の組成物。
- 中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制する方法であって、前記中鎖脂肪酸を含む油脂に長鎖脂肪酸を含む油脂を配合させた組成物を摂取することを特徴とする方法。
- 中鎖脂肪酸を含む油脂を摂取することに起因する消化管症状を抑制する方法であって、請求項1~7のいずれかに記載の組成物を摂取することを特徴とする方法。
