【배경기술】
코로나바이러스는 외피가 있는 단일가닥의 양성 RNA 바이러스이 며 게놈 크기는 25-32kb로 지금까지 알려진 RNA바이러스 중에서는 .비 교적 큰 바이러스에 속한다. 외피에 곤봉모양의 돌기인 스파이크 (spike) 단백질이 박혀 있어 화염 또는 왕관 모양의 특이적 구조를 가 지고 있고, 라틴어 Corona로부터 바이러스 이름이 유래되었다고 한다.
1937년 닭에서 처음으로 발견된 후 박쥐, 새, 고양이, 개, 소 ᅳ 돼지 , 쥐 등 다양한 조류와 동물에서 발견된 코로나바이러스는 4개의 그룹 (Alphaᅳ, Beta-, Gamma-, De 1 tacoronavi rus )으로 나누어진다. 알 파와 베타코로나바이러스 그룹은 포유류에 주로 감염되고 감마와 델타 코로나바이러스 그룹은 조류에서 찾을 수 있다. 코로나바이러스는 동 물들에서 위장병 및 호흡기질환과 같은 다양한 질환을 일으킬 수 있다 고 알려져 있다. 사람에게 감염되는 사람 코로나바이러스는 1960대 발견된 HCoV一 229E와 HCoV-0C43, 사스 대유행 이후 발견된 HCoV-NL63(2004년)과 HCoV-MUl(2005년)로 이들은 일반적으로 상기도감염증과 관계가 있다 고 알려져 있으나 면역결핍환자들에게는 심각한 폐질환을 유도하기도 한다. 주로 겨울이나 이른 봄에 코로나바이러스에 대한 감염률이 증가 된다고 보고되고 있고, 성인 감기환자 중 코로나바이러스가 원인 병원 체인 비율이 상당히 높다고 알려져있다 . 2003년 중증급성호흡기증후군 (Severe Acute Respiratory Syndrome, SARS)을 일으키는 사스 코로나 바이러스 (SARS-CoV)가 처음 발견되었으며 세계보건기구 (WHO)보고에 따 르면 2002년에서 2003년 동안 전 세계적으로 8, 273명의 환자와 775명 의 사망자 (치사율 약 10%)가 발생하였고, 2004년까지 추가적인 환자 발생과 사망자가 보고되었다.
2012년 9월 사스와 유사한 고열, 기침, 호흡곤란 등의 호흡기 증 상을 나타내는 중중 호흡기질환 환자가 발생하였고, 이 원인 병원체는 기존의 알려진 바이러스와는 다른 신종 코로나바이러스 (HCoV-EMC)로 밝혀졌다. 신종 코로나바이러스는 2013년 5월 국제바이러스 분류위원회
(International Committee on Taxonomy of Viruses)의 코로나바이러스 스타디그룹에서 Middle East respiratory syndr ome-coronavi rus (MERS-CoV)이라는 이름으로 분류하였다. 이 바이러스는 유전자 염기서 열 (genetic sequencing)이 박쥐에서 발견되는 Pi i strel lus Bat CoV一 HKU5, HKU4와 유사하다 하여 박쥐가 가장 유력한 감염원일 것으로 추 정하였다. 그러나 최근 란셋전염병 (The Lancet Infectious Diseases) 학술지를 통해 발표된 논문에서는, 오만의 단봉 낙타 50마리의 혈액샘 플을 분석한 결과 모두 MERS-CoV 항체 흔적이 발견됐다고 밝혔다. 그 러나 카나리아제도에서는 105마리 가운데 15마리에서 항체 양성 반웅 을 보였다. 바이러스 자체가 발견되지는 못했지만 연구 결과는 이 낙타들이 어느 시점에 메르스 또는 유사 바이러스에 감염되었음을 의미하며, 바 이러스 숙주가 낙타일 가능성이 매우 높음을 보여주었다. 증동호흡기증후군 코로나바이러스 (MERS-CoV) 감염은 2012년 9월 사우디아라비아에서 첫 감염사례가 확인된 후 2014년 6월까지 WHO에 공식적으로 보고된 환자는 전 세계적으로 808명이며 , 313명이 사망하 였다 (34.5%). 중동호흡기증후군 코로나바이러스 (MERS-CoV) 초기 확진환자 발생 당시 환자의 임상증세가 사스와 상당히 유사했기에 사스 코로나바이러 스와 상당한 연관성을 가지고 있지 않을까 하는 우려가 있었으나, 전 장 유전자가 분석된 이후 분자 유전학적으로 사스 바이러스와는 상당 한 차이를 보이는 새로운 바이러스라는 것이 밝혀졌다. 우선 중동호흡기증후군 코로나바이러스 (MERS-CoV)는 사스 코로나 바이러스와는 55% 정도의 낮은 뉴클레오타이드상동성올 보였으며 , 계 통학적 분석에 있어서도 두 바이러스는 서로 다른 진화적 차이를 보였 는데, 사스 코로나바이러스는 베타코로나바이러스의 B 그룹에 중동호 흡기증후군 코로나바이러스 (MERS-CoV)는 베타코로나바이러스의 C 그룹 에 속해 있었다. 베타코로나바이러스 그룹에 속하는 것으로는 사스 코 로나바이러스 그리고 사람 코로나바이러스인 HCoV— HKU1과 HCoV-0C43이 알려져 있다. 또한 사스와 증동호흡기증후군 코로나바이러스 (MERS-CoV)는 세포 내로 들어가는 경로도 차이를 보이는데 사스 바이러스는 사람 기도의 섬모세포에 분포되어 있는 ACE2 수용체를, 중동호흡기증후군 코로나바 이러스 (MERS-CoV)는 비섬모세포내 DDP4(CD26이라고도 불림 )를 이용한 다. 두 수용체 모두 사람의 상기도 보다는 하기도에 많이 분포하고 있 고, 증동호흡기증후군 코로나바이러스 (MERS-CoV)와 사스 바이러스가 높은 치사율을 나타내기는 하나 폐렴으로 진행된 후 큰 비말을 통해서
전파가 이루어져야 하기에 인플루엔자나 흥역 경우처럼 에어로졸에 전 파되는 질환에 비교하여 전염성은 그만큼 떨어질 수 있다는 것을 제시 한다 . 그러나 사스환자 발생의 경우를 보면 사스환례에 밀접하게 노출 되는 경우나 호흡기비말의 에어로졸화를 유발하는 환경에서는 그 전파 력은 상당히 높아질 수 있기에 감염성 위험은 여전히 높다고 할 수 있 다. 중동호흡기증후군 코로나바이러스 (MERS-CoV)의 최초 감염원이 무— 엇인지 어떤 방법으로 사람에게 전파되어 병을 일으키게 되었는지에 대해서는 아직까지 밝혀져 있지 않다 . 다만 발생한 신종 코로나바이러 스 확진 .환자 중 2명의 사례는 농장방문과 동물접촉 사실이 밝혀졌다. 이것은 신종 코로나바이러스의 인수공통감염 가능성을 보여주고 있다. 주요 임상증상은 발열 (8.7%), 기침 (89%), 호흡곤란 등으로 폐렴 증상을 보이며, 일부 환자는 구토와 설사 (35%)도 나타난다. 면역기능 이 저하된 환자에서 신부전 (renal fai lure)도 일으키는데 사망를은 34.5%로 매우 높다. 중동지역 (사우디아라비아, 카타르 등)에서 많이 발생하여 이들 지역을 감염지역으로 추정되고 있으나 정확한 감염경로 는 정확하게 밝히지 못하고 있다. 그러나 그동안 사람끼리 광범위하게 감염이 나타난다는 증거는 없었으나 가족이나 의료진과 밀접하게 접촉 하는 경우 (close contact) 전파된다는 사실이 확인되었다. 잠복기는 9 일〜 12일로 추정하는데 사람마다 잠복기의 차이가 많다. 아직까지 중동호흡기증후군 코로나바이러스 (MERS-CoV)의 치료 효 과가 확인된 약물 및 예방 백신이 개발되어 있지 않은 상황이다. 따라 서 현재까지는 바이러스 감염이 의심되는 환자와의 접촉을 피하고 , 개 인위생을 철저히 하는 것 이외에는 특별한 치료 및 예방방법이 없는 실정이다. 현재, 중동호흡기증후군 코로나바이러스 (MERSCoV)의 감염 치료를 위하여 항생제, 리바비린, 인터페론 그리고 환자의 혈청 등이 사용되기도 하였으나 이들 치료제는 특이적인 효과를 발휘하지 못하고 있다 . 다만, 최근에는 라도티닙이 호흡기 바이러스성 질환의 치료에 가능성이 있음이 공지된 바 있다 (대한민국 공개특허 10-2017—0009276), 따라서 중동호흡기증후군 코로나바이러스 (MERS-CoV)에 대한 질병 치료 및 예방을 위한 대체 약물의 개발이 매우 시급한 실정이다.

본 발명의 일 목적은 항바이러스용 약학적 조성물을 제공하는 것 이다. 본 발명의 다른 목적은 바이러스 감염으로 인한 질환의 예방 또

<실시예 1> 2-((4-페녹시페녹시 )메틸)벤젠아민
하기 반웅식에 따라 실시예 1 화합물을 제조하였으며 구체적
단계 1: 1-니트로 2-((4-페녹시페녹시 )메틸)벤젠의 합성 2-니트로벤질 브로마이드 (2.5g, 11.57mmol), 4—페녹시페놀 ᅵ (2.17g, 11.68瞧01), 포타슘 카보네이트 (3.19g, 23.08瞧 ol)를 디메틸 포름아미드 (30 niL)에 흔합하고 실온에서 14시간 교반하였다. 반웅물 에 얼음물을 가하고 에틸 아세테이트 (60 mL)로 추출 후 증류수 (2 X 30 niL)로 세척하고 , 수분을 제거 후 (Na2S04) 감압농축하였다. 잔류물 을 플래시 컬럼 크로마토그래피 (에틸아세테이트: 핵산 = 1:10)로 분리 하여 1-니트로 -2-((4-페녹시페녹시 )메틸)벤젠 (3.05g, 수율 83%)을 제 조하였다.

Vero (CCL-81)세포는 ATCC에서 구입하였고, Huh7 (human hepatocellular carcinoma)는 Japanese Collection of Research Biosources Cell Bank (JCRB 세포주은행 )에서 구입하여 10% 우태아혈 청이 첨가된 Dulbecco's modified Eagle's medium (DMEM) 배지에서 37 °C의 5% C02 배양기를 이용하여 배양하였다.

plaque assay를 이용하여 바이러스 역가를 측정하였다. <1-3> 세포기반 항코로나바이러스 활성 평가
항바이러스 활성 시험을 위해서 Huh-7 세포를 96— we 11 p 1 at e에 배양하고 다음날 준비된 세포에 메르스 코로나바이러스 (M0I 으 1)로 1 시간 감염시켰다 . 감염 후 바이러스를 제거하고 3배수로 희석한 실시예 1 내지 34 화합물, 또는 대조 화합물인 gemci tabine으로 처리한다 . 감염 48시간 후에 CeUTiter 96® AQueous One Solution Cel 1 Prol i ferat ion Assay (Pr이 nega, USA) 를 이용하여 약물의 세포 독성 (50% cytotoxicity concentrat ion, CC50)과 항바이러스 효과에 의한 ceU viability (50% effective concentration, EC50)를 측정하였다. CC50와 EC5o의 통계분 석은 GraphPad Prism 6 (GraphPad Software, USA) 프로그램을 사용하 였다. 그 결과를 하기 표 2에 정리하여 나타내었다.

EC50는 항바이러스 효과에 의한 cell viability로서 값이 낮을수 록 해당 화합물의 항바이러스 효과가 우수한 것이고;
CC50은 해당 농도에서 세포독성을 보이는 지표로 값이 높을수록 해당 화합물의 독성이 낮은 것이고;
S.I 값은 EC50/CC50 값을 바탕으로 SI 값을 계산한 것이며 값이 높을수록 해당 화합물의 효과는 우수하며 독성은 낮음을 의미하고; 및 n.cl.는 약효가 없음을 의미한다. 상기 표 2에 나타난 바와 같이,
본 발명에 따른 실시예 화합물들은 우수한 항코로나바이러스 활 성을 나타냄을 알 수 있다. 그중에서도 특히 실시예 1, 5, 6, 12, 17, 21 및 34 화합물은 다른 실시예 화합물과 비교하여 현저히 우수한 항 코로나바이러스 활성을 나타내는 것을 알 수 있다.
<1-4> 면역 블롯 분석
Huh— 7 세포를 6— well plate에 배양한 후 메르스 코로나바이러스 (M0I 0.02)로 1시간 감염시켰다. 감염 후 0.1 y M, 1 μ Μ, 10 μ Μ 후 보화합물 (실시예 1, 5, 6)로 처리하거나, 대조 약물 gemcitabine을 처 리하였다. 음성대조군은 0.02% DMS0로 처리하였다. 감염 24시간 후 , 30 μ g의 세포용해액을 사용하여 12% SDS-PAGE (sodium dodecyl SLII fat e-polyacryl amide gel e 1 ect r ophor es i s )에서 전기영동 루 PVDF (polyvinyl idene difluoride membrane , Mi 11 i pore , USA)로 전이하였다 메르스 코로나바이러스 NP 단백질은 항메르스 NP 단클론 항체 (Cat. 100211-RP02, Sino Biological Inc. , China)와 horseradish peroxidase(HRP)가 결합된 2차 항체를 이용하여 검출한다. 동일한 농 도의 단백질 loading 확인을 위하여 항 β -actin 항체를 이용하였다. SuperSi gna 1 West Pico Chemi luminescence Substrate (Pierce, USA)를
이용하여 단백질을 발색하고, 이미지는 LAS-4000 Luminescent Image Analyzer (Fuj if i lm, Japan)로 현상하였다. 그 결과를 도 1에 나타내 었다. 도 1은 메르스 코로나바이러스 감염된 Huh-7세포에서 실시예 화 합물의 항바이러스 활성효과의 면역블롯 분석을 보여준다ᅳ 도 1에 나타난 바와 같이, 본 발명에 따른 실시예 1, 5, 6 화합 물은 1 μΜ 이하의 농도에서 메르스 ΝΡ 단백질 발현 감소로 항메르스 활성을 나타내는 것으로 확인하였다ᅳ

화학식 1의 화합물을 포함하는 약학적 조성물은 코로나 바이러스 에 대해 우수한 억제 활성을 나타내므로 코로나 바이러스로 인해 유발 되는 메르스, 사스 등 질환의 예방 또는 치료에 유용하다.

하기 화학식 1로 표시되는 화합물, 이의 광학 이성질체 , 또는 이 의 약학적으로 허용 가능한 염:
(상기 화학식 1에서,
-L-은 단일결합 , -0- 또 -0-(CH2)-이고
R1은 -H, -0H 쇄 측쇄 Ci-io의 직쇄 이상의 할로
여기서, 상기 Ρ 및 q는 독립적으로 0 내지 3의 정수이고, 상기 A1은 -C00H, 또는 -NA2A3이고, 이때 상기 A2 및 A3는 독립적으로 -H, 비치환 또는 하나의 -CN이 치환된 d 의 직쇄 또는 측쇄 알킬이거나 A2 및 A3는 함께 연결되어 N을 하나 이상 포함하며 비치환 또는 하나 의 -0H가 치환된 5 내지 10 각환의 헤테로사이클로알킬이고,
여기서 , 상기 B1은 N, 0 및 S로 이루어지는 군으로부터 선택되는 1종 이상의 헤테로원자를 포함하는 비치환 또는 치환된 5 내지 10 각 환의 해테로사이클로알킬이고, 상기 치환된 5 내지 10 각환의 헤테로 사이클로알킬은 =0 및 =≤로 이루어지는 군으로부터 선택되는 1종 이상 의 치환기가 치환된 5 내지 10 각환의 헤테로사이클로알킬이고;
R2는 또는 할로겐이고
여기서 , 상기 치환된 페닐옥시는 할로겐, 나이트로, — C00H, Ci-5 의 직쇄 또는 측쇄 알킬, d-5의 직쇄 또는 측쇄 알콕시 및 d— 5의 직 쇄 또는 측쇄 알콕시카보닐로 이루어지는 군으로부터 선택되는 1종 이 상의 치환기가 치환된 페닐옥시이고,
여기서 , 상기 E1 및 E2는 독립적으로 d-5의 직쇄 또는 측쇄 알콕시카보닐, d-5의 직쇄 또는 측쇄 알킬카보닐 Ci-5의 직쇄 또는 측 쇄 알킬이고, .
여기서, 상기 B2는 하나 이상의 N을 포함하는 비치환 또는 치환 된 5 내지 10 각환의 헤테로아릴이고, 상기 치환된 5 내지 10 각환의 헤테로아릴은 하나 이상의 =0가 치환된 5 내지 10 각환의 헤테로아릴

R6은 -H, 할로겐, -10의 직쇄 또는 측쇄 알킬, (시의 직쇄 또 는 측쇄 알콕시, 또는 d 10의 직쇄 또는 측쇄 알킬설파닐이고; R7은 -H, 할로겐 , 또는 비치환 또는 하나 이상의 할로겐이 치환 된 C O의 직쇄 또는 측쇄 알킬이고 ;
1^는 할로겐, ^-^의 직쇄 또는 측쇄 알킬, C 0의 직쇄 또 는 측쇄 알콕시, C 0의 직쇄 또는 측쇄 알콕시카보닐 , (:6-20의 아릴카 보닐, 하나 이상의 메틸기로 치환된 (:6-10의 아릴 C -5의 직쇄 알킬카보 닐 d 3의 직쇄 알킬이거나 상기 R7과 함께 연결되어 비치환된 (:6ᅳ10의 아릴을 형성하고;
R9는 -H, 또는 할로겐이고; 및
R10은 -H, -CN, 할로겐 아민 . -10의 직쇄 또는 측쇄 알콕시카 보닐이거나 상기 R9와 함께 연결되어 비치환된 (:6_10의 아릴을 형성한 다 ). 【청구항 14】
