Abstract
Description
特許文献1は、iPS細胞等の多能性幹細胞の未分化状態を維持しつつ増殖させるために、アクチビン存在下で多能性幹細胞を培養することを開示する。
本開示は、本願発明者らが、さらなる鋭意研究を重ねた結果、ヘマグルチニンに多能性幹細胞の増殖促進に寄与する効果があることを見出したことに基づく。具体的には、ヘマグルチニンは、細胞間接着を緩和するように機能し、培養される多能性幹細胞のコロニーや細胞集塊内の細胞環境が増殖しやすいものとなり、増殖が促進されるものと推測される。さらに、細胞間接着が緩和されることで、培養される多能性幹細胞のコロニーの細胞間接着力が均一化し、「未分化状態から逸脱した細胞」の発生を抑制できると推測される。但し、本開示はこれらのメカニズムに限定して解釈されなくてもよい。
本開示において、多能性(pluripotent)幹細胞は、限定されない一又は複数の実施形態において、ヒ卜多能性幹細胞である。本開示においてヒ卜多能性幹細胞は、限定されない一又は複数の実施形態において、ヒ卜iPS細胞又はヒ卜ES細胞である。
本開示において未分化状態を逸脱した細胞(未分化逸脱細胞)とは、限定されない一又は複数の実施形態において、細胞の形態が未分化状態の細胞のそれとは異なり、判別可能である。さらに未分化逸脱細胞となったことは、限定されない一又は複数の実施形態において、未分化マーカーの消失で確認できる。未分化マーカーは、限定されない一又は複数の実施形態において、Oct3/4、Nanog、SSEA-4、TRA-1-60である。
本開示において、ヘマグルチニンは、限定されない一又は複数の実施形態において、ポツリヌス菌(Clostridiumbotulinum)のヘマグルチニンである。本開示において、ヘマグルチニンは、特に言及がない場合、神経毒素複合体(ヘマグルチニン複合体ともいう)をいう。多能性を有する細胞の増殖促進の点から、ヘマグルチニンは、ポツリヌス菌(Clostridiumbotulinum)の神経毒素複合体における3つのヘマグルチニンサブコンポーネントHA1(HA33)、HA2(HA17)及びHA3(HA70)からなる群から選択される2又は3成分で構成される複合体又はその複合体を含むものが挙げられる。ヘマグルチニンは、HA2(HA17)及びHA3(HA70)で構成される複合体若しくは3成分で構成される複合体又はその複合体を含むものである。
本開示において、ヘマグルチニンは、多能性幹細胞の増殖促進に寄与する効果を有する範囲で、ヘマグルチニンの改変体であってもよい。該改変体の一又は複数の実施形態として、サブコンポーネントの1つ、2つ又は3つのアミノ酸配列が、少なくとも1つのアミノ酸変異を有するものが挙げられる。該改変体としては、WO2015/199243に挙げるものが使用できる。
本開示において、特に言及のない場合、「ヘマグルチニン」は、「ヘマグルチニン改変体」を含む。また、本開示において、特に言及のない場合、「ヘマグルチニン改変体」は、多能性幹細胞の増殖促進に寄与する効果を有するものをいう。
本開示は、一態様として、ヘマグルチニン又はその改変体を含む、多能性幹細胞の増殖を促進するための組成物に関する。
多能性を有する細胞の増殖促進の点から、本開示に係る増殖促進組成物は、有効量のヘマグルチニン又はその改変体を含むことが好ましい。
本開示は、一態様として、本開示に係る増殖促進組成物が添加された培地で多能性幹細胞を培養することを含む、多能性幹細胞の増殖促進方法に関する。本開示に係る増殖促進組成物が添加される培地としては、特に制限されず、多能性幹細胞の培養に使用されうる培地でよい。本開示に係る増殖促進化合物の添加濃度は、上述のとおり、有効量が挙げられる。一又は複数の実施形態において、添加後の培地におけるヘマグルチニン又はその改変体の濃度が、5nM以上、10nM以上、又は15nM以上となる濃度が挙げられる。また、同様の観点から、200nM以下、150nM以下、又は100nM以下となる濃度が挙げられる。
本開示に係る増殖促進方法の一又は複数の実施形態として、単一細胞状態、又は分散状態の多能性幹細胞を播種すること、及び、ヘマグルチニン又はその改変体が添加された培地で多能性幹細胞を培養することを含む、多能性幹細胞の増殖を促進する方法に関する。単一細胞状態又は分散状態の多能性幹細胞集団をヘマグルチニン又はその改変体と接触させることで、一又は複数の実施形態において、培養した際、コロニーの中心であっても細胞増殖能を維持する細胞の存在数が増加し、均一性が向上した培養が可能となる。その結果細胞の増殖促進が効率化され、最終的に従来法と比べて高い増殖率での培養が可能となる。
本開示に係る増殖促進方法の一又は複数の実施形態として、多能性幹細胞の増殖を促進する方法であって、多能性幹細胞の細胞集塊の浮遊培養の培地にヘマグルチニン又はその改変体を添加し、かつ、前記細胞集塊を小塊に分割しないで培養することを含む増殖促進方法が挙げられる。本開示において、細胞集塊とは、一又は複数の実施形態において、多能性幹細胞を浮遊培養すると形成されるスフェロイド状の細胞塊をいう。本開示において、「細胞集塊を小塊に分割しない」とは、一又は複数の実施形態において、ヘマグルチニン又はその改変体の添加後に、一部又は全部の培養が終了するまで添加前の細胞集塊の大きさより小さな細胞集塊、分散細胞、及び単一細胞等を産生する作業若しくは処理をしないことをいう。
本実施形態では、均一性が高い培養が可能となるため、細胞に与えるダメージやストレスが小さいままに増殖を促進でき、結果的に細胞塊が分割することなく、効率よく細胞を増殖させることが可能となる。
本開示は、その他の態様において、本開示の増殖促進方法を行うことを含む、多能性幹細胞の培養方法に関する。本開示の増殖促進方法については、上記を参照できる。
本開示に係る培養方法によれば、多能性幹細胞の増殖を促進することができる。本開示に係る培養方法によれば、一又は複数の実施形態において、さらに、均一性の高い培養が可能となる。
[1] 多能性幹細胞の増殖を促進するための組成物であって、ヘマグルチニン又はその改変体を含む、増殖促進組成物。
[2] [1]に記載の増殖促進組成物が添加された培地で多能性幹細胞を培養することを含む、多能性幹細胞の増殖を促進する方法。
[3] 多能性幹細胞の増殖を促進する方法であって、単一細胞状態、又は分散状態の多能性幹細胞を播種すること、及び、ヘマグルチニン又はその改変体が添加された培地で前記多能性幹細胞を培養することを含む、方法。
[4] ヘマグルチニン又はその改変体を、多能性幹細胞のコロニーが形成される前に、培地に添加することを含む、[3]記載の増殖促進方法。
[5] 多能性幹細胞の増殖を促進する方法であって、多能性幹細胞の細胞集塊の浮遊培養の培地にヘマグルチニン又はその改変体を添加し、かつ、前記細胞集塊を小塊に分割しないで培養することを含む、方法。
[6] [2]から[5]のいずれかに記載の増殖促進方法を行うことを含む、多能性幹細胞の培養方法。
ヒトiPS細胞の単一細胞に由来するクローン細胞培養におけるヘマグルチニン(HA)の効果を確認した。iMatrixコート培養面にiPS細胞(単一細胞)を播種して下記条件で培養し、HAの有無による影響を確認した。

添加濃度:50 nM
添加頻度(培養1日目から毎日)
〔培養条件〕
37℃、5%CO2雰囲気下
培養開始時(t=0)からHA複合体が添加された培地を使用した。
〔観察〕
t=24 h、t=216 hにおいて、IN Cell Analyzer 2000(商品名、GEヘルスケアバイオサイエンス社製)で培養細胞を観察し画像を取得した。得られた顕微鏡観察写真を図1に示す。
図1に示されるとおり、HAが添加された培地を使用した培養では、HAが添加されていない培地を使用した培養に比べて、増殖の速度が高いことが認められた。また、HAが添加された培地を使用した培養では、HAが添加されていない培地を使用した培養に比べて、未分化を逸脱した細胞の出現頻度が抑制されていた。
iPS細胞の浮遊培養におけるヘマグルチニン(HA)の効果を確認した。

30 ml バイオリアクター(Able co. Ltd.)

1 x 105 細胞/mL

HA:Type B
添加濃度:10 nM
〔培養条件〕
37℃、5%CO2雰囲気下
培養開始時(t=0)の72時間後から24時間ごとに3日間、ヘマグルチニン濃度が10nMとなるように添加した(図2)。
〔観察〕
t=0から24時間ごとにおいてIN Cell Analyzer 2000(商品名、GEヘルスケアバイオサイエンス社製)で培養細胞を観察し画像を取得するとともに、培地の体積当たりの細胞密度を測定した(図3及び4)。
t=0からt=72 hまでを第1相、t=72 hからt=168 hまでを第2相として、培養時間を2つに分け、それぞれの相における細胞生存率(α)と、見かけ上の比増殖速度(μ)を算出した(表1)。下記表1に示されるとおり、HAが添加された第2相では、第1相及びHAが添加されてない第2相に比べて、増殖のスピードが優れていた。また、HAが添加された第2相では、第1相及びHAが添加されてない第2相に比べて、細胞集塊の大きさが均一であり、また、未分化を逸脱した細胞の出現頻度が抑制されていた。

ヒトiPS細胞集塊の増殖に対するHAの添加効果として、細胞増殖特性と、集塊内における細胞分裂の局在性の2点について評価を行った。

添加濃度(培養24h以降):0, 5, 10 nM
〔培養条件〕
37℃、5%CO2雰囲気下
播種細胞数:3.0 x 103 cells/well
培養時間240 時間
〔測定項目〕
細胞増殖特性:集塊細胞数と、培養24 hから168 hにおける見かけ上の比増殖速度(μ)を測定した。
集塊内特性:凍結切片に対する蛍光染色法による観察(核=DAPI,分裂可能細胞=抗ki-67抗体)により細胞分裂の局在性を観察した。
測定頻度:培養 24 h以降 48 h毎
Tic株を、HAの添加濃度を0, 5, 10 nM含む培地を用いて、上記の培養条件で培養を行った。増殖特性として、集塊細胞数、及び、比増殖速度(μ)を測定した。
集塊細胞数の結果を図5に示す。同図のおとり、HA添加条件(添加濃度:△=5 nM、□=10 nM)は、無添加条件(〇=0 nM)に比べて集塊細胞数が増加することが確認された。 比増殖速度(μ)は、培養24 hから168 hにおける見かけ上の比増殖速度として下記式に基づき算出した。その結果を下記表2に示す。同表のとおり、比増殖速度(μ)の値は、HA添加時に無添加時に比べて高くなり、増殖が促進されていることが確認された。

集塊内特性として、蛍光染色法による顕微鏡観察を行った。核をDAPIで染色(赤)し、分裂可能細胞を抗Ki-67抗体で染色(緑)して集塊内での分裂可能細胞の局在性を確認した。その結果の一例を図6に示す。
HA無添加条件では、培養24 hにおいて分裂可能細胞(Ki-67陽性細胞)は、集塊内にてほぼ均一に存在していいた(黄色い部分)。その後、集塊内部においては、分裂可能細胞は観察されず、周辺部のみの局在性を示した。すなわち、集塊周辺部のみで分裂が行われることで細胞集塊の細胞数が増えることが確認された。
HA添加条件(5, 10 nM)では、培養24 hにおいてHA無添加での培養同様の傾向がみられたが、その後の培養において、集塊周辺部での分裂可能細胞の頻度がHA無添加に比べて高くなった。10 nM HA添加での培養では、集塊内部でも分裂可能細胞が存在することが確認された。以上により,HAの添加により,集塊内の細胞分裂頻度が向上し,増殖速度が上昇することが示された。また、HAによる増殖速度上昇が、HA濃度依存性を示すことが確認された。

- 多能性幹細胞の増殖を促進するための組成物であって、
ヘマグルチニン又はその改変体を含む、増殖促進組成物。 - 請求項1に記載の増殖促進組成物が添加された培地で多能性幹細胞を培養することを含む、多能性幹細胞の増殖を促進する方法。
- 多能性幹細胞の増殖を促進する方法であって、
単一細胞状態、又は分散状態の多能性幹細胞を播種すること、及び、
ヘマグルチニン又はその改変体が添加された培地で前記多能性幹細胞を培養することを含む、方法。 - ヘマグルチニン又はその改変体を、多能性幹細胞のコロニーが形成される前に、培地に添加することを含む、請求項3記載の方法。
- 多能性幹細胞の増殖を促進する方法であって、
多能性幹細胞の細胞集塊の浮遊培養の培地にヘマグルチニン又はその改変体を添加し、かつ、前記細胞集塊を小塊に分割しないで培養することを含む、方法。 - 請求項2から5のいずれかに記載の増殖促進方法を行うことを含む、多能性幹細胞の培養方法。
