Abstract

水への溶解性に優れるノビレチン組成物およびそれを簡便に製造する方法の提供。 ノビレチンを含有する固体分散体の製造方法であって、ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程と、溶融した溶融物を冷却し、固化させる工程を含む、製造方法。当該方法により製造されるノビレチン含有組成物。

Description

ノビレチン含有固体分散体の製造方法
 本発明は、ノビレチンを含有する固体分散体の製造方法に関する。
 ノビレチンは、温州みかんやシークワーサー等の柑橘類に含まれるポリメトキシフラボノイドの一種である。ノビレチンは、PPAR活性化作用及びアディポネクチン分泌促進作用(特許文献1)、神経突起伸長促進作用(特許文献2)、記憶障害抑制作用(非特許文献1)等の種々の生理活性を有することが知られていることから、健康食品の重要な成分として認識されている。
 しかしながら、ノビレチンは難水溶性であり、その原体そのものの生理機能を飲食品・医薬品等で有効活用することは難しい。
 そこで、ノビレチンを水に可溶化させる技術が検討され、例えばシクロデキストリン包接物とする方法が報告されている(特許文献3)。しかしながら、この製造方法では、溶媒としてエタノール水溶液を用いているため、その除去のためにプロセスが煩雑化し、コスト上昇の一因になるという問題があり、また水溶性の面でも十分ではない。
 一方、難水溶性ポリフェノール類に水溶性ポリフェノールを混合し、加熱溶融した後に冷却、固化した場合に難水溶性ポリフェノール類の水への溶解性が高まることが報告されている(特許文献4)。しかしながら、当該文献にはノビレチンの可溶化については全く記載されておらず、その効果は明らかではない。

 本発明は、以下の(1)~(5)に係るものである。
(1)ノビレチンを含有する固体分散体の製造方法であって、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程と、溶融した溶融物を冷却し、固化させる工程を含む、製造方法。
(2)ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を含有し、ノビレチンがX線回折スペクトルから算出される結晶化度10%以下の状態で分散してなるノビレチン含有固体分散体。
(3)ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を含有し、ノビレチンが分散してなる、X線回折スペクトルから算出される結晶化度10%以下のノビレチン含有固体分散体。
(4)(1)の製造方法により得られ、X線回折スペクトルから算出されるノビレチンの結晶化度が10%以下であるノビレチン含有固体分散体。
(5)(2)~(4)のいずれかのノビレチン含有固体分散体を含有する飲食品、医薬品又は医薬部外品。

発明の詳細な説明
 本発明は、水への溶解性に優れるノビレチン組成物を簡便に製造する方法を提供することに関する。
 本発明者は、上記課題に鑑み鋭意検討したところ、ノビレチン又はノビレチン含有物に水溶性ヘスペリジン誘導体を混合し、加熱溶融させた後、冷却、固化させると、ノビレチンが非晶質の状態で分散した固体分散体が得られること、斯かる固体分散体は、ノビレチンの水への溶解性が極めて高く、且つ水中での高い溶解性が安定的に維持され、また高い生体膜透過性を有すること、更には、動物の経口投与において、高い血中移行性、組織移行性を与えることを見出した。
 本発明によれば、ノビレチンの水への溶解性が飛躍的に向上したノビレチン含有固体分散体を提供することができる。本発明の固体分散体を用いることにより、ノビレチンの生体膜透過性、経口摂取時の血中移行性並びに組織移行性を向上でき、ノビレチンの生理機能を増強することなどが期待される。また、本発明の固体分散体は、製造工程中に有機溶媒を用いないため飲食品への使用に好適である。
 本発明において、「ノビレチン」とは、3’,4’,5,6,7,8-ヘキサメトキシフラボンであり、以下の構造を有する。

 ノビレチンとしては、例えば、化学合成したもの、天然物から分離したもの、化学合成や天然物から分離した後に精製したもの、市販品等、何れのものでも良い。ノビレチンの純度は特に限定されず、例えば、固体分散体とした際に所望の薬理効果が発揮できる程度の純度であればよい。例えば、柑橘類の果実や搾汁残渣を、メタノールやエタノール等のノビレチン可溶性の有機溶媒で抽出処理し、適宜分離・精製して得られる、ノビレチンを高含有率又は高含有量で含む「ノビレチン含有物」を用いることもできる。ノビレチン含有物としては、例えばノビレチンを高含有量で含むシークワーサーエキス(例えば「PMF90」<株式会社沖縄リサーチセンター製、ノビレチン含有率約60重量%>等)が挙げられる。
 本発明において用いられる「水溶性ヘスペリジン誘導体」としては、ヘスペリジンを酵素又は化学処理することにより水溶性を高めたヘスペリジン類が挙げられ、例えばグルコシルヘスペリジン等のヘスペリジンの糖部分(ルチノース部分)に別の糖を結合させた糖転移ヘスペリジンやメチルヘスペリジンが挙げられる。このうち水溶性の点から、メチルヘスペリジン、グルコシルへスペリジンが好ましい。
 「メチルヘスペリジン」としては、主に、カルコン型化合物(1)及びフラバノン型化合物(2)が含まれることが知られており、その構成成分として、例えば以下に示す構造のものが挙げられる。

 ここで、医薬品添加物および食品添加物としてのメチルヘスペリジンは、主に、化合物(3)及び(4)の混合物として取り扱われている。

 (式中、Glは、グルコース残基、Rhは、ラムノース残基を表す。また、Gl-2は、グルコース残基の2位((3-1)の場合、3位も含む)、Rh-2は、ラムノース残基の2位を表す。)
 また、化粧品原料としてのヘスペリジンメチルカルコンは、(5)で示される化合物として取り扱われている。なお、カルコン型化合物を多く含む組成の場合、ヘスペリジンメチルカルコンとも呼ばれる。

 本発明において用いられるメチルヘスペリジンは、上記で示したカルコン型化合物(1)とフラバノン型化合物(2)の両方を含むものでもよいし、また、それぞれの片方のみを含むものでもよい。
 本発明において、より好適なメチルヘスペリジンとしては、化合物(3)と化合物(4)の混合物が挙げられる。
 メチルヘスペリジンは、公知の方法、例えば、ヘスペリジンを水酸化ナトリウム水溶液に溶かし、そのアルカリ溶液に対応量のジメチル硫酸を作用させ、反応液を硫酸で中和し、n-ブチルアルコールで抽出し、溶媒を留去したのち、イソプロピルアルコールで再結晶することにより製造できるが(崎浴、日本化學雑誌、79、733-6(1958))、その製造法はこれに限るものではない。
 メチルヘスペリジンとして市販のメチルヘスペリジン含有製剤を使用してもよく、例えば、「メチルヘスペリジン」(東京化成工業(株))、「ヘスペリジンメチルカルコン」(Sigma社)、「メチルヘスペリジン」(浜理薬品工業(株))、「メチルヘスペリジン」(昭和電工(株))、「メチルヘスペリジン」(アルプス薬品工業(株))が挙げられる。
 また、グルコシルヘスペリジンとしては、例えばモノグルコシルヘスペリジン(「αGヘスペリジン PA-T」(東洋製糖(株))、「林原ヘスペリジン(登録商標)S」(林原株式会社)等が挙げられる。
 本発明のノビレチンを含有する固体分散体は、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程と、溶融した溶融物を冷却し、固化させることにより製造できる。また、必要であるならば、加熱溶融前のノビレチンと水溶性ヘスペリジン誘導体に各種可塑剤や高分子を混合することもできる。
 ここで、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合する際、ノビレチンの含有量は、固体分散体中のノビレチンの含有量を高める点から、混合物中に1質量%以上、更に3質量%以上、更に4質量%以上、更に5質量%以上、更に7.5質量%以上、更に10質量%以上、更に15質量%以上が好ましく、また、加工しやすい点から、90質量%以下、更に70質量%以下、更に60質量%以下、更に50質量%以下、更に45質量%以下、更に40質量%以下、更に35質量%以下が好ましい。また、混合物中のノビレチンの含有量は、1~90質量%、更に3~70質量%、更に4~60質量%、更に5~50質量%、更に7.5~45質量%、更に10~40質量%、更に15~35質量%が好ましく、25%が殊更好ましい。
 水溶性ヘスペリジン誘導体の含有量は、その種類によって異なるが、ノビレチンの水への溶解性の点から、混合物中に10質量%以上、更に20質量%以上、更に25質量%以上、更に30%以上、更に45%以上、更に60%以上、更に65%以上が好ましく、また、固体分散体の調製がしやすい点から、99質量%以下、更に97質量%以下、更に96質量%以下、更に92.5%以下、更に90%以下、更に85%以下が好ましい。また、前記混合物中のメチルヘスペリジンの含有量は、10~99質量%、更に20~97質量%、更に25~96質量%、更に30~92.5%質量%、更に45~90%質量%、更に60~85質量%、更に65~85質量%が好ましく、75%が殊更好ましい。
 本発明において、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合する際、水溶性ヘスペリジン誘導体に対するノビレチンの質量比[ノビレチン/水溶性ヘスペリジン誘導体]は、得られる固体分散体中のノビレチンの含有量を高める点、また、固体分散体を調製しやすさの点から、0.01以上が好ましく、0.03以上がより好ましく、0.04以上が更に好ましく、0.07以上が更に好ましく、0.1以上が更に好ましい。また、得られるノビレチンの水への溶解性を高める点から、9以下が好ましく、更に4以下がより好ましく、3以下が更に好ましく、1以下が更に好ましく、0.67以下が更に好ましい。また、0.01~9が好ましく、0.03~4がより好ましく、0.04~3がより好ましく、更には0.07~1が、更には0.1~0.67が好ましく、0.33が殊更好ましい。
 ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体の混合物には、可塑剤として糖や糖アルコールを含有させることができる。例えば、グルコース、フルクトース、マルトース、マンノース、ラムノース、リボース、キシロース、トレハロース、キシリトール、ソルビトール、マンニトール、エリスリトール、アラビノース、イノシトール、グルコサミン、スクラロース、ソルビトールが好ましい。
 また、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体の混合物には、コスト低減や溶解挙動の改善、また粉体物性の改善を目的として各種高分子を含有させることが出来る。例えば、アマシードガム、アラビアガム、アラビノガラクタン、ウェランガム、カシアガム、ガディガム、カードラン、カラギナン、キサンタンガム、キチン、キトサン、グァーガム、ジェランガム、シクロデキストリン、タマリンドシードガム、タラガム、トラガントガム、微結晶セルロース、微小繊維状セルロース、ナイシン、プルラン、ペクチン、マクロホモプシスガム、ラムザンガム、アセチル化アジピン酸架橋デンプン、アセチル化酸化デンプン、アセチル化リン酸架橋デンプン、アルギン酸及びその塩、オクテニルコハク酸デンプンナトリウム、カゼイン及びその塩、カルボキシメチルセルロース及びその塩、コンドロイチン硫酸ナトリウム、ヒドロキシプロピル化リン酸架橋デンプン、ヒドロキシプロピルセルロース、ヒドロキシプロピルデンプン、ヒドロキシプロピルメチルセルロース、メチルセルロース、リン酸架橋デンプン、リン酸化デンプンなど、多糖及びその誘導体やたんぱく質、ポリアクリル酸ナトリウム、ポリビニルピロリドン、ポリビニルポリピロリドンなどの合成高分子が好ましい。
 可塑剤及び各種高分子の含有量は、その種類によって異なるが、ノビレチンの溶解度の点から混合物中に5質量%以上、更に10質量%以上、更に15質量%以上が好ましく、更に25質量%以上が好ましく、また、固体分散体の調製がしやすい点から、90質量%以下、更に85質量%以下、更に80質量%以下、更に70質量%以下、更に65質量%以下、更に50質量%以下が好ましい。また、前記混合物中の可塑剤及び各種高分子の含有量は、5~90質量%、更に10~90質量%、更に15~85質量%、更に15~80質量%、更に25~70質量%が好ましく、更に25~65質量%、更に25~50質量%が好ましい。
 ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体と可塑剤・各種高分子を混合する際のノビレチンと可塑剤・各種高分子成分の質量比[可塑剤・各種高分子/ノビレチン]は、固体分散体を調製しやすい点から、0.2以上が好ましく、0.3以上がより好ましい。また、ノビレチンの水への溶解度を高める点から、19以下が好ましく、17以下がより好ましく、9以下がより好ましく、6以下がより好ましく、2以下がより好ましい。また、斯かる質量比は、0.2~19、更に0.2~17、更に0.2~9、更に0.3~6、更に0.3~2が好ましい。
 ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体と可塑剤・各種高分子を混合する際の水溶性ヘスペリジン誘導体と可塑剤・各種高分子の質量比[可塑剤・各種高分子/水溶性ヘスペリジン誘導体]は、固体分散体を調製しやすい点から、0.05以上、0.06以上、より好ましくは0.08以上、更に好ましくは0.11以上である。また、ノビレチンの水への溶解度を高めるための点から、100以下が好ましく、17以下が好ましく、4以下がより好ましく、2以下がよりいっそう好ましい。また、斯かる質量比は、0.05~100、0.05~17、更に0.08~4、更に0.11~2が好ましい。
 ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体と可塑剤・各種高分子を混合する際の水溶性ヘスペリジン誘導体と可塑剤・各種高分子の合計量に対するノビレチンの質量比[ノビレチン/(水溶性ヘスペリジン誘導体+可塑剤・各種高分子)]は、固体分散体中のノビレチンの含有量を高める点から、0.01以上が好ましく、0.02以上がより好ましく、0.04以上が更に好ましく、0.07以上が更に好ましく、0.1以上が更に好ましく、0.17以上が更に好ましく、0,25以上がより更に好ましい。また、得られるノビレチンの水への溶解性を高める点から、9以下が好ましく、更に4以下がより好ましく、3以下が更に好ましく、1以下が更に好ましく、0.67以下が更に好ましく、0.55以下が更に好ましく、0.43以下が更に好ましい。また、0.01~9が好ましく、0.03~4がより好ましく、0.04~3が、更には0.07~1が、更には0.1~0.67が、更には0.17~0.55が、更には0.25~0.43が好ましい。
 ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体の混合物を加熱して溶融させる方法は、特に制限されず、公知の方法を適用できるが、混合物を攪拌しながら加熱溶融させることが好ましい。例えば、エクストルーダー、ニーダー等の混練機、押出機を用いることができる。また、リボンミキサー等の攪拌機を用いることができる。例えば、HAAKE製のエクストルーダー、Thermo scientific製のエクストルーダー、テクノベル(株)製のKZW134T、(株)栗本鉄工所のKRCニーダー、浅田鉄工製のミラクルK.C.K、スエヒロEPM製のEA-20、エヌピー食品(株)製のMC-1102等が挙げられる。加熱の手段は、例えば、水蒸気、電気が挙げられる。
 なかでも、均一な組成の固体分散体を得る観点から、混練と加熱溶融を同時に行うことのできるスクリューを備えた押出機を用いるのが好ましい。スクリューを備えた押出機としては、単軸、二軸のどちらの形式でもよいが、搬送能力を高める等の観点から、二軸押出機が好ましい。二軸押出機としては、シリンダの内部に2本のスクリューが回転自在に挿入された押出機が好ましく、従来から公知のものを使用できる。2本のスクリューの回転方向は、同一でも逆方向でもよいが、搬送能力を高める観点から、同一方向の回転が好ましい。また、スクリューの噛み合い条件としては、完全噛み合い、部分噛み合い、非噛み合いの各形式の押出機のいずれでもよいが、処理能力を向上させる観点から、完全噛み合い型、部分噛み合い型が好ましい。
 また、スクリューを備えた押出機としては、強い圧縮せん断力を加える観点から、スクリューのいずれかの部分に、いわゆるニーディングディスク部を備えることが好ましい。ニーディングディスク部とは、複数のニーディングディスクで構成され、これらを連続して、一定の位相で、例えば90度ずつ、ずらしながら組み合わせたものであり、スクリューの回転にともなって、狭い隙間に、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体の混合物を強制的に通過させることで極めて強いせん断力を付与することができる。スクリューの構成としては、ニーディングディスク部と複数のスクリューセグメントとが交互に配置されることが好ましい。二軸押出機の場合、2本のスクリューが、同一の構成を有することが好ましい。
 スクリューを備えた押出機を用いる際は、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体の混合物、好ましくは当該混合物を粗粉砕したものを押出機に投入し、スクリューを回転させることにより、連続的に処理する方法が好ましい。
 スクリュー回転数は、30~500r/minが好ましく、50~300r/minがより好ましく、50~250r/minが更に好ましく、80~200r/minが更に好ましく、80r/minが殊更好ましい。
 また、せん断速度としては、10sec-1以上が好ましく、20~30000sec-1がより好ましく、50~3000sec-1が更に好ましい。せん断速度が10sec-1以上であれば、有効に粉砕が進行するため好ましい。
 スクリューを備えた押出機中で溶融した溶融物は、押出し、成形される。
 押出機によるパス回数は、1パスでも十分効果を得ることができるが、ノビレチンの分散性を向上させる点から、2パス以上行うことが好ましい。また、生産性の点からは、1~10パスが好ましい。パスを繰返すことにより、粗大粒子が粉砕され、粒径のばらつきが少ないノビレチンを含有する固体分散体を得ることができる。2パス以上行う場合、生産能力を考慮し、複数の押出機を直列に並べて処理を行ってもよい。
 加熱温度は、水溶性ヘスペリジン誘導体の軟化点以上であり、好ましくは水溶性ヘスペリジン誘導体の融点以上である。本発明において、軟化点とは固体物質を加熱していく際に物質が軟化し、変形し始める温度であり、例えばメチルヘスペリジンの軟化点は98℃、グルコシルヘスペリジンの融点は150℃である。加熱によりメチルヘスペリジンが軟化或いは溶融すると、そこにノビレチンが溶融する。具体的には、例えば通常のメチルヘスペリジンの融点は135℃である。
 本発明において、加熱温度は、95℃以上が好ましく、更に100℃以上が好ましく、更に105℃以上が好ましく、また、ノビレチンの熱安定性の点から、200℃以下が好ましく、更に185℃以下、更に180℃以下、更に160℃以下、更に140℃以下が好ましい。また、加熱温度は、95~200℃、更に95~180℃、更に100~185℃、更に100~160℃、更に105~140℃が好ましく、130℃が殊更好ましい。
 加熱の時間は、水溶性ヘスペリジン誘導体とノビレチンの熱安定性と生産性の点から、水溶性ヘスペリジン誘導体が溶融する温度に達してから30分以下が好ましく、更に15分以下が好ましく、更に10分以下が好ましく、また、水溶性ヘスペリジン誘導体とノビレチンの混合、およびノビレチンの水への溶解性の点から、1分以上が好ましく、更に3分以上が好ましく、更に5分以上が好ましい。また、加熱の時間は、水溶性ヘスペリジン誘導体が溶融する温度に達してから1~30分が好ましく、更に3~15分、更に5~10分が好ましく、10分が殊更好ましい。
 次いで、溶融物を冷却し、固化させる。斯かる処理によって、ノビレチンが非晶質化し、ノビレチンを非晶質の状態で含む固体分散体となる。
 非晶質とは、分子配列に一定の規則性を欠いている状態である。非晶質(アモルファス)であることは、粉末X線回折によって確認できる。
 本発明において固体分散体中のノビレチンは、結晶化度が50%以下であるものが好ましく、更に結晶化度が40%以下であるものが好ましく、更に結晶化度が20%以下であるのものが好ましく、更に結晶化度が10%以下であるのものが好ましく、更に結晶化度が5%以下であるのものが好ましく、更に結晶化度が0%の完全な非晶質であることが殊更好ましい。
 本発明の固体分散体は、粉末X線回折測定において、ノビレチンの結晶性回折ピークが検出されないのが好ましい。
 ノビレチンの結晶化度は、以下の方法で算出できる。まず、X線回折法による回折強度値から、非干渉性散乱や格子の乱れ等の影響を考慮しないで、プロファイル・フィッティングの手法を用いて結晶性回折線と非晶ハローにピーク分離する。次に、そこで得られた各ピークの積分強度よりノビレチンの結晶化度を下記計算式〔1〕により算出する。
 ノビレチンの結晶化度(%)=[ΣIα/(ΣIα+ΣIam)]×100 〔1〕
 [ΣIαは、結晶性回折線の各ピークの積分強度の和、ΣIamは、非晶部の回折線の各ピークの積分強度の和]
 溶融物冷却時の冷媒温度は、水溶性ヘスペリジン誘導体が溶融する温度よりも低い温度であり、50℃以下が好ましく、更に30℃以下が好ましい。冷却方法は、例えば、固体分散体を50℃以下、更に、30℃以下、更に室温(25℃)の雰囲気下におくことが好ましい。また、加熱処理後の固体分散体に冷風を吹き付けて急冷することが好ましい。加熱処理温度から50℃まで低下するのに要した時間から算出される固体分散体の冷却速度は0.1℃/s以上、更に0.2℃/s以上、更に0.3℃/s以上が好ましく、製造設備の制約等の観点から、例えば100℃/s以下、更には50℃/s以下が好ましい。冷却時間は30分以下が好ましく、更に20分以下が好ましく、更に10分以下が好ましく、更に5分以下が好ましい。
 冷却により固化したノビレチンおよび水溶性ヘスペリジン誘導体を含有する固体分散体は、任意の形、大きさに成形可能であり、例えばペレット状、顆粒状等が挙げられる。更に、必要に応じて粉砕してもよい。
 斯くして得られる本発明のノビレチンを含有する固体分散体(「ノビレチン含有固体分散体」)は水への溶解性(初期溶解性、経時的溶解性)に極めて優れる。
 例えば後記実施例に示すように、ノビレチンの経時的溶解性(例えば、ノビレチン含有固体分散体を水に添加して6時間撹拌した場合のノビレチン溶解濃度(攪拌開始6時間後までの曲線下面積(横軸:時間(単位=分)、縦軸:溶解濃度(単位=ppm))は、約2.20×10ppm・minと極めて高く、高い溶解濃度が長時間維持される(図1参照)。斯かる高い溶解性は、ノビレチンと同様に難溶性のポリフェノールとして知られている化合物(例えばヘスペリジン、セサミン、エラグ酸)を水溶性ヘスペリジン誘導体と混合して製造される固体分散体では得られない予想外の効果である。
 また、本発明のノビレチン含有固体分散体は、ノビレチンのヒト小腸上皮細胞由来の細胞膜に対する透過性が極めて優れており、当該効果もまた上記他の難溶性のポリフェノールの固体分散体からは予測できないものである。
 また、固体分散体中の水分含量は、微細化し易く、ハンドリング性が良好な点から、20質量%以下、更に10質量%以下、更に7質量%以下、更に5質量%以下が好ましい。
 本発明の製造方法で得られるノビレチンを含有する固体分散体は、様々な飲食品や医薬品、医薬部外品、化粧品等に使用することができる。とりわけ、水系の製品に利用するのが有用である。
 例えば、飲食品としては、飲料、パン類、麺類、クッキー等の菓子類、スナック類、ゼリー類、乳製品、冷凍食品、粉末コーヒー等のインスタント食品、でんぷん加工製品、加工肉製品、その他加工食品、調味料、栄養補助食品等の液状、固形状又は半固形状の飲食品が挙げられる。また、医薬品又は医薬部外品としては、錠剤(チュアブル錠等)、カプセル剤、粉末剤等の剤型が挙げられる。また、化粧品としては、洗浄料、化粧水、メイクアップ用化粧料、日焼け止め用化粧料、ニキビ用化粧料、デオドラント用化粧料、美白用化粧料、洗髪剤、育毛剤等が挙げられる。
 本発明の態様及び好ましい実施態様を以下に示す。
<1>ノビレチンを含有する固体分散体の製造方法であって、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程と、溶融した溶融物を冷却し、固化させる工程を含む、製造方法。
<2><1>において、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体混合物中の、ノビレチンの含有量は、混合物中に好ましくは1質量%以上、更に好ましくは3質量%以上、更に好ましくは4質量%以上、更に好ましくは7.5質量%以上、更に好ましくは10質量%以上、更に好ましくは15質量%以上であり、好ましくは90質量%以下、更に好ましくは80質量%以下、更に好ましくは75質量%以下、更に好ましくは50質量%以下、更に好ましくは40質量%以下、更に好ましくは35質量%以下である。また、好ましくは1~90質量%、更に好ましくは3~80質量%、更に好ましくは4~75質量%、更に好ましくは7.5~50質量%、更に好ましくは10~40%、更に好ましくは15~35%であり、殊更好ましくは25%である。

<4><1>~<3>において、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合する際のノビレチンと水溶性ヘスペリジン誘導体質量比[ノビレチン/水溶性ヘスペリジン誘導体]は、好ましくは0.01以上、更に好ましくは0.03以上、更に好ましくは0.04以上、更に好ましくは0.07以上、更に好ましくは0.1以上であり、好ましくは9以下、更に好ましくは4以下、更に好ましくは3以下、更に好ましくは1以下、更に好ましくは0.67以下である。また、好ましくは0.01~9、更に好ましくは0.03~4、更に好ましくは0.04~3、更に好ましくは0.07~1、更に好ましくは0.1~0.67であり、殊更好ましくは0.33である。
<5>スクリューを備えた押出機を用いて、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程が行われる<1>~<4>のいずれかの固体分散体の製造方法。

〔難水溶性物質の定量〕
 難水溶性ポリフェノール類の定量は、アジレント・テクノロジー株式会社製高速液体クロマトグラフを用い、化学物質評価研究機構製カラムL-Column ODS―2 (4.6mmφ×50mm、2μm)を装着し、カラム温度40℃でグラジエント法により行った。
 試料注入量は10μL、移動相A液は0.1重量%トリフルオロ酢酸水溶液、B液はアセトニトリルとし、1.0mL/分で送液した。グラジエント条件は以下のとおりである。

 検出は、ノビレチンは波長320nm、エラグ酸は波長254nm、ヘスペリジン、セサミンは波長280nmの吸光度により定量した。
〔モノグルコシルヘスペリジン(mGHes)の精製方法〕
 mGHesの精製には、ジーエルサイエンス株式会社製高速液体分取クロマトグラフを用い、ジーエルサイエンス株式会社製カラムInertsil ODS―3(14mmφ×250mm、5μm)を装着し、カラム温度40℃でイソクラティック法により行った。
 分取試料はαGヘスペリジン PA-T(東洋製糖株式会社製)の20w/v%水溶液を用い、一回の分取における試料注入量は250μLとした。また、移動相はA液を0.1重量%トリフルオロ酢酸水溶液、B液をアセトニトリルとし、A:B=25:75の混合液を用い、15.0mL/分で送液した。検出は波長280nmの吸光度を用いた。
 本法により得られたmGHes含有溶液を減圧濃縮し、アセトニトリルを除去した。本濃縮液をダイヤイオンHP-20(三菱化学株式会社製樹脂)を用いたカラムクロマトグラフィーにより精製、減圧濃縮した後に凍結乾燥することでmGHesを粉末として得た。本品を固体分散体の原料として使用した。
〔X線回折分析〕
 X線回折強度は、株式会社リガク製の「MiniFlexII」を用いて、X線源:Cu/Kα-radiation、管電圧:30kV、管電流:15mA、測定範囲:回折角:5~40°、X線スキャンスピード:10°/minの条件で測定した。測定用試料は、面積400mm2×厚さ0.5mmのペレットを圧縮して作製した。
〔経時的な溶解性の評価〕
 50mLのスクリュー管(株式会社マルエム製、No.2、褐色)に、イオン交換水を30mL加えた後、固体分散体100mg(難水溶性物質25mg含有)添加、2cmスターラーチップを用いて500rpmで攪拌した。攪拌開始を0分とし、攪拌6時間までの間、適宜その一部を0.45μmセルロースアセテートフィルターでろ過し、前記〔難水溶性物質の定量〕に記載した方法によりノビレチン溶解量を定量した。
〔Caco-2細胞膜透過性評価〕
 試験例1 ヒト大腸癌由来上皮細胞を用いた、難水溶性物質原体およびその固体分散体の小腸上皮の透過促進試験
 Caco-2細胞(ヒト大腸癌由来上皮細胞、DSファーマバイオケミカル社より入手)を37℃、5%CO存在下で培養した。培養にはCaco-2細胞分化培地セット(Corning社製)、及びバイオコートフィブリラーコラーゲンHTSマルチウェルインサート(24ウェル、膜孔1μm、Corning社製)を用いた。MITO+TMシーラム・エクステンダーを添加したシーディング基本培地にCaco-2細胞を懸濁してインサート上部(頂端膜側)に播種し、インサート下部(基底膜側)にも同培地を添加した。24時間のCaco-2細胞培養後に、前記シーディング基本培地をMITO+TMシーラム・エクステンダーを添加したEntero―STIM腸上皮分化培地に交換した。その後、さらに48時間前記細胞を培養し、小腸上皮細胞シート様に分化させた。
 腸管のタイトジャンクションの形成を確認するため、難水溶性物質原体および固体分散体の小腸上皮の透過促進試験直前にMillicell ERS(Millipore社製)を用いて経上皮電気抵抗(TEER)値を測定し、規定値(350Ω・cm)以上のCaco-2細胞を用いた。
 評価サンプルの調製は以下の通り行った。20mLのスクリュー管(株式会社マルエム製、No.2、褐色)に、HBSS(Invitrogen社製、10mM MES、5mM グルコース、10mM グルタミン、1mM アスコルビン酸を添加しpH6.0に調製)を6mL加えた後、難水溶性物質原体10mgもしくは固体分散体40mg(難水溶性物質10mg含有)添加し、2cmスターラーチップを用いて300rpmで10分間攪拌した。攪拌後、0.45μmセルロースアセテートフィルターでろ過した溶液、およびその希釈液を評価サンプルとして用いた。なお、表1には、本ろ液をHBSSで30倍に希釈した溶液をインサート上部仕込み液に使用した評価の結果を示した。
 TEER測定による細胞選別後、インサート上部及び下部をHBSSで置換して2回洗浄した。その後、インサート上部には、前記方法で調製した難水溶性物質およびその固体分散体水溶液、その希釈液を添加した。各抽出物の評価濃度を表2に示す。インサート下部には、HBSS(10mM HEPES、5mM グルコース、10mM グルタミン、pH7.4)を添加した。そしてCaco-2細胞を37℃、5%CO存在下で4時間培養した。
 その後、インサート下部のHBSSを回収し、下記の方法で小腸上皮を透過した難水溶性物質を、液体クロマトグラフ・タンデム型質量分析法(LC-MS/MS)により定量した。

 アジレント・テクノロジー株式会社製高速液体クロマトグラフを用い、化学物質評価研究機構製カラムL-Column ODS―2 (4.6mmφ×50mm、2μm)を装着し、カラム温度40℃でグラジエント法により行った。
 ノビレチン、セサミン、ヘスペリジンの測定では、試料注入量は10μL、移動相A液は0.1重量%ぎ酸水溶液、B液はアセトニトリルとし、0.6mL/分で送液した。グラジエント条件は以下の通りである。

 質量分析装置はQTRAP(R)4500(株式会社エービーサイエックス製)を用いた。スキャン条件はMRM,パラメータテーブルはアナライトで測定した。ノビレチンはポジティブイオンモードによりMRM(m/Z)404.0→374.0のイオンを、セサミンはポジティブイオンモードによりMRM(m/Z)355→276.9のイオンを、ヘスペリジンはポジティブイオンモードによりMRM(m/Z)611.2→71.0のイオンを、エラグ酸はネガティブモードによりMRM(m/Z)300.9→284.0のイオンをそれぞれ検出することで定量した。
[LDH活性測定試験による細胞生存率評価]
試験例2 LDH活性測定試験
 試験例1のインサート上部のHBSSを回収し、下記の方法で、細胞膜を構成するCaco-2細胞から放出された溶液中のLDH(Lactate dehydrogenase)活性測定から求まる、各評価サンプルの細胞傷害性から細胞生存率を評価した。
 細胞傷害性の評価は、LDH Cytotoxicity Assay Kit(Cayman Chemical Company製)を用い、所定の方法により行った。評価終了後のインサート上部の試験溶液をサンプルとし、含有LDHにより産生されるホルマザン量(490nmの吸光強度から定量)からLDH活性を算出した。なお、細胞生存率算出に当たり、Caco-2細胞膜透過性評価(試験例1)のサンプルとして1.0%Triton X-100のHBSS溶液を用いた際の、評価終了後のインサート上部の試験溶液でのLDH活性を生存率0%に相当する値とした。そして、下記式により、LDH活性の変化倍率を算出した。(算出式)細胞生存率(%)=(1.0%Triton X HBSS溶液 評価終了後のインサート上部の試験溶液でのLDH活性-各サンプル 評価終了後のインサート上部の試験溶液のLDH活性)÷(1.0%Triton X HBSS溶液 評価終了後のインサート上部の試験溶液でのLDH活性)
 算出式により求まる細胞生存率の結果の平均値及び標準誤差(N=3―4)を表1に示す。
[マウス経口投与時のノビレチン血中移行性評価]
(動物)
 C57BL/6Jマウス(雄性、6週齢)を日本クレア(株)より購入した。1週間以上の環境馴化を行った後、試験に供した。試験群一群当たりの動物数は、本試験系で統計的優位を得るのに最小で必要と考えられた8匹とした。(試験溶液の調製)本試験では、ノビレチン原体、ノビレチン-メチルヘスペリジン混合物(比較例5組成物)、ノビレチン-メチルヘスペリジン(実施例1組成物)を投与サンプルとした。ノビレチン原体および比較例5組成物、実施例1組成物を用いて、いずれもノビレチン濃度20mg/mLの水溶液もしくは水懸濁液を調製した。
(経口投与及び血漿の調製)
 固形食(CE-s)の自由摂食下にて1週間当該施設で飼育したマウスを用い試験を行った。マウスを16時間絶食させ、イソフルラン麻酔下で初期採血を眼下静脈叢より行った(ヘパリン処理ヘマトクリット微量採血管、VITREX製)後、マウス1個体あたり200mg/kg体重の投与量で、各サンプル溶液をゾンデを用いて胃内投与した。溶液投与後、10、30、60、90、120分後にイソフルラン麻酔下で眼下静脈叢より採血を行った。
 採血した血液を、12000rpmで10分間遠心分離し、エッペンドルフチューブに血漿を30μLずつ回収した。得られた血漿は、ノビレチン含有量測定まで-80℃で保管した。血漿中のノビレチン濃度はLC-MS/MSにより測定した。
(血漿サンプル処理)
 回収した血漿30μLにアセトニトリル30μLを加え、ボルテックスミキサーで5秒間攪拌後、室温下12000rpmで15分間遠心分離した。得られた上清30μLをLC測定用バイアルに回収後、さらにアセトニトリルを30μL添加、ボルテックスミキサーで混和した後にLC-MS/MSでノビレチン濃度を測定した。
[マウス経口投与時のノビレチン組織移行性評価]
(動物)
 C57BL/6Jマウス(雄性、6週齢)を日本クレア(株)より購入した。1週間以上の環境馴化を行った後、試験に供した。試験群一群当たりの動物数は、本試験系で統計的優位を得るのに最小で必要と考えられた5匹以上とした。
(試験溶液の調製)
 本試験では、ノビレチン原体、ノビレチン-メチルヘスペリジン混合物(比較例5組成物)、ノビレチン-メチルヘスペリジン(実施例1組成物)の3点を投与サンプルとした。ノビレチン原体および比較例5組成物、実施例1組成物を用いて、いずれもノビレチン濃度15mg/mLの水溶液もしくは水懸濁液を調製した。
(経口投与及び血漿の調製)
 固形食(CE-s)の自由摂食下にて1週間当該施設で飼育したマウスを用い試験を行った。マウスを16時間絶食させ、イソフルラン麻酔下で、マウス1個体あたり50mg/kg体重の投与量で、各サンプル溶液をゾンデを用いて胃内投与した。溶液投与後、30分後にイソフルラン麻酔下で腹部大静脈からの全採血を行い、安楽死させた。なお採血には、ヘパリン処理ヘマトクリット微量採血管、VITREX製を使用した。
 その後、全組織灌流下にて脱血し、各組織(肝臓、白色脂肪組織、褐色脂肪組織、腓腹筋、ヒラメ筋、脛骨、大腿骨、左脳、右脳各部位(大脳皮質、海馬))を採取した。また、採血した血液を、12000rpmで10分間遠心分離し、エッペンドルフチューブに血漿を30μLずつ回収した。得られた血漿および各組織サンプルは、ノビレチン含有量測定まで-80℃で保管した。
(骨組織以外の組織サンプル処理)
 回収した組織10mgにアセトニトリル300μLを加え、ホモジナイザー(ヒスコトロンNS-310E(株式会社マイクロテック・ニチオン製))で3分間攪拌後、4℃、15000rpmで20分間遠心分離した。得られた上清50μLをLC測定用バイアルに回収し、LC-MS/MSでノビレチン濃度を測定した。
(骨組織サンプル処理)
 回収した骨組織10mgを解剖ばさみで切断、室温下10000rpmで1分間遠心分離し脱血した。本サンプルを液体窒素を用いて凍結後、破砕機(クライオプレスCP-100W(株式会社マイクロテック・ニチオン製))にて破砕した後に、アセトニトリル300μLを加え、超音波処理(超音波洗浄機FU-9H(東京硝子器械株式会社製))を15分行った。得られた上清50μLをLC測定用バイアルに回収し、LC-MS/MSでノビレチン濃度を測定した。
実施例1
 ノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合し、その混合物を2軸のエクストルーダー(HAAKE社製)を用いて、加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/minの条件で処理し、25℃の冷風を吹き付けて5分で25℃まで冷却を行って(冷却速度:0.52℃/s)、固体分散体を調製した。このとき、押出機の内部での閉塞はみられず、固体分散体の成形体の排出は良好であった。本固体分散体の粉末X線回折を図1に示す。ノビレチン由来の回折ピークが消失していることから、本固体分散体中でノビレチンが非晶質化していることが確認され、本固体分散体の結晶化度は0%であると見積もられた。メチルヘスペリジンはX線回折でピークを与えないことから、ノビレチンの結晶化度は0%であると見積もられる。
 前記〔経時的な溶解性の評価〕により固体分散体のノビレチンの溶解濃度を測定した結果、攪拌開始6時間後までの曲線下面積(横軸:時間(単位=分)、縦軸:溶解濃度(単位=ppm))は2.20×10ppm・minであり、極めて高い溶解濃度を長時間維持することが示された(図6及び表1-1)。
 また、前記〔Caco-2細胞膜透過性評価〕による、本固体分散体試験時のインサート上部評価開始時ノビレチン濃度は132μM、インサート下部評価終了時ノビレチン濃度4.33μMであり、高い溶解性によるノビレチン膜透過量の向上が確認された(表1-1)。
実施例2
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ10質量%、90質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図2)から、本固体分散体中でノビレチンが非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例3
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ15質量%、85質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図3)から、本固体分散体中でノビレチンが非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例4
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ20質量%、80質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図4)から、本固体分散体中でノビレチンが非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例5
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ33質量%、67質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図5)から、本固体分散体中でノビレチンが非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例6
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ40質量%、60質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図6)から、本固体分散体中でノビレチンが非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例7
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製、ノビレチン60%含有)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図7)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例8
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ10質量%、90質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図8)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例9
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ33質量%、67質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図9)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例10
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ40質量%、60質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図10)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例11
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ60質量%、40質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図11)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例12
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ67質量%、33質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図12)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例13
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、モノグルコシルヘスペリジン(以下mGHes、東洋製糖株式会社製品αGヘスペリジン PA-Tを用いて、前記方法によりHPLCを用いた精製品)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図13)から、本固体分散体中でPMF90に含有される結晶性物質が非晶質化されていることが確認され、本固体分散体の結晶化度は0%であると見積もられ、同様にノビレチンの結晶化度も0%であると見積もられる。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を表1-1に示す。
実施例14
 難水溶性物質としてPMF90(株式会社沖縄リサーチセンター製)及び、mGHesをそれぞれ33質量%、67質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。

比較例1
 難水溶性物質としてエラグ酸二水和物(東京化成工業株式会社)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。
 実施例1と同様に、粉末X線回折測定の結果(図17)から、本固体分散体中でエラグ酸が非晶質化されていることが確認された。前記〔経時的な溶解性の評価〕、及び〔Caco-2細胞膜透過性評価〕の結果を図6及び表1-2に示す。
比較例2
 難水溶性物質としてエラグ酸二水和物(東京化成工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:180℃、加熱時間:10分、スクリューの回転数:80r/min)。実施例1と同様に、粉末X線回折測定の結果(図18)から、本固体分散体中でエラグ酸が非晶質化されていることが確認された。前記〔経時的な溶解性の評価〕及び〔Caco-2細胞膜透過性評価〕の結果を図6及び表1-2に示す。
比較例3
 難水溶性物質としてセサミン(CHROMADEX INC.製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。実施例1と同様に、粉末X線回折測定の結果(図19)から、本固体分散体中でセサミンが非晶質化されていることが確認された。前記〔経時的な溶解性の評価〕及び〔Caco-2細胞膜透過性評価〕の結果を図6及び表1-2に示す。
比較例4
 難水溶性物質としてヘスペリジン(東京化成工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを、実施例1と同様にして固体分散体を調製した(加熱温度:160℃、加熱時間:10分、スクリューの回転数:80r/min)。実施例1と同様に、粉末X線回折測定の結果(図20)から、本固体分散体中でヘスペリジンが非晶質化されていることが確認された。前記〔経時的な溶解性の評価〕及び〔Caco-2細胞膜透過性評価〕の結果を図6及び表1-2に示す。
 比較例5
 難水溶性物質としてノビレチン(和光純薬工業株式会社製)及び、メチルヘスペリジン(東京化成工業株式会社製)をそれぞれ25質量%、75質量%の割合で混合したものを評価した。粉末X線回折測定の結果(図21)から、本混合物中ではノビレチンが結晶として存在することを確認した。前記〔経時的な溶解性の評価〕及び〔Caco-2細胞膜透過性評価〕の結果を表1-2に示す。
比較例6

[マウス経口投与時のノビレチン血中移行性]
 ノビレチン、比較例5記載の組成物および実施例1記載の組成物を用いた、マウス経口投与時のノビレチン血中移行性について、投与後180分までのノビレチン血中濃度推移を図7に、血中濃度推移曲線における最高血中濃度(Cmax)と曲線下部面積(AUC)を表2に示した。ノビレチン原体ではCmax=3.7μg/mL(投与後60分)、AUC=323μg/(mL・min)、比較例5記載の組成物ではCmax=11.1μg/mL(投与後30分)、AUC=956μg/(mL・min)であるのに対し、固体分散体である実施例1記載の組成物では、Cmax=36.4μg/mL(投与後30分)、AUC=3550μg/(mL・min)と大幅に血中移行量が増加した。
[マウス経口投与時のノビレチン組織移行性]
 ノビレチン、比較例5記載の組成物および実施例1記載の組成物を用いた、マウス経口投与時のノビレチン組織移行性について、投与後30分での各組織におけるノビレチン移行量を表3に示した。ノビレチン原体、比較例5記載の組成物に対し、固体分散体である実施例1記載の組成物では、検討した組織に対する移行量が大幅に増加した。

 以上より、ノビレチン含有固体分散体におけるノビレチンの経時的溶解性は、例えば実施例1の如く2.20×10ppm・minと極めて高く、高い溶解濃度が長時間維持されていた(表1-1及び図23)。
 斯かる高い溶解性は、ノビレチンと同様に難溶性のポリフェノールとして知られているヘスペリジン、セサミン又はエラグ酸をそれぞれメチルヘスペリジンと混合して製造される固体分散体と比較して、固体分散体中及び水溶液中におけるノビレチン-メチルヘスペリジンの特異的な分子間相互作用が存在するためと推察される。
 また、ノビレチン含有固体分散体は、ヒト小腸上皮細胞由来の細胞膜に対する透過性も極めて優れており、上記他の難溶性のポリフェノールの固体分散体と比較して、水溶性ヘスペリジン誘導体のノビレチン可溶化能が著しく高いこと、さらにノビレチンの過飽和現象が高濃度かつ長時間発現しているためと推察される。Biopharmaceutical Classification System(BCS)においてClass 4に分類されるような各種化合物では、高溶解性を実現したとしても細胞膜透過性・経口吸収性は向上しない。また、記載全ての実施例・比較例における細胞膜透過性評価時のインサート上部添加液は、同じ方法により調製したものであることから、ノビレチンの高溶解こそが本固体分散体の極めて優れたノビレチンの細胞膜透過性を実現していると考えられ、更に動物を用いた経口投与実験での高い血中移行性、組織移行性を実現していると考えられる。
 したがって、本発明のノビレチン含有固体分散体を用いることにより、ノビレチンの経口吸収性を高めることが期待される。

Claims (11)

  1.  ノビレチンを含有する固体分散体の製造方法であって、ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程と、溶融した溶融物を冷却し、固化させる工程を含む、製造方法。
  2.  加熱温度が95~200℃、好ましくは100~185℃である請求項1記載の固体分散体の製造方法。
  3.  加熱温度が95~180℃、好ましくは100~160℃である請求項1記載の固体分散体の製造方法。
  4.  ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合する際の水溶性ヘスペリジン誘導体に対するノビレチンの質量比[ノビレチン/水溶性ヘスペリジン誘導体]が0.01~0.67である請求項1~3のいずれか1項記載の固体分散体の製造方法。
  5.  スクリューを備えた押出機を用いて、ノビレチン又はノビレチン含有物と水溶性ヘスペリジン誘導体を混合した後、加熱して溶融させる工程が行われる請求項1~4のいずれか1項記載の固体分散体の製造方法。
  6.  水溶性ヘスペリジン誘導体がメチルヘスペリジンである請求項1~5のいずれか1項記載の固体分散体の製造方法。
  7.  ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を含有し、ノビレチンがX線回折スペクトルから算出される結晶化度10%以下の状態で分散してなるノビレチン含有固体分散体。
  8.  ノビレチン又はノビレチン含有物と、水溶性ヘスペリジン誘導体を含有し、ノビレチンが分散してなる、X線回折スペクトルから算出される結晶化度10%以下のノビレチン含有固体分散体。
  9.  水溶性ヘスペリジン誘導体がメチルヘスペリジンである請求項7又は8記載のノビレチン含有固体分散体。
  10.  請求項1~6のいずれか1項記載の製造方法により得られ、X線回折スペクトルから算出されるノビレチンの結晶化度が10%以下であるノビレチン含有固体分散体。
  11.  請求項7~10のいずれか1項記載のノビレチン含有固体分散体を含有する飲食品、医薬品又は医薬部外品。
