Abstract
본 발명은 인삼의 사포닌으로부터, 원래 인삼 내에 미량으로 존재하는 컴파운드 K 및 컴파운드 Y를 선택적으로 제조하는 방법에 관한 것으로서, 보다 상세하게는 인삼으로부터 얻은 사포닌에 특정 효소를 처리하여, 상기 사포닌을 구조 전환시킴으로써 원하는 목표 화합물, 즉 컴파운드 K 및 컴파운드 Y를 고수율로 수득할 수 있는 방법에 관한 것이다.
Description
본 발명은 인삼의 사포닌으로부터, 원래 인삼 내에 미량으로 존재하는 컴파운드 K 및 컴파운드 Y를 선택적으로 제조하는 방법에 관한 것으로서, 보다 상세하게는 인삼으로부터 얻은 사포닌에 특정 효소를 처리하여, 상기 사포닌을 구조 전환시킴으로써 원하는 목표 화합물, 즉 컴파운드 K 및 컴파운드 Y를 고수율로 수득할 수 있는 방법에 관한 것이다.
인삼 사포닌은 다른 식물에서 발견되는 사포닌과는 다른 특이한 화학구조를 가지고 있으며, 이로 인하여 약리효능도 특이하므로, 인삼(ginseng) 배당체(glycoside)란 의미로 “진세노사이드(ginsenoside)”라고도 불린다. 인삼 사포닌의 구체적인 종류로는 파낙사디올계인 진세노사이드 Rb1, Rb2, Rc, Rd, 컴파운드 K, 컴파운드 Mc, 컴파운드 O 등, 및 파낙사트리올계인 진세노사이드 Re, Rf, Rg1, Rg3, Rg5, Rh1, Rh2 등이 있으며, 이들 인삼 사포닌은 각각 다른 효능을 나타낸다.

상기 반응식 1에 나타낸 바와 같이, 특히 파낙사디올계 사포닌인 진세노사이드 Rb1, Rb2, Rc 등은 미생물 대사에 의해 다른 인삼 사포닌으로 전환될 수 있으므로, 인삼 사포닌으로부터 다른 종류의 특정 인삼 사포닌으로 전환시키는 방법으로서 효소를 이용하는 방법이 이전부터 사용되어 왔다.
그러나, 종래 효소를 이용한 전환 반응은 기질에 대한 효소의 비특이성이 커서 사용되는 사포닌 기질 대비 매우 많은 양의 효소를 사용하여야 하였으며, 또한 생성되는 원하는 인삼 사포닌들의 수율이 매우 낮았다.
종래의 인삼 사포닌을 얻는 방법들은 원하는 특정 인삼 사포닌만으로 전환시키는 것이 아닌, 전환된 다양한 인삼 사포닌을 추출 등을 통해 수득한 다음, 이를 정제하여 원하는 인삼 사포닌만을 분리하는 것을 기술적 해결책으로 하고 있다.
그러나, 이와 같은 종래 방법들은 순수한 특정 인삼 사포닌을 획득하기 위해 정제에 따른 추가적인 비용, 시간 등이 소요되기 때문에, 인삼 사포닌의 판매가가 상승할 수 밖에 없고, 그에 따라 관련 제품에 인삼 사포닌을 다량으로 적용하기에는 한계점이 있다.
특히, 컴파운드 K의 경우에는 인삼 사포닌으로부터 전환되는 화합물의 경로에 있어서 마지막에 생성되는 화합물에 해당하므로, 중간 대사체들을 모두 반응시킬 수 있는 다양한 효소가 필요하며, 또한 반응 중간에 생성되는 대사산물에 의해 대사체와 효소간의 반응성이 떨어지게 된다. 뿐만 아니라, 중간 대사체간의 반응액 내의 뭉침 현상으로 인해 수율이 낮아지는 문제가 발생한다.

종래의 방법으로 특정 인삼 사포닌을 얻기 위해서는 비용이 많이 들 뿐만 아니라, 원하는 인삼 사포닌을 대량으로 입수하기가 어려웠다. 따라서, 목표 인삼 사포닌을 대량 생산할 수 있으면서 비용을 절감할 수 있는 제조 방법에 대한 필요성이 존재하였다.
그러므로, 본 발명은 원하는 특정 인삼 사포닌을 고수율로 획득할 수 있으면서 실시가 용이한 인삼 사포닌 전환 방법을 제공하는 것을 목적으로 한다.
상기한 목적을 달성하기 위하여, 본 발명은 아스퍼질러스 속에서 분리한 펙티나제, 나린지나제, 셀룰라제 및 헤미셀룰라제로 이루어진 군에서 선택되는 1종 이상; 및 트리코데르마 속에서 분리한 펙티나제 및 셀룰라제로 이루어진 군에서 선택되는 1종 이상을 이용하여 인삼의 사포닌을 전환시켜서 컴파운드 K 및 컴파운드 Y를 고수율로 제조하는 방법을 제공한다.
본 발명에 따른 인삼 사포닌으로의 전환 방법을 이용함으로써 원하는 인삼 사포닌을 고수율로 용이하게 획득할 수 있다.
도 1은 인삼 사포닌으로의 전환 반응 후 생성된 컴파운드 K 및 컴파운드 Y를 실리카겔 칼럼크로마토그래피에 의해 확인한 결과를 나타낸다.
본 발명은 효소적 방법에 의해 인삼의 사포닌으로부터 컴파운드 K(화학식 1) 및 컴파운드 Y(화학식 2)를 선택적으로 제조하는 방법에 관한 것이다.
본 발명의 방법에서는 미생물 유래의 효소를 사용함으로써, 인삼의 사포닌, 구체적으로는 인삼의 파낙사디올계 사포닌, 보다 구체적으로는 진세노사이드 Rb1, Rb2, Rc, Rd 또는 이들의 혼합물로부터 원하는 인삼 사포닌으로의 전환이 효율적으로 일어나게 하여 고수율로 컴파운드 K 및 컴파운드 Y를 획득할 수 있다.
구체적으로, 본원발명에서 사용되는 효소는 바람직하게는 아스퍼질러스 속의 미생물 및 트리코데르마(Trichoderma) 속의 미생물로부터 얻은 것이며, 바람직하게 아스퍼질러스 나이저(Aspergillus
niger), 아스퍼질러스 아큘레아투스(Aspergillus
aculeatus), 아스퍼질러스 루츄엔시스(Aspergillus
luchuensis) 및 아스퍼질러스 오리자에(Aspergillus
oryzae)로 이루어진 군에서 선택되는 1종 이상의 아스퍼질러스 속의 미생물, 및 트리코데르마(Trichoderma) 속의 미생물은 트리코데르마 아그레시붐(Trichoderma aggressivum), 트리코데르마 하르지아눔(Trichoderma
harzianum), 트리코데르마 레세이(Trichoderma
reesei) 및 트리코데르마 비리데(richoderma viride)로 이루어진 군에서 선택되는 1종 이상의 트리코데르마 속의 미생물로부터 얻은 것이며, 가장 바람직하게는 아스퍼질러스 아큘레아투스 및 트리코데르마 레세이로부터 얻은 것이다.
또한, 본 발명에서 사용되는 효소는 상기한 미생물로부터 분리한 나린지나제, 헤미셀룰라제, 베타-글루카나제, 락타제, 셀룰라제, 베타-갈락토시다제, 펙티나제일 수 있으며, 바람직하게는 펙티나제, 나린지나제, 헤미셀룰라제 또는 셀룰라제이다.
대부분 동일한 작용을 하는 같은 종류의 효소라 하더라도, 효소가 유래한 미생물의 종에 따라서 구체적으로 효소가 기질 내에서 작용하는 부위가 상이하여 기질 특이성에 차이가 생긴다. 따라서, 본 발명에서는 가장 바람직하게는 아스퍼질러스 아큘레아투스로부터 얻은 펙티나제, 나린지나제, 셀룰라제 및 헤미셀룰라제로 이루어진 군에서 선택되는 1종 이상; 및 트리코데르마 레세이로부터 얻은 펙티나제, 셀룰라제, 또는 이들의 혼합물로 이루어진 군에서 선택되는 1종 이상을 동시에 사용하는 것이 좋다.
본 발명에서는 인삼의 사포닌을 용매에 0.01~20중량%의 양으로 용해시킨 후, 상기 효소를 사용하여 효소적 방법에 의해 사포닌을 원하는 다른 사포닌으로 전환시킨다. 이 때 사용되는 용매로는 효소의 활성을 저하시키지 않는 것을 사용하는 것이 바람직하며, 예를 들어 물 또는 완충용액과 같은 수성용매, 또는 물이나 완충용액과 같은 수성용매와 유기용매의 혼합액을 사용할 수 있다. 구체적으로 이 때 사용되는 완충용액은 아세트산, 시트레이트, 인산 또는 시트레이트-인산 등일 수 있고, 유기용매는 아세토니트릴, 디록산, 디메틸 설폭사이드, 메탄올, 에탄올, 1-프로판올, 2-프로판올 등이 있다. 바람직하게, 사용되는 용매는 pH 범위가 2.5~7.5, 바람직하게는 3~6, 더 바람직하게는 3.5~5.5이다.
본 발명의 방법에서, 사용되는 효소의 전체 양은 사용되는 기질의 양에 대하여 1~500중량%의 양으로 첨가됨이 바람직하며, 더 바람직하게는 10~400중량%, 보다 더 바람직하게는 10~200중량%의 양으로 첨가된다.
반응 온도는 효소의 불활성화가 일어나지 않는 온도 조건이어야 하는데, 바람직하게는 30~60℃, 더 바람직하게는 35~60℃, 보다 더 바람직하게는 40~55℃의 범위로 온도를 유지하는 것이 좋다.
또한, 반응 시간 역시 효소의 활성이 유지되는 기간이라면 특별히 제한을 받지 않으나, 1~120시간, 바람직하게는 1~96시간, 더 바람직하게는 24~96시간, 보다 더 바람직하게는 24~72시간 동안 교반하면서 반응시키는 것이 바람직하다.
효소 반응은, 2가지 효소를 기질에 동시에 첨가하거나, 하나의 효소를 먼저 반응시킨 다음 나머지 효소를 이후에 투입하는 순차적 방식으로 수행할 수 있다.
이후 비등 수욕조에서의 가열과 같은 공지된 방법을 사용하여, 효소를 불활성화시킴으로써 컴파운드 K 및 컴파운드 Y가 다량 함유된 반응액을 얻을 수 있다.
이하 본 발명을 실시예에 의하여 보다 상세히 설명한다. 다만, 다음 실시예로 본 발명의 범주가 한정되는 것은 아니다.
[참고예 1] 인삼 정제 사포닌의 제조
홍삼, 백삼, 수삼, 미삼 또는 이들의 인삼엽, 인삼꽃, 인삼열매 2㎏에 에탄올 20ℓ를 넣고, 3회 환류 추출한 후, 15℃에서 6일간 침적시켰다. 그 후, 여과포 여과와 원심분리를 통해 잔사와 여액을 분리하고, 분리된 여액을 감압농축하여 얻은 엑기스를 물에 현탁한 후에, 에테르 1ℓ로 5회 추출하여 색소를 제거하고, 수층을 1-부탄올 1ℓ로 3회 추출하였다. 이로부터 얻은 총 1-부탄올층을 5% KOH로 처리한 다음 증류수로 세척한 뒤, 감압농축하여 1-부탄올 엑기스를 얻고, 이를 소량의 메탄올에 녹인 다음, 대량의 에틸아세테이트에 추가하여, 생성된 침전물을 건조함으로써, 인삼 정제 사포닌(진세노사이드 Rb1, Rb2, Rc, Rd, Re, Rg1, Rf 등을 포함함) 40~80g을 얻었다.
[실시예 1] 효소반응 통한 컴파운드 K 및 컴파운드 Y의 제조
상기 참고예 1의 인삼 정제 사포닌(진세노사이드 Rb1, Rb2, Rc, Rd, Re, Rg1, Rf 등을 포함함) 10g을 물 1ℓ에 넣고 용해하였다.
그런 다음 상기 혼합액에 아스퍼질러스 아큘레아투스에서 분리한 펙티나제와 트리코데르마 레세이에서 분리한 펙티나제를 동시에 첨가하였으며, 이 때 상기 효소는 각각 기질대비 100중량%씩 첨가하였고, 30℃에서 72시간을 반응시켰다. 박층크로마토그래피에 의해 주기적으로 확인하여 기질이 완전히 소실되면 비등 수욕조에서 10분간 가열하여 효소를 불활성시켜 반응을 종료시켰다. 마지막으로 반응액에 에틸아세테이트를 1:1의 비율(반응액에 대한 부피비)로 넣고 3회 추출한 후 농축한 다음 실리카겔 칼럼크로마토그래피(클로로포름:메탄올=9:1)에 의해 컴파운드 K 및 컴파운드 Y를 분리하였다(도 1).
인삼사포닌 10g에는 진세노사이드 Rb1 2.66g과 진세노사이드 Rb2 0.73g, 진세노사이드 Rc 1.23g, 진세노사이드 Rd 0.38g이 존재한다. 컴파운드 K는 진세노사이드 Rb1과 진세노사이드 Rc, 진세노사이드 Rd로부터 95% 이상의 수득율로 전환하였고, 컴파운드 Y는 진세노사이드 Rb2로부터 95% 이상의 수득율로 전환하였다.

- 제1항에 있어서, 상기 방법은1) 기질인 인삼의 사포닌을 수성용매 또는 수성용매와 유기용매의 혼합액 중에 용해시켜 효소로서 아스퍼질러스 속에서 분리한 펙티나제, 나린지나제, 셀룰라제 및 헤미셀룰라제로 이루어진 군에서 선택되는 1종 이상; 및 트리코데르마 속에서 분리한 펙티나제 및 셀룰라제로 이루어진 군에서 선택되는 1종 이상을 첨가하여 가온 상태의 수욕조상에서 교반하면서 반응시키는 단계;2) 상기 기질이 완전히 소실되면 효소를 불활성화시켜 반응을 종료시키는 단계;3) 반응액에 에틸아세테이트를 첨가하여 추출한 후 농축시켜 컴파운드 K 및 컴파운드 Y를 분리하는 단계;를 포함하는 방법.
- 제2항에 있어서, 상기 인삼의 사포닌은 진세노사이드 Rb1, Rb2, Rc, Rd 또는 이들의 혼합물 중에서 선택되는 1종 이상임을 특징으로 하는 방법.
- 제2항에 있어서, 상기 아스퍼질러스 속의 미생물은 아스퍼질러스 나이저(Aspergillus niger), 아스퍼질러스 아큘레아투스(Aspergillus aculeatus), 아스퍼질러스 루츄엔시스(Aspergillus luchuensis) 및 아스퍼질러스 오리자에(Aspergillus oryzae)로 이루어진 군에서 선택되는 1종 이상이고,상기 트리코데르마 속의 미생물은 트리코데르마 아그레시붐(Trichoderma aggressivum), 트리코데르마 하르지아눔(Trichoderma harzianum), 트리코데르마 레세이(Trichoderma reesei) 및 트리코데르마 비리데(richoderma viride)로 이루어진 군에서 선택되는 1종 이상임을 특징으로 하는 방법.
- 제2항에 있어서, 상기 아스퍼질러스 속에서 분리한 펙티나제, 나린지나제, 셀룰라제 및 헤미셀룰라제로 이루어진 군에서 선택되는 1종 이상; 및 트리코데르마 속에서 분리한 펙티나제 및 셀룰라제로 이루어진 군에서 선택되는 1종 이상은 동시에 또는 순차적으로 첨가됨을 특징으로 하는 방법.
- 제2항에 있어서, 상기 효소 전체의 양은 기질의 양에 대하여 10~400중량%에 해당함을 특징으로 하는 방법.
- 제2항에 있어서, 상기 효소의 반응은 35~60℃의 온도에서 일어나는 것을 특징으로 하는 방법.
- 제2항에 있어서, 상기 효소의 반응은 24~96시간 동안 반응하는 것을 특징으로 하는 방법.
- 제2항에 있어서, 상기 용매는 pH가 3~5.5의 범위임을 특징으로 하는 방법.
