Abstract
Description
このような貼付製剤には、皮膚に貼付する際には充分な粘着性を示し、使用後には皮膚表面を汚染する(例えば、糊残りやベタツキなどを生じる)ことなく剥離除去できることが求められている。また、貼付製剤は、皮膚に対して低刺激性であることが望ましい。
[1]支持体と、支持体の片面に形成された粘着剤層を含む貼付製剤であって、当該粘着剤層が下記の(A)成分~(C)成分を含有するビソプロロール含有貼付製剤。
(A)成分:水酸基含有単量体と(メタ)アクリル酸アルキルエステル単量体とを含む単量体成分を共重合してなるポリマー;
(B)成分:メチルメタクリレート単量体とブチルメタクリレート単量体とを含む単量体成分を共重合してなるポリマー;および
(C)成分:ビソプロロール
[2]前記粘着剤層が、有機液状成分をさらに含む、上記[1]に記載のビソプロロール含有貼付製剤。
[3]前記粘着剤層に含まれる(A)成分と(B)成分との含有量比((A)成分:(B)成分)が重量比で1:0.05~1:1である上記[1]または[2]に記載のビソプロロール含有貼付製剤。
本発明の一態様のビソプロロール含有貼付製剤は、経皮吸収型製剤として提供されるものであり、具体的には、マトリックス型貼付製剤、リザーバー型貼付製剤等として提供される。
上記多孔質無機粒子としては、例えば、多孔質シリカ粒子、多孔質アルミナ粒子、多孔質チタニア粒子などが挙げられる。なかでも、製造工程や分散液中においても粒子が壊れにくいことから多孔質シリカ粒子が好ましく用いられ得る。なお、多孔質無機粒子としては、1種のみを用いてもよく、2種以上を組み合わせて用いてもよい。
上記下塗り溶液を支持体表面に塗布し、乾燥および/または硬化させることによって、下塗り層を形成することができる。
水酸基含有単量体としては、中でも、N-(2-ヒドロキシエチル)アクリルアミド、N-(2-ヒドロキシエチル)メタクリルアミドが好ましく、N-(2-ヒドロキシエチルアクリル)アミドが最も好ましい。
水酸基含有単量体は一種または二種以上を使用できる。
ポリマー(A1)の調製
冷却管、窒素ガス導入管、温度計、滴下漏斗および攪拌機を備えた反応容器に、アクリル酸2-エチルヘキシル(以下「2-EHA」と表すことがある)70重量部、N-(2-ヒドロキシエチル)アクリルアミド(以下「HEAA」と表すことがある)5重量部、N-ビニル-2-ピロリドン(以下「N-VP」と表すことがある)25重量部、および溶媒として酢酸エチル333.3重量部を入れ、室温において窒素ガスバブリング(100mL/分)を行いながら1時間攪拌した。その後、反応容器の内容物を加熱し、60℃に達した時点で、重合開始剤として2,2’-アゾビスイソブチロニトリル(AIBN)0.2部を加えた。内容物の温度を60℃に保つよう制御し、窒素ガス流中で6時間重合を行い、次いで76℃に15時間保持した。上記方式の溶液重合により、アクリル系共重合体(2-EHA/HEAA/N-VP=70/5/25(重量比)、Mw:2,200,000)の溶液である粘着剤組成物(実質的には、粘着剤層を形成するための塗工液、実施例2~3および比較例1~3において同様)を得た。
2-EHAを55重量部、HEAAを5重量部、N-VPを40重量部とした以外は、ポリマー(A1)と同様にして調製した。Mwは2,000,000であった。
(B)成分として、メチルメタクリレート単量体とブチルメタクリレート単量体との含有量比が重量比で1:3であるコポリマー(ローム・ファルマ社製Plastoid B(商品名)、重量平均分子量150,000)(ポリマー(B1))を用いた。
有機液状成分として、ミリスチン酸イソプロピル(IPM)、アジピン酸ジイソプロピル(DIPA)、オレイルアルコール(OA)を用いた。
(C)成分として、ビソプロロールのフリー体を用いた。
(A)成分としてポリマー(A1)55重量部(固形分の量として)、(B)成分としてポリマー(B1)10重量部、(C)成分としてビソプロロールのフリー体5重量部、有機液状成分としてミリスチン酸イソプロピル30重量部を混合し、濃度調整用の酢酸エチルを適量加えて撹拌し、均一な粘着剤溶液を得た。得られた粘着剤溶液を厚み75μmのポリエステル製剥離シートの剥離処理面上に、乾燥後の厚みが45μmとなるよう塗布、80℃、5分で乾燥して粘着剤層を形成した。次いで、上記粘着剤層に、支持体として厚さ4μmのポリエステルフィルムと目付量12g/m2のポリエステル製不織布との積層体を用い、その不織布面を圧着して貼り合わせ、本発明の一態様のビソプロロール含有貼付製剤を得た。
表1に示す組成で、実施例1と同様の手順で、実施例2、3、比較例1に係るビソプロロール含有貼付製剤を調製した。
(A)成分としてポリマー(A2)52重量部(固形分の量として)、(B)成分としてポリマー(B1)5重量部、(C)成分としてビソプロロール13重量部、有機液状成分としてミリスチン酸イソプロピル30重量部を混合し、濃度調整用の酢酸エチルを適量加えて撹拌し、均一な粘着剤溶液を得た。得られた粘着剤溶液を厚み75μmのポリエステル製剥離シートの剥離処理面上に、乾燥後の厚みが17μmとなるよう塗布、80℃、5分乾燥して粘着剤層を形成した。
一方、エチレン-ビニルアルコール共重合樹脂および多孔質シリカ粒子(富士シリシア製 サイリシア350)を1:1の配合比(重量基準)でn-プロパノールと水との混合溶媒(n-プロパノール/水=7/3(体積基準))中にそれぞれ、溶解および分散して、下塗り剤を得た。得られた下塗り剤を支持体として厚さ15μmのエチレン-ビニルアルコール共重合体フィルムの片面に乾燥後の塗膜(下塗り層)の目付量が0.7g/m2となるように塗布および乾燥して下塗り層を形成し、これにより、支持体を得た。
上記粘着剤層に、上記支持体の下塗り層表面を圧着して貼り合わせ、本発明の一態様のビソプロロール含有貼付製剤を得た。
表1に示す組成で、実施例4と同様の手順で、実施例5~13、比較例2~5に係るビソプロロール含有貼付製剤を調製した。
貼付製剤の剥離ライナーを剥離後、粘着剤層の露出面を指で触った際の粘着剤層の凝集性を次の指標で目視評価した。
○:糸曳きはなく、指に糊残りしなかった。
×:糸曳きがひどく、指に糊残りした。
各貼付製剤を幅12mm×長さ40mm以上に裁断したものを試験片に用いた。試験片の剥離ライナーを除去し、JISZ0237:2009に準拠して、試験板(ベークライト板)に2kgのゴムローラーを一往復して圧着し、23℃雰囲気下で30分間放置した。その後、引張試験機に取り付け、試験片の導きを180度に折り返して、毎分300mmの速度で引き剥がした。ほぼ等間隔に3点の荷重を読み取り、平均値を粘着力(N/12mm幅)とした。また、試験後のベークライト板への糊残りの有無について目視評価し、凝集性の良し悪しを判断した。
各貼付製剤を幅10mm×長さ50mmに裁断したものを試験片に用いた。試験片の剥離ライナーを除去し、JISZ0237:2009に準じて、試験板(ベークライト板)の一端(幅10mm、長さ20mm)に重さ2kgのゴムローラーを一往復させて圧着し、逆の一端を補助紙で補強した。これを40±5℃の温度で安定した装置内のフックに取り付け30分間放置した。その後、100gの荷重をかけ、落下するまでの時間を保持力(分)とした。試験数はn=3で行い、計3回の測定値を平均した。
実施例5、9および比較例3、5にて得られた貼付製剤について、直径8mmの略円形に切断し、剥離ライナーを剥がした後、直径20mmの略円形に打ち抜いたヘアレスマウス摘出皮膚の角質面に貼り付けた。
ヘアレスマウス摘出皮膚を皮膚透過実験用セル(適用面積:6mmφ)に角質層側がドナー相、真皮側がレセプター相になるように装着し、レセプター相には、生理食塩水を入れ、32℃下で12時間皮膚透過実験を行った。24時間後におけるレセプター液を採取し、その中の薬物濃度を高速液体クロマトグラフィー(HPLC)にて測定した。試験数はn=3で行い、計3回の測定値を平均した。

これに対し、比較例2~5では凝集性が×となり、凝集破壊が生じることが判明し、実施例よりも指に糊残りがしやすいと評価された。比較例3、5では放出試験において良好な結果が得られたが、粘着特性において凝集破壊が生じ、貼付製剤としては使用に適さないものであった。

Claims (3)
- 支持体と、支持体の片面に形成された粘着剤層を含む貼付製剤であって、当該粘着剤層が下記の(A)成分~(C)成分を含有する、ビソプロロール含有貼付製剤。
(A)成分:水酸基含有単量体と(メタ)アクリル酸アルキルエステル単量体とを含む単量体成分を共重合してなるポリマー
(B)成分:メチルメタクリレート単量体とブチルメタクリレート単量体とを含む単量体成分を共重合してなるポリマー
(C)成分:ビソプロロール - 前記粘着剤層が、有機液状成分をさらに含む、請求項1に記載のビソプロロール含有貼付製剤。
- 前記粘着剤層に含まれる(A)成分と(B)成分との含有量比((A)成分:(B)成分)が重量比で1:0.05~1:1である請求項1または2に記載のビソプロロール含有貼付製剤。
