Abstract
본 발명은 마키아신 (Maackiasin)을 유효성분으로 함유하는 피부 미백용 조성물에 관한 것으로, 더 상세하게는 마키아신이 멜라닌 생성과 관련된 효소들의 발현을 억제하는 효과를 이용한 마키아신을 유효성분으로 함유하는 피부 미백용 화장료 조성물에 관한 것이다. 본 발명의 화장료 조성물에 유효성분으로 함유되는 마키아신은 알부틴 (arbutin)과 비교해 봤을 때, 훨씬 적은 농도에서도 멜라닌 합성 저해효과가 더 큰 바, 피부 미백 효과가 뛰어난 화장료 조성물 및 기능성 화장품을 제조하는데 효과적이다.

【기술분야】
본 발명은 마키아신 (Maacki as in)의 미백제제로서의 용도에 관한 것으로, 더 상세하게는 마키아신이 멜라닌 생성과 관련된 인자들의 발현을 억제하는 효과를 이용한 마키아신의 미백제제로서의 용도 및 마키아신을 유효성분으로 함유하는 피부 미백용 화장료 조성물에 관한 것이다.
【배경기술】
사람의 피부, 모발, 눈 등에서 발현하는 색은 멜라닌, 카로틴 및 헤모글로빈에 의해 결정되며 특히 피부에 있어 색깔을 결정하는 가장 증요한 요인은 멜라닌으로 멜라닌의 양과 성질 그리고 분포의 정도에 따라 피부색이 결정된다.
멜라닌의 주요한 기능은 이러한 유해 라디칼을 제거함으로써, 유해 라디칼에 의한 손상으로부터 피부를 보호하는 것이다. 따라서 멜라닌이 많다는 것은 물리적, 화학적 독성물질로부터 피부를 보호하기 위한 효과적인 대웅체계를 가지고 있다는 것을 의미한다. 이러한 멜라닌의 생성을 촉진하는 요인들로서는 햇빛 (자외선)뿐 만 아니라 에스트로겐, 프로스타글라딘 등과 같은 호르몬이 있다.
멜과닌은 햇빛 (자외선)에 의해 멜라노사이트 (me l anocyt e)에서 티로신이 티로시나제라는 효소에 의해 도파와 도파크름으로 변환된 뒤 복잡한 산화와 축합 반응을 거쳐 생성된다. 생성된 멜라닌은 피부 세포에 전달되고 표피 박리와 함께 멜라닌이 상실되어 소멸되는 순환 작용을 한다. 멜라닌 생성 메카니즘의 특징은 티로시나제라는 단 한개의 효소가 관여하는 것이기 때문에 티로시나제 활성을
억제하여 멜라닌 생성을 막아줌으로서 미백 효과를 기대할 수 있다. 미백 화장품에 있어 멜라닌 생성을 억제하는 방법은 크게 다음과 같이 나눌 수 있다. 첫번째 방법은 멜라닌 생성의 주원인인 자외선올 차단하여 제거하는 것으로써, 이 방법은 화장품 조성물에 광 산란제 또는 광 차단제를 함유시킴으로써 효과적으로 멜라닌 생성을 억제할 수 있다. 두 번째 방법은 글루코사민과 같이 티로시나제가 활성을 나타내기 위해 필요한 코어 탄수화물의 합성을 저해함으로써 멜라닌 생성을 억제시키는 것이다. 세 번째 방법은 효소산 (코직산) 또는 알부틴과 같이 멜라닌 생성에 관여하는 티로시나제의 기능을 방해하는 것이다. 네 번째 방법은 하이드로퀴논과 같이 멜라닌을 생성하는 세포인 멜라노사이트에 대하여 특이적인 독성을 가지고 있어 세포 분열을 방해하는 것이다. 다섯 번째 방법은 비타민 C와 같이 티로신이 도파와 도파크름을 거쳐 복잡한 산화와 축합반응을 거치는 동안에 강한 항산화 효과를 발휘함으로써 미백효과를 기대하는 것이다.
최근들어 가장 이상적으로 여겨지는 미백 화장료는 일차적으로 자외선을 차단해주고, 체내에 생성되는 라디칼을 소거해주면서 자외선 등에 의해 발생할 수 있는 염증 유발 물질 생성을 억제해주며 궁극적으로는 멜라닌 생합성을 억제하는 효과를 갖는 물질들을 복합적으로 사용하는 것으로 알려져 있다.
그렇지만 천연 식물들은 많은 양을 사용하는 경우에는 오히려 피부에 자극을 주어 피부색소 침착을 야기하는 등의 부작용을 야기시킬 수 있으므로 낮은 농도에서도 미백 효과를 극대화 시켜 줄 수 있는 물질 및 방법의 개발이 요구되고 있다. 본 명세서 전체에 걸쳐 다수의 논문 및 특허문헌이 참조되고 그 인용이 표시되어 있다. 인용된 논문 및 특허문헌의 개시 내용은 그 전체로서 본 명세서에 참조로 삽입되어 본 발명이 속하는 기술 분야의 수준 및 본 발명의 내용이 보다 명확하게 설명된다.
【발명의 내용】
【기술적 과제】
본 발명자들은 다롭나무 am!rensis) 부^ 추출한 마키아신이 세포독성이 적으면서도 시중에서 미백원료로 많이 쓰이는 알부틴보다도 멜라닌 합성 저해율이 훨씬 높은 것을 발견하여 본 발명을 완성하였다.
따라서 본 발명의 목적은 피부 미백용 화장료 조성물 제조를 위한 하기 화학식 1로 표시되는 마키아신의 용도. 마키아신을 유효성분으로 함유하는 피부 미백용 화장료 조성물 및 상기 조성물을 포함하는 피부 미백용 기능성 화장품을 제공하는 것이다.

【기술적 해결방안】
상기 본 발명의 목적을 달성하기 위해, 본 발명은 피부 미백용 화장료 조성물 제조를 위한 상기 화학식 1로 표시되는 마키아신의 용도, 마키아신을
유효성분으로 함유하는 피부 미백용 화장료 조성물 및 상기 조성물을 포함하는 피부 미백용 기능성 화장품을 제공한다. ᅳ
또한, 본 발명의 다른 목적을 달성하기 위해, 본 발명은 피부 미백용 기능성 화장품의 제조를 위한 상기 피부 미백용 화장료 조성물의 용도를 제공한다.
또한, 본 발명의 또 다른 목적을 달성하기 위해, 본 발명은 마키아신을 유효성분으로 함유하는 화장료 조성물을 객체 (subject )에 투여하는 단계를 포함하는 피부 미백 방법을 제공한다. 이하, 본 발명을 상세히 설명한다. 본 발명은 피부 미백용 화장료 조성물 제조를 위한 상기 화학식 1로 표시되는 마키아신의 용도, 마키아신을 유효성분으로 함유하는 피부 미백용 화장료 조성물 및 상기 조성물을 포함하는 피부 미백용 기능성 화장품을 제공한다.
마키아신 (maackias in)은 하기 화학식 1과 같은 화학구조를 갖고 있고, 화학구조식은 7H-Pyr ano [ 2, 3-f ] - 1 , 4-benzod i ox i η-7-one , 2-(3 , 4-di hydroxypheny 1 ) - 3-(3 , 5-di hydroxypheny l )-2 , 3-dihydro-8-( 4-me t hoxypheny 1 ) - ( 9C I )로 표시할 수 있다.

마키아신의 미백 효과를 확인하기 위해 B16F1 멜라노마 세포를 이용하여 마키아신 처리 농도별 멜라닌 합성 저해 효과를 측정하였다. 측정 결과, 농도의존적으로 멜라닌의 합성이 저해되었다. 이때 알부틴 (ar but in)을 50O«g/ 처리한 것을 양성대조군으로 사용했다. 마키아신은 10 /m.e만 처리하였는데도 알부틴보다 멜라닌 합성비율이 매우 유의적으로 낮게 나왔다 (실험예 2 , 표 2 및 도 4 참조) . 양성대조군으로 사용된 알부틴은 월글나무열매 (bearberry)에서 추출한 글라이코실레이티드 하이드로퀴논으로서 티로시나제 (tyros inase)를 저해하여 멜라닌 색소의 형성을 방해한다. 현재 시중에서 피부 미백용 원료로 많이 쓰이고 있다.
마키아신이 멜라닌 합성에 관여하는 효소들인 Tyros inase (티로시나아제) , TRP( tyros inase rel ated protein)— 1 , TRP-2 및 이를 조절하는 전사인자인 MITF(micr ophthalmia assoc iated transcr ipt ion factor)의 발현을 억제하는지 여부를 웨스턴블랏팅 (western blott ing)을 통해 밝혔다. 상기 MITF는 티로시나아제 및 TRPs에 결합하여 이 효소들의 발현을 조절하는 것으로 잘 알려져 있기 때문에 멜라닌 합성을 조절하는 피부 미백 성분을 개발하기 위한 주요 타깃으로서 여겨지고 있다. 웨스턴 블랏팅 결과에서 마키아신을 10 /ΙΙΙ£ 처리한 실험군은 알부틴 500/zg/ 처리한 양성 대조군보다 단백질들의 발현이 억제된 것을 확인할 수 있다 (실험예 3 및 도 5 참조) .
따라서, 본 발명에 따른 상기 화학식 1의 마키아신은 멜라닌 합성을 효과적으로 저해함으로써 우수한 미백효과를 나타내므로 피부 미백용 조성물로 유용하게 사용될 수 있다.
본 발명에 따른 화장료 조성물에 포함되는 성분은 유효성분으로서의 마키아신 이외에 화장료 조성물에 통상적으로 이용되는 성분들을 포함하며, 예컨대 안정화제, 용해화제, 미타민, 안료 및 향료와 같은 통상적인 보조제, 그리고
담체를 포함한다.
본 발명의 피부 미백용 화장료 조성물은 당업계에서 통상적으로 제조되는 어떤 제형으로도 제조될 수 있으며 , 예를 들어, 용액, 현탁액, 유탁액 , 페이스트, 겔 , 크림, 로션, 파우더. 비누, 계면활성제 -함유 클린싱, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파운데이션 및 스프레이 등으로 제형화될 수 있으나, 이에 한정되는 것은 아니다. 보다 상세하게는, 유연 화장수, 영양 화장수, 영양 크림, 마사지 크림, 에센스. 아이 크림, 클렌징 크림, 클렌징 폼, 클렌징 워터, 팩 . 스프레이 또는 파우더의 제형으로 제조될 수 있다. 본 발명의 미백용 화장료 조성물은 마키아신 이외에 화장품 제제에 있어서 수용가능한 담체를 포함할 수 있으며, 이미 공지되어 사용되고 있는 피부 미백 효과가 있는 화합물 또는 조성물을 포함할 수 있다.
본 발명의 제형이 페이스트, 크림 또는 겔인 경우에는 담체 성분으로서 동물성유, 식물성유, 왁스, 파라핀, 전분, 트라칸트, 셀를로오스 유도체, 폴리에틸렌 글리콜, 실리콘, 벤토나이트, 실리카, 탈크 또는 산화아연 등이 이용될 수 있다.
본 발명의 제형이 파우더 또는 스프레이인 경우에는 담체 성분으로서 락토스 탈크, 실리카, 알루미늄 히드록시드, 칼슘 실리케이트 또는 폴리아미드 파우더가 이용될 수 있고, 특히 스프레이인 경우에는 추가적으로 클로로플루오로히드로카본, 프로판 /부탄또는 디메틸 에테르와 같은 추진체를 포함할 수 있다.
본 발명의 제형이 용액 또는 유탁액인 경우에는 담체 성분으로서 용매.. 용해화제 또는 유탁화제가 이용되고, 예컨대 물 , 에탄올, 이소프로판올, 에틸 카보네이트, 에틸 아세테이트, 벤질 알코을, 벤질 벤조에이트, 프로필렌글리콜, 1 , 3- 부틸글리콜 오일, 글리세롤 지방족 에스테르, 폴리에틸렌 글리콜 또는 소르비탄의 지방산 에스테르가 있다. 본 발명의 제형이 현탁액인 경우에는 담체 성분으로서
물, 에탄올, 또는 프로필렌 글리콜과 같은 액상의 희석제, 에록실화 이소스테아릴 알코올, 폴리옥시에틸렌 소르비를 에스테르 및 폴리옥시에틸렌 소르비탄 에스테르와 같은 현탁제, 미소결정성 셀롤로오스, 알루미늄 메타히드록시드, 벤토나이트, 아가, 또는 트라칸트 등이 이용될 수 있다.
본 발명의 제형이 계면-활성제 함유 클린징인 경우에는 담체 성분으로서 지방족 알코올 설페이트 , 지방족 알코올 에테르 설페이트 . 설포숙신산 모노에스테르, 이세티오네이트, 이미다졸리늄 유도체 , 메틸타우레이트. 사르코시네이트, 지방산 아미드 에테르 설페이트, 알킬아미도베타인 , 지방족 알코올, 지방산 글리세리드, 지방산 디에탄을아미드, 식물성 유 , 라놀린 유도체 또는 에톡실화 글리세를 지방산 에스테르 등이 이용될 수 있다.
상기 마키아신은 분리 또는 합성될 수 있고 , 다롭나무
<3搬/^/75/5)로부터 추출된 것일 수 있다. 다롭나무는 한국, 중국, 일본, 만주 등지에 분포하는 낙엽 활엽교목으로 산중턱, 산기슭, 골짜기 등 흙이 깊은 곳에서 잘 자란다. 큰 나무는 지름 1 .5m , 높이 15m에 달하며 줄기는 가지를 많이 치고 성장이 빠르다. 잎은 어긋나고 1회 기수 우상복엽이며 작은 잎은 타원형 또는 장난형으로 양면에 털이 없다. 마키아신을 다롭나무로부터 분리하는 방법은 실시예 1에 자세히 기재하였다. 본 발명의 화장료 조성물은 마키아신을 이에 한정하지 않지만, 0.0001 내지 90 중량 % 함유할 수 있고. 바람직하게는 0.001 내지 90중량 % 함유할 수 있다. 상기 마키아신 함량이 0.0001 중량 % 미만인 경우에는 피부미백 효과가 나타나지 않으며. 90중량 %를 초과하게 되면 함유량 증가 대비 효과 증가 정도가 미미하며 . 제형상의 안정 및 안정성에 문제가 있을 수 있고, 경제적이지 못하다. 마키아신의 이러한 조성은 적용 대상의 연령, 성별. 체중, 증상, 제품형태, 적용부위 및 기간 등에 따라 조절될 수 있으며, 따라서 본 발명의' 상기 범위는 이에 제한되는 것은 아니다.
본 발명의 화장료 조성물은 피부 미백을 목적으로 하는 기능성 화장품에 광범위하게 사용될 수 있으며, 따라서 본 발명은 상기 화장료 조성물을 포함하는 피부 미백용 기능성 화장품을 제공한다.

본 발명은 마키아신을 포함하는 조성물을 객체 (subj ect )에 투여하는 단계를 포함하는 피부 미백 방법을 제공한다. 본 발명의 방법은 미백를 요하는 피부에 대해 1일 1회 이상 수행할 수 있다. 피부 외관의 개선 효과는, 피부 상태, 본 발명의 방법에 사용된 활성 성분의 농도, 조성물의 사용량, 적용 빈도 및 달성 이익에 따라 통상적으로 1일 내지 6개월 후에 나타난다.
【유리한 효과】

다롭나무 ( /^c/sr/ amurensis Rupr . et Max.) 2800 g을 99.5 % 메탄올에 침적시켜 3 일 동안 실은에서 침출하였고, 이 침출물을 여과지로 여과하고, 침출 여과과정을 3 회 더 반복한 후, 이 여과액을 감압농축함으로써 메탄올 추출물을 수득하였다. 다롭나무의 메탄을 추출물에 대한 용매분획은 다음과 같이 수행하였다. 먼저, 다롭나무 메탄올 추출물 (99.5% methanol extract)을 증류수에 현탁시켜 분별 깔대기를 이용해 /Hiexane ?— Hex) , me t hy 1 enech 1 or i de ( CH2C 12 ) . ethylacetateCEtOAc), /H itanol 广 BuOH)을 사용하여 분획한 뒤 여과, 감압 농축하여 각각의 용매별 분획물 yrhexane 1.36 g, CH2C124.56g. EtOAc 92.28g. n- BuOH 37.96g„ H20 36.46g을 얻었다.
에틸아세테이트 분획물 (10.0 g)을 가지고 순상 실리카겔과 메틸렌클로라이드와 메탄올을 용매기울기 조건으로 사용해 17개의 분획을 얻었다. 분획— 10에 대하여 C¾CN-Water용매로 prep-LC를 실시하여 화합물을 분리하였다.
얻어진 화합물은丽 R을 이용해 확인한 결과 순수하게 분리되었음을 확인하였다. 핵자기공명기 (Bruker Co. 400 M Hz, 匪 R)를 이용하여 , 13C - NMR스펙트럼 (도 1 및 도 2 참조)을 얻었으며, 이들 스펙트럼을 종합적으로 분석하여 구조를 결정하였다.
그 결과, 화합물은 하기 화학식 1의 마키아신 (maackiasin)으로 동정하였다.

(C-3' "), 113.6 (C-3* , C-5'), 106.3 (06' '), 79.6 (C_a), 79.3 (Οβ), 55.1 (C-OMe) 실험예 1 : 세포 독성 시험 (NRU assay) 상기 실시예의 방법으로 수득한 화합물의 세포독성을 측정하였다. 본 발명의 상기 화합물에 대한 세포독성을 측정하기 위하여 B16Fl(niouse melanoma cell)을 소혈청 10%와 항생제 1%가 포함된 DMEM/high glucoseCDubeco' s Modified Eagle Medium. Hyclone)를 이용한 배지를 이용하여 96-well plate에 200/1!씩 나누어 넣고 37°C, C02 인큐베이터에서 24시간 배양한 후 상기 마키아신을 농도 별로 처리하여 같은 배양 조건에서 72시간 배양하였다. 배양이 끝난 후 배양액을 제거하고 5mg/m Neutral red 시약을 첨가한 배양액을 넣고 37°C에서 2시간 동안 배양하였다. 그 후 배양액을 제거하고 1% CaCl2 & Formaldehyde를 처리하고 1분간 400—500 rpm으로 흔들어 준 뒤 이를 다시 제거하였다. 그리고 1¾ Acetic acid & 50% Ethanol을 각 well에 처리하고 10분간 잘 흔들어 준 다음 마이크로 플레이트 계수기 (Microplate reader)를 이용하여 570 nm/655 nm의 흡광도 측정으로 결과를 측정하였으며, 그 결과는 표 1과 같다.

마키아신 농도에 따른 세포생존율 (¾)
그 결과, 상기 표 1 및 도 4에 나타난 바와 같이 마키아신의 세포독성은 나타나 않았다.
실험예 2 : 멜라닌 생합성 억제 효과측정 상기 실시예의 방법으로 수득한 화합물의 멜라닌 합성율을 측정하였다.
마우스 유래 B16F1 멜라노마 (melanoma) 내 멜라닌 (melanin) 생합성을 억제하는 효과를 측정하기 위해 다음과 같이 실험을 진행하였다. B16Fl(mouse melanoma cell)을 세포를 소혈청 10%와 항생제 1%가 포함된 DMEM/high glucose(Dulbeco 1 s Modified Eagle Medium, Hyclone)에서 배양한 후, 이를 6 well plate에 분주한 다음, 37 °C, 5% C¾조건에서 24시간 배양 하였다. 24시간 배양 후. 배지를 제거하고 a-MSH(Melanocyte stimulating hormone, 멜라닌 세포 자극 호르몬)를 배지에 흔입하여 각 well에 첨가하고 농도별 추출시료도 함께 첨가하였다. 이때 양성대조구는 500«/m 농도의 알부틴을 사용하였다. 72시간 배양한 후 각 plate 내의 세포 내 멜라닌 생성량을 측정하기 위해 우선 plate 내 나머지 배양액을 제거하고, lxPBS(phosphate buffered saline)로 1회 세척하였다. 티로신 (trypsin)을 각 well에 처리하고 5분간 37'C의 C02배양기에서 배양하여 세포를 떼어낸 후, 각 well에 배양액을 처리하고 2πι.Γ마이크로튜브 (microtube)로 옮겼다. 이를 10,000 rpm에 10분간 원심 분리한 후 튜브 안의 배양액을 제거하고 PBS 처리하고, 다시 원심분리와 제거를 통해 세척하였다. 그 후 PBS를 제거하고 70°C의 건조기에서 튜브의 뚜¾올 연 채로 1시간 보관하였다. 건조기에서 꺼낸 튜브에 각각 IN Na0H(10 DMS0)를 처리하고, 다시 70°C 건조기에서 10분간 류브의 뚜껑을 닫은 채 반응시켰다. 반응 후 1,000 rpm에 약 10분간 흔들어주고. 96-well plate에 분주한 후, 마이크로플레이트 계수기를 이용하여 490nm/655nm에서 흡광도를 측정하였다. α-MSH를 첨가하지 않은 a-MSH (-) 그룹의 흡광도를 1로 놓고, 다른 그룹은 이에 대한 상대적인 비율로 측정하여 하기 표 2 및 도 4에 기재하였다. 음성대조군 ( a-MSH (-)로 표시)에는 a-MSH. 마키아신 등을 첨가하지

그 결과, 상기 표 2및 도 4에 나타난 바와 같이 마키아신은 농도의존적으로 멜라닌 합성이 저해되고 있는 것을 확인하여 미백 효과를 나타냄을 알 수 있다. 실험예 3 : 멜라닌 생성 관련 효소의 생성 억제 효과
B16Fl(mouse melanoma cell)을 10% FBS—DMEM에 배양하고 이를 취하여 적정 농도 세포액을 조제하였다. 이를 6-well plate에 분주한 후, 3TC , 5% C02 인큐베이터에서 24시간 배양시켰다. 24시간 배양 후, 배지 제거 후 배지와 농도별 시료를 흔합한 뒤, 다시 동일 조건에 '서 72시간 배양시켰다. Blank의 경우, 배지만을 넣었다. 72시간 배양 후, 각 플레이트 내의 세포를 회수하기 위하여 trypsine-EDTA 용액을 넣고 세포를 분리하여 microtube에 획득한 뒤 원심 분리하여 상등액을 제거하였다. 분리된 pellet을 lxPBS (ᅳ)로 1회 세척하고 원심분리 한 후, pellet에 PRO-PREP™ Protein Extract Solut ionUntron, Inc. )을 처리하여 -20°C에서 15분 반응시킨다. 원심분리 진행 후, 상등액을 tube로 옮겨 -
20°C에 보관하였다. 상기 방법으로 정제한 단백질을 8% SDS-PAGE를 이용 전기영동을 실행하고 이를 PVDF membrane으로 이전하였다. 5% skim mi lk가 함유된 tris 완층용액애서 1시간 blocking 과정을 거친 후에 tyrosinase, TRP-1, TRP-2 ,
B-act in 항체와 각각 반응시켰다. 반응 후, donkey ant i -goat lgG-HRP 가 결합된 항체를 가하고 , Immobi Ion™ Western Chemi luminescent HRP Substrate(Mi l l ipore)를 사용하여 일정 시간 동안 반응 시킨 후 현상하였다. 상기 실험 결과는 하기 도 5에 기재한 바와 같이 마키아신은 10 /zg/ 농도에서 Tyros inase , TRP-1 , TRP-2 , . 및 MITF 발현을 저해하는 것을 확인할 수 있었다. 따라서 마키아신은 멜라닌 생성 단계 관여 효소들의 발현이 '억제되어 나타나는 것 임을 알수 있다.
한편, 본 발명에 따른 상기 화학식 1로 표시되는 마키아신은 목적에 따라 여러 형태로 제형화가 가능하다. 하기에 본 발명의 조성물을 위한 제조예를 예시한다. 제조예 1 : 유연화장수의 제조
소듐히알루로네이트는 프로펠러믹서 (3000rpm)로 정제수에 분산하여 1%용액상태로 준비하였다. 소듐히알루로네이트를 제외한 원료 1 내지 8은 프로펠러믹서를 사용하여 수상 용해조에 500rpm으로 균일화시키고 75°C에서 가온하여 완전 용해한 후, 실온으로 넁각하였다. 이후, 별도의 용해조에 원료 9 내지 11을 완전 용해시킨 후, 상기 수상 용해조에 투입하고 교반 혼합하였다. 여기에 마키아신을 투입하여 충분히 교반혼합하여 유연화장수를 제조하였다.

프로펠러믹서를 이용하여 카보머를 4000rpm으로 분산하여 Ά 용액상태로 준비하였다. 수상 용해조에 원료 1 내지 6을 투입하여 호모믹서 (2000rpm)로 교반하여 분산 후, 75°C까지 가온하였다. 유상 용해조에 원료 7 내지 14를 투입하여 75 T까지 가온 용해하였다. 그 다음 수상 용해조에 용해된 유상을 투입하여 유화 (3000rpm/5분)시킨 후, 실은으로 냉각하였다. 여기에 마키아신을 투입하여 층분히 교반 혼합하여 영양화장수를 제조하였다.

소듬히알루로네이트, 히드록시에칠셀롤로이즈는 각각 정제수에 프로펠러믹서 (2000rpm)로 분산하여 1% solution상태로 준비하였다. 또 카보머는 정제수에 프로펠러믹서 (4000rpm)로 분산하여 2% solution상태로 준비하였다. 한편 수상 용해조에 원료 1 내지 12를 투입하여 호모믹서 (2000rpm)로 교반하여 분산 후 75°C까지 가온하였고, 가온된 수상을 다시 실온으로 넁각하였다. 별도의 용해조에 원료 13 내지 15를 완전 용해시킨 후, 상기 수상 용해조에 투입하여 교반 흔합하였다. 여기에 마키아신을 투입하여 층분히 교반 흔합하여 에센스를 제조하였다.

수상용해조에 원료 1 내지 8을 투입하여 호모믹서 (2000 rpm)로 교반하여 분산후, 75°C까지 가은하였다. 별도의 유상 용해조에 원료 9 내지 16을 투입하여 80°C까지 가온 용해하였다. 수상 용해조에 용해된 유상을 투입하여 유화 (3000 rpm/10분)시킨 후 실은으로 냉각하였다. 여기에 마키아신을 투입하여 층분히

본 발명의 화장료 조성물에 유효성분으로 함유되는 마키아신은 알부틴 (arbLit in)과 비교해 봤을 때, 훨씬 적은 농도에서도 멜라닌 합성 저해효과가 더 큰 바, 피부
미백 효과가 뛰어난 화장료 조성물 및 기능성 화장품을 제조할 수 있어, 산업상 이용가능성이 매우 높다.
