以下の実施例1から5により製造された本化合物の物性における融点はYamato MP-21型融点測定器を使用して測定し、温度計の補正は行っていない。NMRスぺクトルはBruker AVANCEIII500型核磁気共鳴装置で測定し、内部標準としてテトラメチルシランを用いた。シリカゲルカラムクロマトグラフィーはシリカゲルPSQ100B(富士シリシア化学)を用いて行った。薄層クロマトグラフィーはSilica gel F254(Merck、No. 5715)を使用し、検出はUVランプ及び5 w/v%リンモリブデン酸-エタノール発色試薬、ヨウ素‐シリカゲルを用いた。また、試薬及び溶媒は市販品をそのまま用いた。
(1)3-アセチルアミノ-2-オキソクロメン-8-イルベンゼンスルホネートの製造
Mp. 229-231℃. 1H-NMR (DMSO-d6) δ: 2.15 (s, 3H), 7.22-7.34 (m, 2H), 7.58-7.89 (m, 6H), 8.58 (s, 1H), 9.81 (s, 1H).
Mp. 129-131℃. 1H-NMR (DMSO-d6) δ : 5.88 (s, 2H), 6.64 (s, 1H), 6.93-7.87 (m, 8H).

3-[(4-ヒドロキシフェニル)スルホニルアミノ]-2-オキソクロメン-8-イル 4-ヒドロキシベンゼンスルホネート [化合物19] の製造
塩酸2-オキソ-3-[[3-(トリフルオロメチル)フェニル]スルホニルアミノ]クロメン-8-イル 4-アミノベンゼンスルホネート [化合物46] の製造
4-[3-[(3-ニトロフェニル)スルホニルアミノ]-2-オキソクロメン-8-イル]オキシスルホニル安息香酸 [化合物49] の製造
(1)ウシ軟骨片の培養
ウシ中手指節関節軟骨片(直径4 mm)を24ウェルプレートの各ウェルに6~8個ずつ入れ、基礎培地1 mLで37℃、5 vol% CO2に設定したCO2インキュベーター中で一晩培養した。その後、20μmol/Lの被験物質を含む基礎培地で2時間培養し、ヒトリコンビナントIL-1α(3 ng/mL)を添加して更に8時間培養した。培養終了後、培養液を廃棄し、軟骨片にRNA安定化剤を添加して4℃で一晩インキュベートした後、軟骨片をはさみで細切し、マイクロチューブに入れてRNA抽出時まで凍結保存した。
Tissue Lyser(QIAGEN)を用いて、マイクロチューブに入った軟骨片を粉砕し(25Hz、5分、2回)、1 mLのQIAzol Lysis Reagent(QIAGEN)を加えて懸濁し、200μLのクロロホルムを添加・混和後、遠心(4℃、1500 rpm)して上清を回収した。Total RNAは、上清をQIACube(QIAGEN)と RNeasy Micro Kit(QIAGEN)を用いて、抽出・精製した。cDNAは、Omniscript RT kit(QIAGEN)とOligo(dT)12-18 Primer(Invitrogen)を用いて、Total RNAより作製した。
上記(2)で抽出したTotal RNAから合成したcDNAを用いて、Light Cycler LC480(ロシュ・ダイアグノスティックス)による蛍光色素SYBR GREEN I(同)を用いた常法のリアルタイムPCR法により、炎症関連因子〔IL-1β、IL-6、COX-2、iNOS(inducible nitric oxide synthase; 誘導型一酸化窒素合成酵素)〕の遺伝子発現量を測定した。検量線はDNA定量キットPicoGreen dsDNA Quantitation kit(Molecular Probes)で定量したPCR産物を102~107 copies/μLとなるように希釈し作成した。得られた各測定値より、内部標準のGlyceraldehyde-3-phosphate dehydrogenase(GAPDH)遺伝子10000 コピー当たりのコピー数を算出した。
(1)THP-1細胞の培養
THP-1細胞をPMA(phorbol 12-myristate 13-acetate; 100 nM/L)で72時間分化誘導した後、THP-1細胞(5×105 cells/mL)を24ウェルプレート内のRPMI1640+10%FCS培養液中で24時間培養した。その後、最終濃度が12.5μmol/mLになるように被験物質を添加し2時間前培養した後、最終濃度が10 mg/mLになるようにリポ多糖(LPS)を添加し、3時間後に細胞を回収した。
1mLのQIAzol Lysis Reagent(QIAGEN)を加えて懸濁し、200μLのクロロホルムを添加・混和後、遠心(4℃、1500rpm)して上清を回収した。Total RNAは、上清をQIACube(QIAGEN)と RNeasy Micro Kit(QIAGEN)を用いて、抽出・精製した。cDNAは、Omniscript RT kit(QIAGEN)とOligo(dT)12-18 Primer(Invitrogen)を用いて、Total RNAより作製した。
(3)遺伝子発現量に対する作用の測定
上記(2)で抽出したTotal RNAから合成したcDNAを用いて、Light Cycler LC480(ロシュ・ダイアグノスティックス)による蛍光色素SYBR GREEN I(同)を用いた常法のリアルタイムPCR法により、炎症関連因子〔IL-1β、IL-6、単球走化性タンパク質(MCP)-1〕の遺伝子発現量を測定した。検量線はDNA定量キットPicoGreen dsDNA Quantitation kit(Molecular Probes)で定量したPCR産物を102~107 copies/μLとなるように希釈し作成した。得られた各測定値より、内部標準のGAPDH遺伝子10000 コピー当たりのコピー数を算出した。
(1)NHLH細胞の培養
NHLF細胞を12ウェルプレートの各ウェルに4×104 cells/wellになるよう播種し、線維芽細胞基本培地(FBM、Lonza)に線維芽細胞添加因子セット-2(FGM-2 SingleQuots、Lonza)を添加した培地中で80%コンフルエントまで培養した。その後、25μmol/Lの被験物質を含むダルベッコ改変イーグル(DMEM)培地で2時間培養し、最終濃度が5 ng/mLになるように腫瘍増殖因子(TGF)-βを添加して更に24時間培養した。
1mLのQIAzol Lysis Reagent(QIAGEN)を加えて懸濁し、200μLのクロロホルムを添加・混和後、遠心(4℃、1500 rpm)して上清を回収した。Total RNAは、上清をQIACube(QIAGEN)と RNeasy Micro Kit(QIAGEN)を用いて、抽出・精製した。cDNAは、Omniscript RT kit(QIAGEN)とOligo(dT)12-18 Primer(Invitrogen)を用いて、Total RNAより作製した。
上記(2)で抽出したTotal RNAから合成したcDNAを用いて、Light Cycler LC480(ロシュ・ダイアグノスティックス)による蛍光色素SYBR GREEN I(同)を用いた常法のリアルタイムPCR法により、線維化関連因子〔結合組織増殖因子(CTGF)、α-平滑筋アクチン(SMA)、フィブロネクチン、エンドセリン、I型コラーゲン(Col1)〕の遺伝子発現量を測定した。検量線はDNA定量キットPicoGreen dsDNA Quantitation kit(Molecular Probes)で定量したPCR産物を102~107 copies/μLとなるように希釈し作成した。得られた各測定値より、内部標準のGAPDH遺伝子10000 コピー当たりのコピー数を算出した。
(1)一過性発現細胞の作製
NF-κBレポーターベクターのpGL4.32 [luc2P/NF-κB-RE/Hygro](Promega)、コントロールベクターのpGL4.74 [hRluc/TK] (Promega)及び遺伝子導入試薬 Lipofectamine LTX(Life Technologies)を含む調製液を、100 mm Dishに播種したヒト胎児腎臓細胞(HEK293T細胞)に添加し、レポータータンパク質を一過性に発現する細胞を作製した。
遺伝子導入24時間後に一過性発現細胞をトリプシン処理により剥がし、ポリリシンコート白色96ウェルプレートに播種し、1晩培養した。最終濃度が25μmol/mLになるように被験物質を添加し、1時間プレインキュベートした後、最終濃度が10 ng/mLになるようにTNF-αを添加し、更に5時間培養した。細胞を溶解し、Dual-Glo Luciferase Assay System(Promega)を用いてホタルルシフェラーゼ及びウミシイタケルシフェラーゼの活性を測定した。TNF-α刺激による発光強度に対する各薬剤添加による発光強度の割合を算出した。
ICR雄性マウスにブレオマイシン2 mg/kgを5日間反復静脈内投与した。正常対照群、発症対照群、被験物質投与群、ピルフェニドン(Pirfenidone)投与群の4群に群編成した。
被験物質は0.5w/v%のカルボキシメチルセルロ一スナトリウム(CMC)で懸濁し、撹拌しながらラット用経ロゾンデを用いて50 mg/kgを1日2回2週間反復経口投与した。ピルフェニドンは0.5 w/v%のCMCで懸濁し、撹拌しながらラット用経ロゾンデを用いて200 mg/kgを1日2回2週間反復経口投与した。正常対照群及び発症対照群には、同様の投与スケジュールで0.5 w/v%のCMCを反復経口投与した。投与期間終了後、マウスより両肺を摘出し、湿重量を測定した。
(1)ニコチン誘発椎間板変性症ラットの作製
SD雄性ラットを入荷16日後に体重、機械刺激に対する50%反応閾値(測定方法は後述)及び歩行持続時間から正常対照群とニコチン群に編成し、入荷17日後にニコチン群にはニコチン曝露を開始した。ニコチンは生理食塩液で344.8 mg/mLに調製して浸透圧ポンプに封入し、ラットの腰部皮下に留置した。また、留置21日後に、正常対照群以外の5群のラットの皮下に、新たに調製したニコチン溶液を封入した浸透圧ポンプを留置した。ニコチンは生理食塩液で357.0 mg/mLに調製して浸透圧ポンプに封入し、SD雄性ラットの腰部皮下に留置した。
ニコチン曝露開始13日後に機械刺激に対する50%反応閾値、歩行持続時間及び体重を測定して正常対照群、発症対照群、化合物57投与群、化合物58投与群、化合物60投与群、化合物68投与群の6群に群編成した。
被験物質は0.5 w/v%のCMCで懸濁し、撹拌しながらラット用経ロゾンデを用いて10 mL/kg をニコチン曝露開始14から52又は53日後まで1日1回反復経口投与した。正常対照群及び発症対照群には、同様の投与スケジュールと投与方法で0.5 w/v%のCMCを反復経口投与した。
機械刺激に対する50%反応閾値は、ニコチン曝露開始前及びニコチン曝露開始41日後に測定した。底が金網の透明アクリルゲージに、上記(2)記載の6群のラットを入れ、約3分間馴化させた後に、機械刺激に対する50%反応閾値を、被験薬の投与1時間後に測定した。
測定は、Chaplanら(Journal of Neuroscience Methods、53巻、1号、55-63頁、1994年)及びDixonら(Annual Review of Pharmacology and Toxicology、20巻、441-462頁、1980年)の方法に準じ、フォン・フライ フィラメント(von Frey filament、North Coast Medical Inc.製)を用いて行った。8本のフィラメント〔刺激荷重(g):0.4、0.6、1.0、2.0、4.0、6.0、8.0、15.0〕のうち、2.0gのフィラメントより開始し、軽度にフィラメントが湾曲する程度の力で2~3秒間、足底に対し垂直に当て、後肢が逃避反応を示した場合を陽性反応とした。陽性反応が見られた場合は一つ下の、反応がなかった場合は一つ上の強さのフィラメントで同様に刺激し、反応が陰性から陽性へ又は陽性から陰性へ変化した時点を最初の2反応とし、その後4回連続してup-down法により刺激を行った。合計6回の刺激に対する反応を用いて、機械刺激に対する50%反応閾値を測定し、各群の平均値を算出した。なお、陽性反応がないまま15.0 gの刺激まで行った場合は15.0 g、逆に陽性反応が0.4 gまで続いた場合は0.25 gを各々の閾値(疼痛閾値)とした。
ニコチン曝露開始前及びニコチン曝露開始48日後にラット・マウス兼用ロータロッドを用いてラットの歩行持続時間を測定した。回転速度は、ロータが停止した状態から回転を開始して、等加速度で300秒後に15回転/分になるように設定した。ラットは、ロータが停止した状態でロータの上に乗せてからロータを回転させ、落下するまでの時間を測定(最高300秒)し、3試行の平均値を求めた。
(1)NHLH細胞の培養
NHLF細胞を24ウェルプレートの各ウェルに5×104 cells/wellになるよう播種し、線維芽細胞基本培地(FBM、Lonza)に線維芽細胞添加因子セット-2(FGM-2 SingleQuots、Lonza)を添加した培地中で80%コンフルエントまで培養した。その後、30μMの被験物質、200μg/mLのピルフェニドン、30μMのデキサメタゾン(DEX)、1.0μMのTPCA-1又は10μMのフルチカゾンプロピオン酸エステル(FP)を含むDMEM培地で2時間培養し、最終濃度が0.5 ng/mLになるようにTGF-βを添加して更に4時間培養した。

- R1及びR2が同一又は異なって、アルコキシ、アルキル、シアノ、ニトロ、ヒドロキシ、トリフルオロメチル、アミノ、カルボキシ、アルコキシカルボニル、フェニル又は1若しくは2のハロゲンで置換されていてもよいフェニルである請求項1に記載の予防又は治療剤。
- R1及びR2が同一又は異なって、アルコキシ、アルキル、シアノ、ニトロ、ヒドロキシ、トリフルオロメチル、アミノ、カルボキシ、アルコキシカルボニル、フェニル又は1若しくは2のハロゲンで置換されているフェニルである請求項2に記載の予防又は治療剤。
- R1及びR2の両方がトリフルオロメチルで置換されているフェニルである請求項3に記載の予防又は治療剤。
- R1又はR2の一方がトリフルオロメチルで置換されているフェニルである請求項3に記載の予防又は治療剤。
- R1又はR2の一方がトリフルオロメチルで置換されているフェニルであり、他方がハロゲンで置換されているフェニルである請求項5に記載の予防又は治療剤。
- ハロゲンが臭素である請求項6に記載の予防又は治療剤。
- 炎症性疾患が、慢性閉塞性肺疾患又は肺線維症である請求項1乃至7のいずれか一項に記載の予防又は治療剤。
- 経口剤である請求項1乃至8のいずれか一項に記載の予防又は治療剤。
- 注射剤である請求項1乃至8のいずれか一項に記載の予防又は治療剤。
- 外用剤である請求項1乃至8のいずれか一項に記載の予防又は治療剤。
