Abstract

화석 에너지 사용의 꾸준한 증가에 의해 대기 중 이산화탄소의 농도가 높아지고, 이러한 결과 나타나는 지구 온난화 문제와 관련해 이산화탄소의 농도를 점차 줄여나가려는 노력이 점점 가속화되고 있다. 환경 친화적인 신.재생 에너지의 개발과 더불어, 끊임없이 증가하는 화석 연료 사용에 의한 이산화탄소의 빠른 증가를 억제하기 위하여 이산화탄소를 포집 .저장하는 기술아크게 주목 받고 있다. 이러한 이산화탄소의 포집 방법에는 여러 가지 화학적 흡수제, 물리적 흡수 방법 등이 있지만, 부식, 높은 열 에너지 등의 문제를 지닌다. 생명체는 이산화탄소를 빠르게 전환시킬 수 있는 효소를 지니고 있는데, 이러한 효소를 이용한 생물 모방 이산화탄소 포집 방법은 친환경적인 이산화탄소 저감 기술로 주목 받고 있다. 상기 효소는 탄산무수화효소이며 내부에 Zinc 이온을 포함하고 있는 금속 효소 (metalloenzyme)로써 이산화탄소의 수화 반응을 빠르,게 촉진시킨다. 상기 효소는 1900년대 중반부터 포유류에서 집중적으로 연구되었으나, 박테리아와 고세균를 포함하여 실질적으로 거의 모든 생명체에 존재하며 광합성, 호흡, 항상성 유지, 바이오미네랄 형성 등에서 다양한 생리적인 역할을 수행하는 것으로 밝혀졌다.
기체 형태의 이산화탄소는 물에 녹고, 이러한 액상의 이산화탄소는 수화 반웅을 통해 탄산을 형성한다. 적정 pH 조건에서는 최종적으로는 carbonate ion (C03 2 )이 형성된다. 이것은 이후 금속 양이은과 반응하여 탄산 미네랄이라는 고체 침전물을 형성할 수 있다ᅳ 전체적인 반웅은 하기 [화학식 1] 내지 [화학식 5]와 같다.

5]의 반응의 율속 단계 (rate-limiting step)이며, 탄산무수화효소는 이산화탄소와 물을 탄산수소이온 (HCCV)과 수소이온 (H+)으로 바꾸는 것을 촉매하여, 상기 반웅을 자연적인 반응 속도보다 최대 천만 배 더 빠르게 촉진시킨다. 따라서 탄산무수화효소를 통해 액상에서 이산화탄소를 빠르게 포집할 수 있으며, 이러한 과정은 응용에 따라 중탄산 이온으로의 포집과 탄산 미네랄로의 전환을 포함한다. 탄산무수화효소를 이용한 이러한 생물 모방 기술은 친환경적이고
효율적이지만 실제 공정 적용에 있어서 해결해야 할 부분이 있는데, 그것은 탄산무수화효소의 생산 단가를 낮추는 것과 효소 안정성 (stability)을 확보하는 것이다. 포집 공정에 탄산무수화효소를 적용하려면 탄산무수화효소를 대량으로 값싸게 얻을 수 있어야 한다. 그 동안 연구에서 주로 사용되어 왔던 소 혈청에서 추출한 탄산무수화효소는 g당 약 2백만원이며 이러한 고비용 문제로 실제 공정에는 적용되기 힘들다. 또한, 포집 공정은 대량으로 이산화탄소를 배출하는 발전소, 제철소 등에 적용될 것으로 기대되는데, 이산화탄소를 포함한 배가스가 지닌 열의 넁각 (cooling)과정이 필요하며 흡수탑에서는 녹아 들어가는
이산화탄소에 의해 열이 방출된다. 또한 녹아 들어간 이산화탄소를 기체상태로 분리하기 위해서는 재생탑이 고온으로 유지되어야 한다. 탄산무수화효소를 흡수탑 공정에 적용하기 위해서는 기본적으로 40 내지 60°C 정도에서 안정해야 하며 재생탑 공정에 적용하기 위해서는 이보다 더 높은 온도에서도 활성을 오래도록 유지할 수 있어야 한다.
【발명의 상세한 설명】
【기술적 과제】
고온에서 안정성이 높고 이산화탄소 포집 활성을 나타내는
탄산무수화효소를 생산하기 위하여, 본 발명은 탄산무수화효소, 상기
탄산무수화효소를 코딩하는 핵산분자, 상기 핵산분자를 포함하는 재조합 백터, 및 상기 재조합 백터로 형질전환된 숙주세포 및 상기 숙주세포를 이용하여
탄산무수화효소를 제조하는 방법을 제공하는 것을 목적으로 한다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 국한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업자에게 명확하게 이해될 수 있을 것이다.
【기술적 해결방법】
상기 목적을 이루기 위한 수단으로, 본 발명은 탄산무수화효소, 상기 탄산무수화효소를 코딩하는 핵산분자, 상가핵산분자를 포함하는 재조합 백터, 및 상기 재조합 백터로 형질전환된 숙주세포 및 상기 숙주세포를 이용하여
탄산무수화효소를 제조하는 방법을 제공한다.
본 발명의 일 구현예는 탄산무수화효소를 제공한다.
다른 일 구현예는 상기 탄산무수화효소를 코딩하는 핵산분자를 제공한다. 또 다른 일 구현예는 상기 핵산분자를 포함하는 재조합 백터를 제공한다. 또 다른 일 구현예는 상기 재조합 백터로 형질전환된 숙주세포를 제공한다.
또 다른 일 구현예는 상기 탄산무수화효소를 포함하는 이산화탄소 포집제를 제공한다.

본 발명자들은 해저 열구수에서 발견된 고온성 세균의 유전체 정보로부터 탄산무수화효소 유전자를 탐색하고 이를 토대로 재조합 발현 백터를 제작한 뒤, 대장균 내에서 성공적으로 대량 발현함으로써 본 발명을 완성하였다. 재조합 탄산무수화효소가 발현된 세포의 파쇄액은 높은 이산화탄소 포집 활성을 지니고 있으며 정제된 탄산무수화효소의 kinetic parameter는 알려져 있는 고세균 유래의 탄산무수화효소보다 더 뛰어나며, 95 °C 온도에서도 효소활성을 갖는 것으로서, 이산화탄소 포집 공정의 높은 온도 조건에서는 상온에서보다 더욱 뛰어난 활성을
지닐 수 있다는 것을 확인하였다. 또한 발현된 탄산무수화효소는 고온에서 대부분의 활성을 유지하며 상당히 안정성이 높은 것이 특징이다.
【유리한 효과】
본 발명의 탄산무수화효소는 고온에서의 안정성이 높아 고온에서도 이산화탄소 포집 활성을 나타내어 실제 고온에서 수행되는 이산화탄소 포집 공정에 적용될 수 있다. 또한 발현 시스템올 이용하여 대량생산이 가능하므로 경제성 측면에서 많은 이점을 줄 것으로 기대된다.

【발명의 실시를 위한 최선의 형태】
이하, 본 발명을 상세히 설명한다.
일 구현예는 탄산무수화효소를 제공한다.
상기 탄산무수화효소는 고온성 세균으로부터 유래된 것일 수 있으며, 고온성 세균으로부터 유래됨에 따라 높은 온도에서도 이산화탄소 포집 활성이 유지되는 특징을 가진다. 예컨대, 상기 탄산무수화효소는 T. ammonificans, P. marina, 또는 C mediatlanticus로―부^ 유래된 것일 수 있으며, 바람직하게는 T.
ammom Za s로부터 유래된 것일 수 있다.
상기 탄산무수화효소는 분자량이 약 27 kDa인 효소로서, 반응속도 상수는
Lineweaver-Burk plot을 이용하여 각각 KM= 10 내지 38 mM, Kcat= 2.8x l05 내지 6.8x l 05/s인 것을 특징으로 한다. 또한 이산화탄소 이외에 에스테르를 분해할 수 있는 특이성을 지니며 0.9 내지 3.2 mol ?-nitrophenyl acetate/mol enzyme min≤ 활성을 지닌다.
상기 탄산무수화효소는 내열성 효소로서 80 °C에서도 적어도 15분간 그 활성을 그대로 유지하며, 95 °C 에서도 수분간 효소활성을 유지하며, 구체적으로 4 °C에서의 이산화탄소 포집 활성을 100% 기준으로 하여 40 내지 70 °C에서 이산화탄소 포집 활성이 60% 이상, 바람직하게는 70% 이상, 가장 바람직하게는 80% 이상일 수 있다. 이때, 이산화탄소 포집 활성은 탄산무수화효소가
이산화탄소와 물을 탄산수소이온 (HC V)과 수소이온 (H+)으로 바꾸는 활성을 의미한다.
본 발명에 따른 T.am 유래의 탄산무수화효소는 P.ma 또는 C.me 유래의 탄산무수화효소보다 고온 안정성이 뛰어남을 확인하였다 (도 4, 도 5 및 도 6) 상기 탄산무수화효소는 서열번호 1로 표시되는 아미노산 서열로 이루어진 것일 수 있으며, 또는 상기 탄산무수화효소는 상기 서열번호 1의 아미노산 서열의 말단에 제한효소인식 부위 및 /또는 정제를 위한 을리고펩타이드를 추가로 포함할 수 있다. 상기 제한효소인식 부위는 재조합 효소 생산시 사용하는 백터의 제한효소 부위와 일치되도록 다양한 서열을 포함할 수 있으며, 정제를 위한 올리고펩타이드는 다양한 Tag을 사용할 수 있으며 예를 들면 6X(His) tag일 수 있다. 본 발명의 일예에 따른 탄산무수화효소는 서열번호 1의 아미노산 서열의 말단에 제한효소인식 부위 및 /또는 정제를 위한 올리고펩타이드가 결합된 펩타이드인, 서열번호 2의 아미노산 서열을 포함하는 펩타이드일 수 있다. 본 발명에서 서열번호 1로 이루어지는 탄산무수화효소는 Γ. amwo ?cara에서 유래된 것이며, 상기 서열번호 3로 이루어지는 P. manna 유래 탄산무수화효소 이며, 상기 서열번호 4로 이루어지는 C. mediatlantic 유래된 탄산무수화효소와 비교된다. 다른 일 구현예는 상기 탄산무수화효소를 코딩하는 핵산분자를 제공한다. 상기 핵산분자는 고온성 세균의 탄산무수화효소의 신호 염기서열올 제거한 것일 수 있다. 상기 신호 염기서열을 포함하고 있는 경우
탄산무수화효소가 세포 간극으로 이동하여 대장균과 같은 발현 시스템에서 원하는 양만큼의 발현이 이루어지지 않게 된다. 상기 핵산분자는 숙주세포,
바람직하게는 대장균의 코돈 편향성을 고려하여 적절히 조절될 수 있다.
상기 핵산분자는 서열번호 1로 표시되는 아미노산 서열에 의해서
암호화되는 염기서열을 포함할 수 있다. 또는 상기 서열번호 1의 아미노산 서열의 말단에 제한효소인식 부위 및 /또는 정제를 위한 올리고펩타이드를 추가로 포함하는 아미노산 서열에 의해서 코딩되는 핵산분자 일 수 있으며 예를 들면 서열번호 2의 아미노산 서열에 의해서 코딩되는 핵산분자일 수 있다.
또 다른 일 구현예는 상기 핵산분자를 포함하는 재조합 백터를 제공한다. 상기 백터는 전형적으로 외부 DNA가 삽입될 수 있는 전달 DNA를 포함하며, 핵산분자의 일종으로 또 다른 핵산과 결합하여 숙주세포로 이송되어 목적 단백질을 발현할 수 있는 것을 의미한다. 예컨대, 상기 백터는 플라스미드 백터, 코스미드 백터, 박테리오파지 백터 및 바이러스 백터 등을 포함한 통상의 모든 백터를 포함한다.
또 다른 일 구현예는 상기 재조합 백터로 형질전환된 숙주세포를
제공한다. 상기 재조합 백터는 숙주세포 내부로 도입될 수 있으며, 도입 방법은 전기충격 유전자 전달법 (electroporation), 인산칼슘 (CaP04) 침전, 염화칼슘 (CaCl2) 침전, PEG, 텍스트란 설페이트, 리포펙타민 등의 공지된 방법으로 수행할 수 있다. 상기 숙주세포는 원핵세포일 수 있다. 상기 원핵세포는 외래 유전자로 형질전환 될 수 있는 원핵세포이면 가능하며, 예를 들어, 대장균 (Escherichia coli), 로도코커스 (Rhodococcus), 슈도모나스 (Pseudomonas), 스트렙토마이세스
(Streptomyces), 스타필로코커스 (Staphylococcus), 시폴러버스 (Syfolobus),
써모플라즈마 (Thermoplasma), 써모프로테우스 (Thermoproteus) 등의 다양한 미생물을 포함할 수 있으며, 바람직하게는 대장균 및 사카로미세스
세리비시아 (Saccharomyces cerevisiae)로 이루어진 군으로부터 선택될 수 있다.
바람직하게는 대장균일 수 있으며, 구체적으로 대장균 XLl-blue, 대장균 BL21(DE3), 대장균 JM109, 대장균. DH 시리즈, 대장균 TOP 10 및 대장균 HB101 등을 포함할 수 있다. .
또 다른 일 구현예는 상기 숙주세포를 이용하여 탄산무수화효소를 제조하는 방법을 제공한다.
배지성분, 배양온도 및 배양시간 등의 조건을 적절히 조절하여, 상기 형질전환된 숙주세포를 배양할 수 있다. 구체적으로, 배양 배지는 탄소원, 질소원,
미량원소 성분 등과 같은 미생물의 성장과 생존에 필수적인 모든 영양소를 포함할 수 있다. 배지의 pH는 적절히 조정할 수 있고, 항생제 등의 성분을 포함할 수 있다.
또한, 이소프로필 베타 디 티오갈락토피라노사이드
(isopropyl- -D-thiogalactopyranoside, 이하 IPTG라고 함) 등의 유도제 (inducer)를 처리하여 탄산무수화효소의 발현을 유도할 수 있다. 처리되는 유도제의 종류는 백터 시스템에 따라 결정될 수 있으며, 유도제 투여시간 및 투여량 등의 조건은 적절하게 조절될 수 있다. 배지성분, 배양온도 및 배양시간 등의 조건의 선택은 사용하는 숙주세포의 종류에 따라 적절히 결정될 수 있다.
발현된 탄산무수화효소는 통상의 방법으로 회수 및 정제된다. 예를 들어, 원심분리 방법으로 회수한 세포를, 프렌치 프레스, 초음파 파쇄기 등을 이용하여 파쇄 할 수 있다. 배양액으로 탄산무수화효소가 분비되는 경우에는 배양 상등액을 수집할 수 있다. 과발현에 의해 웅집되는 경우, 적절한 용액에서
탄산무수화효소를 용해시키고 및 변성시키고 재폴딩시켜서 얻을 수 있다.
글루타티온, 디티오트레이를 , β-머갑토에탄을 , β-머갑토메탄을, 시스틴 및
시스타민의 산화 및 환원 시스템을 사용하고 요소, 구아니딘, 아르기닌 등의 재폴딩제 등을 이용할 수 있다. 재폴딩제와 함께 염의 일부를 사용할 수 있다. 이 때, 70 내지 85 °C에서 10 내지 60분간 열처리하는 공정을 추가할 수도 있으며, 상기 탄산무수화효소의 제조규모는 목적에 맞도록 조절할 수 있다.
또 다른 일 구현예는 상기 탄산무수화효소를 포함하는 이산화탄소 포집제를 제공한다. 상기 포집제는 이산화탄소 포집제로 알려진 것들올
추가적으로 포함할 수 있으며, 예컨대 암모니아 수용액 , MEA(mono-ethanol-amine) 수용액, 탄산칼륨 수용액 등을 포함할 수 있다.
또 다른 일 구현예는 상기 탄산무수화효소를 이용하여 이산화탄소를 포집하는 방법을 제공하며, 구체적으로 상기 방법은 40 내지 70°C에서 상기 탄산무수화효소를 이용하여 포집하는 방법일 수 있다.

실시예 1. 형질전환된 숙주세포 제조

mediatlantic S^ 탄산무수화효소를 코딩하는 핵산을 각 미생물의 유전체 핵산을 주형으로 PCR 프라이머를 사용하여 각각 증폭하였다.
구체적으로 탄산무수화효소가 세포질에서 발현될 수 있게 Γ.
ammonificam^ 신호서열이 제거된 탄산무수화효소의 염기서열, P. war/«a의 신호서열이 제거된 탄산무수화효소의 염기서열로 증폭하였고, C.

상기 각각의 증폭산물을 NdeI, XhoI 제한 효소를 이용하여 pET-22b (+)
백터에 도입하여, 최종적으로 3개의 발현백터를 제조하였으며 , pET-22b(+) 백터에서 각각의 탄산무수화효소 유전자들은 C-말단에 Ni 이은과 결합이 가능한 histidine tag를 갖는다. 구체적으로 , T. ammonificans 탄산무수화효소는 아미노산 서열은 서열번호 2, P. marina 탄산무수화효소의 아미노산 서열은 서열번호 3, C. mediatlanticus탄산무수화효소의 아미노산 서열을 서열번호 4를 갖는다.
42 °C에서 2분간 열층격하는 방법으로 제조된 각각의 백터를 숙주세포인 대장균 BL21 (DE3)에 도입하였고, 백터가 도입된 각각의 숙주세포는
암피실린 (ampidllin)이 첨가된 LB 배지에서 선별하여 최종적으로 유래가 다른 탄산무수화효소를 발현하는 3종류의 숙주세포를 제조하였다.
실시예 2. 단백질 발현 분석
상기 실시예 1에서 각각 제조된 숙주세포를 50 g/mL 암피실린 첨가된 LB 배지에서 37 °C로 배양하여 배양액의 흡광도 (OD600)가 0.6~0.8 정도가 되었을 때 유도물질인 IPTG(isopropyl-13-D-thiogalactopyranoside, 1 mM)을 첨가하여 단백질의 발현을 유도하였다. IPTG 첨가 후 추가로 12시간 동안 37 °C에서 숙주세포를 배양한 후, 배양된 숙주세포를 4000xg에서 10분간 원심 분리한 다음 상등액을 제거하고 숙주세포를 회수하였다. 회수된 숙주세포는 파쇄를 위한 용액 (50 mM sodium phosphate buffer, 300mM NaCl, pH 8, 10 mM imidazole)에 부유시킨 다음 초음파 분쇄기를 이용하여 파쇄하였다. 이후 파쇄된 숙주세포의 단백질 총체를 SDS-PAGE를 통해 분석하였으며, 그 결과를 도 1에 나타내었다.
도 1은 3종류의 탄산무수화효소를 세포질에 발현한 후 SDS-PAGE로 분석한 결과로서 , Μ은 분자량 표준 마커, C. e는 Caminibacter mediatlanticus 유래 탄산무수화효소 , .wa는 Persephonella marina 유래 탄산무수화효소 , r.aw는

도 1에 나타난 바와 같이, 탄산무수화효소는 각각의 숙주세포에서 성공적으로 발현됨을 확인하였다.
실시예 3. 이산화탄소 포집 활성 확인
실시예 2의 숙주세포의 파쇄액을 이용하여 발현된 탄산무수화효소의 이산화탄소 포집에 관한 활성올 측정하였다. 3 mL의 20 mM Tris sulfate buffer(pH 8.3)에 100 의 숙주세포의 파쇄액을 첨가한 후 이산화탄소 포화수용액 (C02 saturated ¾0 solution)을 넣어줌으로써 반웅을 시작시켰고, 탄산무수화효소의 pH가
감소하는 것을 관찰하였고, 그 결과를 도 2에 나타내었다.
도 2는 숙주세포의 파쇄액을 이용하여 이산화탄소 포집에 관한 3종류의 탄산무수화효소의 활성을 측정한 결과를 나타낸 것으로, Blank는
탄산무수화효소가 들어 있지 않은 비교군 , P.ma CA는 Persephonella marina 유래 탄산무수화효소 , T.am CA는 Thermovibrio ammonificans 유래 탄산무수화효소, C.me CA는 Caminibacter mediatlanticus 유래 탄산무수화효소를 의미한다.
도 2에 나타난 바와 같이, Persephone!!a marina 유래 탄산무수화효소, Thermovibrio ammonificans 유래 탄산무수화효소, 및 Caminibacter mediatlanticus 유래 탄산무수화효소의 pH는 비교군에 비해 pH가 빨리 떨어지는데, 이는
탄산무수화효소의 이산화탄소 포집에 관한 활성이 높다는 것을 의미한다.
실시예 4. 탄산무수화효소의 정제
실시예 2의 숙주세포의 파쇄액을 10,000xg에서 20분간 원심 분리한 후 상등액의 탄산무수화효소를 분리, 정제하였다. 구체적으로, 니켈 레진이 층진된 컬럼에 상등액을 적용하여 탄산무수화효소와 컬럼이 결합하도록 하고, 세척 버퍼 (50 mM sodium phosphate buffer, 300mM NaCl, 30mM imidazole, pH 8.0)로 컬럼에 결합하지 않은 탄산무수화효소를 씻어 내었다. 컬럼으로부터 탄산무수화효소의 용출은 50 mM sodium phosphate buffer, 300mM NaCl, 250mM imidazole (pH 8.0)을 이용하여 진행하였고, 상기 정제된 용액은 투석을 이용하여 20mM Tris-sulfate(300 mM NaCl, pH8.3)으로 바뀌며 imidazole이 제거되었으며, 최종적으로 3종류의 탄산무수화효소를 정제하였다.
상기 정제된 3종류의 탄산무수화효소를 실시예 2와 같은 방법으로
SDS-PAGE를 통해 분석하였으며, 그 결과를 도 3에 나타내었다.
도 3은 3종류의 탄산무수화효소를 각각 분리, 정제한 후 SDS-PAGE로 분석한 결과를 나타낸 것으로 , P.ma는 Persepho lla marina ^캑、 탄산무수화효소, me는 Caminibacter mediatlanticus 유래 탄산무수화효소, T.am는 Thermovibrio ammonificans 유래 탄산무수화효소를 의미한다.
도 3에 나타난 바와 같이, SDS-PAGE 분석을 통해 각 탄산무수화효소가 완전하게 정제되었음을 확인하였다.
실시예 5. 고온 안정성 비교
고온에서의 탄산무수화효소의 안정성을 확인하기 위해, 실시예 4에서 분리,
정제된 3종류의 탄산무수화효소를 70 °C에서 16시간 방치한 후 4 °C에 보관해둔 실험군 (No treatment)과 비교하여 활성의 감소 정도를 측정하였고, 대조군으로는 상업적으로 판매되는 소 혈청 유래 탄산무수화효소를 사용하였다. 활성은 실시예 3에서 사용한 이산화탄소 포집 활성으로 측정하였으며, 그 결과는 도 4에 나타내었다.
도 4는 3종류의 정제된 탄산무수화효소의 70 °C에서의 고온 안정성을 측정한 결과를 나타낸 것으로, 무처리 (no treatment)는 열처리를 하지 않은 대조군, T.am는 Thermovibrio ammonificans 유래 탄산무수화효소, P. ma는 Persephonella marina 유래 탄산무수화효소, C. me는 Caminibacter mediatlanticus 유래
탄산무수화효소, bCA는 소 혈청 유래 탄산무수화효소를 의미하며 ,
잔류활성 (Residual activity)(%)는 초기 열처리 하지 않았을 때와 비교하여 열처리를 하였을 때 얼만큼 활성을 유지하는지를 나타낸 것이디-.
도 4에 나타난 바와 같0 Thermovibrio ammonifica 유래 탄산무수화효소는 80% 이상의 Residual activity를 나타내어 고온에서도 안정성이 유지됨을
확인하였다.
또한, 실질적인 포집 온도 조건에서 오랜 시간 동안의 열 안정성을 확인하기 위하여 40 °C와 60 °C에서의 열 안정성을 60일간 테스트하였다.
마찬가지로 각 온도에서 일정시간 효소를 방치한 뒤 4 °C에 보관해둔 실험군과 비교하여 활성의 감소정도를 측정하였고, 그 결과를 도 5에 나타내었다.
도 5a 및 도 5b는 Persephonella marina 유래 탄산무수화효소와 Thermovibrio ammonificans 유래 탄산무수화효소를 각각 40 °C 및 60 °C에서 시간에 따른 고온 안정성을 측정한 그래프를 나타낸다ᅳ
도 5a 및 도 5b에 나타난 바와 같이, The vibrio ammonifwa 유래와 Persephonella marina 유래의 탄산무수화효소는 높은 열 안정성을 보였다. 특히, Thermovibrio ammonificans 유래 탄산무수화효소는 40 °C의 경우 60일 후 초기 활성의 91 %를 지녔으며 (도 5a), 6( C의 경우에도 60일 후 초기 활성을 62%를 유지하였다 (도 5b).
실시예 6. Kinetic parameter 즉정
C02에 대한 정밀한 kinetic 측정을 위하여 stopped-flow spectroscopy를 사용하였다. 10 nM에서 100 nM의 탄산무수화효소를 포함하는 l OO mM
N-Tris(hydroxymethyl)methyl-3-aminopropanesulfonic acid (TAPS)/NaOH buffer (57.2 mM Na2S04와 97.2 μΜ m-cresol purple를 포함. !^ 8.5)를 다양한 농도의 C02 용액과 섞은 후 초기 pH의 변화를 25 °C에서 578 nm의 흡광도 변화를 통해 관찰하였다. 얻어진 데이터로 Michadis-menten 식을 사용하여 KM, Kcat 값을 구하였다.
상기 탄산무수화효소는 C02에 관한 기질 특이성 이외에 ester를 분해할 수 있는 esterase의 능력을 지닌다 . 100 μΐ 의 30 mM p-nitrophenyl acetate 를 800 μΐ의 buffer (50 mM potassium phosphate; pH 7.0)와 100 μΐ의 탄산무수화효소를 지닌 용액에 넣고 잘 섞은 후 25 °C에서 3분간 348nm에서의 흡광도 변화를 측정함으로써 esterase 활성을 보았다. 표 1은 bCA, T.am CA, P.ma CA에 대한 kinetic parameter를 나타낸다. 표 1은 실시예 6에 따른 정제된 탄산무수화효소의 반응속도 값을 나타낸 것이다.

실시예 6에서 사용한 esterase활성 측정 방법을 25 °C이외의 더 높은 온도에도 적용함으로써, 온도에 따라 탄산무수화효소의 활성 변화를 측정하였다. 도 6에서 볼 수 있듯이 P. marina와 T. ammonificans 유래의 탄산무수화효소는 모두 높은 온도에서 더욱 높은 활성을 지니고 있다. 95 °C에서 가장 높은 활성을 보였으며, 더 높은 온도는 실험 특성상 측정할 수 없었다. 두 미생물의 최적 생장 온도 (70 °C부근)를 고려했을 때 두 탄산무수화효소의 최적 반웅 온도는 95 °C보다 크게 높지 않을 것으로 추측된다.
