Abstract
Description
ある種のMMPを阻害する化合物は、TNF-αの産生も阻害することが報告されている(非特許文献4参照)。また、TACEとMMPは、共に触媒部位に亜鉛を有する酵素であり、その3次元構造も類似していることもあって、これまでMMPおよびTACEを共に阻害する化合物が報告されている(非特許文献5参照)。しかしながら、多種類のMMPを同時に阻害する薬剤を連続投与したラットでは軟骨である成長板の肥層化が起こること(非特許文献6参照)や、MT1-MMP(MMP-14)ノックアウトマウスでは関節炎症状が観察されること(非特許文献7参照)などの報告から、MMP阻害により種々の副作用を生じる懸念がある。更に、MMPの多くは、生体構造の基本となる細胞外マトリックスの維持、恒常性に関与していることから、非選択的に多種のMMP酵素活性を阻害することは、生体にとって重大な悪影響を与える危険性がある。したがって、TACE阻害に基づくTNF-α産生抑制を目的とした化合物は、MMPに対する阻害作用を実質的に示さないことが好ましい。
本発明は、このようなTNF-αを介する疾患の治療および予防を鑑みてなされたものである。すなわち本発明の目的は、MMPに対する阻害作用が弱い、選択的なTACE阻害作用を示す新規化合物またはその塩を提供すること、およびこれらを有効成分とする医薬を提供することにある。
すなわち本発明は、少なくとも以下の各発明に関する:
(1)(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩。
(3)(-)異性体を実質的に含まない前記(2)に記載の医薬。
(4)前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の95重量%以上が(+)異性体である前記(2)または(3)に記載の医薬。
(5)前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の98重量%以上が(+)異性体である前記(4)に記載の医薬。
(6)前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の99.5重量%以上が(+)異性体である前記(5)に記載の医薬。
(8)前記外用製剤がローション剤、クリーム剤、液剤および軟膏剤からなる群から選択される剤形である前記(7)に記載の医薬。
(9)薬学的に許容されるグリコール類を含む前記(7)または(8)に記載の医薬。
(11)製剤中に水を含む前記(7)から(10)のいずれかに記載の医薬。
(12)有効成分として、1w/w%以上の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩を含有する前記(7)から(11)のいずれかに記載の医薬。
(13)有効成分として、3w/w%以上の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩を含有する前記(12)に記載の医薬。
(14)経皮投与により投与される前記(2)から(13)のいずれかに記載の医薬。
(15)皮膚疾患の予防または治療剤である前記(2)から(14)のいずれかに記載の医薬。
(16)皮膚疾患が、限局性強皮症、アトピー性皮膚炎、接触性皮膚炎、乾癬およびざ瘡からなる群より選択される1つ以上の疾患である前記(15)に記載の医薬。
(18)ラセミ体の(±)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンを光学分取用のカラムを用いたクロマトグラフィーによって光学分割して得られる、(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン。
(20)前記混合物のうち、(+)異性体が(-)異性体に対して96%ee以上で存在する前記(19)に記載の混合物。
(21)前記混合物のうち、(+)異性体が(-)異性体に対して99%ee以上で存在する前記(19)または(20)に記載の混合物。
まず、本発明の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンの塩としては、薬学的に許容される塩であればとくに制限されず、例えば、無機塩基との塩、有機塩基との塩などがあげられる。無機塩基との塩の例としては、リチウム塩、ナトリウム塩、カリウム塩、マグネシウム塩、カルシウム塩、バリウム塩などのアルカリ金属塩およびアルカリ土類金属塩などがあげられる。有機塩基との塩の例としては、トリエチルアミン塩、ピリジン塩、エタノールアミン塩、シクロヘキシルアミン塩、ジシクロヘキシルアミン塩、ジベンジルエタノールアミン塩、ベンジルアミン塩、2-メチルベンジルアミン塩、α-メチルベンジルアミン塩、ブルシン塩、キニーネ塩、キニジン塩、シンコニン塩、シンコニジン塩、アルギニン塩などがあげられる。
化合物(+)-(I)は、例えば下記のスキーム1(工程1~工程3)に示される方法により製造することができる。
(式中、Xは塩素原子、臭素原子、ヨウ素原子を表す。アスタリスク(*)は光学的に純粋な形態であること意味する。)
工程1においては、一般式(II)で表される化合物と化合物(III)を塩基の存在下反応し、化合物(IV)を製造する。化合物(III)の代わりに、その互変異性体である化合物(V)
を用いても同様に製造することができる。好適な塩基としては、炭酸カリウム、炭酸ナトリウム、炭酸セシウムまたは水素化ナトリウムなどがあげられる。反応を円滑に行うために添加物を共存させてもよく、添加物として、ヨウ化カリウム、ヨウ化ナトリウム、テトラブチルアンモニウムヨージド、臭化カリウム、臭化ナトリウムまたはテトラブチルアンモニウムブロミドなどを加えることができる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、N,N-ジメチルホルムアミド、ジメチルスルホキシド、アセトン、アセトニトリル、メタノール、エタノール、テトラヒドロフラン、1,4-ジオキサン、2-メトキシエタノールまたはそれらの混合溶媒などが好ましい。また、反応溶媒に水を添加することもできる。水を添加する場合、水の添加量は特に限定されず、例えば、反応溶媒に対して10%以下が好ましい。反応温度はとくに限定されず、例えば室温~60℃が好ましい。反応時間は、1時間~2日間が好ましい。
工程2においては、化合物(IV)を塩存在下、シアン化物との反応を経てラセミ体である化合物(±)-(I)を製造する。好適な塩としては、炭酸アンモニウムまたは炭酸水素アンモニウムなどがあげられる。好適なシアン化物としては、シアン化カリウムまたはシアン化ナトリウムなどがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、水、アンモニア水、メタノール、エタノール、テトラヒドロフラン、1,4-ジオキサン、N,N-ジメチルホルムアミドまたはそれらの混合溶媒などが好ましい。反応温度はとくに限定されず、例えば50℃~120℃が好ましい。反応時間は、1時間~10日間が好ましい。本工程において得られる化合物(±)-(I)は、反応終了後の処理方法次第で、その塩の形態として得ることもできる。
工程3においては、化合物(±)-(I)を、キラルカラムを用いて光学分割し化合物(+)-(I)を製造する。キラルカラムを用いた光学分割は、当業者における公知の方法(例えば、『光学異性体の分離』(季刊化学総説No.6 1989 日本化学会編 学会出版センター)など参照)に準じて行えばよい。また、用いるキラルカラムは多様なものが市販されており、適宜、適切なものを選択すれば良いが、両光学異性体の分離能が良い点で、(株)ダイセル社製のCHIRALPAK ADが好ましい。
工程4においては、化合物(III)と一般式(VI)で表される化合物を塩基の存在下反応し、一般式(VII)で表される化合物を製造する。化合物(III)の代わりに、その互変異性体である化合物(V)
を用いても同様に製造することができる。好適な塩基としては、炭酸カリウム、炭酸ナトリウム、炭酸セシウムまたは水素化ナトリウムなどがあげられる。反応を円滑に行うために添加物を共存させてもよく、添加物として、ヨウ化カリウム、ヨウ化ナトリウム、テトラブチルアンモニウムヨージド、臭化カリウム、臭化ナトリウムまたはテトラブチルアンモニウムブロミドなどを加えることができる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、N,N-ジメチルホルムアミド、ジメチルスルホキシド、アセトン、アセトニトリル、メタノール、エタノール、テトラヒドロフラン、1,4-ジオキサン、2-メトキシエタノールまたはそれらの混合溶媒などが好ましい。反応温度はとくに限定されず、例えば室温~60℃が好ましい。反応時間は、1時間~2日間が好ましい。
工程5においては、一般式(VII)で表される化合物を無機塩基水溶液の存在下に加水分解して、化合物(VIII)を製造する。好適な無機塩基水溶液としては、水酸化ナトリウム水溶液、水酸化カリウム水溶液または水酸化リチウム水溶液などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、水、メタノール、エタノール、テトラヒドロフラン、1,4-ジオキサンまたはそれらの混合溶媒などが好ましい。反応温度はとくに限定されず、例えば室温~60℃が好ましい。反応時間は、1~96時間が好ましい。本工程において、化合物(VIII)は、カルボン酸、カルボン酸ナトリウム、カルボン酸カリウム、カルボン酸リチウム、無機塩(塩化ナトリウム、塩化カリウムまたは塩化リチウム)とカルボン酸との混合物などの形態で得られる。
工程6においては、工程5で得られた化合物(VIII)を、活性化されたカルボン酸誘導体に導き、アミンまたはその塩と反応させて、化合物(IX)を製造する。活性化されたカルボン酸誘導体としては、例えば、化合物(VIII)を塩化チオニル、オキシ塩化リン、五塩化リン、塩化オキサリル、臭化チオニル等で処理することにより得られる酸ハロゲン化物;化合物(VIII)を1-エチル-3’-(3’-ジメチルアミノプロピル)カルボジイミド塩酸塩またはジシクロへキシルカルボジイミド等の縮合剤と縮合することにより得られる活性エステル;化合物(VIII)をクロロ炭酸エチル、ピバロイルクロリド、クロロ炭酸イソブチル等と反応させることにより得られる混合酸無水物等があげられる。また、この反応においては、必要に応じて塩基を添加しても良い。塩基としては、例えば、トリエチルアミン、tert-ブチルアミン、ピリジン、N-メチルモルホリン等の有機アミンがあげられ、好ましくはトリエチルアミン、ピリジンまたはN-メチルモルホリンである。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、N,N-ジメチルホルムアミド、テトラヒドロフラン、ジクロロメタンまたはクロロホルムなどが好ましい。反応温度はとくに限定されず、例えば0℃~60℃が好ましい。反応時間は、1~96時間が好ましい。
工程7においては、工程6で得られた化合物(IX)と一般式(X)で表される化合物を反応させて化合物(IV)を製造する。化合物(X)としては、一般式(XI)で表される化合物
(構造式中、X1は、塩素原子、臭素原子またはヨウ素原子を表す。)
を原料とし、ノルマルブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウムなどの塩基を用いたハロゲン-金属交換反応により調製したリチウム試薬;マグネシウム、イソプロピルマグネシウムブロミドまたはイソプロピルマグネシウムクロリドなどを用いて調製したグリニャール試薬;活性化した亜鉛、臭化亜鉛または塩化亜鉛などを用いて調製した亜鉛試薬などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、テトラヒドロフラン、ジエチルエーテル、1,4-ジオキサンまたはジメトキシエタンなどが好ましい。反応温度はとくに限定されず、例えば-100℃~室温が好ましい。反応時間は、1~24時間が好ましい。
工程8においては、工程7と同様にして、一般式(VII)で表される化合物と一般式(X)で表される化合物を反応させて化合物(IV)を製造する。
また、前記化合物(II)は、以下に示すように、スキーム3(工程9~工程11)に示される方法によって製造することができる。

工程9においては、化合物(XII)を、一般式(XIV-a)で表される中間体と反応し、化合物(II)を製造する。中間体(XIV-a)としては、酸ハライドとルイス酸から得られる活性中間体;酸無水物とルイス酸から得られる活性中間体;カルボン酸と脱水剤から得られる活性中間体があげられる。酸ハライドとしては、クロロアセチルクロリド、クロロアセチルブロミド、ブロモアセチルブロミド、ブロモアセチルクロリドまたはヨードアセチルクロリドなどがあげられる。酸無水物としては、クロロ酢酸無水物、ブロモ酢酸無水物またはヨード酢酸無水物などがあげられる。カルボン酸としては、クロロ酢酸、ブロモ酢酸またはヨード酢酸があげられる。ルイス酸としては、塩化アルミニウム、塩化亜鉛などがあげられる。脱水剤としては、五酸化リンなどがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、ジクロロメタン、ジクロロエタンなどが好ましい。また、反応溶媒を用いなくてもよい。反応温度はとくに限定されず、例えば0℃~100℃が好ましい。反応時間は、1~24時間が好ましい。
工程10においては、化合物(XII)を、一般式(XIV-b)で表される中間体と反応し、化合物(XIII)を製造する。中間体(XIV-b)としては、酢酸ハライドとルイス酸から得られる活性中間体;酢酸無水物とルイス酸から得られる活性中間体;酢酸と脱水剤から得られる活性中間体があげられる。酢酸ハライドとしては、塩化アセチル、臭化アセチルまたはヨウ化アセチルがあげられる。ルイス酸としては、塩化アルミニウム、塩化亜鉛などがあげられる。脱水剤としては、五酸化リンなどがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、ジクロロメタン、ジクロロエタンなどが好ましい。また、反応溶媒を用いなくてもよい。反応温度はとくに限定されず、例えば0℃~100℃が好ましい。反応時間は、1~24時間が好ましい。
工程11においては、化合物(XIII)を、ハロゲン化剤と反応し、化合物(II)を製造する。ハロゲン化剤としては、N-クロロコハク酸イミド、N-ブロモコハク酸イミド、N-ヨードコハク酸イミドまたはベンジルトリメチルアンモニウムトリブロミドなどがあげられる。この反応は、適当な酸を用いることで反応を促進させることができる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、テトラヒドロフラン、ジクロロメタン、ジクロロエタンなどが好ましい。反応温度はとくに限定されず、例えば0℃~100℃が好ましい。反応時間は、1~72時間が好ましい。
また、化合物(+)-(I)は、下記のスキーム4(工程12~工程15)に示される方法により製造することもできる。

工程12においては、化合物(IV)を、(R)-tert-ブタンスルフィンアミドと反応させて、化合物(XV)を製造する。この反応は、適当な酸を用いることで反応を促進させることができる。酸としては、チタンテトライソプロポキシド、チタンテトラエトキシド、チタンテトラブトキシド、パラトルエンスルホン酸などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、トルエン、キシレン、ベンゼン、テトラヒドロフラン、ジメトキシエタン、1,4-ジオキサンなどがあげられる。反応温度はとくに限定されず、例えば0℃~130℃が好ましい。反応時間は、1~72時間が好ましい。本工程において得られる化合物(XV)は、E体、Z体またはそれらの混合物として得られる。
工程13においては、化合物(XV)を、シアノ化剤と反応させて、化合物(XVI)を製造する。シアノ化剤としては、シアン化ナトリウム、シアン化カリウム、トリメチルシリルシアニドなどがあげられる。この反応は、ルイス酸や陽イオン捕捉剤を用いることで反応を促進させることができる。ルイス酸としては、トリメチルアルミニウムなどがあげられる。陽イオン捕捉剤としては、クラウンエーテル(15-クラウン-5、18-クラウン-6など)、ヘキサメチルリン酸トリアミド、N,N’-ジメチルプロピレン尿素などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、トルエン、キシレン、ベンゼン、テトラヒドロフラン、ジメトキシエタン、1,4-ジオキサンなどがあげられる。反応温度はとくに限定されず、例えば-40℃~60℃が好ましい。反応時間は、1~120時間が好ましい。
なお、場合によっては、工程12から工程13にかけては、特段、反応の後処理をせずにワンポットで実施しても良い。
工程14においては、化合物(XVI)を、酸を用いて、tert-ブチルスルフィニル基を除去した後、イソシアン酸トリクロロアセチルと反応させて、次いでトリエチルアミンを作用させて、化合物(XVII)を製造する。酸としては、塩化水素、硫酸、トリフルオロ酢酸などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、塩化メチレン、クロロホルム、トルエン、キシレン、ベンゼン、テトラヒドロフラン、ジメトキシエタン、1,4-ジオキサンなどがあげられる。反応温度はとくに限定されず、例えば0℃~60℃が好ましい。反応時間は、1~72時間が好ましい。
工程15においては、化合物(XVII)を、酸を用いて加水分解を行い、化合物(+)-(I)を製造する。酸としては、塩酸、硫酸、臭化水素酸などがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されず、例えば、塩化メチレン、クロロホルム、トルエン、キシレン、ベンゼン、テトラヒドロフラン、ジメトキシエタン、1,4-ジオキサン、水などがあげられる。また、無溶媒で行うこともできる。反応温度はとくに限定されず、例えば0℃~120℃が好ましい。反応時間は、1~72時間が好ましい。
さらに、本発明の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩は、後述する試験例3および4に示されるように経皮投与によっても優れた薬理効果や経皮吸収性を示すことから、本発明の医薬は、上記TNF-αが関与する疾患の中で皮膚に症状が現れる疾患すなわち皮膚疾患に対する予防または治療剤としてとりわけ有用である。当該皮膚疾患として、例えば、限局性強皮症、アトピー性皮膚炎、接触性皮膚炎、乾癬、ざ瘡などがあげられる。
油性基剤としては、例えば、鉱物由来のワセリンやパラフィン、ポリエチレン樹脂を流動パラフィンでゲル化したプラスチベース、生物由来のミツロウ等があげられる。
乳剤性基剤としては、例えば、ラノリン、ステアリルアルコール等があげられる。乳剤性基剤を用いる場合には、乳化剤をさらに加えることが好ましく、このような乳化剤としては、例えば、ポリオキシエチレン硬化ヒマシ油、モノステアリン酸グリセリン等があげられる。
なお、以下に示す1H-NMRスペクトルは、重クロロホルム(CDCl3)または重ジメチルスルホキシド(DMSO-d6)を溶媒とし、テトラメチルシラン(TMS)を内部標準として、JNM-ECA400型スペクトルメーター(400MHz、日本電子(株)製)で測定した。化学シフトの測定結果は、δ値をppmで示し、結合定数のJ値をHzで示した。略号のsはsinglet、dはdoublet、tはtriplet、mはmultipletを意味する。質量スペクトル(エレクトロスプレーイオン化法:ESI-MS)測定には、サーモフィッシャーサイエンティフィック(株)製Exactiveを使用した。比旋光度の測定には、旋光度計(SEPA-300型、(株)堀場製作所)を使用した。
(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン((+)-(I))の製造
(1)製造法-1(キラルカラムを用いた製造方法)による製造
(式中、アスタリスク(*)は、光学的に純粋な形態であることを意味する。)
化合物(III)(133 mg、1.2 mmol)のN,N-ジメチルホルムアミド(10mL)溶液に、炭酸セシウム(436 mg、1.3 mmol)と3’,4’-ジフルオロフェナシルブロミド(II-1)(300 mg、1.3 mmol)を加え、室温で3時間撹拌した。反応溶液を水で希釈し、析出した固体をろ取した。水で洗浄することで、化合物(IV)(収量206 mg、収率64%)を黄色固体として得た。
前記化合物(IV)(206mg、0.78mmol)、シアン化カリウム(61mg、0.94mmol)及び炭酸アンモニウム(301 mg、3.13 mmol)のエタノール(0.8 mL)懸濁液に、水(0.8 mL)を加え密閉し、100℃で67時間撹拌した。放冷後、反応液を水で希釈し、酢酸エチルで抽出した。無水硫酸ナトリウムで乾燥し、減圧下で溶媒を留去した。残渣にクロロホルムを加え、析出した固体をろ取することで、化合物(±)-(I)(収量119 mg、収率46%)を無色固体として得た。物性値を以下に示す。

なお、工程2を複数回行うことにより得られた化合物(±)-(I)を合わせ、続く工程3に供試した。
高速液体クロマトグラフ(LaChrom Elite、日立ハイテクノロジーズ(株))に分取用カラム(CHIRALPAK AD)を取り付け、ノルマルヘキサン-エタノール(30:70)をカラム温度40℃、流速8 mL/minで1時間通液し、平衡化した。前記化合物(±)-(I)(14 mg)のノルマルヘキサン-エタノール(30:70、2 mL)溶液を注入後、UV検出器を観察しながら第一ピーク(およそ12.5分後~およそ15.5分後)および第二ピーク(およそ18分後~およそ23分後)をそれぞれ分取した。同様の操作を繰り返した後、それぞれ減圧下溶媒を留去した。第一ピークを示す化合物として、本発明化合物である(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン(収量 86 mg、光学純度 >99.9%ee)を得た。なお、第二ピークを示す化合物として、本発明化合物の鏡像体である(-)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン(収量 84 mg、光学純度 99.8%ee)が得られた。

前記化合物(IV)(500 mg、1.89 mmol)及び(R)-tert-ブタンスルフィンアミド(345 mg、2.85 mmol)のトルエン(1.9 mL)溶液にチタンテトライソプロポキシド(596 μL、2.85 mmol)を加え、100℃で16時間撹拌した。室温まで放冷後、飽和塩化アンモニウム水溶液を加えて撹拌した。析出した固体をセライトろ過によりろ別し、酢酸エチルで洗浄した。ろ液と洗液の有機層を分取し、飽和食塩水で洗浄した。無水硫酸ナトリウムで乾燥し、減圧下で溶媒を留去した。残渣をカラムクロマトグラフィー(シリカゲル)で精製し、化合物(XV)(収量447 mg、収率64%)を得た。
-20℃で冷却下、前記化合物(XV)(244 mg、0.67 mmol)のトルエン(1.3 mL)溶液にトリメチルアルミニウムの1.8 mol/Lトルエン溶液(556 μL、1.00 mmol)及びトリメチルシリルシアニド(126 μL、1.00 mmol)を加え、-20℃で2時間撹拌した。飽和塩化アンモニウム水溶液を加え、析出した固体をセライトろ過によりろ別し、酢酸エチルで洗浄した。ろ液と洗液の有機層を分取し、飽和食塩水で洗浄した。無水硫酸ナトリウムで乾燥し、減圧下で溶媒を留去した。残渣をカラムクロマトグラフィー(シリカゲル)で精製し、化合物(XVI)(収量145 mg、収率55%)を得た。

氷冷下、前記化合物(XVI)(130 mg、0.33 mmol)の酢酸エチル(3.3 mL)溶液に4 mol/L 塩化水素-酢酸エチル溶液(1.5 mL)を加え、30分間撹拌した。反応液を減圧濃縮した後、塩化メチレン(3.3 mL)に溶解し、イソシアン酸トリクロロアセチル(47 μL、0.40 mmol)を加え、室温下で40分間撹拌した。トリエチルアミン(46 μL、0.33 mmol)を加え、室温下で2時間撹拌した。反応液を減圧濃縮し、残渣をカラムクロマトグラフィー(シリカゲル)で精製することにより、化合物(XVII)(収量142 mg、収率90%)を得た。

前記化合物(XVII)(125 mg、0.26 mmol)に6 mol/L塩酸(5.2 mL)を加え、100℃で16時間撹拌した。室温まで放冷後、反応液を水で希釈し、酢酸エチルで2回抽出した。有機層をあわせて飽和食塩水で洗浄し、無水硫酸ナトリウムで乾燥した。減圧下で溶媒を留去し、残渣をカラムクロマトグラフィー(シリカゲル)で精製することで、化合物(+)-(I)(収量80 mg、収率92%、光学純度97.6%ee)を無色固体として得た。

試験例1
TACE阻害実験(インビトロ)
TACEの塩基配列はモス(Moss)らにより報告されている(Moss, M. L. et al., Nature 1997, 385, 733-736)。したがって、THP-1細胞などから定法にしたがってTACEのcDNAを取得し、これを発現ベクターに組み込み、次いでこのベクターを哺乳動物細胞あるいは昆虫細胞に形質転換し、TACEを発現取得した。
TACE阻害実験は、前記のようにして得られたTACEを酵素として用い、また基質としては、膜結合型TNFのTACE切断配列を含んだ蛍光合成基質 Nma(N-メチルアントラニル酸)-Leu-Ala-Gln-Ala-Val-Arg-Ser-Ser-Lys(Dnp(ジニトロフェニル))-D-Arg-NH2 を用いて、被験物質存在下または非存在下におけるTACEの活性を測定することにより行った。以下にTACE阻害実験の方法を示した。
すなわち、アッセイバッファーA(200mM塩化ナトリウム、5mM塩化カルシウム、10μM硫酸亜鉛、2mg/mL牛血清アルブミンを含む、50mMトリス-塩酸緩衝液(pH7.5))にて調製した酵素液90μL、及びアッセイバッファーB(200mM塩化ナトリウム、5mM塩化カルシウム、10μM硫酸亜鉛、0.05%PLURONIC F-68を含む、50mM トリス-塩酸緩衝液(pH7.5))にて20μMに調製した蛍光合成基質90μLを混合し、37℃で1.5時間反応させた。この後、蛍光強度計(Labsystems, Fluoroskan Ascent)にて、励起波長355nm、測定波長460nmの条件で測定し、酵素活性を求めた。
被験物質の存在下及び非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
その結果、本発明の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンは、IC50値が100nM以下のTACE阻害活性を示した。
MMP阻害実験
MMP阻害実験は、例えばビケット(Bickett)ら(D. Mark Bickett et al., Anal. Biochem., 1993, 212, 58-64)及びナガセ(Nagase)ら(H. Nagase et al., J. Biol. Chem., 1994, 269, 20952-20957)の方法に準じて、蛍光合成基質を用いて行うことができる。以下に各MMPの阻害実験の方法を示した。
ヒトMMP-1(Calbiochem#444208)180μL(100ng)は、10mMのp-アミノフェニル水銀アセテート(APMA)20μLと混和し、37℃で1時間反応させることによって活性化した。この酵素液20μLをアッセイバッファーAにて90μLに希釈し、これをアッセイバッファーBにて調製した20μMの蛍光基質(Dnp-Pro-Cha(β-シクロヘキシルアラニル)-Gly-Cys(Me)-His-Ala-Lys(Nma)-NH2)90μLに添加して、37℃で5時間反応させた。この後、蛍光強度計(Labsystems, Fluoroskan Ascent)にて、励起波長355nm、測定波長460nmの条件で測定し、酵素活性を求めた。
被験物質の存在下及び非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
ヒトMMP-2(Calbiochem#444213)90μL(5ng)は、10mMのAPMA10μLと混和し、37℃で1時間反応させることによって活性化した。この酵素液10μLをアッセイバッファーAにて90μLに希釈し、これをアッセイバッファーBにて調製した20μMの蛍光基質
(MOCAc((7-methoxycoumarin-4-yl)acetyl)-Pro-Leu-Gly-Leu-A2pr(Dnp)-Ala-Arg-NH2、ペプチド研究所#3163-v)90μLに添加して、37℃で5時間反応させた。この後、蛍光強度計(Labsystems、Fluoroskan Ascent)にて、励起波長320nm、測定波長405nmの条件で測定し、酵素活性を求めた。
被験物質の存在下及び非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
アッセイバッファーAにて調製したヒトMMP-3(Calbiochem#444217)90μL(1.5ng)を、アッセイバッファーBにて調製した20μMの蛍光基質NFF-3(MOCAc((7-methoxycoumarin-4-yl)acetyl)-Arg-Pro-Lys-Pro-Val-Glu-Nva-Trp-Arg-Lys(Dnp)-NH2、ペプチド研究所#3168-v)90μLに添加して、37℃で4時間反応させた。この後、蛍光強度計(Labsystems, Fluoroskan Ascent)にて、励起波長320nm、測定波長405nmの条件で測定し、酵素活性を求めた。

被験物質の存在下及び非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
本試験により得られた、本発明の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンのMMPsに対する50%阻害濃度を表1に示す。
マウスTPA(12-O-テトラデカノイルホルボール-13-アセタート)単回塗布により誘発される耳介浮腫の抑制実験(TNF-αが関与する皮膚炎症モデルでのインビボ薬効試験)
BALB/cマウスの左耳の内外両側に、TPAの54μmol/Lアセトン溶液をそれぞれ10μLずつ塗布(1.08nmol TPA/耳介)して、耳介浮腫を誘発した。非誘発群には、TPAの54μmol/Lアセトン溶液の代わりにアセトンを同様に塗布した。被験物質は、10vol% DMSO含有アセトン(経皮投与媒体)で被験物質の1w/v%溶液を調製し、TPA塗布前1時間にマウス左耳の内外両側にそれぞれ10μLずつ塗布した。対照群には、被験物質の溶液の代わりに経皮投与媒体を同様に塗布した。エタネルセプト群には、TPA塗布前日及び塗布前1.5時間に5mg/mLのエタネルセプト溶液を0.2mL静脈内投与(1mg/匹)した。ヒトIgG(hIgG)群(エタネルセプトの対照群)には5mg/mLのhIgG溶液を0.2mL静脈内投与(1mg/匹)した。TPA塗布前日及び塗布後6時間に、エーテル麻酔下で耳介厚さを測定し、耳介厚さの増加量を指標に被験物質の耳介浮腫抑制作用を評価した。
被験物質の耳介浮腫抑制率(%)=(C-A)/(C-B)×100
A:被験物質を投与した群の耳介厚さの増加量の平均値
B:非誘発群の耳介厚さの増加量の平均値
C:対照群の耳介厚さの増加量の平均値
エタネルセプトの耳介浮腫抑制率(%)は、非誘発群の耳介厚さの増加量の平均値(B)、エタネルセプト群の耳介厚さの増加量の平均値(D)及びhIgG群の耳介厚さの増加量の平均値(E)を用い、次式により算出した。
エタネルセプトの耳介浮腫抑制率(%)=(E-D)/(E-B)×100
D:エタネルセプト群の耳介厚さの増加量の平均値
E:hIgG群の耳介厚さの増加量の平均値
また、被験物質の耳介浮腫抑制率(%)を、陽性対照とした同一試験中でのエタネルセプトの耳介浮腫抑制率(%)と比較して、次式によりエタネルセプト比を算出した。

本発明の化合物は、経皮投与によって、すでに上市されているTNF-αを介する疾患用の薬剤であるエタネルセプトの静脈内投与よりも優れた効果を示した。
ヘアレスマウス 経皮投与PK試験
静脈内投与方法
ジエチルエーテル麻酔下のヘアレスマウス(摂食)に尾静脈より被験物質(0.1~0.5mg/5mL/kg)を単回静脈内投与した。
経皮投与方法
ジエチルエーテル麻酔下のヘアレスマウス(摂食)の背部皮膚に油性ペンを用いて投与部位4cm2(2cm×2cm)をマーキングした。投与部位に被験物質50μL/animal(1w/v%マクロゴール400溶液)を塗布した。約2cm×2cmのガーゼ(BEMCOT(登録商標))を約4cm×4cmのポリエチレンシートに両面テープを用い固定し、ガーゼ面を被験物質塗布面の上に被せる。その上に粘着性伸縮包帯(エラストポア、約10cm)を貼付し、被験物質塗布面を固定、保護した。その後ケージへ戻し個別飼育を行う。投与後24時間に、閉塞塗布が正常に行われていることを確認した。
採血方法
無麻酔下のマウスの尾静脈をカミソリで切り、マイクロピペットを用いて尾静脈から採血した。静脈内投与の採血時点は投与後5分、15分、30分、1時間、3時間および6時間とし、各2匹のマウスから採血を行った。経皮投与の採血時点は投与後30分、1時間、3時間、6時間および24時間とし、各2匹のマウスから採血を行った。各時点での採血量はそれぞれ約30~50μLとした。血液はヘパリンナトリウム(1000単位/mL)2μLを添加したチューブに移し、遠心分離(4℃、19,200×g、10min)して血漿を得た。血漿は設定温度-30℃のフリーザー内で凍結保存した。
血漿中の被験物質濃度の測定方法
前記方法によって得た凍結保存した血漿は、室温で解凍し、メタノールを用いて除タンパクしたのち、被験物質の血漿中濃度を測定した。血漿中濃度の測定には、CTC Analytics製HTC PALハイスループットLC注入システム、サーモフィッシャーサイエンティフィック(株)製AccelaおよびTSQ Quantum Ultraを用いた。
経皮吸収率の算出方法
前記方法によって得た血漿中の被験物質濃度より、AUC(血漿中濃度曲線下面積)を算出し、以下の式を用いて経皮吸収率を算出した。
経皮吸収率(%)=((Div×AUCpc)/(Dpc×AUCiv))×100
Div:静脈内投与時の被験物質の投与量
Dpc:経皮投与時の被験物質の投与量
AUCiv:静脈内投与後の被験物質の血漿中濃度曲線下面積
AUCpc:経皮投与後の被験物質の血漿中濃度曲線下面積
日本薬局方崩壊試験液第2液に対する溶解性試験
検量線用試料溶液の調製
被験物質10.0mgを正確に量りとり、メタノールを加え10mLとした後、10倍希釈し、100μg/mLの溶液を調製する。
溶解度測定用試料溶液の調製
遠沈管に被験物質5.0mgを量り、日本薬局方崩壊試験液第2液5.0mLを加え、3分間超音波をかけた後、2時間振とうする。この溶液をメンブランフィルター(0.2μm)でろ過後、正確にろ液を500μL量りとり、アセトニトリル500μLを加えて良く撹拌し、試料溶液とする。
日本薬局方崩壊試験液第2液(PH6.8)の調製方法
0.2mol/Lリン酸二水素カリウム水溶液250mLに0.2mol/L水酸化ナトリウム水溶液118mL及び水を加えて1000mLに調整する。
日本薬局方崩壊試験液第2液に対する溶解度の測定方法
高速液体クロマトグラフィーを用いて、検量線用試料溶液および溶解度測定用試料溶液を各2回測定し、平均面積をそれぞれ算出する。
日本薬局方崩壊試験液第2液に対する溶解度の算出方法
前記方法によって得たそれぞれの平均面積を基に、以下の式を用いて溶解度を算出した。
日本薬局方崩壊試験液第2液に対する溶解度(μg/mL)=((P×2)/S)×100
S:検量線用試料溶液の平均面積
P:溶解度測定用試料溶液の平均面積
上記試験により、本発明化合物はそのラセミ体と比較して、日本薬局方崩壊試験液第2液に対してはるかにより高い溶解度を有することが確認された。よって、本発明化合物は、製剤を調製するにあたって、優位な性質を有することが明らかになった。
有機溶媒に対する溶解性試験
室温条件下、0.5mLポリプロピレン製チューブ中に予め秤量した適量の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン及び(±)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンを用意し、それぞれ種々の重量(例えば、被験化合物に対して、10倍量、20倍量、100倍量など)の溶媒(プロピレングリコール(PG)またはブチレングリコール(BG))を添加し、ボルテックスミキサーで撹拌した。37℃設定の卓上型超音波洗浄機で10分間超音波処理した。80℃設定の恒温槽で1時間温めた後、室温にて24時間静置した。試験液を目視で観察し、その状態を下記判定基準に従って評価した。
溶媒に対する溶解性の判定基準
判定基準は、
「Soluble」:被験化合物が溶解し、不溶物が目視で確認されない状態
「Insoluble」:被験化合物が溶け残り、不溶物が目視で確認される状態
の2段階とした。
軟膏1g中における用量
(1)化合物(+)-(I) 10mg
(2)白色ワセリン 990mg
水浴上で(1)および(2)を加温融和させ(80℃)、固まるまで冷却しながらかき混ぜて製する。
製剤例2(3w/w%軟膏剤)
軟膏1g中における用量
(1)化合物(+)-(I) 30mg
(2)白色ワセリン 970mg
水浴上で(1)および(2)を加温融和させ(80℃)、固まるまで冷却しながらかき混ぜて製する。
クリーム1g中における用量
(1)化合物(+)-(I) 10mg
(2)白色ワセリン 250mg
(3)ステアリルアルコール 200mg
(4)プロピレングリコール 120mg
(5)ポリオキシエチレン硬化ヒマシ油 40mg
(6)モノステアリン酸グリセリン 10mg
(7)パラオキシ安息香酸メチル 1mg
(8)パラオキシ安息香酸プロピル 1mg
(9)精製水 適量
水浴上で(1)から(3)を加温融和させ(80℃)、これにあらかじめ(4)から(8)を精製水に溶かして80℃に加温した液を加え、均一になるように攪拌し、固まるまで冷却しながらかき混ぜて製する。
製剤例4(3w/w%クリーム剤)
クリーム1g中における用量
(1)化合物(+)-(I) 30mg
(2)白色ワセリン 250mg
(3)ステアリルアルコール 200mg
(4)プロピレングリコール 120mg
(5)ポリオキシエチレン硬化ヒマシ油 40mg
(6)モノステアリン酸グリセリン 10mg
(7)パラオキシ安息香酸メチル 1mg
(8)パラオキシ安息香酸プロピル 1mg
(9)精製水 適量
水浴上で(1)から(3)を加温融和させ(80℃)、これにあらかじめ(4)から(8)を精製水に溶かして80℃に加温した液を加え、均一になるように攪拌し、固まるまで冷却しながらかき混ぜて製する。
錠剤中における用量
(1)化合物(+)-(I) 100mg
(2)乳糖 308mg
(3)カルボキシメチルセルロースカルシウム 30mg
(4)ヒドロキシメチルセルロース 7mg
(5)ステアリン酸マグネシウム 5mg
(6)結晶セルロース 50mg
(1)から(4)および(6)を均一になるように混合し、造粒および篩過を行う。さらに(5)を加えて混合した後、打錠し製する。
液1g中における用量
(1)化合物(+)-(I) 10mg
(2)エタノール 500μg
(3)グリセリン 200μg
(4)プロピレングリコール 200μg
(5)精製水 適量
(1)から(5)をかき混ぜて製する。
製剤例7(3w/w%液剤)

Claims (21)
- (+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩。
- 請求項1に記載の化合物またはその塩を有効成分として含有する医薬。
- (-)異性体を実質的に含まない請求項2に記載の医薬。
- 前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の95重量%以上が(+)異性体である請求項2または3に記載の医薬。
- 前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の98重量%以上が(+)異性体である請求項4に記載の医薬。
- 前記医薬中、5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩の全量の99.5重量%以上が(+)異性体である請求項5に記載の医薬。
- 外用製剤である請求項2から6のいずれかに記載の医薬。
- 前記外用製剤がローション剤、クリーム剤、液剤および軟膏剤からなる群から選択される剤形である請求項7に記載の医薬。
- 薬学的に許容されるグリコール類を含む請求項7または8に記載の医薬。
- 薬学的に許容されるグリコール類がプロピレングリコールまたはブチレングリコールである請求項9に記載の医薬。
- 製剤中に水を含む請求項7から10のいずれかに記載の医薬。
- 有効成分として、1w/w%以上の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩を含有する請求項7から11のいずれかに記載の医薬。
- 有効成分として、3w/w%以上の(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンまたはその塩を含有する請求項12に記載の医薬。
- 経皮投与により投与される請求項2から13のいずれかに記載の医薬。
- 皮膚疾患の予防または治療剤である請求項2から14のいずれかに記載の医薬。
- 皮膚疾患が、限局性強皮症、アトピー性皮膚炎、接触性皮膚炎、乾癬およびざ瘡からなる群より選択される1つ以上の疾患である請求項15に記載の医薬。
- ラセミ体の(±)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンを光学分取用のカラムを用いたクロマトグラフィーによって光学分割することを含む、(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンの製造方法。
- ラセミ体の(±)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンを光学分取用のカラムを用いたクロマトグラフィーによって光学分割して得られる、(+)-5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオン。
- 5-(3,4-ジフルオロフェニル)-5-[(3-メチル-2-オキソピリジン-1(2H)-イル)メチル]イミダゾリジン-2,4-ジオンの(+)及び(-)異性体の混合物であって、(+)異性体が(-)異性体に対して90%ee以上で存在する混合物。
- 前記混合物のうち、(+)異性体が(-)異性体に対して96%ee以上で存在する請求項19に記載の混合物。
- 前記混合物のうち、(+)異性体が(-)異性体に対して99%ee以上で存在する請求項19または20に記載の混合物。
