Definitions
- the present technology relates to a flow path device for circulating a fluid containing a sample, an analysis apparatus using the flow path device, and a fluid apparatus.
- Patent Document 1 discloses a cartridge used for performing a chemical reaction of a sample.
- This cartridge is composed of a rigid substrate and an elastic container. In the container, a flow path and two or more chambers connected by the flow path are formed.
- this cartridge has a chamber for storing two kinds of solutions, a reaction chamber in which a reaction occurs when they are mixed, and a waste liquid storage chamber.
- the roller or the like is moved while rotating and an external force is applied to the elastic body of the cartridge, the elastic body is crushed and the solution in the flow path or the chamber moves (for example, the paragraph in the specification of Patent Document 1). [0013], [0016], see FIGS. 2 and 3 etc.).
- An object of the present technology is to provide a flow channel device suitable for sample injection and recovery, a sorting device and a fluid device using the flow channel device.
- a flow channel device includes a container layer and a flow channel layer.
- the container layer has an injection container part into which a fluid containing a sample is injected, and a recovery container part that recovers the fluid.
- the channel layer has a channel connected to the injection container part and the recovery container part, and is joined to the container layer.
- the flow path layer is joined to the container layer, and the injection container part and the recovery container part are provided in the same container layer, the injection and recovery of the fluid containing the sample is simplified and the workability is improved.
- the injection container part may be formed by a through-hole provided in the container layer with the surface of the flow path layer as a bottom, or may include a recess provided in the container layer.
- the collection container part may be formed by a through hole provided in the container layer with the surface of the flow path layer as a bottom, or may include a recess provided in the container layer.
- the collection container may have a concave reservoir provided in the through hole or the recess.
- the recovery container part may have a recovery container inflow passage connected to a region outside the reservoir part in the through hole or the recess.
- the flow path of the flow path layer may further include a recovery container outflow path connected to the flow path, provided at a position facing the recovery container inflow path. Since the fluid flowing at a speed higher than the speed of the settling sample easily proceeds toward the recovery container outflow path provided at a position facing the recovery container inflow path, it can smoothly flow out from the recovery container outflow path.

At this time, the valve connected to the port C5 remains closed, but the flow control mechanism 76 may also open the valve connected to the port C5 at or immediately after opening the valve of the port C4 (FIGS. 14A and 14B). reference).
- the fluid flowing through the second flow path 12 is indicated by a thick white arrow in FIG.
- This fluid merges with the fluid flowing through the first flow path 11 on the downstream side of the constriction path K. That is, the flow of the fluid in the second flow path 12 including the velocity component (horizontal velocity component, here the x component) perpendicular to the flow direction (z direction) of the fluid containing particles in the constriction path K ( Cross flow) is formed.
- the flow mode after the second channel 12 is governed by the cross flow, and the flow of particles in the second channel 12 can be stabilized.
- the particles that have exited the constriction path K quickly leave the constriction path K and flow through the second flow path 12, so that the particles can be prevented from returning to the vicinity of the constriction path K.
- the flow rate ratio in the first flow path 11 and the second flow path 12 is set, for example, in the range of 1: 5 to 1: 1000.
- the processing unit 70 uses the measurement voltage signal of the measurement electrode unit 25 to measure the amount of electricity of the fluid containing the particles in the constriction path K. Then, the type and state of the particles are specified.
- the measurement accuracy of each particle in the measurement electrode unit 25 can be increased.
- the processing unit 70 determines whether to generate and output an action voltage signal according to the type and state of the identified particle.
- the action signal generation unit 73 generates an action voltage signal, which is generated in the working electrode unit 29 (see FIGS. 7 and 8). Apply. Then, in the region where the working electrode portion 29 is disposed in the second flow path 12, a migration force that applies a component force in the y direction to the particles is applied to the particles and guided to the branch path 16.
- the determination unit 72 determines that the identified particle is not a particle to be accumulated in the collection container unit 53, the determination unit 72 does not generate an action voltage signal and the particle is guided to the branch path 17.
- the structure and operation of the second flow path 12 are such that the particles are guided to the branch path 17 by generating and outputting an action voltage signal.
- the structure or the like of the electrode part 29 may be designed.
- a pressure tank that is a part of the flow control mechanism 76 is connected to the ports C4 and C5, for example.
- the flow control mechanism 76 can increase the stability of the flow rate ratio of the branch paths 16 and 17 by controlling the pressure ratio between these pressure tanks.
- FIG. 13 shows a cross section at the collection container 53, and the outflow path (second outflow path) connected to the branch path 17 and the port C4 is not drawn.
- the branch paths 16 and 17 overlap with each other as seen in FIG. 13, and the second outflow path and the recovery container inflow path 54 overlap. Therefore, in order to make FIGS. 13 and 14 easier to understand, the outflow path of the fluid from the second outflow path is drawn by a broken line white arrow by shifting the outflow path from the recovery container inflow path 54.
- the particles guided to the branch path 16 flow into the recovery container portion 53 via the recovery container inflow path 54.
- region outside the reservoir part 53a which is an exit of the collection container inflow path 54 is shallower than the reservoir part 53a. It is desirable that the cross-sectional area of the flow channel immediately after the outlet of the recovery container inflow passage 54 is several tens of times the cross-sectional area of the outlet of the recovery container inflow passage 54.
- the velocity component of the particles flowing out from the recovery container inflow passage 54 is larger in the gravitational direction than the fluid flow direction toward the recovery container outflow passage 55, that is, in the x direction. Since the speed of the particles decreases sufficiently, the particles settle toward the reservoir 53a. In this way, the particles can be efficiently stored in the reservoir 53a.
- recovery container outflow path 55 is disposed at a position facing the recovery container inflow path 54, the fluid that has flowed into the recovery container section 53 from the recovery container inflow path 54 toward the recovery container outflow path 55. Since it advances easily, it can flow out smoothly from the collection container outflow passage 55.
- the operation by the flow control mechanism 76 is stopped, and the flow of the fluid in the flow channel device 100 is stopped.
- the operator peels off the sealing material S1 from the container layer 50 and uses a device such as a pipette 60 to store the fluid containing particles at a high concentration stored in the reservoir 53a of the recovery container 53. Can be taken out.
- the flow path device 100 As shown in FIGS. 14A and 14B, when particles are clogged in the narrow path K or the flow path in the vicinity thereof, the flow path is closed as shown in FIGS. 17A and 17B.
- the clogging can be released by the control mechanism 76 temporarily opening the port C3.
- the electric means as described above, that is, the processing unit 70 measures the amount of electricity, whereby the clogging of the particles can be detected.
- the processing unit 70 continuously obtains a higher resistance value than usual, and therefore can detect that the particles are clogged.
- the clogging of particles can be detected not only by electrical means but also by optical means such as photographing near the narrow path K by a camera or analyzing reflected light or scattered light near the narrow path K by light irradiation. .
- the flow channel layer 40 is joined to the container layer 50, and the injection container 51 and the recovery container 53 are provided in the same container layer 50. Fluid injection and recovery are simplified and workability is improved.
- the inlet ports C1 and C2 and the outlet ports C3 to C5 connected to the flow control mechanism 76 are provided on the surface of the container layer 50, the operator can connect the flow control mechanism 76 and the flow channel device. 100 can be easily mechanically connected.
- the containers for injecting or collecting the samples are individually made, and are made separately from the devices constituting the flow path.
- the injection container unit 51, the flow channel of the flow channel layer 40, and the recovery container unit 53 are integrally configured, and this single flow channel device 100 is from the injection to the recovery of the sample. It is completed with.
- the flow channel device 100 can be reused after being cleaned, sterilized, or the like.
