Description
本発明は、L-ヒスチジンを含有する水性液体組成物、該液体組成物の製造方法、及び該液体組成物の用途などに関する。
近年、アミノ酸の様々な生理機能が明らかとなっており、アミノ酸を含有する飲食品等が多数上市されている。アミノ酸を含有する飲料や液剤では、高濃度でアミノ酸を含有する場合に析出や沈殿が起こることが知られており、アミノ酸を高濃度で含有する飲食品の製造方法について研究開発が行われている。例えば、特許文献1には、pH3.0~6.0に調整した40~100℃の温水に、高濃度のアミノ酸を溶解させることを特徴とする、アミノ酸含有食品の製造方法が開示されている。該文献には、アミノ酸を0.1~20重量%の範囲で含有する食品を製造できることが記載されており、実施例にはアミノ酸濃度が1.5重量%のアミノ酸混合物を含有する飲料が開示されている。また特許文献2には、イソロイシン、ロイシン及びバリンからなる3種の分岐鎖アミノ酸を有機酸及び/又は無機酸の存在下に水に溶解させることを特徴とする、当該分岐鎖アミノ酸を高濃度で含有する、医薬用又は健康飲料用の液剤の製造方法が開示されている。該文献の実施例には、分岐鎖アミノ酸の濃度が4.0g/61~94mLの液剤が開示されている。
必須アミノ酸の一つであるヒスチジンは、食品添加物としても認められており、多くの食品に含まれている。ヒスチジンの生理機能としては、例えば抗酸化作用、肥満予防等が知られている。ヒスチジンを含む組成物として、特許文献3には、固形分100に対するヒスチジンまたはヒスチジン塩酸塩の配合比率が15~95重量部で、ヒスチジン換算1重量%水溶液のpHが5.0~7.5になるようにpH調整剤または/および矯味剤が含有されている組成物が開示されている。
アミノ酸を含有する飲食品、サプリメント等を製造する場合、高濃度のアミノ酸を含有する溶液を調製することができれば、高用量のアミノ酸を容易に摂取できる、従来にない形態の飲食品、サプリメント等を製造することが可能となる。また溶液中のアミノ酸濃度は高いほど好ましく、それにより製造上の取り扱い易さの向上、輸送コストの低減なども期待される。
しかしながらヒスチジンに関しては、水への溶解性が低いため高濃度の水溶液を調製できないという問題があった。
ところで、前記特許文献1には0.1~20重量%のアミノ酸を含有する食品の製造方法が記載されている。しかしながら、当該文献における唯一の実施例においては、17種のアミノ酸が配合されており、その濃度は17種のアミノ酸濃度として1.5重量%に過ぎず、しかも配合された17種アミノ酸の全量100重量部中、ヒスチジン配合量は僅か2重量部に過ぎない。
しかしながらヒスチジンに関しては、水への溶解性が低いため高濃度の水溶液を調製できないという問題があった。
ところで、前記特許文献1には0.1~20重量%のアミノ酸を含有する食品の製造方法が記載されている。しかしながら、当該文献における唯一の実施例においては、17種のアミノ酸が配合されており、その濃度は17種のアミノ酸濃度として1.5重量%に過ぎず、しかも配合された17種アミノ酸の全量100重量部中、ヒスチジン配合量は僅か2重量部に過ぎない。
そこで本発明は、高用量での摂取に好適な、高濃度でヒスチジンを含有する水性液体組成物、該液体組成物の製造方法、及び該液体組成物の用途などを提供することを目的とする。
本発明者らは、上記目的を達成すべく鋭意検討した結果、ヒスチジンを20重量%を超える濃度で含有する液体組成物を調製することに初めて成功した。
塩基性アミノ酸であるヒスチジンは、酸性下で水溶性が上昇するが、意外にも、酸性物質として2価以上の有機酸を含有する酸性の水溶液中では、飲食品に適用可能な他の酸性物質(例えば、酢酸などの1価の有機酸、リン酸などの無機酸など)を含有する酸性の水溶液よりも、同程度のpHであってもヒスチジンの水溶性が顕著に高くなることを見出した。
本発明者らは、かかる知見に基づいて更に研究を重ねた結果、本発明を完成するに至った。
塩基性アミノ酸であるヒスチジンは、酸性下で水溶性が上昇するが、意外にも、酸性物質として2価以上の有機酸を含有する酸性の水溶液中では、飲食品に適用可能な他の酸性物質(例えば、酢酸などの1価の有機酸、リン酸などの無機酸など)を含有する酸性の水溶液よりも、同程度のpHであってもヒスチジンの水溶性が顕著に高くなることを見出した。
本発明者らは、かかる知見に基づいて更に研究を重ねた結果、本発明を完成するに至った。
即ち、本発明は以下の通りである。

本発明により提供される水性液体組成物は、従来公知のものよりも高濃度のヒスチジンを含有するため、少量で高用量のヒスチジンの摂取が可能な飲食品、サプリメント等の提供が可能となる。また当該水性液体組成物の成分はいずれも経口摂取に適しており、安全性に優れた飲食品、サプリメント、医薬品等の提供が可能となる。
本発明の水性液体組成物は、飲食品加工原料として使用する場合には、ヒスチジンを原料中に添加するときに粉体を用いるよりも原料中に容易に分散させやすいので加工が容易になる。
また飲食品や医薬品の製造工程で、原料中に本発明の水性液体組成物として添加することで、加熱や乾燥により水分を減量してもヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることがなく、またヒスチジンが析出することにより口当たりのざらつき等が発生することがなく製品の品質を一定にすることができる。
さらに本発明の水性液体組成物は冷蔵または冷凍下に静置しても、ヒスチジンが析出することがないため、飲食品や医薬品の原料として使用することで、その製造及びその保管工程で冷蔵や冷凍を経ても、製品中でヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることなく、またヒスチジンが析出することによる口当たりのざらつき等が発生することもなく、優れた品質を保つことができる。
本発明の水性液体組成物は、飲食品加工原料として使用する場合には、ヒスチジンを原料中に添加するときに粉体を用いるよりも原料中に容易に分散させやすいので加工が容易になる。
また飲食品や医薬品の製造工程で、原料中に本発明の水性液体組成物として添加することで、加熱や乾燥により水分を減量してもヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることがなく、またヒスチジンが析出することにより口当たりのざらつき等が発生することがなく製品の品質を一定にすることができる。
さらに本発明の水性液体組成物は冷蔵または冷凍下に静置しても、ヒスチジンが析出することがないため、飲食品や医薬品の原料として使用することで、その製造及びその保管工程で冷蔵や冷凍を経ても、製品中でヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることなく、またヒスチジンが析出することによる口当たりのざらつき等が発生することもなく、優れた品質を保つことができる。
本発明は、ヒスチジンを20重量%を超える濃度で含有する水性液体組成物、特に2価以上の有機酸を更に含有する水性液体組成物(以下、本発明の水性液体組成物とも称する)、これを含む飲食品及び当該液体組成物の製造方法(以下、本発明の製造方法とも称する)を提供する。
本発明において、水性液体組成物における水性液体とは、水を含む液体を意味し、水のみよりなる液体の他、水を主成分とし他の溶媒を含むものを包含する概念である。例えば、水、含アルコール水等が例示され、通常水を1~100重量%含有するものである。

本発明の水性液体組成物に含まれるヒスチジンは、その由来及び製法に制限はなく、天然に存在する動植物等から抽出し精製したもの、或いは化学合成法、発酵法、酵素法又は遺伝子組換え法によって得られるもののいずれを使用してもよい。またヒスチジンは、L-体、D-体又はDL-体のいずれも使用することができるが、L-体が好適に使用される。またヒスチジンは、遊離体または塩(塩酸塩等)を用いることができるが、遊離体が好適に用いられ、酸性の水性溶液に添加され溶解される。
本発明の水性液体組成物に含まれるヒスチジンの濃度は、20重量%を超える濃度、例えば20.5重量%以上、好ましくは28重量%以上、より好ましくは30重量%以上、より好ましくは36重量%以上、更により好ましくは40重量%以上、更により好ましくは45重量%以上である。ヒスチジンの濃度が20重量%を超える濃度であることにより、所望の量のヒスチジンを摂取するために必要な水性液体組成物の量を下げることができ、効率よくヒスチジンを摂取することが可能となる。
またヒスチジンの濃度の上限値は、ヒスチジンが溶解する限り特に限定されないが、通常60重量%以下、好ましくは57重量%以下である。ヒスチジン濃度が60重量%以下であれば、ヒスチジンの析出や沈殿がみられず、良好な液体組成物を得ることができる。

また本発明の水性液体組成物において、水含量に対するヒスチジン含量の割合(対水ヒスチジン含量%(W/W))は、ヒスチジンが溶解する限り特に限定されないが、通常60~900%程度の範囲で適宜設定することができる。
本発明の水性液体組成物は、例えば、2価以上の有機酸を含有する酸性の水性溶液に、ヒスチジンを溶解させることによって調製することができる。
従って、本発明の水性液体組成物は、通常ヒスチジンの他に2価以上の有機酸を含有するものである。
2価以上の有機酸は、由来及び製法に制限はない。2価以上の有機酸としては、例えば、2価の有機酸(例えば、リンゴ酸、酒石酸、コハク酸、フマル酸など)、3価の有機酸(例えば、クエン酸など)などが挙げられるがこれらに限定されない。2価以上の有機酸は、これらから1種又は2種以上を選択して使用することができる。当該有機酸は3価以下が好ましい。2価以上の有機酸としては、好ましくはクエン酸、リンゴ酸及び酒石酸のいずれか1種以上である。かかる2価以上の有機酸をさらに含有することにより、経口摂取に好適な、高濃度のヒスチジンを含有する水性液体組成物の製造が可能となる。
従って、本発明の水性液体組成物は、通常ヒスチジンの他に2価以上の有機酸を含有するものである。

本発明の水性液体組成物中の2価以上の有機酸の含有量は、前記濃度範囲のヒスチジンが水性液体組成物中に溶解した状態を維持することが可能な範囲であれば特に限定されない。ここで溶解した状態とは、20℃で24時間静置した場合においてヒスチジンが析出又は沈殿していない状態を意味する。2価以上の有機酸の含有量は、水性液体組成物中のヒスチジンの濃度をA重量%としたときに、水性液体組成物中のヒスチジン1重量部に対して(0.0109A+0.0511)重量部以上である。本願実施例に示すデータより、水性液体組成物中のヒスチジンの濃度と、該ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能な2価以上の有機酸の含有割合(対ヒスチジン)の下限値との間には、上記関数で表される関係があることが見出された。即ち、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量が、(0.0109A+0.0511)重量部以上であれば、20重量%を超える濃度のヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が20.5重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.27重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が30重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.38重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が40重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.49重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が45重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.54重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が20.5重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.27重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が30重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.38重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が40重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.49重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
例えば、水性液体組成物中のヒスチジンの濃度が45重量%である場合、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量を、0.54重量部程度以上にすれば、ヒスチジンが水性液体組成物中に溶解した状態を維持することが可能である。
また、本発明の水性液体組成物中のヒスチジンの含有量を高める観点から、2価以上の有機酸の含有量は、ヒスチジン1重量部に対して、通常2.0重量部以下、好ましくは1.2重量部以下、より好ましくは1.0重量部以下、更により好ましくは0.8重量部以下である。尚、ヒスチジンと2価以上の有機酸の含有量は、本発明の水性液体組成物中のそれらの合計濃度が100重量%未満となる範囲で適宜設定される。
本発明の水性液体組成物は、必要に応じて、ヒスチジン及び有機酸以外の他の成分を含んでもよい。他の成分としては、飲食品、医薬品等の製造に際して通常使用される原料が挙げられ、特に制限されないが、例えば、増粘剤、甘味剤、矯味剤、保存剤、香料、賦形剤、安定化剤等が挙げられる。
増粘剤の例としては、デキストリン、アルギン酸ナトリウム、アルギン酸プロピレングリコールエステル、トラガント末、キサンタンガム、カルボキシメチルセルロースナトリウム、ヒドロキシプロピルセルロース、ポリビニルアルコール、ポリビニルピロリドン等の高分子が挙げられる。
甘味剤の例としては、ブドウ糖、果糖、転化糖、ソルビトール、キシリトール、グリセリン、単シロップなどが挙げられる。また他の成分の含有量を減らして高濃度でヒスチジンを提供するため、高甘味度甘味料を用いることもできる。高甘味度甘味料としては、例えば、アスパルテーム(α-L-アスパラチルフェニルアラニンメチルエステル)、アセスルファムK(6-メチル-1,2,3-オキサチアジン-4(3H)-オン-2,2-ジオキシドカリウム)、スクラロース(4,1,6-トリクロロガラクトスクロース)、グリチルリチン、ソーマチン、モネリンなどが挙げられる。
矯味剤の例としては、アスパルテーム、サッカリン、サッカリンナトリウム、グリチルリチン酸、グリチルリチン酸モノアンモニウム、グリチルリチン酸二アンモニウム、グリチルリチン酸二カリウム、グリチルリチン酸二ナトリウム、グリチルリチン酸三ナトリウム、アセスルファムK、マンニトール、エリスリトール、ソルビトール、キシリトール、トレハロース、カカオ末などが挙げられる。
保存剤の例としては、中鎖脂肪酸モノグリセライド、グリシン、エタノールなどが挙げられる。
香料の例としては、レモンフレーバー、オレンジフレーバー、グレープフルーツフレーバー、チョコレートフレーバー、アップルフレーバー、dl-メントール、l-メントールなどが挙げられる。
賦形剤の例としては、アルコール、グリセロール、転化糖、グルコース、植物油、ワックス、脂肪、半固体及び液体ポリオールなどが挙げられる。
安定化剤の例としては、酢酸、亜硫酸水素ナトリウム、チオ硫酸ナトリウム、エデト酸ナトリウム、アスコルビン酸、ジブチルヒドロキシトルエンなどが挙げられる。
次に本発明の水性液体組成物の製造方法(以下、本発明の製造方法ともいう)を説明する。尚、以下の製造方法は、本発明において一例を示すものであって、特に限定されるわけではなく、当業者であれば所望により適宜変更及び修飾することができる。
本発明の製造方法は、例えば、2価以上の有機酸を含有する酸性の水性溶液に、ヒスチジンを溶解させる工程を含むものである。
2価以上の有機酸を含有する酸性の水性溶液とは、上述した2価以上の有機酸(好ましくはクエン酸、リンゴ酸及び酒石酸のいずれか1種以上)を含有する酸性の水性溶液である。2価以上の有機酸を溶解させる溶媒は、飲食品の製造に際して通常使用されるものであれば特に限定されず、通常前記水性液体が使用される。
前記溶媒に、本発明の水性液体組成物中の含有量が上述したものになるように2価以上の有機酸を添加し、適宜撹拌して溶解させ、酸性の水性溶液を調製する。該溶媒は、2価以上の有機酸の溶解を促進するために、加温されていることが好ましく、例えば水の場合、80~100℃であることが好ましい。
また酸性の水溶液のpHは、上述した通り2価以上の有機酸を溶媒に溶解することにより低下するため特に調整する必要はないが、通常5以下、好ましくは3以下、より好ましくは1以下である。

上記調製した酸性の水溶液に、上述した本発明の水性液体組成物中の含有量となるようにヒスチジンを添加し、適宜撹拌して溶解させる。酸性の水溶液は、ヒスチジンの溶解を促進するために、加温されていることが好ましく、例えば酸性水溶液である場合、80~100℃であることが好ましい。
加温された酸性の水性溶液にヒスチジンを溶解させた場合、放冷し24時間20℃で静置することにより、ヒスチジンの溶解性を確認してもよい。ヒスチジン及び2価以上の有機酸を上述した含有量で添加した場合には、ヒスチジンの溶解性は良好であり、ヒスチジンの析出及び沈殿は認められない。
本発明の水性液体組成物中のヒスチジン及び有機酸以外の他の成分(例えば、上述の増粘剤、甘味剤、矯味剤、保存剤、香料、賦形剤、安定化剤など)は、任意の段階で添加することができ、例えば酸性の水性溶液に添加しておいてもよいし、ヒスチジンの添加後に添加してもよい。
本発明の水性液体組成物は、それ自体で、飲食品、医薬品等として供することができ、また任意の濃度のヒスチジンを含有する飲食品、医薬品等の製造のための原料として有用である。本発明の水性液体組成物を原料として利用する場合、20重量%以下の任意のヒスチジン濃度に希釈した後に利用してもよい。本発明の水性液体組成物又はその希釈液を用いれば、固形のヒスチジンを直接取り扱う場合よりも、容易に飲食品、医薬品等を製造することが可能となる。
ヒスチジンは、成長に関与し、神経機能補助の役割も果たし、さらに抗酸化作用やストレスを抑制する作用があることから、本発明の水性液体組成物は、慢性関節炎の症状改善、ストレスの軽減、性的欲求の向上、集中力や記憶力の向上、肥満の予防、抗疲労のための飲食品、医薬品等に適用することができる。
本発明の水性液体組成物は、飲食品加工原料として使用する場合には製造や保存の際には以下の観点から有用である。
(1)飲食品や医薬品の製造において、ヒスチジンを原料中に添加するときに粉体を用いるよりも液剤として用いる方が原料中に容易に分散させやすい。特に水分が少ない系や原料の粘度が高い系でその効果をより発揮する。
(2)飲食品や医薬品の製造工程で、原料中にヒスチジン液剤として添加することで、加熱や乾燥により水分を減量してもヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることがなく、またヒスチジンが析出することにより口当たりのざらつき等が発生することがない。
(3)液剤は冷蔵または冷凍下に静置しても、ヒスチジンが析出することがない。そのためヒスチジンを液剤として飲食品や医薬品の原料として使用することで、その製造及びその保管工程で冷蔵や冷凍を経ても、製品中でヒスチジンが析出することなく、均一性を維持することから、製品中でのヒスチジンの濃度に偏りが生じることなく、またヒスチジンが析出することによる口当たりのざらつき等が発生することもない。

また本発明の水性液体組成物は、上記のような飲食品、医薬品等の他、本発明の水性液体組成物に含有されるヒスチジンが20重量%以下の濃度に希釈されない態様で本発明の水性液体組成物を含有する飲食品などの製造に使用される。
本発明は、そのような本発明の水性液体組成物を含有する飲食品を更に提供する。また、該水性液体組成物を含有する医薬品を提供する。
本発明は、そのような本発明の水性液体組成物を含有する飲食品を更に提供する。また、該水性液体組成物を含有する医薬品を提供する。
本明細書において「飲食品」とは、飲料を含む食品全般を意味し、消費者庁の特別用途食品制度に規定される特別用途食品(病者用食品、特定保健用食品)や、保健機能食品制度に規定される特定保健用食品や栄養機能食品、さらにダイエタリーサプリメント(栄養補助食品)も包含するものである。
本発明の飲食品は、経口摂取可能な形態であれば特に限定されないが、例えば、飲料、ゼリー、グミ、キャンディー、ガム、チョコレートなどの食品、栄養食品、カプセル等が挙げられる。これらの形態のうち、高用量のヒスチジンを簡便に摂取することができることから、カプセルが好ましい。これらは、本発明の水性液体組成物が他の原料と混じり合って20重量%以下の濃度に希釈されないように、当該液体組成物をゼリー状にして配合したり、当該液体組成物を適当なカプセルに封入したりすることにより、当該分野において公知の方法で調製することができる。
本発明の飲食品は、本発明の水性液体組成物に含有されるヒスチジンが20重量%以下の濃度に希釈されない態様で本発明の水性液体組成物を含有する。好ましくは、本発明の飲食品中には、本発明の水性液体組成物と他の原料とが明確に分かれた状態で存在する。そのような飲食品としては、例えば、本発明の水性液体組成物を封入したカプセル、ゼリー、グミ、キャンディー、ガム、チョコレート、或いは油脂分が多く、水分が少ない食品であって水相にヒスチジンを高濃度に溶解し、油脂成分中にその水相を分散、乳化させたもの(例えばマヨネーズ、ドレッシング、マーガリン、クリーム、調味油など)などが挙げられるがこれらに限定されない。
本発明の飲食品中の本発明の水性液体組成物の含有量は、特に限定されないが、所望の量のヒスチジンを本発明の飲食品により摂取し得るように適宜設定することができる。
また本発明の医薬品は、本発明の水性液体組成物が他の原料と混じり合って20重量%以下の濃度に希釈されないように、常套手段に従って医薬組成物として製剤化することができる。本発明の医薬品は、そのまま液剤として、又は適当な剤形の医薬組成物として、対象に対して経口的又は非経口的に投与することができる。本発明の医薬品は、通常、経口的に投与される。
本発明の医薬品中に所望の量のヒスチジンが含まれるように、本発明の水性液体組成物が含有される。その含有量は、通常、医薬組成物全体の約0.1~100重量%、好ましくは約1~99重量%、さらに好ましくは約10~90重量%程度である。
本発明の飲食品及び医薬品は、少量で高用量のヒスチジンの摂取が可能であるため有用である。
以下、実施例により、本発明を更に詳細に説明するが、本発明は以下の実施例に限定されるものではない。
実施例1:ヒスチジンの溶解性への酸性物質の種類の影響
80~100℃の温度下で、表1に記載した量の酸性物質(酢酸、リン酸、クエン酸、リンゴ酸又は酒石酸)を水に添加し溶解させ、酸性の水溶液を調製した。酸性の水溶液のpHは、表1に示す通り、いずれも1.20以下であった。次いで、調製した酸性の水溶液に、表1に記載した量のヒスチジン(HIS)を添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。
80~100℃の温度下で、表1に記載した量の酸性物質(酢酸、リン酸、クエン酸、リンゴ酸又は酒石酸)を水に添加し溶解させ、酸性の水溶液を調製した。酸性の水溶液のpHは、表1に示す通り、いずれも1.20以下であった。次いで、調製した酸性の水溶液に、表1に記載した量のヒスチジン(HIS)を添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。
結果を表1に示す。酸性物質としてクエン酸、リンゴ酸又は酒石酸を用いてpH1.0以下の酸性の水溶液を調製した場合、添加したヒスチジンは、組成物中の濃度が45重量%以上となる量であっても良好に溶解し(表1中、溶解性○)、高濃度のヒスチジンを含有する液体組成物を調製することができた。一方、酸性物質として酢酸又はリン酸を用いてpH1.0以下の酸性の水溶液を調製した場合、添加したヒスチジンは溶解しないか又は溶解しても24時間室温で静置した後に析出し(表1中、溶解性×)、高濃度のヒスチジンを含有する液体組成物を調製することはできなかった。
これらの結果から、酸性の水溶液のpHを一定値以下に下げるのみでは高濃度のヒスチジンを含有する液体組成物を調製することはできず、クエン酸、リンゴ酸及び酒石酸などの2価以上の有機酸を用いる必要があることが示された。

実施例2:ヒスチジンに対する酸性物質の添加比率の影響(45重量%以上のヒスチジンを含有する液体組成物を調製する場合)
80~100℃の温度下で、表2に記載した量の酸性物質(クエン酸、リンゴ酸又は酒石酸)を水に添加し溶解させ、酸性の水溶液を調製した。次いで、調製した酸性の水溶液に、45重量%以上の濃度となるように表2に記載した量のヒスチジン(HIS)を添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した.表2中pH欄の「-」は測定不能を示す。但し、表2中、*2の組成物は、*1の組成物を調製した後、減圧蒸留にて水分量を減量することにより調製した。

結果を表2に示す。1重量部のヒスチジンに対して0.65重量部以上のクエン酸、0.67重量部以上のリンゴ酸、又は0.60重量部以上の酒石酸を用いて酸性の水溶液を調製した場合、ヒスチジンの濃度が45重量%以上の液体組成物を調製することができた。一方、1重量部のヒスチジンに対して0.56重量部のクエン酸又はリンゴ酸を用いて酸性の水溶液を調製した場合、ヒスチジンの濃度が47.4重量%以上の液体組成物を調製することはできなかった。
表2中、*2の組成物は、*1の組成物(ヒスチジン高濃度液剤)から水分を留去し液剤中のヒスチジン濃度が上昇してもヒスチジンが析出することなく、液剤中で溶解性と均一性を維持した。
また*1の組成物(ヒスチジン高濃度液剤)を冷凍温度下(-80℃、24時間)においても、ヒスチジンが析出することなく、液剤中で溶解性と均一性を維持した。また室温に戻しても凍結前と性状に変化が認められなかった。
表2中、*2の組成物は、*1の組成物(ヒスチジン高濃度液剤)から水分を留去し液剤中のヒスチジン濃度が上昇してもヒスチジンが析出することなく、液剤中で溶解性と均一性を維持した。
また*1の組成物(ヒスチジン高濃度液剤)を冷凍温度下(-80℃、24時間)においても、ヒスチジンが析出することなく、液剤中で溶解性と均一性を維持した。また室温に戻しても凍結前と性状に変化が認められなかった。
実施例3:水、酸性物質及びヒスチジンの添加量の影響
80~100℃の温度下で、表3~5に記載した量のクエン酸を水に添加し溶解させ、酸性の水溶液を調製した。次いで、調製した酸性の水溶液に、表3~5に記載した量のヒスチジン(HIS)を添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。但し、表5中、*2の組成物は、*1の組成物を調製した後、減圧蒸留にて水分量を減量することにより調製した。
80~100℃の温度下で、表3~5に記載した量のクエン酸を水に添加し溶解させ、酸性の水溶液を調製した。次いで、調製した酸性の水溶液に、表3~5に記載した量のヒスチジン(HIS)を添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。但し、表5中、*2の組成物は、*1の組成物を調製した後、減圧蒸留にて水分量を減量することにより調製した。
表3に示すように、酸性の水溶液(水50g、クエン酸60g)に添加するヒスチジンの量を下げてもヒスチジンの溶解性は良好であった。
また表4に示すように、ヒスチジンと酸性物質の比率が一定(酸性物質/HIS=0.67(w/w))となるようにヒスチジンと酸性物質の添加量を下げた場合にもヒスチジンの溶解性は良好であった。
また表5に示すように、ヒスチジンと水の比率が一定(対水ヒスチジン含量=180%(w/w))となるようにヒスチジンと水の添加量を下げた場合にもヒスチジンの溶解性は良好であった。
また表4に示すように、ヒスチジンと酸性物質の比率が一定(酸性物質/HIS=0.67(w/w))となるようにヒスチジンと酸性物質の添加量を下げた場合にもヒスチジンの溶解性は良好であった。
また表5に示すように、ヒスチジンと水の比率が一定(対水ヒスチジン含量=180%(w/w))となるようにヒスチジンと水の添加量を下げた場合にもヒスチジンの溶解性は良好であった。
実施例4:ヒスチジンに対する酸性物質の添加比率の影響(20重量%又は30重量%のヒスチジンを含有する液体組成物を調製する場合)
80~100℃の温度下で、表6又は7に記載した量の酸性物質(クエン酸又はリンゴ酸)を水に添加し溶解させ、酸性の水溶液を調製した。次いで、調製した酸性の水溶液に、20重量%又は30重量%の濃度となるように表6又は7に記載した量のヒスチジンを添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。
80~100℃の温度下で、表6又は7に記載した量の酸性物質(クエン酸又はリンゴ酸)を水に添加し溶解させ、酸性の水溶液を調製した。次いで、調製した酸性の水溶液に、20重量%又は30重量%の濃度となるように表6又は7に記載した量のヒスチジンを添加し、十分に撹拌した。混合物を24時間室温で静置し、ヒスチジンの溶解性を検討した。
表6に示すように、ヒスチジンを20重量%の濃度で添加した場合、ヒスチジン1重量部に対して酸性物質を0.17重量部添加しても、ヒスチジンは溶解しなかった。一方、ヒスチジン1重量部に対して酸性物質を0.27重量部以上添加すると、ヒスチジンの溶解性は良好であった。
また表7に示すように、ヒスチジンを30重量%の濃度で添加した場合、ヒスチジン1重量部に対して酸性物質を0.33重量部添加しても、ヒスチジンは溶解しなかった。一方、ヒスチジン1重量部に対して酸性物質を0.40重量部以上添加すると、ヒスチジンの溶解性は良好であった。
また表7に示すように、ヒスチジンを30重量%の濃度で添加した場合、ヒスチジン1重量部に対して酸性物質を0.33重量部添加しても、ヒスチジンは溶解しなかった。一方、ヒスチジン1重量部に対して酸性物質を0.40重量部以上添加すると、ヒスチジンの溶解性は良好であった。
以上の実施例1~4において2価以上の有機酸を用いて得られたデータについて、「液剤中HIS濃度%(W/W)」をx、「酸性物質/HIS(W/W)」をyとしてグラフ上にプロットすると、良好なヒスチジンの溶解性が得られたもの(表中、○)と、良好なヒスチジンの溶解性が得られなかったもの(表中、×)とは、関数(y=0.0109x+0.0511)で表される線によってxy平面上で明確に区分された。従って、「液剤中HIS濃度%(W/W)」と「酸性物質/HIS(W/W)」とを、当該xy平面上で、本実施例において良好なヒスチジンの溶解性が得られたものが含まれる領域に含まれるように設定することにより、高濃度のヒスチジンを含有する液体組成物を調製することができることが明らかとなった。
本発明は、従来公知のものよりも高濃度のヒスチジンを含有する液体組成物を提供するため、飲食品、保健機能食品、特別用途食品、医薬品などの分野において有用である。
本出願は、日本で出願された特願2013-045654を基礎としており、その内容は本明細書に全て包含されるものである。
Claims (13)
- ヒスチジンを20重量%を超える濃度で含有する水性液体組成物。
- ヒスチジンの濃度が20重量%を超え、60重量%以下である、請求項1に記載の組成物。
- 20重量%を超える濃度のヒスチジンと2価以上の有機酸とを含有する、水性液体組成物。
- ヒスチジンの濃度が20重量%を超え、60重量%以下である、請求項3に記載の組成物。
- 2価以上の有機酸が、クエン酸、リンゴ酸及び酒石酸から選ばれる1種以上である、請求項3又は4に記載の組成物。
- 水性液体組成物中のヒスチジンの濃度をA重量%としたときに、水性液体組成物中のヒスチジン1重量部に対する2価以上の有機酸の含有量が、(0.0109A+0.0511)重量部以上である、請求項3~5のいずれか一項に記載の組成物。
- 請求項1~6のいずれか一項に記載の組成物を、該組成物に含有されるヒスチジンが20重量%以下の濃度に希釈されない態様で含有する飲食品。
- 請求項1~6のいずれか一項に記載の組成物を、該組成物に含有されるヒスチジンが20重量%以下の濃度に希釈されない態様で含有する医薬品。
- 2価以上の有機酸を含有する酸性の水性溶液に、ヒスチジンを20重量%を超える濃度で溶解させる工程を含むことを特徴とする、ヒスチジンを含有する水性液体組成物の製造方法。
- ヒスチジンを20重量%を超えて60重量%までの濃度で溶解させることを特徴とする、請求項9に記載の製造方法。
- 2価以上の有機酸が、クエン酸、リンゴ酸及び酒石酸のいずれか1種以上である、請求項9又は10に記載の製造方法。
- 酸性の水性溶液が、水性液体組成物中のヒスチジンの濃度をA重量%としたときに、水性液体組成物中のヒスチジン1重量部に対して2価以上の有機酸を(0.0109A+0.0511)重量部以上含有する、請求項9~11のいずれか一項に記載の製造方法。
- 酸性の水性溶液のpHが、1.0以下である、請求項9~12のいずれか一項に記載の製造方法。
