前記大腸菌(E.coli NBRC3972)を、SCD寒天培地(日本製薬株式会社製)を用いて、好気条件下で37℃にて18時間静置培養した。SCD寒天培地上に形成されたコロニーを適量かきとり、生理食塩水(大塚製薬株式会社製)に懸濁して108cfu/mLの菌濃度に調整し、試験菌液とした。この試験菌液0.005mLを、実施例及び比較例の各透明ゲル状手指殺菌剤0.495mLに摂取し、30秒間接触させた後、直ちにこの混合液をSCDLP液体培地(日本製薬株式会社製)で10倍に希釈し、不活化した。不活化した溶液(以下、「不活化液」と称することがある)を、SCDLP培地で更に100倍希釈、1,000倍希釈、10,000倍希釈と10倍段階希釈した。SCDLP寒天培地(日本製薬株式会社製)(9cmシャーレ)に、各濃度に希釈した不活化液を、各々1mL播種(n=3)し、混釈平板培養法により好気条件下で37℃にて24時間静置培養した。培養後、形成された全コロニー数をカウントし、残存生菌数を測定した。コロニーが観測されない時は、検出限界の10cfu/mL未満とした。測定した残存生菌数を下記評点で判定し、その評点の平均値に基づき、下記評価基準で評価した。
なお、cfuとは、コロニー形成単位(colony forming unit)である。
