Abstract
Description
即ち、本発明の3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する化合物は抗菌活性を有し、更に、それらの中には、多剤耐性菌であるMRSA、VRE、クロストリジウム ディフィシルの少なくともいずれかに対する抗菌活性を有するものも多数含まれていることを知見し、本発明の完成に至った。

本発明の抗菌化合物またはその塩によると、前記従来における諸問題を解決することができ、3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属し、抗菌作用を有する抗菌化合物またはその塩を提供することができる。
本発明の化合物、または抗菌化合物の製造方法によると、前記従来における諸問題を解決することができ、前記化合物、その互変異性体、幾何異性体、乃至それらの塩、または前記抗菌化合物またはその塩の効率的な製造方法を提供することができる。

本発明の化合物は、下記一般式(1)~一般式(3)のいずれかで表される新規化合物、及び下記一般式(4)、及び一般式(5)のいずれかで表される抗菌化合物である。下記一般式(1)~一般式(3)のいずれかで表される新規化合物、及び下記一般式(4)及び一般式(5)のいずれかで表される抗菌化合物は、それぞれ、3-アシルオキシインドール化合物、あるいは3-アシル-4-ヒドロキシクマリン化合物に属する化合物であり、以下、一般式(1)で表される化合物を化合物AI、一般式(2)で表される化合物を化合物AII、一般式(3)で表される化合物を化合物B、一般式(4)で表される化合物を化合物BI、一般式(5)で表される化合物を化合物BIIと称することがある。

本発明の化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法、抗菌化合物またはその塩の製造方法は、下記の合成法1~合成法5のいずれかである。
前記化合物、並びに抗菌化合物は、前記一般式(1)~(5)で示される、3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する化合物(化合物AI、AII、B、BI、BII)である。
前記その互変異性体、幾何異性体としては、前記一般式(1)で表される化合物AI及び前記一般式(2)で表される化合物AIIは、互変異性体、幾何異性体を有しており、したがって、前記化合物AI及びAIIのいずれかの互変異性体、幾何異性体が含まれる。前記化合物A及びCは、例えば前記化学式(1)に示されるような数種類の構造パターンをとり得、ある一定の状態では固定されていない状態で存在しているものと考えられる。
前記塩としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ナトリウム、カリウム等とのアルカリ金属塩;カルシウム、マグネシウム等とのアルカリ土類金属塩;メチルアミン、エチルアミン、ジエタノールアミン等との有機アミン塩;などが挙げられる。
前記合成法1は、前記一般式(1)~一般式(3)のいずれかで表される化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法であって、基質である、一般式(6)または一般式(7)で表されるオキシインドール化合物(以下、それぞれ「オキシインドール(Aa)、オキシインドール(Ab)」と称することがある)、及び一般式(8)で表される4-ヒドロキシクマリン化合物(以下「4-ヒドロキシクマリン(Ba)」と称することがある)のいずれかを、有機溶媒中、縮合剤及びアミン塩基の存在下で、カルボン酸と混合することで反応させる工程を含有してなり、更に必要に応じてその他の工程を含有してなる。
Z1は、ハロゲン原子を有していてもよい炭素数8~30の環状のアルキル基、直鎖または分岐アルキル基のいずれかを表す。
X2は、単結合、-CO-、-CONH-、及び-COO-のいずれかを表す。
Y2は、-CN、-NO2、アルコキシ基及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基、及び水素原子のいずれかを表す。
X3は、単結合、及び-NH-のいずれかを表す。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
前記オキシインドール化合物(オキシインドール(Aa)、及びオキシインドール(Ab))としては、前記一般式(6)あるいは一般式(7)で表されるものであれば特に制限はなく、目的に応じて適宜選択することができる。市販品を用いてもよいし、新たに合成した合成品を用いてもよい。前記オキシインドール化合物として、例えば、下記の構造式(85)~(106)で表されるオキシインドール化合物などが挙げられる。
前記オキシインドール(A-e)としては、前記一般式(9)で表されるものであれば、特に制限はなく、目的に応じて適宜選択することができる。前記オキシインドール(A-e)として、市販品を用いても良いし、新たに合成した合成品を用いてもよいが、市販品であることが好ましい。前記オキシインドール(A-e)として、例えば2-オキシインドール、6-クロロ-2-オキシインドール、5-ブロモ-2-オキシインドール、5-フルオロ-2-オキシインドールなどが挙げられる。
前記4-ヒドロキシクマリン化合物(4-ヒドロキシクマリン(Ba))としては、一般式(8)で表されるものであれば特に制限はなく、目的に応じて適宜選択することができる。市販品を用いても良いし、新たに合成した合成品を用いてもよい。前記4-ヒドロキシクマリン(Ba)として、例えば6-クロロ-4-ヒドロキシクマリン、7-メトキシ-4-ヒドロキシクマリン、6-フルオロ-4-ヒドロキシクマリン、4-ヒドロキシクマリン、6-メチル-4-ヒドロキシクマリンなどが挙げられる。
前記有機溶媒としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ジクロロメタン、ジクロロエタン等の塩化アルカン類、アセトニトリル、プロピオニトリル等のニトリル類、ジメチルスルホキシド等のスルホキシド類、ジメチルホルムアミド等のアミド類、酢酸エチル等のエステル類、テトラヒドロフラン、ジオキサン等のエーテル類などが挙げられる。これらの中でも、ジクロロメタン、アセトニトリルが好ましい。
前記縮合剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、N,N'-ジシクロヘキシルカルボジイミド等の置換カルボジイミド類などが挙げられる。これらの中でも、N-[3-(ジメチルアミノ)]プロピル-N'-エチルカルボジイミド及びその塩酸塩のいずれかが好ましい。
前記アミン塩基としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、トリエチルアミン、ジイソプロピルエチルアミン等の3級アミン、ピリジン、4-(ジメチルアミノ)ピリジン等のピリジン類などが挙げられる。これらの中でも、4-(ジメチルアミノ)ピリジンが好ましい。
前記カルボン酸としては、特に制限はなく、目的に応じて適宜選択することができる。市販品を用いてもよいし、新たに合成した合成品を用いてもよい。前記カルボン酸としては、例えば、下記の構造式(107)~(114)で表される化合物(D-a-01~D-a-5、D-b-01、D-c-01、D-c-02)などが挙げられる。
前記その他の工程としては、本発明の効果を損なわない範囲内であれば、特に制限はなく、目的に応じて適宜選択することができ、例えば、3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する前記化合物、その互変異性体、幾何異性体、並びに抗菌化合物に塩を付加する工程や、それらをさらに修飾する工程、並びに3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する前記化合物、その互変異性体、幾何異性体、並びに抗菌化合物、乃至それらの塩を精製する工程などが挙げられる。
前記合成法2は、前記一般式(1)~一般式(3)のいずれかで表される化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法であって、基質である前記一般式(6)または一般式(7)で表されるオキシインドール化合物、及び前記一般式(8)で表される4-ヒドロキシクマリン化合物のいずれかを、縮合剤を用いることなく、有機溶媒中、アミン塩基の存在下で、カルボン酸クロリドと混合することで反応させる工程を含有してなり、更に必要に応じてその他の工程を含有してなる。

本発明の抗菌剤は、本発明の化合物、その互変異性体、幾何異性体、乃至それらの塩、及び本発明の抗菌化合物またはその塩の少なくともいずれかを含有してなり、更に必要に応じてその他の成分を含有してなる。
前記抗菌剤における本発明の化合物、その互変異性体、幾何異性体、乃至それらの塩、及び本発明の抗菌化合物またはその塩の少なくともいずれかの含有量としては、特に制限はなく、目的に応じて適宜選択することができる。また、前記抗菌剤は、本発明の化合物、その互変異性体、幾何異性体、乃至それらの塩そのものであってもよいし、本発明の抗菌化合物またはその塩そのものであってもよいし、両者からなるものであってもよい。
前記その他の成分としては、本発明の効果を損なわない範囲内であれば、特に制限はなく、目的に応じて適宜選択することができ、例えば、エタノール、水、デンプン等の薬理学的に許容され得る担体などが挙げられる。
前記抗菌剤中の、前記その他の成分の含有量としては、特に制限はなく、前記化合物、並びに抗菌化合物の効果を損なわない範囲内で、目的に応じて適宜選択することができる。
前記抗菌剤が抗菌活性を示す対象としては、細菌であれば特に制限はなく、目的に応じて適宜選択することができるが、薬剤耐性菌であることが好ましく、多剤耐性菌であることがより好ましく、MRSA、VRE及びC.difficileの少なくともいずれかであることがさらに好ましい。
前記抗菌剤の剤型としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、経口固形剤、経口液剤、注射剤、吸入散剤などが挙げられる。
前記経口固形剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、錠剤、被覆錠剤、顆粒剤、散剤、カプセル剤などが挙げられる。
前記経口固形剤の製造方法としては、特に制限はなく、常法を使用することができ、例えば、前記化合物に、賦形剤、及び必要に応じて、前記その他の成分、各種添加剤を加えることにより、製造することができる。ここで、前記賦形剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、乳糖、白糖、塩化ナトリウム、ブドウ糖、デンプン、炭酸カルシウム、カオリン、微結晶セルロース、珪酸などが挙げられる。また、前記添加剤としても、特に制限はなく、目的に応じて適宜選択することができ、例えば、結合剤、崩壊剤、滑沢剤、着色剤、矯味/矯臭剤などが挙げられる。
前記結合剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、水、エタノール、プロパノール、単シロップ、ブドウ糖液、デンプン液、ゼラチン液、カルボキシメチルセルロース、ヒドロキシプロピルセルロース、ヒドロキシプロピルスターチ、メチルセルロース、エチルセルロース、シェラック、リン酸カルシウム、ポリビニルピロリドンなどが挙げられる。
前記崩壊剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、乾燥デンプン、アルギン酸ナトリウム、カンテン末、炭酸水素ナトリウム、炭酸カルシウム、ラウリル硫酸ナトリウム、ステアリン酸モノグリセリド、乳糖などが挙げられる。
前記滑沢剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、精製タルク、ステアリン酸塩、ホウ砂、ポリエチレングリコールなどが挙げられる。
前記着色剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、酸化チタン、酸化鉄などが挙げられる。
前記矯味/矯臭剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、白糖、橙皮、クエン酸、酒石酸などが挙げられる。
前記経口液剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、内服液剤、シロップ剤、エリキシル剤などが挙げられる。
前記経口液剤の製造方法としては、特に制限はなく、常法を使用することができ、例えば、前記化合物、及び必要に応じて、前記その他の成分に、添加剤を加えることにより、製造することができる。ここで、前記添加剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、矯味/矯臭剤、緩衝剤、安定化剤などが挙げられる。
前記緩衝剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、クエン酸ナトリウムなどが挙げられる。
前記安定化剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、トラガント、アラビアゴム、ゼラチンなどが挙げられる。
前記注射剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、溶液、懸濁液、用事溶解用固形剤などが挙げられる。
前記注射剤の製造方法としては、特に制限はなく、常法を使用することができ、例えば、前記化合物、及び必要に応じて、前記その他の成分に、pH調節剤、緩衝剤、安定化剤、等張化剤、局所麻酔剤などを添加することにより、製造することができる。ここで、前記pH調節剤及び前記緩衝剤としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、クエン酸ナトリウム、酢酸ナトリウム、リン酸ナトリウムなどが挙げられる。また、前記安定化剤としても、特に制限はなく、目的に応じて適宜選択することができ、例えば、ピロ亜硫酸ナトリウム、EDTA、チオグリコール酸、チオ乳酸などが挙げられる。前記等張化剤としても、特に制限はなく、目的に応じて適宜選択することができ、例えば、塩化ナトリウム、ブドウ糖などが挙げられる。前記局所麻酔剤としても、特に制限はなく、目的に応じて適宜選択することができ、例えば、塩酸プロカイン、塩酸リドカインなどが挙げられる。
前記抗菌剤の投与方法、投与量、投与時期、及び投与対象としては、特に制限はなく、目的に応じて適宜選択することができる。
前記投与方法としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、経口投与法、注射による方法、吸入による方法などが挙げられる。
前記投与量としては、特に制限はなく、投与対象個体の年齢、体重、体質、症状、他の成分を有効成分とする医薬の投与の有無など、様々な要因を考慮して適宜選択することができる。
前記投与対象となる動物種としては、特に制限はなく、目的に応じて適宜選択することができ、例えば、ヒト、サル、ブタ、ウシ、ヒツジ、ヤギ、イヌ、ネコ、マウス、ラット、トリなどが挙げられるが、これらの中でもヒトに好適に投与される。
本発明の抗菌剤が抗菌活性を示す対象としては、前記のように、特に制限はなく、目的に応じて適宜選択することができ、薬剤耐性菌であることが好ましく、多剤耐性菌であることがより好ましく、MRSA、VRE及びC.difficileの少なくともいずれかであることがさらに好ましいが、前記対象が、特定の薬剤に対する耐性菌(例えばバンコマイシン耐性菌)である場合、前記抗菌剤は、前記薬剤に対する感受性菌(例えばバンコマイシン感受性菌)には無効であることが好ましい。
前記MICを求める方法としては、特に制限はなく、公知の方法を適宜用いることができる。
また、本発明において、本発明の抗菌剤が薬剤感受性菌に無効であるとは、前記薬剤感受性菌に対する抗菌活性が低い場合も含むものとする。前記抗菌剤の前記薬剤感受性菌に対するMICとしては、特に制限はなく、細菌の種類などに応じて適宜選択することができるが、8mg/L以上が好ましい。
前記抗菌剤は、1種単独で使用してもよいし、他の成分を有効成分とする医薬と併せて使用してもよい。また、前記抗菌剤は、他の成分を有効成分とする医薬中に、配合された状態で使用してもよい。
前記抗菌剤は、薬剤感受性菌を耐性化させないために、薬剤感受性菌には抗菌活性を示さないことが好ましいことから、薬剤感受性菌にのみ抗菌活性を有する他の成分を有効成分とする薬剤と併用又は前記他の成分を有効成分とする薬剤に配合することが特に好ましい。
前記抗菌剤は、前記一般式(1)~(5)で表される、3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する化合物の少なくともいずれかを含有し、優れた抗菌活性を示すことから、本発明の感染症治療薬に好適に利用可能である。
本発明の感染症治療薬は、本発明の抗菌剤を含有してなり、更に必要に応じてその他の成分を含有してなる。
前記感染症治療薬における本発明の抗菌剤の含有量としては、特に制限はなく、目的に応じて適宜選択することができる。また、前記感染症治療薬は、本発明の抗菌剤そのものであってもよい。
前記その他の成分としては、本発明の効果を損なわない範囲内であれば、特に制限はなく、目的に応じて適宜選択することができ、例えば、エタノール、水、デンプン等の薬理学的に許容され得る担体などが挙げられる。
前記感染症治療薬中の、前記その他の成分の含有量としては、特に制限はなく、前記一般式(1)~前記一般式(5)で表される、3-アシルオキシインドール化合物、及び3-アシル-4-ヒドロキシクマリン化合物のいずれかに属する化合物の効果を損なわない範囲内で、目的に応じて適宜選択することができる。
前記感染症治療薬は、1種単独で使用してもよいし、他の成分を有効成分とする医薬と併せて使用してもよい。また、前記感染症治療薬は、他の成分を有効成分とする医薬中に、配合された状態で使用してもよい。
前記感染症治療薬は、薬剤感受性菌を耐性化させないために、薬剤感受性菌には抗菌活性を示さないことが好ましいことから、薬剤感受性菌にのみ抗菌活性を有する他の成分を有効成分とする薬剤と併用又は前記他の成分を有効成分とする薬剤に配合することが好ましい。
<D-a-01(デシロキシ酢酸)の合成>
ブロモデカン(市販品、3.00g、13.6mmol)、グリコール酸メチル(市販品、1.34g、15.0mmol、1.1equiv.)をジメチルホルムアミド(5mL)-テトラヒドロフラン(市販品、5mL)混合溶媒に溶解し、水素化ナトリウム(0.60g、1.1equiv.)を室温で加えた。
60℃で12時間撹拌した後、溶媒を減圧下で留去した。

N-アセチルオキシインドール(A-b-01)(100mg、0.57mmol)、デシロキシ酢酸(D-a-01)(123mg、0.57mmol、1.0equiv.)、4-(ジメチルアミノ)ピリジン(市販品、105mg、0.85mmol、1.5equiv.)をジクロロメタン(4mL)に溶解し、縮合剤としてN-[3-(ジメチルアミノ)]プロピル-N’-エチルカルボジイミド塩酸塩(市販品、165mg、0.85mmol、1.5equiv.)を室温で加えた。
室温で12時間撹拌した後、5重量%クエン酸水溶液(10mL)を加え、酢酸エチル(20mL)を用い抽出した。
得られた有機層を分離し、無水硫酸ナトリウム上で乾燥させた。溶液を濃縮後、酢酸エチル-ヘキサン混合溶媒を用いたシリカゲルカラムクロマトグラフィーにより精製することで、A013(N-アセチル-3-デシロキシアセチル-2-オキシインドール)(160mg、76%)が得られた。
<B003(6-クロロ-3-デカノイル-4-ヒドロキシクマリン)の合成>
6-クロロ-4-ヒドロキシクマリン(市販品、100mg、0.51mmol)、4-(ジメチルアミノ)ピリジン(市販品、93mg、0.76mmol、1.5equiv.)をジクロロメタン(4mL)に溶解し、デカン酸クロリド(0.12mL、0.56mmol、1.1 equiv.)を室温で加えた。
室温で12時間撹拌した後、5重量%クエン酸水溶液(10mL)を加え、酢酸エチル(20mL)を用い抽出した。
得られた有機層を分離し、無水硫酸ナトリウム上で乾燥させた。溶液を濃縮後、酢酸エチル-ヘキサン混合溶媒を用いたシリカゲルカラムクロマトグラフィーにより精製することで、B003(6-クロロ-3-デカノイル-4-ヒドロキシクマリン)(134mg、71%)が得られた。
<A033(N-(フェニルアミノカルボニル)-5-ブロモ-3-デカノイル-2-オキシインドール)の合成>
A-c-03(N-(フェニルアミノカルボニル)-5-ブロモ-2-オキシインドール)(100mg、0.30mmol)、4-(ジメチルアミノ)ピリジン(市販品、55mg、0.45mmol、1.5equiv.)をジクロロメタン(4mL)に溶解し、デカン酸無水物(108mg、0.33mmol,1.1equiv.)を室温で加えた。
室温で12時間撹拌した後、5重量%クエン酸水溶液(10mL)を加え、酢酸エチル(20mL)を用い抽出した。
得られた有機層を分離し、無水硫酸ナトリウム上で乾燥させた。溶液を濃縮後、酢酸エチル-ヘキサン混合溶媒を用いたシリカゲルカラムクロマトグラフィーにより精製することで、A033(N-(フェニルアミノカルボニル)-5-ブロモ-3-デカノイル-2-オキシインドール)(127mg、87%)が得られた。
<B020(4-ヒドロキシ-3-(オクチルアミノカルボニル)クマリン)の合成>
4-ヒドロキシクマリン(市販品、103mg、0.64mmol)、4-(ジメチルアミノ)ピリジン(市販品、94mg、0.77mmol、1.2equiv.)をジクロロメタン(4mL)に溶解し、オクチルイソシアネート(市販品、120mg、0.77mmol、1.2equiv.)を室温で加えた。
室温で12時間撹拌した後、5重量%クエン酸水溶液(10mL)を加え、酢酸エチル(20mL)を用い抽出した。
得られた有機層を分離し、無水硫酸ナトリウム上で乾燥させた。溶液を濃縮後、酢酸エチル-ヘキサン混合溶媒を用いたシリカゲルカラムクロマトグラフィーにより精製することで、B020(4-ヒドロキシ-3-(オクチルアミノカルボニル)クマリン)(165mg、82%)が白色固体として得られた。

<黄色ブドウ球菌、大腸菌に対するMICの測定>
実施例1~84で得られた各化合物の黄色ブドウ球菌及び大腸菌に対する最小発育阻害濃度(MIC)の測定を行った。
培養終了後、菌の増殖の有無を濁度にて目視して判定し、各菌株のMICを求めた。結果を表1~表3に示す。

実施例1~84で得られた各化合物の腸球菌に対する最小発育阻害濃度(MIC)の測定を行った。

<クロストリジウム ディフィシル菌及びウェルシュ菌に対するMICの測定>
実施例1~84で得られた各化合物のクロストリジウム ディフィシル菌及びウェルシュ菌に対する最小発育阻害濃度(MIC)の測定を行った。
培養終了後、菌の増殖の有無を濁度にて目視して判定し、各菌株のMICを求めた。結果を表1~表3に示す。

X3は、単結合、及び-NH-のいずれかを表す。
Y3は、-O-、-NH-、-NHCO-、-NHCOO-、-S-、-SO-、及び-SO2-のいずれかを表す。
X4は、炭素数1~8のアルキレン基のいずれかを表す。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
R1は、アルコキシ基、-OH、-CN、-NO2及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基で表される基のいずれかを表す。
<2> X1及びX4がメチレン基であり、X2が-CO-、及び-CONH-のいずれかであり、X3が単結合であり、Y1及びY3が-O-であり、Y2がメチル基、及び置換基としてアルコキシ基、トリフルオロメチル基、及びハロゲン原子の少なくともいずれかを有していてもよいフェニル基のいずれかであり、Zが水素原子、及びハロゲン原子のいずれかであり、Z1及びZ2が炭素数8~12の環状のアルキル基、直鎖または分岐アルキル基のいずれかである前記<1>に記載の化合物、その互変異性体、幾何異性体、乃至それらの塩である。

Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
Z2は、ハロゲン原子を有していてもよい炭素数5~30の炭化水素基で表される基のいずれかを表す。
Z3は、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
<6> 前記<1>に記載の一般式(1)から一般式(3)のいずれかで表される化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法であって、
下記一般式(6)または一般式(7)で表されるオキシインドール化合物、及び下記一般式(8)で表される4-ヒドロキシクマリン化合物のいずれかを、有機溶媒中、縮合剤及びアミン塩基の存在下で、カルボン酸と混合することで反応させる工程を含むことを特徴とする化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法である。
Z1は、ハロゲン原子を有していてもよい炭素数8~30の環状のアルキル基、直鎖または分岐アルキル基のいずれかを表す。
X2は、単結合、-CO-、-CONH-、及び-COO-のいずれかを表す。
Y2は、-CN、-NO2、アルコキシ基及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基、及び水素原子のいずれかを表す。

下記一般式(6)または一般式(7)で表されるオキシインドール化合物、及び下記一般式(8)で表される4-ヒドロキシクマリン化合物のいずれかを、有機溶媒中、アミン塩基の存在下で、カルボン酸クロリドと混合することで反応させる工程を含むことを特徴とする化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法である。
Z1は、ハロゲン原子を有していてもよい炭素数8~30の環状のアルキル基、直鎖または分岐アルキル基のいずれかを表す。
X2は、単結合、-CO-、-CONH-、及び-COO-のいずれかを表す。
Y2は、-CN、-NO2、アルコキシ基及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基、及び水素原子のいずれかを表す。

X3は、単結合、及び-NH-のいずれかを表す。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
<9> 前記<5>に記載の一般式(5)表される抗菌化合物またはその塩の製造方法であって、
下記一般式(8)で表される4-ヒドロキシクマリン化合物を、有機溶媒中、縮合剤及びアミン塩基の存在下で、カルボン酸と混合することで反応させる工程を含むことを特徴とする抗菌化合物またはその塩の製造方法である。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
<10> 前記<5>に記載の一般式(4)表される抗菌化合物またはその塩の製造方法であって、
下記一般式(8)で表される4-ヒドロキシクマリン化合物を、有機溶媒中、アミン塩基の存在下で、イソシアネートと混合することで反応させる工程を含むことを特徴とする抗菌化合物またはその塩の製造方法である。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
<11> 前記<1>から<4>のいずれかに記載の化合物、その互変異性体、幾何異性体、乃至それらの塩を含有することを特徴とする抗菌剤である。
<12> 前記<5>に記載の抗菌化合物またはその塩を含有することを特徴とする抗菌剤である。
<13> 前記<11>から<12>の少なくともいずれかに記載の抗菌剤を含有することを特徴とする感染症治療薬である。
Claims (13)
- 下記一般式(1)から一般式(3)のいずれかで表されることを特徴とする化合物、その互変異性体、幾何異性体、乃至それらの塩。
X1は、単結合、及び炭素数1~8のアルキレン基のいずれかを表す。
Y1は、単結合、-O-、-NHCO-、-NHCOO-、-S-、-SO-、及び-SO2-のいずれかを表す。
Z1は、ハロゲン原子を有していてもよい炭素数8~30の環状のアルキル基、直鎖または分岐アルキル基のいずれかを表す。
X2は、単結合、-CO-、-CONH-、及び-COO-のいずれかを表す。
Y2は、-CN、-NO2、アルコキシ基及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基、及び水素原子のいずれかを表す。
Z2は、ハロゲン原子を有していてもよい炭素数5~30の炭化水素基で表される基のいずれかを表す。
X3は、単結合、及び-NH-のいずれかを表す。
Y3は、-O-、-NH-、-NHCO-、-NHCOO-、-S-、-SO-、及び-SO2-のいずれかを表す。
X4は、炭素数1~8のアルキレン基のいずれかを表す。
Zは、水素原子、ハロゲン原子、炭素数1~6のアルコキシ基、及び炭素数1~20の炭化水素基のいずれかを表す。
R1は、アルコキシ基、-OH、-CN、-NO2及びハロゲン原子の少なくともいずれかを有していてもよい炭素数1~30の炭化水素基で表される基のいずれかを表す。 - X1及びX4がメチレン基であり、X2が-CO-、及び-CONH-のいずれかであり、X3が単結合であり、Y1及びY3が-O-であり、Y2がメチル基、及び置換基としてアルコキシ基、トリフルオロメチル基、及びハロゲン原子の少なくともいずれかを有していてもよいフェニル基のいずれかであり、Zが水素原子、及びハロゲン原子のいずれかであり、Z1及びZ2が炭素数8~12の環状のアルキル基、直鎖または分岐アルキル基のいずれかである請求項1記載の化合物、その互変異性体、幾何異性体、乃至それらの塩。
- X1及びX4がメチレン基であり、X2が-CO-、及び-CONH-のいずれかであり、X3が単結合であり、Y1及びY3が-S-であり、Y2がメチル基、及び置換基としてアルコキシ基、トリフルオロメチル基、及びハロゲン原子の少なくともいずれかを有していてもよいフェニル基のいずれかであり、Zが水素原子、及びハロゲン原子のいずれかであり、Z1及びZ2が炭素数8~12の環状のアルキル基、直鎖または分岐アルキル基のいずれかである請求項1記載の化合物、その互変異性体、幾何異性体、乃至それらの塩。
- X1、X3、及びY1が単結合であり、X2が-CO-、及び-CONH-のいずれかであり、X4がメチレン基であり、Y3が-O-、及び-S-のいずれかであり、Y2がメチル基、及び置換基としてアルコキシ基、トリフルオロメチル基、及びハロゲン原子の少なくともいずれかを有していでもよいフェニル基のいずれかであり、Zが水素原子、及びハロゲン原子のいずれかであり、Z1及びZ2が炭素数8~12の環状のアルキル基、直鎖または分岐アルキル基のいずれかである請求項1記載の化合物、その互変異性体、幾何異性体、乃至それらの塩。
- 請求項1に記載の一般式(1)から一般式(3)のいずれかで表される化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法であって、
下記一般式(6)または一般式(7)で表されるオキシインドール化合物、及び下記一般式(8)で表される4-ヒドロキシクマリン化合物のいずれかを、有機溶媒中、縮合剤及びアミン塩基の存在下で、カルボン酸と混合することで反応させる工程を含むことを特徴とする化合物、その互変異性体、幾何異性体、乃至それらの塩の製造方法。
