また、出願人は、これまでにα2δリガンド及びその製造方法として、特許文献5及び特許文献6を報告している。
特許文献5又は特許文献6には、Scheme1に記載のような化合物6の製造方法が報告されている。
本出願の発明者らは、化合物6の製造方法における不斉炭素の立体制御の方法に着目して、その効率的な方法を開発するために鋭意研究を続けてきた。これまでの製法では、最終工程の一つ前の工程(Step 4)で光学分割を行っているが、光学分割を早期の工程で実施することに、より効率的な製造方法を確立することが可能であろうと考えた。
すなわち、本発明が解決しようとする課題は、化合物6の製造の早期の工程において、その中間体を光学活性化合物とするような製造方法を開発することである。本出願の発明者らは、この課題を解決するために鋭意研究を継続し、その結果として課題を解決し、本発明を完成させた。
[式中、各置換基は以下のように定義される。R1a:水素原子またはC1-C6アルキル基]
[1] 一般式(I)を有する化合物又は一般式(II)を有する化合物を製造する方法であり、
(1)一般式(I)を有する化合物と一般式(II)を有する化合物との混合物を、
(i)還元酵素、又は、アルコール脱水素酵素、
(ii)補酵素、及び、
(iii)ギ酸又はその塩の存在下、及び/又は、グルコースの存在下、
緩衝液中で反応させることによって、
一般式(I)を有する化合物又は一般式(II)を有する化合物のどちらか一方を、一般式(I’)を有する化合物、又は、一般式(II’)を有する化合物へと変換させた後、
(2)一般式(I)を有する化合物を、一般式(II’)を有する化合物から分離することによって、又は、一般式(II)を有する化合物を、一般式(I’)を有する化合物から分離することによって、

本発明の好適な態様としては、以下の発明である。
[2] 一般式(I)を有する化合物を製造する方法であり、
(1)一般式(I)を有する化合物と一般式(II)を有する化合物とのラセミ混合物を、
(i)還元酵素、又は、アルコール脱水素酵素、
(ii)補酵素、及び、
(iii)ギ酸又はその塩の存在下、及び/又は、グルコースの存在下、
緩衝液中で反応させることによって、
一般式(II)を有する化合物を、一般式(II’)を有する化合物へと変換させた後、
(2)一般式(I)を有する化合物を、一般式(II’)を有する化合物から分離することによって、
[各式中、R1:水素原子又はC1-C6アルキル基]
一般式(I)を有する化合物を製造する方法。
[3] 還元酵素、又はアルコール脱水素酵素としてE039(株式会社ダイセル製)、補酵素としてニコチンアミドアデニンジヌクレオチド(NAD+又はNADH)を用いることを特徴とする、[1]又は[2]に記載の方法。
[4] (2)の工程において、一般式(I’)を有する化合物又は一般式(II’)を有する化合物を、塩基の存在下、環状酸無水物と反応させた後に分離することを特徴とする、[1]-[3]から選択されるいずれか1項に記載の方法。
[5] R1が、水素原子、メチル基又はエチル基である、[1]-[4]から選択されるいずれか1項に記載の方法。
[6] (1)工程における反応温度を20-40℃で行うことを特徴とする、[1]-[5]から選択されるいずれか1項に記載の方法。
[7] (1)工程における還元酵素又は、アルコール脱水素酵素の使用量が、一般式(I)を有する化合物及び/又は一般式(II)を有する化合物に対して、4-20重量%であり、補酵素の使用量が、0.001当量以下であることを特徴とする、[1]-[6]から選択されるいずれか1項に記載の方法。
[8] ギ酸又はその塩が、ギ酸ナトリウムであることを特徴とする、[1]-[7]から選択されるいずれか1項に記載の方法。
[9] 緩衝液がリン酸緩衝液であり、そのリン酸緩衝液の濃度が、50mmol以上である、[1]-[8]から選択されるいずれか1項に記載の方法。
[10] (2)の工程における、環状酸無水物が、無水コハク酸、無水マレイン酸、又は、無水フタル酸であり、塩基が、3級アミンである、[3]-[9]から選択されるいずれか1項に記載の方法。
[11] [2]-[10]から選択されるいずれか1項により製造された一般式(I)を有する化合物を用いて、一般式(IV)を有する化合物又はその塩を製造する方法。
本発明は、その製造の早期の工程において、その中間体を光学活性化合物とするような製造方法であることから効率的である。
また、一般式(II’)を有する化合物に環状酸無水物を反応させることにより、精製工程をより効率的に行うことができる。
一般式(I)又は一般式(II)を有する化合物として、好適には、R1が水素原子、メチル基又はエチル基を有する化合物である。好適には、R1がエチル基を有する化合物である。
反応温度は、20-40℃が好適であり、20-35℃がより好適である。
使用する還元酵素、又は、アルコール脱水素酵素の量は、反応基質である一般式(I)を有する化合物及び一般式(II)を有する化合物に対して、4-20重量%であり、好適には、5-10重量%である。
使用するギ酸又はその塩として、好適には、ギ酸ナトリウムであり、そのギ酸ナトリウムの使用量は、反応基質である一般式(I)を有する化合物及び一般式(II)を有する化合物に対して、好適には、1-3等量である。
使用する補酵素の量は、反応基質である一般式(I)を有する化合物及び一般式(II)を有する化合物に対して、好適には、0.0001-0.003等量であり、より好適には、0.0001-0.001等量である。
使用する補酵素としては、ニコチンアミドアデニンジヌクレオチド(NAD+、又は、NADH)、ニコチンアミドアデニンジヌクレオチドリン酸(NADP+、又は、NADPH)等を使用できるが、好ましくは、ニコチンアミドアデニンジヌクレオチド(NAD+、又は、NADH)であり、例えば、和光純薬工業株式会社で販売されている、カタログNo.308-50446(β-NAD+:β-ニコチンアミドアデニンジヌクレオチド、酸化型、オリエンタル酵母工業株式会社製)等である。
使用する緩衝液は、特に限定はされないが、例えば、リン酸緩衝液であり、好適には、50-200mmolリン酸緩衝液であり、10-20倍量(v/w、基質に対して)用いることが好ましい。
環状酸無水物としては、無水コハク酸、無水マレイン酸、無水フタル酸が好適であり、特に、無水コハク酸が好ましい。
塩基としては、一般的な3級アミン(例えば、トリエチルアミン、ピリジン等)であれば好適であるが、特に好適には、ピリジンと触媒量のDMAP(4-ジメチルアミノピリジン)を用いる場合である。
溶媒は、非プロトン性の溶媒(エーテル類、エステル類、炭化水素類、ハロゲン化炭化水素類等)であれば好適であるが、特に好適には、TBME(t-ブチルメチルエーテル)とDMAc(N,N-ジメチルアセトアミド)の混合溶媒を用いる場合である。
反応温度は40-60℃程度が好ましい。
本工程によって、例えば、一般式(II’)を有する化合物を環状酸無水物と反応させて、カルボン酸化合物である一般式(II’’)を有する化合物に変換すれば、通常のアルカリ条件での分液操作によって、容易に、一般式(I)を有する化合物と、一般式(II’’)を有する化合物とを分離することができる。
(1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オン
ラセミ体の3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オン(100 g, 0.743 mol)に対し、0.1 Mリン酸緩衝液 (pH 7, 1.5 L)、ギ酸ナトリウム (50.0 g, 50 wt%)、β-NAD+ (125 mg, 0.125 wt%, 0.00025等量)、還元酵素E039 (6.00 g, 6.0 wt%, 株式会社ダイセル製)を順次加えた。
30℃にて24時間攪拌した後、Celite(登録商標)535 (20.0 g, 20 wt%)を加え、ろ過した。残渣はTBME (800 mL)にて洗浄し、ろ液とあわせた。ろ液の水層を除去した後、有機層を20%食塩水(160 mL)にて洗浄した。
この溶液にDMAc (200 mL)、ピリジン(145 g, 2.5 eq.)、DMAP (8.97 g, 0.10 eq.)、無水コハク酸 (73.5 g, 1.0 eq.)を順次加え、60℃にて8時間攪拌した。室温まで冷却した後、水(600 mL)にて洗浄し、有機層を得た。水層はTBME (各200 mL)で2回抽出した。合致した有機層を2 M塩酸(1.0 L)で洗浄した後、さらに5%重曹水(各1.0 L)で2回洗浄し、有機層を濃縮した。濃縮残渣をヘキサン(500 mL)で希釈し、5%重曹水(500 mL)、20%食塩水(200 mL)で順次洗浄後、濃縮した。残渣を蒸留し(圧力2.8~3.0 kPa, 留出温度102~104℃)、(1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オン(41.3 g, 収率41%, 97.7%ee)を得た。

オーブン温度:130 °C (0-13 min) → 230 °C (18-20 min)

(1S,5R)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オン (9.5 min)
3-エチルビシクロ[3.2.0]へプタ-3-エン-6-オール(還元体) (9.2 min)
(実施例2)
[(1R,5S,6S)-6-(アミノメチル)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イル] 酢酸tert-ブチル=D-マンデル酸塩
窒素雰囲気下、テトラヒドロフラン(50 mL)にカリウムt-ブトキシド(8.66 g)を溶解させた後、5°C程度まで冷却した。 (ジメトキシホスホリル)酢酸tert-ブチル(17.30 g)を15°C以下で添加し、5~15°Cで1時間撹拌した。(1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オン(10.00 g)とテトラヒドロフラン(25 mL)を追加し、5~15°Cで1.5時間撹拌した。更にカリウムt-ブトキシド(0.90 g)、(ジメトキシホスホリル)酢酸tert-ブチル(1.73 g)を追加し、約1時間撹拌後、トルエン(85 mL)及び水(40 mL)を加えて分液した。有機層を水(20 mL)で二回洗浄した後、減圧下濃縮し、黄色油状物質として [(1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イリデン]酢酸tert-ブチル(23.09 g)を得た。
窒素雰囲気下、上記のようにして得た [(1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イリデン]酢酸tert-ブチル(23.09 g)にジメチルスルホキシド(40 mL)、1,8-ジアザビシクロ[5.4.0]-7-ウンデセン(22.36 g)、ニトロメタン(17.93 g)を加え、60°Cにて加熱撹拌した。5時間後に加熱を停止し、氷冷した後、酢酸エチル(70 mL)及び水(80 mL)を加えて分液した。有機層を水(60 mL)で洗浄後、減圧下濃縮し、赤色油状物質として [(1R,5S,6S)-3-エチル-6-(ニトロメチル)ビシクロ[3.2.0]ヘプタ-3-エン-6-イル]酢酸tert-ブチル(20.89 g、含量95.1%)を得た(ジアステレオマー混合物; dr=87/13)。
窒素雰囲気下、上記のようにして得た[(1R,5S)-3-エチル-6-(ニトロメチル)ビシクロ[3.2.0]ヘプタ-3-エン-6-イル]酢酸tert-ブチル(20.86 g)にエタノール(167 mL)、スポンジニッケル触媒PL-9T(川研ファインケミカル株式会社製, 4.28 g, エタノールで3回洗浄後に使用)を加えた後、ヒドラジン・一水和物(13.48 g)をゆっくりと滴下した。30~35°Cで2時間撹拌した後、ろ過 により触媒を除去し、ろ液を減圧下濃縮し、黄橙色油状物質として[(1R,5S,6S)-6-(アミノメチル)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イル]酢酸tert-ブチル(22.37 g)を得た(ジアステレオマー混合物; dr=88/12)。
窒素雰囲気下、上記のようにして得た[(1R,5S,6S)-6-(アミノメチル)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イル]酢酸tert-ブチル(15.03 g)にアセトニトリル(300 mL)を加えて内温40~45°Cに加熱し、D-マンデル酸 (6.07 g)を添加した。結晶析出を確認した後、40~45°Cで約1時間撹拌し、約1時間かけて内温0~5°Cまで徐冷し、そのままの温度で約1時間撹拌した。その後、結晶をろ過により取得し、0~5°Cに冷却したアセトニトリル (60 mL)で洗浄後、減圧下乾燥を行い、白色結晶として [(1R,5S,6S)-6-(アミノメチル)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-イル]酢酸tert-ブチル=D-マンデル酸塩(13.22 g)を得た (99.1% de, 99.8% ee, (1R,5S)-3-エチルビシクロ[3.2.0]ヘプタ-3-エン-6-オンからの収率79%)。
Claims (11)
- 一般式(I)を有する化合物又は一般式(II)を有する化合物を製造する方法であり、
(1)一般式(I)を有する化合物と一般式(II)を有する化合物との混合物を、
(i)還元酵素、又は、アルコール脱水素酵素、
(ii)補酵素、及び、
(iii)ギ酸又はその塩の存在下、及び/又は、グルコースの存在下、
緩衝液中で反応させることによって、
一般式(I)を有する化合物又は一般式(II)を有する化合物のどちらか一方を、一般式(I’)を有する化合物、又は、一般式(II’)を有する化合物へと変換させた後、
(2)一般式(I)を有する化合物を、一般式(II’)を有する化合物から分離することによって、又は、一般式(II)を有する化合物を、一般式(I’)を有する化合物から分離することによって、
[各式中、R1:水素原子又はC1-C6アルキル基]
一般式(I)を有する化合物又は一般式(II)を有する化合物を製造する方法。 - 還元酵素、又はアルコール脱水素酵素としてE039(株式会社ダイセル製)、補酵素としてニコチンアミドアデニンジヌクレオチド(NAD+又はNADH)を用いることを特徴とする、請求項1又は2に記載の方法。
- (2)の工程において、一般式(I’)を有する化合物又は一般式(II’)を有する化合物を、塩基の存在下、環状酸無水物と反応させた後に分離することを特徴とする、請求項1-3から選択されるいずれか1項に記載の方法。
- R1が、水素原子、メチル基又はエチル基である、請求項1-4から選択されるいずれか1項に記載の方法。
- (1)工程における反応温度を20-40℃で行うことを特徴とする、請求項1-5から選択されるいずれか1項に記載の方法。
- (1)工程における還元酵素、又は、アルコール脱水素酵素の使用量が、一般式(I)を有する化合物及び/又は一般式(II)を有する化合物に対して、4-20重量%であり、補酵素の使用量が、0.001当量以下であることを特徴とする、請求項1-6から選択されるいずれか1項に記載の方法。
- ギ酸又はその塩が、ギ酸ナトリウムであることを特徴とする、請求項1-7から選択されるいずれか1項に記載の方法。
- 緩衝液がリン酸緩衝液であり、そのリン酸緩衝液の濃度が、50mmol以上である、請求項1-8から選択されるいずれか1項に記載の方法。
- (2)の工程における、環状酸無水物が、無水コハク酸、無水マレイン酸、又は、無水フタル酸であり、塩基が、3級アミンである、請求項3-9から選択されるいずれか1項に記載の方法。
