- Primer sets designed from transcript sequences for human beta-globin genes. Using a primer 5'-CAAGGTGAACGTGGATGAAG-3 'and a reverse primer 5'-ATTAGCCACACCAGCCACCAC-3', it was separated in the state bound to the silica membrane by the method of Example 3 (1).
- a 2 mm diameter silica membrane was taken from the entire nucleic acid portion and used for real-time reverse transcriptase chain reaction.
- Real-time reverse transcriptase polymerase chain reaction was performed under the conditions of 35 cycles and 95 ° C (20 sec) -55 ° C (40 sec) and -72 ° C (1 min).
- the globulin (beta-globin) RNA transcript was detected, and both types of real-time quantitative PCR spectrometers were able to confirm that the analysis using the separated nucleic acid by the ultrafast nucleic acid separation method according to the present invention.
