 本発明に係る生物材料の透明化方法は、尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を所定の濃度で含む溶液を、生物材料中に浸潤する第一浸潤工程、次いで、第一浸潤工程で用いた上記溶液より高濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液を、上記生物材料中に浸潤する第二浸潤工程を含んでなる。

Description

生物材料を透明化する方法、及びその利用
 本発明は、生物材料を透明化する方法、及びその利用に関する。
 光学顕微鏡を用いて不透明な組織内部を観察する際には、透明化試薬を用いた前処理(透明化処理)が行われる。
 透明化試薬及び前処理方法としては、例えば、特許文献1及び非特許文献1に記載のAnn-Shyn ChiangによるFocus Clear(商品名)溶液、或いは非特許文献2に記載のHans-Ulrich Dodtらのtissue clearing methodが代表的なものとして知られている。これらは何れも、組織を透明化することで、組織中に存在する蛍光物質を観察する目的で使用される。

 上記の方法は何れも、透明化処理の有効成分等として有機溶剤が必須であるため、生組織に用いることは難しく、適用対象が専ら固定化サンプルに限定されるという問題がある。また、透明化処理の対象となる生物材料の収縮を引き起こしうるという問題がある。
 本発明に先立ち、本願発明者らは、尿素を用いれば、生物材料を透明化することが出来ることを見出している。尿素は、生物親和性に優れた成分であるため、透明化処理の有効成分として尿素又は尿素誘導体を用いれば、上記の問題を解決しうる。
 本願発明者らは、さらに、尿素又は尿素誘導体を用いた、より一層効率的な生物材料の透明化処理方法の開発という課題に着手した。
 本願発明は、上記の課題を解決するためになされたものであり、生物親和性に優れた成分を用いた、生物材料を透明化するより効率的な方法、及びその利用を提供することを目的とする。
 上記の課題を解決するために、本願発明者らは鋭意検討を行なった。その結果、有効成分たる尿素等の濃度が異なる溶液の組み合わせを用いることで、不所望な体積変化を抑制しつつ透明化処理をより効率的に行いうることを新たに見出し、本願発明を想到するに至った。
 すなわち、本発明に係る生物材料の透明化方法は、尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を所定の濃度で含む溶液を、生物材料中に浸潤する第一浸潤工程、次いで、第一浸潤工程で用いた上記溶液より高濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液を、上記生物材料中に浸潤する第二浸潤工程を含んでなる、ことを特徴としている。
 本発明に係る透明化方法は、さらに、第二浸潤工程に次いで、当該第二浸潤工程で用いた上記溶液より低濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液を、上記生物材料中に浸潤する第三浸潤工程を含んでいてもよい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程で用いる溶液と、上記第三浸潤工程で用いる溶液とが実質的に同濃度で上記化合物を含むことが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程で用いる溶液と、上記第三浸潤工程で用いる溶液とが同じ溶液であることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程、第二浸潤工程、及び、必要に応じて行なわれる第三浸潤工程で用いる溶液が何れも上記化合物として尿素を含むことが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程、第二浸潤工程、及び、必要に応じて行なわれる第三浸潤工程で用いる溶液の少なくとも一つが水溶液であることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程、第二浸潤工程、及び、必要に応じて行なわれる第三浸潤工程で用いる溶液の少なくとも一つが界面活性剤を含むことが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記界面活性剤が非イオン性の界面活性剤であることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記非イオン性の界面活性剤が、TritonX(登録商標)、Tween(登録商標)、及びNP-40(商品名)からなる群より選択される少なくとも一種であることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記水溶液が水溶性の高分子化合物をさらに含むことが好ましい。
 本発明に係る透明化方法は、上記の方法において、水溶性の上記高分子化合物が、パーコール(登録商標)、フィコール(登録商標)、ポリエチレングリコール、及びポリビニルピロリドンからなる群より選択される少なくとも一種であることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程、第二浸潤工程、及び、必要に応じて行なわれる第三浸潤工程で用いる溶液の少なくとも一つが、さらに、グリセロール、カルボキシビニルポリマー、ヒドロキシプロピルメチルセルロース、プロピレングリコール、及びマクロゴールからなる群より選択される少なくとも一種を含むことが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記生物材料として、多細胞動物由来の組織又は器官、或いはヒトを除く多細胞動物を透明化するものであることが好ましい。
 本発明に係る透明化方法は、上記の方法において、上記第一浸潤工程で用いる溶液における上記化合物の濃度が2.5M以上で5.5M以下の範囲内であり、かつ、上記第二浸潤工程で用いる溶液における上記化合物の濃度が6M以上で8.5M以下の範囲内であることが好ましい。
 本発明に係る生物材料用透明化処理キットは、尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を所定の濃度で含む第一溶液と、上記第一溶液より高濃度で、尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む第二溶液と、からなることを特徴としている。
 本発明に係る生物材料用透明化処理キットは、上記第一溶液が、尿素を2.5M以上で5.5M以下の範囲内の濃度で、界面活性剤を0.025(w/v)%以上で5(w/v)%以下の範囲内の濃度でそれぞれ含むことが好ましい。
 本発明に係る生物材料用透明化処理キットは、上記第二溶液が、尿素を5.5M以上で8.5M以下の範囲内の濃度で、界面活性剤を0.025(w/v)%以上で5(w/v)%以下の範囲内の濃度でそれぞれ含むことが好ましい。
 本発明に係る生物材料用透明化処理キットは、上記第一溶液が、グリセロール、カルボキシビニルポリマー、ヒドロキシプロピルメチルセルロース、プロピレングリコール、及びマクロゴールからなる群より選択される少なくとも一種を含むことが好ましい。
 本発明は、生物親和性に優れた成分を用いた、より効率的な生物材料透明化法、及びその利用を提供することが出来るという効果を奏する。

 以下、本発明の実施の形態について詳細に説明する。
 〔1.生物材料用透明化試薬を用いた透明化処理方法〕
 (透明化処理方法の概要)
 本発明に係る透明化処理法は、生物材料を透明化する方法であって、以下に示す第一浸潤工程と第二浸潤工程とをこの順に含む方法である。本発明に係る透明化処理法は、さらに、第二浸潤工程の後に第三浸潤工程を含むことが好ましい。
1)尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物(尿素系化合物と総称する場合がある)を所定の濃度で含む溶液(「第一生物材料用透明化試薬」)を、生物材料中に浸潤する第一浸潤工程、
2)第一浸潤工程で用いた上記溶液より高濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液(「第二生物材料用透明化試薬」)を、上記生物材料中に浸潤する第二浸潤工程、
3)第二浸潤工程で用いた上記溶液より低濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液(「第三生物材料用透明化試薬」)を、上記生物材料中に浸潤する第三浸潤工程。
 なお、以下の説明では、第一生物材料用透明化試薬、第二生物材料用透明化試薬、及び第三生物材料透明化試薬を総称する場合は、単に「生物材料用透明化試薬」と称する。従って、単に「生物材料用透明化試薬」の説明としてなされる記載は、第一~第三生物材料透明化試薬に共通の説明に相当する。
 本発明にかかる生物材料の透明化処理方法は、透明化処理の有効成分たる尿素系化合物の濃度が異なる「生物材料用透明化試薬」を、「生物材料」に対して所定の順序で浸潤させる。このプロセスにより、生物材料の不所望な体積変化を抑制しつつ透明化処理に要する時間を大幅に短縮可能とすることを利点の一つとする。
 すなわち、上記第一浸潤工程及び第二浸潤工程をこの順序で行なうことで、生物材料の不所望な体積変化の抑制と、透明化処理に要する時間の大幅な短縮とを両立可能とする。必要に応じて、さらに上記第三浸潤工程を行なうことで、生物材料の不所望な体積変化をより一層確実に抑制可能となり、かつ周辺温度の低下時(寒冷時)であっても尿素系化合物の結晶析出等の発生によって生物材料が損傷する虞をより一層確実に抑制可能となる。
 (第一浸潤工程)
 第一浸潤工程は、上記第一生物材料用透明化試薬を、生物材料中に浸潤する工程である。より具体的には、例えば、透明化処理用の容器内で、上記の「生物材料」に対して「第一生物材料用透明化試薬」を浸潤させる。
 上記の第一浸潤工程を行う処理温度は特に限定されないが、15℃以上で45℃以下の範囲内であることが好ましい。また、第一浸潤工程を行う処理時間は、特に限定されないが、2時間以上で1ヶ月以内の範囲内であることが好ましく、12時間以上で3日以内の範囲内であることがより好ましい。また、透明化処理を行う圧力は特に限定されない。
 上記第一生物材料用透明化試薬に含まれる上記尿素系化合物の濃度は、上記第二生物材料用透明化試薬に含まれる上記尿素系化合物の濃度より低ければ特に限定されない。ただし、当該尿素系化合物の濃度は、1M以上で5.5M以下の範囲内であることが好ましく、2.5M以上で5.5M以下の範囲内であることがより好ましく、3.5M以上で4.5M以下の範囲内であることがさらに好ましく、3.7M以上で4.3M以下の範囲内であることが特に好ましい。なお、本発明において、「尿素系化合物の濃度」とは、透明化する必須の有効成分としての尿素系化合物が、生物材料用透明化試薬中に複数種含まれる場合にはその合計濃度を指す。
 (第二浸潤工程)
 第二浸潤工程は、上記第二生物材料用透明化試薬を、生物材料中に浸潤する工程である。より具体的には、例えば、透明化処理用の容器内で、上記の「生物材料」に対して「第二生物材料用透明化試薬」を浸潤させる。第二生物材料透明化試薬は、第一生物材料透明化試薬、及び、必要に応じて用いる第三生物材料透明化試薬より高濃度で上記尿素系化合物を含むことによって、生物材料に対する尿素系化合物の浸透を促進すると推定される。
 上記の第二浸潤工程を行う処理温度は特に限定されないが、15℃以上で45℃以下の範囲内であることが好ましい。また、第二浸潤工程を行う処理時間は、特に限定されないが、2時間以上で1ヶ月以内の範囲内であることが好ましく、12時間以上で3日以内の範囲内であることがより好ましい。また、透明化処理を行う圧力は特に限定されない。
 上記第二生物材料用透明化試薬に含まれる上記尿素系化合物の濃度は、上記第一生物材料用透明化試薬に含まれる上記尿素系化合物の濃度より高いという前提を満たせば特に限定されない。ただし、当該前提を満たし、かつ、5.5M以上で8.5M以下の範囲内であることが好ましく、6M以上で8.5M以下の範囲内であることがより好ましく、6.5M以上で8.5M以下の範囲内であることがさらに好ましく、7.7M以上で8.2M以下の範囲内であることが特に好ましい。また、第一生物材料用透明化試薬との濃度差という観点では、第二生物材料用透明化試薬の方が、2M以上高い濃度で上記尿素系化合物を含むことが好ましく、3M以上高い濃度で上記尿素系化合物を含むことがより好ましく、3.5M以上高い濃度で上記尿素系化合物を含むことがさらに好ましい。
 (第三浸潤工程)
 必要に応じて行われる第三浸潤工程は、上記第三生物材料用透明化試薬を、生物材料中に浸潤する工程である。より具体的には、例えば、透明化処理用の容器内で、上記の「生物材料」に対して「第三生物材料用透明化試薬」を浸潤させる。
 上記の第三浸潤工程を行う処理温度は特に限定されないが、15℃以上で45℃以下の範囲内であることが好ましい。また、第三浸潤工程を行う処理時間は、特に限定されないが、2時間以上で1ヶ月以内の範囲内であることが好ましく、12時間以上で3日以内の範囲内であることがより好ましい。また、透明化処理を行う圧力は特に限定されない。
 上記第三生物材料用透明化試薬に含まれる上記尿素系化合物の濃度は、上記第二生物材料用透明化試薬に含まれる上記尿素系化合物の濃度より低ければ特に限定されない。ただし、当該尿素系化合物の濃度は、1M以上で5.5M以下の範囲内であることが好ましく、2.5M以上で5.5M以下の範囲内であることがより好ましく、3.5M以上で4.5M以下の範囲内であることがさらに好ましく、3.7M以上で4.3M以下の範囲内であることが特に好ましい。
 また、特に好ましい態様は、第一生物材料用透明化試薬と第三生物材料用透明化試薬とで、上記尿素系化合物を実質的に同濃度で含むことがより好ましい。ここで、実質的に同濃度とは、好ましくは濃度差が0.5M以内であることを指し、より好ましくは0.3M以内であることを指し、さらに好ましくは0.1M以内であることを指す。さらに他の態様では、第一生物材料用透明化試薬と第三生物材料用透明化試薬とが同一の溶液である。
 上記の第一浸潤工程~第三浸潤工程において、透明化処理用の容器内に「生物材料用透明化試薬」と「生物材料」とを格納する順序は特に限定されない。第一浸潤工程~第三浸潤工程を同一の容器内で連続的に行う場合の一例では、まず「第一生物材料用透明化試薬」を容器内に格納し、次いで「生物材料」を格納する。次いで、第一生物材料用透明化試薬の廃液、第二生物材料用透明化試薬の容器への格納、第二生物材料用透明化試薬の廃液、第三生物材料用透明化試薬の容器への格納、の順に処理を行う。また、必要に応じて、各浸潤工程の間で、容器及び生物材料を洗浄処理等する工程を設けてもよい。
 上記の各浸潤工程で用いた、透明化処理された生物材料を格納した処理容器は、後述する観察工程に供されるまで、例えば、室温又は低温環境下で保存してもよい(透明化試料保存工程)。
 (生物材料用透明化試薬の有効成分)
 本発明では、生物材料を透明化する必須の有効成分として「尿素」を含む溶液を用いる。本発明にかかる他の態様では、生物材料を透明化する必須の有効成分として「尿素誘導体」を含む溶液を用いる。これら溶液は、上記「生物材料用透明化試薬」に相当する。
 上記尿素誘導体の種類は特に限定されないが具体的には例えば、各種のウレイン、又は、下記の一般式(1)に示す化合物である。なお、一般式(1)に示す化合物には一部のウレインが含まれる。本発明に用いる生物材料用透明化試薬は、尿素及び尿素誘導体からなる群から選択される少なくとも1種の化合物を有効成分として含んでいればよいが、これらの中で尿素を含むことがより好ましい。

 一般式(1)に示す尿素誘導体において、R1、R2、R3、R4は、互いに独立に水素原子(但し、R1~R4の全てが水素原子の場合は尿素自体に相当するので除かれる)、ハロゲン原子、又は炭化水素基であり、炭化水素基を構成する炭素原子が複数個ある場合には当該炭素原子の一部が、窒素原子、酸素原子、硫黄原子等のヘテロ原子により置換されていてもよい。炭化水素基としては、鎖状炭化水素基、及び環状炭化水素基が含まれる。
 上記鎖状炭化水素基として、例えば、鎖状アルキル基、鎖状アルケニル基、及び鎖状アルキニル基等が例示される。鎖状炭化水素基を構成する炭素数は特に限定されないが、例えば、6以下の直鎖状又は分岐状のものが挙げられ、好ましくは炭素数が1~3のアルキル基である。鎖状炭化水素基は、例えば、ハロゲン原子等の置換基を有していてもよい。鎖状アルキル基の例としては、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基、iso-ブチル基、sec-ブチル基、tert-ブチル基、ヘキシル基、オクチル基等が挙げられる。
 上記環状炭化水素基として、例えば、シクロアルキル基、及びシクロアルケニル基等が例示される。環状炭化水素基は、例えばハロゲン原子等の置換基を有していてもよい。シクロアルキル基の例としては、シクロプロピル基、シクロブチル基、シクロペンチル基、シクロヘキシル基、等の、炭素数が3以上で、好ましくは6以下のものが挙げられる。シクロアルケニル基の例としては、シクロヘキセニル基等の、炭素数が3以上で、好ましくは6以下のものが挙げられる。
 上記ハロゲン原子としては、フッ素原子、塩素原子、臭素原子、ヨウ素原子等が挙げられる。
 また、一般式(1)に示す尿素誘導体のより好ましい具体例は、以下の1)~2)に示す通りである。
1)R1~R4より選択される任意の3つの基が水素原子であり、残りの1つの基がハロゲン原子、又は炭素数1~6以下の鎖状炭化水素基である。より好ましくは残りの1つの基が、炭素数1~3、又は炭素数が1~2のアルキル基である。
2)R1~R4より選択される任意の2つの基が水素原子であり、残りの2つの基が互いに独立にハロゲン原子、又は炭素数1~6以下の鎖状炭化水素基である。より好ましくは残りの2つの基が何れも、炭素数1~3、又は炭素数が1~2のアルキル基である。なお、水素原子となる2つの基の一方はR1、R2の何れかから選ばれ、他方はR3、R4の何れかから選ばれることがより好ましい。
 (有効成分として尿素等を用いる利点)
 尿素は、毒性が極めて低い生体由来の成分である。そのため、本発明に用いる「生物材料用透明化試薬」は、1)固定化された生物材料はもとより、固定化されていない(生きた)生物材料の透明化にも用いうるものとなる。2)また、尿素は、蛍光タンパク質へのダメージ、及びその蛍光の消失が比較的少なく、蛍光タンパク質を用いた生物材料の観察にも適用しうるものとなる。3)尿素は、極めて安価で入手容易でもあり、かつ取扱い性に優れるため、極めて低コストかつ簡単な手順で透明化処理を行いうるものとなる。
 また、上記の利点に加えて、4)従来の生物材料用透明化試薬と比較して、光散乱性が高い不透明な生物材料の透明性を格段に向上させることができ、超深部組織に存在する種々の蛍光タンパク質および蛍光物質の観察が可能となる。5)特に脳組織では、これまで深部観察のバリアーであった白質層を透明化することが可能となり、白質層より深部に位置する領域(例えば脳梁)の観察が可能となる。6)本発明にかかる透明化処理は可逆的である。具体的には、透明化処理後の生物材料を平衡塩類溶液に浸漬するのみで、透明化処理前の状態に戻すことが可能である。透明化処理の前後においてタンパク質等の抗原性も変化せずに保存されるため、通常の組織染色、及び免疫染色の手法を用いた分析が可能である。
 なお、尿素に代えて上記した尿素誘導体を用いることによっても、同様の利点が得られうる。
 (界面活性剤)
 本発明で用いる上記「生物材料用透明化試薬」は、必要に応じて界面活性剤を含んでいてもよい。界面活性剤は、生物組織への本試薬の侵入を緩やかに向上させるという理由から、非イオン性の界面活性剤が好ましい。非イオン性の界面活性剤として、ポリオキシエチレンソルビタンモノラウレート、ポリオキシエチレンソルビタンモノパルミテート、ポリオキシエチレンソルビタンモノステアレート、及びポリオキシエチレンソルビタンモノオレエート等の脂肪酸系;ポリビニルアルコール等の高級アルコール系;ポリオキシエチレンオクチルフェニルエーテル等のアルキルフェノール系の界面活性剤が挙げられる。具体的には、例えば、TritonX-100、及びTritonX-140等のTritonX(登録商標)シリーズ;Tween-20、Tween-40、Tween-60、及びTween-80等のTween(登録商標)シリーズ;NP-40(商品名);からなる群より選択される少なくとも一種が挙げられる。界面活性剤は、必要に応じて、二種以上を混合して使用することもできる。
 これら界面活性剤は、尿素の、生物材料に対する浸透性を高め、透明化処理の効率を向上させうる。特に、脳組織の白質層等、のように、透明化処理が比較的困難な生物材料の透明化を行う場合には、「生物材料用透明化試薬」は界面活性剤を含むことが好ましい。
 なお、上記界面活性剤は、尿素に対するのと同様に、生物材料に対する尿素誘導体の浸透性を高めうる。
 「生物材料用透明化試薬」が界面活性剤を含む場合は、第一生物材料用透明化試薬、第二生物材料用透明化試薬、及び、必要に応じて用いる第三生物材料用透明化試薬の少なくとも一つが界面活性剤を含んでいればよい。ただし、第一~第二生物材料用透明化試薬(又は、第一~第三生物材料用透明化試薬)が何れも同じ界面活性剤を含むことが好ましく、同じ界面活性剤を実質的に同濃度で含むことがより好ましい。ここで、実質的に同濃度とは、濃度差が0.01(w/v)%以内であることを指す。
 (水溶性の高分子化合物)
 本発明で用いる上記「生物材料用透明化試薬」は、必要に応じて水溶性の高分子化合物をさらに含んでいてもよい。ここで、高分子化合物とは、分子量が例えば5万~6万程度以上の大きさであって、細胞内に実質的に侵入しないものである。また、高分子化合物は、生物材料の変性等を引き起こさないものが好ましい。水溶性の高分子化合物として、具体的には、例えば、架橋型シュークロース高分子物質、ポリエチレングリコール、ポリビニルピロリドン、又はパーコール(商品名。コロイド状シリカをポリビニルピロリドン皮膜で被覆した高分子物質)等が挙げられる。架橋型シュークロース高分子物質として、具体的には例えば、フィコール(Ficoll)PM70(商品名))のような、シュークロースをエピクロルヒドリンで架橋(共重合)した重量平均分子量約7万の高分子物質等が挙げられる。
 これら水溶性の高分子化合物は、尿素及び尿素誘導体とは異なり細胞内に浸入することが実質的になく、また水溶性であるから、細胞内外の浸透圧差の調整に寄与すると考えられる。そのため、透明化処理の対象となる生物材料の原型維持に寄与し、とりわけ、生物材料の膨張の防止に寄与する。特に限定されないが、本発明に係る「生物材料用透明化試薬」の浸透圧が比較的高い場合には、当該試薬はこれら水溶性の高分子化合物を含むことが好ましい。
 「生物材料用透明化試薬」が上記水溶性の高分子を含む場合は、第一生物材料用透明化試薬、第二生物材料用透明化試薬、及び、必要に応じて用いる第三生物材料用透明化試薬の少なくとも一つが当該高分子を含んでいればよい。ただし、第二生物材料用透明化試薬は、第一及び第三生物材料用透明化試薬より高濃度で上記尿素系化合物を含むため、生物材料の膨潤をより確実に抑制する観点では、少なくとも第二生物材料用透明化試薬が上記水溶性の高分子化合物を含むことが好ましい場合がある。なお、第一~第二生物材料用透明化試薬(又は、第一~第三生物材料用透明化試薬)が何れも上記高分子を含む場合は何れも同じ高分子を含むことが好ましく、同じ高分子を実質的に同濃度で含むことがより好ましい。ここで、実質的に同濃度とは、濃度差が0.01M以内であることを指す。
 (その他の成分)
 本発明で用いる上記「生物材料用透明化試薬」は、必要に応じて、グリセロール、カルボキシビニルポリマー、ヒドロキシプロピルメチルセルロース、プロピレングリコール、ならびにマクロゴールから選択される少なくとも一種の化合物を「乾燥抑制成分」として含むことができる。乾燥抑制成分は、透明化処理の対象となる生物材料の乾燥を防止する。とりわけ、透明化処理後に光学顕微鏡による観察に供されるまでの時間が比較的長い場合、或いは、光学顕微鏡による長時間観察に供される場合には、本発明の試薬は、上記の乾燥抑制成分を含むことが好ましい。また、上記列挙した乾燥抑制成分は、生物材料透明化試薬の浸透圧を調節し、生物材料の膨潤をある程度抑制する効果も示す。
 「生物材料用透明化試薬」が上記列挙した乾燥抑制成分を含む場合は、第一生物材料用透明化試薬、第二生物材料用透明化試薬、及び、必要に応じて用いる第三生物材料用透明化試薬の少なくとも一つが当該乾燥抑制成分を含んでいればよい。また、乾燥抑制効果に加えて、生物材料の軽微な膨潤を抑制するという観点で、第一生物材料用透明化試薬、及び必要に応じて用いる第三生物材料用透明化試薬のみに上記乾燥抑制成分を添加する場合もある。なお、第一~第二生物材料用透明化試薬(又は、第一~第三生物材料用透明化試薬)が何れも上記乾燥抑制成分を含む場合は、何れも同じ乾燥抑制成分を含むことが好ましく、同じ乾燥抑制成分を実質的に同濃度で含むことがより好ましい。
 また、本発明で用いる上記「生物材料用透明化試薬」は、上述した「尿素及び/又は尿素誘導体」、「界面活性剤」及び「水溶性の高分子化合物」以外に、例えば、pH調整剤、及び浸透圧調整剤等の添加剤を必要に応じて含んでいてもよい。
 (溶媒)
 本発明で用いる「生物材料用透明化試薬」は、尿素が可溶な溶媒を含む溶液である。溶媒の種類は、尿素が可溶な限り特に限定されないが、水を主溶媒として用いることが好ましく、水のみを溶媒として用いることが特に好ましい。なお、本発明において、「水を主溶媒として用いる」とは、使用される全溶媒に占める水の体積の割合が他の溶媒と比較して最も多いことを指し、好ましくは使用される全溶媒の体積の合計の50%を超え100%以下の量の水を用いることを指す。また、水を主溶媒として用いて調製された「生物材料用透明化試薬」を、水溶液としての「生物材料用透明化試薬」と称する。
 なお、水を主溶媒として用いた場合には、例えば、固定化した標本にはジメチルスルホキシド(DMSO)を水と混合し用いてもよい。例えば固定化した標本にDMSOを混合して用いれば、生物材料に対する試薬の浸透性の向上、及び角質表面を有する組織の透明化処理促進等の効果が期待される。
 溶媒として水を用いる主な利点は、以下の通りである。1)本発明に用いる上記「生物材料用透明化試薬」の有効成分である尿素は水への溶解性に優れているため、生物材料用透明化試薬の調製が容易かつ低コストとなる。2)有機溶剤を主溶媒として用いる場合と比較して、透明化処理時に処理対象となる生物材料の脱水を伴わない。そのため、生物材料が収縮するという問題を抑制可能となる。3)有機溶剤を主溶媒として用いる場合と比較して、蛍光タンパク質に損傷を与える可能性が著しく低減される。そのため、透明化処理を受けた生物材料を、蛍光タンパク質を用いて観察可能となる。4)固定化された材料に限定されず、生材料の透明化処理に適用可能となる。5)後述するように透明化処理が可逆的となり、透明化処理後の生体試料を必要に応じて透明化処理前の状態に戻すことができる。6)有機溶剤を主溶剤として用いる場合と比較して、取り扱いの安全性がより高くなる。
 また、本発明で用いる上記「生物材料用透明化試薬」は、透明化処理の対象となる生物材料に好適なpHの維持が可能な緩衝液であってもよい。さらに、本発明に係る「生物材料用透明化試薬」は、透明化処理の対象となる生物材料の変形を招来せず、かつ生物材料内に尿素が充分に浸透する程度に、その浸透圧が調整されていてもよい。
 なお、尿素誘導体に関しても、尿素と同様の上記溶媒を用いることができる。
 「生物材料用透明化試薬」が水溶液である場合は、第一生物材料用透明化試薬、第二生物材料用透明化試薬、及び第三生物材料用透明化試薬の少なくとも一つが水溶液であればよい。ただし、第一~第二生物材料用透明化試薬(又は、第一~第三生物材料用透明化試薬)が何れも水溶液であることが好ましい。
 (組成物の量的関係)
 本発明で用いる上記「生物材料用透明化試薬」における「尿素」の含有量は、生物材料の透明化処理が進展する限りにおいて特に限定されない。なお、「尿素」の含有量の上限、すなわち第二生物材料用透明化試薬における尿素の含有量の上限は、使用する溶媒に対する尿素の溶解度により決定される。対象とする生物材料の種類にも依存するが、例えば、「生物材料用透明化試薬」における尿素の含有量が比較的少ない場合には処理時間を長くし、尿素の含有量が比較的多い場合には処理時間を短くすることで必要な透明化処理を行うことができる。
 また、「界面活性剤」を使用する場合、その含有量は特に限定されないが、0.025(w/v)%以上で5(w/v)%以下の範囲内の濃度で含むことが好ましく、0.05(w/v)%以上で0.5(w/v)%以下の範囲内の濃度で含むことがより好ましく、0.05(w/v)%以上で0.2(w/v)%以下の範囲内の濃度で含むことが特に好ましい。なお、単位(w/v)%とは、「生物材料用透明化試薬」の体積(v(ミリリットル))に対する、使用する界面活性剤の重量(w(グラム))の百分率である。
 また、「水溶性の高分子化合物」を使用する場合、その含有量は特に限定されないが、2.5(w/v)%以上で40(w/v)%以下の範囲内の濃度で含むことが好ましい。また、生物材料の膨張抑制効果と透明化処理後の屈折率とのバランスという観点では、上記含有量は、5(w/v)%以上で25(w/v)%以下の範囲内の濃度で含むことがより好ましく、10(w/v)%以上で20(w/v)%以下の範囲内の濃度で含むことがより好ましく、10(w/v)%以上で15(w/v)%以下の範囲内の濃度で含むことが特に好ましい。なお、単位(w/v)%とは、「生物材料用透明化試薬」の体積(v(ミリリットル))に対する、使用する「水溶性の高分子化合物」の重量(w(グラム))の百分率である。
 また、グリセロール等の「乾燥抑制成分」を使用する場合、その含有量は特に限定されないが、2.5(w/v)%以上で20(w/v)%以下の範囲内の濃度で含むことが好ましく、5(w/v)%以上で15(w/v)%以下の範囲内の濃度で含むことがより好ましく、8(w/v)%以上で12(w/v)%以下の範囲内の濃度で含むことが特に好ましい。なお、単位(w/v)%とは、「生物材料用透明化試薬」の体積(v(ミリリットル))に対する、使用する「乾燥抑制成分」の重量(w(グラム))の百分率である。
 (対象となる生物材料)
 上記「生物材料用透明化試薬」を用いた透明化処理の対象となる生物材料の種類は特に限定されないが、植物由来の材料又は動物由来の材料が好ましく、魚類、両生類、爬虫類、鳥類又は哺乳類(哺乳動物)等の動物由来の材料がより好ましく、哺乳動物由来の材料が特に好ましい。また、哺乳動物の種類は特に限定されないが、マウス、ラット、ウサギ、モルモット、ヒトを除く霊長類等の実験動物;イヌ、ネコ等の愛玩動物(ペット);ウシ、ウマ等の家畜;ヒト;が挙げられる。
 また、生物材料は、個体そのものであってもよく(生きているヒト個体そのものは除く)、多細胞生物の個体から取得した器官、組織、或いは細胞であってもよい。本発明で用いる「生物材料用透明化試薬」は優れた透明化処理能力を有するので、生物材料が、多細胞動物由来の組織又は器官(例えば、脳全体又はその一部)、或いはヒトを除く多細胞動物の個体(例えば、胚等)そのものであっても透明化処理を適用可能である。
 また、上記の生物材料は、顕微鏡観察用に固定化(fixed)処理された材料であってもよく、固定化されていない材料であってもよい。なお、固定化された材料を用いる場合には、固定化処理後に、例えば20(v/w)%ショ糖-PBS溶液に、充分に(例えば、24時間以上)浸漬する処理を行うことが好ましい。さらに、当該材料をOCT compundに包埋して液体窒素で凍結後、PBS中で解凍し、4(v/w)%PFA-PBS液で再度固定する処理を行うことが好ましい。
 上記生物材料は、具体的には例えば、蛍光性化学物質を注入した生体組織、蛍光性化学物質で染色を行った生体組織、蛍光タンパク質を発現した細胞を移植した生体組織、または蛍光タンパク質を発現した遺伝子改変動物の生体組織等であってもよい。
 (生物材料用透明化試薬の調製)
 本発明で用いる「生物材料用透明化試薬」の調製方法は、「尿素及び/又は尿素誘導体」、並びに、必要に応じて用いる「界面活性剤」、「水溶性の高分子化合物」、及び「乾燥抑制成分」等を、溶媒中に溶解することで調製される。溶媒中に溶解、又は混合する手順は特に限定されない。
 (透明化処理された生物材料の観察工程)
 透明化処理された生物材料は、次いで、例えば、光学顕微鏡による観察工程に供される。観察工程に供される生物材料は、必要に応じて、上記透明化処理工程の事前に、又は透明化処理工程後で観察工程前に、染色或いはマーキング等の可視化処理工程が施されてもよい。
 例えば、可視化処理工程に蛍光タンパク質を用いる場合には、透明化処理工程の事前に、生きた生物材料に対して蛍光タンパク質遺伝子を導入して、蛍光タンパク質を発現させる。
 また、可視化処理工程として、蛍光性化学物質(蛍光タンパク質は除く)の生物材料への注入、又は蛍光性化学物質を用いた生物材料の染色を行う場合には、上記透明化処理工程の事前に行うことが好ましいが、上記透明化処理工程後に行うこともできる。さらに、可視化処理工程として、蛍光性化学物質以外の化学物質を用いた染色を行うこともできる。
 観察工程は、あらゆる種類の光学顕微鏡を用いて行うことができる。例えば、観察工程は、3次元超分解顕微鏡技術(例えば、STED、3D PALM、FPALM、3D STORM、及びSIM)を適用して行うこともできる。また、観察工程は、多光子励起型(一般的には2光子励起型)の光学顕微鏡技術を適用して行うことが好ましい。
 (その他の応用)
 上記「生物材料用透明化試薬」を用いた透明化処理は可逆的である。そのため、透明化処理された生物材料は、例えば、平衡塩類溶液に浸漬することにより、生物材料透明化試薬の成分を取り除き、透明化処理前の状態に戻すことが可能である。ここで、平衡塩類溶液とは、具体的には例えば、PBS、HBSSなどリン酸塩によって緩衝液化された平衡塩類溶液;トリス塩酸塩によって緩衝液化された平衡塩類溶液(TBS);人工脳脊髄液(ACSF);MEM, DMEM, 及びHam’s F-12などの細胞培養用の基礎培地;等が挙げられる。
 上記「生物材料用透明化試薬」を用いた場合、透明化処理の前後において、或いは、透明化処理後に透明化処理前の状態に戻す場合において、生物材料に含まれるタンパク質等の変性等を招来しない。そのため、生物材料に含まれるタンパク質等の抗原性も変化せずに保存される。そのため、例えば、生物材料を透明化処理して光学顕微鏡による観察を行った後に、当該生物材料を透明化処理前の状態に戻して汎用の組織染色、又は免疫染色の手法を用いた詳細分析を行うことなども可能となる。
 〔2.生物材料用透明化処理キット、及び処理システム〕
 (生物材料用透明化処理キット)
 本発明にかかる「生物材料用透明化処理キット」は、上記尿素系化合物を所定の濃度で含む第一溶液と、当該第一溶液より高濃度で上記尿素系化合物を含む第二溶液とを備える。ここで、第一溶液は上記第一及び第三生物材料用透明化試薬に相当し、第二溶液は上記第二生物材料用透明化試薬に相当する。また、第一・第三生物材料用透明化試薬として異なるものを用いる場合、「生物材料用透明化処理キット」は、上記尿素系化合物を第二溶液より低濃度で含み、第一溶液とは組成が異なる第三溶液を備えていてもよい。
 「生物材料用透明化処理キット」は、さらに、上記の透明化処理工程で用いる「処理容器」、「生物材料把持器具(ピンセット等)」、透明化処理後の生物材料を透明化処理前の状態に戻す「平衡塩類溶液」、及び「キットの取扱説明書」から選択される少なくとも一つを備えていてもよい。なお、キットの取扱説明書には、例えは、上記(透明化処理方法の概要)欄に記載したような、第一~第二生物材料用透明化試薬、より好ましくは第一~第三生物材料用透明化試薬を用いた透明化処理方法の手順等が記載されている。
 以下、第一溶液~第三溶液のより具体的な組成の一例を示す。これらの透明化試薬は、特に、マウス等の哺乳動物の脳全体、脳の一部(例えば、海馬、大脳全体又は大脳の一部)、及び胚を透明化処理する目的で、極めて好適である。
・第一溶液(1)、又は第三溶液(1):
 水に、尿素を2.5M以上で5.5M以下の範囲内の濃度で溶解した水溶液。より好ましくは、尿素の濃度が、3.5M以上で4.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、3.7M以上で4.3M以下の範囲内である。
・第一溶液(2)、又は第三溶液(2):
 水に、尿素を2.5M以上で5.5M以下の範囲内の濃度で、非イオン性の界面活性剤(例えば、TritonX-100)を0.05(w/v)%以上で0.2(w/v)%以下の範囲内の濃度で、さらに「乾燥抑制成分(例えば、グリセロール)」を8(w/v)%以上で12(w/v)%以下の範囲内の濃度で含ませてなる水溶液。より好ましくは、尿素の濃度が、3.5M以上で4.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、3.7M以上で4.3M以下の範囲内である。
・第一溶液(3)、又は第三溶液(3):
 水に、尿素を2.5M以上で5.5M以下の範囲内の濃度で、非イオン性の界面活性剤(例えば、TritonX-100)を0.05(w/v)%以上で0.2(w/v)%以下の範囲内の濃度で、「乾燥抑制成分(例えば、グリセロール)」を8(w/v)%以上で12(w/v)%以下の範囲内の濃度で、さらに「水溶性の高分子化合物(例えば、Ficoll)」を5(w/v)%以上で25(w/v)%以下の範囲内の濃度で含ませてなる水溶液。より好ましくは、尿素の濃度が、3.5M以上で4.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、3.7M以上で4.3M以下の範囲内である。
・第一溶液(4)、又は第三溶液(4):
 水に、尿素を2.5M以上で5.5M以下の範囲内の濃度で、非イオン性の界面活性剤(例えば、TritonX-100)を0.05(w/v)%以上で0.2(w/v)%以下の範囲内の濃度で、ジメチルスルホキシド(DMSO)を8(w/v)%以上で12(w/v)%以下の範囲内の濃度で含ませてなる水溶液。より好ましくは、尿素の濃度が、3.5M以上で4.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、3.7M以上で4.3M以下の範囲内である。
・第二溶液(1):
 水に、尿素を5.5M以上で8.5M以下の範囲内の濃度で溶解した水溶液。より好ましくは、尿素の濃度が、6M以上で8.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、7.7M以上で8.2M以下の範囲内である。
・第二溶液(2):
 水に、尿素を5.5M以上で8.5M以下の範囲内の濃度で、非イオン性の界面活性剤(例えば、TritonX-100)を0.05(w/v)%以上で0.2(w/v)%以下の範囲内の濃度で含ませてなる水溶液。より好ましくは、尿素の濃度が、6M以上で8.5M以下の範囲内であり、さらに好ましくは、尿素の濃度が、7.7M以上で8.2M以下の範囲内である。
 なお、尿素に代えて尿素誘導体(又は尿素と尿素誘導体との混合物)を用いる場合には、上記した尿素と同じ濃度で尿素誘導体等を用いて、上記第一・第三溶液(1)~(4)、及び第二溶液(1)~(2)を調製すればよい。
 (生物材料透明化処理システム)
 本発明に係る生物材料透明化処理システムは、本発明に用いる「生物材料用透明化試薬」と、単離された上記「生物材料」とを含んでなり、この「生物材料」を透明化するために当該「生物材料」の内部に「生物材料用透明化試薬」が浸潤したものである。すなわち、当該処理システムとは、例えば、透明化処理の途中段階にある生物材料を含む処理システム、又は、透明化処理が完了した生物材料を含む処理システムを包含する概念である。
 本発明について、以下の、参考例、実施例、及び比較例等に基づいてより具体的に説明するが、本発明はこれに限定されない。なお、各参考例に示す、尿素を有効成分として含む生物材料用透明化試薬は、本発明における第一~第三生物材料用透明化試薬として好適に使用できる。
 〔参考例1:海馬の観察〕
 (全身固定工程)
 神経系のニューロンに蛍光タンパク質YFPを発現したトランスジェニックマウス(YFP-H line:米国Harvard大学 Josh Sanes教授より供与されたもの。[参考文献] Feng et al. Neuron, 28: 41-51, 2000)の生後7-8週齢のものを用い、ペリスタポンプを用いて左心室より氷冷PBSを灌流した後、氷冷固定液(4% パラホルムアルデヒド-PBS, pH7.4)を灌流してマウスの全身を完全に固定した。
 (生物材料の摘出及び固定工程)
 次いで、マウスの頭蓋骨を除去して、海馬を注意深く摘出した。そして、摘出した海馬を氷冷固定液(4% パラホルムアルデヒド-PBS, pH7.4)に一晩、4℃の環境下で浸漬した。その後、この海馬を、20% シュークロース-PBS溶液中に移し24時間、4℃の環境下で緩やかに振盪した。
 次いで、海馬の組織を20% シュークロース-PBS溶液で完全に置換した後、OCT compound中に包埋し、液体窒素を用いて凍結した。そして、凍結後の海馬を、PBS中に移し室温で解凍した。解凍後の海馬は、再び、4% パラホルムアルデヒド-PBS中で1時間再固定した。
 (生物材料の透明化処理及び観察工程)
 最後に、再固定した海馬を、本参考例に係る生物材料用透明化試薬に3日間、浸漬し、室温で振盪した。生物材料用透明化試薬は、4M濃度の尿素を純水に溶解した水溶液である。透明化が進んだ時点(処理開始から3日後)で、アガロースゲル(濃度0.5%(w/v)以下)中に海馬を封入し、920nmの波長のレーザーを備えた正立型2光子顕微鏡(商品名FV1000MPE:オリンパス社製)で、海馬の基底側から表面側まで観察を行った。なお、観察に用いた対物レンズの作動距離(working distance)は2mmであった。
 観察の結果は、図1に示す。図1に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、歯状回(Dentate Gyrus:DG)、CA1、及び海馬表面までをカバーした約1.9mmの深度(生物材料の厚みに相当)にわたるクリアな観察が可能であった。
 〔参考例2:海馬の観察、及び試薬の検討〕
 上記(生物材料の透明化処理工程)で用いる生物材料用透明化試薬として、4M濃度の尿素、0.1%(w/v)濃度のTritonX-100、10%(w/v)濃度のグリセロールを純水に溶解した水溶液(SCALE-A2試薬と呼称)を用いた点、及び、海馬の表面側から基底側へ観察を行った点以外は、上記参考例1と同じ方法に従い、マウスの海馬の観察を行った。
 観察の結果は、図2に示す。図2に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、海馬表面、CA1、歯状回(DG)、及び海馬基底までをカバーした約2mmの深度(生物材料の厚みに相当)にわたるクリアな観察が可能であった。
 〔参考例3:大脳の観察〕
 以下の点1)~3)以外は、参考例1と同じ方法に従い、マウスの大脳の観察を行った。
1) 上記(生物材料の摘出及び固定工程)において大脳全体を摘出し、この大脳を固定したものを生物材料として用いた点。
2) 上記(生物材料の透明化処理及び観察工程)において、参考例2と同一のSCALE-A2試薬を生物材料用透明化試薬として用いた点。
3) 上記(生物材料の透明化処理及び観察工程)において、作動距離が3mmの対物レンズを用いて大脳の表面(大脳皮質)側から海馬側に向けて観察を行った点。
 観察の結果は、図3に示す。図3に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、大脳皮質、V-VI層はもちろん、大脳皮質下部の白質領域(corpus callosum)を越え、さらに海馬の繊維叢(fiber tract)を越えて、CA1及び歯状回(DG)等を構成する細胞群までをカバーした約3mmの深度(対物レンズの作動限界)にわたるクリアな観察が可能であった。
 なお、従来の透明化試薬では、大脳皮質下部の白質領域を透明化することが出来なかったため、白質領域より深部の領域を観察することは不可能であった。
 〔参考例4:試薬の検討〕
 以下の条件に従い、SCALE-A2試薬が生物材料を膨潤させる程度の評価、及びFicollを併用した膨潤抑制の効果について評価をした。
1)まず、透明化処理の期間を2日間とした以外は、参考例3に記載の方法に従い、SCALE-A2試薬を用いてマウスの大脳の透明化処理を行った。そして、透明化処理後の大脳の体積を測定した。なお、透明化処理の期間を2日間確保すれば、マウスの大脳は、観察可能な程度に透明化されていた。
2)また、透明化試薬として、SCALE-A2試薬にFicollを5%(w/v)濃度、或いは20%(w/v)濃度、添加したものを用いた以外は、上記1)と同様にしてマウスの大脳の透明化処理を行った。そして透明化処理後の大脳の体積をそれぞれ測定した。
3)コントロールとして、PBS中に浸漬した直後(0日間)のマウスの大脳の体積を測定した。
 観察の結果は、図4に示す。図4に示すように、コントロール(図中でPBSと示す)と比較して、上記1)の処理を行ったもの(図中でSCALE-A2と示す)は膨潤し、上記2)の処理を行ったものはFicollの添加量に応じて膨潤が抑制された。特に、20%(w/v)濃度のFicollを添加した透明化試薬を用いることで、膨潤が85%以上抑制された(図中でSCALE-A2+20%Ficollと示す)。
 なお、膨潤又は退縮が見られないものが特に好ましいが、膨潤又は退縮が見られた大脳も、光学顕微鏡観察用の試料として用いることは可能である。
 〔参考例5〕
 以下の点1)・2)以外は、参考例1と同じ方法に従い、マウスの大脳の観察を行った。
1) (生物材料の透明化処理及び観察工程)において、4M濃度の尿素、0.1%(w/v)濃度のTritonX-100、10%(w/v)濃度のグリセロール、及び10%(w/v)濃度のFicollを純水に溶解した水溶液を生物材料用透明化試薬として用い、3日間の透明化処理を行った点。
2) (生物材料の透明化処理及び観察工程)において、作動距離が2mmの対物レンズを用いて大脳の表面(大脳皮質)側から海馬側に向けて観察を行った点。
 観察の結果は、図5に示す。図5に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、大脳皮質、V-VI層はもちろん、大脳皮質下部の白質領域(corpus callosum)を越え、さらに海馬の繊維叢(fiber tract)を越えて、CA1等を構成する細胞群までをカバーした約2.16mmの深度(対物レンズの作動限界)にわたるクリアな観察が可能であった。
 なお、従来の透明化試薬では、大脳皮質下部の白質領域を透明化することが出来なかったため、白質領域より深部の領域を観察することは不可能であった。
 〔参考例6:光透過率の評価〕
 水、PBS、8M濃度の尿素水溶液(透明化試薬)、4M濃度の尿素水溶液(透明化試薬)、透明化試薬SCALE-A2(4M濃度の尿素、0.1%(w/v)濃度のTritonX-100、10%(w/v)濃度のグリセロールを含む水溶液)、及び透明化試薬SCALE-D2(4M濃度の尿素+0.1%(w/v)濃度のTriton X-100、10%(w/v)濃度のDMSOを含む水溶液)、の各々で1日間処理を行ったマウス脳切片(厚さ60μm)の大脳皮質部分を用いて、波長200nm~1000nmの光の透過率を測定した。なお、光の波長帯200nm~1000nmは、2光子顕微鏡観察で常用する励起波長領域と、観察対象となる蛍光タンパク質の主たる蛍光波長の波長領域との双方をカバーする。
 なお、光の透過率測定は、マルチチャンネル検出PMA-2(浜松ホトニクス社)を用いて行った。測定の結果は、図6に示す。同図に示すように、8M濃度の尿素水溶液、4M濃度の尿素水溶液、透明化試薬SCALE-A2、及び透明化試薬SCALE-D2は何れも光の透過率が80%以上であり、水又はPBSを用いた処理とは異なり顕著な透明化処理の効果を奏した。
 〔参考例7:マウス胎仔の透明化処理〕
 Fucci-S-Green(Fucci-S/G/M)を発現するトランスジェニックマウス(理化学研究所・脳科学総合研究センター・細胞機能探索技術開発チームが作製したマウスを自家繁殖し得たもの。参考文献Sakaue-Sawano et al., Cell, 132(3): 487-98, 2008.)の胎生13.5日目の胎仔を、4(v/w)% PFA-PBSで2日固定後に20(v/w)%ショ糖-PBS溶液に浸漬し、一旦凍結後に解凍を行った。この後、再び4(v/w)%PFA-PBSで1時間固定を行った。その後、参考例2に記載のSCALE-A2に7日間浸漬することで透明化処理を行った。
 次いで、アガロースゲル(濃度0.5%(w/v)以下)中に上記胎仔を封入し、蛍光実体顕微鏡(商品名MZ10F:Leica社製)で観察を行った。なお、観察に用いた対物レンズの作動距離は70mmであった。
 観察の結果は、図7に示す。同図に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、肝臓だけはやや黄色が残ったもののマウス胎仔の全身が顕著に透明になった。なお、同図中に示すmr(Mounting rubber)は、マウス胎仔の頭部を支えるために用いたものである。
 〔参考例8:マウス海馬における神経幹細胞の観察〕
 (全身固定工程)
 Fucci-S-Green(Fucci-S/G/M)を発現するトランスジェニックマウス(理化学研究所・脳科学総合研究センター・細胞機能探索技術開発チームが作製したマウスを自家繁殖し得たもの。[参考文献] Sakaue-Sawano et al., Cell, 132(3): 487-98, 2008.)の生後6週齢のものを用い、ペリスタポンプを用いて左心室より氷冷PBSを灌流した後、氷冷固定液(4% パラホルムアルデヒド-PBS, pH7.4)を灌流してマウスの全身を完全に固定した。
 (生物材料の摘出及び固定工程)
 次いで、マウスの頭蓋骨を除去して、海馬の歯状回を注意深く摘出した。そして、摘出した海馬の歯状回を氷冷固定液(4% パラホルムアルデヒド-PBS, pH7.4)に一晩、4℃の環境下で浸漬した。その後、この海馬の歯状回を、20% シュークロース-PBS溶液中に移し24時間、4℃の環境下で緩やかに振盪した。
 次いで、海馬の歯状回を20% シュークロース-PBS溶液で完全に置換した後、OCT compound中に包埋し、液体窒素を用いて凍結した。そして、凍結後の海馬の歯状回を、PBS中に移し室温で解凍した。解凍後の海馬の歯状回は、再び、4% パラホルムアルデヒド-PBS中で1時間再固定した。
 なお、本参考例で用いた海馬の歯状回は、レクチン-テキサスレッド(Lectin-Texas Red)コンジュゲートによりその血管が染色されている。
 (生物材料の透明化処理及び観察工程)
 最後に、再固定した海馬の歯状回を、本参考例に係る生物材料用透明化試薬に3日間、浸漬し、室温で振盪した。生物材料用透明化試薬は、4M濃度の尿素、0.1%(w/v)濃度のTritonX-100、10%(w/v)濃度のグリセロールを純水に溶解した水溶液である。透明化が進んだ時点(処理開始から3日後)で、アガロースゲル(濃度0.5%(w/v)以下)の薄膜中に海馬の歯状回を封入し、920nmの波長のレーザーを備えた正立型2光子顕微鏡(商品名FV1000MPE:Olympus社製)で観察を行った。なお、観察に用いた対物レンズの作動距離(working distance)は2mmであった。また、観察は、海馬腹側表面から1.7 mmの深さまで18個の連続視野について行い、当該観察の結果を統合画像として再構成した。
 観察の結果は、図8に示す。図8に示すように、本参考例に係る生物材料用透明化試薬で透明化処理を行うことで、Fucci-S-Green(Fucci-S/G/M)陽性の神経幹細胞が血管周囲に見られることが確認された。
 〔参考例9:神経幹細胞移植の観察〕
 蛍光タンパクVenusを発現させた大人のラット海馬由来の神経幹細胞を取得し、この神経幹細胞をラット系統Fisher(F344)(日本SLCより購入)の大脳皮質部分(第V層付近、深さ約1.7 mmの部位)に移植した。
 神経幹細胞の移植後、3日が経過したラット系統Fisher(F344)を、上記参考例8の(全身固定工程)に記載の方法に従い、固定をした。次いで、上記参考例8の(生物材料の摘出及び固定工程)に記載の方法に従い、ラット系統Fisher(F344)の大脳皮質を固定した。なお、本参考例で用いたラットの大脳皮質は、レクチン-テキサスレッド(Lectin-Texas Red)コンジュゲートによりその血管が染色されている。
 次いで、上記参考例8の(生物材料の透明化処理及び観察工程)に記載の方法に従い、大脳皮質の観察を行った。観察の結果は、図11に示す。同図に示すように、本参考例に係る生物材料透明化試薬で透明化処理を行うことで、蛍光タンパクVenusを発現した神経幹細胞が血管(Lectin Texas-Redコンジュゲートで染色されている)付近に集まっている様子が観察された。本方法は移植後の細胞の移動及び形態変化などを観察することにも適している。
 〔参考例10:BABB法との比較(1)〕
 神経系に蛍光タンパク質YFPを発現したトランスジェニックマウス(YFP-H line:米国Harvard大学 Josh Sanes教授より供与されたもの。参考文献: Feng et al. Neuron, 28: 41-51, 2000)の生後12週齢のものから大脳を摘出して、SCALE-A2試薬(参考例2参照)を用いた透明化処理と、非特許文献2に記載のDodtらによるベンジルベンゾエート/ベンジルアルコール(benzylbenzoate / benzylalchol (BABB))法による透明化処理との比較を行った。
 なお、透明化処理は、何れも2日間、大脳を透明化処理液に浸漬することにより行った。また、上記BABB法で用いた透明化処理液の組成は、benzylalchol、およびbenzylbenzoate(両者の組成比=1:2)であった。
 結果を図9に示す。同図中で、Transmissionと示す列及びYFPと示す列の上段はSCALE-A2試薬による処理結果であり、下段はBABB法による処理である。なお、「Transmission」は透過型顕微鏡による観察結果を、「YFP」は蛍光顕微鏡を用いたYFPからの蛍光を観察した結果を示す。
 図9に示すように、SCALE-A2試薬による透明化処理では、YFPが発する蛍光を明確に観察することが可能であった。一方、BABB法による透明化処理を行った大脳は著しく縮小し、かつYFPからの蛍光が著しく減弱した。
 〔参考例11:BABB法との比較(2)〕
 各種の蛍光タンパク質をコードする遺伝子をHeLa細胞(東京大学医科学研究所より分与されたもの。Scherer et al., J. Exp. Med. 97(5):695-710, 1953)に導入して対応する蛍光タンパク質を発現させた。ここで、発現させた蛍光タンパク質は、SECFP、EGFP、mAG1、Venus、及びDsRedである。EGFPおよび DsRedはClontech社より購入。mAG1はMBL社より購入。VenusおよびSECFPは理化学研究所・脳科学総合研究センター・細胞機能探索技術開発チームがGFPをもとに遺伝子的に変異を導入し作製した。参考文献:SECFP, Hadjantonakis et al., BMC Biotechnol., 2:11, 2002.: EGFP, Zhang et al. Biochem. Biophys. Res. Commun., 227: 707-711, 1996.: Venus, Nagai et al., Nat. Botech., 20: 87-90, 2002.: mAG1, Karasawa et al., J. Biol. Chem., 278(36): 34167-34171, 2003.: Baird et al., PNAS., 97(22): 11984-11989, 2000.。
 次いで、参考例10と同様にして、SCALE-A2試薬(参考例2参照)を用いた透明化処理と、非特許文献2に記載のDodtらによるベンジルベンゾエート/ベンジルアルコール(benzylbenzoate / benzylalchol (BABB))法による透明化処理とを行い、両者を比較した。
 観察の結果は、図10に示す。同図では、透明化処理前の蛍光タンパク質由来の蛍光強度を100とした、各透明化処理後の蛍光タンパク質由来の蛍光強度の相対値を示している。図10より明らかな通り、SCALE-A2試薬による透明化処理では蛍光タンパク質由来の蛍光強度は何れの場合も80%以上維持されたが、BABB法による透明化処理では蛍光タンパク質由来の蛍光強度は何れの場合も顕著に減弱させた。
 〔参考例12:透明化処理後の組織の回復〕
 GFPトランスジェニックマウスおよびYFPトランスジェニックマウス(GFP-M lineおよびYFP-H line, 米国ハーバード大学Josh Sanes教授より供与されたもの)の海馬より2.5 mm厚のスライスを作製し、一旦SCALE-A2試薬(参考例2参照)による透明化処理を5日間行って透明化した後、PBSで3回リンスして不透明状態の組織に戻した際の免疫染色性を、SCALE-A2試薬による透明化処理前のものと比較した。
 免疫染色には幼若な神経細胞の表面に存在するPSA-NCAMのポリシアル酸を認識するマウス由来抗PSA-NCAM(polysialilated neural cell adhesion molecule)モノクローナル抗体(Millipore社)と、アストログリアに特異的な中間径フィラメントGFAP(glial fibrillary acidic protein)を認識するウサギ由来抗GFAPポリクローナル抗体(Sigma社)とを1次抗体として用い、24時間4℃で反応させた後にPBSでリンスした。その後、抗PSA-NCAMモノクローナル抗体は、Alexa Fluor 546で標識された抗マウスIgM抗体(Invitrogen, Molecular Probes社)を、GFAPポリクローナル抗体はAlexa Fluor 633で標識された抗ウサギIgG抗体を2次抗体(Invitrogen, Molecular Probes社)として室温で3時間反応させることで免疫組織染色を行った。これらのスライスの蛍光はこの間減衰することはなかったため、GFPあるいはYFPの蛍光を含めた3重蛍光像が得られた。観察は倒立型共焦点レーザー顕微鏡(FV500, Olympus社)を用いて20倍の対物レンズ(UplanApo20, Olympus社)により行った。
 図12に示すように、SCALE-A2の処理前(図中でBefore SCALE)、処理後(図中でAfter Recovery)ともに、抗PSA-NCAMモノクローナル抗体によって、海馬歯状回に存在する幼若な神経細胞とその細胞からCA3領域へと伸長した苔状線維とが実質的な問題なく染色された。また同様に、抗GFAPポリクローナル抗体によって、アストログリアも問題なく染色された。
 〔実施例1:大脳の観察〕
 (生物材料の摘出及び固定工程)
 全身麻酔によって眠らせたマウスを開腹開胸し、左心室より4% パラホルムアルデヒド-PBS溶液を注入して全身を灌流固定した後、大脳を摘出した。摘出した大脳を氷冷固定液(4% パラホルムアルデヒド-PBS溶液)に浸漬して8~12時間固定を行った。その後、この大脳を20% シュークロース-PBS溶液に2日間浸漬して完全に置換をした。
 次いで、この大脳をOCT compound中に包埋し、液体窒素を用いて凍結した。そして、凍結した大脳をOCT compoundごとPBS中で振盪しながら解凍した。PBSを交換しリンスを行った後、約30分、4% パラホルムアルデヒド-PBSを用いて大脳の再固定を行った。
 (透明化処理工程)
 次いで、再固定後の大脳を、参考例2で示す生物材料用透明化試薬(SCALE-A2)中に移し2日間、室温で振盪した(第一浸潤工程)。次いで、より高濃度で尿素を含む生物材料用透明化試薬(試薬Bとする)中に大脳を移し、2日間、室温で振盪した(第二浸潤工程)。試薬Bは、8M濃度の尿素、及び0.1%(w/v)濃度のTritonX-100を純水に溶解した水溶液である。次いで、新鮮なSCALE-A2液中に大脳を移し、2~3日間、室温で振盪して透明化した(第三浸潤工程)。なお、大脳の湿重量1gに対して用いるSCALE-A2の液量ならびに試薬Bの液量はそれぞれ約30mlであった。
 上記のように、尿素の濃度が高い試薬Bでの透明化処理を中間に挟む方法により、SCALE-A2を単独で用いて透明化処理を行う場合と比較して、極めて短い処理時間(凡そ半分程度)で同等の透明化度を達成することができた。
 〔実施例2:海馬の観察〕
 以下の点1)~3)以外は、参考例1と同じ方法に従い、マウスの海馬の観察を行なった。
1) 生後6週齢の野生型(非遺伝子改変)の正常C57BL6系統雄マウスを用いた点。
2) (生物材料の透明化処理及び観察工程)において、透明化処理を次のように行なった点。まず、再固定した海馬を、SCALE-A2試薬(参考例2)で48時間、浸漬し、室温で振盪した後に(第一浸潤工程)、次いで、試薬B(実施例1)で48時間、浸漬し、室温で振盪した。
3) 観察工程は、模様つきの基板上に海馬を配置して、模様が海馬を介して透けて見えるか否かで透明化度を判定した。
 なお、参考例として、再固定した海馬を、PBSで96時間、浸漬し、室温で振盪したものと、SCALE-A2試薬で96時間、浸漬し、室温で振盪したものも準備した。
 観察の結果は、図13に示す。同図中のAは、PBSで海馬を処理したもの(参考例)、Bは、SCALE-A2試薬で海馬を処理したもの(参考例)、Cは、SCALE-A2試薬と試薬Bとで海馬を処理したもの(実施例)に相当する。同図に示すように、同じ処理時間で比較した場合、実施例は何れの参考例と比較しても透明化度が明らかに高くなっていることが確認された。また、処理前の海馬の大きさは何れもほぼ同じであったので、同図中のBに示す海馬と比較してCに示す海馬がより体積膨張をしたということもなかった。
 〔実施例3:脳切片の観察〕
 以下の点1)~3)以外は、参考例1と同じ方法に従い、マウスの脳切片の観察を行なった。
1) 生後6週齢の野生型(非遺伝子改変)の正常C57BL6系統雄マウスを用いた点。
2) (生物材料の摘出及び固定工程)において、マウスの脳全体を摘出して、参考例1と同様に固定及び再固定した点。
3) (生物材料の透明化処理及び観察工程)において、透明化処理を次のように行なった点。まず、再固定した脳から海馬を含む冠状断スライス(厚さ3mm)を作成した。次いで、この冠状断スライスを、中央線で右半球側スライスと左半球側スライスとに切断した。そして一方の半球側スライスをSCALE-A2試薬で(参考例2)で24時間、浸漬し、室温で振盪した後に(第一浸潤工程)、次いで、試薬B(実施例1)で24時間、浸漬し、室温で振盪した(第二浸潤工程)。次いで、この冠状断スライスから、海馬を含む厚さ1.5mmの冠状断スライスを切り出した。このようにして得た冠状断スライスを一度目の観察工程(次の段落参照)に供した。
次いで、一度目の観察工程を終えた冠状断スライスをSCALE-A2試薬で二度リンスした後、SCALE-A2試薬で24時間、浸漬し、室温で振盪した(第三浸潤工程)。このようにして得た冠状断スライスを二度目の観察工程(次の段落参照)に供した。
3) 観察工程は、分光光度計(日立製:製品名U-3310 Spectrophotometer)を用いて、1.5mm厚の上記冠状断スライスの、300nm~920nmの波長域の光の透過スペクトルを測定した。
 なお、参考例として、再固定した脳から海馬を含む冠状断スライス(厚さ3mm)を作成した。次いで、この冠状断スライスを、中央線で右半球側スライスと左半球側スライスとに切断した。そして一方の半球側スライスをSCALE-A2試薬で72時間、浸漬し、室温で振盪した後に得た、海馬を含む厚さ1.5mmの冠状断スライスを準備した。
 観察の結果は、図14に示す。同図中の「B」は、本実施例の一度目の観察の結果であり、「B-A2 replaced」は二度目の観察の結果であり、「SCALE-A2」は参考例の結果である。同図に示すように、実施例は、参考例と比較して、同一又はより短い処理時間で、透明化度が明らかに高くなっている(約2倍)ことが確認された。
 なお、データは示さないが、SCALE-A2試薬、試薬B、及び水は、300nm~920nmの波長域の光の透過スペクトルがほぼ等しい。
 〔参考例13:尿素濃度が脳組織の透明化に及ぼす影響〕
 8週齢の野生型C57BL6/J系統マウス(日本エスエルシーより購入)の大脳のスライスを、PBS、4M、6M、又は8Mの尿素を含む水溶液中に浸漬し、室温で12時間振盪することでインキュベーションした。
 大脳スライスは、まず、アクリル製のbrain slicer(室町機械製)を用いて、厚さ1mmで、大脳の前後方向に連続したものを2枚調製した。次いで、2枚のスライスを左右にほぼ均等に分断して4つのスライスとした。そして、4つのスライスそれぞれを上記した各種水溶液に浸漬してインキュベーションした。結果を図15に示す。PBSの場合は組織の透明化が生じていないことが認められた。これに対して8M尿素を含む溶液の場合は組織に膨潤があるものの、背景のパターンが最もよく組織を透過して確認することができた。4Mから8Mの濃度範囲では尿素濃度に依存的に透明化が生じることが認められた。
 本発明は上述した各実施形態及び実施例に限定されるものではなく、請求項に示した範囲で種々の変更が可能であり、異なる実施形態にそれぞれ開示された技術的手段を適宜組み合わせて得られる実施形態についても本発明の技術的範囲に含まれる。
 本発明は、生物親和性に優れた成分を用いた、より効率的な生物材料透明化法、及びその利用を提供することが出来る。

Claims (18)

  1.  生物材料を透明化する方法であって、
     尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を所定の濃度で含む溶液を、生物材料中に浸潤する第一浸潤工程、
     次いで、第一浸潤工程で用いた上記溶液より高濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液を、上記生物材料中に浸潤する第二浸潤工程を含んでなる、ことを特徴とする生物材料の透明化方法。
  2.  上記第二浸潤工程に次いで、当該第二浸潤工程で用いた上記溶液より低濃度で尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む溶液を、上記生物材料中に浸潤する第三浸潤工程を含んでなることを特徴とする請求項1に記載の生物材料の透明化方法。
  3.  上記第一浸潤工程で用いる溶液と、上記第三浸潤工程で用いる溶液とが実質的に同濃度で上記化合物を含むことを特徴とする請求項2に記載の生物材料の透明化方法。
  4.  上記第一浸潤工程で用いる溶液と、上記第三浸潤工程で用いる溶液とが同じ溶液であることを特徴とする請求項3に記載の生物材料の透明化方法。
  5.  上記溶液が何れも上記化合物として尿素を含むことを特徴とする請求項1から4の何れか一項に記載の透明化方法。
  6.  上記溶液の少なくとも一つが水溶液であることを特徴とする請求項1から5の何れか一項に記載の透明化方法。
  7.  上記溶液の少なくとも一つが界面活性剤を含むことを特徴とする請求項1から6の何れか一項に記載の透明化方法。
  8.  上記界面活性剤が非イオン性の界面活性剤であることを特徴とする請求項7に記載の透明化方法。
  9.  上記非イオン性の界面活性剤が、TritonX(登録商標)、Tween(登録商標)、及びNP-40(商品名)からなる群より選択される少なくとも一種であることを特徴とする請求項8に記載の透明化方法。
  10.  上記水溶液が水溶性の高分子化合物をさらに含むことを特徴とする請求項6から9の何れか一項に記載の透明化方法。
  11.  水溶性の上記高分子化合物が、パーコール(登録商標)、フィコール(登録商標)、ポリエチレングリコール、及びポリビニルピロリドンからなる群より選択される少なくとも一種であることを特徴とする請求項10に記載の透明化方法。
  12.  上記溶液の少なくとも一つが、さらに、グリセロール、カルボキシビニルポリマー、ヒドロキシプロピルメチルセルロース、プロピレングリコール、及びマクロゴールからなる群より選択される少なくとも一種を含むことを特徴とする請求項1から11の何れか一項に記載の透明化方法。
  13.  上記生物材料として、多細胞動物由来の組織又は器官、或いはヒトを除く多細胞動物を透明化するものであることを特徴とする請求項1から12の何れか一項に記載の透明化方法。
  14.  上記第一浸潤工程で用いる溶液における上記化合物の濃度が2.5M以上で5.5M以下の範囲内であり、かつ、上記第二浸潤工程で用いる溶液における上記化合物の濃度が6M以上で8.5M以下の範囲内であることを特徴とする請求項1から13の何れか一項に記載の透明化方法。
  15.  生物材料用透明化処理キットであって、
     尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を所定の濃度で含む第一溶液と、
     上記第一溶液より高濃度で、尿素及び尿素誘導体からなる群から選択される少なくとも一種の化合物を含む第二溶液と、からなることを特徴とする生物材料用透明化処理キット。
  16.  上記第一溶液が、尿素を2.5M以上で5.5M以下の範囲内の濃度で、界面活性剤を0.025(w/v)%以上で5(w/v)%以下の範囲内の濃度でそれぞれ含むことを特徴とする請求項15に記載の生物材料用透明化処理キット。
  17.  上記第二溶液が、尿素を5.5M以上で8.5M以下の範囲内の濃度で、界面活性剤を0.025(w/v)%以上で5(w/v)%以下の範囲内の濃度でそれぞれ含むことを特徴とする請求項15又は16に記載の生物材料用透明化処理キット。
  18.  上記第一溶液が、グリセロール、カルボキシビニルポリマー、ヒドロキシプロピルメチルセルロース、プロピレングリコール、及びマクロゴールからなる群より選択される少なくとも一種を含むことを特徴とする請求項15から17の何れか一項に記載の生物材料用透明化処理キット。
