本発明の、検体中のHDL3中のコレステロール(以下、HDL3-Cと記す)の測定方法は、遠心分離などの物理的方法によるリポ蛋白の分離除去を必要としない方法であり、また、HDL3-Cの測定に先立って、検体中のHDL3以外のリポ蛋白中のコレステロールを消去することなく、検体中のHDL3-Cを測定する方法である。
(1)検体と、コレステロール測定用酵素との反応を、(a)2価の金属塩、(b)硫酸塩、硝酸塩、炭酸塩、酢酸塩、及び、ハロゲン化物からなる群より選ばれるアルカリ金属塩、及び、(c)デキストラン硫酸又はその塩を含有する水性媒体中で反応させる工程;
(2)HDL3以外のリポ蛋白を分離除去することなく、工程(1)の反応により生成する物質又は消費される物質を測定する工程;
(3)予め、既知濃度のHDL3-Cを用いて作成された、HDL3-C濃度と、該生成する物質又は該消費される物質由来の情報量との関係を表す検量線と、上記(2)での測定値とを相関付ける工程;及び、
(4)検体中のHDL3-C濃度を決定する工程
を含む。ここで、工程(1)におけるコレステロール測定用酵素としては、前述のコレステロール測定用酵素等が挙げられる。
本発明のHDL3-Cの測定方法における反応時間は、本発明のHDL3-Cの測定方法を可能とする反応時間であれば特に制限はないが、1~60分間が好ましく、2~30分間がより好ましい。
HDLは、HDL2とHDL3の2つの亜分画からなるリポ蛋白であるので、検体中のHDLコレステロール(総HDLコレステロール)を測定し、当該総HDLコレステロール濃度から、本発明のHDL3-Cの測定方法により測定される検体中のHDL3-C濃度を差し引くことにより、当該検体中のHDL2コレステロール(以下、HDL2-Cと記す)濃度を測定することができる。
(1)検体中のHDL中のコレステロールを測定する工程;
(2)本発明のHDL3-Cの測定方法により、検体中のHDL3-Cを測定する工程;
(3)上記(1)工程での測定の測定値から、(2)工程での測定の測定値を差し引く工程。
工程(2)におけるHDL3-Cの測定は、前述のHDL3-C測定方法により行うことができる。
本発明のHDL3-C測定用試薬は、本発明のHDL3-C測定方法に用いられる試薬である。
本発明のHDL3-C測定用試薬の態様を以下に記す。

HEPES緩衝液(pH8.0,0.1mol/L)にCOO322を50g/Lとなるように加え、15℃に冷却した後、サンブライトVFM-4101(日油社製)を6.25g/Lとなるように加え、さらに2時間反応させた。得られた修飾酵素溶液を精製分離せずそのまま化学修飾COO322として使用した。
以下の第一試薬及び第二試薬からなるHDL3-C測定用キットを調製した。第1表に示す濃度の2価の金属塩(硝酸マグネシウム6水和物又は塩化カルシウム2水和物)、及び、硫酸ナトリウムを含むキットを実施例1(1)~1(11)のキットとした。

日立7170S形自動分析装置を用いて、以下の操作により「反応吸光度」を算出した。
上記(1)と同じヒト血清検体を用いて、Journal of Lipid Research vol.49, p.1130-1136 (2008)に記載の方法(分画法)にて各検体中のHDL3を分離し、得られたHDL3分画中のコレステロール量をデタミナーL TCII(協和メデックス社製)を用いて測定した。
実施例1(1)のキットを用いた測定における「反応吸光度」と、(2)の分画法による測定値との間の相関係数を第1表に記す。
同様に、実施例1(1)のキットの代わりに、実施例1(2)~1(11)のキットを用いて、分画法による測定値との間の相関係数を決定した。相関関係を第1表に記す。
実施例1(1)の代わりに、比較例1(1)~1(3)の各キットを用いる以外は実施例2の方法と同様の方法により、比較例1(1)~1(3)の各キットを用いた測定における「反応吸光度」と、分画法による測定値との間の相関係数を決定した。相関係数を第1表に記す。
以下の第一試薬及び第二試薬からなるHDL3-C測定用キットを調製した。第2表に示す濃度の硝酸マグネシウム6水和物、及び、硫酸ナトリウムを含むキットを実施例3(1)~3(12)のキットとした。

ヒト新鮮血清5検体について、各検体中のHDL3-C濃度を、分画法、及び、本発明の実施例3(6)及び実施例3(11)のキットを用いる方法により、以下の手順に従い決定した。
Journal of Lipid Research vol.49, p.1130-1136(2008)に記載の方法(分画法)にて各検体中のHDL3を分離し、得られたHDL3分画中のコレステロールをデタミナーL TCII(協和メデックス社製)を用いて測定し、各検体中のHDL3-C濃度を決定した。
分画法による測定より、HDL3-C濃度が16.1mg/dLであることが判明している血清標準液を検量線作成用サンプルとした。実施例2の(1)記載の測定方法と同様にして日立7170S自動分析装置により、実施例3(6)のキットを用いて、検量線作成用サンプル検体の反応吸光度を測定し、HDL3-C濃度と反応吸光度との間の関係を示す検量線を作成した。
分画法を用いて決定されたHDL3-C濃度、実施例3(6)及び実施例3(11)のキットを用いて決定されたHDL3-C濃度を第4表に示す。
Claims (19)
- 検体と、(1)コレステロールエステル加水分解酵素及びコレステロール酸化酵素の組み合わせ、又は、(2)コレステロールエステル加水分解酵素、酸化型補酵素及びコレステロール脱水素酵素の組み合わせとを、(a)2価の金属塩、(b)硫酸塩、硝酸塩、炭酸塩、酢酸塩、及び、ハロゲン化物からなる群より選ばれるアルカリ金属塩、及び、(c)デキストラン硫酸又はその塩を含有する水性媒体中で反応させ、HDL3以外のリポ蛋白を分離除去することなく、生成する物質又は消費される物質を測定することを特徴とする、検体中のHDL3中のコレステロールの測定方法。
- 2価の金属塩がマグネシウム塩又はカルシウム塩である請求項1記載の方法。
- デキストラン硫酸又はその塩の反応液中の濃度が、0.75~2.6g/Lである請求項1または2記載の方法。
- 反応液中の2価の金属塩由来の2価の金属イオンの濃度が、12~20mmol/Lであり、反応液中のアルカリ金属塩由来のアルカリ金属イオンの濃度が、5~21mmol/Lである、請求項1~3のいずれかに記載の方法。
- 以下の工程を含むことを特徴とする、検体中のHDL2中のコレステロールの測定方法。

- 請求項1~4のいずれかに記載の測定方法により、HDL3以外のリポ蛋白を分離除去することなく、検体中のHDL3中のコレステロールを測定するための試薬であって、コレステロールエステル加水分解酵素、酸化型補酵素、コレステロール脱水素酵素、(a)2価の金属塩、(b)硫酸塩、硝酸塩、炭酸塩、酢酸塩、及び、ハロゲン化物からなる群より選ばれるアルカリ金属塩、及び、(c)デキストラン硫酸又はその塩を含有することを特徴とする、検体中のHDL3中のコレステロールの測定用試薬。
- さらに、還元型補酵素測定用試薬を含む、請求項7記載の試薬。
- 2価の金属塩がマグネシウム塩又はカルシウム塩である請求項6~8のいずれかに記載の試薬。
- デキストラン硫酸又はその塩が、反応液中での濃度が0.75~2.6g/Lとなる含量で含まれる請求項6~9のいずれかに記載の試薬。
- 2価の金属塩が、反応液中での2価の金属塩由来の2価の金属イオンの濃度が12~20mmol/Lとなる含量で含まれ、アルカリ金属塩が、反応液中でのアルカリ金属塩由来のアルカリ金属イオンの濃度が5~21mmol/Lとなる含量で含まれる請求項6~10のいずれかに記載の試薬。
- 第一試薬および第二試薬を含有する、請求項1~4のいずれかに記載の測定方法により、HDL3以外のリポ蛋白を分離除去することなく、検体中のHDL3中のコレステロールを測定するためのキットであって、(a)2価の金属塩、(b)硫酸塩、硝酸塩、炭酸塩、酢酸塩、及び、ハロゲン化物からなる群より選ばれるアルカリ金属塩、及び、(c)デキストラン硫酸又はその塩を第一試薬に含有し、コレステロール酸化酵素を第二試薬に含有し、過酸化水素測定用試薬を第一試薬、第二試薬のいずれか又は両方に含有し、コレステロールエステル加水分解酵素を第一試薬、第二試薬のいずれか又は両方に含有することを特徴とするキット。
- 第一試薬および第二試薬を含有する、請求項1~4のいずれかに記載の測定方法により、HDL3以外のリポ蛋白を分離除去することなく、検体中のHDL3中のコレステロールを測定するためのキットであって、(a)2価の金属塩、(b)硫酸塩、硝酸塩、炭酸塩、酢酸塩、及び、ハロゲン化物からなる群より選ばれるアルカリ金属塩、及び、(c)デキストラン硫酸又はその塩を第一試薬に含有し、コレステロール脱水素酵素を第二試薬に含有し、酸化型補酵素を第一試薬、第二試薬のいずれか又は両方に含有し、コレステロールエステル加水分解酵素を第一試薬、第二試薬のいずれか又は両方に含有することを特徴とするキット。
- さらに、還元型補酵素測定用試薬を第一試薬、第二試薬のいずれか又は両方に含有する請求項13記載のキット。
- 2価の金属塩が、マグネシウム塩又はカルシウム塩である請求項12~14のいずれかに記載のキット。
- デキストラン硫酸又はその塩が、反応液中での濃度が0.75~2.6g/Lとなる含量で含まれる請求項12~15のいずれかに記載のキット。
- 2価の金属塩が、反応液中での2価の金属塩由来の2価の金属イオンの濃度が、12~20mmol/Lとなる含量で含まれ、アルカリ金属塩が、反応液中でのアルカリ金属塩由来のアルカリ金属イオンの濃度が、5~21mmol/Lとなる含量で含まれる請求項12~16のいずれかに記載のキット。
- 請求項6~11のいずれかに記載のHDL3中のコレステロール測定用試薬と、HDLコレステロール測定用試薬とを含むことを特徴とする、検体中のHDL2中のコレステロールの測定用キット。
- 請求項12~17のいずれかに記載のHDL3中のコレステロール測定用キットの第一試薬及び第二試薬と、HDLコレステロール測定用試薬とを含むことを特徴とする、検体中のHDL2中のコレステロールの測定用キット。
