センサチップ(300)を用意する工程(a)、ここで、

ここで、前記ハロゲン化銀は、前記ストリッピング電極(34)の表面上に堆積し、
前記第1作用電極(31a)および前記第2作用電極(31b)に電位が印加されない状態でストリッピング電極(34)に電位を印加し、ストリッピング電極(34)を流れた電流量を測定する工程(d)、および
前記電流量に基づいて前記酸化還元物質(還元体)の濃度を算出し、前記算出された濃度に基づいて前記化学物質を定量する工程(e)。
まず、センサチップ300が用意される。
図3は、実施の形態1によるセンサチップ300を示す。センサチップ300は、基板30、一対のくし型作用電極31a/31b、参照極32、対極33、ストリッピング電極34、ストリッピングゲル35、保護ゲル36、およびカバー37を具備している。カバー37は、試料溶液を注入するための注入口38および空気孔39を具備する。
一対のくし型作用電極31a/31bは、第1作用電極31aおよび第2作用電極31bから構成される。
参照極32は、必要に応じて具備される。高い定量精度を考慮すれば、センサチップ300は参照極32を具備することが好ましい。
ストリッピングゲル35は、基準電解質を含有する疎水性もしくは親水性イオン液体をゲル化して得られる。すなわち、ストリッピングゲル35は基準電解質および疎水性もしくは親水性イオン液体を含有する。疎水性もしくは親水性イオン液体は支持電解質として機能する。疎水性もしくは親水性イオン液体をゲル化する方法は特に限定されない。例として、高分子化合物がゲル化のために用いられる。高分子化合物の例は、フッ化ビニリデン‐ヘキサフルオロプロピレン共重合体、ポリメチルメタクリレート、ポリアクリロニトリル、またはポリブチルアクリレートである。
保護ゲル36は、ストリッピングゲル35を覆う。すなわち、ストリッピングゲル35は、保護ゲル36およびストリッピング電極34の間に挟まれる。図3に示すように、ストリッピングゲル35が保護ゲル36によって完全に隠されるように、保護ゲル36はストリッピングゲル35の全体を覆うことが好ましい。
保護ゲル36は、ストリッピングゲル35と同様、疎水性イオン液体をゲル化して得られる。保護ゲル36は、基準電解質を含有しない。ストリッピングゲル35と同様、疎水性イオン液体は支持電解質として機能する。保護ゲル36は、親水性イオン液体を含有しない。

(1)1-R1-2-R2-3-R3-イミダゾリウムイオン(R1は水素または1以上6以下の炭素数を有する低級アルキル基であり、R2は水素またはメチル基であり、R3はアルキル基であり、R3はヘテロ原子を含んでもよい)、

N-ヘキシルピリジニウム クロライド
溶解度の観点から、基準電解質は、ストリッピングゲルを構成する疎水性もしくは親水性イオン液体のカチオンと同一または類似のカチオンを有することが好ましい。例えば、イオン液体が1-ブチル-3-メチルイミダゾリウムビストリフルオロメタンスルフォニルイミドである場合は、基準電解質は1-ブチル-3-メチルイミダゾリウムハライドであることが好ましい。
本実施形態によるセンサチップを作製する手順が以下、説明される。
フォトレジストにより被覆された絶縁性基板が、パターン作製用のマスクを通して紫外光に照射され、続いてアルカリ現像を行う。パターンが形成された基板上に、金属がスパッタされる。有機溶媒中で残りのフォトレジストが溶解され、不要な金属を除去する。基板の全面が絶縁膜により被覆される。電極上の絶縁膜がドライエッチングによって除去され、一対のくし型作用電極31a/31b、参照極32、対極33、およびストリッピング電極34を形成する。銀/塩化銀電極は、銀/塩化銀のペーストを参照電極32上に塗布して形成される。
ストリッピングゲル35は、以下のように形成され得る。
まず、密閉容器中においてフッ化ビニリデン-ヘキサフルオロプロピレン共重合体が超音波照射によって、氷冷しながらアセトンに溶解され、アセトン溶液を調製する。1-ブチル-3-メチルイミダゾリウムヨージドを含有する1-ブチル-3-メチルイミダゾリウムビストリフルオロメタンスルフォニルイミドがアセトン溶液に添加される。その後、アセトン溶液は攪拌され、ストリッピング電極34上に滴下される。最後に、アセトンは蒸発されて、ストリッピングゲル35を形成する。
保護ゲル36は、以下のように形成され得る。
ストリッピングゲル35の場合と同様に、密閉容器中においてフッ化ビニリデン-ヘキサフルオロプロピレン共重合体が超音波照射によって、氷冷しながらアセトンに溶解され、アセトン溶液を調製する。1-ブチル-3-メチルイミダゾリウムビストリフルオロメタンスルフォニルイミドがアセトン溶液に添加される。その後、アセトン溶液は攪拌され、ストリッピングゲル35上に滴下される。最後に、アセトンは蒸発されて、保護ゲル36を形成する。
上述されたセンサチップ300の表面に試料溶液が供給され、前記表面を前記試料溶液により被覆する。試料溶液は、本発明により定量される化学物質を含有する。当該化学物質の例は、抗原、抗体、核酸、細胞、細菌、ウィルス、ハプテン、糖類である。
より好ましくは、試料溶液は、カバー37とセンサチップ300との間の空間を満たす。これにより、試料溶液の容量を一定にすることが可能になる。
(工程(c))
工程(c)においては、スイッチ41aおよび41bは、いずれも各端子Aに接続され、第1作用電極31aに一定電位が印加される。さらに、第2作用電極31bはストリッピング電極34に電気的に接続され、一対のくし型作用電極31a・31bの間におけるレドックスサイクルを形成する。
酸化還元物質がフェロセンカルボン酸である場合、くし型作用電極31a・31bおよびストリッピング電極34上では、以下の化学式(I)~(III)に示される反応が生じる。
くし型作用電極31a:
Fc2+COOH(還元体) → Fc3+COOH(酸化体) + e-・・・(I)
くし型作用電極31b:
Fc3+COOH(酸化体) +e- → Fc2+COOH(還元体)・・(II)
ストリッピング電極34:
Ag+X- → AgX↓+e-・・・(III)
XはI、Br、またはClである。
ハロゲン化銀が、銀からなるストリッピング電極34の表面上に堆積される。
工程(d)では、スイッチ41aおよび41bは、いずれも端子Bに接続される。第1作用電極31aおよび第2作用電極31bのいずれにも、電位は印加されない。ポテンショスタット42により、ストリッピング電極34が掃引され、以下の化学式(IV)に示されるように、工程(c)時に析出したハロゲン化銀が試料溶液に溶解する。
ストリッピング電極34:
AgX+e- → Ag+X-・・・(IV)
工程(d)の溶解時において流れる電流量は、ハロゲン化銀の析出量に比例する。ハロゲン化銀の析出量は、酸化還元物質(還元体)の濃度および工程(c)において電位が印加された時間の積に比例する。すなわち、以下の等式が充足される。
(ハロゲン化銀の析出量)=(酸化還元物質(還元体)の濃度)・(工程(c)において電位が印加された時間)
以下、本発明の参考実施例が説明される。
図4(a)は、本参考実施例による第一作用電極50を示す。第一作用電極50は、銀プレート51、ストリッピングゲル52、および保護ゲル53を具備した。
第一作用電極50は、以下のように作製した。
電気化学測定は、以下のように行われた。
図5は、参考実施例による電気化学測定セル70を示す。電気化学測定セル70は、作用電極71、参照極72、対極73、電解液74およびポテンショスタット75を具備した。
作用電極71は第一作用電極50であった。参照極72は銀/塩化銀電極であった。対極73は白金ワイヤであった。電解液74は、Dulbecco’s Phosphate Buffered Saline (D-PBS:7mM NaCl、2.7mM KCl、10mM Na2HPO4、1.8mM KH2PO4)であった。
以下、本発明の比較例が説明される。
第二作用電極60は、保護ゲルを形成しなかったこと以外、第一作用電極50の場合と同様に作製された。
電気化学測定は、図5に示した電気化学セル70を用いて第一作用電極50の場合と同様に行った。第二作用電極60を、作用電極71に用いた。
プロット602は、測定サイクル数が増加すると、酸化電流ピーク値は徐々に減少した。この結果は、ストリッピングゲル62に含有されたヨウ化物イオンが電解液74に溶出したことを示す。

Claims (17)
- 試料溶液に含有される化学物質を定量する方法であって、以下の工程(a)~(e)を具備する:
センサチップを用意する工程(a)、ここで、
前記センサチップは、基板、一対の作用電極、対極、ストリッピング電極、ストリッピングゲル、および保護ゲルを具備し、
一対の作用電極は、第1作用電極および第2作用電極から構成され、
前記ストリッピング電極の表面は銀を具備し、
前記ストリッピングゲルは、前記ストリッピング電極を覆い、
前記ストリッピングゲルは、一対の作用電極および対極のいずれをも覆わず、
前記ストリッピングゲルは、基準電解質および、疎水性もしくは親水性イオン液体を含有し、
前記ストリッピングゲルは、水を含有せず、
前記疎水性もしくは親水性イオン液体は不揮発性であり、
前記疎水性もしくは親水性イオン液体は、陽イオンおよび陰イオンから構成され、
前記基準電解質は、前記陽イオンおよびハロゲン化物イオンから構成され、
前記保護ゲルは、前記ストリッピングゲルを覆い、
前記保護ゲルは、疎水性イオン液体を含有するが、前記基準電解質も水も含有せず、
センサチップの表面に、前記試料溶液を供給し、前記表面を前記試料溶液により被覆する工程(b)、ここで、
前記試料溶液は、前記化学物質および酸化還元物質を含有するか、または酸化還元物質により修飾された化学物質を含有し、
ポテンシオスタットにより前記第1作用電極に電位を印加し、かつ前記第2作用電極を前記ストリッピング電極に電気的に接続し、第1作用電極、前記第2作用電極、および前記ストリッピング電極の各表面上において以下の化学式(I)~(III)によって表される反応をそれぞれ生じさせる工程(c)、
第1作用電極:
酸化還元物質(還元体) → 酸化還元物質(酸化体) + e-・・・(I)
第2作用電極:
酸化還元物質(酸化体) +e- → 酸化還元物質(還元体)・・(II)
ストリッピング電極:
銀+ハロゲン化物イオン- →ハロゲン化銀↓+e-・・(III)
ここで、前記ハロゲン化銀は、前記ストリッピング電極の表面上に堆積し、
前記第1作用電極および前記第2作用電極に電位が印加されない状態でストリッピング電極に電位を印加し、ストリッピング電極を流れた電流量を測定する工程(d)、および
前記電流量に基づいて前記酸化還元物質(還元体)の濃度を算出し、前記算出された濃度に基づいて前記化学物質を定量する工程(e)。 - 請求項1に記載の方法であって、
前記センサチップは、注入口を有するカバーをさらに具備し、
前記カバーと前記センサチップとの間には空間が形成されており、
前記工程(b)において、前記試料溶液は前記注入口を介して前記センサチップの表面に供給される。 - 請求項2に記載の方法であって、
前記カバーはさらに空気孔を具備し、
前記工程(b)において、前記空間に満たされていた空気が、前記空気孔から排出される。 - 請求項2に記載の方法であって、
前記工程(b)の後には、前記空間に試料溶液が満たされる。 - 請求項1に記載の方法であって、
前記疎水性イオン液体を構成する陽イオンおよび前記陰イオンは、それぞれ以下の群Iおよび群IIから選択される。
群I:
(1)1-R1-2-R2-3-R3-イミダゾリウムイオン(R1はメチル基またはブチル基を表し、R2は水素またはメチル基を表し、R3はアルキル基を表し、R3はヘテロ原子を含んでもよい)、
(2)N-R4-イソキノリウムイオン(R4はアルキル基を表し、ヘテロ原子を含んでもよい)、
(3)N-R5-ピリジニウムイオン(R5はアルキル基を表し、ヘテロ原子を含んでもよい)、
(4)N,N-R6,R7-ピロリジニウムイオン(R6はメチル基、エチル基、プロピル基、イソプロピル基、またはブチル基を表し、R7はアルキル基を表し、R7はヘテロ原子を含んでもよい)、
(5)N,N-R8,R9-ピペリジニウムイオン、または
(R8はメチル基またはブチル基を表し、R9はアルキル基を表し、R9はヘテロ原子を含んでもよい)
(6)R10,R11,R12,R13-アンモニウムイオン
(R10,R11,R12,R13は独立してアルキル基、フェニル基、またはベンジル基であり、ヘテロ原子を含んでもよい)
群II:
(1)ビストリフルオロメタンスルフォニルイミドイオン、
(2)トリフレートイオン、または
(3)ビスペンタフルオロエタンスルフォニルイミドイオン - 請求項1に記載の方法であって、
前記疎水性イオン液体は、以下から選択される:
1,3-ジメチルイミダゾリウム ビストリフルオロメタンスルフォニルイミド
1-エチル-3-メチルイミダゾリウム ビストリフルオロメタンスルフォニルイミド
1-エチル-3-メチルイミダゾリウム トリフレート
1-エチル-3-メチルイミダゾリウム ビスペンタフルオロエタンスルフォニルイミド
1,3-ジエチルイミダゾリウム ビストリフルオロメタンスルフォニルイミド
1,3-ジエチルイミダゾリウム トリフレート
1-ブチル-3-エチルイミダゾリウム トリフレート
1,2-ジメチル-3-エチルイミダゾリウム ビストリフルオロメタンスルフォニルイミド
1-ブチル-3-メチルイミダゾリウム ビストリフルオロメタンスルフォニルイミド

前記親水性イオン液体を構成する陽イオンおよび前記陰イオンは、それぞれ以下の群Iおよび群IIIから選択される。
群I:
(1)1-R1-2-R2-3-R3-イミダゾリウムイオン(R1はメチル基またはブチル基を表し、R2は水素またはメチル基を表し、R3はアルキル基を表し、R3はヘテロ原子を含んでもよい)、
(2)N-R4-イソキノリウムイオン(R4はアルキル基を表し、ヘテロ原子を含んでもよい)、
(3)N-R5-ピリジニウムイオン(R5はアルキル基を表し、ヘテロ原子を含んでもよい)、
(4)N,N-R6,R7-ピロリジニウムイオン(R6はメチル基、エチル基、プロピル基、イソプロピル基、またはブチル基を表し、R7はアルキル基を表し、R7はヘテロ原子を含んでもよい)、
(5)N,N-R8,R9-ピペリジニウムイオン、または
(R8はメチル基またはブチル基を表し、R9はアルキル基を表し、R9はヘテロ原子を含んでもよい)
(6)R10,R11,R12,R13-アンモニウムイオン
(R10,R11,R12,R13は独立してアルキル基、フェニル基、またはベンジル基であり、ヘテロ原子を含んでもよい)
群III:
(4)テトラフルオロボレートイオン、または
(5)ハライドイオン。 - 請求項1に記載の方法であって、
前記親水性イオン液体は、以下から選択される:
1-エチル-3-メチルイミダゾリウム テトラフルオロボレート、
1-ブチル-3-メチルイミダゾリウム テトラフルオロボレート、
1-メチル-3-プロピルイミダゾリウム テトラフルオロボレート、
1-ブチル-3-メチルイミダゾリウム ヨージド、
1-ヘキシル-3-メチルイミダゾリウム ブロマイド、
1-ヘキシル-3-メチルイミダゾリウム クロライド、
1-オクチル-3-メチルイミダゾリウム クロライド、または
N-ヘキシルピリジニウム クロライド。 - 請求項1に記載の方法であって、
前記ストリッピングゲルが前記保護ゲルによって完全に隠されるように、前記保護ゲルは前記ストリッピングゲルの全体を覆っている。 - 変換ストリッピング法のためのセンサチップであって、以下を具備する:
基板、
一対のくし型作用電極、
対極、
ストリッピング電極、および
ストリッピングゲル、
ここで、
前記ストリッピング電極の表面は銀を具備し、
前記ストリッピングゲルは、前記ストリッピング電極を覆い、
前記ストリッピングゲルは、一対のくし型作用電極および対極のいずれをも覆わず、
前記ストリッピングゲルは、基準電解質および、疎水性もしくは親水性イオン液体を含有し、
前記疎水性もしくは親水性イオン液体は、陽イオンおよび陰イオンを含有し、
前記基準電解質は、前記陽イオンおよびハロゲン化物イオンを含有し、
前記保護ゲルは、前記ストリッピングゲルを覆い、
前記保護ゲルは、疎水性イオン液体を含有するが、前記基準電解質も水も含有しない。 - 請求項10に記載のセンサチップであって、
前記センサチップは、注入口を有するカバーをさらに具備し、
前記カバーと前記センサチップとの間には空間が形成されている。 - 請求項11に記載のセンサチップであって、
前記カバーはさらに空気孔を具備する。 - 請求項10に記載のセンサチップであって、
前記疎水性イオン液体を構成する陽イオンおよび前記陰イオンは、それぞれ以下の群Iおよび群IIから選択される。
群I:

1-ヘキシル-3-メチルイミダゾリウム クロライド、
1-オクチル-3-メチルイミダゾリウム クロライド、または
N-ヘキシルピリジニウム クロライド。 - 請求項10に記載のセンサチップであって、
前記ストリッピングゲルが前記保護ゲルによって完全に隠されるように、前記保護ゲルは前記ストリッピングゲルの全体を覆っている。
