[1] 被検細胞を含む末梢血含有試料と標識用物質とをin vitroにおいて接触させること、前記被検細胞に前記標識用物質を取り込ませて、前記被検細胞を標識させること、及び、前記被検細胞から、末梢血含有試料中の正常細胞の標識度合いよりも、前記標識用物質による標識度合いが強い細胞を、癌細胞として検出すること、を含む癌細胞の検出方法。
[2] 前記標識用物質が蛍光物質で標識された物質又は蛍光物質の前駆物質であり、前記検出が前記蛍光物質により標識された前記被検細胞の蛍光強度を得ることを含む[1]に記載の検出方法。

従って、本発明によれば、癌細胞の表面抗原の有無や特別な担体を用意する必要がなく、末梢血細胞中の被検細胞が癌細胞であるか正常細胞であるかの判断を、標識用物質による標識の度合いに基づいて短時間で且つ簡便に行うことができる。
また、本明細書において「~」を用いて示された数値範囲は、「~」の前後に記載される数値をそれぞれ最小値及び最大値として含む範囲を示す。
また、本明細書において、組成物中の各成分の量は、組成物中に各成分に該当する物質が複数存在する場合には、特に断らない限り、組成物中に存在する当該複数の物質の合計量を意味する。
本明細書において、偽陽性とは、癌細胞の検出の際に、癌細胞以外の細胞が検出されることを意味する。
本発明において、「末梢血含有試料」とは、末梢血試料を含有する試料を意味する。
以下、本発明について説明する。
これにより、末梢血試料を含有する末梢血含有試料中の癌細胞の有無を、標識用物質による標識度合いに基づいて短時間で且つ簡便に行うことができる。
前記標識用物質としては、検出時に認識可能な物質であればよく、癌細胞内に取り込まれる前から認識可能な物質(例えば、蛍光物質で標識された物質)及び癌細胞内に取り込まれた後に認識可能な物質(例えば、蛍光物質の前駆物質)のいずれであってもよい。
癌細胞内に取り込まれる前から認識可能な物質としては、例えば、公知の標識物質により標識された糖(標識化糖)、アミノ酸(標識化アミノ酸)、核酸(標識化核酸)、酢酸(標識化酢酸)、コリン(標識化コリン)、葉酸(標識化葉酸)等が挙げられる。
また、癌細胞内に取り込まれた後に認識可能になる物質としては、例えば、細胞内に取り込まれてプロトポルフィリン9に代謝される物質が挙げられる。
前記糖としては、単糖、二糖又は、糖単位が3~10個程度で構成されたオリゴ糖等を挙げることができる。また、細胞のエネルギー源となる栄養素であり、常時取り込みが行われており、短時間の培養でも充分量の取り込みが行われるため、前記糖は単糖であってもよい。単糖以外の糖は、細胞へ取り込み可能な形態に修飾されていてもよい。前記糖の例示としては、例えば、グルコース、デオキシグルコース、グルコサミン、ガラクトース、マンノース、フルクトース、アラビノース、スクロース、ラクトース、マルトース等を挙げることができる。また、例えば細胞への取り込み等の観点から、グルコースが挙げられる。
また、前記酵素に対する基質としては、ジアミノベンチジンなどが使用可能である。
標識物質としてビオチンを用いた場合には、さらにアビジンを結合させた酵素や蛍光物質を結合させることにより、検出を行うことができる。これらの標識物質による標識化は、常法により行うことができる。
このようなD-グルコース誘導体としては、例えば2-[N-(7-ニトロベンズ-2-オキサ-1,3-ジアゾール-4-イル)アミノ]-2-デオキシ-D-グルコース(2-NBDG、Yamada K. et al., J. Biol. Chem. 275:22278-22283, 2000)、6-[N-(7-ニトロベンズ-2-オキサ-1,3-ジアゾール-4-イル)アミノ]-6-デオキシ-D-グルコース(6-NBDG:Speizer L. et al., Biochim. Biophys.Acta 815:75-84, 1985))等が挙げられる。
前記標識化糖は、単独で用いてもよく、二種以上を併用してもよい。
前記天然アミノ酸としては、アラニン(Ala)、グリシン(Gly)、バリン(Val)、ロイシン(Leu)、イソロイシン(Ile)、フェニルアラニン(Phe)、チロシン(Tyr)、トリプトファン(Trp)、プロリン(Pro)、セリン(Ser)、トレオニン(Thr)、システイン(Cys)、メチオニン(Met)、アスパラギン(Asn)、グルタミン(Gln)等の中性アミノ酸、アスパラギン酸(Asp)、グルタミン酸(Glu)等の酸性アミノ酸、リシン(Lys)、アルギニン(Arg)、ヒスチジン(His)等の塩基性アミノ酸が挙げられる。
前記非天然アミノ酸としては、前記天然アミノ酸と同様のアミノ酸トランスポーターを介して細胞内に輸送されるアミノ酸誘導体であればよい。前記非天然アミノ酸としては、例えば、α-aminocyclobutane carboxylic acid(ACBC)、α-aminocyclopetane carboxylic acid(ACPC) α-aminoisobutyric acid(AIB)等が挙げられるが、これに限定されない。
前記非天然アミノ酸は、天然アミノ酸と同様に、その化学的性質により、中性アミノ酸、酸性アミノ酸又は塩基性アミノ酸に分類することができ、その分類に従い、本発明に係るアミノ酸として、天然アミノ酸と同様に使用することができる。
また、陽性率の向上等の観点より、前記アミノ酸としては、中性アミノ酸及び塩基性アミノ酸からなる群より選ばれるアミノ酸を用いることができる。
また、偽陽性の低下及び陽性率の向上等の観点の観点より、前記アミノ酸としては、中性アミノ酸からなる群より選ばれるアミノ酸を用いることができる。
前記アミノ酸は、前記標識化糖と同様の標識物質により標識されていればよい。好ましくは被検細胞内への取り込み効率等の観点からBenzoacrydony(Bacd)基、acridine(Acd)、7-Dimethylaminocoumarin-4-acetic acid(DMACA)、5-((2-Aminoethyl)amino)naphthalene-1-sulfonic acid(EDANS)、nathphalene (Nap)、anthracene (Ant)により標識することができるが、これに限定されない。
前記標識化アミノ酸は、単独で用いてもよく、二種以上を併用してもよい。
前記アミノレブリン酸及びその誘導体としては、アミノレブリン酸及びその誘導体、アミノレブリン酸及びその誘導体の塩、アミノレブリン酸及びその誘導体のエステル、並びにアミノレブリン酸のエステル及びその誘導体のエステルの塩が挙げられる。
具体的には、アミノレブリン酸、アミノレブリン酸メチル、アミノレブリン酸エチル、アミノレブリン酸プロピル、アミノレブリン酸ヘキシル、アミノレブリン酸ヘプチル、アミノレブリン酸オクチル等のアミノレブリン酸及びその誘導体が挙げられ、アミノレブリン酸メチルエステル、アミノレブリン酸エチルエステル、アミノレブリン酸プロピルエステル、アミノレブリン酸ヘキシルエステル、アミノレブリン酸ヘプチルエステル、アミノレブリン酸オクチルエステル等のアミノレブリン酸及びその誘導体のエステルが挙げられ、あるいはこれらの塩酸塩、スルホン酸塩、エステルリン酸塩、エステルスルホン酸塩、リン酸塩、硝酸塩、硫酸塩等のアミノレブリン酸及びその誘導体の塩、並びにアミノレブリン酸のエステル及びその誘導体のエステルの塩が挙げられる。
市販品としては、例えば、コスモ・バイオ株式会社のアミノレブリン酸(商品名:5-アミノレブリン酸塩酸塩)、東京化成工業株式会社のアミノレブリン酸メチルエステル(商品名:5-アミノレブリン酸メチルエステル塩酸塩)などが挙げられるが、これに限定されない。
前記標識用物質は、前記標識化糖及び標識化アミノ酸を併用することができる。前記標識化糖及び標識化アミノ酸を併用することにより、癌細胞の種類によって異なると推測される標識化糖又は標識化アミノ酸の取り込み速度の影響を受ける可能性が低減される。これにより、癌細胞の検出が簡便となる。また、様々な種類の癌細胞を検出することが可能となる。
また、例えば後述する検出工程の簡便化等の観点から、標識化糖の標識に用いる標識物質と、標識化アミノ酸の標識に用いる標識物質とは、同一の標識物質であることが挙げられる。
前記溶血剤としては、公知のものを用いることができ、例えば、塩化アンモニウム、塩化ナトリウム、シュウ酸アンモニウム、サポニン、ラウリル硫酸ナトリウム(SDS)のような界面活性剤、その他市販の溶血剤、例えばイムノプレップ(ベックマンコールター社)、イムノライズ(ベックマンコールター社)、BD FACS(登録商標) Lysing Solution(ベクトン・ディッキンソン社)などが挙げられる。また、例えば前記被検細胞による前記標識用物質の取り込み効率や標識化効率等の観点より、溶血剤としては、塩化アンモニウム、サポニン、溶血力の弱い市販の溶血剤を使用することができる。
前記溶血剤の量としては、例えば前記溶血剤の濃度(最終濃度)が0.01質量%~10質量%となるように前記末梢血試料に添加すること等が挙げられる。また、好ましくは例えば偽陽性率の低下等の観点より溶血剤の量としては、最終濃度として0.05質量%~5質量%の範囲とすることができる。また、より好ましくは例えば偽陽性率の低下及び癌細胞の検出の簡便化などの観点より溶血剤の量としては、最終濃度として0.1質量%~1質量%の範囲とすることができる。
例えば、前記標識用物質の濃度(最終濃度)が0.1μM~100mMとなるように、前記末梢血含有試料に添加すればよい。例えば、好ましくは前記標識用物質の濃度(最終濃度)が0.3μM~75mMとなるように、前記末梢血含有試料に添加すればよい。例えば、より好ましくは前記標識用物質の濃度(最終濃度)が1μM~50mMとなるように、前記末梢血含有試料に添加すればよい。
前記被検細胞を含む前記末梢血含有試料に、細胞種識別用抗体を接触させることにより、末梢血試料中に存在する様々な種類の細胞を、個別に標識し、認識することができる。これにより、例えば偽陽性率の低下や癌細胞の検出簡便化等が可能となる。
偽陽性率低下のために用いる前記細胞種別用抗体としては、血球分画に含まれる癌細胞以外の細胞を認識する抗体であれば、いずれの抗体を用いてもよい。例えば、抗CD45抗体、抗CD34抗体、抗CD3抗体、抗CD4抗体、抗CD14抗体、抗CD20抗体、抗CD36抗体、抗CD41抗体、抗CD309抗体等が挙げられる。例えば溶血剤により除去困難な白血球、血管内皮細胞、血管内皮前駆細胞、未分化白血球、造血系細胞の包括的な認識等の観点より、前記偽陽性率低下のために用いる前記細胞種別用抗体としては、抗CD45抗体及び抗CD34抗体からなる群より選ばれる少なくとも一種を用いることができる。
前記細胞種識別用抗体は、単独で用いてもよく、二種以上を併用してもよい。
前記標的細胞と、前記アミノレブリン酸及びその誘導体との前記接触工程を、鉄イオン及び還元剤の存在下において行うことにより、還元剤により、鉄イオンが2価の状態で安定化されるために、正常細胞では、一旦生成されたプロトポルフィリン9が速やかに代謝されてしまうので、偽陽性が抑制できると推測される。
また、好ましくは例えば偽陽性率の抑制等の観点より鉄イオンは、2価の鉄イオンを用いることができ、該2価の鉄イオンの濃度は、0.05mM~50mMであればよく、アスコルビン酸の濃度0.05mM~50mMであればよい。また、より好ましくは例えば偽陽性率の抑制等の観点より鉄イオンは、2価の鉄イオンを用いることができ、該2価の鉄イオンの濃度は、0.1mM~10mMであればよく、アスコルビン酸の濃度は0.1mM~10mMであればよい。
前記標的細胞と、前記アミノレブリン酸およびその誘導体との前記接触工程を、前記エンドサイトーシスを誘導する物質、及び前記細胞膜タンパク質の安定化作用を有するイオンからなる群より選ばれる少なくとも一種の存在下において行うことにより、アミノレブリン酸およびその誘導体の標的細胞への取り込みが促進されると推測される。なお、アミノレブリン酸およびその誘導体を使用する場合、検出工程のためにはこれらがプロトポルフィリン9に変換される必要があり、前記標的細胞と前記アミノレブリン酸およびその誘導体とを接触させ、かつ、インキュベートしてよい。そして、アミノレブリン酸への取り込みが促進されることにより、アミノレブリン酸およびその誘導体と標的細胞との接触(インキュベート)時間を短縮することができ、アミノレブリン酸およびその誘導体の反応性が向上する。
市販品としては、例えば、GeneTools社の両親媒性ペプチド(商品名Endo-Porter)、GeneTools社の両親媒性ペプチド(商品名Endo-Porter Aqueous)、invitrogen社のカチオン性脂質(商品名Lipofectin Reagent)、invitrogen社のカチオン性脂質(商品名Lipofectamine 2000 Reagent)、Roche社のカチオン性脂質(商品名DOTAP Liposomal Transfection Reagent)等が挙げられるが、これに限定されない。
前記エンドサイトーシスを誘導する物質の濃度としては、前記エンドサイトーシスを誘導する物質の最終濃度が0.1μM~20mMとなるように、前記接触前の細胞懸濁液に添加することができる。また、好ましくは例えばアミノレブリン酸の被検細胞への取り込み効率の上昇等の観点より、前記エンドサイトーシスを誘導する物質の濃度(最終濃度)としては、1μM~10mMを添加することができる。また、より好ましくは例えばアミノレブリン酸の被検細胞への取り込み効率の上昇等の観点より、前記エンドサイトーシスを誘導する物質の濃度(最終濃度)としては、5μM~2mMを添加することができる。
前記細胞膜タンパク質の安定化作用を有するイオンとしては、Mgイオン、Caイオンが挙げられる。
癌細胞は、被検細胞中の正常細胞と比較して3倍~8倍の代謝速度と考えられているため、前記標識用物質の取り込み量が正常細胞よりも多くなる。また、癌細胞は、正常の細胞とは異なる代謝経路を有するため、標識の度合いが、癌細胞と正常細胞とでは異なる。従って、検出工程では、このような前記標識用物質による標識の度合いが正常細胞より強い細胞を正常細胞と識別し、癌細胞として検出する。これにより、末梢血細胞中に、癌細胞が存在しているか否かがわかる。
ここで発色の程度又は蛍光強度に基づく癌細胞の検出は、目視観察によって行ってもよく、それぞれ吸光度又は蛍光強度を測定することによって得られた測定値によって行ってもよい。
また、前記細胞の検出は、癌細胞の検出精度の観点から、前記蛍光物質により標識された前記被検細胞の平均蛍光強度の検出及び前記蛍光物質の合計蛍光強度の検出の少なくとも一種であってもよい。
前記細胞の検出に加えて、簡便なノイズ又は血小板の除去の観点より、前記蛍光物質により標識された前記被検細胞のサイズに基づき、前記細胞の検出を実行することができる。具体的には、前記標識用物質により標識された光点の集合の直径が例えば、1μm~10μmより大きいもの、好ましくは2μm~9μmより大きいもの、より好ましくは3μm~8.5μmより大きいものをオブジェクトとして抽出することにより検出を実行することができる。
検出された癌細胞を単離する方法としては、前記標識用物質の種類に応じた単離方法を選択してもよく、前記標識用物質の種類に拘束されない単離方法を選択してもよい。前記標識用物質の種類に応じた単離方法としては、例えば、前記標識用物質が蛍光物質で標識化されている場合には、セルソーターなど、蛍光強度に基づいて細胞を分離する当業界で既知の方法を適用することができる。また、前記標識用物質の種類に拘束されない単離方法としては、細胞のサイズや形態に基づく単離方法を適用することができる。
次に、ステップ108で、上記ステップ106で算出した各オブジェクトの平均蛍光強度に基づいて、癌細胞を検出するための検出閾値αを設定する。検出閾値αとしては、例えば、各オブジェクトの平均蛍光強度の平均や最小値等を設定することができ、例えば100~3000、好ましくは150~2500、より好ましくは180~2200と設定することが出来る。
次に、ステップ112で、上記ステップ100で取得した明視野画像内の全細胞数をカウントする。
なお、上記ステップ108では、各オブジェクトの平均蛍光強度を算出する場合について説明したが、各オブジェクトに含まれる各画素の蛍光強度の和である合計蛍光強度を算出するようにしてもよい。
次に、ステップ208で、上記ステップ200で取得した明視野画像内の全細胞数をカウントする。
その他試薬類の例としては、抗体の特異性を高めるための物質例えば、各種レセプターのブロッキング剤;偽陽性を抑制するための物質、例えば鉄イオン;鉄イオンの安定性を高めるための物質、例えば還元剤;細胞膜タンパク質の安定性を高めるための物質、例えばMgイオンやCaイオン;および、細胞への物質の取込を促進させる物質、例えばエンドサイトーシス誘導物質、等が含まれる。
また、本発明における癌細胞検出キットは、前記標識用物質と、前記標識用物質による標識度合いに基づいて癌細胞を検出する方法を記載した取扱説明書とを含む。
本癌細胞検出キットによれば、末梢血試料中の癌細胞を短時間で且つ簡便に検出することができる。本キットにおける取扱説明書に記載された癌細胞を検出する方法としては、前記癌細胞検出方法において記述した事項がすべて適用される。
健常人の静脈血から、常法に従って、遠心分離法により血清試料と沈殿物とを分離し、血清試料から、白血球層と赤血球層の一部を回収して、PBSに再懸濁して、白血球試料及び赤血球試料を調製した。
DSファーマバイオメディカル社より購入した乳癌細胞株MCF-7を、αMEM培地(インビトロジェン社、10容量%FBS、L-グルタミン2mM、ペニシリン100U/mL及びストレプトマイシン100μg/mL添加)を用いて37℃、5容量%CO2濃度環境下で培養した。試験直前に、0.25%トリプシン含有EDTAを用いて細胞を回収し、PBSにて洗浄後、癌細胞株試料を調製した。
30分後に、1000μLのPBSを用いて洗浄し、2000rpm×5分間の遠心を行って常法に従って各細胞を回収した。
回収された細胞を50μLのPBSに再度、懸濁し、96ウェルプレートに全量を移し、観察用の試料を調製した。
各観察用の試料を、蛍光顕微鏡(ニコン社)にて観察し、2-NBDGによる取り込み量を目視にて確認した。また、同様に、抗EpCAM抗体-PEによる染色像を観察して、癌細胞を確認した。試料1の観察結果を、それぞれ図4~図6に示す。
一方、白血球細胞はやや蛍光顕微鏡視野で観察されるが癌細胞の蛍光像よりも遙かに弱く、蛍光強度として半分以下であった。赤血球細胞は、ほとんど活性がないため蛍光像はまったく観察されなかった。
<血球の調製>
真空採血管(EDTA・2K入り)を用いてヒトから採血を行う。HetaSep(STEMCELL社)の添付文書に従い、血球分離を行う。
<癌細胞の調製>
常法に従い、培養したヒト線維肉腫細胞株(HT1080:DSファーマバイオメディカル社)をトリプシン(インビトロジェン社)により回収した。なお、浮遊系であるヒト胃癌細胞株(SNU-1:ATCC)は、トリプシン処理を行うことなく、そのまま回収する。回収した癌細胞の細胞懸濁液を遠心し、上清を除く。ダルベッコPBS(-)(ニッスイ社)で再懸濁して、遠心し、再度上清を除く。
溶血処理を施す場合には、上記のようにして回収した血球分画に対して、塩化アンモニウム(STEMCELL社)の添付文書に従い、塩化アンモニウムを用いて溶血処理を行う。
<染色>
常法に従い、血球計算盤を用いて、各細胞の細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いてから、ダルベッコPBS(-)で溶解した2-NBDG(和光純薬社、表3又は表4に記載の最終濃度)を添加し、細胞を十分に懸濁させる。表3又は表4に記載の温度及び時間、反応(培養)する。反応後、ダルベッコPBS(-)を加えて、遠心し、上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く工程を2~3回行い、細胞を十分に洗浄する。ダルベッコPBS(-)を用いて再懸濁し、濃度調整をして384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。蛍光顕微鏡(ニコン社)で撮影を行う。蛍光フィルタセット:B-2A(ニコン社)を用いる。B-2Aの波長は次の通りである。 EX(励起波長) 475±15nm、DM 505nm以上、BA(蛍光波長)520nm以上。
画像解析ソフト(製品名NIS‐Elements ニコン社)を用いて、取得した画像を解析し、細胞の蛍光強度を評価する。
結果を表3及び表4に示す。表3は、上述の溶血処理を行った末梢血試料を使用しており、表4は、溶血処理を行っていない末梢血試料を使用した。

常法に従い、血球計算盤を用いて、各細胞の細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いてから、H-Ala(2-Bacd)-OH(渡辺化学工業社)を添加(表5又は表6に記載の最終濃度)し、細胞を十分に懸濁させる。表5又は表6に記載の温度及び時間、反応(培養)する。反応後、ダルベッコPBS(-)を加えて、遠心し、上清を除く。ダルベッコPBSを用いて再懸濁し、遠心し、再度上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く工程を2~3回行い、細胞を十分に洗浄する。ダルベッコPBS(-)を用いて再懸濁し、濃度調整をして384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。蛍光顕微鏡(ニコン社)で撮影を行う。蛍光フィルタセット:B-2A(ニコン社)を用いる。B-2Aの波長は実施例2と同様である。画像解析ソフト(製品名NIS‐Elements ニコン社)を用いて、取得した画像を解析し、細胞の蛍光強度を評価する。
結果を表5及び表6に示す。表5は、上述の溶血処理を行った末梢血試料を使用しており、表6は、溶血処理を行っていない末梢血試料を使用した。

常法に従い、血球計算盤を用いて、細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いてから、ダルベッコPBS(+)に溶解させたアミノレブリン酸(商品名5-アミノレブリン酸塩酸塩、コスモ・バイオ社、表7又は表8に記載の最終濃度)、アミノレブリン酸と同濃度のFeCl2とアミノレブリン酸と同濃度のアスコルビン酸を添加し、細胞を十分に懸濁させる。表7又は表8に記載の温度及び時間、反応(培養)する。細胞数を調整して384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。蛍光顕微鏡(ニコン社)で撮影を行う。
蛍光フィルタセットの波長:EX(励起波長)402±7.5、DM 430以上、BA(蛍光波長)625±15。
画像解析ソフト(製品名NIS‐Elements ニコン社)を用いて、取得した画像を解析し、細胞の蛍光強度を評価した。
結果を表7及び表8に示す。表7は、上述の溶血処理を行った末梢血試料を使用しており、表8は、溶血処理を行っていない末梢血試料を使用した。
実施例2の<染色>において、標識用物質を2-NBDGから6-NBDG(インビトロジェン社)に変更した以外は、実施例2と同様にして、細胞の蛍光強度を評価した。その結果を表9に示す。なお、2-NBDG及び6-NBDGの最終濃度は表9に記載した通りである。
実施例2と同様にして、ヒト乳腺上皮癌(細胞株名 MCF-7:DSファーマバイオメディカル社)、ヒト結腸腺癌(細胞株名 SW620:DSファーマバイオメディカル社)、ヒト前立腺癌(細胞株名 PC3:DSファーマバイオメディカル社)、ヒト肺胞上皮癌(細胞株名 NCI-H358:DSファーマバイオメディカル社)、ヒト胃癌(細胞株名 SNU-1:ATCC)及びヒト繊維肉腫(細胞株名 HT1080:DSファーマバイオメディカル社)のそれぞれを用いて、標識化糖(2-NBDG)による癌細胞の検出を行った。
その結果を表10に示す。なお、2-NBDGの最終濃度は表10に記載した通りである。
実施例3と同様にして、ヒト乳腺上皮癌(細胞株名 MCF-7:DSファーマバイオメディカル社)、ヒト結腸腺癌(細胞株名 SW620:DSファーマバイオメディカル社)、ヒト前立腺癌(細胞株名 PC3:DSファーマバイオメディカル社)、ヒト肺胞上皮癌(細胞株名 NCI-H358:DSファーマバイオメディカル社)、ヒト胃癌(細胞株名 SNU-1:ATCC)及びヒト繊維肉腫(細胞株名 HT1080:DSファーマバイオメディカル社)のそれぞれを用いて、標識化アミノ酸による癌細胞の検出を行った。
その結果を表11に示す。なお、標識化アミノ酸H-Ala(2-Bacd)-OHの最終濃度は表11に記載した通りである。
実施例4と同様にして、ヒト乳腺上皮癌(細胞株名 MCF-7:DSファーマバイオメディカル社)、ヒト結腸腺癌(細胞株名 SW620:DSファーマバイオメディカル社)、ヒト前立腺癌(細胞株名 PC3:DSファーマバイオメディカル社)、ヒト肺胞上皮癌(細胞株名 NCI-H358:DSファーマバイオメディカル社)、ヒト胃癌(細胞株名 SNU-1:ATCC)及びヒト繊維肉腫(細胞株名 HT1080:DSファーマバイオメディカル社)のそれぞれを用いて、アミノレブリン酸による癌細胞の検出を行った。
その結果を表12に示す。なお、アミノレブリン酸の最終濃度は表12に記載した通りである。

<溶血処理>
上記のようにして回収した血球分画に対して、塩化アンモニウム(STEMCELL社)の添付文書に従い、室温にて塩化アンモニウムを用いて溶血処理を行う。
<染色>
常法に従い、血球計算盤を用いて、細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いた後で、H-Ala(2-Bacd)-OH(渡辺化学工業社)及び2-NBGD(和光純薬社)を添加(表13に記載の最終濃度)し、細胞を十分に懸濁させる。37℃、30分反応させる。反応後、ダルベッコPBS(-)を加えて、遠心し、上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く工程を2~3回行い、細胞を十分に洗浄する。ダルベッコPBS(-)を用いて再懸濁し、濃度調整をして384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。蛍光顕微鏡(ニコン社)で撮影(露光時間1秒)を行う。蛍光フィルタセット:B-2A(ニコン社)を用いる。
画像解析ソフト(製品名NIS‐Elements ニコン社)を用いて、取得した画像を解析し、細胞の蛍光強度を評価する。その結果を表13に示す。
<細胞の調製>
常法に従い、培養しているヒト繊維肉腫細胞株(HT-1080:DSファーマバイオメディカル社)およびヒト正常皮膚繊維芽細胞株(NHDF:ロンザ社)を、トリプシン(インビトロジェン社)を用いて回収する。回収した癌細胞の細胞懸濁液を遠心し、上清を除く。その後、ダルベッコPBS(+)(ニッスイ社)で再懸濁して、遠心し、再度上清を除く。
<染色>
常法に従い、血球計算盤を用いて、細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いた後に、鉄イオンと還元剤を添加する条件では、ダルベッコPBS(+)に溶解させた0.3mMのアミノレブリン酸(商品名5-アミノレブリン酸塩酸塩、コスモ・バイオ社)、0.3mMのFgCl2、0.3mMのアスコルビン酸を添加し、細胞を十分に懸濁させる。鉄イオンと還元剤を添加しない条件では、ダルベッコPBS(+)に溶解させた0.3mMのアミノレブリン酸のみを添加し、細胞を十分に懸濁させる。37℃で60分間反応させる。細胞数を調整して384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。
<蛍光撮影>
蛍光顕微鏡(ニコン社)で明視野(対物×10倍)および蛍光(対物×10倍)を同一視野での観察を行い、偽陽性の程度について確認を行った。その結果を表14に示す。
蛍光フィルタセット:EX(励起波長) 402±7.5、DM 430以上、BA(蛍光波長) 625±15。
<細胞の調製>
常法に従い、培養しているヒト乳腺上皮癌細胞株(細胞株名 MCF-7:DSファーマバイオメディカル社)を、トリプシン(インビトロジェン社)を用いて回収する。回収した癌細胞懸濁液を遠心し、上清を除く。ダルベッコPBS(+)(ニッスイ社)で再懸濁して、遠心し、再度上清を除く。
<染色>
常法に従い、血球計算盤を用いて、細胞数をカウントする。1サンプルに2.8×105個の細胞を分取する。Mgイオン及びCaイオンが配合されたダルベッコPBS(+)に、アミノレブリン酸(商品名5-アミノレブリン酸塩酸塩、コスモ・バイオ社)を1mMになるように添加し、細胞を十分に懸濁させる。37℃で反応させる。
<蛍光検出>
反応開始後、15分、30分、60分、90分での細胞懸濁液の蛍光値をi-densy(アークレイ社)を用いて測定した。蛍光値の測定条件:音調&光量モードにて40℃、ゲイン「3」、LED1(390±20nm:励起)- PD3(642.5±57.5nm:検出)。
結果を図7に示す。図7中、縦軸は蛍光値を示し、横軸は反応時間を示す。黒塗り四角は、Mgイオン及びCaイオンが配合されていないダルベッコPBS(-)を用いた場合の蛍光値である。黒塗り三角は、Mgイオン及びCaイオンが配合されたダルベッコPBS(+)を用いた場合の蛍光値である。なお、黒塗り四角で表された結果は、実施例11において、ダルベッコPBS(+)を、ダルベッコPBS(-)に変更した以外は、実施例11と同様に行った結果である。
この結果より、アミノレブリン酸に加えて、Mgイオン及びCaイオンを添加することにより、アミノレブリン酸の反応性が向上することが明らかとなった。
実施例11の染色の際に、ダルベッコPBS(+)に溶解させたアミノレブリン酸を1mMになるように添加し、10μMのEndo-poter(GeneTools社)を添加した以外は、実施例11と同様にして、細胞懸濁液の蛍光値を測定した。
結果を図8に示す。図8中、縦軸は蛍光値を示し、横軸は反応時間を示す。黒塗り四角は、Mgイオン及びCaイオンが配合されていないダルベッコPBS(-)を用いた場合の蛍光値である。黒塗り丸は、Mgイオン及びCaイオンが配合されたダルベッコPBS(+)にEndo-poterを添加した場合の蛍光値である。
この結果より、アミノレブリン酸、Mgイオン及びCaイオンに加えて、Endo-porterを添加することにより、アミノレブリン酸の反応性が、さらに向上することが明らかとなった。
なお、図9は、実施例11と実施例12との結果を一図にまとめたものである。これにより、アミノレブリン酸のみのもの(黒塗り四角)に比べて、アミノレブリン酸にMgイオン及びCaイオンを加える(黒塗り三角)ことにより、アミノレブリン酸の反応性が向上し、アミノレブリン酸に、Mgイオン、Caイオン及びEndo-porterを加える(黒塗り丸)ことにより、アミノレブリン酸の反応性がさらに向上することが明らかとなった。
実施例3の<染色>において、標識用物質をH-Ala(2-Bacd)-OHから、H-Lys(DMACA)-OH(渡辺化学工業社)又はH-Glu(EDANS)-OH(渡辺化学工業社)に変更した。
また、蛍光物質の蛍光特性に合わせて、蛍光フィルタセットの波長をEX(励起波長)365/±10、DM 400以上、BA(蛍光波長)470±20に変更した以外は、実施例2と同様にして、細胞の蛍光強度を評価した。結果を表15に示す。
<血球の調製>
真空採血管(EDTA・2K入り)を用いてヒトから採血を行う。HetaSep(STEMCELL社)の添付文書に従い、血球分離を行う。
<癌細胞の調製>
常法に従い、培養したヒト前立腺癌細胞株(PC3:DSファーマバイオメディカル社)を、トリプシン(インビトロジェン社)を用いて回収する。回収した癌細胞の細胞懸濁液を遠心し、上清を除く。ダルベッコPBS(+)(ニッスイ社)で再懸濁して、遠心し、再度上清を除く。
<血球と癌細胞の混合液の調整>
血球分離を行った血球細胞懸濁液の細胞数を常法に従い、血球計算盤を用いてカウントする。また、ヒト前立腺癌細胞株(PC3)の細胞数も同様にカウントし、血球細胞数に対して1%の細胞数になるように血球細胞懸濁液に添加する。ヒト前立腺癌細胞株(PC3)を添加した血球細胞懸濁液に対して、塩化アンモニウム(STEMCELL社)の添付文書に従い、室温にて塩化アンモニウムを用いて溶血処理を行う。
<染色>
常法に従い、血球計算盤を用いて、細胞数をカウントし、サンプルを105個の~106個の試料(細胞懸濁液)に分取する。遠心し、上清を除いてから、ダルベッコPBS(+)に溶解させた0.3mMのアミノレブリン酸(商品名5-アミノレブリン酸塩酸塩、コスモ・バイオ社)、0.3mMのFeCl2と0.3mMのアスコルビン酸を添加し、細胞を十分に懸濁させる。37度、60分間反応(培養)する。反応後、遠心し、上清を除き、FcR Blocking Reagent(Miltenyi Biotec社)を添付文書に従い添加し、4度、30分間反応する。反応後、FITC標識抗CD45抗体(Miltenyi Biotec社社)及び、FITC標識抗CD34抗体(BioLegend社)、PE標識抗EpCAM抗体(Miltenyi Biotec社社)をそれぞれ添付文書に従い添加し、23度、30分間反応する。反応後、ダルベッコPBS(-)を加えて、遠心し、上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く。ダルベッコPBS(-)を用いて再懸濁し、遠心し、再度上清を除く工程を2~3回行い、細胞を十分に洗浄する。細胞数を調整して384ウェルプレートに滴下する。プレートを遠心して、細胞をウェル底面に落とす。蛍光顕微鏡(ニコン社)で撮影を行う。プロトポルフィリン9は蛍光フィルタセット1で、PE標識抗EpCAM抗体は蛍光フィルタセット2で、FITC標識抗CD45抗体及び、FITC標識抗CD34抗体は蛍光フィルタセット3で検出する。
蛍光フィルタセット1の波長:EX(励起波長)402±7.5nm、DM 430nm以上、BA(蛍光波長)625±15nm。
蛍光フィルタセット2の波長:EX(励起波長)535±25nm、DM 575nm以上、BA(蛍光波長)590nm以上。
蛍光フィルタセット3の波長:EX(励起波長)475±15nm、DM 505nm以上、BA(蛍光波長)525±10nm。
画像解析ソフト(製品名NIS‐Elements ニコン社)を用いて、取得した画像を解析し、細胞の蛍光強度を評価した。その結果を表16に示す。

Claims (21)
- 被検細胞を含む末梢血含有試料と標識用物質とをin vitroにおいて接触させること、
前記被検細胞に前記標識用物質を取り込ませて、前記被検細胞を標識させること、及び、
前記被検細胞から、末梢血含有試料中の正常細胞の標識度合いよりも、前記標識用物質による標識度合いが強い細胞を、癌細胞として検出すること、
を含む癌細胞の検出方法。 - 前記標識用物質が蛍光物質で標識された物質又は蛍光物質の前駆物質であり、前記検出が前記蛍光物質により標識された前記被検細胞の蛍光強度を得ることを含む請求項1に記載の検出方法。
- 前記標識用物質が蛍光物質で標識された物質又は蛍光物質の前駆物質であり、前記検出が前記蛍光物質により標識された前記被検細胞の平均蛍光強度及び合計蛍光強度からなる群より選ばれる少なくとも一種を得ることを含む請求項1又は請求項2に記載の検出方法。
- 前記検出が、さらに前記蛍光物質により標識された前記被検細胞のサイズに基づいて行われる請求項1から請求項3のいずれか一項に記載の検出方法。
- 前記標識用物質が、標識化糖、標識化アミノ酸及び細胞内に取り込まれてプロトポルフィリン9に代謝される物質からなる群より選ばれる少なくとも一種である請求項1から請求項4のいずれか一項に記載の検出方法。
- 前記標識用物質が、標識化糖及び標識化アミノ酸である請求項1から請求項5のいずれか一項に記載の検出方法。
- 前記標識用物質が前記標識化糖であり、前記標識化糖が、標識化単糖である請求項1から請求項6のいずれか一項に記載の検出方法。
- 前記標識用物質が前記標識化糖であり、前記標識化糖が、蛍光標識化グルコース誘導体である請求項1から請求項7のいずれか一項に記載の検出方法。
- 前記標識用物質が標識化アミノ酸であり、前記アミノ酸が、天然アミノ酸及び非天然アミノ酸からなる群より選ばれる少なくとも一種である請求項1から請求項8のいずれか一項に記載の検出方法。
- 前記標識用物質が標識化アミノ酸であり、前記アミノ酸が、中性アミノ酸及び酸性アミノ酸からなる群より選ばれる少なくとも一種である請求項1から請求項9のいずれか一項に記載の検出方法。
- 前記標識用物質が細胞内に取り込まれてプロトポルフィリン9に代謝される物質であり、前記物質が、アミノレブリン酸及びその誘導体からなる群より選ばれる少なくとも一種である請求項1から請求項10のいずれか一項に記載の検出方法。
- 前記標識用物質が細胞内に取り込まれてプロトポルフィリン9に代謝される物質であり、前記物質がアミノレブリン酸及びその誘導体であり、前記アミノレブリン酸及びその誘導体が、それらの塩、それらのエステル、及びそれらのエステルの塩からなる群より選ばれる少なくとも一種である請求項1から請求項11のいずれか一項に記載の検出方法。
- 前記被検細胞を含む前記末梢血含有試料が溶血処理されている請求項1から請求項12のいずれか一項に記載の検出方法。
- 前記被検細胞を含む前記末梢血含有試料に、細胞種識別用抗体をin vitroにおいて接触させることを含む請求項1から請求項13のいずれか一項に記載の検出方法。
- 前記細胞種識別用抗体が、抗CD45抗体及び抗CD34抗体からなる群より選ばれる少なくとも一種である請求項14に記載の検出方法。
- 鉄イオン及び還元剤の存在下において、前記被検細胞を含む前記末梢血含有試料と前記標識用物質とをin vitroにおいて接触させる請求項1から請求項15のいずれか一項に記載の検出方法。
- エンドサイトーシスを誘導する物質及び細胞膜タンパク質の安定化作用を有するイオンからなる群より選ばれる少なくとも一種の存在下において、前記被検細胞を含む前記末梢血含有試料と前記標識用物質とをin vitroにおいて接触させる請求項1から請求項16のいずれか一項に記載の検出方法。
- 0.1μM~100mMの濃度の前記標識用物質を用いて、前記被検細胞を含む前記末梢血含有試料と前記標識用物質とをin vitroにおいて接触させる請求項1から請求項17のいずれか一項に記載の検出方法。
- 1℃~42℃の条件下において、前記被検細胞に前記標識用物質を取り込ませ、標識させる請求項1から請求項18のいずれか一項に記載の検出方法。
- 1分~5時間、前記被検細胞に前記標識用物質を取り込ませ、標識させる請求項1から請求項19のいずれか一項に記載の検出方法。
- 標識用物質と、細胞腫識別用抗体及び洗浄液からなる群より選ばれる少なくとも一方とを含む、癌細胞検出用キット。
