Abstract
Description
から選択される1つもしくは複数のアミノ酸が置換されていることが好ましく、次の群;2番目がイソロイシン、46番目がメチオニン、68番目がセリン、および95番目がグリシン、
から選択される1つもしくは複数のアミノ酸置換が導入されていることがより好ましい。
(a)配列表の配列番号1に示すアミノ酸配列と配列同一性が85%以上であり、(b)3-キヌクリジノンを還元して(R)-3-キヌクリジノールを生成し、もしくは、3-クロロ-1-(2-チエニル)プロパン-1-オンを還元して(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールを生成し、(c)配列表の配列番号1に示すアミノ酸配列からなるカルボニル還元酵素と比較して、ハロゲン原子存在下でのカルボニル化合物に対する反応性が高い。
なお、本明細書において、アミノ酸、ペプチド、タンパク質は下記に示すIUPAC-IUB生化学命名委員会(CBN)で採用された略号を用いて表される。また、特に明示しない限り、ペプチドおよびタンパク質のアミノ酸残基の配列は、左端から右端にかけてN末端からC末端となるように表される。また、参照を容易にするため、一般的に用いられている下記の命名法を適用する。1つは、“もとのアミノ酸;位置;置換したアミノ酸”と記述する方法であり、例えば、位置64におけるチロシンのアスパラギン酸への置換は「Y84D」と表される。多重変異については、ハイフン記号“-”により分けることで表記する。例えば、「S41A-Y64D」とは、位置41のセリンをアラニンへ、かつ、位置64のチロシンをアスパラギン酸へ置換することを示す。

ポリペプチドやポリヌクレオチドの「配列同一性」とは、対比される2つのポリペプチドまたはポリヌクレオチドを最適に整列させ、アミノ酸又は核酸塩基(例えば、A、T、C、G、U、またはI)が両方の配列で一致した位置の数を比較塩基総数で除し、そして、この結果に100を乗じた数値で表される。
2番目がイソロイシン、46番目がメチオニン、68番目がセリン、および95番目がグリシン、から選択される1つもしくは複数のアミノ酸置換が導入されていることが好ましい。
0.1M 3-キヌクリジノン塩酸塩及び0.1M 還元型ニコチンアミド・アデニンジヌクレオチドリン酸(以下、NADPH)を含む0.5M リン酸緩衝液(pH6.5)に、本発明のポリペプチドを加えて30℃で反応させる。反応後、ジクロロメタンなどの有機溶媒で抽出操作を行い、下記のガスクロマトグラフィー条件で分析することにより、生成した3-キヌクリジノールの、その立体配置及び光学純度を確認することができる。

[塩化物に対する酵素の安定性の評価方法]
酵素を含む無細胞抽出液に任意の濃度(例えば0.01%~50%)の塩化物を含む緩衝液(好ましくはpH5~8の0.01~1Mリン酸緩衝液)を加えて任意の温度(例えば4~40℃)でインキュベートする。塩化物と緩衝液が不均一な場合は振とう、または攪拌しながらインキュベートする。塩化物を加えた直後(0時間目)と任意の処理時間(0.1~48時間)でサンプリングし、それを0.1Mのリン酸カリウム水溶液(pH7.0)で希釈し、その希釈液を用いて下記の[カルボニル化合物に対する還元能力の評価方法]で酵素の活性を測定する。残存活性は下記式で算出することができる。
残存活性(%)=[任意の処理時間での酵素活性]÷[0時間目の酵素活性]×100
ジメチルスルホキシド0.3%(v/v)を含む100mMリン酸カリウム緩衝液(pH6.5)にNADPHもしくは還元型ニコチンアミド・アデニンジヌクレオチド(以下、NADH)0.25mM、還元活性を評価したいカルボニル化合物(例えば3-キヌクリジノンもしくは3-クロロ-1-(2-チエニル)プロパン-1-オン)1~50mMおよび本発明のポリペプチドを含む反応液を30℃で反応させ、NADPHもしくはNADH量の減少に伴う波長340nmの吸光度の減少を測定することにより、還元反応の進行を容易に評価することができる。吸光度が減少した場合、本発明のペプチドは評価対象のカルボニル化合物を還元する能力を有する、と判断することができる。なお、吸光度の減少速度が速いほど、評価対象のカルボニル化合物に対する還元能力が高いといえる。また、ポリペプチドの還元能力は数値化することも可能であり、還元活性1Uは1分間に1μmolのNADPHの消費を触媒する酵素量とした。
[塩化物による反応阻害に対する抵抗性の評価方法]
まず、前記[カルボニル化合物に対する還元能力の評価方法]で塩化物が存在しない条件での酵素の活性を測定する。これを[塩化物非存在下での酵素活性]とする。次に任意の濃度(例えば0.01%~50%)の塩化物と[カルボニル化合物に対する還元能力の評価方法]と同じ濃度のカルボニル化合物とジメチルスルホキシド0.3%(v/v)を含む100mMリン酸カリウム緩衝液(pH6.5)で同様に活性を測定する。これを[塩化物存在下での酵素活性]とする。
相対活性は下記式で算出することができる。
相対活性(%)=[塩化物存在下での酵素活性]÷[塩化物非存在下での酵素活性]×100
[補酵素に対する親和性の評価方法]
任意の濃度(例えば、1~0.01mMの範囲で4つ以上の異なる濃度)の還元型補酵素(例えば、NADPH)存在下で前記[カルボニル化合物に対する還元能力の評価方法]と同様の方法で、任意のカルボニル化合物(例えば3-キヌクリジノンもしくは3-クロロ-1-(2-チエニル)プロパン-1-オン)に対する還元酵素活性を測定する。測定した還元酵素活性の値を用いてラインウィーバー・バーク・プロットを作成し、これよりKm値を算出することができる。
エラープローンPCR法(Leung et al., Technique 1, 11-15(1989))、あるいは同様の原理に基づいたキットを用いて、配列表の配列番号2に示す塩基配列(野生型酵素遺伝子)に1つ以上の塩基配列の置換、挿入、欠失、付加が導入されたDNA断片を得ることができる。例えば、野生型酵素遺伝子をテンプレートにプライマー1:5’-GGATAACAATTTCATAAGGAGGTTACATATG-3’(配列表の配列番号3)、およびプライマー2:5’-CTAGAGGATCCCCGGGTACCTTA-3’(配列表の配列番号4)とDiversify PCR Random Mutagenesis Kit(Clontech社製)を用いて、野生型酵素遺伝子全長にランダムに変異が導入され、かつ開始コドンにNdeI認識部位が付加され、終始コドンの直後に新たな終止コドン(TAA)とKpnI認識部位が付加された複数種類の二本鎖DNA(変異型酵素遺伝子)を得ることができる。この増幅断片をNdeI及びKpnIで消化し、プラスミドpUCN18(PCR法によりpUC18(タカラバイオ社製)の185番目のTをAに改変してNdeIサイトを破壊し、更に471-472番目のGCをTGに改変することにより新たにNdeIサイトを導入したプラスミド)のlacプロモーターの下流のNdeI認識部位とKpnI認識部位の間に挿入し、このプラスミドを用いてエシェリヒア・コリ(Escherichia coli)HB101株(以下、E.coli HB101)を形質転換する。形質転換した大腸菌を100μg/mLのアンピシリンを含むLBプレート培地に塗布し、シングルコロニーの大腸菌を得る。また、野生型遺伝子の代わりに前記方法で得られた変異型酵素遺伝子を用いて、同様の操作でさらに変異を導入した変異酵素ライブラリーを作製することもできる。

変異酵素ライブラリーの各組換え菌及び野生型酵素を生産する組換え菌(例えば、参考例3に示すE.coli HB101(pNBS))を適当な培地(例えば200μg/mlのアンピシリンを含む2×YT培地(トリプトン1.6%、イーストエキス1.0%、塩化ナトリウム 0.5%、pH7.0))に接種し、37℃で24時間振盪培養する。得られた各培養液を菌体破砕処理し、遠心分離後、沈殿を除去し、無細胞抽出液を得る。各酵素を含む無細胞抽出液に適当な濃度の塩化物(好ましくは終濃度0.64Mの塩化ナトリウム、もしくは終濃度0.01~0.02%のクロロアセトン)を含む緩衝液(好ましくはpH5~8の0.01~1Mリン酸緩衝液)を加えて適当な温度(例えば4~40℃)でインキュベートする。0.1~48時間程度インキュベートした後、処理した各無細胞抽出液を96穴プレート(AGCテクノグラス社製)に分注し、NADPH(好ましくは1.5mM)とカルボニル化合物(好ましくは20mMの3-キヌクリジノン塩酸塩)を含むリン酸緩衝液(pH5~7)を加えて、10~40℃で反応する。経時的にUVサンプル撮影装置FAS-III(東洋紡績社製)でNADPHの蛍光を観察する。この時、反応が進行しない酵素液はNADPHの蛍光が残存するが、反応が進行した無細胞抽出液はNADPHの減少に伴い、蛍光がなくなる。対照である野生型酵素に比べ、短時間で蛍光がなくなった酵素を塩化物に対する安定性が向上した酵素として選抜した。選抜した酵素の培養液からプラスミドを抽出し、BigDye Terminator Cycle Sequencing Kit(アプライドバイオシステムズジャパン社製)およびApplied Biosystems 3130xlジェネティックアナライザ(アプライドバイオシステムズジャパン社製)を用いて改変型カルボニル還元酵素遺伝子の塩基配列を決定し、変異部位を特定することができる。

或いは、形質転換体として、本発明のポリペプチドをコードするポリヌクレオチドを染色体中に導入して得られる形質転換体も挙げられる。

バークホルデリア・エスピー(Burkholderia sp.)YT株(NITE P-613)より、3-キヌクリジノン及び3-クロロ-1-(2-チエニル)プロパン-1-オンに対する還元活性を有するポリペプチド(以下、本ポリペプチドをRBSと呼ぶ)をコードするDNAをPCRにより取得した。
500ml容坂口フラスコに、バクトトリプトン16g、酵母エキス10g、塩化ナトリウム5g、アデカノールLG-109(日油(株)製)0.1g(いずれも1L当たり)の組成からなる液体培地(pH7)50mlを調製し、120℃で20分間蒸気殺菌をおこなった。この培地に、予め同培地にて前培養しておいたバークホルデリア・エスピー(Burkholderia sp.)YT株の培養液を5ml接種し、30℃で18時間振盪しながら培養を行った。この培養液から、Murray等の方法(Nucl.Acids Res.8,4321(1980))に記載の方法に従って染色体DNAを抽出した。
プライマー3:5’-ATATATACATATGACCCACCCGCAACCCCG-3’(配列表の配列番号5)、プライマー4:5’-TATAGGTACCTTATCAAAGCTTGTCGAACGCGAG-3’(配列表の配列番号6)を用いて、バークホルデリア・エスピー(Burkholderia sp.)YT株の染色体DNAを鋳型としてPCRを行った。その結果、配列表の配列番号2に示す塩基配列からなる遺伝子の開始コドン部分にNdeI認識部位が付加され、かつ終始コドンの直後に新たな終止コドン(TAA)とKpnI認識部位が付加された二本鎖DNA(RBS遺伝子)が得られた。PCRは、DNAポリメラ-ゼとして、PrimeSTAR HS DNA polymerase(タカラバイオ社製)を用いて行い、反応条件はその取り扱い説明書に従った。
参考例1で得られたRBS遺伝子をNdeI及びKpnIで消化し、プラスミドpUCN18(PCR法によりpUC18(タカラバイオ社製)の185番目のTをAに改変してNdeIサイトを破壊し、更に471-472番目のGCをTGに改変することにより新たにNdeIサイトを導入したプラスミド)のlacプロモーターの下流のNdeI認識部位とKpnI認識部位の間に挿入し、組換えベクターpNBSを構築した。
参考例2で構築した組換えベクターpNBSを用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、組換え生物E.coli HB101(pNBS)を得た。また、pUCN18を用いてE.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、組換え生物E.coli HB101(pUCN18)を得た。
参考例3で得た2種類の組換え生物(E.coli HB101(pUCN18)、E.coli HB101(pNBS))を、200μg/mlのアンピシリンを含む2×YT培地(トリプトン1.6%、イーストエキス1.0%、塩化ナトリウム 0.5%、pH7.0)5mlに接種し、37℃で24時間振盪培養した。上記の培養で得られたそれぞれ培養液について、遠心分離により菌体を集め、5mlの100mMリン酸緩衝液(pH6.5)に懸濁した。これを、UH-50型超音波ホモゲナイザー(SMT社製)を用いて破砕した後、遠心分離により菌体残渣を除去し、無細胞抽出液を得た。これら無細胞抽出液の3-キヌクリジノン還元活性を測定した。
参考例4と同様の方法で野生型酵素の無細胞抽出液を得た。この無細胞抽出液の3-キヌクリジノン還元活性を測定し、これを0時間目の活性とした。この無細胞抽出液に表1に示した化合物を添加し、硫酸もしくは水酸化ナトリウムでpH6.5に調整後、30℃で24時間インキュベートした。また、対照として何も添加しない無細胞抽出液も同様にインキュベートした。24時間後、各無細胞抽出液を希釈し、参考例4と同様の方法で3-キヌクリジノン還元活性を測定した。残存活性は下記式より算出し、この値を各種化合物に対する安定性の指標とした。
残存活性(%)=[24時間目の酵素活性]÷[0時間目の酵素活性]×100
結果を表1に示す。
参考例4と同様の方法で野生型酵素の無細胞抽出液を得た。この無細胞抽出液の3-キヌクリジノン還元活性を測定し、これを阻害剤なしの活性とした。表2に示した化合物及び5mM 3-キヌクリジノン、0.25mM補酵素NADPHを含む100mMリン酸緩衝液(pH6.5)に、無細胞抽出液を添加して30℃で1分間反応を行い、波長340nmにおける吸光度の減少速度より3-キヌクリジノンに対する還元活性を算出した。

結果を表2に示す。
参考例2で作製したRBS遺伝子を含むプラスミドpNBSをテンプレートに、プライマー1:5’-GGATAACAATTTCATAAGGAGGTTACATATG-3’(配列表の配列番号3)およびプライマー2:5’-CTAGAGGATCCCCGGGTACCTTA-3’(配列表の配列番号4)を用いてエラープローンPCR法(Leung et al.Technique 1,11-15(1989))を用いて、RBS遺伝子全長にランダムな変異を導入したDNA増幅断片を得た。この増幅断片を制限酵素NdeIおよびKpnIで消化したのち、同酵素で処理した高発現ベクターpUCN18に組み込み、複数の変異酵素発現プラスミドを作製した。このプラスミドを用いてE.coli HB101を形質転換し、100μg/mLのアンピシリンを含むLBプレート培地に塗布した。生育したコロニーは変異導入されたRBS遺伝子を有する組換え大腸菌であり、この組換え菌群を変異酵素ライブラリー1とした。
変異酵素ライブラリーより無機塩化物の1つである塩化ナトリウムに対する安定性が向上した改変型カルボニル還元酵素を選抜した。実施例1で作製した変異酵素ライブラリー1の各組換え菌及び参考例3で作製したE.coli HB101(pNBS)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液60μlに10mM EDTA・2Na及び1% Triton X-100を含むリン酸緩衝液(pH7.0)240μlを加え、37℃で1時間インキュベートした。各処理液を遠心し、上清を無細胞抽出液とした。各無細胞抽出液100μlに1.24M 塩化ナトリウムを含むリン酸緩衝液(pH6.5)を加え、30℃で18時間インキュベートした(塩化ナトリウム処理)。塩化ナトリウム処理した各無細胞抽出液を96穴プレート(AGCテクノグラス社製)に50μl分注し、6mMのNADPHを含むリン酸緩衝液(pH6.5)50μlと20mM 3-キヌクリジノン塩酸塩を含むリン酸緩衝液(pH6.5)100μlを加え30℃で反応した。経時的にUVサンプル撮影装置FAS-III(東洋紡績社製)でNADPHの蛍光を観察した。反応が進行しない酵素液はNADPHの蛍光が残存するが、反応が進行した無細胞抽出液はNADPHの減少に伴い、蛍光がなくなった。対照であるE.coli HB101(pNBS)の無細胞抽出液(野生型酵素)に比べ、早く蛍光がなくなった酵素を塩素原子存在下で反応性の高い酵素、すなわち塩化ナトリウムに対する安定性が向上した改変型カルボニル還元酵素として選抜した。選抜した酵素の培養液からプラスミドを抽出し、BigDye Terminator Cycle Sequencing Kit(アプライドバイオシステムズジャパン社製)およびApplied Biosystems 3130xlジェネティックアナライザ(アプライドバイオシステムズジャパン社製)を用いて変異型RBS遺伝子の塩基配列を決定し、変異部位を特定した。塩化ナトリウムに対する安定性の向上した改変型カルボニル還元酵素を表3に示す。

実施例2で取得したT2I-L46M変異酵素の変異型RBS遺伝子を含むプラスミドpNBSm1をテンプレートに実施例1と同様の方法で変異酵素ライブラリーを作製し、これを変異酵素ライブラリー2とした。
変異酵素ライブラリー2より有機塩化物の1つであるクロロアセトンに対する安定性が向上した改変型カルボニル還元酵素を選抜した。実施例5で作製した変異酵素ライブラリー2の各組換え菌及び実施例2で取得したT2I-L46M変異酵素を生産するE.coli HB101(pNBSm1)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液60μlに10mM EDTA・2Na及び1% Triton X-100を含むリン酸緩衝液(pH7.0)240μlを加え、37℃で1時間インキュベートした。各処理液を遠心し、上清を無細胞抽出液とした。各無細胞抽出液100μlに0.02% クロロアセトンを含むリン酸緩衝液(pH6.5)を加え、30℃で18時間インキュベートした(クロロアセトン処理)。クロロアセトン処理した各無細胞抽出液を96穴プレート(AGCテクノグラス社製)に50μl分注し、6mMのNADPHを含むリン酸緩衝液(pH6.5)50μlと20mM 3-キヌクリジノン塩酸塩を含むリン酸緩衝液(pH6.5)100μlを加え30℃で反応した。経時的にUVサンプル撮影装置FAS-III(東洋紡績社製)でNADPHの蛍光を観察した。対照であるE.coli HB101(pNBSm1)の無細胞抽出液(T2I-L46M変異酵素)に比べ、早く蛍光がなくなった酵素をクロロアセトンに対する安定性が向上した酵素として選抜した。実施例2と同様の方法で変異型RBS遺伝子の塩基配列を決定し、変異部位を特定した。得られたクロロアセトンに対する安定性が向上した改変型カルボニル還元酵素を表6に示す。
変異酵素ライブラリー2より無機塩化物の1つである塩化ナトリウムによる反応阻害に対する抵抗性が向上した改変型カルボニル還元酵素を選抜した。実施例5で作製した変異酵素ライブラリー2の各組換え菌及び実施例2で取得したT2I-L46M変異酵素を生産するE.coli HB101(pNBSm1)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液60μlに10mM EDTA・2Na及び1% Triton X-100を含むリン酸緩衝液(pH7.0)240μlを加え、37℃で1時間インキュベートした。各処理液を遠心し、上清を無細胞抽出液とした。無細胞抽出液を96穴プレート(AGCテクノグラス社製)に50μl分注し、6mMのNADPHを含むリン酸緩衝液(pH6.5)50μlと1.24M塩化ナトリウムと20mM 3-キヌクリジノン塩酸塩を含むリン酸緩衝液(pH6.5)100μlを加え30℃で反応した。経時的にUVサンプル撮影装置FAS-III(東洋紡績社製)でNADPHの蛍光を観察した。対照であるE.coli HB101(pNBSm1)の無細胞抽出液(T2I-L46M変異酵素)に比べ、早く蛍光がなくなった酵素を塩化ナトリウムによる反応阻害に対する抵抗性が向上した改変型カルボニル還元酵素として選抜した。実施例2と同様の方法で変異型RBS遺伝子の塩基配列を決定し、変異部位を特定した。得られた塩化ナトリウムによる反応阻害に対する抵抗性が向上した改変型カルボニル還元酵素を表7に示す。
実施例2で取得したプラスミドpNBSm01を鋳型として、プライマー5:5’-ATATATACATATGATCCACCCGCAACCCCG-3’(配列表の配列番号7)、プライマー6:5’-CGGCGATCCGCGGCAGGGAGCCGGCCCAGT-3’(配列表の配列番号8)を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちT2I、L46M、F68Sのアミノ酸置換を有するN末端側のポリペプチドをコードする二本鎖DNAを得た(N末端側DNA)。同様にプラスミドpNBSm01を鋳型として、プライマー7:5’-ACTGGGCCGGCTCCCTGCCGCGGATCGCCG-3’(配列表の配列番号9)、プライマー8:5’-TATAGGTACCTTATCAAAGCTTGTCGAACG-3’(配列表の配列番号10)を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちF68Sのアミノ酸置換を有するC末端側のポリペプチドをコードする二本鎖DNAを得た(C末端側DNA)。N末端側DNAとC末端側DNAを混合し、これを鋳型として、プライマー5、プライマー8を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちT2I、L46M、F68Sのアミノ酸置換を有するポリペプチドをコードする二本鎖DNAを得た。参考例2と同様の方法でpUCN18に導入し、pNBSm57を作製した。このpNBSm57を用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、改変型カルボニル還元酵素T2I-L46M-F68Sを生産する組換え生物E.coli HB101(pNBSm57)を得た。
実施例2で取得したプラスミドpNBSm01を鋳型として、プライマー9:5’-ATATATACATATGACCCACCCGCAACCCCG-3’(配列表の配列番号11)、プライマー6を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちL46M、F68Sのアミノ酸置換を有するN末端側のポリペプチドをコードする二本鎖DNAを得た(N末端側DNA)。同様にプラスミドpNBSm01を鋳型として、プライマー7、プライマー8を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちF68Sのアミノ酸置換を有するC末端側のポリペプチドをコードする二本鎖DNAを得た(C末端側DNA)。N末端側DNAとC末端側DNAを混合し、これを鋳型として、プライマー9、プライマー8を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちL46M、F68Sのアミノ酸置換を有するポリペプチドをコードする二本鎖DNAを得た。参考例2と同様の方法でpUCN18に導入し、pNBSm58を作製した。このpNBSm58を用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、改変型カルボニル還元酵素L46M-F68Sを生産する組換え生物E.coli HB101(pNBSm58)を得た。
参考例2で取得したプラスミドpNBSを鋳型として、プライマー5:5’-ATATATACATATGATCCACCCGCAACCCCG-3’(配列表の配列番号7)、プライマー8:5’-TATAGGTACCTTATCAAAGCTTGTCGAACG-3’(配列表の配列番号10)を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちT2Iのアミノ酸置換を有するポリペプチドをコードする二本鎖DNAを得た。参考例2と同様の方法でpUCN18に導入し、pNBSm59を作製した。このpNBSm59を用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、改変型カルボニル還元酵素T2Iを生産する組換え生物E.coli HB101(pNBSm59)を得た。
参考例2で取得したプラスミドpNBSm01を鋳型として、プライマー9:5’-ATATATACATATGACCCACCCGCAACCCCG-3’(配列表の配列番号11)、プライマー8:5’-TATAGGTACCTTATCAAAGCTTGTCGAACG-3’(配列表の配列番号10)を用いてPCRを行い、配列番号1に示すアミノ酸配列のうちL46Mのアミノ酸置換を有するポリペプチドをコードする二本鎖DNAを得た。参考例2と同様の方法でpUCN18に導入し、pNBSm60を作製した。このpNBSm60を用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、改変型カルボニル還元酵素L46Mを生産する組換え生物E.coli HB101(pNBSm60)を得た。
実施例2で取得した改変型カルボニル還元酵素の各組換え菌及び参考例3で作製したE.coli HB101(pNBS)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液について、遠心分離により菌体を集め、培養液と等量の100mMリン酸緩衝液(pH6.5)に懸濁した。これを、UH-50型超音波ホモゲナイザー(SMT社製)を用いて破砕した後、遠心分離により菌体残渣を除去し、無細胞抽出液を得た。無細胞抽出液500μlに1.24M塩化ナトリウムを含むリン酸緩衝液(pH7.0)500μlを加え、30℃でインキュベートした(塩化ナトリウム処理)。0時間目と20時間目の塩化ナトリウム処理液をサンプリングし、希釈して参考例4に示した方法で3-キヌクリジノンに対する還元活性を測定した。下記式より残存活性は算出し、この値を塩化ナトリウムに対する安定性の指標とした。

野生型酵素及び改変型カルボニル還元酵素の残存活性を表8に示す。
実施例2~9で取得した改変型カルボニル還元酵素の各組換え菌及び参考例3で作製したE.coli HB101(pNBS)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液について、遠心分離により菌体を集め、培養液と等量の100mMリン酸緩衝液(pH6.5)に懸濁した。これを、UH-50型超音波ホモゲナイザー(SMT社製)を用いて破砕した後、遠心分離により菌体残渣を除去し、無細胞抽出液を得た。無細胞抽出500μlに0.02%クロロアセトンを含むリン酸緩衝液(pH7.0)500μlを加え、30℃でインキュベートした(クロロアセトン処理)。0時間目と20時間目のクロロアセトン処理液をサンプリングし、希釈して参考例4に示した方法で3-キヌクリジノンに対する還元活性を測定した。下記式より残存活性は算出し、この値をクロロアセトンに対する安定性の指標とした。

野生型酵素及び改変型カルボニル還元酵素の残存活性を表9に示す。
実施例2~11で取得した改変型カルボニル還元酵素の各組換え菌及び参考例3で作製したE.coli HB101(pNBS)(対照)をそれぞれ参考例4と同様の方法で培養した。得られた各培養液について、遠心分離により菌体を集め、培養液と等量の100mMリン酸緩衝液(pH6.5)に懸濁した。これを、UH-50型超音波ホモゲナイザー(SMT社製)を用いて破砕した後、遠心分離により菌体残渣を除去し、無細胞抽出液を得た。塩化ナトリウム存在下での3-キヌクリジノンに対する還元活性を、0.62M 塩化ナトリウム、5mM 3-キヌクリジノン、0.25mM補酵素NADPHを含む100mMリン酸緩衝液(pH6.5)に、無細胞抽出液を添加して30℃で1分間反応を行い、波長340nmにおける吸光度の減少速度より算出した。また、塩化ナトリウム非存在下での3-キヌクリジノンに対する還元活性についても参考例4に示した方法で算出した。下記式より相対活性を算出し、この値を塩化ナトリウムによる反応阻害に対する抵抗性の指標とした。

野生型酵素及び改変型カルボニル還元酵素の相対活性を表10に示す。
参考例4と同様の方法で野生型酵素及び改変型カルボニル還元酵素T2I-L46Mを生産するE.coli HB101(pNBS01)およびT2I-R32H-L46Mを生産するE.coli HB101(pNBS43)を培養し、無細胞抽出液を得た。この無細胞抽出液の3-キヌクリジノン還元活性を測定し、これを阻害剤なしの活性とした。表11に示した化合物及び5mM 3-キヌクリジノン、0.25mM補酵素NADPHを含む100mMリン酸緩衝液(pH6.5)に、無細胞抽出液を添加して30℃で1分間反応を行い、波長340nmにおける吸光度の減少速度より3-キヌクリジノンに対する還元活性を算出した。

実施例2~11で取得した改変型カルボニル還元酵素を生産する各組換え菌をそれぞれ参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液1mlを得た。この無細胞抽出液1mlに、NADPH51mg、3-キヌクリジノン塩酸塩10mgを添加し、6N水酸化ナトリウムでpH6.5に調製し、30℃で1時間攪拌した。反応液の一部をサンプリングし、6N 水酸化ナトリウムでpH13以上とした後にn-ブタノールで抽出した。抽出液を後述の[ガスクロマトグラフィーによる分析条件(2)]の条件で分析することにより、3-キヌクリジノールの生成を確認した。その結果、取得したすべての改変型カルボニル還元酵素が3-キヌクリジノンを立体選択的に還元し、(R)-3-キヌクリジノールを生成していた。
野生型酵素を生産するE.coli HB101(pNBS)を参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液100mlを得た。この無細胞抽出液100mlに、グルコース脱水素酵素(商品名:GLUCDH”Amano”II、天野エンザイム社製)1000U、グルコース24g、NADP+3mg、塩化物として塩酸を含む3-キヌクリジノン塩酸塩19gを添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.5に調整しながら、30℃で20時間攪拌した。反応液の一部をサンプリングし、6N水酸化ナトリウムでpH13以上とした後にn-ブタノールで抽出した。抽出液を下記の[ガスクロマトグラフィーによる分析条件(1)]で分析し、(R)-3-キヌクリジノールへの変換率を測定した。また、抽出液を下記の[ガスクロマトグラフィーによる分析条件(2)]の条件で分析することにより、(R)-3-キヌクリジノールの光学純度を測定した。その結果、(R)-3-キヌクリジノールへの変換率は45%であり、その光学純度は99%e.e.以上であった。野生型酵素は塩化物存在下で反応阻害、酵素失活を受けるため変換率が低かった。

塩化物非存在下で野生型酵素を用いて、(R)-3-キヌクリジノールを製造した。3-キヌクリジノン塩酸塩を等量の水に溶解し、水酸化ナトリウムでpH12に調整した後、2倍量のn-ブタノールで3回抽出した。抽出液を減圧濃縮し、フリー体3-キヌクリジノンの結晶を得た。野生型酵素を生産するE.coli HB101(pNBS)を参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液100mlを得た。この無細胞抽出液100mlに、グルコース脱水素酵素(商品名:GLUCDH”Amano”II、天野エンザイム社製)1000U、グルコース24g、NADP+3mgを添加し、硫酸でpH6.5に調整しながら3-キヌクリジノン15gを加えた。5Nの水酸化ナトリウム水溶液を滴下することによりpH6.5に調整しながら、30℃で20時間攪拌した。反応液の一部をサンプリングし、6N水酸化ナトリウムでpH13以上とした後にn-ブタノールで抽出した。抽出液を前記の[ガスクロマトグラフィーによる分析条件(1)]で分析し、(R)-3-キヌクリジノールへの変換率を測定した。また、抽出液を下記の[ガスクロマトグラフィーによる分析条件(2)]の条件で分析することにより、(R)-3-キヌクリジノールの光学純度を測定した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。野生型酵素は塩化物非存在下では比較例1に比べ変換率が高かった。しかし、本法ではフリー体3-キヌクリジノンを調製する工程が必要であり、効率的な製造法ではなかった。
実施例2で取得した改変型カルボニル還元酵素T2I-L46Mを生産するE.coli HB101(pNBSm01)を参考例4と同様の方法で無細胞抽出液を調製し、比較例1と同様の方法でキヌクリジノン還元反応を実施した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。
実施例2で取得した改変型カルボニル還元酵素F68Sを生産するE.coli HB101(pNBSm05)について、実施例17と同様の方法でキヌクリジノン還元反応を実施した。その結果、変換率は100%、光学純度は99%e.e.以上であった。
実施例2で取得した改変型カルボニル還元酵素D95Gを生産するE.coli HB101(pNBSm10)について、実施例17と同様の方法でキヌクリジノン還元反応を実施した。その結果、変換率は100%、光学純度は99%e.e.以上であった。
実施例6で取得した改変型カルボニル還元酵素T2I-R32H-L46Mを生産するE.coli HB101(pNBSm43)について実施例20と同様の方法でキヌクリジノン還元反応を実施した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。
実施例6で取得した改変型カルボニル還元酵素T2I-A45S-L46M-G90Sを生産するE.coli HB101(pNBSm44)について実施例20と同様の方法でキヌクリジノン還元反応を実施した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。
実施例6で取得した改変型カルボニル還元酵素T2I-A45T-L46Mを生産するE.coli HB101(pNBSm45)について実施例20と同様の方法でキヌクリジノン還元反応を実施した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。
プライマー10:5’-GCCGAATTCTAAGGAGGTTAACAATGTATAAAGATTTAGAAGG-3’(配列表の配列番号12)と、プライマー11:5’-CTGCAGGTCGACTTATCCGCGTCCTGCTTGG-3’(配列表の配列番号13)を用い、プラスミドpGDK1(Eur.J.Biochem.186,389(1989))に記載の方法で取得及び調製可能)を鋳型としてPCRを行い、バシラス・メガテリウム(Bacillus megaterium)IAM1030株由来のグルコース脱水素酵素(以後、GDHと呼ぶ)遺伝子の開始コドンから5塩基上流に大腸菌のリボゾーム結合配列に、さらにその直前にEcoRI認識部位が付加され、かつ、終止コドンの直後にSalIおよびPstI認識部位が付加された二本鎖DNA(GDH遺伝子)を取得した。得られたGDH遺伝子をEcoRIおよびPstIで消化し、プラスミドpSTV28(タカラバイオ社製)のEcoRI-PstI部位に挿入して、組換えベクターpSTVGを構築した。
参考例2で構築した組換えベクターpNBSおよび参考例7で構築したpSTVGの2つの組換えベクターを用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、組換え生物E.coli HB101(pNBS,pSTVG)を得た。また、pUCN18及びpSTV28を用いてE.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、組換え生物E.coli HB101(pUCN18,pSTV28)を得た。
参考例8で得た2種の組換え生物(E.coli HB101(pNBS,pSTVG)、E.coli HB101(pUCN18,pSTV28))、それぞれを、200μg/mlのアンピシリンおよび100μg/mlのクロラムフェニコールを含む2×YT培地(トリプトン1.6%、イーストエキス1.0%、塩化ナトリウム0.5%、pH7.0)5mlに接種し、37℃で24時間振盪培養した。
それぞれの組換え生物の3-キヌクリジノン還元活性およびGDH活性を以下に示す。
実施例2で取得した改変型カルボニル還元酵素T2I-L46Mをコードする遺伝子を含む組換えベクターpNBSm01および参考例5で構築したpSTVGの2つの組換えベクターを用いて、E.coli HB101コンピテントセル(タカラバイオ社製)を形質転換し、組換え生物E.coli HB101(pNBSm01,pSTVG)を得た。
実施例24で取得したE.coli HB101(pNBSm01,pSTVG)を参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液100mlを得た。この無細胞抽出液100mlにグルコース24g、NADP+3mg、3-キヌクリジノン塩酸塩19gを添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.5に調整しながら、30℃で20時間攪拌した。反応液の一部をサンプリングし、6N水酸化ナトリウムでpH13以上とした後にn-ブタノールで抽出した。抽出液を比較例1と同様の方法で分析した。その結果、(R)-3-キヌクリジノールへの変換率は100%であり、その光学純度は99%e.e.以上であった。
野生型酵素を生産するE.coli HB101(pNBS)を参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液100mlを得た。この無細胞抽出液100mlに、グルコース脱水素酵素(商品名:GLUCDH”Amano”II、天野エンザイム社製)1000U、グルコース30g、トルエン10g、NADP+10mg、3-クロロ-1-(2-チエニル)プロパン-1-オンを15g添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.25に調整しながら、30℃で攪拌した。反応開始後24時間目に反応液の一部をサンプリングし、酢酸エチルで抽出した。抽出液を下記の高速液体クロマトグラフィーの条件で分析し、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールへの変換率とその光学純度を測定した。その結果、変換率は78%であり、その光学純度は99%e.e.であった。

実施例2で取得した改変型カルボニル還元酵素T2I-L46Mを生産するE.coli HB101(pNBSm01)について比較例3と同様の方法で3-クロロ-1-(2-チエニル)プロパン-1-オン還元反応を実施した。反応終了後、比較例2と同様の方法で分析した。その結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの変換率は95%であり、その光学純度は99%e.e.であった。
実施例2で取得した改変型カルボニル還元酵素F68Sを生産するE.coli HB101(pNBSm05)について実施例26と同様の方法で3-クロロ-1-(2-チエニル)プロパン-1-オン還元反応を実施した。その結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの変換率は92%であり、その光学純度は99%e.e.以上であった。
実施例2で取得した改変型カルボニル還元酵素D95Gを生産するE.coli HB101(pNBSm10)について実施例26と同様の方法で3-クロロ-1-(2-チエニル)プロパン-1-オン還元反応を実施した。その結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの変換率は96%であり、その光学純度は99%e.e.以上であった。
実施例6で取得した改変型カルボニル還元酵素T2I-R32H-L46Mを生産するE.coli HB101(pNBSm43)について実施例26と同様の方法で3-クロロ-1-(2-チエニル)プロパン-1-オン還元反応を実施した。その結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの変換率は98%であり、その光学純度は99%e.e.以上であった。
実施例24で取得したE.coli HB101(pNBSm01,pSTVG)を参考例4と同様に培養し、培養液100mlを得た。この培養液100mlにグルコース30g、トルエン10g、NADP+10mg、3-クロロ-1-(2-チエニル)プロパン-1-オンを15g添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.25に調整しながら、30℃で攪拌した。反応開始後24時間目に反応液の一部をサンプリングし、酢酸エチルで抽出した。抽出液を比較例2と同様の方法で分析した。その結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの変換率は96%であり、その光学純度は99%e.e.以上であった。
(工程1)
実施例24で取得したE.coli HB101(pNBSm01,pSTVG)を参考例4と同様に培養後、超音波ホモゲナイザーによる菌体破砕を実施し、無細胞抽出液200mlを得た。この無細胞抽出液200mlにグルコース48g、NADP+6mg、3-キヌクリジノン塩酸塩38gを添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.5に調整しながら、30℃で20時間攪拌した。20時間後、反応液を比較例1と同様の方法で分析した。その結果、(R)-3-キヌクリジノールの光学純度は99%e.e.以上であった。反応液を6N水酸化ナトリウムでpH13以上とした後にn-ブタノールで抽出し、抽出液を減圧乾燥し、結晶を得た。この結晶を再結晶化し、(R)-3-キヌクリジノール21gの結晶を得た。
ジクロロメタン200mlに(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリン10.6gと炭酸カリウム10.6gを加え、氷中で冷却しながら、ジクロロメタン20mlとクロロぎ酸エチル5.8mlを混合した溶液を添加し、室温で一晩攪拌した。一晩後、反応液に水を添加し、室温で30分攪拌した。有機層を分離した後、水と飽和食塩水で洗浄し、硫酸マグネシウムで脱水した後、減圧濃縮した。濃縮液をシリカゲルクロマトグラフィーで精製し、(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリン-2-カルボン酸を得た。
トルエン120mlに精製した(S)-1-フェニル-1,2,3,4-テトラヒドロイソキノリン-2-カルボン酸12gと(R)-3-キヌクリジノール16.3gを加え、水素化ナトリウム1.7gを添加した後、140℃で3時間加熱した。反応後、室温まで冷却し、飽和食塩水を添加した後、酢酸エチルで抽出した。抽出液を水で洗浄した後、20%塩酸で抽出した。水層を1M水酸化ナトリウムでpH10に調整した後、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄した後、硫酸マグネシウムで脱水した。抽出液を減圧濃縮した後、塩酸を加えたアセトニトリルとジエチルエーテル中で結晶化し、無色の結晶の(1S,3´R)-キヌクリジン-3-イル-1-フェニル-1,2,3,4-テトラヒドロイソキノリン-2-カルボン酸塩酸塩10.1gを得た。
(工程1)
実施例24で取得したE.coli HB101(pNBSm01,pSTVG)を参考例4と同様に培養し、培養液200mlを得た。この培養液100mlにグルコース60g、トルエン20g、NADP+20mg、3-クロロ-1-(2-チエニル)プロパン-1-オンを30g添加し、5Nの水酸化ナトリウム水溶液を滴下することによりpH6.25に調整しながら、30℃で24時間攪拌した。反応後、トルエンで抽出し、有機層を減圧濃縮し、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オール26gを得た。濃縮液を比較例2と同様の方法で分析した結果、(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールの光学純度は99%e.e.以上であった。
40%メチルアミン/メタノール溶液93gに(S)-3-クロロ-1-(2-チエニル)プロパン-1-オール21.6gを室温で滴下し、30分間反応させた。メタノール及びメチルアミンを減圧下で留去した後、水を加えメチル-t-ブチルエーテルで抽出した。有機層を食塩水で洗浄した後、減圧濃縮した。濃縮物にn-ヘプタンを加え、析出した固体をろ別し、(S)-3-N-メチルアミノ-1-(2-チエニル)プロパン-1-オールの微黄色結晶17.4gを得た。
ペンタンで洗浄した水素化ナトリウム5gをジメチルアセトアミドに懸濁し、(S)-3-N-メチルアミノ-1-(2-チエニル)プロパン-1-オール17gを添加した後、70℃で加熱しながら攪拌した。溶液が透明になった後、1-フルオロナフタレン26gを添加し、90℃で2時間攪拌した。反応後、水を添加し、ジエチルエーテルで抽出した。有機層を飽和食塩水で洗浄した後、減圧濃縮した。濃縮液をカラムクロマトグラフィーで精製し、(S)-N-メチル-3-(1-ナフタレニルオキシ)-3-(2-チオフェン)-プロパンアミン17gを得た。次に(S)-N-メチル-3-(1-ナフタレニルオキシ)-3-(2-チオフェン)-プロパンアミンを酢酸エチルに溶解した後、0.44M塩酸/酢酸エチル溶液を滴下し、結晶化させた。室温で1時間攪拌後、冷却し、ジエチルエーテルを混合して、結晶をろ別し、(S)-N-メチル-3-(1-ナフタレニルオキシ)-3-(2-チオフェン)-プロパンアミン塩酸塩15gを得た。
参考例3で作製したE. coli HB101(pNBS)(対照)を参考例4と同様の方法で培養した。得られた培養液について、遠心分離により菌体を集め、培養液と等量の100mMリン酸緩衝液(pH6.5)に懸濁した。これを、UH-50型超音波ホモゲナイザー(SMT社製)を用いて破砕した後、遠心分離により菌体残渣を除去し、無細胞抽出液を得た。3-キヌクリジノンに対する還元活性を、5mM 3-キヌクリジノン、0.025mM~0.25mM 補酵素NADPHを含む100mMリン酸緩衝液(pH6.5)に、無細胞抽出液を添加して30℃で1分間反応を行い、波長340nmにおける吸光度の減少速度より算出した。また、0.62M 塩化ナトリウム存在下での3-キヌクリジノンに対する還元活性についても同様に補酵素濃度を変えて測定した。各測定結果のラインウィーバー・バーク・プロットを図2に示す。
実施例2及び実施例11で取得した改変型カルボニル還元酵素T2I-L46M、F68S、D95G及びL46Mをそれぞれ生産するE.coli HB101(pNBSm01)、E.coli HB101(pNBSm05)、E.coli HB101(pNBSm10)、E.coli HB101(pNBSm60)について比較例4と同様の方法で塩化ナトリウム非存在下でのNADPHに対するKm値を算出した。

から選択される1つもしくは複数のアミノ酸置換が導入されており、配列表の配列番号1に示すアミノ酸配列からなるカルボニル還元酵素と比較して、塩化物イオン存在下での安定性が向上している請求項4に記載のポリペプチド。 - 配列表の配列番号1に示すアミノ酸配列に下記の(1)~(17);

- 前記(a)~(c)に加え、さらに
(d)配列表の配列番号1に示すアミノ酸配列からなるカルボニル還元酵素と比較して、補酵素に対する親和性が高い、
の性質を示す、請求項1または2に記載のポリペプチド。 - 配列表の配列番号1に示すアミノ酸配列のうち次の群;
2、46、68、および95番目、
から選択される1つもしくは複数のアミノ酸が置換されている、請求項13に記載のポリペプチド。 - 配列表の配列番号1に示すアミノ酸配列のうち次の群;
2番目がイソロイシン、46番目がメチオニン、68番目がセリン、および95番目がグリシン、
から選択される1つもしくは複数のアミノ酸置換が導入されている、請求項14に記載のポリペプチド。 - 請求項1~15のいずれか1項に記載のポリペプチドをコードするポリヌクレオチド。
- 請求項16に記載のポリヌクレオチドを含むベクター。
- 還元型補酵素再生能を有するポリペプチドをコードするポリヌクレオチドをさらに含む、請求17に記載のベクター。
- 還元型補酵素再生能を有するポリペプチドがグルコース脱水素酵素である、請求項18に記載のベクター。
- 請求項17~19のいずれかに記載のベクターにより宿主細胞を形質転換して得られる形質転換体。
- 前記宿主細胞が大腸菌である請求項20記載の形質転換体。
- 請求項1~15のいずれかに記載のポリペプチド、または、請求項20もしくは請求項21に記載の形質転換体および/またはその処理物を、カルボニル化合物に作用させることを特徴とする、アルコール化合物の製造方法。
- 前記カルボニル化合物が非対称ケトンであり、生成物が光学活性アルコールである、請求項22に記載の製造方法。
- 前記カルボニル基を有する化合物が、3-キヌクリジノンもしくはその塩、3-クロロ-1-(2-チエニル)プロパン-1-オン、3-クロロ-1-(2-フェニル)プロパン-1-オンのいずれかである請求項23に記載の製造方法。
- 請求項1~15のいずれかに記載のポリペプチド、または、請求項20もしくは請求項21に記載の形質転換体および/またはその処理物を、カルボニル化合物に作用させて、光学活性アルコールを得る工程を有することを特徴とする、医薬品原薬の製造方法。
- 前記光学活性アルコールが(R)-3-キヌクリジノンであり医薬品原薬がソリフェナシン、または、前記光学活性アルコールが(S)-3-クロロ-1-(2-チエニル)プロパン-1-オールであり医薬品原薬がデュロキセチンである、請求項26に記載の製造方法。
